La présente invention a pour objet un procédé de préparation de l'acide N- (2, 3-diméthyl-phényl)-anthrani lique et de ses sels par hydrolyse d'un composé hétéro- cyclique de formule

cal hydrocarboné non substitué ou substitué, par exem-
ple un radical alcoyle, phényle ou phénylalcoyle, substi
tué ou non.
Le procédé selon l'invention est caractérisé en ce
que l'on hydrolyse un composé hétérocyclique correspon-
dant à l'une des formules ci-dessus. Dans la plupart des
cas, il est préférable d'effectuer l'hydrolyse au moyen
d'un hydroxyde de métal alcalin, notamment d'hy
droxyde de sodium ou d'hydroxyde de potassium, bien
que d'autres bases fortes ou modérément fortes, telles
que les hydroxydes de métaux alcalino-terreux ou par
fois l'ammoniac ou une amine, soient utilisables. Dans
certains cas, notamment pour l'hydrolyse d'un anhydride
d'acide, un acide fort tel que l'acide chlorhydrique peut
également tre utilisé.
Il est préférable d'employer un
excès de l'agent hydrolytique et de mettre en oeuvre le
procédé dans un solvant hydroxylé tel qu'un alcanol
inférieur, un glycol,, de l'eau ou un mélange de ces substances, avec ou sans un co, solvant non hydroxylé tel qu'un éther de glycol. La température nécessaire pour la réaction dépend dans une certaine mesure des réactifs choisis, mais en général la réaction s'effectue à vitesse satisfaisante à une température de 80 à 150 C. Dans le cas de l'hydrolyse d'un anhydride d'acide, la réaction peut tre effectuée à une température d'environ 25 à 1500 C. Dans les conditions indiquées, l'hydrolyse est pratiquement achevée en moins de 24 h.
Le produit de réaction peut tre isolé directement ou après transformation en acide. libre ou en un sel.
L'acide N- (2,3-diméthyl-phényl)-anthranilique peut tre transformé en divers sels par réaction avec des bases organiques ou inorganiques, telles que l'hydroxyde de sodium, l'hydroxyde de potassium, l'hydroxyde de calcium, le carbonate, de, sodium, la 2-hydroxy-éthyl- amine, la choline, l'ammoniac et la diéthylamine. Les sels peuvent tre transformés en acide libre par traitement avec un acide fort, par exemple l'acide chlorhydrique en solution aqueuse.
L'acide N- (2, 3-diméthyl-phényl)-anthranilique et ses sels pharmaceutiquement acceptables sont utiles comme analgésiques et pour combattre les symptômes associés aux états rhumatismaux, arthritiques et autres états in flammatoires. Ces substances sont efficaces par voie orale.
Exemple 1
On chauffe à reflux pendant 10h une solution de 6,9 g de 1, 3-di (2,3-diméthyl-phényl)-2, 4-(lH, 3H) quinazoline-dione dans 100 ml d'éthanol contenant 37 g d'une solution aqueuse d'hydroxyde de sodium à 50 /o. On chasse l'éthanol sous vide, on dilue le concentré avec 100 ml d'eau et on ajoute peu à peu la solution ainsi obtenue à 140 ml d'acide chlorhydrique 6 N, en agitant.
On recue : lle l'acide N-(2, 3-diméthylkphényl)-anthranili- que qui a précipité, on le lave à l'eau et on le purifie par recristallisation dans Idu méthanol aqueux ; p. f. 2292300 C. On peut récupérer du chlorhydrate de 2, 3-diméthyl-aniline dans le filtrat acide, comme sousproduit.
La 1,3-di (2, 3-diméthyl-phényl)-2, 4-(lH, 3H) quinazo line-dione utilisée comme matière de départ peut tre préparée comme suit. On prépare du 2,3-diméthyl-phénate de sodium en ajoutant progressivement 24 g de 2,3 diméthyl-phénol à une suspension de 9,6 d'hydrure de sodium à 55 /o dans 90 ml d'éther diméthylique du di éthylèneglycol sec dans un bain de glace. On chauffe le mélange à 500 C jusqu'à cessation du dégagement d'hy- drogène, puis on ajoute 18,0 g de 2, 4-dichloro-quinazo- line en une fois.
Après la réaction exothermique initiale, on chauffe le mélange à reflux pendant 5 h. On refroidit le mélange, on le dilue avec de la glace et de l'eau et on 1'extrait à l'éther. On lave l'extrait éthéré avec 2,51 d'eau en six portions et avec une solution de chlorure de sodium saturée, on le sèche et on le concentre. On triture le produit qui s'est séparé avec de l'éther de pé- trole, et ce produit, qui est la 2,4-di (2,3-dimthyl-plie noxy)-quinazoline, peut tre encore purifié par cristallisation dans de l'éthanol aqueux ; p. f. 177-178 C.
On chauffe 8, 9g de ce produit sous atmosphère d'azote à 320-3330C pendant 3 h pour le transformer en 1, 3-di (2,3-diméthyl-phényl)-2,4-(lH, 3H)-quinazoline-dione, utilisable sans autre purification.
Exemple 2
On chauffe à reflux pendant 24 h une solution de 1, 0 g de 1-(2, 3-diméthyl-phényl)-2, 4-(lH, 3H) quinazoline- dione dans 20 ml d'éthylène-glycol contenant 4,0 g d'hydroxyde de potassium à 85 /0 et 2,8 ml d'eau, on étend la solution à 120 ml avec de l'eau et on l'acidule avec de l'acide chlorhydrique dilué. On recueille l'acide N (2, 3-diméthyl-phényl)-anthranl'ique insoluble ; p. f. 2292300 C après cristallisation dans de l'éthanol aqueux.
La matière de départ peut tre préparée comme suit.
On dilue avec 15ml d'hydroxyde de sodium 2N une solution de 10g de 2,4-di (2,3-diméthyl-phénoxy)-quinazoline dans 30 ml d'éther diméthylique du diéthylèneglycol et on la chauffe brièvement jusqu'à 500 C. On ajoute de la glace, puis de l'eau, on lave la solution alcaline à l'éther et on l'acidule avec de l'acide chlor- hydrique dilué. On recueille la 2- (2, 3-diméthyl-phénoxy)- 4- (3H)-quinazoEnone insoluble et on la cristallise dans de l'éthanol ; p. f. 272-273o C.
On chauffe un mélange de 1 g de ce produit et de 20 ml d'huile lourde minérale pendant 2 h à 328-3350 C, sous azote et en agitant. Après refroidissement, on dilue le mélange avec de l'éther de pétrole et on recueillle la 1-(2, 3-diméthyl-phényl)-2, 4 (lH, 3H) quinazoline-dione insoluble ; p. f. 2700 C après cristallisation dans de l'éthanol.
Exemple 3
On chauffe 10g de 4-, phénoxy-2-(2, 3-diméthyl-phé- noxy)-quinazoline à 320-330 C pendant 3 h, sous azote et en agitant. On refroidit la masse fondue, contenant de la 3-phényl-1-(2, 3-diméthyl-phényl)-2, 4-(lH, 3H) quinazo line-dione, et on la verse dans 150 ml d'éthanol, en utilisant une partie de l'éthanol pour rincer le récipient de réaction. On ajoute 55g d'une solution aqueuse d'hy- droxyde de sodium à 50"/e et on chauffe le mélange à reflux pendant 24 h, on le concentre à un faible volume, on le dilue avec 250ml d'eau et on l'acidulé avec de l'acide chlorhydrique 6 N.
On recueille l'acide N- (2, 3 diméthyl-phényl)-anthranilique qui s'est séparé et on le cristallise dans de l'éthanol aqueux ; p. f. 229-2300 C.
La matière de départ peut tre préparée comme suit.
On ajoute 15 g Ide 2-chloro-4-phénoxy-quinazoline à une solution préparée à partir de 7,3 g de 2,3-diméthyl-phé noi et 2,7 g d'hydrure de sodium à 55 O/o dans 50 ml d'éther diméthylique du diéthylèneglycol. On chauffe le mélange à reflux pendant 5 h, on le dilue avec de la glace et de l'eau et on l'extrait à l'éther. On lave l'ex trait éthéré à l'eau, on le sèche, on l'évapore et on triture le résidu avec de l'éther de pétrole. On obtient ainsi de la 4-phénoxy-2-(2, 3-d. méthyl-phénoxy)-quinazoline, utilisable sans autre purification.
Exemple 4
On chauffe 2,2 g de 3-méthyl-2-(2, 3-diméthyl-phé- noxy)-4- (3H) qumazolinone dans 5 ml d'huile minérale lourde à 320-3350 C pendant 3 h, sous azote et en agitant.
On verse la solution huileuse, contenant de la 3 méthyl-1- (2,3-diméthyl-phényl)-2, 4-(lH, 3H) quinazoline- dione, dans une solution de 38 g d'hydroxyde de sodium aqueux à 50 /o dan, s 100 ml d'éthanol et on chauffe le mélange à reflux pendant 24 h. Onchassel'éthanol sous vide, on d. lue le résidu à l'eau, on le lave à l'éther de pétrole et l'acidule avec de l'acide chlorhydrique dilué.
On recueille l'acide N-(2, 3-diméthyl-phényl)-anthranili- que insoluble et on le cristallise-dans de l'éthanol aqueux ; p. f. 229-2300 C.
La matière de départ peut etre préparée comme suit.
On traite une solution de 3,0g de 2- (2, 3-diméthyl-phé noxy)-4- (3H) quinazolinone dans 50 ml d'hydroxyde de sodium 1 N avec 5 ml de sulfate de diméthyle et on l'agite à la température ordinaire pendant 24 h. On recueille la 3-méthyl-2- (2, 3-diméthyl-phénoxy)-4- (3H) quinazolino- ne, on la lave à l'eau et on la sèche.
Exemple 5
On chauffe à reflux pendant 24 h un mélange de 1 g de 5,11-bis (2,3-diméthyl-phényl)-dibenzo [b, f] [1, 5] diazo- cine-6, 12-; dione, 1 g d'hydroxyde de potassium et 10 ml d'éthylèneglycol. On refroidit le mélange, on le dilue à l'eau, on le filtre, on acidule le filtrat avec de l'acide chlorhydrique dilué et on lechauffe au bain-marie pen- dant 5 mn. On recueille l'acide N- (2, 3-diméthyl-phényl)- anthranilique insoluble ; p. f. 9-230O C après cristallisation dans de l'éthanol.
La matière de départ peut tre préparée comme suit.
On agite une suspension de 0,9 g d'hydrure de sodium à 54"/e (dispersé dans de l'huile minérale) dans 10 ml d'éther bis (2-méthoxy-éthylique) en refroidissant dans un bain de glace. On ajoute 2,5g de 2,3-diméthyl-phéno ! l par petites portions et on agite le mélange pendant plusieurs minutes à la température ordinaire, puis dans un bain d'eau chaude jusqu'à dissolution complète et cessation du dégagement d'hydrogène. On refroidit la solu- tion à la température ordinaire, on ajoute 2,75 g de 6,12dichloro-dibenzo [b, f] [1, idiazocine par petites portions, on agite le mélange pendant 1 h, puis on le chauffe au bain-marie pendant 17 h.
On verse la solution dans 200 ml d'eau et on extrait le mélange au benzène. On lave l'extrait benzénique à l'eau, on le sèche, on l'éva- pore et on triture le résidu avec de l'éther de pétrole.
On recueille le produit soLde qui est la 6,12-bis (2,3-di móthyl-phénoxy)-dibenzo [b, f] [1, 5] diazocine ; p. f. 210 2150 C. On transpose ce produit en 5,11-bis (2,3-dimé thyl-phényl)-dibenzo [b, f] [1, 5] diazocine-6, 12-dioneen le chauffant à 250-2900 C pendant 2 1/2 h sous azote.
Exemple 6
On chauffe à 50-70O C un mélange de 5,0 g de N- (2, 3 diméthyl-phényl)-isatine dans 50 ml d'acide acétique et on le maintient à cette température tout en lui ajoutant 1,33 g d'anhydride chromique par portions. On recueille l'anhydride N- (2, 3-diméthyl-phényl)-isatoïque qui s'est formé et on dilue le filtrat à l'eau pour obtenir une nouvelle quantité du mme produit ; p. f. 197-198 C après cristallisation dans de l'éthanol. On chauffe à reflux le produit brut dans 50 ml d'ethanol avec 10 g d'hydroxyde de so ; d. um à 50 /o pendant 1 h.
On concentre le mélange à un faible volume, on le dilue à l'eau et on l'acidule avec de l'acide chlorhydrique dilué. On recueille l'acide N- (2, 3-diméthyl-phényl)-anthraniiique qui s'est séparé, p. f. 229-230 C après cristallisation dans de l'éthanol. On obtient le mme produit en hydrolysant 1'anhydride N (2,3-diméthyl-phenyl)-isatoique dans un mélange bouillant à reflux de 25ml d'éther diméthylique du diéthylèneglycol et 10 ml d'acide chlorhydrique 6 N.
La matière de départ peut tre préparée comme suit.
On ajoute peu à peu 18 g, de 2,3-diméthyl-! phénol à une suspension refroidie et agitée de 6,9 g d'hydrure de so dium à 54 I/o dans 75 ml d'éther diméthylique du diéthy- lèneglycol. On chauffe le mélange à 500 C jusqu'à cessation du dégagement d'hydrogène, puis on ajoute 25 g de 2-chloro-4-méthyl-quinoléine en une fois.
On chauffe le mélange à reflux pendant 2 h, on le laisse reposer à la température ordinaire jusqu'au lendemain, on le dilue avec de la glace et de l'eau et on l'extrait à l'éther. On lave l'extrait éthéré avec plusieurs portions d'eau, on le sèche et on l'évapore, obtenant ainsi deula 2-^ (2, 3-dimé- thyl-phénoxy)-4-méthyl-quinoléine ; p. f. 123-124 C après cristallisation dans de l'éthanol aqueux. On chauffe 47,8 g de ce produit à 320-3300 C pendant 3 h sous azote, on refroidit partiellement la masse fondue, on la dilue avec 300 ml de cyclohexane et on la chauffe à reflux en agitant.
On recueille le N- (2, 3-diméthyl-phényl)-4-mé- thyl-carbostyriil insoluble ; p. f. 194-195 C après cristalli- sation dans de l'éthanol aqueux. On dissout 2g de ce produit dans 25 ml de pyridine en chauffant et on dilue la solution successivement avec 10 ml d'hydroxyde de sodium 2 N et 15 ml d'eau. On ajoute 5,6 g de permanganate de potassium en l'espace de 2 h, tout en maintenant la température à 35-40O C en refroidissant. On refroidit lentement le mélange jusqu'à la température ordinaire et, 18 h plus tard, on le rend légèrement plus basique et on le filtre.
On lave le filtrat deux fois à l'éther, on l'acidule fortement avec de l'acide chlorhydrique et on le laisse reposer jusqu'au lendemain. On recueille la N (2,3-diméthyl-phényl)-isatine ; p. f. 188 C après cristalli- sation dans de 1'éthanol.
Exemple 7
On traite une solution de 1 g de N-(2, 3-diméthyl-phé- nyl)-1, 2-dihydro-4H-3,1-benzoxazin-4-one dans 2 ml d'éthanol absolu avec 10 ml d'ammoniaque concentrée et on chauffe un peu au-dessous du point d'ébullition pendant 30 mn ou jusqu'à obtention d'une solution limpide.
On acidulé la solution avec de l'acide chlorhydrique di lué, on la refroidit et on recueille l'acide N- (2, 3-dimé- thyl-phényl)-anthranilique insoluble ; p. f. 229-2300C après cristallisation dans de l'éthanol aqueux.
Le sel de sodium de l'acide N-(2, 3-ldiméthzrl-phényl)- anthranilique peut tre préparé en dissolvant l'acide N (2,3-dilméthyl-phényl)-anthranilique dans de l'étnanol, en ajoutant un équivalent d'hydroxyde de sodium aqueux ou alcoolique et en évaporant le mélange sous vide. Les sels de potassium, de calcium, d'ammonium et de 2-hydroxy-éthylamine peuvent tre préparés de manière semblable à partir de carbonate de potassium, d'hydroxyde de calcium, d'ammoniac et de 2-hydroxy-éthylamine.
La matière de départ peut tre préparée comme suit.
On ajoute 5,5 g de 2-bromo-2', 3'-diméthyl-diphényl- amine dans 30 ml d'éther anhydre à un mélange conte nant du magnésium et du bromure d'óthylmagnélsium (préparé à partir de 3,03 g de bromure d'éthyle, 2,53 g de tournure de magnésium et 100 ml d'éther anhydre) et on chauffe le mélange à reflux en agitant et sous atmo- sphère d'azote jusqu'au lendemain.
On chasse la majeure partie de l'éther par distNIation du mélange contenant du magnésium et le sel N-bromomagnésien Ide la 2-bromo2', 3'-diméthyl-di, phénylamine. On ajoute 80 ml de tétrahydrofuranne anhydre, on chauffe le mélange à reflux pendant 16 h et on le refroidit, obtenant ainsi une solu- tion contenant le sel N-bromomagnésien, du bromure de 2-(2', 3'-diméthyl-anilino)-phényl, magnésium. On fait passer sur cette solution à 35O C du formaldéhyde formé par dépolymérisation de 5,0 g de paraformaldéhyde et on maintient la température à 35 C pendant 1 h après la fin de l'addition.
On décante le mélange réactionnel pour le séparer du magnésium éventuellement présent, on le dilue avec 20 ml d'eau, on l'acidule avec de l'acide chlorhydrique 6 N et on l'agite à 40-50 C pendant 1 h.
On le dilue ensuite avec une nouvelle quantité d'eau, on l'extrait à l'éther, on sèche l'extrait éthéré et on l'éva- pore, obtenant ainsi de la 2-hydroxyméthyl-2', 3'-! dimé- thyl-diphénylamme, p. f. 65-670 C après cristallisation dans de l'hexane. On traite une solution de 20 g de ce produit dans 100 mll d'éthanol avec 40 g de formaldéhyde à 37 /o dans du méthanol aqueux, et on agite le mélange à la température ordinaire pendant 2 h et à 600 C pendant 3 h. On refroidit le mélange et on recueille la N- (2,3-diméthyl-phényl)-1, 2-, dihydro-4H-3, 1-benzoxazine ; p. f. 61-64 C après cristallisation dans de ll'hep- tane.
On maintint un mélange de 1,0 g de ce produit, 5 ml de pyridine et 2 ml d'eau à environ 30 C, tout en lui ajoutant 1,7 g de permanganate de potassium en un laps de temps de 2 h. On laisse reposer le mélange jusqu'au lendemain et on ajoute encore 2 ml de pyridine, après quoi on l'agite pendant encore 2 h. On filtre pour séparer le bioxyde de manganèse, on dilue le filtrat à l'eau et on l'extrait à l'éther. On lave la solution éthérée avec de l'acide chlorhydrique 1,5 N, avec de d'eau, avec une solution de carbonate de sodium et avec une solution de chlorure de sodium.
On la sèche et on l'évapore, obtenant ainsi la N-(2, 3-diméthyltphényl)-1, 2-dihydro-4H- 3,1-benzoxazin-4-one ; p. f. 82-830C après cristalllisation dans du cyclohexane.
Les matières'de départ utilisées dans le. procédé selon l'invention peuvent tre préparées de diverses manières, comme illustré dans les exemples précédents. Ainsi, on peut introduire un ou plusieurs groupes 2,3-dannéthylphénoxy dans un noyau de la quinazoline ou de la di benzodiazocine,puistransposer le groupe 2,3-diméthylphényle sur un atome d'azote adjacent par chauSage. On peut oxyder la N-(2, 3-diméthyl-lphényl)-isatine en un anhydride d'acide ; ou on peut oxyder la N-(2, 3-diméthyl phényl)-1, 2-dihydro-4H-3,1-benzoxazine en N- (2, 3-di méthyl-phényI)-l, 2-dihydro-4H-3, l-benzoxazin-4-one.
Les matières de départ peuvent tre utilisées à l'état brut ou purifié, et peuvent également etre Iproduites in situ et utilisées sans tre isolées.

Example 1

Example 2
A solution of 1.0 g of 1- (2, 3-dimethyl-phenyl) -2, 4- (1H, 3H) quinazolin-dione in 20 ml of ethylene glycol containing 4, is refluxed for 24 hours. 0 g of 85/0 potassium hydroxide and 2.8 ml of water, the solution is made up to 120 ml with water and acidified with dilute hydrochloric acid. The starting material can be prepared as follows.
A solution of 10g of 2,4-di (2,3-dimethyl-phenoxy) -quinazoline in 30 ml of diethylene glycol dimethyl ether is diluted with 15 ml of 2N sodium hydroxide and it is heated briefly to 500 ° C. Ice is added followed by water, the alkaline solution is washed with ether and acidified with dilute hydrochloric acid. The insoluble 2- (2, 3-dimethyl-phenoxy) - 4- (3H) -quinazoEnone is collected and crystallized from ethanol; p. f. 272-273o C.
A mixture of 1 g of this product and 20 ml of mineral heavy oil is heated for 2 h at 328-3350 C, under nitrogen and with stirring. After cooling, the mixture is diluted with petroleum ether and the insoluble 1- (2, 3-dimethyl-phenyl) -2, 4 (1H, 3H) quinazolin-dione is collected; p. f. 2700 C after crystallization from ethanol.
Example 3
10 g of 4-, phenoxy-2- (2, 3-dimethyl-phenoxy) -quinazoline are heated at 320-330 C for 3 h, under nitrogen and with stirring. The melt, containing 3-phenyl-1- (2, 3-dimethyl-phenyl) -2, 4- (1H, 3H) quinazo line-dione, is cooled and poured into 150 ml of ethanol, using some of the ethanol to rinse the reaction vessel. 55 g of a 50% aqueous sodium hydroxide solution are added and the mixture is heated under reflux for 24 h, concentrated to a small volume, diluted with 250 ml of water and dissolved. The N- (2,3 dimethyl-phenyl) -anthranilic acid which has separated is collected and crystallized from aqueous ethanol; p. f. 229-2300 C.
The starting material can be prepared as follows.
15 g of 2-chloro-4-phenoxy-quinazoline are added to a solution prepared from 7.3 g of 2,3-dimethyl-phé noi and 2.7 g of sodium hydride at 55 O / o in 50 ml of diethylene glycol dimethyl ether. The mixture was heated under reflux for 5 h, diluted with ice and water and extracted with ether. The ethereal extract is washed with water, dried, evaporated and the residue triturated with petroleum ether. In this way 4-phenoxy-2- (2, 3-d. Methyl-phenoxy) -quinazoline is obtained, which can be used without further purification.

A solution of 3.0g of 2- (2, 3-dimethyl-phé noxy) -4- (3H) quinazolinone in 50 ml of 1N sodium hydroxide is treated with 5 ml of dimethyl sulfate and stirred. at room temperature for 24 h. 3-Methyl-2- (2, 3-dimethyl-phenoxy) -4- (3H) quinazolinone is collected, washed with water and dried.
Example 5
A mixture of 1 g of 5,11-bis (2,3-dimethyl-phenyl) -dibenzo [b, f] [1, 5] diazocine-6, 12- is refluxed for 24 hours; dione, 1 g of potassium hydroxide and 10 ml of ethylene glycol. The mixture is cooled, diluted with water, filtered, the filtrate acidified with dilute hydrochloric acid and heated in a water bath for 5 min. Insoluble N- (2, 3-dimethyl-phenyl) anthranilic acid is collected; p. f. 9-230O C after crystallization from ethanol.
The starting material can be prepared as follows.
A suspension of 0.9 g of 54% sodium hydride (dispersed in mineral oil) in 10 ml of bis (2-methoxy-ethyl) ether is stirred while cooling in an ice bath. 2.5 g of 2,3-dimethyl-phenol are added in small portions and the mixture is stirred for several minutes at room temperature, then in a hot water bath until complete dissolution and cessation of evolution. The solution is cooled to room temperature, 2.75 g of 6,12-dichloro-dibenzo [b, f] [1, idiazocine is added in small portions, the mixture is stirred for 1 hour, then heated. in a bain-marie for 17 h.
The solution is poured into 200 ml of water and the mixture is extracted with benzene. The benzene extract is washed with water, dried, evaporated and the residue triturated with petroleum ether.
The solid product which is 6,12-bis (2,3-di-methyl-phenoxy) -dibenzo [b, f] [1, 5] diazocine is collected; p. f. 210 2150 C. This product is transposed into 5,11-bis (2,3-dimethyl-phenyl) -dibenzo [b, f] [1, 5] diazocine-6, 12-dion by heating it to 250-2900 C for 2 1/2 h under nitrogen.
Example 6
A mixture of 5.0 g of N- (2, 3 dimethyl-phenyl) -isatin in 50 ml of acetic acid is heated to 50-70O C and maintained at this temperature while adding 1.33 g of chromic anhydride in portions. The N- (2,3-dimethyl-phenyl) -isatoic anhydride which has formed is collected and the filtrate is diluted with water to obtain a new quantity of the same product; p. f. 197-198 C after crystallization from ethanol. The crude product is refluxed in 50 ml of ethanol with 10 g of sodium hydroxide; d. um at 50 / o for 1 h.
The mixture is concentrated to a small volume, diluted with water and acidified with dilute hydrochloric acid. Collected N- (2,3-dimethyl-phenyl) -anthraniiic acid which separated, p. f. 229-230 C after crystallization from ethanol. The same product is obtained by hydrolyzing N (2,3-dimethyl-phenyl) -isatoic anhydride in a mixture boiling under reflux of 25 ml of diethylene glycol dimethyl ether and 10 ml of 6 N hydrochloric acid.
The starting material can be prepared as follows.
Gradually 18 g of 2,3-dimethyl-! phenol to a cooled and stirred suspension of 6.9 g of 54 l / o sodium hydride in 75 ml of diethyleneglycol dimethyl ether. The mixture is heated to 500 ° C. until the evolution of hydrogen ceases, then 25 g of 2-chloro-4-methyl-quinoline are added all at once.
The mixture is heated under reflux for 2 h, allowed to stand at room temperature overnight, diluted with ice and water and extracted with ether. The ethereal extract is washed with several portions of water, dried and evaporated, thereby obtaining deula 2- ^ (2, 3-dimethyl-phenoxy) -4-methyl-quinoline; p. f. 123-124 C after crystallization from aqueous ethanol. 47.8 g of this product were heated at 320-3300 C for 3 h under nitrogen, the melt was partially cooled, diluted with 300 ml of cyclohexane and heated to reflux with stirring.
Insoluble N- (2,3-dimethyl-phenyl) -4-methyl-carbostyriil is collected; p. f. 194-195 C after crystallization from aqueous ethanol. 2 g of this product are dissolved in 25 ml of pyridine with heating and the solution is diluted successively with 10 ml of 2N sodium hydroxide and 15 ml of water. 5.6 g of potassium permanganate are added over 2 h, while maintaining the temperature at 35-40O C while cooling. The mixture is slowly cooled to room temperature and, 18 hours later, made slightly more basic and filtered.
The filtrate is washed twice with ether, acidified strongly with hydrochloric acid and left to stand overnight. N (2,3-dimethyl-phenyl) -isatin is collected; p. f. 188 ° C. after crystallization from ethanol.
Example 7
A solution of 1 g of N- (2, 3-dimethyl-phenyl) -1, 2-dihydro-4H-3,1-benzoxazin-4-one in 2 ml of absolute ethanol is treated with 10 ml of Concentrated ammonia and heated a little below the boiling point for 30 minutes or until a clear solution is obtained.
The solution was acidified with dilute hydrochloric acid, cooled and the insoluble N- (2,3-dimethyl-phenyl) -anthranilic acid collected; p. f. 229-2300C after crystallization from aqueous ethanol.
The sodium salt of N- (2, 3-ldimethzrl-phenyl) anthranilic acid can be prepared by dissolving N (2,3-dilmethyl-phenyl) -anthranilic acid in etnanol, by adding a equivalent of aqueous or alcoholic sodium hydroxide and evaporating the mixture in vacuo. The potassium, calcium, ammonium and 2-hydroxy-ethylamine salts can be prepared in a similar manner from potassium carbonate, calcium hydroxide, ammonia and 2-hydroxy-ethylamine.
The starting material can be prepared as follows.
5.5 g of 2-bromo-2 ', 3'-dimethyl-diphenylamine in 30 ml of anhydrous ether are added to a mixture containing magnesium and ethylmagnelsium bromide (prepared from 3.03 g of ethyl bromide, 2.53 g of magnesium turnip and 100 ml of anhydrous ether) and the mixture is heated under reflux with stirring and under a nitrogen atmosphere overnight.
Most of the ether is removed by distillation of the mixture containing magnesium and the N-bromagnesium salt of 2-bromo2 ', 3'-dimethyl-di, phenylamine. 80 ml of anhydrous tetrahydrofuran are added, the mixture is heated under reflux for 16 h and cooled, thereby obtaining a solution containing the N-bromomagnesium salt, 2- (2 ', 3'-dimethyl-anilino bromide. ) -phenyl, magnesium. Formaldehyde formed by depolymerization of 5.0 g of paraformaldehyde is passed through this solution at 35 ° C. and the temperature is maintained at 35 ° C. for 1 hour after the end of the addition.
The reaction mixture is decanted to separate it from any magnesium present, diluted with 20 ml of water, acidified with 6N hydrochloric acid and stirred at 40-50 C for 1 h.
It is then diluted with more water, extracted with ether, the ethereal extract is dried and evaporated, thus obtaining 2-hydroxymethyl-2 ', 3'-! dimethyl diphenylamme, p. f. 65-670 C after crystallization from hexane. A solution of 20 g of this product in 100 ml of ethanol is treated with 40 g of 37% formaldehyde in aqueous methanol, and the mixture is stirred at room temperature for 2 h and at 600 C for 3 h. The mixture was cooled and the N- (2,3-dimethyl-phenyl) -1, 2-, dihydro-4H-3, 1-benzoxazine was collected; p. f. 61-64 C after crystallization from heptane.
A mixture of 1.0 g of this product, 5 ml of pyridine and 2 ml of water was maintained at about 30 ° C., while adding 1.7 g of potassium permanganate to it over a period of 2 h. The mixture is left to stand overnight and a further 2 ml of pyridine is added, after which it is stirred for a further 2 h. It is filtered to separate the manganese dioxide, the filtrate is diluted with water and extracted with ether. The ethereal solution is washed with 1.5N hydrochloric acid, with water, with sodium carbonate solution and with sodium chloride solution.
It is dried and evaporated, thus obtaining N- (2, 3-dimethyltphenyl) -1, 2-dihydro-4H-3,1-benzoxazin-4-one; p. f. 82-830C after crystallization from cyclohexane.
The starting materials used in the. process according to the invention can be prepared in various ways, as illustrated in the preceding examples. Thus, one or more 2,3-dannethylphenoxy groups can be introduced into a nucleus of quinazoline or di-benzodiazocine, puistranspose the 2,3-dimethylphenyl group onto an adjacent nitrogen atom by heating. N- (2, 3-dimethyl-lphenyl) -isatin can be oxidized to an acid anhydride; or we can oxidize N- (2, 3-dimethyl phenyl) -1, 2-dihydro-4H-3,1-benzoxazine to N- (2, 3-di methyl-phenyl) -l, 2-dihydro-4H -3, 1-Benzoxazin-4-one.
The starting materials can be used in the crude or purified state, and can also be produced in situ and used without being isolated.
