Verfahren zur Herstellung neuer Guanidin-Verbindungen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Verbindungen der Formel I, worin X für Wasserstoff, Halogen, die Hydroxymethyl- oder Methylgruppe steht, R1 eine niedere Alkylgruppe, eine Benzyl- oder Phenäthylgruppe, die gegebenenfalls durch einen oder zwei der folgenden Substituenten - Halogen, eine niedere Alkyl- oder Alkoxygruppe - im Phenylrest substituiert sein können, und R für eine niedere Alkylgruppe, die Benzyl- oder Phenäthylgruppe, die gegebenenfalls durch einen oder zwei der folgenden Substituenten - Halogen, eine niedere Alkyl- oder Alkoxygruppe - im Phenylrest substituiert sein können, eine Cycloalkylgruppe mit 3 - 6 Kohlenstoffatomen, die Allyl-, Hydroxy-, Amino- oder Dimethylaminogruppe steht, und ihrer Säureadditionssalze.
Erfindungsgemäss gelangt man zu den Verbindungen der Formel I und ihren Säureadditionssalzen, indem man Verbindungen der Formel II, worin X und R1 obige Bedeutung besitzen und R5 für einen niederen Alkylrest steht, mit Verbindungen der Formel III, worin R2 obige Bedeutung besitzt, umsetzt und gewünschtenfalls die so erhaltenen Verbindungen der Formel I in ihre Säureadditionssalze überführt.
Vorzugsweise arbeitet man in Gegenwart einer Säure, wobei jedoch stets mindestens eine der Reaktionskomponenten teilweise als Base vorliegen soll.
Beispielsweise geht man so vor, dass man Verbindungen der Formel II, vorzugsweise als Säureadditionssalz, z.B. als Hydrochlorid, Hydrobromid, Hydrojodid oder Sulfat, mit etwa 1-10 Äquivalenten einer Verbindung der Formel III umsetzt.
Die Umsetzung wird vorzugsweise bei erhöhter Temperatur, gegebenenfalls im Autoklaven durchgeführt, kann jedoch auch in einem unter den Reaktionsbedingungen inerten, polaren Lösungsmittel, wie z.B. einem niederen Alkanol wie Äthanol oder Isopropanol, einem Gemisch von Wasser mit Dimethylformamid usw., bei Normaldruck und Raumtemperatur oder erhöhter Temperatur erfolgen. Falls die Kondensation ohnelösungs- mittel durchgeführt wird, kann zur rascheren Entfernung des Alkylmercaptans ein Vakuum angelegt werden (ca.
12 Torr). Statt Anlegen eines Vakuums kann auch ein inertes Gas durch oder über das Reaktionsgemisch geleitet werden.
Die nach dem vorstehend beschriebenen Verfahren hergestellten Guanidinverbindungen der Formel l können als freie Basen oder in Form von Säureadditionssalzen auf übliche Weise isoliert und nach bekannten Methoden, z.B. durch Kristallisation, gereinigt werden.
Durch Behandlung mit Alkali, zum Beispiel mit einem alkalisch vorbehandelten Anionenaustauscher, lassen sich aus den Salzen die entsprechenden Basen freiset zen; zur Herstellung von Säureadditionssalzen können die Basen mit anorganischen oder organischen Säuren umgesetzt werden, z.B. mit Chlorwasserstoff, Salpetersäure, Schwefelsäure, Methansulfonsäure, Cyclohexylsulfaminsäure, Maleinsäure, Weinsäure usw. Es ist aber auch möglich, in den Salzen das Anion durch doppelte Umsetzung auszutauschen.
Die Verbindungen der Formel I und ihre Säureadditionssalze, im folgenden kurz als neue Substanzen bezeichnet, sind in der Literatur bisher nicht beschrieben worden. Sie zeichnen sich in der pharmakologischen Prüfung an isolierten Organen und am Ganztier durch vielfältige und stark ausgeprägte Effekte aus und sollen daher als Heilmittel verwendet werden.
Insbesondere können sie als Kreislaufmittel, Psychopharmaca, Salidiuretica und Antiphlogistica verwendet werden, wie z.B. aus der Blutdruck-steigernden, bradycarden, Amin-verstärkenden und Kreislaufreflexhemmenden Wirkung am narkotisierten Hund, aus der Motilitätshemmung, dem Reserpinantagonismus und einer sympathischen Stimulation an der Maus, aus der salidiuretischen Aktivität an der Ratte und aus der Hemmung des Carrageenoedems an der Ratte hervorgeht.
Als Heilmittel können die Verbindungen der Formel I bzw. ihre wasserlöslichen, physiologisch verträglichen Säureadditionssalze allein oder in geeigneten Arzneiformen, wie Tabletten, Dragees, Injektionslösungen, Suppositorien usw., enteral oder parenteral verabreicht werden.
Die neuen Ausgangsverbindungen der Formel II können hergestellt werden, indem man Verbindungen der Formel IV, worin X obige Bedeutung -besitzt, mit Verbindungen der Formel V, worin R1 obige Bedeutung hat, zu den Verbindungen der Formel VI, worin X und R1 obige Bedeutung besitzen, umsetzt und diese durch Reaktion mit Alkylhalogeniden bzw. Alkylsulfaten bei erhöhter Temperatur, vorzugsweise bei Siedetemperatur des Reaktionsgemisches, in die Verbindungen der Formel II überführt.
Zur Herstellung der als Ausgangsprodukt benötigten, bisher unbekannten Verbindung der Formel IV kann man Verbindungen der Formel VII, worin X obige Bedeutung besitzt, reduzieren, z.B. mittels Zink in Ameisensäure oder Essigsäure, vorzugsweise aber mittels eines komplexen Hydrids der Alkalimetalle wie Lithiumaluminiumhydrid, Natrium-dihydro-bis(2-methoxy äthoxy)aluminat usw. in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel, z.B. einem cyclischen oder offenkettigen Äther wie Diäthyläther, bei etwa 5 - 80.
Die Verbindungen der Formel VII sind ebenfalls neu und können auf an sich bekannte Weise aus den Verbindungen der Formel VIII, worin X obige Bedeutung besitzt, erhalten werden. Praktisch geht man z.B. so vor, dass man eine Lösung von Verbindungen der Formel VIII in überschüssiger verdünnter Salzsäure mit Natriumnitrit versetzt.
Die Verbindungen der Formel VIIIa, worin XI Halogen, die Hydroxymethyl- oder Methylgruppe bedeutet, sind auch neu und können folgendermassen hergestellt werden:
1) 6-Chlor-1,2,2a,3,4,5-hexahydrobenz[cd]indol erhält man beispielsweise, indem man l-Acetyl-1,2,2a,3,4,- 5-hexahydrobenz[cd]indol chloriert, z.B. durch Einleiten von Chlor in eine Lösung von 1-Acetyl-1,2,2a,3,4,5-he- xahydrobenz[odjindol in einem unter den vorliegenden Bedingungen inerten organischen Lösungsmittel, z.B.
einem chlorierten Alkankohlenwasserstoff wie Tetrachlorkohlenstoff, bei Raumtemperatur und das spontan kristallisierende 1 -Acetyl-6-chlor- 1 ,2,2a,3,4,5-hexahydro benz[cd]indol deacetyliert, z.B. durch Erhitzen in einem Gemisch von Eisessiglkonz. Salzsäure während ca. 1 - 2 Stunden.
2) Zur Herstellung des 6-Brom-1,2,2a,3,4,5-hexahy- drobenz[cd]indols kann man 1 -Acetyl-l ,2,2a,3,4,5-hexa- hydrobenz[cd]indol bromieren und das so erhaltene 1 -Acetyl-6-brom-1,2,2a,3,4,5-hexahydrobenz[cd]indol, welches spontan auskristallisiert, deacetylieren. Die Bromierung erfolgt z.B. durch Eintropfen von Brom in ein unter den Reaktionsbedingungen inertes organisches Lösungsmittel, z.B. Eisessig, bei ca. 10- 150 in Gegenwart einer katalytischen Menge Bromwasserstoff.
Die Deacetylierung kann wie unter 1) zur Herstellung von 6-Chlor -1 ,2,2a,3,4,5 - hexahydrobenz[cd]indol beschrieben, durch saure Hydrolyse durchgeführt werden.
3) Das 1 ,2,2a,3,4,5-Hexahydrn-6-hydrnxymethylbenz- [odindol wird z.B. hergestellt, indem man l-Acetyl -1,2,2a,3,4,5-hexahydrobenz[cd]indol nitriert, z.B. in Eisessig durch Zusatz von rauchender Salpetersäure bei etwa 100, die gebildete Nitro-Verbindung, die spontan auskristallisiert, zur entsprechenden Amino-Verbindung reduziert, die erhaltene Amino-Verbindung diazotiert, z.B. mittels Natriumnitrit in schwefelsaurer Lösung bei etwa 0- 5 , das entstandene Diazoniumsalz z.B. nach Sandmeyer zum 1 -Acetyl-6-cyan- 1 ,2,2a,3,4,5-hexahydro- benz[cd]indol umsetzt, die gebildete Cyan-Verbindung in das I-Acetyl-o-formyl-l ,2,2a,3,4,5 -hexahydrobenzcdl- indol überführt, diese Formyl-Verbindung reduziert, z.B.
mittels Natriumborhydrid in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel, z.B. einem niederen Alkanol wie Methanol, und das erhaltene 1 -Acetyl- 1 ,2,2a,3,4,5-hexahydro-6-hydroxymethylbenz[cd]indol durch alkalische Hydrolyse, z.B. durch Sieden mit wässerig-methanolischer Natriumhydroxid Lösung, deacetyliert.
Die Reduktion der Nitro-Verbindung erfolgt z.B. mit Hilfe von Hydrazinhydrat in Gegenwart von Raney Nickel in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel, z.B. einem niederen Alkanol wie Methanol, bei erhöhter Temperatur, vorzugsweise 50- 60 , und unter Rühren und dauert etwa 30 Minuten.
Nach beendeter Reaktion filtriert man vom Katalysator ab und verdampft schonend zur Trockne. Die gebildete Amino-Verbindung kann z.B. durch Kristallisation aus einem unter den vorliegenden Bedingungen inerten organischen Lösungsmittel, z.B. einem chlorierten Alkankohlenwasserstoff wie Methylenchlorid, gereinigt werden.
Das 1 -Acetyl-6-formyl- 1,2,2a,3,4,5- hexahydrobenz- [cd]indol kann man z.B. erhalten, indem man die entsprechende Cyan-Verbindung mit Natriumhypophosphit und Raney-Nickel in einem Gemisch von Eisessig/Pyridin/Wasser versetzt und während längerer Zeit bei einer Temperatur von etwa 10 - 250 rührt. Zur Aufarbeitung filtriert man vom Katalysator ab und dampft das Filtrat ein. Der Rückstand kann weiter aufgearbeitet werden, indem man ihn zwischen Wasser und einem damit nicht mischbaren, unter den vorliegenden Bedingungen inerten organischen Lösungsmittel, z.B. einem chlorierten Alkankohlenwasserstoff wie Methylenchlorid, ausschüttelt und die vereinigten organischen Phasen eindampft. Die rohe Formyl-Verbindung kann auf an sich bekannte Weise, z.B. chromatographisch, gereinigt werden.
4) Zur Herstellung des l,2,2a,3,4,5-Hexahydrn-6-me thylbenz[cd]indols kann man z.B. das unter 3) beschriebene 1-Acetyl-6-formyl - 1,2,2a,3,4,5 - hexahydrobenz[cd]indol reduzieren.
Diese Reduktion kann z.B. nach Wolf-Kishner bzw.
nach Modifikationen und Verbesserungen (z.B. Huang Minlon-Verfahren) durchgeführt werden, wobei gleichzeitig die Acetylgruppe hydrolytisch abgespalten wird.
5) 6-Fluor- 1 ,2,2a,3,4,5-hexahydrobenz[cd]indol und 1 ,2,2a,3 ,4,5-Hexahydro-6-jodbenz[cd] indol kann man z.B.
nach Sandmeyer aus dem unter 3) beschriebenen Diazoniumsalz herstellen.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen oder analog zu an sich bekannten Verfahren herstellbar.
In den nachfolgenden Beispielen, welche die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden und sind unkorrigiert.
Es ist klar. dass die C/N-Doppelbindung im Guanidinrest der Formel A nicht am Stickstoffatom in Position 2 (UPAC-Nomenklatur) fixiert ist, sondern auch zwischen dem Stickstoffatom in Position 3 oder 1 und dem Kohlenstoffatom liegen kann, soweit das Stickstoffatom in Position 3 oder 1 durch Wasserstoff substituiert ist.
Nomenklatur und Formeln sind in diesem Sinne zu interpretieren.

Beispiel 1
1-(2,3-Dimethyl uanidino)-1,2,2a,3,4,5-hexahydro- benzccinaoi
Eine Lösung von 10 g 1-(l,2,2a,3,4,5-Hexahydrobenz [cd]indol - 1 - yl)-2,3-dimethyl-isothioharnstoff-hydrojodid in 50 ml Äthanol wird nach Zugabe von 10 ml Methylamin 1 Stunde im Autoklaven bei Raumtemperatur stehengelassen und dann 3 Stunden auf 800 erhitzt. Beim Eindampfen der Lösung erhält man als Eindampfrückstand das Hydrojodid der im Titel genannten Verbindung, das aus Methanol/Äther in Prismen vom Smp.
255 - 2570C kristallisiert.
Das als Ausgangsmaterial benötigte 1-(1,2,2a,3,4,5 -Hexahydrobenz[cd]indol - 1 - yl)-2,3-dimethylisothioharnstoff-hydrojodid erhält man z.B. wie folgt: 1,2,2a,3,4,5-Hexahydrobenz[cd]indol wird in salzsaurer Lösung bei Raumtemperatur mit Natriumnitrit zum 1 ,2,2a,3,4,5-Hexahydro 1 -nitrosobenz[cd]indol nitrosiert (farblose Quader aus Äther/Petroläther vom Smp. 70 - 710). Hieraus erhält man durch Reduktion mit Lithiumaluminiumhydrid in Äther bei 5 - 80 l-Ami- no-1,2,2a,3,4,5-hexahydrobenz[cd]indol (aus Äther/Pe troläther Smp. 59 - 610).
Durch Umsetzung der Aminoverbindung mit Methylisothiocyanat in Benzol bei 600 gelangt man zum 1-(1 ,2,2a,3,4,5-hexahydrobenz[cd]indol- - 1 -yl)-3-methylthioharnstoff, (aus Methanol/Äther Smp.
184-1860) der durch zweistündiges Kochen mit Methyljodid in Methanol ins 1-(1 ,2,2a,3,4,5-Hexahydrobenz [cdjindol - 1 -yl) 2,3- dimethylisothioharnstoff-hydrojodid überführt wird (aus Methanol/Äther, Smp. 180- 1820).
Beispiel 2
1 -(2-Benzyl-3-methylgaamdino)-1,2,2a,3,4,5 -hexahydrobenz[cd]indol
10 g 1-(1,2,2a,3,4,5-Hexahydrobenz[cd]indol-l-yl)- -2,3-dimethylisothioharnstoff-hydrojodid werden in 50 ml Äthanol mit 25 ml Benzylamin 5 Stunden zum Sieden erhitzt. Man verdampft die Reaktionslösung am Vakuum zur Trockne und schüttelt den Eindampfrückstand zwischen Methylenchlorid und Wasser aus. Die über Magnesiumsulfat getrocknete organische Phase wird eingedampft und das als Eindampfrückstand erhaltene rohe 1 -(2-Benzyl -3- methylguanidino)- 1 ,2,2a,3,4,5-hexahydro- benz[cd]indol mit Methylenchlorid + 1% Äthanol an 30 Teilen Aluminiumoxid chromatographiert. Die im Titel genannte Verbindung wird anschliessend direkt in ihr Hydrogensulfat übergeführt, das aus Äthanol/ Äther mit 1 Mol Kristallalkohol kristallisiert.
Smp. 101 bis 1030.
Beispiel 3
1,2,2a,3,4,5-HexaXydro-1-(3-hydroxy-2- -methylguanidino)benz[cdlindol
Eine Lösung von 13 g 1-(1,2,2a3,4,5-Hexahydrobenz [cd]indol- 1 -yl) -2,3 - dimethylisothioharnstoff (freigesetzt aus dem Hydrojodid mit verdünnter Natriumhydroxidlösung) in 450ml Äthanol wird mit 43,8g Hydroxylaminhydrochlorid und 6,6 g Kalium äthylat versetzt und 3 Stunden unter Rückfluss gekocht. Nach dem Abkühlen saugt man Kaliumchlorid und überschüssiges Hydroxylaminhydrochlorid ab und verdampft die Lösung zur Trockne. Der Eindampfrückstand wird in Methylenchlorid gelöst, die Lösung klarfiltriert und dann mit 2 N Salzsäure ausgeschüttelt. Die saure Phase macht man mit konz.
Ammoniaklösung vorsichtig alkalisch, schüttelt 3mal mit Methylenchlorid aus und verdampft die über Magnesiumsulfat getrockneten Methylenchloridlösungen zur Trockne. Beim Verreiben des Eindampfrückstandes mit Äther erhält man die im Titel genannte Verbindung die nach anschliessender Kristallisation aus Methylenchlorid/Äther/Petroläther einen Smp. von 132 bis 1340 aufweist.
Beispiel 4
1-(3-Benzyl-2-hydroxyguanidino)-1,2,2a,3,4,5.
-hexahydrob.enz[cs]indol
Ein Gemisch von 19,1 g 1-(1,2,2a,3,4,5-Hexahydro- benz[cd] indol- 1 -yl)-3 -benzyl-2-methylisothioharnstoff-hydrochlorid, 19,7g Hydroxylaminhydrochlorid und 8,6 g Kaliumäthylat in 300 ml Äthanol wird 30 Minuten zum Sieden erhitzt. Nach dem Abkühlen saugt man das überschüssige Hydroxylamin-hydrochlorid und das bei der Reaktion gebildete Kaliumchlorid ab, verdampft das Filtrat zur Trockne und schüttelt den Eindampfrückstand zwischen Äther/konz. Ammoniaklösung aus. Beim Eindampfen der über Magnesiumsulfat getrockneten Ätherlösung erhält man das l-(3-Benzyl-2-hydroxygua nidino)- 1 ,2,2a,3,4,5-hexahydrobenz[cd]indol. Smp. 122 bis 1240, aus Methylenchlorid/Petroläther.
Das als Ausgangsmaterial benötigte 1-(1,2,2a,3,4,5 -Hexahydrobenz[cd] indol- 1 -yl)-3-benzyl-2-methylisothio- harnstoff-hydrochlorid erhält man z.B. wie folgt:
Durch Umsetzung von l-Amino-1,2,2a,3,4,5-hexa- hydrobenz[cd]indol mit Benzylisothiocyanat in siedendem Methylenchlorid während 8 Stunden gelangt man zum 1 -(1 ,2,2a,3,4,5.Hexahydrobenz[cd]indol- 1 -yl)-3-ben- zylthioharnstoff (aus Methylenchlorid/Äther, Smp. 149 bis 1510). Der Thioharnstoff wird 21/2 Stunden in Methanol mit Methyljodid unter Rückfluss gekocht. Den beim Eindampfen der Reaktionslösung erhaltenen Rückstand schüttelt man zwischen Äther und konz. Natriumhydroxidlösung aus. Die Ätherlösung wird über Natriumsulfat getrocknet und zur Trockne verdampft.
Den als Eindampfrückstand erhaltenen 1-(1 ,2,2a,3,4,5-Hexa- hydrobenz[cd]indol- 1- yl) -3- benzyl-2-methylisothioharn stoff überführt man direkt in sein Hydrochlorid. Smp.

Process for the production of new guanidine compounds
The present invention relates to a process for the preparation of new compounds of the formula I in which X is hydrogen, halogen, the hydroxymethyl or methyl group, R1 is a lower alkyl group, a benzyl or phenethyl group, optionally substituted by one or two of the following substituents - halogen , a lower alkyl or alkoxy group - may be substituted in the phenyl radical, and R represents a lower alkyl group, the benzyl or phenethyl group, which may be substituted by one or two of the following substituents - halogen, a lower alkyl or alkoxy group - in the phenyl radical can, a cycloalkyl group with 3 - 6 carbon atoms, the allyl, hydroxy, amino or dimethylamino group, and their acid addition salts.
According to the invention, the compounds of the formula I and their acid addition salts are obtained by reacting compounds of the formula II in which X and R1 have the above meaning and R5 stands for a lower alkyl radical with compounds of the formula III in which R2 has the above meaning, and if desired the compounds of the formula I thus obtained are converted into their acid addition salts.
It is preferable to work in the presence of an acid, but at least one of the reaction components should always be partially present as a base.
For example, one proceeds in such a way that compounds of formula II are used, preferably as acid addition salts, e.g. as hydrochloride, hydrobromide, hydroiodide or sulfate, with about 1-10 equivalents of a compound of the formula III.
The reaction is preferably carried out at elevated temperature, if appropriate in an autoclave, but can also be carried out in a polar solvent which is inert under the reaction conditions, such as e.g. a lower alkanol such as ethanol or isopropanol, a mixture of water with dimethylformamide, etc., at normal pressure and room temperature or elevated temperature. If the condensation is carried out without a solvent, a vacuum can be applied to remove the alkyl mercaptan more quickly (approx.
12 Torr). Instead of applying a vacuum, an inert gas can also be passed through or over the reaction mixture.
The guanidine compounds of the formula I prepared by the process described above can be isolated as free bases or in the form of acid addition salts in a customary manner and according to known methods, e.g. by crystallization.
Treatment with alkali, for example with an anion exchanger pretreated with an alkali, enables the corresponding bases to be liberated from the salts; for the preparation of acid addition salts the bases can be reacted with inorganic or organic acids, e.g. with hydrogen chloride, nitric acid, sulfuric acid, methanesulphonic acid, cyclohexylsulphamic acid, maleic acid, tartaric acid etc. It is also possible, however, to exchange the anion in the salts by double conversion.
The compounds of the formula I and their acid addition salts, referred to below as new substances for short, have not yet been described in the literature. In pharmacological testing on isolated organs and whole animals, they are characterized by diverse and highly pronounced effects and should therefore be used as medicinal products.
In particular, they can be used as circulatory drugs, psychopharmacas, salidiuretics and anti-inflammatory drugs, e.g. arises from the blood pressure-increasing, bradycardic, amine-increasing and circulatory reflex inhibiting effect in the anesthetized dog, from the inhibition of motility, the reserpine antagonism and a sympathetic stimulation in the mouse, from the salidiuretic activity in the rat and from the inhibition of carrageenedema in the rat.
The compounds of the formula I or their water-soluble, physiologically tolerable acid addition salts can be administered enterally or parenterally as medicaments, such as tablets, coated tablets, injection solutions, suppositories, etc., alone or in suitable medicinal forms.
The new starting compounds of the formula II can be prepared by combining compounds of the formula IV in which X has the above meaning with compounds of the formula V in which R1 has the above meaning to give the compounds of the formula VI in which X and R1 have the above meaning , and these are converted into the compounds of the formula II by reaction with alkyl halides or alkyl sulfates at elevated temperature, preferably at the boiling point of the reaction mixture.
To prepare the previously unknown compound of the formula IV required as a starting material, compounds of the formula VII in which X has the above meaning can be reduced, e.g. by means of zinc in formic acid or acetic acid, but preferably by means of a complex hydride of the alkali metals such as lithium aluminum hydride, sodium dihydro bis (2-methoxy ethoxy) aluminate etc. in an organic solvent which is inert under the reaction conditions, e.g. a cyclic or open-chain ether such as diethyl ether, at about 5 - 80.
The compounds of the formula VII are likewise new and can be obtained in a manner known per se from the compounds of the formula VIII, in which X has the above meaning. In practice one goes e.g. in such a way that a solution of compounds of the formula VIII in excess dilute hydrochloric acid is mixed with sodium nitrite.
The compounds of the formula VIIIa in which XI denotes halogen, the hydroxymethyl or methyl group are also new and can be prepared as follows:
1) 6-chloro-1,2,2a, 3,4,5-hexahydrobenz [cd] indole is obtained, for example, by adding l-acetyl-1,2,2a, 3,4,5-hexahydrobenz [cd] indole chlorinated, e.g. by passing chlorine into a solution of 1-acetyl-1,2,2a, 3,4,5-hexahydrobenz [odjindole in an organic solvent which is inert under the present conditions, e.g.
a chlorinated alkane hydrocarbon such as carbon tetrachloride, at room temperature and the spontaneously crystallizing 1-acetyl-6-chloro-1,2,2a, 3,4,5-hexahydrobenz [cd] indole deacetylated, e.g. by heating in a mixture of glacial acetic acid. Hydrochloric acid for approx. 1 - 2 hours.
2) For the preparation of 6-bromo-1,2,2a, 3,4,5-hexahydrobenz [cd] indole, 1-acetyl-1,2,2a, 3,4,5-hexahydrobenz [ cd] indole and deacetylate the 1-acetyl-6-bromo-1,2,2a, 3,4,5-hexahydrobenz [cd] indole thus obtained, which spontaneously crystallizes out. The bromination takes place e.g. by dripping bromine into an organic solvent which is inert under the reaction conditions, e.g. Glacial acetic acid, at about 10-150 in the presence of a catalytic amount of hydrogen bromide.
The deacetylation can be carried out as described under 1) for the preparation of 6-chloro -1, 2,2a, 3,4,5-hexahydrobenz [cd] indole, by acid hydrolysis.
3) The 1,2,2a, 3,4,5-hexahydron-6-hydrnxymethylbenz- [odindole is e.g. prepared by nitrating 1-acetyl -1,2,2a, 3,4,5-hexahydrobenz [cd] indole, e.g. in glacial acetic acid by adding fuming nitric acid at about 100, the nitro compound formed, which crystallizes out spontaneously, is reduced to the corresponding amino compound, the amino compound obtained is diazotized, e.g. by means of sodium nitrite in sulfuric acid solution at about 0-5, the resulting diazonium salt e.g. According to Sandmeyer, it is converted into 1-acetyl-6-cyano-1,2,2a, 3,4,5-hexahydrobenz [cd] indole, and the cyano compound formed into I-acetyl-o-formyl-1,2 , 2a, 3,4,5 -hexahydrobenzcdl- indole transferred, this formyl compound reduced, eg
by means of sodium borohydride in an organic solvent which is inert under the reaction conditions, e.g. a lower alkanol such as methanol, and the obtained 1-acetyl-1,2,2a, 3,4,5-hexahydro-6-hydroxymethylbenz [cd] indole by alkaline hydrolysis, e.g. deacetylated by boiling with aqueous-methanolic sodium hydroxide solution.
The reduction of the nitro compound takes place e.g. with the aid of hydrazine hydrate in the presence of Raney nickel in an organic solvent which is inert under the reaction conditions, e.g. a lower alkanol such as methanol, at an elevated temperature, preferably 50-60, and with stirring and takes about 30 minutes.
After the reaction has ended, the catalyst is filtered off and gently evaporated to dryness. The amino compound formed can e.g. by crystallization from an organic solvent which is inert under the present conditions, e.g. a chlorinated alkane hydrocarbon such as methylene chloride.
The 1-acetyl-6-formyl-1,2,2a, 3,4,5-hexahydrobenz- [cd] indole can e.g. obtained by adding sodium hypophosphite and Raney nickel in a mixture of glacial acetic acid / pyridine / water to the corresponding cyano compound and stirring for a prolonged period at a temperature of about 10-250. For work-up, the catalyst is filtered off and the filtrate is evaporated. The residue can be worked up further by dividing it between water and an immiscible organic solvent which is inert under the present conditions, e.g. a chlorinated alkane hydrocarbon such as methylene chloride, shaken out and the combined organic phases evaporated. The crude formyl compound can be prepared in a manner known per se, e.g. chromatographically, be purified.
4) For the preparation of the 1,2,2a, 3,4,5-hexahydron-6-methylbenz [cd] indole, e.g. reduce the 1-acetyl-6-formyl - 1,2,2a, 3,4,5 - hexahydrobenz [cd] indole described under 3).
This reduction can e.g. after Wolf-Kishner resp.
after modifications and improvements (e.g. Huang Minlon process), whereby the acetyl group is hydrolytically split off at the same time.
5) 6-fluoro-1, 2,2a, 3,4,5-hexahydrobenz [cd] indole and 1, 2,2a, 3, 4,5-hexahydro-6-iodobenz [cd] indole can be e.g.
Manufacture according to Sandmeyer from the diazonium salt described under 3).
If the preparation of the starting compounds is not described, they are known or can be prepared by processes known per se or analogously to those described here or analogously to processes known per se.
In the following examples, which explain the invention in more detail but are not intended to restrict its scope in any way, all temperatures are given in degrees Celsius and are uncorrected.

1- (2,3-Dimethyl uanidino) -1,2,2a, 3,4,5-hexahydrobenzccinaoi
A solution of 10 g of 1- (1,2,2a, 3,4,5-hexahydrobenz [cd] indol - 1 - yl) -2,3-dimethylisothiourea hydroiodide in 50 ml of ethanol is added after 10 ml Methylamine left to stand in the autoclave at room temperature for 1 hour and then heated to 800 for 3 hours. From this, reduction with lithium aluminum hydride in ether at 5-80 gives 1-amino-1,2,2a, 3,4,5-hexahydrobenz [cd] indole (from ether / petroleum ether, mp 59-610).
Reaction of the amino compound with methyl isothiocyanate in benzene at 600 gives 1- (1,2,2a, 3,4,5-hexahydrobenz [cd] indol- - 1 -yl) -3-methylthiourea, (from methanol / ether mp .
184-1860) which is converted into 1- (1, 2,2a, 3,4,5-hexahydrobenz [cdjindol - 1 -yl) 2,3-dimethylisothiourea-hydroiodide by boiling with methyl iodide in methanol for two hours (from methanol / ether , M.p. 180-1820).
Example 2
1 - (2-Benzyl-3-methylgaamdino) -1,2,2a, 3,4,5 -hexahydrobenz [cd] indole
10 g of 1- (1,2,2a, 3,4,5-hexahydrobenz [cd] indol-1-yl) -2,3-dimethylisothiourea hydroiodide are heated to boiling in 50 ml of ethanol with 25 ml of benzylamine for 5 hours . The reaction solution is evaporated to dryness in vacuo and the evaporation residue is shaken out between methylene chloride and water. The organic phase, dried over magnesium sulfate, is evaporated and the crude 1 - (2-benzyl -3-methylguanidino) -1, 2,2a, 3,4,5-hexahydrobenz [cd] indole obtained as evaporation residue with methylene chloride + 1% Ethanol chromatographed on 30 parts of aluminum oxide. The compound mentioned in the title is then converted directly into its hydrogen sulfate, which crystallizes from ethanol / ether with 1 mol of crystal alcohol.
M.p. 101 to 1030.
Example 3
1,2,2a, 3,4,5-HexaXydro-1- (3-hydroxy-2-methylguanidino) benz [cdlindole
A solution of 13 g of 1- (1,2,2a3,4,5-hexahydrobenz [cd] indol- 1 -yl) -2,3 - dimethylisothiourea (released from the hydroiodide with dilute sodium hydroxide solution) in 450 ml of ethanol is mixed with 43, 8 g of hydroxylamine hydrochloride and 6.6 g of potassium ethylate are added and the mixture is refluxed for 3 hours. After cooling, the potassium chloride and excess hydroxylamine hydrochloride are filtered off with suction and the solution is evaporated to dryness. The evaporation residue is dissolved in methylene chloride, the solution is filtered clear and then extracted with 2N hydrochloric acid. The acidic phase is made with conc.
Ammonia solution carefully alkaline, extracted 3 times with methylene chloride and evaporated the methylene chloride solutions dried over magnesium sulfate to dryness. When the evaporation residue is triturated with ether, the compound mentioned in the title is obtained which, after subsequent crystallization from methylene chloride / ether / petroleum ether, has a melting point of 132 to 1340.
Example 4
1- (3-benzyl-2-hydroxyguanidino) -1,2,2a, 3,4,5.

A mixture of 19.1 g of 1- (1,2,2a, 3,4,5-hexahydro- benz [cd] indol-1-yl) -3-benzyl-2-methylisothiourea hydrochloride, 19.7 g of hydroxylamine hydrochloride and 8.6 g of potassium ethylate in 300 ml of ethanol is heated to boiling for 30 minutes. After cooling, the excess hydroxylamine hydrochloride and the potassium chloride formed during the reaction are suctioned off, the filtrate is evaporated to dryness and the evaporation residue is shaken between ether / conc. Ammonia solution. Evaporation of the ether solution dried over magnesium sulfate gives l- (3-benzyl-2-hydroxygua nidino) - 1,2,2a, 3,4,5-hexahydrobenz [cd] indole. M.p. 122 to 1240, from methylene chloride / petroleum ether.
The 1- (1,2,2a, 3,4,5-hexahydrobenz [cd] indol-1 -yl) -3-benzyl-2-methylisothiourea hydrochloride required as starting material is obtained e.g. as follows:
Reaction of l-amino-1,2,2a, 3,4,5-hexa-hydrobenz [cd] indole with benzyl isothiocyanate in boiling methylene chloride for 8 hours gives 1 - (1, 2.2a, 3.4, 5. Hexahydrobenz [cd] indol-1-yl) -3-benzylthiourea (from methylene chloride / ether, m.p. 149 to 1510). The thiourea is refluxed in methanol with methyl iodide for 21/2 hours. The residue obtained on evaporation of the reaction solution is shaken between ether and conc. Sodium hydroxide solution. The ether solution is dried over sodium sulfate and evaporated to dryness.
The 1- (1,2,2a, 3,4,5-hexa-hydrobenz [cd] indol-1-yl) -3-benzyl-2-methylisothiourea obtained as evaporation residue is converted directly into its hydrochloride. M.p.
199 - 2010, from methanol / ether.
