Ces composés A2-tétrazolines nouveaux, à l'état de base libre, sont de nature basique et forment avec les acides, tant organiques que minéraux, des sels d'addition bien définis. On peut citer, à titre d'exemple de ces sels d'addition les chlorhydrate, bromhydrate, sulfate, acétate, citrate, sulfamates, etc.
Conformément à l'invention, on obtient les composés de la formule ci-dessus mentionnée en condensant un 5-amino-tétrazole de formule

avec un halogénure de formule : Hal - X, formules dans lesquelles<I>Hal</I> est un halogène et des deux restes<I>Y</I> et<I>X</I> l'un est un radical alcoyle contenant 5 à-17 atomes de carbone inclus et l'autre est un radical aralcoyle de formule : Ar - Z - où Z et<I>Ar</I> ont la signification ci-dessus.

Dans ces formules, R,<I>Ar,</I> Z et<I>Hal</I> ont la signification indiquée. La condensation ci-dessus peut être effec tuée en présence d'un solvant organique inerte dans la réaction, comme le benzène, le xylène, le toluène, l'éthanol, le butanoi; l'isopropanol, etc. La réaction peut également être effectuée en présence ou en l'absence d'un catalyseur ba sique.
Si l'on fait intervenir un catalyseur basi que, la 1-alcoyl-4-ara-Icoyl-5-imino-A2-tétra- zoline est présente dans le milieu réactionnel à l'état de base libre; dans le cas contraire, elle figure dans le mélange réactionnel à l'état d'halohydrate. Le procédé peut être mis en oeuvre à l'aide d'un catalyseur organique ou minéral. Quelques exemples de matières pro pres à ce but sont la pyridine, la triéthylamine, le carbonate de potassium, le bicarbonate de sodium, la N-éthylpipéridine, etc.
La tempéra ture peut varier entre de très larges limites sui vant le mode de condensation, variant de 50 à 1600 C. En l'absence de solvant ou de cataly seur basique, on peut obtenir les résultats les meilleurs en opérant à une température voisine de 110 - 150a C, et de préférence comprise entre 120 et 1300 C.
Les produits obtenus par le procédé selon l'invention possèdent une activité germicide ex ceptionnellement élevée. Ils exercent cette ac tion même à des dilutions élevées et sont donc particulièrement intéressants comme agents sté- rilisants. De plus, ils possèdent une grande acti- vité à l'égard de<I>Trichomonas vaginales,
Tri-</I> <I>chomonas f</I> cetus et<I>Candida</I> albicans. Il est pré- férable en thérapeutique d'utiliser les sels d'ad dition avec les acides en raison de leurs pro priétés de solubilité.
Les produits préférés sont les 4-benzyl- et 4-(p - chlorobenzyl) - 5 -imino-1-n-octyl-42-té- trazolines sous forme de sels d'addition avec les halo-acides.
Les exemples suivants illustrent la présente invention. <I>Exemple 1</I> 1-n-octyl-4-benzyl-5-imino-42-tétrazoline.
(a) On mélange 15 g de 1-n-octyl-5-amino- tétrazole avec 11,4 g de chlorure de benzyle et l'on chauffe le mélange au bain d'huile à 120 - 1250 C pendant environ huit heu res. On refroidit le mélange et l'on dissout le gâteau solide dans 100 cc d'éthanol à 951%. On chauffe la solution, on l'étend à l'aide de 300 cc d'eau, on la décolore à l'aide de charbon de bois, et l'on filtre. Le produit cristallise par refroidissement et est séparé par filtration, lavé à l'aide d'éthanol étendu, puis séché à l'air.
On lave le produit brut à l'aide de benzène pour enlever le chlorure de benzyle inaltéré et on le fait recristalliser au sein d'un mélange consis tant en 170 cc d'eau, 85 cc d'éthanol à 95 % et 2,5 cc d'acide chlorhydrique con- centré de poids spécifique 1,18.
Le produit ainsi obtenu est le chlorhydrate de 1-n- octyl-4-benzyl-5-imino-A2-tétrazoline, de point de fusion de 163 - 1640 C. Ce produit a pour formule

On traite 2 g de 1-n-octyl-4-benzyl-5- imino-A2-tétrazoline (préparée, par exem ple, en dissolvant le chlorhydrate dans l'eau chaude, additionnée d'une petite quantité d'éthanol, alcalinisant la solution à l'aide d'hydroxyde de sodium aqueux à 20%, épuisant la base libre à l'aide de benzène, séchant l'extrait et enlevant le benzène par distillation sous vide)
à l'aide d'un excès d'une solution aqueuse concentrée d'acide tartrique et l'on concentre le mélange jus qu'à cristallisation. Le sel complexe hydraté est la 1-n-octyl-4-benzyl-5-imino-A2-tétra- zoline contenant deux molécules d'acide tartrique et une mol d'eau par mol de base ; isolé et recristallisé au sein d'acétate d'éthyle, il fond à 79 - 800 C.
(b) On chauffe un mélange formé de 4 g de 1-n-octyl-5-amino-tétrazole et de 4 g de bromure de benzyle à 125o C pendant six heures. Le mélange réactionnel passe à l'état de produit fondu homogène dont le bromhydrate se sépare peu à peu à l'état cristallisé. Le bromhydrate brut -de la 1-n- ootyl-4-benzyl-5-imino-A2-tétrazoline ainsi obtenu est recueilli et purifié par recristalli- sation au sein <RTI

La réaction peut être également effec tuée à l'aide d'halogénures d'aralcoyle con tenant à titre de substituants nucléaires des groupes. hydroxy, nitro, alcoyle inférieur et alcoxy inférieur, ces deux derniers termes désignant des groupes ne possédant pas plus de quatre atomes de carbone alcoyli- que. <I>Exemple 2</I> 1-n-octyl-4-p-chlorobenzyl-5-imino-A2- tétrazoline.
(a) On chauffe à 120 - 125,, C pendant huit à dix heures un mélange consistant en 15 g de 1-n-octyl-5-amino-tétrazole et 15 g de chlorure de p-chlorobenzyle. On refroidit le mélange réactionnel, on le reprend à l'aide d'un minimum d'éthanol chaud et on étend la solution à 400 cc à l'aide d'eau. On distille le mélange réactionnel pour chasser l'éthanol et le chlorure de p-chlorobenzyle inaltéré.
On alcalinise fortement le résidu aqueux au tournesol à l'aide .d'une solution d'hydroxyde de sodium à 25 '0 /0 et on l'épuise à l'aide de benzène. On concentre les extraits benzéniques sous vide et l'on reprend 1e résidu au moyen de 75 cc d'éthanol à 95 %. On traite la solution à l'ébullition, puis on l'étend au moyen de 75 cc d'eau.
Le chlorhydrate brut de 1-n- octyl-4-p -chlorobenzyl-5-imino-A-'-tétrazo- line qui se sépare par refroidissement est purifié par recristallisation au sein. d'éthanol, aqueux. Point de fusion 165,5 - 166,5o C. (b) On chauffe le mélange de 15,8 g de 1-n- octyl-5-aminotétrazole et de 14,1 g de chlorure de p-chlorobenzyle à 1250 C pen dant quatre heures.
On refroidit le mélange réactionnel, on le reprend à l'aide de 75 cc d'éthanol à 95'%, on l'étend à 500 cc au moyen d'eau et on le distille de manière à chasser l'éthanol et le chlorure de p-chloro- benzyle inaltéré. On alcalinise le résidu aqueux par addition d'une solution de 15 g d'hydroxyde de sodium dans 50 cc d'eau et l'on épuise successivement au moyen de quatre portions (1 de l00 et 3 de 50 cc) de benzène. On sèche le mélange des ex traits benzéniques sur du carbonate de po tassium anhydre, on filtre et l'on concentre par distillation sous vide.
Le produit rési duel ainsi obtenu est la 1-n-octyl-4-p-chlo- robenzyl-5-iminotétrazoline. On peut, si on le veut, purifier le produit par recristalli sation au sein du n-hexane. Point de fusion 52 - 53o C.
<I>Exemple 3</I> 1-n-octyl-4-m-xylyl-5-imino-A2-tétrazoline. On chauffe un mélange de 8 g de 1-n-octyl- 5-amino-tétrazole et de 8,9 g de m-bromure de xylyle à 125 - 1300 C pendant six heures envi ron. Au cours de la période<B>dé</B> chauffage, les réactifs fondent ensemble sous forme d'un pro duit limpide et homogène duquel le brom- hydrate de 1-n-octyl-4-xylyl-5-imino-A2-tétra- zoline cristallise progressivement.
On reprend le bromhydrate chaud au moyen d'éthanol chaud, on étend d'eau la solution et on la sou met à l'entraînement par la vapeur d'eau. On alcalinise le résidu par addition d'une solution d'hydroxyde de sodium et l'on épuise au moyen de benzène. On chasse le benzène de l'extrait par distillation et l'on reprend le résidu à l'aide d'isopropanol, puis on le convertit en chlor- hydrate par addition d'un léger excès d'acide chlorhydrique concentré.
On recueille le chlor- hydrate brut qui se sépare par dilution de la solution à l'aide d'un égal volume d'eau, on le lave au moyen de benzène et on le fait recris- talliser au sein d'éthanol à 40 % contenant une petite quantité d'acide chlorhydrique. Le pro duit obtenu est le chlorhydrate de 1-n-octyl-4- m-xylyl-5-imino-A2-tétrazoline, de point de fu sion de 163 - 164,1 C.

<I>Exemple 4</I> 1-n-octyl-4-o-xylyl-5-imino-A2-tétrazoline.
(a) On chauffe à 1250 C pendant six heures un mélange de 8 g de 1-n-octyl-5-amino- tétrazole et 8,9 g de bromure d'o-xylyle. On reprend le bromhydrate brut qui se sé pare du mélange réactionnel au moyen d'éthanol et l'on étend d'eau la solution. On soumet ce mélange à l'entraînement par la vapeur d'eau de manière à éliminer le bro mure d'o-xylyle et l'éthanol.
On alcalinise la solution résiduelle par addition d'une solution d'hydroxyde de sodium à 20 % et l'on épuise au moyen de benzène. On sèche les extraits benzéniques et l'on chasse le benzène par distillation. On reprend le résidu par de l'isopropanol contenant un léger excès d'acide chlorhydrique concen tré.
On étend la solution, on sépare par fil tration le précipité et on le fait recristalliser au sein d'éthanol à 30'% acidifié à l'aide d'acide chlorhydrique. Le produit, qui est le chlorhydrate de 1-n-octyl-4-o-xylyl-5- imino-A2-tétrazoline, fond à 161 - 162,1 C.
(b) On dissout 5 g de chlorhydrate de 1-n octyl - 4 - o-xylyl-5-imino-A2-tétrazoline dans de l'eau chaude contenant une petite quan tité d'éthanol, et l'on alcalinise la solution ainsi obtenue au moyen d'une solution à 20 % d'hydroxyde de sodium. On épuise la base libre de la 1-n-octyl-4-o-xylyl-5- imino-A2-tétrazoline au moyen de benzène, on sèche l'extrait et l'on chasse le benzène par distillation sous vide.
On reprend le résidu au moyen d'alcool anhydre et on le traite à l'aide d'une solution aqueuse con tenant de l'acide tartrique. On concentre la solution à cristallisation et l'on recueille le sel complexe désiré de 1-n-octyl-4-o-xylyl- 5-imino-A2-tétrazoline contenant deux mo lécules d'acide tartrique et une mol d'eau par mol de base. <I>Exemple 5</I> 1-n-undécyl-4-p-chlorobenzyl-5-imino-A2- tétrazoline. On chauffe à 120-1250 C pendant 12 heures un mélange formé de 12 g de 1-n-undécyl-5- aminotétrazole et de 10 g de chlorure de p chlorobenzyle.
Au cours de la période de chauffage, le chlorhydrate brut de la 1-n- undécyl - 4-p -chlorobenzyl- 5-imino-A2-tétrazo- line se sépare de la solution. On recueille le chlorhydrate brut, on le dissout dans l'éthanol, on l'étend d'eau et on le soumet à l'entraine- ment par la vapeur d'eau. On alcalinise forte ment le résidu à l'aide d'une solution d'hydr oxyde de sodium à 25 0/0, on le refroidit, on l'épuise à l'aide de benzène et l'on chasse le benzène par distillation.
On obtient la 1-n undécyl-4-p-chlorobenzyl-5-imino-A2-tétrazo- line. On reprend la base libre au moyen de 50 cc d'isopropanol, et l'on traite à l'aide de 10 cc d'acide chlorhydrique concentré. On étend la solution au moyen de 40 cc d'eau, ce qui provoque la précipitation du chlorhydrate. On recueille le chlorhydrate et on le - fait re- cristalliser au sein d'éthanol à 40,% acidifié à l'aide d'acide chlorhydrique.
Le produit obtenu est le chlorhydrate de 1-n-undécyl-4-p-chloro- benzyl-5-imino-A2-tétrazoline, de point de fu sion de 145 - 1460 C. <I>Exemple 6</I> 1-n-octyl-4-benzyl-5-imino-A2-tétrazoline. On chauffe à<B>1350</B> C pendant huit heures un mélange formé . de 8,75 g de 1-benzyl-5- aminotétrazole et de 10,5 g de bromure de n- octyle. On refroidit le mélange réactionnel, on le reprend au moyen de 75 cc d'éthanol à 95 0/0, on étend d'eau à environ 500 cc et l'on soumet à l'entraînement à la vapeur d'eau.
On alcalinise le résidu par addition d'une solution d'hydroxyde de sodium et l'on épuise à l'aide de trois portions successives de benzène. On sèche le mélange des extraits, on chasse le sol vant par évaporation et l'on reprend le résidu à l'aide d'isopropanol, la conversion en -chlor hydrate étant effectuée par l'addition d'un léger excès d'acide chlorhydrique concentré.
Le chlorhydrate brut qui se sépare par dilution de la solution à l'aide de 150 cc d'eau est recueilli, lavé à l'aide de benzène et recristallisé au sein d'éthanol contenant une petite quantité d'acide chlorhydrique. Le produit obtenu est le chlor- hydrate de 1-n-octyl-4-benzyl-5-imino-A2-tétra- zoline identique au produit de l'exemple 1. <I>Exemple 7</I> 1-n-octyl-4-p-chlorobenzyl-5-imino-A2- tétrazoline.
On chauffe à<B>1250</B> C pendant environ vingt heures un mélange formé de 10 g de 1-p-chloro- benzyl-5-aminotétrazole et de 12 g de bro= mure de n-octyle. On refroidit le mélange ré actionnel et on le traite conformément à l'exemple 6. Le produit formé est le chlor hydrate de 1-n-octyl-4-p-chlorobenzyl-5-imino- A2-tétrazoline identique à celui préparé dans l'exemple 2. <I>Exemple 8</I> 1-n-octyl-4-(bêta-phényléthyl)-5-imino-A2- tétrazoline.
(a) On chauffe à 125 - 130o C pendant sept heures un mélange formé de 9,9 g de 1-n- octyl-5-aminotétrazole et de 10,2 g de bro- mure de bêta-phényléthyle. On refroidit le mélange réactionnel, on le reprend au moyen de 50 cc d'éthanol à 95 %,
on étend à l'aide de 500 cc d'eau et l'on dis tille pour chasser l'éthanol et l'excès de bromure de bêta-phényl-éthyle. On alcali nise le résidu par addition d'une solution d'hydroxyde de sodium à 20,1% et l'on épuise au moyen de benzène. On sèche les extraits benzéniques sur du carbonate de potassium anhydre et l'on chasse le ben zène par distillation sous vide.
Le produit résiduel est la 1-n-octyl-4-(bêta-ph6nyl- éthyl)-5-imino-42-tétrazoline. (b) On reprend le produit (a) au moyen de 30 cc d'isopropanol, on mélange avec 10 cc d'acide chlorhydrique concentré, on chauffe à l'ébullition et l'on étend à l'aide de 60 cc d'eau. On recueille le chlorhydrate qui se sépare par refroidissement, on le fait recristalliser une première fois au sein de benzène, puis au sein d'eau, et l'on sè che à<B>1000</B> C.
Le produit est le chlor- hydrate de 1-n-octyl-4-(bêta-ph6nyléthyl)- 5-imino-A2-tétrazoline, de point de fusion de 207 - 208q C. <I>Exemple 9</I> 1-n-octyl-4-(gamma-phénylpropyl)-5- imino-A2-tétrazoline.
(a) On chauffe à 125 -<B>1300</B> C pendant sept heures un mélange de 9,9 g de 1-n-octyl-5- aminotétrazole et de 11 g de bromure de gamma-phénylpropyle. On refroidit le mé lange réactionnel, on le reprend à l'aide de 50 ce, d'éthanol à 95,%, on étend au moyen de 500 cc d'eau,
et l'on distille de manière à éliminer l'éthanol et l'excès de bromure de gamma-phénylpropyle. On al calinise le résidu par addition \d'une solu- tion d'hydroxyde de sodium à 20%, on refroidit et l'on épuise à l'aide de benzène. On sèche les extraits benzéniques sur du carbonate de potassium anhydre et l'on chasse le benzène par distillation sous vide.
Le produit résiduel est la 1-n-octyl-4- (gamma -phénylpropyl) - 5 -imino-A2-tétrazo- line.
(b) On reprend le produit (a) au moyen de 30 cc d'isopropanol, on mélange avec 10 cc d'acide chlorhydrique concentré, on chauffe à l'ébullition et l'on étend au moyen de 60 cc d'eau. On recueille le chlorhydrate qui se sépare, on le fait re- cristalliser d'abord au sein de benzène, puis au sein d'eau, et l'on sèche à 100 C. Le produit obtenu est le chlorhydrate de 1-n- octyl - 4 - (gamma -phénylpropyl)-5-imino-42- tétrazoline, de point de fusion de 153 154q C.
<I>Exemple 10</I> 1-n-amyl-4-p-chlarobenzyl-5-imino-42- tétrazoline.
(a) On chauffe à<B>1250</B> C pendant six heures un mélange de 12,4 g de 1-n-amyl-5- aminotétrazole et de 14,1 g de chlorure de p-chlorobenzyle. On refroidit le mélange réactionnel, on le reprend à l'aide d'étha nol à 951)/û, on étend au moyen de 500 cc d'eau,
et l'on distille de manière à chasser l'éthanol et l'excès de chlorure de p-chloro- benzyle. On alcalinise le résidu par addi- tion d'une solution à 20 % d'hydroxyde de sodium, on refroidit, et l'on épuise au moyen de benzène. On sèche les extraits benzéniques sur du carbonate de potas sium anhydre et l'on chasse le benzène par distillation sous vide.
Le produit résiduel est la 1-n-amyl-4-p-chlorobenzyl-5-imino- 42-tétrazoline. (b) On reprend le produit de (a) au moyen d'isopropanol additionné de 10 cc d'acide chlorhydrique concentré, on chauffe au point d'ébullition, et l'on étend au moyen de 60 cc d'eau. On recueille le chlorhydrate qui> - se sépare, on le fait recristalliser d'abord au sein de benzène, puis au sein d'eau, et l'on sèche à<B>1000</B> C.
Le produit formé est le chlorhydrate de 1-n-amyl-4-p- chlorobenzyl-5-imino-42-tétrazoline, de point de fusion de 151,5 - 152,5q C. <I>Exemple 11</I> 1-n-hexyl-4-(bêta-phényléthyl)-5-imino- A2-tétrazoline.
(a) On chauffe à 125 - 1301, C pendant sept heures un mélange de 8,4 g de 1-n-hexyl-5- aminotétrazole et de 11,1 g de bromure de bêta-phényléthyle. On refroidit le mélange réactionnel, on le reprend au moyen de 50 cc d'éthanol à 95 %, on étend à l'aide de 500 cc d'eau,
et l'on distille de manière à chasser l'éthanol et l'excès de bromure de bêta-phényléthyle. On alcalinise le ré sidu par addition d'une solution à 20 0/0 d'hydroxyde de sodium et l'on épuise à l'aide de benzène. On sèche les extraits benzéniques sur du carbonate de potassium anhydre et l'on chasse le benzène par distil lation sous vide. Le produit résiduel est la 1-n-hexyl-4-(bêta-ph6nyléthyl)-5-imino- 42-tétrazoline. (b) On reprend le produit (a) par de l'isopro- panol, on le mélange avec 10 cc d'acide chlorhydrique concentré, - on chauffe à l'ébullition et l'on étend à l'aide de 60 cc d'eau.
On recueille le chlorhydrate qui se sépare par refroidissement, on le fait re- cristalliser d'abord dans le benzène, puis au sein d'eau et on le sèche à 1001, C. Le pro duit obtenu est le chlorhydrate de 1-n- hexyl -4-(bêta-phényléthyl)-5-imino-42-tétra- zoline, de point de fusion de 229 - 2301, C. <I>Exemple 12</I> 1-n-hexyl-4-(gamma-phénylpropyl)-5- imino-02-tétrazoline.
(a) On chauffe un mélange de 5,1 g de 1-n hexyl-5-amina-t6trazole et de 7,2 g de bro mure de gamma-phénylpropyle à 125 130o C pendant sept heures: On refroidit le .mélange réactionnel, on le reprend au moyen d'éthanol à 95,%, on étend à l'aide de 500 cc d'eau et l'on distille de manière à chasser l'éthanol et l'excès de bromure de gamma-phénylpropyle. On alcalinise le résidu par addition de solution d'hydr oxyde de sodium à 20 0/0, on refroidit et l'on épuise à l'aide de benzène.
On sèche les extraits benzéniques et l'on chasse le benzène par distillation sous vide ; il reste la 1-n-hexyl-4-(gamma-phénylpropyl)-5- imino-42-tétrazoline. (b) On reprend le produit (a) à l'aide d'iso- propanol additionné d'un excès d'acide chlorhydrique concentré, on chauffe à l'ébullition et l'on étend au moyen de 60 cc d'eau. On recueille le chlorhydrate qui se sépare, on le fait recristalliser d'abord au sein d'un mélange 2/1 de benzène-cyclo- hexane, puis au sein d'eau, et l'on sèche à 1000 C.
Le produit formé est le chlor hydrate de 1-n-hexyl-4-(gamma-phényl- propyl)-5-imino-42-tétrazoline, de point de fusion de 171 - 1720 C. <I>Exemple 13</I> 1-n-amyl-4-(delta-phénylbutyl)-5-imino- 42-tétrazoline.
On chauffe à 125 - 130o C pendant sept heures un mélange de 12,4 g de 1-n-amyl-5- aminotétrazole et de 18,7 g de bromure de delta-phénylbutyle. On refroidit le mélange ré actionnel, on le reprend à l'aide d'une quantité minimum d'éthanol à 95 0./o,
on étend au moyen de 500 cc d'eau et l'on distille pour chasser l'éthanol et l'excès de bromure de delta-phényl- butyle. On alcalinise le résidu par addition d'une solution à 20 % d'hydroxyde de sodium, on refroidit et l'on épuise à l'aide de benzène. On concentre les extraits benzéniques sous vide et- l'on reprend le résidu au moyen d'étha- nol à 95 0/0.
On traite la solution à l'aide de 15 cc d'acide chlorhydrique concentré, on chauffe au point d'ébullition et l'on étend à l'aide de 60 cc d'eau. On recueille le chlor- hydrate qui se sépare, _ on le fait recristalliser au sein de benzène et au sein d'eau, puis on sèche.
Le produit obtenu est le chlorhydÊate de 1-n-amyl-4-(delta-phénylbutyl)-5-imino-02- tétrazoline. <I>Exemple 14</I> En procédant dans l'ensemble comme décrit dans les exemples ci-dessus, on peut encore préparer les dérivés substitués suivants de la 5-imino-à2-tétrazoline. Pour chacun des dérivés ci- dessous, le radical alcoyle est un radical alcoyle à chaîne droite.
