Verfahren zur Herstellung von neuen Derivaten des Sulfanilamids
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen Derivaten des Sulfanilamids.
Verbindungen der allgemeinen Formel I

in welcher Rl eine Alkyl- oder Alkenylgruppe von höchstens 5 Koh lenstoffatomen oder eine Cycloalkyl- oder Cycloalke nylgruppe von höchstens 7 Kohlenstoffatomen und R die Methyl- oder Äthylgruppe bedeutet, und ihre Additionssalze mit anorganischen oder organischen Säuren sind bisher nicht bekannt geworden.
Wie nun gefunden wurde, besitzen diese Verbindungen wertvolle pharmakologische Eigenschaften, insbesondere das l-Sulfanilyl-2-imino-3-butyl-5-äthyl-, das 1 -Sulfanilyl-2-imino-3-tertbutyl-4-methyl- und das l-Sulf anilyl - 2 - imino-3-tert.butyl-5-methyl-imidazolidin weisen bei peroraler oder parenteraler Verabreichung hypoglykämische Wirkung auf, die sie als geeignet zur Behandlung der Zuckerkrankheit charakterisieren. Die hypoglykämische Wirkung wird an Standardversuchen an Warmblütern, z.B. an Kaninchen und Ratten, nachgewiesen.
In den Verbindungen der allgemeinen Formel I kann Rl als niedere Alkylgruppen z.B. die Methyl-, Äthyl-, Propyl-, Isopropyl-, Butyl-, sek.Butyl-, tert.Butyl-, Isobutyl-, Pentyl-, Isopentyl-, 2,2-Dimethyl-propyl-, l-Me- thyl-butyl-, l-Äthyl-propyl-, 1 ,2-Dimethyl-propylgruppe und als Alkenylgruppe z.B. die Allyl-, die 1-, 2- oder 3-Buten-l-yl-, die 2-Methyl-propenyl-, Methallyl- oder die 3- oder 4-Penten-l-ylgruppe sein.
Ferner kann Rl beispielsweise sein: als Cycloalkylgruppe die Cyclopropyl-, Cyclopropylmethyl-, 2-Cyclopropyl-äthyl-, 3-Cyclopropyl -propyl-, Cyclobutyl-, Cyclobutylmethyl-, 2-Cyclobutyl -äthyl-, 3-Cyclobutyl-propyl-, Cyclopentyl-, l-Methyl-cy- clopentyl-, 2-Methyl-cyclopentyl-, 3-Methyl-cyclopentyl-, l-Äthyl-cyclopentyl-, 2-Äthyl-cyclopentyl-, 3-Äthyl-cyclopentyl-, Cyclopentylmethyl-, 2-Methyl-cyclopentylmethyl-, 3-Methyl-cyclopentylmethyl-, Cyclohexyl-, l-Methyl-cyclohexyl-, 2-Methyl-cyclohexyl-, 3-Methyl-cyclohexyl-, 4 -Methyl-cyclohexyl-,

Rest bedeutet oder, falls R Wasserstoff ist, auch die freie Aminogruppe bedeutet, mit einem reaktionsfähigen Cyansäurederivat umsetzt und cyclisiert, nötigenfalls das Reaktionsprodukt zur Umwandlung der Gruppe X in die freie Aminogruppe hydrolysiert, reduziert oder reduktiv spaltet und gegebenenfalls die erhaltene Verbindung mit einer anorganischen oder organischen Säure in ein Additionssalz überführt.
R. ist als Arylmetllyl-, Diarylmethyl- oder Triarylmethylgruppe beispielsweise die Benzyl-, Benzhydrylbzw. die Tritylgruppe.
Als reaktionsfähige Cyansäurederivate eignen sich Ha logencyane, wie Chlorcyan oder Bromcyan, oder Cyansäureester, insbesondere Cyansäurephenylester. Die Umsetzung erfolgt vorzugsweise in Gegenwart eines mit 'Wasser mischbaren oder nicht mischbaren inerten organi edlen Lösungsmittels in An- oder Abwesenheit von Wasser. Geeignete inerte organische Lösungsmittel sind beispielsweise Kohlenwasserstoffe, wie Benzol, Toluol oder Xylol, niedere Alkanole, wie Methanol oder Äthanol, ätherartige Flüssigkeiten, wie Äther, Dioxan oder Tetrahydrofuran, chlorierte Kohlenwasserstoffe, wie Methylenchlorid, niedere Ketone, wie Aceton oder Methyläthylketon, Carbonsäureester. wie Aceton oder Methyläthylketon, Carbonsäureester, wie Essigsäureäthylester.
Carbonsäurenitrile, wie Acetonitril, oder Sulfone, wie Tetra llvdro-thiophen-l,l-dioxid. Die Umsetzung kann in Anwesenheit oder Abwesenheit eines säurebindenden Mittels vorgenommen werden. Als säurebindende Mittel eignen sich anorganische Basen oder Salze, z.B. Alkalimetallhydroxide, Alkalimetallhydrogencarbonate, Alkalimetallcarbonate oder Alkalimetallphosphate, wie die entsprechenden Natrium- oder Kaliumverbindungen. Ferner können auch Calciumcarbonate sowie Calciumphosphate und Magnesiumcarbonat eingesetzt werden.
Die anschliessende Umwandlung der Gruppe X des Reaktionsproduktes in die freie Aminogruppe, welche dieses in eine Verbindung der allgemeinen Formel I überführt, wird je nach der Art der Gruppe X durch eine Hydrolyse, Reduktion oder reduktive Spaltung vorgenommen.
Durch Hydrolyse in die freie Aminogruppe überführbare Reste X sind beispielsweise Acylaminoreste, wie z.B. die Acetamidogruppe. Ferner sind solche Reste nie dere Alkoxyearbonylaminoreste, wie z.B. die Äthoxycarbonylaminogruppe, Aryloxycarbonylaminoreste, wie der Phenoxycarbonylaminorest, oder Arylmethoxycarbo- nylaminoreste, wie der Benzyloxycarbonylaminorest, oder Reste von entsprechenden Thiokohlensäurederivaten.
Weitere Beispiele sind substituierte Methylenaininoreste wie z.B. die Benzylidenamino- oder die p-Dimethylamino -benzylidenaminogruppe. Die Hydrolyse zur Freisetzung der Aminogruppe kann in saurem Medium, z.B. in methanolischer Salzsäure, oder in verdünnter wässriger Salzsäure oder Schwefelsäure erfolgen oder, falls X durch einen Alkoxycarbonylaminorest verkörpert ist, auch unter milden alkalischen Bedingungen, z.B. mittels l-n oder
2-n Natronlauge, vorgenommen werden.
Ein Beispiel für einen durch Reduktion in die Aminogruppe überführbaren Rest X ist die Nitrogruppe und Beispiele für solche Reste, die durch reduktive Spaltung zur Aminogruppe führen, sind die Phenylazo- oder p-Di methylamino-phenylazogruppen. Die Reduktion dieser Reste kann allgemein katalytisch, z.B. mittels Wasserstoff in Gegenwart von Raney-Nickel, Palladium- oder Platin Kohle, in einem inerten Lösungsmittel, wie z.B. Äthanol, erfolgen. Neben diesen kommen auch andere übliche Rcduktionsverfahren in Betracht, beispielsweise die Reduktion von Nitrogruppen oder die reduktive Spaltung von Azogruppen mittels Eisen in Essigsäure oder Salzsäure.
Als Ausgangsstoffe der allgemeinen Formel II eignen sich solche Verbindungen, deren Symbole Rl, R., R5 und X mit den Gruppen übereinstimmen, die anschliessend an Formel I bzw. II aufgezählt sind. Solche Ausgangsstoffe sind beispielsweise N-(2-Amino-äthyl)-benzosulfonamide, deren Benzolring durch den Rest X in para-Stellung, deren Aminogruppe durch die Reste R1 sowie Rs und deren Äthylgruppe in 1- oder 2-Stellung durch die Methyl- oder Äthylgruppe substituiert ist.
Eine Gruppe von solchen Verbindungen kann z.B. hergestellt werden, wenn man von einem l-Pilenylsulfonyl-2-methyl- oder l-Phenylsulfonyl-2-äthyl-aziridin ausgeht welches durch den Rest X substituiert ist, und das Aziridin mit einem primären oder sekundären Amin, das die Reste R1 und R, enthält, umsetzt. Vertreter der genannten Aziridine sind in der Literatur beschrieben [vgl. A. Weissenberger, Heterocyclic Compounds with Three- and Four-Membered Rings, Part. One. John Wiley & Sons Inc. London (l964). Weitere Aziridine von diesem Typus können analog hergestellt werden.
Die nach dem erfindungsgemässen Verfahren erhaltenen Verbindungen der allgemeinen Formel I werden anschliessend gewünschtenfalls in ihre Salze mit anorganischen sowie organischen Säuren übergeführt. Die Herstellung dieser Salze erfolgt z.B. durch Umsetzung der Verbindungen der allgemeinen Formel 1 mit der äquivalenten Menge einer Säure in einem geeigneten wässrigorganischen oder organischen Lösungsmittel, wie z.B.
Methanol, Äthanol, Diäthyläther, Chloroform oder Methylenchlorid.
Zur Verwendung als Arzneistoffe können anstelle der freien Verbindungen der allgemeinen Formel I deren pharmazeutisch annehmbare Salze mit Säuren eingesetzt werden. Geeignete Additionssalze sind z.B. Salze mit Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Methansulfonsäure, Äthansulfonsäure, -Hydroxyäthansulfonsäure, Essigsäure, Milchsäure, Oxalsäure, Bernsteinsäure, Fumarsäure, Maleinsäure, Äpfelsäure, Weinsäure, Citronensäure, Benzoesäure, Salicylsäure, Phenylessigsäure, Mandelsäure und Embonsäure.
Die nachfolgenden Beispiele erläutern die Herstellung der neuen Verbindungen der allgemeinen Formel I und von bisher nicht beschriebenen Zwischenprodukten näher, stellen jedoch keineswegs die einzigen Ausführungsformen derselben dar. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel I a) 32,7 g N1 - (1 - Methyl-2-tert.butylamino-äthyl)-N4 -acetyl-sulfanilamid werden in 100 ml 2-n Natronlauge gelöst und unter Kühlung bei 20 - 300 mit 10,6 g Bromcyan versetzt. Das Rohprodukt fällt gleich kristallin aus.
Es wird nach 30 Minuten abgenutscht und aus N,N-Di methylformamid umkristallisiert. Das erhaltene 4'-(2-Imino - 3 - tert.butyl- 5-methyl-imidazolidin-1 -ylsuifonyl)-acet- anilid schmilzt bei 243 - 2440, b) Zu 125 g auf 400 erwärmte 50%ige Schwefelsäure werden unter Rühren 35,2 g des nach a) erhaltenen Acetanilids gegeben. Man erwärmt das Gemisch 2 Stunden auf 500 und giesst die Lösung anschliessend unter Rühren zu 750 ml Wasser. Das Rohprodukt fällt als Sulfat aus. Das pH des Reaktionsgemisches wird mit
10-n Natronlauge auf 6,3 eingestellt. Das Gemisch löst sich bis auf eine geringe Verunreinigung; es wird mit 2 g Aktivkohle 15 Minuten gerührt und über Hyflo (gereinigte Diatomeenerde) filtriert.
Man stellt das Filtrat mit 10-n Natronlauge alkalisch. Das Rohprodukt fällt aus.
Es wird abfiltriert, mit Wasser gewaschen, bei 900 getrocknet und aus Essigsäureäthylester umkristallisiert. Das erhaltene 1 -Sulfanilyl-2-imino-3-tert.butyl-5-methyl-imid- azolidin schmilzt bei 145 - 1460.
c) Man erhält die unter a) genannte Ausgangsverbindung wie folgt: Eine Lösung von 23,3 g N-Acetyl-sulfanilylchlorid in 70ml Aceton wird unter Rühren bei -100 bis 0 zu 5,6 g 2-Methyl-aziridin in 30 ml 4-n Natronlauge getropft. Man rührt das Reaktionsgemisch bei 0 2 Stunden weiter und giesst es anschliessend auf 400 ml Eiswasser. Die erhaltene Suspension wird abfiltriert, der Niederschlag, das rohe 4-(2-Methyl-aziridin-1 -ylsulfonyl)-acetanilid, mit Wasser gewaschen und gleich weiter verarbeitet, da es leicht polymerisiert.
d) Man löst das nach c) erhaltene Acetanilid in der Kälte in 100ml Dioxan und 15 mol Wasser und tropft zur Lösung 100 ml tert. Butylamin. Dann wird das Reaktionsgemisch eine Stunde unter Rückfluss gekocht und das überschüssige tert. Butylamin sowie das Dioxan abdestilliert. Man kristallisiert den kristallinen Rückstand aus Essigsäureäthylester um, wonach das reine N'-(1-Methyl- -2-tert.butylamino-äthyl)-N4-acetyl-sulfanilamid bei 93 bis 950 schmilzt.
Beispiel 2 a) Analog Beispiel 1 a) erhält man ausgehend von 28,5 g N1-(1 -Methyl-2-methylamino-äthyl)-N4-acetyl-sulf- anilamid (Rohprodukt) mit 10,6g Bromcyan das 4'-(2 -Imino -3,5- dimethyl-imidazolidin-1-ylsulfonyl)-acetanilid vom Smp. 258 - 2600, von dem 31,0g analog Beispiel 1 b) mit 125 g 50%iger Schwefelsäure zum Endprodukt 1 -Sulfanilyl-2-imino-3,5-dimethyl-imidazolidin vom Smp.
196 - 1970 hydrolysiert werden.
b) Die Ausgangssubstanz von a) erhält man analog Beispiel 1 c) aus 25,4 g 4'-(2-Methyl-aziridin- 1 -ylsulfonyl)- -acetanilid (vgl. Beispiel 1 c) und 200 ml 33%igem Methylamin in Äthanol.
Beispiel 3 a) Analog Beispiel 1 a) erhält man ausgehend von 29,9 g N1 - (1 - Methyl-2-äthylamino-äthyl)-N4-acetyl-sulfanilamid (Rohprodukt) mit 10,6g Bromcyan das 4'-(2 -Imino - 3 - äthyl-5-methyl-imidazolidin-1-ylsulfonyl) - acetanilid vom Smp. 229 - 2500, von dem 324 g analog Beispiel 1 b) mit 125 g 50%iger Schwefelsäure zum Endprodukt 1 - Sulfanilyl-2-imino-3-äthyl-5-methyl-imidazolidin hydrolysiert werden.
b) Die Ausgangssubstanz von a) erhält man analog Beispiel 1 c) aus 25,4 g 4-(2-Methyl-aziridin-1-ylsulfonyl)- -acetanilid (vgl. Beispiel 1 c) und 250 ml 20%igem Äthylamin in Benzol.
Beispiel 4 a) Analog Beispiel 1 a) erhält man ausgehend von 31,3 g NÚ-(1-Methyl-2-propylamino-äthyl)-NÚ-acetyl-sul- fanilamid vom Smp. 162 - 1630 mit 10,6 g Bromcyan das 4' -(2-Imino-3 -propyl-5-methyl-imidazolidin- 1 -ylsulfonyl) -acetanilid vom Smp. 254-2550, von dem 33,8 g analog Beispiel 1 b) mit 125 g 50%iger Schwefelsäure zum Endprodukt l-Sulfanilyl-2-imino-3-propyl-5-methyl-imidazol- idin vom Smp. 146-1480 hydrolysiert werden.
b) Die Ausgangssubstanz von a) erhält man analog Beispiel 1 c) aus 25,4 g 4'-(2-Methyl-aziridin-1-ylsulfonyl) -acetanilid (vgl. Beispiel 1 c) und 100 ml Propylamin.
Beispiel 5 a) Analog Beispiel 1 a) erhält man ausgehend von 32,7 g N1 -(1 -Methyl-2-butylamino-äthyl)-N4-acetyl-sulfanilamid vom Smp. 166-1670 mit 10,6 g Bromcyan das 4'-(2 -Imino-3-butyl-5-methyl-imidazolidin- 1 -ylsulfonyl) - acetanilid vom Smp. 243-244 , von dem 35,2 g analog Beispiel 1 b) mit 125 g 50%iger Schwefelsäure zum Endprodukt 1 -Sulfanilyl-2-imino-3 -butyl-5 - methyl-imidazolidin vom Smp. 125.1260 hydrolysiert werden.
b) Die Ausgangssubstanz von a) erhält man analog Beispiel 1 c) aus 25,4 g 4'-(2-Methyl-aziridin-1-ylsulfonyl)- -acetanilid (vgl. Beispiel 1 c) und 100 ml Butylamin.
Beispiel 6 a) Analog Beispiel 1 a) erhält man ausgehend von 32,7 g N'-(1-Methyl-2-sek.butylamino-äthyl)-N4-acetyl- -sullanilamid vom Smp. 118-1200 mit 10,6 g Bromcyan das 4' - (2 - Ii-nino - 3- sek.butyl-5-methyl-imidazolidin-1-yl sulfonyl)-acetanilid vom Smp. 242.2440, von dem 35,2 g analog Beispiel 1 b) mit 125 g 50%iger Schwefelsäure zum Endprodukt 1-Sulfanilyl-2-imino-3-sek.butyl-5-me- thyl-imidazolidin vom Smp. 194-1950 hydrolysiert werden.
b) Die Ausgangssulistanz von a) erhält man analog Beispiel 1 c) aus 25,4 g 4'-(2-Methyl-aziridin- l-ylsulfonyl)- acetanilid (vgl. Beispiel 1 c) und 100 ml sek. Butylamin.
Beispiel 7 a) Analog Beispiel 1 a) erhält man ausgehend von 34,1 g N'-(l-Methyl-2-pentylaminoWäthyl)-N4-acetyl-sulf- anilamid (Rohprodukt) mit 10,6 g Bromcyan das 4'-(2 -Imino-3 -pentyl-5 -methyl-imidazolidin- 1 -ylsulfonyl)-acetanilid vom Smp. 228.2290, von dem 36,4 g analog Beispiel 1 b) mit 125 g 50%iger Schwefelsäure zum Endprodukt 1 -Sulfanilyl-2-im ino3 -pentyl-5-methvl-im idazolidin vom Smp. 127.1280 hydrolysiert werden.
b Die Ausgangssubstanz von a) erhält man analog Beispiel 1 c) aus 25,4 g 4'.(2-Methyl-aziridin.1-ylsulfonyl)- -acetanilid (vgl. Beispiel 1 c) und 100 ml Pentylamin.
Beispiel 8 a) Analog Beispiel 1 a) erhält man ausgehend von 35,1 g Nl -Methyl-2-cyclohexylamino-äthyl)-N4-acetyl- -sulfanilamid (Rohprodukt) mit 10,6 g Bromcyan das 4' .(2-Imino-3-cyclohexyl-5-methyl-imidazolidin- 1 -ylsulfonyl)-acetanilid vom Smp. 235.2370, von dem 37,6 g analog Beispiel 1 b) mit 125 g 50%iger Schwefelsäure zum Endprodukt 1 -Sulfanilyl-2-iinino-3 -cydohexyl-5.methyl-imid- azolidin vom Smp. 217.2180 hydrolysiert werden.
b) Die Ausgangssubstanz von a) erhält man analog Beispiel 1 c) aus 25,4 g 4'-(2-Methyl-aziridin- 1 -ylsulfonyl).
-acetanilid (vgl. Beispiel 1 c) und 100 ml Cyclohexylamin.
Beispiel 9 a) Analog Beispiel 1 aj erhält man ausgehend von 34,9 g N1 -[1 -Methyl-2-(3 -cyclohexen- 1 -ylamino)-äthyU- -N4-acetyl-sulfanilamid vom Smp. 139-1410 mit 10,6 g Bromcyan das 4'-[2-Imino-3-(3-cyclohexen-1-yl)-5-me- thyl-imidazolidin- 1-ylsulfonyl]-acetanilid vom Smp. 270 2710, von dem 37,4 g analog Beispiel 1 c) mit 125 g 50%iger Schwefelsäure zum Endprodukt 1 .Sulfanilyl-2-imino.
-3-(3 -cyclohexen- 1 -yl)-5-methyl-imidazolidin vom Smp.
208-2090 hydrolysiert werden.
b) Die Ausgangssubstanz von a) erhält man analog Beispiel 1 c) aus 25,4 g 4'-(2-Methyl-aziridin-1-ylsulfonyl) -acetanilid (vgl. Beispiel 1 c) und 100 ml (3-Cyclohexen -l -yl).amin.
c) Ferner wird analog Beispiel 1 a) das 1-Sulfanilyl-2- -imino-3-tert.butyl-4- ethyl-imidazolidin vom Smp. 1781800 (aus Isopropanol) und das 1 .Sulfanilyl-2-imino-3- butyl-5-äthyl-imidazolidin vom Smp. 161-1630 hergestellt.
Beispiel 10 a) Analog Beispiel 1 a) erhält man ausgehend von 32,7 g N1-(1 -Methyl-2.tert.butylamino.äthyl).N4.acetyl- -sulfanilamid vom Smp. 93-950 (aus Essigsäureäthylester) mit 6,5 g Chlorcyan oder mit 11,9 g Phenylcyanat anstelle von Bromcyan das 4'-(2-Imino-3-tert.butyl-5-methyl-imid- azolidin- 1 -ylsulfonyl)-acetanilid vom Smp. 243-2440 (aus N,N-Dimethylformamid), von dem 35,2 g nach Beispiel 1 b) zum Endprodukt 1-Sulfanilyl.2.imino.3.tert.butyl.5- -methyl-imidazolidin vom Smp. 145-1460 (aus Essigsäure äthylester) hydrolysiert werden.
b) Die Ausgangssubstanz von a) erhält man nach Beispiel lc - d) aus 24,0 g 4'-(2.Methyl-aziridin. 1 -ylsulfonyl).

As has now been found, these compounds have valuable pharmacological properties, in particular l-sulfanilyl-2-imino-3-butyl-5-ethyl-, 1-sulfanilyl-2-imino-3-tert-butyl-4-methyl- and that When administered orally or parenterally, l-sulf anilyl - 2 - imino-3-tert-butyl-5-methyl-imidazolidine have hypoglycemic effects that characterize them as being suitable for the treatment of diabetes. The hypoglycemic effect is demonstrated in standard tests on warm-blooded animals, e.g. in rabbits and rats.

The subsequent conversion of the group X of the reaction product into the free amino group, which converts it into a compound of the general formula I, is carried out, depending on the nature of the group X, by hydrolysis, reduction or reductive cleavage.
X radicals which can be converted into the free amino group by hydrolysis are, for example, acylamino radicals, e.g. the acetamido group. Furthermore, such radicals are never other alkoxy carbonylamino radicals, e.g. the ethoxycarbonylamino group, aryloxycarbonylamino groups such as the phenoxycarbonylamino group, or aryl methoxycarbonylamino group such as the benzyloxycarbonylamino group, or groups of corresponding thiocarbonic acid derivatives.
Further examples are substituted methylene amino groups such as e.g. the benzylideneamino or the p-dimethylamino benzylideneamino group. The hydrolysis to liberate the amino group can be carried out in an acidic medium, e.g. in methanolic hydrochloric acid, or in dilute aqueous hydrochloric acid or sulfuric acid or, if X is represented by an alkoxycarbonylamino radical, also under mild alkaline conditions, e.g. using l-n or
2-N sodium hydroxide solution.
An example of a radical X which can be converted into the amino group by reduction is the nitro group and examples of such radicals which lead to the amino group by reductive cleavage are the phenylazo or p-dimethylamino-phenylazo groups. The reduction of these residues can generally be catalytic, e.g. by means of hydrogen in the presence of Raney nickel, palladium or platinum carbon, in an inert solvent such as e.g. Ethanol. In addition to these, other customary reduction processes can also be used, for example the reduction of nitro groups or the reductive cleavage of azo groups by means of iron in acetic acid or hydrochloric acid.
Suitable starting materials of the general formula II are those compounds whose symbols Rl, R., R5 and X correspond to the groups which are listed after formula I or II. Such starting materials are, for example, N- (2-amino-ethyl) -benzosulfonamides, their benzene ring through the radical X in the para position, their amino group through the radicals R1 and Rs and their ethyl group in the 1- or 2-position through the methyl or Ethyl group is substituted.
A group of such compounds can e.g. be prepared if one starts from a l-pilenylsulfonyl-2-methyl- or l-phenylsulfonyl-2-ethyl-aziridine which is substituted by the radical X, and the aziridine with a primary or secondary amine which contains the radicals R1 and R , contains, implements. Representatives of the aziridines mentioned are described in the literature [cf. A. Weissenberger, Heterocyclic Compounds with Three- and Four-Membered Rings, Part. One. John Wiley & Sons Inc. London (1964). Further aziridines of this type can be prepared analogously.
The compounds of general formula I obtained by the process according to the invention are then, if desired, converted into their salts with inorganic and organic acids. These salts are produced e.g. by reacting the compounds of general formula 1 with the equivalent amount of an acid in a suitable aqueous, organic or organic solvent, such as e.g.
Methanol, ethanol, diethyl ether, chloroform or methylene chloride.
For use as medicinal substances, their pharmaceutically acceptable salts with acids can be used instead of the free compounds of the general formula I. Suitable addition salts are e.g. Salts with hydrochloric acid, hydrobromic acid, sulfuric acid, phosphoric acid, methanesulfonic acid, ethanesulfonic acid, -hydroxyethanesulfonic acid, acetic acid, lactic acid, oxalic acid, succinic acid, fumaric acid, maleic acid, malic acid, tartaric acid, citric acid, benzoic acid, salicylic acid, phenyl acetic acid.
The following examples explain the preparation of the new compounds of general formula I and of hitherto not described intermediates in more detail, but by no means represent the only embodiments of the same. The temperatures are given in degrees Celsius.
Example I a) 32.7 g of N1- (1-methyl-2-tert-butylamino-ethyl) -N4 -acetylsulfanilamide are dissolved in 100 ml of 2N sodium hydroxide solution and, while cooling at 20-300, with 10.6 g Cyanogen bromide added. The crude product immediately precipitates in crystalline form.
It is suction filtered after 30 minutes and recrystallized from N, N-dimethylformamide. The 4 '- (2-imino - 3 - tert-butyl-5-methyl-imidazolidin-1-ysulfonyl) -acetanilide melts at 243 - 2440, b) To 125 g of 50% sulfuric acid heated to 400 are added Stirring 35.2 g of the acetanilide obtained according to a) are added. The mixture is heated to 500 for 2 hours and the solution is then poured into 750 ml of water with stirring. The crude product precipitates as sulfate. The pH of the reaction mixture is with
10N sodium hydroxide solution adjusted to 6.3. The mixture dissolves except for a small amount of contamination; it is stirred with 2 g of activated charcoal for 15 minutes and filtered through Hyflo (purified diatomaceous earth).
The filtrate is made alkaline with 10 N sodium hydroxide solution. The crude product precipitates.
It is filtered off, washed with water, dried at 900 and recrystallized from ethyl acetate. The 1-sulfanilyl-2-imino-3-tert-butyl-5-methyl-imidazolidine obtained melts at 145-1460.
c) The starting compound mentioned under a) is obtained as follows: A solution of 23.3 g of N-acetyl-sulfanilyl chloride in 70 ml of acetone is converted into 5.6 g of 2-methyl-aziridine in 30 ml of 4 while stirring at -100 to 0 -n caustic soda dripped. The reaction mixture is stirred for a further 2 hours and then poured into 400 ml of ice water. The suspension obtained is filtered off, the precipitate, the crude 4- (2-methyl-aziridin-1 -ylsulfonyl) -acetanilide, washed with water and immediately processed further, since it polymerizes easily.
d) The acetanilide obtained according to c) is dissolved in the cold in 100 ml of dioxane and 15 mol of water and 100 ml of tert is added dropwise to the solution. Butylamine. Then the reaction mixture is refluxed for one hour and the excess tert. Butylamine and the dioxane distilled off. The crystalline residue is recrystallized from ethyl acetate, after which the pure N '- (1-methyl--2-tert-butylamino-ethyl) -N4-acetylsulfanilamide melts at 93 to 950.
Example 2 a) Analogously to Example 1 a), starting from 28.5 g of N1- (1-methyl-2-methylamino-ethyl) -N4-acetyl-sulfanilamide (crude product) with 10.6 g of cyanogen bromide the 4'- (2-Imino -3,5-dimethyl-imidazolidin-1-ylsulfonyl) -acetanilide of melting point 258-2600, of which 31.0 g as in Example 1 b) with 125 g of 50% sulfuric acid to give the end product 1-sulfanilyl-2 -imino-3,5-dimethyl-imidazolidine of m.p.
196 - 1970 are hydrolyzed.
b) The starting substance from a) is obtained analogously to Example 1 c) from 25.4 g of 4 '- (2-methyl-aziridin-1 -ylsulfonyl) - -acetanilide (cf. Example 1 c) and 200 ml of 33% methylamine in ethanol.
Example 3 a) Analogously to Example 1 a), starting from 29.9 g of N1 - (1-methyl-2-ethylamino-ethyl) -N4-acetyl-sulfanilamide (crude product) with 10.6 g of cyanogen bromide the 4 '- (2 -Imino - 3 - ethyl-5-methyl-imidazolidin-1-ylsulfonyl) - acetanilide of melting point 229-2500, of which 324 g as in Example 1 b) with 125 g of 50% sulfuric acid to the end product 1 - sulfanilyl-2- imino-3-ethyl-5-methyl-imidazolidine are hydrolyzed.
b) The starting substance of a) is obtained analogously to Example 1 c) from 25.4 g of 4- (2-methyl-aziridin-1-ylsulfonyl) - -acetanilide (see. Example 1 c) and 250 ml of 20% ethylamine in Benzene.
Example 4 a) Analogously to Example 1 a), 31.3 g of NÚ- (1-methyl-2-propylamino-ethyl) -NÚ-acetyl-sulphanilamide of melting point 162-1630 with 10.6 g of cyanogen bromide are obtained the 4 '- (2-imino-3-propyl-5-methyl-imidazolidin-1-ysulfonyl) -acetanilide of melting point 254-2550, of which 33.8 g as in Example 1 b) with 125 g of 50% sulfuric acid hydrolyzed to the end product l-sulfanilyl-2-imino-3-propyl-5-methyl-imidazolidine with a melting point of 146-1480.
b) The starting substance of a) is obtained analogously to Example 1 c) from 25.4 g of 4 '- (2-methyl-aziridin-1-ylsulfonyl) acetanilide (cf. Example 1 c) and 100 ml of propylamine.
Example 5 a) Analogously to Example 1 a), starting from 32.7 g of N1 - (1-methyl-2-butylamino-ethyl) -N4-acetylsulfanilamide with a melting point of 166-1670 with 10.6 g of cyanogen bromide, the 4th '- (2-Imino-3-butyl-5-methyl-imidazolidin-1 -ylsulfonyl) - acetanilide of melting point 243-244, of which 35.2 g as in Example 1 b) with 125 g of 50% sulfuric acid to the end product 1-sulfanilyl-2-imino-3-butyl-5-methyl-imidazolidine with a melting point of 125.1260 can be hydrolyzed.
b) The starting substance of a) is obtained analogously to Example 1 c) from 25.4 g of 4 '- (2-methyl-aziridin-1-ylsulfonyl) - -acetanilide (cf. Example 1 c) and 100 ml of butylamine.
Example 6 a) Analogously to Example 1 a), starting from 32.7 g of N '- (1-methyl-2-sec.butylamino-ethyl) -N4-acetyl-sullanilamide of melting point 118-1200 with 10.6 g of cyanogen bromide 4 '- (2 - Ii-nino - 3-sec-butyl-5-methyl-imidazolidin-1-yl sulfonyl) -acetanilide of melting point 242.2440, of which 35.2 g as in Example 1 b) with 125 g of 50% sulfuric acid to the end product 1-sulfanilyl-2-imino-3-sec-butyl-5-methyl-imidazolidine with a melting point of 194-1950 are hydrolyzed.
b) The starting sulfate of a) is obtained analogously to Example 1 c) from 25.4 g of 4 '- (2-methyl-aziridin-1-ylsulfonyl) -acetanilide (cf. Example 1 c) and 100 ml of sec. Butylamine.
Example 7 a) Analogously to Example 1 a), starting from 34.1 g of N '- (1-methyl-2-pentylaminoWäthyl) -N4-acetyl-sulfanilamide (crude product) with 10.6 g of cyanogen bromide the 4'- (2-Imino-3-pentyl-5 -methyl-imidazolidin-1 -ylsulfonyl) -acetanilide of melting point 228.2290, of which 36.4 g as in Example 1 b) with 125 g of 50% sulfuric acid to the end product 1 -sulfanilyl- 2-im ino3 -pentyl-5-methvl-im idazolidine with a melting point of 127.1280 can be hydrolyzed.
b The starting substance of a) is obtained analogously to Example 1 c) from 25.4 g of 4 '. (2-methyl-aziridin.1-ylsulfonyl) - acetanilide (cf. Example 1 c) and 100 ml of pentylamine.
Example 8 a) Analogously to Example 1 a), starting from 35.1 g of Nl-methyl-2-cyclohexylamino-ethyl) -N4-acetyl-sulfanilamide (crude product) with 10.6 g of cyanogen bromide the 4 'is obtained. Imino-3-cyclohexyl-5-methyl-imidazolidin-1 -ylsulfonyl) acetanilide of melting point 235.2370, of which 37.6 g analogously to Example 1 b) with 125 g of 50% sulfuric acid to give the end product 1-sulfanilyl-2-iinino -3 -cydohexyl-5.methyl-imid-azolidine with a melting point of 217.2180 can be hydrolyzed.
b) The starting substance from a) is obtained analogously to Example 1 c) from 25.4 g of 4 '- (2-methyl-aziridin-1 -ylsulfonyl).

208-2090 are hydrolyzed.
b) The starting substance of a) is obtained analogously to Example 1 c) from 25.4 g of 4 '- (2-methyl-aziridin-1-ylsulfonyl) acetanilide (see Example 1 c) and 100 ml (3-cyclohexene - l -yl) amine.
c) Furthermore, analogously to Example 1 a), the 1-sulfanilyl-2- imino-3-tert-butyl-4-ethyl-imidazolidine with a melting point of 1781800 (from isopropanol) and the 1-sulfanilyl-2-imino-3- butyl-5-ethyl-imidazolidine of m.p. 161-1630.
Example 10 a) Analogously to Example 1 a), starting from 32.7 g of N1- (1-methyl-2.tert.butylamino.ethyl) .N4.acetyl-sulfanilamide of melting point 93-950 (from ethyl acetate) are obtained 6.5 g of cyanogen chloride or, with 11.9 g of phenyl cyanate instead of cyanogen bromide, 4 '- (2-imino-3-tert-butyl-5-methyl-imidazolidine-1-ysulfonyl) acetanilide with a melting point of 243-2440 (from N, N-dimethylformamide), of which 35.2 g according to Example 1 b) to the end product 1-sulfanilyl.2.imino.3.tert.butyl.5- -methyl-imidazolidine with a melting point of 145-1460 (from Ethyl acetate) are hydrolyzed.
b) The starting substance of a) is obtained according to Example 1c - d) from 24.0 g of 4 '- (2.Methyl-aziridine. 1 -ylsulfonyl).
