On effec tue de préférence la réaction dans un solvant exempt d'alcool, à environ 00 C.

Cette oxydation peut être effectuée par les procédés conventionnels d'oxydation, par exemple au moyen d'acide chromi- que dans l'acide acétique glacial.
Le présent procédé permet notamment de pré parer la 9a-fluorohydrocortisone, la 9a-fluorocorti- costérone, les esters de ces composés, la 9a-fluoro- 11(3-hydroxyprogestérone et la 9a-fluôro-11(3,17a-di- hydroxyprogestérone. Comme mentionné, ces 9a- fluoro-11#-hydroxy-stéroïdes peuvent alors être oxy dés en composés 11-céto. correspondants.
Les stéroïdes du groupe pregnane ayant un subs tituant 9a-fluoro et un substituant 11-céto ou 11p hydroxy (ainsi que les esters de ceux ayant un groupe 21-hydroxy), contrairement à ce que l'on aurait prédit, possèdent une activité corticoïde à l'essai du glycogène du foie.
Cette activité est bien plus grande que celle des stéroïdes correspondants dans lesquels un autre reste halo est substitué au lieu du reste 9a-fluoro, et est de même plus grande que celle des stéroïdes corres pondants non substitués en position 9. Les 9a-fluoro- stéroïdes qui ont un groupe 21-hydroxy peuvent être obtenus directement sous forme de leurs esters d'aci des gras, spécialement d'esters d'acides alcanoïques inférieurs, ou être facilement convertis en ceux-ci.
Comme 9p, llp-oxydo-stéroïde de départ, on peut notamment utiliser la 9p,11 p-oxydo-04-pre- gnène-17a,21-diol-3,20-dione (ou 9p,llp-oxydo-17(X, 21-dihydroxyprogestérone), la 9p,11 p-oxydo-04-pre- gnène-21-ol-3,20-dione (ou 9p,llp-oxydo-21-hydr- oxy-progestérone), les esters de ces composés, la 9p, 11 p-oxydoprogestérone et la 9p,
11 p-oxydo-17a-hy- droxyprogestérone.

04-pregnène 9p,11p-oxydo-17a-ol-3,20-dione (VIII) <I>(a) 11</I> a-tosyloxy-17a-hydroxyprogestérone <I>à partir de 11</I> a-17a-dihydroxy- progestérone On dissout 1,2 g de lla,17a-dihydroxyprogesté- rone dans 12 ml de pyridine anhydre, et à la solu tion résultante, immergée dans un bain de glace, on ajoute goutte à goutte en agitant une solution de 1,4g de chlorure de p-toluène-sulfonyle dans du chloroforme exempt d'alcool.
Cette addition demande environ deux heures, après quoi on laisse le mélange de réaction à 00 pendant encore deux heures, puis éventuellement jusqu;au lendemain, à la température ordinaire. On ajoute de la glace et, après une demi- heure, la solution est concentrée à un faible volume sous vide. Le résidu résultant est traité par du chlo roforme et de l'eau et la solution dans le chloroforme est lavée avec de l'acide chlorhydrique dilué froid, avec une solution diluée de bicarbonate de sodium et finalement avec de l'eau. La solution dans le chloroforme est séchée sur du sulfate de sodium puis évaporée à siccité dans le vide.

La solution dans le chloroforme, qui contient le pro duit cherché, est séchée sur du sulfate de sodium, puis le chloroforme est éliminé dans le vide. Le ré sidu (environ 693 mg) cristallise facilement à partir d'acétone, en donnant environ 520 mg de A4,9(11)_ pregnadiène-17a-ol-3,20-dione pure, ayant les pro priétés suivantes<B>:</B> P.

<I>(c)</I> 9a-bromo-11 p,17a-dihydroxyprogestérone (IV), <I>à partir de</I> A4,9(11)=pregnadiène-17a-ol- 3,20-dione On traite 330 mg de A4,9(11)-pregnadiène-17a-ol- 3,20-dione, en suspension dans du dioxane et de l'eau, avec 200 mg de N-bromo-acétamide et 3 ml d'acide perchlorique aqueux 1 N ; le mélange est doucement agité de temps en temps jusqu'à ce qu'il soit devenu parfaitement limpide.
Après une durée totale de réaction d'une heure, on ajoute une solu tion diluée de sulfite de sodium jusqu'à ce que la solution jaune soit presque complètement décolorée. On ajoute alors du chloroforme, puis on élimine la faible phase aqueuse qui surnage. On lave la phase chloroforme-dioxane, qui contient la bromohydrine (IV), avec du bicarbonate de sodium dilué et avec de l'eau, puis on la sèche sur du sulfate de sodium. <I>(d) Préparation de</I> A4-pregnène-9(3,11(3-oxydo- 17a-ol-3,20-dione (VIII),<I>à partir de</I> 9a- bromo-11(3,17a-dihydroxyprogestérone (IV) Une solution de 247 mg de 9a-bromo-11(3,17a- dihydroxyprogestérone et 630 mg d'acétate de potas sium anhydre dans 19 ml d'alcool absolu est chauf fée à reflux pendant 90 minutes. Après addition de 2 ml d'eau, la solution est concentrée dans le vide jusqu'à cristallisation naissante.
On ajoute de nou veau de l'eau (5 ml) et la cristallisation est terminée dans un réfrigérant ; on obtient une seconde récolte par concentration de la liqueur-mère. Le produit brut obtenu est recristallisé à partir d'acétone-hexane, et donne la A4-pregnène-9(3,11p-oxydo-17a-ol-3,20- dione pure, ayant les propriétés suivantes : P.

Les exemples ci-après illustrent le procédé selon l'invention. Exemple 1 <I>(a) Acétate de</I> 9a-fluorohydrocortisone <I>(X),</I> <I>à partir de</I> 21-acétate <I>de</I> A4-pregnène- 9(3,11(3-oxydo-17a,21-diol-3,20-dione <I>(V)</I> De l'acide fluorhydrique anhydre est ajouté à une solution de 15 g de 21-acétate de A4-pregnène-9p, 11(3-oxydo-17a,21-diol-3,20-dione dans 300 ml de chloroforme (contenu dans un récipient de polyéthy lène muni d'un tube d'introduction en cuivre).
Pen dant l'adjonction, la solution est maintenue dans un bain de glace et agitée par action magnétique, jus qu'à ce que la solution prenne une couleur prédomi nante rouge. Le tube d'introduction est alors rem placé par un capuchon (de polyéthylène), et on laisse la réaction se faire, tout en agitant pendant 4 heures et demie à 00. On ajoute alors une solution aqueuse concentrée de bicarbonate de sodium jusqu'à ce que la solution soit légèrement alcaline ; il se sépare deux couches. La solution dans le chloroforme, qui est maintenant légèrement jaune, est lavée avec de l'eau ; après séchage sur du sulfate de sodium, on l'évapore à siccité dans le vide.
Le résidu (environ 17,0 g) est alors repris avec 125 ml d'acétate d'éthyle chaud, et la suspension résultante est filtrée, puis le précipité (sur le filtre) et le filtrat sont traités comme décrit ci-après. (On a obtenu occasionnellement des échantillons qui se ramollissaient à environ 205 208o et qui, resolidifiés, fondaient éventuellement à 226 - 2280, probablement du fait d'un polymor phisme). Elle a les propriétés suivantes : [a123 = -I- 1230 (C = 0,64 dans du CHC13) ; a mâ@.= 238 mu, (s = 16,800) ; INu?_ol 2,86 R., 3,01 R., 5,62 [,, 5,78 R,, 5,83 R,, 6,05 R. Analyse:
calculé pour C23H310cF (422) : C = 65,39 ; H = 7,39 ; F = 4,52. Trouvé (approximativement) : C = 65,32 ; H = 7,26 ; F = 4,50.
Par fractionnement de la liqueur-mère d'acétate d'éthyle, on obtient une nouvelle quantité d'acétate de 9a-fluoro-hydrocortisone, ce qui donne un rende- ment total de 50 %.
L'acétate de 9a-fluorohydrocortisone est environ 11 fois plus actif que l'acétate de cortisone à l'essai du glycogène de foie de rat, et environ 1 - 2 fois aussi actif que l'acétate de désoxycorticostérone pour produire la rétention du sodium chez le rat.
La substance insoluble dans l'acétate d'éthyle, restée sur le filtre (environ 1,35 g) est recristallisée dans l'acétone. Le composé pur a les propriétés sui vantes<B>:</B> P. (b) On peut obtenir la 9a-f luorohydrocor-tisone (XI) <I>à partir de l'acétate de</I> 9 -f luorohydrocorti- sone (X), en opérant comme suit A une solution de 103,8 mg d'acétate de 9a- fluorohydrocortisone dans 10 ml de méthanol absolu, on ajoute sous une atmosphère d'azote une solution de 11,4 mg de sodium dans 1,9 ml de méthanol an hydre. On laisse reposer le mélange pendant 30 mi nutes à la température ordinaire, puis on l'acidifie par addition de quelques gouttes d'acide acétique glacial.
On ajoute alors de l'eau; après avoir éliminé le méthanol dans le vide, on extrait la 9a-fluoro- hydrocortisone avec de l'acétate d'éthyle. L'extrait est lavé avec une solution de bicarbonate de sodium et avec de l'eau, puis il est séché sur du sulfate de sodium. Après évaporation du solvant, il reste un résidu cristallin (environ 92 mg), lequel, après re- cristallisation dans de l'alcool à 95 0/0, a les proprié tés suivantes: P.

La 9a-fluorohydrocortisone a approximativement la même activité biologique que son acétate.
(c) On peut obtenir l'acétate de 9a-f lciorocortisone (XVI) <I>à partir de l'acétate de</I> 9a-fluorohydro- cortïsone (X), en opérant comme suit A une solution de 31,2 mg d'acétate de 9a-fluoro- hydrocortisone dans 3 ml d'acide acétique glacial, on ajoute une solution de 10 mg d'acide chromique dans 2 ml d'acide acétique Une demi-heure après, on ajoute 1 ml de méthanol et le mélange résultant est concentré dans le vide.
Le résidu est réparti entre du chloroforme et de l'eau, et l'extrait résultant dans le chloroforme est lavé avec de l'eau, une solution de bicarbonate de sodium et de nouveau avec de l'eau. Après séchage sur du sulfate de sodium et éva poration du solvant dans le vide, le résidu (environ 25 mg) est cristallisé à partir d'alcool à 95 0/0. L'acé tate pur de 9a-fluorocortisone a les propriétés sui vantes<B>:</B> P.

En opérant comme à la section b de l'exemple 1, l'acétate de 9a-fluorocorticostérone peut être hydro lysé pour donner la 9a-fluorocorticostérone (XIII). En opérant comme décrit à la section c de l'exemple 1, l'acétate de 9a-fluorocorticostérone peut être oxydé en acétate de 9a-fluoro-11-déhydrocorticostérone (XVIII). 04-pregnene 9p, 11p-oxydo-17a-ol-3,20-dione (VIII) <I> (a) 11 </I> a-tosyloxy-17a-hydroxyprogesterone <I> from 11 </I> a-17a-dihydroxyprogesterone 1.2 g of 11a, 17a-dihydroxyprogesterone are dissolved in 12 ml of anhydrous pyridine, and to the resulting solution, immersed in an ice bath, is added dropwise with stirring with solution of 1.4g of p-toluenesulfonyl chloride in chloroform free of alcohol.
This addition takes about two hours, after which the reaction mixture is left at 00 for a further two hours, then optionally overnight at room temperature. Ice is added and after half an hour the solution is concentrated to a small volume in vacuo. The resulting residue is treated with chloroform and water and the chloroform solution is washed with cold dilute hydrochloric acid, with dilute sodium bicarbonate solution and finally with water. The chloroform solution is dried over sodium sulfate and then evaporated to dryness in vacuo.

After evaporating most of the acetic acid in vacuo, the residue is taken up in chloroform and extracted with water then with dilute sodium bicarbonate, then again with water. The chloroform solution, which contains the desired product, is dried over sodium sulfate, then the chloroform is removed in vacuo. The residue (about 693 mg) crystallizes readily from acetone, yielding about 520 mg of pure A4,9 (11) _ pregnadiene-17a-ol-3,20-dione, having the following properties <B> : </B> P.

<I> (c) </I> 9a-bromo-11 p, 17a-dihydroxyprogesterone (IV), <I> from </I> A4,9 (11) = pregnadiene-17a-ol- 3.20 -dione 330 mg of A4,9 (11) -pregnadiene-17a-ol-3,20-dione, suspended in dioxane and water, are treated with 200 mg of N-bromo-acetamide and 3 ml of 1N aqueous perchloric acid; the mixture is gently stirred from time to time until it has become perfectly clear.
After a total reaction time of one hour, a dilute sodium sulphite solution is added until the yellow solution is almost completely discolored. Chloroform is then added, then the weak aqueous phase which supernates is removed. The chloroform-dioxane phase, which contains the bromohydrin (IV), is washed with dilute sodium bicarbonate and with water, then dried over sodium sulfate. Removing the solvents in vacuum leaves an amorphous residue, which is dissolved in acetone. Crystallization occurs rapidly.
The resulting crude product (approximately 450 mg) is recrystallized from acetone-chloroform; it gives about 362 mg (85%) of pure 9α-bromo-11p, 17a-dihydroxy-progesterone, having the following properties P. The examples below illustrate the process according to the invention. Example 1 <I> (a) </I> 9a-fluorohydrocortisone acetate <I> (X), </I> <I> from </I> 21-acetate <I> of </I> A4 -pregnene- 9 (3,11 (3-oxydo-17a, 21-diol-3,20-dione <I> (V) </I> Anhydrous hydrofluoric acid is added to a solution of 15 g of 21 -A4-pregnene-9p, 11 acetate (3-oxydo-17a, 21-diol-3,20-dione in 300 ml of chloroform (contained in a polyethylene container fitted with a copper introduction tube) .
During the addition, the solution is kept in an ice bath and stirred by magnetic action, until the solution takes on a predominantly red color. The introduction tube is then replaced by a cap (of polyethylene), and the reaction is allowed to proceed, while stirring for 4 and a half hours at 00. A concentrated aqueous solution of sodium bicarbonate is then added until that the solution is slightly alkaline; it separates two layers. The chloroform solution, which is now slightly yellow, is washed with water; after drying over sodium sulfate, it is evaporated to dryness in vacuo.
The residue (about 17.0 g) is then taken up in 125 ml of hot ethyl acetate, and the resulting suspension is filtered, then the precipitate (on the filter) and the filtrate are treated as described below. The ethyl acetate filtrate deposits, on cooling, a crystalline precipitate formed of long needles mixed with small heavy prisms. This substance (about 6.2 mg) melts at about 228 2300, and is essentially pure 9α-fluorohydrocortisone acetate. For its analysis, it is recrystallized from ethyl acetate; it then melts at about 233 - 2340.
(Samples were occasionally obtained which softened at about 205208o and when resolidified eventually melted at 226-2280, probably due to polymorphism). It has the following properties: [a123 = -I-1230 (C = 0.64 in CHCl3); a ma @. = 238 mu, (s = 16.800); INu? _Ol 2.86 R., 3.01 R., 5.62 [,, 5.78 R ,, 5.83 R ,, 6.05 R. Analysis:
calcd for C23H310cF (422): C = 65.39; H = 7.39; F = 4.52. Found (approximately): C = 65.32; H = 7.26; F = 4.50.
By fractionation of the ethyl acetate mother liquor, a further quantity of 9α-fluorohydrocortisone acetate is obtained, giving a total yield of 50%.
9α-Fluorohydrocortisone acetate is approximately 11 times more active than cortisone acetate in rat liver glycogen test, and approximately 1 - 2 times as active as deoxycorticosterone acetate in producing sodium retention in rats.
The substance insoluble in ethyl acetate, which remains on the filter (approximately 1.35 g) is recrystallized from acetone. The pure compound has the following properties <B>: </B> P. F. approximately 259 - 2620, with browning; [a] 23 = -I-2620 (C = 0.53 in 95% ethanol); X ## g. = 239 mR (e = 18,000); lmu # ol = 2.94 R, 3.03 R ,, 5.75 L., 5.82 R, 6.07 [,, 6.11 R ,. Ana lysis:
calcd for C23H3006 (40-2): C = 68.63; H = 7.51. Found (approximately): C = 68.45; H, 7.17.
(b) The 9α-fluorohydrocor-tisone (XI) <I> can be obtained from the acetate of </I> 9 -f luorohydrocortisone (X), by operating as follows With a solution of 103, 8 mg of 9α-fluorohydrocortisone acetate in 10 ml of absolute methanol, a solution of 11.4 mg of sodium in 1.9 ml of anhydrous methanol is added under a nitrogen atmosphere. The mixture is left to stand for 30 minutes at room temperature, then it is acidified by adding a few drops of glacial acetic acid.
Water is then added; after removing the methanol in vacuo, the 9α-fluorohydro-cortisone is extracted with ethyl acetate. The extract is washed with sodium bicarbonate solution and with water, then it is dried over sodium sulfate. After evaporation of the solvent, a crystalline residue (about 92 mg) remains, which, after recrystallization from 95% alcohol, has the following properties: P.

9α-Fluorohydrocortisone has approximately the same biological activity as its acetate.
(c) 9a-fluorocortisone acetate (XVI) <I> can be obtained from 9a-fluorohydrocortisone acetate (X), by operating as follows A solution of 31, 2 mg of 9a-fluorohydrocortisone acetate in 3 ml of glacial acetic acid, a solution of 10 mg of chromic acid in 2 ml of acetic acid is added Half an hour later, 1 ml of methanol is added and the resulting mixture is concentrated in vacuo.
The residue is partitioned between chloroform and water, and the resulting extract in chloroform is washed with water, sodium bicarbonate solution and again with water. After drying over sodium sulfate and evaporating the solvent in vacuo, the residue (approximately 25 mg) is crystallized from 95% alcohol. Pure 9a-fluorocortisone ace tate has the following properties <B>: </B> P.

9α-Fluorocortisone acetate is approximately 10 times more active than cortisone acetate in rat liver glycogen assay.
(d) 9 -f luorocortisone (XVII), <I> from 9a-fluorocortisone acetate </I> (XVI) can be obtained by operating as follows 25 mg of 9a-fluorocortisone acetate are de-acetylated as described in section b of Example 1. Approximately 18.4 mg of the product is obtained which, by crystallization from 95% alcohol, gives approximately 14 mg of 9α-fluorocortisone having the following characteristics. following properties <B>: </B> P.

Example 2 <I> Acetate of </I> 9a-fluorocorticosterone (XII) <I> from </I> 21-acetate <I> of </I> A4-pregnene-90, ll (3-oxydo -21-ol-3,20-dione (VI): Anhydrous hydrofluoric acid is added to an ice-cold solution of 100 mg of 21-acetate A4-pregnene-9p, 11a-oxydo-21-ol -3,20-dione in 10 ml of chloroform After one hour at 0o, more chloroform is added and the solution is extracted with sodium bicarbonate solution and finally dried over sodium sulfate.
The residue (approximately 125 mg) is then taken up in hot ethyl acetate and purified as described in section a of Example 1. Pure 9α-fluorocorticosterone acetate has the following properties: Mp about 212-2140 C; <B> [] 23 </B> = -I-1840 (C = 0.11 in CHCl3); activity: 4 times that of corticosterone acetate in the liver glycogen test, and more than twice that of deoxycorticosterone acetate in the sodium retention test.
By operating as in section b of Example 1, 9a-fluorocorticosterone acetate can be hydrolysed to give 9a-fluorocorticosterone (XIII). By operating as described in section c of Example 1, 9a-fluorocorticosterone acetate can be oxidized to 9a-fluoro-11-dehydrocorticosterone acetate (XVIII). By operating as in section b of example 1,
9a-fluoro-11-dehydrocorticosterone acetate can be hydrolyzed to give 9a-fluoro-11-dehydrocorticosterone (XIX). Example 3 9a-fluoro-11 p-hydroxyprogesterone (XIV) <I> from </I> 9 (3,11 (3-oxydoprogesterone (VII) 63.2 mg of 9 (3,11 ( 3-oxydoprogesterone in 6,
3 ml of chloroform with hydrofluoric acid gas at Oo for one hour. The reaction mixture is worked up as described in section a of Example 1, and the resulting residue (approx. 61 mg) is crystallized from acetone-hexane. Pure 9α-fluoro-11 [3-hydroxyprogesterone is obtained by further crystallization from absolute ethanol; it has the following properties: P.
F. about 216-2170 C; [] p = -I-191o (C = 0.74 in chloroform); activity equal to that of cortisone acetate in the liver glycogen test, and twice that of deoxycorticosterone acetate in the sodium retention test.
By oxidation of 9α-fluoro-11 (3-hydroxyprogesterone with chromic acid as described in section c of Example 1, 9α-fluoro-11-ketoprogesterone (XX) can be obtained. Example 4 9a-fluoro-11 (3,17a-dihydroxyprogestérohe (XV) <I> from </I> A4-pregnene-9 (3,11 (1-oxydo-17a-ol-3,20-dione ( VIII) 42 mg of A4-pregnene-9 (3,11 (3-oxydo-17a-ol-3,
20-dione in 3 ml of chloroform and reacted with hydrofluoric acid gas at 0o for one hour. The reaction mixture is worked up as described in section a of Example 1, and the resulting residue (about 36 mg), after crystallization from acetone-chloroform-hexane, gives 9a-fluoro-11 (3, Pure 17a-dihydroxyprogesterone, which has the following properties <B>: </B> PF about 274 - 275o C; [a] D = -I- <B> 1360 </B> (C = 0.3 in dioxane ); activity: one third of that of cortisone acetate to the liver glycogen.
9a-fluoro-11 (3,17a-dihydroxyprogesterone can be oxidized to 9a-fluoro-11-keto-17a-hydroxyprogesterone (XXI) by the method described in section c of example 1.
