Verfahren zur Herstellung von neuen Derivaten des Sulfanilamids
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Derivate des Sulfanilamids.
Verbindungen der allgemeinen Formel I

in welcher 31 eine Alkylgruppe mit höchstens 5 Kohlenstoffatomen, oder eine Cycloalkyl- oder Cycloalkenylgruppe von höchstens 7 Kohlenstoffatomen bedeutet, und ihre Additionssalze mit anorganischen oder organi schen Säuren sind bisher nicht bekannt geworden.
Wie nun gefunden wurde, besitzen diese Verbindun gen wertvolle pharmakologische Eigenschaften, insbeson dere das 1 -Sulfanilyl-2-imino-3-butyl-, das 1-Sulfanilyl- -2-imino.3-tert.butyl- und das 1-Sulfanilyl-2-imino-3-cy- clohexyl-imidazolidin weisen bei peroraler oder parente raler Verabreichung hypoglykämische Wirkung auf, die sie als geeignet zur Behandlung der Zuckerkrankheit cha rakterisieren. Die hypoglykämische Wirkung wird an
Standardversuchen an Warmblütern, z.B. Kaninchen und
Ratten, nachgewiesen.
In den Verbindungen der allgemeinen Formel I kann Rl als niedere Alkylgruppen z.B. die Methyl-, Äthyl-,
Propyl-, Isopropyl-, Butyl-, sek.-Butyl-. tert.Butyl-, Iso butyl-, Pentyl-, Isopentyl-, 2,2-Dirnethyl-propyl-, 1-Me- thyl-butyl-, 1 -Äthyl-propyl-, 1,2-Dimethyl-propylgruppe sein.
Ferner kann R1 beispielsweise sein: als Cycloalkyl gruppe die Cyclopropyl-, Cyclopropylmethyl-, 2-Cyclo propyl-äthyl-, 3-Cyclopropyl-propyl-, Cyclobutyl-, Cyclo butylmethyl-. 2-Cyclobutyl-äthyl-, 3-Cyclobutylpropyl-,

in welcher R1 die unter Formel I angegebene Bedeutung hat, oder mit einem Alkalimetall- oder Erdalkalimetallderivat einer solchen Verbindung kondensiert und cycli siert. nötigenfalls das Reaktionsprodukt zur Umwandlung der Gruppe X in die freie Aminogruppe hydrolysiert. reduziert oder reduktiv spaltet und gegebenenfalls die erhaltene Verbindung mit einer anorganischen oder organischen Säure in ein Additionssalz überführt.
Geeignete Alkalimetall- oder Erdalkalimetallderivate der allgemeinen Formel III sind Natrium-, Kalium-, Lithium- bzw. Calciumderivate. Die Kondensation wird vorzugsweise in einer ätherartigen Flüssigkeit, z.B. in Äther, Tetrahydrofuran, Dioxan, Anisol oder Äthylen glykoldimethyläther, vorgenommen.
Die anschliessende Umwandlung der Gruppe X des Reaktionsproduktes in die freie Aminogruppe, welche dieses in eine Verbindung der allgemeinen Formel I überführt, wird je nach der Art der Gruppe X durch eine Hydrolyse, Reduktion oder reduktive Spaltung vorgenommen.
Durch Hydrolyse in die freie Aminogruppe überführbare Reste X sind beispielsweise Acylaminoreste, wie z.B. die Acetamidogruppe, Ferner sind solche Reste niedere Alkoxycarbonyl-aminoreste. wie z.B. die Äthoxy c.irbonylaminogruppe, Aryloxycarhonylaminoreste, wie der Phenoxycarbonvlaminorest. oder Arylmethoxycarbonylaminoreste, wie der Benzyloxycarbonylaminorest, oder Reste v. entsprechenden Thiokohlensäurederivaten. Weitere Beispiele sind substituierte Methylenaminoreste, wie z.B. die Benzylidenamino- oder die p-Dimethylamino -benzylidenaminogruppe.
Die Hydrolyse zur Freisetzung der Aminogruppe kann in saurem Medium, z.B. in me Ihanolischer Salzsäure, oder in verdünnter wässriger Salzsäure oder Schwefelsäuren erfolgen oder, falls X durch einen Alkoxycarbonylaminorest verkörpert ist, auch un ter milden alkalischen Bedingungen, z.B. mittels l-n. oder 2-n. Natronlauge, vorgenommen werden.
Ein Beispiel für einen durch Reduktion in die Ami nogruppe überführbaren Rest X ist die Nitrogruppe und Beispiele für solche Reste. die durch reduktive Spaltung zur Aminogruppe führen, sind die Phenylazo- oder p-Di methylamino-phenylazogruppen. Die Reduktion dieser
Reste kann allgemein katalytisch, z.B. mittels Wasserstoff in Gegenwart von Raney-Nickel, Palladium- oder Platin-Kohle, in einem inerten Lösungsmittel, wie z.B.
Äthanol, erfolgen. Neben diesen kommen auch andere übliche Reduktionsverfahren in Betracht, beispielsweise die Reduktion von Nitrogruppen oder die reduktive Spaltung von Azogruppen mittels Eisen in Essigsäure oder Salzsäure.
Als Ausgangsstoffe der allgemeinen Formel II eignen sich solche Verbindungen. deren Symbol X mit den Gruppen übereinstimmen, die anschliessend an Formel II allgemein erwähnt und vorstehend näher definiert sind.
Einzelne Vertreter solcher Verbindungen sind in der Literatur beschrieben, z.B. das 4'-(Aziridin-l-ylsulfonyl)- -acetanilid [vgl. R. Lehmann et al., Bull. Soc. Chim. Belges 55. 52-97 (1946: C.A. 41, 3475 f (1947)]. Weitere Aziridine von diesem Typus können analog hergestellt werden.
Die nach dem erfindungsgemässen Verfahren erhalte nen Verbindungen der allgemeinen Formel I werden an seliliessend gewünschtenfalls in ihre Salze mit anor,eani- aschen sowie organischen Säuren übergeführt. Die Her stellung dieser Salze erfolgt z.B. durch Umsetzung der
Verbindungen der allgemeinen Formel I mit der äquiva lenten Menge einer Säure in einem geeigneten wässrig or.rí nischen oder organischen Lösungsmittel, wie z.B.
N-Tethanol, Äthanol. Diäthyläther, Chloroform oder Me thylenchlond.
Zur Verwendung als Arzneistoffe können anstelle der freien Verbindungen der allgemeinen Formel I deren pharmazeutisch annehmbare Salze mit Säuren eingesetzt werden. Geeignete Additionssalze sind z.B. Salze mit Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsaure, Phosphorsäure, Methansulfonsäure, Äthansulfonsäure, ,B-Hydroxyäthansulfonsäure, Essigsäure, Milchsaure, Oxalsäure, Bernsteinsäure, Fumarsäure, Maleinsäure, Äpfelsäure. Weinsäure, Citronensäure, Benzoesäure, Salicylsäure, Phenylessigsäure. Mandelsäure und Embonsäure.
Die nachfolgenden Beispiele erläutern die Herstellung der neuen Verbindungen der allgemeinen Formel I und von bisher nicht beschriebenen Zwischenprodukten näher, stellen jedoch keineswegs die einzige Ausführungsform desselben dar. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel I a) Eine Lösung von 14,6 g Butylamin in 100 inl abs.
Äther wird unter Rühren bei --100 bis 5 innerhalb 30 Minuten mit einer Lösung von 10,6 g Bromcyan in 50 ml abs. Äther versetzt. Man rührt das Reaktionsgemisch noch 30 Minuten weiter und filtriert das ausve- fallene Butylamin-hydrochlorid ab. Man versetzt das Filtrat, in welchem das erhaltene Butylcyanamid gelöst ist, unter weiterer Kühlung mit einer Suspension von 2,8 g Natriumhydrid in 40 ml abs. Äther. Man rührt die Suspension, welche weiter auf dieselbe Temperatur gekühlt wird, noch 30 Minuten und fügt dann zum entstandenen, suspendierten Natriumderivat des Butylcyanamids 24,0 g 4'-(Aziridin-l-ylsulfonyl).acetanilid zu. Man erwärmt das Gemisch auf Raumtemperatur und rührt 15 Stunden bei dieser Temperatur. Anschliessend versetzt man das Reaktionsgemisch langsam mit 2-n.
Salzsäure und trennt die beiden entstandenen Phasen. Die saure, wässrige Phase wird zweimal mit Äther gewaschen, mit Aktivkohle gereinigt, filtriert und bei 0 mit konz. Natronlauge alkalisch gestellt. Man filtriert das ausgefallene 4'-(2-Imino -3-butyl-imidazolidin- 1 -ylsulfonyl)-acetanilid ab und kristallisiert es aus Methanol um. wonach es bei 243-244 schmilzt.
b) 33,8 g des nach a) erhaltenen Acetanilids werden mit 100 ml 2-n. Salzsäure eine Stunde auf 800 erwärmt.
Anschliessend kühlt man das Reaktionsgemisch auf 200 ab und stellt es mit 2-n. Natronlauge alkalisch. Die ausgefallene. rohe Base wird abfiltriert, mit Wasser gewaschen und aus Äthanol umkristallisiert. Das erhaltene 1 -Sulfani- lyl-2-imino-3-butyl-imidazolidin schmilzt bei 179-1810.
Beispiel 2
Analog Beispiel 1 erhält man: a) aus 6,2 g Methylamin in 100 ml Äther mit 10,6 g Bromcyan das N-Methyl-cyanamid, welches mit 2,8 g Natriumhydrid in 40 ml Äther in das Natriumderivat übergeführt wird; dieses liefert mit 24.0 g 4'-(Aziridin-l -ylsulfonyl)-acetanilid das 4'-(2-Imino-3-methyl-imidazoli din-l-ylsulfonyl)-acetanilid vom Smp. 266-2670C, welches nach Beispiel 1 b) zum l-Sulfanilyl-2-imino-3-methyl -imidazolidin vom Smp. -09-7110 hydrolysiert wird.
b) aus 9,0 g Äthylamin in 100 ml Äther mit 10,6 g Bromcyan das N-Äthyl-cyanamid, welches mit 2,8 g Natriumhydrid in 40 ml Äther in das Natriumderivat übergeführt wird; dieses liefert mit 24,0 g 4'-(Aziridin-l-yl- sulfonyl)-acetanilid das 4'-(2-Imino-3-Åathyl-imidazolidin- -l-ylsulfonyl)-acetanilid vom Smp. 267-2690, welches ana log Beispiel lb) zum 1 -Sulfanilyl-2-imino-3 -äthyl-imid- azolidin vom Smp. 171-1720, hydrolysiert wird; c) aus 11,8 g Propylamin in 100 ml Äther mit 10,6 g Bromcyan das Propyl-cyanamid, welches mit 2,8 g Natriumhydrid in 40 ml Äther in das Natriumderivat übergeführt wird;
dieses liefert mit 24,0 g 4'-(Aziridin-1-ylsulfonyl)-acetanilid das 4'-(2-Imino-3-propyl-imidazolidin-1-ylsulfonyl)-acetanilid vom Smp. 253-2550, welches analog Beispiel lb) zum 1-Sulfanilyl-2-imino-3-propyl -imidazolidin vom Smp. 164-1660 hydrolysiert wird; d) aus 14,6 g tert.
Butylamin in 100 ml Äther mit 10,6 g Bromcyan das tert.Butyl-cyanamid, welches mit 2.8 g Natriumhydrid in 40 ml Äther in das Natriumderivat übergeführt wird; dieses liefert mit 24,0 g 4'-(Aziri din-l-ylsulfonyl)-acetanilid das 4'-(2-Imino-3 -tert.butyl- -imidazolidin- 1 -ylsulfonyl)-acetanilid vom Smp. 245-2470, welches nach Beispiel lb) zum l-Sulfanilyl-2-imino-3- -tert.butyl-imidazolidin vom Smp. 187-1890 hydrolysiert wird; e) aus 17,0 g Cyclopentylamin in 100 ml Äther mit 10,6 g Bromcyan das Cyclopentyl-cyanamid, welches mit 2,8 g Natriumhydrid in 40 ml Äther in das Natriumderivat übergeführt wird;
dieses liefert mit 24,0 g 4' -(Aziridin- 1 -ylsulfonyl)-acetanilid das 4'-(2-Imino-3-cy clopentyl-imidazolidin- 1 -ylsulfonyl)-acetanilid vom Smp.
261-2630C, welches nach Beispiel lb) zum l-Sulfonilyl -2-imino-3.cyclopentyl.imidazolidin vom Smp. 192-1930 hydrolysiert wird; f) aus 19,8 g Cyclohexylamin in 100 ml Äther mit 10,6 g Bromcyan das Cyclohexyl-cyanamid, welches mit 2,8 g Natriumhydrid in 40 ml Äther in das Natriumderivat übergeführt wird;
dieses liefert mit 24,0 g 4'-(Aziri din-l-ylsulfonyl)-acetanilid das 4'-(2-Imino-3-cyclohexyl -imidazolidin- 1 -ylsulfonyl)-acetanilid vom Smp. 283-2840, welches analog Beispiel lb) zum l-Sulfanilyl-2-imino-3- -cyclohexyl-imidazolidin vom Smp. 178-179 hydrolysiert wird. und g) aus 17,4 g Pentylamin in 100 ml Äther mit 10,6 g Bromcyan das Pentvlcyanamid, welches mit 2,8 g Natriumhydrid in 40 ml Äther in das Natriumderivat übergeführt wird;
dieses liefert mit 24,0 g 4'-(Aziridin-1-ylsulfonyl)-acetanilid das 4'-(2-lmino-3-pentyl-imidazolidin- - 1 -ylsulfonyl)-acetanilid vom Smp. 248-2500 welches analog Beispiel lb) zum 1 -Sulfanilyl-2-imino-3-pentyl-imid azolidin vom Smp. 167-1680 hydrolysiert wird.
Beispiel 3
Unter Rühren und Kühlen auf 5 bis - 100 gibt man zu einer Lösung von 10,6 g Bromcyan in 50 ml abs.
Äther tropfenweise eine Lösung von 14,6 g Butylamin in 100 ml abs. Äther zu. Nach einigen Minuten fällt das Butylamin-hydrochlorid aus. Es wird abfiltriert. Das Filtrat, in welchem das Butyl-cyaniamid gelöst ist, wird portionenweise mit 2.8 g Natriumhydrid in-40 ml abs. Äther versetzt. Ein Niederschlag fällt aus. Man verdünnt die Suspension mit 200 ml abs. Äther und 200 ml Dioxan und rührt sie 10 Minuten bei Raumtemneratur. Anschlies send fügt man 19,8 g l-Sufanilyl-aziridin zu, rührt die graue Suspension 15 Stunden weiter und gibt noch 30 ml 2-n. Salzsäure zu. Man trennt die organische Phase ab.
Sie wird zweimal mit 2-n. Salzsäure extrahiert. Die wässrigen, sauren Phasen werden vereinigt, mit Aktivkohle gereinigt und filtriert. Man neutralisiert das Reaktionsgemisch mit festem Natriumhydrogencarbonat, wobei ein braunes Harz ausfällt. Das Harz wird über Cellit (gereinigte Diatomeenerde) abfiltriert und die farblose, klare Lösung mit konz. Natronlauge alkalisch gestellt. Man lässt das Reaktionsgemisch eine Stunde bei 0 stehen, filtriert die ausgefallenen Kristalle, die aus Äthanol umkristallisiert werden. Man erhält das 1 -Sulfanilyl-2-imi- no-3-butyl-imidazolidin vom Smp. 179-1810.
Beispiel 4
Analog Beispiel 3 erhält man: a) aus 6,2 g Methylamin in 100 ml Äther mit 10,6 g Bromcyan das Methyl-cyanamid, welches mit 2,8 g Natriumhydrid in 40 ml Äther in das Natriumderivat übergeführt wird; dieses liefert mit 19,8 g l-Sulfanilyl-aziridin das l-Sulfanilyl-2-imino-3-methyl-imidazolidin vom Smp.
209-2110: b) aus 9,0 g Äthylamin in 100 ml Äther mit 10,6 g Bromcyan das Äthy]-cyanamid, welches mit 2,8 g Natriumhydrid in 40 ml Äther in das Natriumderivat übergeführt wird; dieses liefert mit 19,8 g 1 -Sulfanilyl-aziridin das 1-Sulfanilyl-2.imino.3-äthyl-imidazolidin vom Smp.
171.1720: c) aus 11,8 g Propylamin in 100 ml Äther mit 10,6 g Bromcyan das Propyl-cyanamid, welches mit 2,8 g Natriumhydrid in 40 ml Äther in das Natriumderivat übergeführt wird; dieses liefert mit 19,8 g 1-Sulfanilyl-aziridin das 1 .Sulfanilyl-2.imino.3.propyl-imidazolidin vom Smp.
164-1660; d) aus 14,6 g tert.Butylamin in 100 ml Äther mit 10,6 g Bromcyan das tert.Butyl-cyanamid, welches mit 2,8 g Natriumhydrid in 40 ml Äther in das Natriumderivat übergeführt wird; dieses liefert mit 19,8 g l-Sulfanilyl-aziridin das 1 -Sulfanilyl-2-imino-3 -tert.butyl-imid- azolidin vom Smp. 187-1890; e) aus 17,0 g Cyclopentylamin in 100 ml Äther mit 10,6 g Bromcyan das Cyclopentyl-cyanamid, welches mit 2,8 g Natriumhydrid in 40 ml Äther in das Natriumderivat übergeführt wird; dieses liefert mit 19,8 g 1-Sulfanilyl- -aziridin das I-Sulfanilyl l-Sulfanilyl-2-imino-3-cyclopentyl-imidazoli- din vom Smp. 192-1930, und f) ausl9,8 g Cyclohexylamin in 100 ml Äther mit 10,6 g Bromcyan das Cyclohexyl-cyanamid, welches mit 2,8 g Natriumhydrid in 40 ml Äther in das Natriumderivat übergeführt wird;
dieses liefert mit 19,8 g 1-Sulfanilyl -aziridin das 1-Sulfanilyl-2-imino-3-cyclohexyl-imidazoli- din vom Smp. 178-1790.

Suitable alkali metal or alkaline earth metal derivatives of the general formula III are sodium, potassium, lithium and calcium derivatives. The condensation is preferably carried out in an ethereal liquid, e.g. in ether, tetrahydrofuran, dioxane, anisole or ethylene glycol dimethyl ether.
The subsequent conversion of the group X of the reaction product into the free amino group, which converts it into a compound of the general formula I, is carried out, depending on the nature of the group X, by hydrolysis, reduction or reductive cleavage.
X radicals which can be converted into the free amino group by hydrolysis are, for example, acylamino radicals, e.g. the acetamido group, such radicals are lower alkoxycarbonyl-amino radicals. such as. the ethoxy c.irbonylamino group, aryloxycarhonylamino radicals, such as the phenoxycarbonylamino radical. or Arylmethoxycarbonylaminoreste, such as the Benzyloxycarbonylaminorest, or radicals v. corresponding thiocarbonic acid derivatives. Further examples are substituted methylene amino radicals, e.g. the benzylideneamino or the p-dimethylamino benzylideneamino group.
The hydrolysis to liberate the amino group can be carried out in an acidic medium, e.g. in methanolic hydrochloric acid, or in dilute aqueous hydrochloric acid or sulfuric acids or, if X is represented by an alkoxycarbonylamino radical, also under mild alkaline conditions, e.g. using l-n. or 2-n. Caustic soda, can be made.
An example of a radical X which can be converted into the amino group by reduction is the nitro group and examples of such radicals. which lead to the amino group through reductive cleavage are the phenylazo or p-dimethylamino-phenylazo groups. The reduction of this
Residues can generally be catalytic, e.g. by means of hydrogen in the presence of Raney nickel, palladium or platinum carbon, in an inert solvent such as e.g.
Ethanol. In addition to these, other customary reduction processes can also be used, for example the reduction of nitro groups or the reductive cleavage of azo groups using iron in acetic acid or hydrochloric acid.
Such compounds are suitable as starting materials of the general formula II. whose symbol X corresponds to the groups which are mentioned in general following formula II and are defined in more detail above.
Individual representatives of such compounds are described in the literature, e.g. the 4 '- (aziridin-1-ylsulfonyl) - acetanilide [cf. R. Lehmann et al., Bull. Soc. Chim. Belges 55, 52-97 (1946: C.A. 41, 3475 f (1947)]. Further aziridines of this type can be prepared analogously.
The compounds of the general formula I obtained by the process according to the invention are optionally converted into their salts with inorganic, eanic and organic acids. These salts are produced e.g. by implementing the
Compounds of the general formula I with the equivalent amount of an acid in a suitable aqueous, organic or organic solvent, such as e.g.
N-tethanol, ethanol. Diethyl ether, chloroform or methylene chloride.
For use as medicinal substances, their pharmaceutically acceptable salts with acids can be used instead of the free compounds of the general formula I. Suitable addition salts are e.g. Salts with hydrochloric acid, hydrobromic acid, sulfuric acid, phosphoric acid, methanesulfonic acid, ethanesulfonic acid,, B-hydroxyethanesulfonic acid, acetic acid, lactic acid, oxalic acid, succinic acid, fumaric acid, maleic acid, malic acid. Tartaric acid, citric acid, benzoic acid, salicylic acid, phenylacetic acid. Mandelic acid and emboxylic acid.
The following examples explain the preparation of the new compounds of general formula I and of hitherto not described intermediates in more detail, but by no means represent the only embodiment of the same. The temperatures are given in degrees Celsius.
Example I a) A solution of 14.6 g of butylamine in 100 inl abs.
Ether is mixed with a solution of 10.6 g of cyanogen bromide in 50 ml of abs within 30 minutes while stirring at -100 to 5. Ether shifted. The reaction mixture is stirred for a further 30 minutes and the butylamine hydrochloride which has precipitated out is filtered off. The filtrate, in which the butyl cyanamide obtained is dissolved, is added, with further cooling, to a suspension of 2.8 g of sodium hydride in 40 ml of abs. Ether. The suspension, which is cooled further to the same temperature, is stirred for a further 30 minutes and then 24.0 g of 4 '- (aziridin-1-ylsulfonyl) acetanilide are added to the suspended sodium derivative of butyl cyanamide. The mixture is warmed to room temperature and stirred for 15 hours at this temperature. The reaction mixture is then slowly mixed with 2-n.
Hydrochloric acid and separates the two phases formed. The acidic, aqueous phase is washed twice with ether, cleaned with activated charcoal, filtered and at 0 with conc. Sodium hydroxide solution made alkaline. The precipitated 4 '- (2-imino -3-butyl-imidazolidin-1 -ylsulfonyl) acetanilide is filtered off and it is recrystallized from methanol. after which it melts at 243-244.
b) 33.8 g of the acetanilide obtained according to a) are mixed with 100 ml of 2-n. Hydrochloric acid heated to 800 for one hour.
The reaction mixture is then cooled to 200 and set to 2-n. Caustic soda alkaline. The unusual one. The crude base is filtered off, washed with water and recrystallized from ethanol. The 1-sulfanilyl-2-imino-3-butyl-imidazolidine obtained melts at 179-1810.
Example 2
Analogously to Example 1, the following is obtained: a) from 6.2 g of methylamine in 100 ml of ether with 10.6 g of cyanogen bromide, N-methyl-cyanamide, which is converted into the sodium derivative with 2.8 g of sodium hydride in 40 ml of ether; With 24.0 g of 4 '- (aziridine-l -ylsulfonyl) -acetanilide this gives the 4' - (2-imino-3-methyl-imidazolydin-l-ylsulfonyl) -acetanilide of melting point 266-2670C, which is obtained according to Example 1 b) is hydrolyzed to the l-sulfanilyl-2-imino-3-methylimidazolidine with a melting point of -09-7110.

Example 3
While stirring and cooling to 5 to -100 are added to a solution of 10.6 g of cyanogen bromide in 50 ml of abs.
