Verfahren zur Herstellung neuer Guanidin-Verbindungen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Verbindungen der Formel I, worin X für Wasserstoff, Halogen, die Hydroxymethyl- oder Methylgruppe steht, R1 Wasserstoff, eine niedere Alkylgruppe, eine Benzyl- oder Phenäthylgruppe, die gegebenenfalls durch einen oder zwei der folgenden Substituenten - Halogen, niedere Alkyl- oder niedere Alkoxygruppe - im Phenylrest substituiert sein können, oder eine Cycloalkylgruppe mit 3 bis 6 Kohlenstoffatomen bedeutet und Ro für Wasserstoff, eine niedere Alkylgruppe oder die Hydroxygruppe steht und, falls R1 für Wasserstoff steht, auch die Dimethylamino. oder Allylgruppe bedeuten kann, oder R1 und R2 zusammen mit dem Stickstoffatom, woran sie gebunden sind,
die Morpholino-, 1 -Pyrrolidinyl-, Pip & dino-, 1-Azepinyl-, 4-Methylpiperazinyl- 1-, 4- (2-Hydroxyäthyl)piperazinyl -1-, 4-(2-Hydroxyäthyl)piperazinyl-1- oder eine 4-Phe nylpiperazinyl- 1-gruppe, welche gegebenenfalls im Phenylrest durch Halogen, die Methyl- oder Nitrogruppe monosubstituiert sein kann.
Erfindungsgemäss gelangt man zu den Verbindungen der Formel I und ihren Säureadditionssalzen, indem man Verbindungen der Formel II, worin X obige Bedeutung besitzt und R5 einen niederen Alkylrest bedeutet, mit Verbindungen der Formel III, worin R1 und R.
obige Bedeutung besitzen, umsetzt und gegebenenfalls die so erhaltenen Verbindungen der Formel T durch Umsetzung mit anorganischen oder organischen Säuren in die entsprechenden Salze überführt. Vorzugsweise arbeitet man in Gegenwart einer Säure, wobei jedoch stets mindestens eine der beiden Reaktionskomponenten teilweise als freie Base vorliegen soll.
Man geht beispielsweise so vor, dass man die Verbindungen der Formel II bzw. ihre Säureadditionssalze, z.B. das Hydrochlorid, Hydrobromid, Hydrojodid, Sulfat usw., mit etwa 1,1-10 Äquivalenten einer Verbindung der Formel III umsetzt. Diese Umsetzung wird vorzugsweise bei erhöhter Temperatur, gegebenenfalls im Autoklaven, durchgeführt, kann jedoch auch in einem unter den Reaktionsbedingungen inerten, polaren Lösungsmittel, wie z.B. Gemische von Wasser mit Dimethylformamid oder niederen Alkanolen, bei Normaldruck und Raumtemperatur oder erhöhter Temperatur erfolgen.
Falls die Kondensation ohne Lösungsmittel durchgeführt wird, kann zur rascheren Entfernung des Alkylmercaptans ein Vakuum angelegt werden (ca. 12 Torr.). Statt Anlegen eines Vakuums kann zu diesem Zwecke auch ein inertes Gas durch oder über das Reaktionsgemisch geleitet werden.
Die nach dem vorstehend beschriebenen Verfahren hergestellten Guanidinverbindungen der Formel I können als freie Basen oder in Form von Säureadditionssalzen auf übliche Weise isoliert und nach bekannten Methoden, z.B. durch Kristallisation, gereinigt werden.
Durch Behandlung mit Alkali, zum Beispiel mit einem alkalisch vorbehandelten Anionenaustauscher, lassen sich aus den Salzen die entsprechenden Basen freiset zen; zur Herstellung von Säureadditionssalzen können die Basen mit anorganischen oder organischen Säuren umgesetzt werden. Es ist aber auch möglich, in den Salzen das Anion durch doppelte Umsetzung auszutauschen.
Die Verbindungen der Formel I und ihre Säureadditionssalze, im folgenden kurz als neue Substanzen bezeichnet, sind, in der Literatur bisher nicht beschrieben worden. Sie zeichnen sich in der pharmakologischen Prüfung an isolierten Organen und am Ganztier durch vielfältige und stark ausgeprägte Effekte aus und sollen daher als Heilmittel verwendet werden.
Insbesondere können sie als Kreislaufmittel, Salidiuretica, Antiphlogistica und Psychopharmaca verwendet werden, wie z.B. aus der Blutdruck-steigernden, bradycarden, Amin-verstärkenden und Kreislaufreflex-hemmenden Wirkung am narkotisierten Hund, aus der Senkung des künstlich erhöhten Blutdrucks bei der hypertonen Grollmann-Ratte, aus der vasokontriktorischen Aktivität an der isolierten, perfundierten Kaninchen Extremität, aus der positiv inotropen Wirksamkeit am isolierten Katzenherzen, aus der salidiuretischen Wirkung an der Ratte, aus der Hemmwirkung auf das Carrageen-Oedem an der Ratte sowie aus der Motolitätshemmung, Reserpin-antagonistischen, analgetischen und dem sympathisch stimulierenden Effekt an der Maus hervorgeht.
Als Heilmittel können die neuen Verbindungen der Formel I bzw. ihre physiologisch verträglichen Säureadditionssalze allein oder in geeigneten Arzneiformen verabreicht werden.
Die neuen Ausgangsverbindungen der Formel II können hergestellt werden, indem man Verbindungen der Formel IV, worin X obige Bedeutung besitzt, mit N-Benzoylisothiocyanat oder einem Gemisch von Ammoniumrhodanid und Benzoylchlorid in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel, z.B. einem cyclischen Äther wie Tetrahydrofuran, bei erhöhter Temperatur, vorzugsweise bei Siedetemperatur des Reaktionsgemisches zu Verbindungen der Formel V, worin X obige Bedeutung besitzt, umsetzt, die Verbindungen der Formel V zu den Verbindungen der Formel VI, worin X obige Bedeutung hat, hydrolysiert, z.B. durch kurzes Erwärmen mit wässriger Natriumhydroxidlösung, und die Verbindungen der Formel VI z.B. mit Alkylhalogeniden bzw. Alkylsulfaten bei erhöhter Temperatur, vorzugsweise bei Siedetemperatur des Reaktionsgemisches, zu den Verbindungen der Formel II umsetzt.
Zur Herstellung der als Ausgangsprodukt benötigten, bisher unbekannten Verbindungen der Formel IV kann man Verbindungen der Formel VII, worin X obige Bedeutung besitzt, reduzieren, z.B. mittels Zink in Ameisensäure, vorzugsweise aber mittels eines komplexen Hydrids der Alkalimetalle wie Lithiumaluminiumhydrid, Natrium-dihydro-bis(2-methoxyäthoxy)aluminat usw. in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel, z.B. einem cyclischen oder offenkettigen Äther wie Diäthyläther, bei etwa 5 - 80.
Die Verbindungen der Formel VII sind ebenfalls neu und können auf an sich bekannte Weise aus den Verbindungen der Formel VIII, worin X obige Bedeutung besitzt, erhalten werden. Praktisch geht man z.B.
so vor, dass man eine Lösung von Verbindungen der Formel VIII in überschüssiger verdünnter Salzsäure mit Natriumnitrit versetzt.
Die Verbindungen der Formel VIIIa, worin XI Halogen, die Hydroxymethyl- oder Methylgruppe bedeutet, sind auch neu und können folgendermassen hergestellt werden:
1) 6-Chlor- 1 ,2,2a,3,4,5-hexahydrobenz[cd]indol erhält man beispielsweise, indem man 1-Acetyl-1,2,2a,3,4,- 5-hexahydrobenz[cd]indol chloriert, z.B. durch Einleiten von Chlor in eine Lösung von l-Acetyl-1,2,2a,3,4,5-he- xahydrobenz[cd]indol in einem unter den vorliegenden Bedingungen inerten organischen Lösungsmittel, z.B.
einem chlorierten Alkankohlenwasserstoff wie Tetrachlorkohlenstoff, bei Raumtemperatur und das spontan kristallisierende 1 -Acetyl-6-chlor- 1 ,2,2a,3,4,5-hexahydro- benz[cd]indol deacetyliert, z.B. durch Erhitzen in einem Gemisch von Eisessig/konz. Salzsäure während ca. 1 - 2 Stunden.
2) Zur Herstellung des 6-Brom-1,2,2a.3,4,5-hexahy- drobenztcd]indols kann man 1-Acetyl-1,2,2a,3,4,5-hexa- hydrobenz[cd]indol bromieren und das so erhaltene 1 -Acetyl-6-brom-1,2,2a,3,4,5-hexahydrobenz[cdlindol, welches spontan auskristallisiert, deacetylieren. Die Bromierung erfolgt z.B. durch Eintropfen von Brom in ein unter den Reaktionsbedingungen inertes organisches Lösungsmittel, z.B. Eisessig, bei ca. 10- 150 in Gegenwart einer katalytischen Menge Bromwasserstoff.
Die Deacetylierung kann wie unter 1) zur Herstellung von 6- Chlor -1 ,2,2a,3 4,5- hexahydrobenz[ed]indol beschrieben, durch saure Hydrolyse durchgeführt werden.
3) Das 1 ,2,2a,3,4,5-Hexahydro.6-hydroxymethylbenz- [cd]indol wird z.B. hergestellt, indem man l-Acetyl -1,2,2a,3,4,5-hexahydrobenz[cd]indol nitriert, z.B. in Eisessig durch Zusatz von rauchender Salpetersäure bei etwa 100, die gebildete Nitro-Verbindung, die spontan auskristallisiert, zur entsprechenden Amino-Verbindung reduziert, die erhaltene Amino-Verbindung diazotiert, z.B. mittels Natriumnitrit in schwefelsaurer Lösung bei etwa 0- 5 , das entstandene Diazoniumsalz z.B.
nach Sandmeyer zum 1-Acetyl-6-cyan- 1 ,2,2a,3,4,5-hexahydro- benz[cd]indol umsetzt, die gebildete Cyan-Verbindung in das 1-Acetyl-6-formyl-1,2,2a,3,4,5-hexahydrobenz[cd]- indol überführt, diese Formyl-Verbindung reduziert, zB. mittels Natriumborhydrid in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel, z.B. einem niederen Alkanol wie Methanol, und das erhaltene 1 -Acetyl- 1 ,2,2a,3,4,5-hexahydro-6-hydroxymethylbenz[cd]indol durch alkalische Hydrolyse, z.B. durch Sieden mit wässerig-methanolischer Natriumhydroxid Lösung, deacetyliert.
Die Reduktion der Nitro-Verbindung erfolgt z.B. mit Hilfe von Hydrazinhydrat in Gegenwart von Raney Nickel in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel, z.B. einem niederen Alkanol wie Methanol, bei erhöhter Temperatur, vorzugsweise 50- 600, und unter Rühren und dauert etwa 30 Minuten.
Nach beendeter Reaktion filtriert man vom Katalysator ab und verdampft schonend zur Trockne. Die gebildete Amino-Verbindung kann z.B. durch Kristallisation aus einem unter den vorliegenden Bedingungen inerten organischen Lösungsmittel, z.B. einem chlorierten Alkankohlenwasserstoff wie Methylenchlorid, gereinigt werden.
Das 1 - Acetyl-6-formyl - 1 ,2,2a,3,4,5 - hexahydrobenz [cd]indol kann man z.B. erhalten, indem man die entsprechende Cyan-Verbindung mit Natriumhypophosphit und Raney-Nickel in einem Gemisch von Eisessig/Pyridin/Wasser versetzt und während längerer Zeit bei einer Temperatur von etwa 10- 250 rührt. Zur Aufarbeitung filtriert man vom Katalysator ab und dampft das Filtrat ein. Der Rückstand kann weiter aufgearbeitet werden, indem man ihn zwischen Wasser und einem damit nicht mischbaren, unter den vorliegenden Bedingungen inerten organischen Lösungsmittel. z.B. einem chlorierten Alkankohlenwasserstoff wie Methylenchlorid, ausschüttelt und die vereinigten organischen Phasen eindampft. Die rohe Formyl-Verbindung kann auf an sich bekannte Weise, z.B. chromatographisch, gereinigt werden.
4) Zur Herstellung des 1 ,2,2a,3,4,5-Hexahydro-6-me- thylbenz[cdindols kann man z.B. das unter 3) beschriebene 1 -Acetyl-6-formyl - 1 ,2.2a,3,4.5 - hexahydrobenz[cd]indol reduzieren.
Diese Reduktion kann z.B. nach Wolf-Kishner bzw.
nach Modifikationen und Verbesserungen (z.B. Huang Minlon-Verfahren) durchgeführt werden, wobei gleichzeitig die Acetylgruppe hydrolytisch abgespalten wird.
5) 6-Fluor-1 ,2,2a,3,4,5-hexahydrobenz[cd]indol und 1,2,2a.3,4,5-Hexahydro-6-jodbenzrcd]indol kann man z.B.
nach Sandmeyer aus dem unter 3) beschriebenen I)i azoniumsalz herstellen.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier be schriebenen oder analog zu an sich bekannten Verfahren herstellbar.
In den nachfolgenden Beispielen, welche die Erfindung näherer läutern, ihren Umfang aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden und sind unkorrigiert.
Es ist klar, dass die C/N-Doppelbindung im Guan dinrest der Formel A nicht am Stickstoffatom in Positio 2 (UPAC-Nomenklatur) fixiert ist, sondern auch zw schen dem Stickstoffatom in Position 3 oder 1 und der Kohlenstoffatom liegen kann, soweit das Stickstoffator in Position 3 oder 1 durch Wasserstoff substituiert is Nomenklatur und Formeln sind in diesem Sinne zu tinte pretieren.

Beispiel I
1,2,2a,3,4,5-Hexahydro-1- < 2-methylguanidino)- benz[cd] indol
Eine Lösung von 2g 1-(1,2,2a,3,4,5-Hexahydrobenz [cd]indol-1-yl)-2- methyllsothiobarnstoff - hydrojodid in l0ml Methanol wird nach Zugabe von 10ml Methyl imin 1 Stunde im Autoklaven bei Raumtemperatur gtehengelassen und dann 2 Stunden auf 800 erhitzt. Beim
Eindampfen der Lösung erhält man als Eindampfrückstand das Hydrojodid der im Titel genannten Verbin Jung, das aus Methanol/Äther in Prismen vom Smp.
ZX)9 - 2110 kristallisiert.
Das als Ausgangsmaterial benötigte 1-(1,2,2a,3,4,5 -Hexahydrobenz [cd]indol- 1 -yl)-2-methylisothioharnstoff- -hydrojodid erhält man wie folgt: 1,2,2a,3,4,5-Hexahydrobenz[cd]indol wird in salzsaurer Lösung bei Raumtemperatur mit Natriumnitrit zum 1,2,2a,3,4,5-Hexahydro-1-nitroso-benz[cd]indol nitrosiert. (Farblose Quader aus Äther/Petroläther vom Smp.
70- 710) Hieraus erhält man durch Reduktion mit Lithiumaluminiumhydrid in Äther bei 5 - 80 l-Amino -1,2,2a,3,4,5-hexahydrobenz[cd]indol. (Aus Äther/Petroläther Smp. 59 - 610). Durch Umsetzung der Aminoverbindung mit N-Benzoylisothiocyanat in siedendem Tetrahydrofuran und anschliessender Verseifung mit verdünnter Natriumhydroxid-Lösung unter Rückfluss während 10 Minuten gelangt man zum 1-(1,2,2a,3,4,5-Hexa hydrobenz[cd]indol- 1 -yl)thioharnstoff. Smp. 182 - 1840.
(Aus Methylenchlorid/Petroläther). Der Thioharnstoff wird durch Kochen während 4 Stunden mit Methyljodid in Methanol in das 1-(1,2,2a,3,4,5-Hexahydrobenz[cd]- indol-1-yl) - 2- methylisothioharnstoff hydrojodid überführt, das aus Methanol/Äther kristallisiert. Smp. 198 bis 2010.
Beispiel 2
1-(3,3-Dimethylguanidino)-1,2,2a,3,4,5-hexa- hydrnbenzc < i\ indol
Hergestellt unter Verwendung von 1-(1,2,2a,3,4,5 -hydrojodid und 33%iger äthanolischer Dimethylamin - Hexahydrobenz[cd]indol- 1 -yl)-2-methylisothioharnstofflösung analog Beispiel 1. Aus dem rohen Hydrojodid setzt man mit 50%iger Kalilauge die Base frei und überführt diese mit 2 N äthanolischer Salzsäure direkt in das Hydrochlorid der im Titel genannten Verbindung.
Smp. 258 - 2610 aus Methanol/Äther.
Beispiel 3
1-(2-Allylguanidino)-1 ,2,2a,3,4,5-hexahydrobenz [cd]indol
12 g 1 - (1,2,2a,3,4,5-Hexahydrobenz[cd]indol-1-yl)-2- -methylisothioharnstoff-hydrojodid werden in 50 ml Methanol gelöst und nach Zugabe von 12 mol Allylamin 6 Stunden zum Sieden erhitzt. Die Lösung verdampft man zur Trockne und schüttelt den Eindampfrückstand zwischen Methylenchlorid und 20%iger Natriumhydroxid Lösung aus. Die organische Phase wird über Magnesiumsulfat getrocknet und eingedampft. Als Eindampfrückstand erhält man 1 -(2-Allylguanidino)- 1,2,2a,3 ,4,5-hexa- hydrobenzcJ]indol, das direkt in sein Hydrogenmaleinat übergeführt wird. Smp. 105- 1070 aus Methanol/ Äther.
Beispiel 4
1,2,2a,3,4,5-Hexahydro-1-(2-phenäthylguanidino)- lenzk < t\ indol
Hergestellt unter Verwendung von 1-(1,2,2a,3,4,5 -Hexahydrobenz [cd] indol- 1 yl)-2-methylisothioharnstoff- -hydrojodid, 2-Phenyläthylamin und Äthanol als Lösungsmittel analog Beispiel 3. Das Hydrogenmaleinat der im Titel genannten Verbindung kristallisiert aus Methanol/Äther vom Smp. 148 - 1500.
Beispiel 5 1-(2-Benzylguanidino)-1 ,2,2a,3,4,5-hexahydrobenz- [cd]indol
Hergestellt unter Verwendung von 1-(1,2,2a,3,4,5 - Hexahydrobenz[cd]indol- 1 -yl)-2-methylisothioharnstoff - -hydrojodid, Benzylamin und Äthanol als Lösungsmittel und einer Reaktionszeit von 3 Stunden analog Beispiel 3. Das Hydrogenmaleinat der im Titel genannten Verbindung kristallisiert aus Methanol/Äther vom Smp.
195 - 1970.
Beispiel 6
1-(2-Cyclopropylguanidino)-1,2,2a,3,4,5-hexa- hyd robenz[cd]indol
12 g 1 -(1 ,2,2a,3,4,5-Hexahydrobenz[ed]indol- 1 -yl)-2 -methylisothioharnstoff-hydrojodid werden in 50 ml Äthanol gelöst und nach Zugabe von 24 ml Cyclopropylamin 5 Stunden im Autoklaven auf 800 erhitzt. Man verdampft die Lösung zur Trockne und schüttelt den Eindampfrückstand zwischen Methylenchlorid und konz Natriumhydroxid-Lösung aus. Die über Magnesiumsulfat getrocknete organische Phase wird eingedampft und das als Eindampfrückstand erhaltene rohe 1 -(2-Cyclo- propylguanidino)- 1 ,2,2a,3,4,5-hexahydrobenz[cd]indol an 30 Teilen Aluminiumoxid chromatographiert.
Die im Titel genannte Verbindung wird mit Methylenchlorid + 1% Äthanol von der Säule gewaschen und anschliessend direkt in sein Hydrogenmaleinat überführt. Smp.
98 - 1000 aus Äthanol/Äther.
Beispiel 7
1-(2- DimetEylaminoguarridino)- 1,2,2a,3, 4,5-hexa- hydroSenz[ctitrdol
12 g 1-(1,2,2a,3,4,5-Hexahydrobenz[cd]indol- 1-yl)-2- -methylisothioharnstoff-hydrojodid werden in 50 ml Methanol gelöst und nach Zugabe von 12 ml N,N-Dimethylhydrazin 31/2 Stunden zum Sieden erhitzt. Man verdampft die Lösung zur Trockne und nimmt den Eindampfrückstand in Äther und Wasser auf. Ein hierbei unlöslicher Teil wird abfiltriert und anschliessend werden die Schichten getrennt. Man macht die wässrige Phase mit konz.
Natriumhydroxid-Lösung alkalisch und schüttelt 3 X mit Methylenchlorid aus. Die über Magnesiumsulfat getrockneten organischen Phasen werden zur Trockne verdampft und das als Eindampfrückstand erhaltene 1 - (2. Dimethylaminoguanidino) - 1,2,2a,3,4,5 hexahydro benz[cd]indol direkt in sein Naphthalin-l,5-disulfonat überführt. Smp. 20{) - 2030 aus Methanol/Äther/Aceton.
Beispiel 8
1,2,2a,3,4,5-Hexahydro-1-(3-hydroxy-3-niethyl- guanidhio)benzkd\ indol
Eine Lösung von 15 g 1-(1,2,2a,3,4,5-Hexahydro- benz[cd]indol-l-yl) - 2 -methylisothioharnstoff-hydrojodid in 200 ml Äthanol wird mit 13,5 g Methylhydroxylamin-hydrogenoxalat versetzt und nach Zugabe von 4,4 g Kaliumhydroxid in 20 ml Wasser 1t/2 Stunden zum Sieden erhitzt. Nach dem Erkalten wird das Reaktionsgemisch filtriert und das Filtrat zur Trockne verdampft.
Man löst den Eindampfrückstand in wenig Äthanol, fügt 3,4 g Kaliumäthylat zu und fällt mit Äther alle ätherunlöslichen Anteile aus. Das Gemisch wird nun klarfiltriert durch eine Talkschicht und das Filtrat zur Trockne verdampft. Das als Eindampfrückstand erhaltene 1,2,2a,3,4,5-Hexahydro-1-(3-hydroxy-3-methylguani- dino)benz[cd]indol wird direkt in sein Hydrogenmaleinat überführt, das aus Methanol/Äther vom Smp. 152 bis 1540 kristallisiert.
Beispiel 9 N-(l ,2,2a,3,4,5-Hexahydrobenz[cd]indol-1 -yl)-4 -methylpiperazin-carboxamidin
10 g 1-(1,2,2a,3,4,5-Hexahydrobenz[cd]indol-1-yl)-2- -methylisothioharnstoff-hydrojodid werden in 50 ml Äthanol gelöst und nach Zugabe von 20 ml N-Methylpiperazin 7 Stunden zum Sieden erhitzt. Man verdampft die Lösung zur Trockne und schüttelt den Eindampfrückstand zwischen Methylenchlorid und Wasser aus.
Beim Eindampfen der über Magnesiumsulfat getrockneten Methylenchloridlösung erhält man als Eindampfrückstand N-(l,2,2a,3,4,5-Hexahydrobenz[ed]indol-l -yl)- -4-methylpiperazincarboxamidin, das man direkt in sein Bis-hydrogenmaleinat überführt. Smp. 153 - 1560 aus Methanol/Äther.
Beispiel 10
1,2,2a,3,4,5-Hexahydro-1-(2-hydroxyguanidino)- benz[cd] indol
9 g 1-(1,2,2a,3,4,5-Hexahydrobenz[cd]indol-1-yl)-2- -methylisothioharnstoff-hydrochlorid und 8,7 g Hydroxylaminhydrochlorid werden in 200 ml Äthanol suspendiert und nach Zugabe von 5,3 g Kaliumäthylat 30 Minuten zum Sieden erhitzt. Nach dem Abkühlen saugt man Kaliumchlorid und überschüssiges Hydroxylaminhydrochlorid ab und verdampft das Filtrat zur Trockne.
Der Eindampfrückstand wird in Methylenchlorid gelöst, die Lösung klarfiltriert und eingeengt. Zu der klaren Lösung fügt man Äther, wobei das 1,2,2a,3,4,5-Hexa- hydro -1 - (2 (2-hydroxyguanidino)benz[cd]indol-hydrochlo- rid auskristallisiert, das nach Umkristallisation aus Methanol/Äther einen Smp. von 171 - 1730 aufweist.

N-(1,2,2a,3,4,5-Hexahydrobenz[cd]indol-1-yl)-4- - tolylpiperazincarboxwnidin
10,2 g 1(1 ,2,2a,3,4,5-Hexahydrobenz[od]indol 1 -yl)-2- -methylisothioharnstoff-hydrojod id und 7,1 g N-o-Tolylpiperazin werden 20 Minuten auf 2000 erhitzt. Das Reaktionsprodukt löst man in Methanol, filtriert die Lösung mit Aktivkohle klar und gibt zu der klaren Lösung Äther, wobei das Hydrojodid der im Titel genannten Verbindung auskristallisiert und nach Umkristallisation aus Methanol/Äther bei 248 - 2510 schmilzt.
Beispiel 12
1-(2-p-Chlorbenzylguanidino)-1,2 ,2,2a,36-hexa- hydrobenz [cd] indol
15 g 1- (1,2,2a,3,4,5-Hexahydrobenz[ed]indol- 1 -yl)-2- -methylisothioharnstoff-hydrojodid und 8,6 g 4-Chlorbenzylamin werden 15 Minuten auf 2000 erhitzt. Das Reaktionsprodukt schüttelt man zwischen Methylenchlo r.id und konz. Natriumhydroxid-Lösung aus und verdampft die über Magnesiumsulfat getrocknete Methylen chloridlösung zur Trockne. Das als Eindampfrückstand erhaltene 1-(2-p-Chlorbenzylguanidino)-1,2,2a,3,4,5-hexa- hydrobenz[cd]indol wird direkt in sein Hydrochlorid übergeführt. Smp. 162- 1650 aus Methanol/Äther.

1,2,2a,3,4,5-Hexahydro-1-(2-p-methylbenzyl- guccnidino)lenz [cdlind.ol
Hergestellt unter Verwendung von 1-(1,2,2a,3,4,5 - Hexahydrobencdindol- 1 -y1)-2-methylisothioharnstoff -hydrojodid und 4-Methylbenzylamin analog Beispiel 12.
Das Hydrochlorid der im Titel genannten Verbindung kristallisiert aus Methanol/Äther. Smp. 142- 1450.

1 ,2,2a,3,4,5-Hexahydro-1-(2-p-methoxybenzyl- guanidino)benz[cd] indol
Hergestellt unter Verwendung von 1-(1,2,2a,3,4,5 - Hexahydrobenz[cd]indol- 1 -yl)-2-methylisothioharnstoff -hydrojodid und 4-Methoxybenzylamin analog Beispiel 12. Das Hydrochlorid der im Titel genannten Verbindung kristallisiert aus Methanol/Äther Smp. 142- 1450.
Beispiel 15 1 -Guanidino- 1,2 ,2a,3 ,4,5-hexahydrolenz [cd]indol
Eine Lösung von 2 g 1-(1,2,2a,3,4,5-Hexahydrobenz- [cd]indol-l-yl)-2-methylisothioharnstoff-hydrojodid in 10 ml Methanol wird nach Zugabe von 10 ml flüssigem Ammoniak 8 Stunden im Autoklaven auf 1000 erhitzt. Anschliessend verdampft man die Reaktionslösung zur Trockne und schüttelt den Eindampfrückstand zwischen Methylenchlorid und Wasser aus. Die über Magnesiumsulfat getrocknete Methylenchloridlösung wird eingedampft und das als Eindampfrückstand erhaltene l-Gua- nidino- 1 ,2,2a,3,4,5-hexahydrobenz[cd]indol aus Methanol/Wasser kristallisiert. Smp. 153 - 1540.
Beispiel 16 6-Brnm-1-guanidino-1,2,2a,3,4,5-h & hydrnhenzkd]indol
Hergestellt unter Verwendung von 1-(6-Brom-1,2,- 2a,3,4,5 - hexahydrobenzEcd]indol - 1 - yl) -2-methylisothioharnstoff-hydrojodid analog Beispiel 15. Das 6-Brom-l -guanidino-1,2,2a,3,4,5-hexahydrobenz[cd]indol wird in sein Hydrochlorid überführt. Smp. 251 - 2530 aus Methanol /Äther.
Das als Ausgangsprodukt verwendete 1-(6-Brom-1,2,- 2a,3,4,5 - hexahydrobenz[cd]indol- 1-yl) - 2 - methylisothioharnstoff-hydrojodid erhält man wie folgt:
1-Acetyl-1,2,2a,3,4,5-hexahydrobenz[cd]indol wird bei 150 in Eisessig in Gegenwart von katalytischen Mengen HBr mit Brom bromiert. Das hierbei erhaltene l-Acetyl- -6-brom- 1 ,2,2a,3,4,5-hexahydrobenz[cd]indol (Smp. 150 bis 1510, aus Methylenchlorid/Petroläther) verseift man durch ll/2stündiges Kochen in konz. Salzsäure/Eisessig zum 6-Brom-1,2,2a,3,4,5-hexahydrobenz[cd]indol. (Smp.
116-1180, aus Methylenchlorid/Petroläther). Dieses wird in salzsaurer Lösung bei 50 mit Natriumnitrit zum 6-Brom- l,2,2a,3,4,5-hexahydro- l-nitrosobenz[cd]in- dol nitrosiert. (Stäbchen aus Methylenchlorid/Petrol äther vom Smp. 110- 1120). Hieraus erhält man durch Reduktion mit Lithiumaluminiumhydrid in Äther bei 5 - 8 1-Amino -6- brom-1,2,2a,3,4,5-hexahydrobenz[ed]- indol. (Aus Äther/Petroläther, Smp. 78 - 800) durch Umsetzung der Aminoverbindung mit N-Benzoylisothiocyanat in siedendem Tetrahydrofuran und anschliessender Verseifung mit verd.
Natriumhydroxid unter Rückfluss während 10 Minuten gelangt man zum 1-(6-Brom - 1 ,2,2a,3 4,5- hexahydrobenz[cd]indol 1 -yl)-thioharnstoff, der direkt durch 4stündiges Kochen mit Methyljodid in Methanol in das 1-(6-Brom-1,2,2a,3,4,5-hexahydrobenz[cd]indol- 1 -yl) -2- methylisothioharnstoff-hydrojodid überführt wird, das ohne Charakterisierung direkt weiterverarbeitet wird.
Beispiel 17
6-Chlor-1.guanidino-1,2,2a,3,4,5-hexdhydrobenz- [cd]indol
Hergestellt unter Verwendung von 1-(6-Chlor-1,2,2a,3,4,5 - hexahydrobenz[cdzindol-1-yl) - 2 - methylisothioharnstoff-hydrojodid analog Beispiel 15. Das 6-Chlor-1- -guanidino- 1 ,2,2a,3,4,5-hexahydrobenz[cd]indol wird direkt in sein Hydrochlorid überführt. Smp. 232- 235, aus Methanol/Äther.
Das als Ausgangsmaterial verwendete 1-(6-Chlor-1,- 2,2a,3,4,5 - hei:ahydrobenz[cdindol- 1 -yl)-2-methylisothioharnstoff-hydrojodid erhält man wie folgt: 1 -Acetyl- 1 ,2,2a,3,4,5-hexahydrobenz[cd]indol wird bei Raumtemperatur in Tetrachlorkohlenstoff mit der berechneten Menge Chlor chloriert. Das hierbei erhaltene
1 -Acetyl - 6 - chlor - 1,2,2a,3,4,5 - hexahydrobenz[cd]indol (Smp. 141 - 1430, aus Methylenchlorid/Petroläther) verseift man durch 11/2stündiges Kochen in konz. Salzsäure/ Eisessig zum 6-Chlor-1 ,2,2a,3,4,5-hexahydrobenz[cd]in- dol. (Smp. 109 - 1110, aus Äther/Petroläther).
Dieses wird in salzsaurer Lösung bei 50 mit Natriumnitrit zum 6-Chlor- 1,2,2a,3,4,5-hexahydro-1 -nitrosobenzfed]indol nitrosiert. (Smp. 123 - 1250, aus Äther.) Hieraus erhält man durch Reduktion mit Lithiumaluminiumhydrid in Äther bei 5 - 80 l-Amino-6-chlor-1,2,2a,3,4,5-hexahydro- beni[cd]indol. (Aus Äther/Petroläther, Smp. 74- 760).
Durch Umsetzung der Aminoverbindung mit N-Benzoylisothiocyanat in siedendem Tetrahydrofuran und anschliessender Verseifung mit verd. Natriumhydroxid Lösung unter Rückfluss während 10 Minuten gelangt man zum 1 -(6-Chlor- 1 ,2,2a,3,4,5-hexahydrobenz[cd]in- dol-l-yl)-thioharnstoff. Aus Methanol/Äther, Smp. 226 bis 2280. Der Thioharnstoff wird durch Kochen während 4 Stunden mit Methyljodid in Methanol in das 1 -(6-Chlor- 1,2,2a,3,4,5-hexahydrobenz[cd]indol-1-yl)-2-me- thylisothioharnstoff-hydrojodid überführt, das aus Methanol/Äther kristallisiert. Smp. 174 - 1770.
Beispiel 18
1-Guanidino-1,2,2a,3,4,5-hexahydro-6-methyl- lenz[cd\indol
Hergestellt unter Verwendung von 1-(1,2,2a,3,4,5 Hexahydro-6-methylbenz[cd]indol- 1 -yl)-2-methylisothio- harnstoff-hydrojodid analog Beispiel 15. Das Hydrochlorid der im Titel genannten Verbindung kristallisiert aus Methanol/Äther. Smp. 238 - 2400.
Das als Ausgangsmaterial benötigte 1-(1,2,2a,3,4,5 -Hexahydro-6-methylbenz[cdTindol- 1- yl)-2methylisothio- harnstoff-hydrojodid erhält man wie folgt: l-Acetyl-1,2,2a,3,4,5-hexahydrobenz[cd]indol wird bei 100 in Eisessig mit rauchender Salpetersäure nitriert.
Das hierbei erhaltene 1 -Acetyl- 1 ,2,2a,3,4,5-hexahydro- 6- -nitrobenz[cd]indol (Smp. 174- 1750, aus Methylenchlo rid/Äther) reduziert man in Methanol bei 50- 600 mit Hydrazinhydrat in Gegenwart von Raney-Nickel zum 1 -Acetyl - 6 -amino - 1,2,2a,3,4,5- hexahydrobenztcd]indol.
(Smp. 147 - 1490, aus Methylenchlorid/Petroläther). Die Aminoverbindung wird in schwefelsaurer Lösung bei 0-50 mittels Natriumnitrit diazotiert und das so erhaltene Diazoniumsalz nach Sandmeyer mit Kupfercyanür bei 600 in das 1-Acetyl-6-cyan-l,2,2a,3,4,5-hexahydro- benz[cd]indol überführt. (Smp. 169 - 1710, aus Methylenchlorid/Petroläther).
Hieraus erhält man durch 20stündiges Rühren mit Natriumhypophosphit und Raney-Nickel in einem Gemisch von Eisessig/Pyridin/ Wasser bei Raumtemperatur das l-Acetyl-6-formyl-1,2,- 2a,3,4,5-hexahydrobenz[cd]indol, (Smp. 145 - 1470, aus Methylenchlorid / Petroläther) das nach dem Huang-Minlon-Verfahren unter gleichzeitiger Abspaltung der Acetylgruppe in das 1 ,2,2a,3,4,5-Hexahydro-6-methylbenz- [cd]indol überführt wird. (Smp. 86- 880, aus Methanol/ Wasser). Dieses wird in salzsaurer Lösung bei 5 mit Natriumnitrit zum l,2,2a,3,4,5-Hexahydro-6-methyl- 1- -nitrosobenz[cd]indol nitrosiert. (Smp. 103 - 1050, aus Äther).
Hieraus erhält man durch Reduktion mit Lithiumaluminiumhydrid in Äther bei 5 - 80 l-Amino -1,2,2a,3,4,5-hexahydro-6-methylbenz[cd]indol. (Als Hydrochlorid aus Methanol/Äther, Smp. 185 - 1870). Durch Umsetzung der Aminoverbindung mit N-Benzoylisothiocyanat in siedendem Tetrahydrofuran und anschliessender Verseifung mit verd. Natriumhydroxid-Lösung unter Rückfluss während 10 Minuten gelangt man zum 1 -(1,2,2a,3,4,5-Hexahydro-6-methylbenz[cd]indol-1-yl)thio- hamstoff. (Smp. 209 - 2110, aus Methylenchlorid/Methanol).
Der Thioharnstoff wird durch Kochen während 2 Stunden mit Methyljodid in Methanol in das 1-(1,2,- 2a,3,4,5 - Hexahydro -6- methylbenz[cd]indol - 1 -yl)-2-methylisothioharnstoff-hydrojodid überführt. das aus Methanol/Äther kristallisiert. Smp. 167 - 1690.
Beispiel 19
I ,2,2a,3,4,5-Hexahydro-1-(2-hydroxyguanidino)-6 -methylbenz[cd]indol
Hergestellt unter Verwendung von 1-(1,2,2a,3,4,5 Hexahydro-6-methylbenz[cd]indol- 1 -yl)-2-methylisothio- hamstoff-hydrochlorid analog Beispiel 10. Das Hydrochlorid der im Titel genannten Verbindung weist nach Umkristallisation aus Methanol/Äther einen Smp. von 165 - 1700 auf.

Beispiel 21 1-(2-Benzylguarridino)-6-chlor-1 ,2 ,2a,3,4,5-kexa- hydrobenz[cd]indol
Hergestellt unter Verwendung von l-(6-Chlor-1,2,- 2a,3,4,5- hexahydrobenz[cd]indol -1- yl) - 2-methylisothio harnstoff-hydrojodid, Benzylamin und Äthanol als Lösungsmittel und einer Reaktionszeit von 4 Stunden analog Beispiel 3. Das Hydrogenmaleinat der im Titel genannten Verbindung kristallisiert aus Methanol/Methy lenchlorid/Äther vom Smp. 179 - 1800.
Process for the production of new guanidine compounds

According to the invention, the compounds of the formula I and their acid addition salts are obtained by mixing compounds of the formula II in which X has the above meaning and R5 is a lower alkyl radical with compounds of the formula III in which R1 and R.

The compounds of the formula I and their acid addition salts, hereinafter referred to as new substances for short, have not yet been described in the literature. In pharmacological testing on isolated organs and whole animals, they are characterized by diverse and highly pronounced effects and should therefore be used as medicinal products.
In particular, they can be used as circulatory agents, salidiuretics, anti-inflammatory drugs and psychopharmacics, e.g. from the blood pressure-increasing, bradycardic, amine-increasing and circulatory reflex-inhibiting effects on the anesthetized dog, from the lowering of the artificially elevated blood pressure in the hypertonic Grollmann rat, from the vasocontractive activity on the isolated, perfused rabbit limb, from the positively inotropic Efficacy on the isolated cat heart, from the salidiuretic effect on the rat, from the inhibitory effect on carrageenan edema in the rat and from the inhibition of motility, reserpine-antagonistic, analgesic and the sympathetically stimulating effect on the mouse.
The new compounds of the formula I or their physiologically tolerable acid addition salts can be administered as medicaments alone or in suitable pharmaceutical forms.
The new starting compounds of the formula II can be prepared by treating compounds of the formula IV, in which X has the above meaning, with N-benzoyl isothiocyanate or a mixture of ammonium thiocyanate and benzoyl chloride in an organic solvent which is inert under the reaction conditions, e.g. a cyclic ether such as tetrahydrofuran, at elevated temperature, preferably at the boiling point of the reaction mixture, to give compounds of the formula V, in which X has the above meaning, hydrolyzes the compounds of the formula V to give the compounds of the formula VI, in which X has the above meaning, e.g. by brief heating with aqueous sodium hydroxide solution, and the compounds of formula VI e.g. with alkyl halides or alkyl sulfates at elevated temperature, preferably at the boiling point of the reaction mixture, to give the compounds of the formula II.
To prepare the previously unknown compounds of the formula IV required as starting material, compounds of the formula VII in which X has the above meaning can be reduced, e.g. by means of zinc in formic acid, but preferably by means of a complex hydride of the alkali metals such as lithium aluminum hydride, sodium dihydro bis (2-methoxyethoxy) aluminate etc. in an organic solvent which is inert under the reaction conditions, e.g. a cyclic or open-chain ether such as diethyl ether, at about 5 - 80.
The compounds of the formula VII are likewise new and can be obtained in a manner known per se from the compounds of the formula VIII, in which X has the above meaning. In practice one goes e.g.
in such a way that a solution of compounds of the formula VIII in excess dilute hydrochloric acid is mixed with sodium nitrite.
The compounds of the formula VIIIa in which XI denotes halogen, the hydroxymethyl or methyl group are also new and can be prepared as follows:
1) 6-chloro-1,2,2a, 3,4,5-hexahydrobenz [cd] indole is obtained, for example, by adding 1-acetyl-1,2,2a, 3,4, - 5-hexahydrobenz [cd] indole chlorinated, e.g. by introducing chlorine into a solution of 1-acetyl-1,2,2a, 3,4,5-hexahydrobenz [cd] indole in an organic solvent which is inert under the present conditions, e.g.
a chlorinated alkane hydrocarbon such as carbon tetrachloride, at room temperature and the 1-acetyl-6-chloro-1, 2,2a, 3,4,5-hexahydrobenz [cd] indole which crystallizes spontaneously is deacetylated, e.g. by heating in a mixture of glacial acetic acid / conc. Hydrochloric acid for approx. 1 - 2 hours.
2) For the preparation of 6-bromo-1,2,2a.3,4,5-hexahydrobenztcd] indole, 1-acetyl-1,2,2a, 3,4,5-hexahydrobenz [cd] Brominate indole and deacetylate the 1-acetyl-6-bromo-1,2,2a, 3,4,5-hexahydrobenz [cdlindole thus obtained, which crystallizes out spontaneously. The bromination takes place e.g. by dripping bromine into an organic solvent which is inert under the reaction conditions, e.g. Glacial acetic acid, at about 10-150 in the presence of a catalytic amount of hydrogen bromide.
The deacetylation can be carried out as described under 1) for the preparation of 6-chloro -1, 2,2a, 3 4,5-hexahydrobenz [ed] indole, by acid hydrolysis.
3) The 1,2,2a, 3,4,5-hexahydro.6-hydroxymethylbenz- [cd] indole is e.g. prepared by nitrating 1-acetyl -1,2,2a, 3,4,5-hexahydrobenz [cd] indole, e.g. in glacial acetic acid by adding fuming nitric acid at about 100, the nitro compound formed, which crystallizes out spontaneously, is reduced to the corresponding amino compound, the amino compound obtained is diazotized, e.g. by means of sodium nitrite in sulfuric acid solution at about 0-5, the resulting diazonium salt e.g.
according to Sandmeyer to 1-acetyl-6-cyano-1,2,2a, 3,4,5-hexahydrobenz [cd] indole, the cyano compound formed into 1-acetyl-6-formyl-1,2 , 2a, 3,4,5-hexahydrobenz [cd] - indole transferred, this formyl compound reduced, eg. by means of sodium borohydride in an organic solvent which is inert under the reaction conditions, e.g. a lower alkanol such as methanol, and the obtained 1-acetyl-1,2,2a, 3,4,5-hexahydro-6-hydroxymethylbenz [cd] indole by alkaline hydrolysis, e.g. deacetylated by boiling with aqueous-methanolic sodium hydroxide solution.
The reduction of the nitro compound takes place e.g. with the aid of hydrazine hydrate in the presence of Raney nickel in an organic solvent which is inert under the reaction conditions, e.g. a lower alkanol such as methanol, at an elevated temperature, preferably 50-600, and with stirring and takes about 30 minutes.
After the reaction has ended, the catalyst is filtered off and gently evaporated to dryness. The amino compound formed can e.g. by crystallization from an organic solvent which is inert under the present conditions, e.g. a chlorinated alkane hydrocarbon such as methylene chloride.
The 1 - acetyl-6-formyl - 1,2,2a, 3,4,5 - hexahydrobenz [cd] indole can be found e.g. obtained by adding sodium hypophosphite and Raney nickel in a mixture of glacial acetic acid / pyridine / water to the corresponding cyano compound and stirring for a prolonged period at a temperature of about 10-250. For work-up, the catalyst is filtered off and the filtrate is evaporated. The residue can be worked up further by dividing it between water and an organic solvent which is immiscible therewith and which is inert under the present conditions. e.g. a chlorinated alkane hydrocarbon such as methylene chloride, shaken out and the combined organic phases evaporated. The crude formyl compound can be prepared in a manner known per se, e.g. chromatographically, be purified.
4) For the preparation of 1, 2,2a, 3,4,5-hexahydro-6-methylbenz [cdindole, e.g. Reduce the 1-acetyl-6-formyl - 1, 2.2a, 3,4.5 - hexahydrobenz [cd] indole described under 3).

5) 6-fluoro-1,2,2a, 3,4,5-hexahydrobenz [cd] indole and 1,2,2a,3,4,5-hexahydro-6-iodobenzrcd] indole can be found e.g.

If the preparation of the starting compounds is not described, they are known or can be prepared by processes known per se or analogously to the processes described here or analogously to processes known per se.
