La présente invention est relative à la synthèse et aux applications à l'industrie pharmaceutique des acides p-alkénylo- xy-, p-alkynyloxy- et p-cycloalkényloxyphénylacétiques et de leurs sels.,notamment leurs sels de sodium et de magnésium.

dans laquelle R1 représente un radical non saturé alkényle, alkynyle. ou cycloalkényle, R2 et R3 représentent chacun un radioal alkyle alkényl inférieur, inférieure/halogène ou alkoxy inférieur, R et R pouvant être iden- tiques ou différents, l'un des R et R3 pouvant aussi être de l'hy- drogène, et R4 représente de l'hydrogène ou un alkyle inférieur.
La demanderesse a découvert que ces composés possèdent' de nombreuses applications en pharmacie à cause de leur activité antipyrétique, anti-inflammatoire, analgésique et antipasmodique.
La toxicité de ces composés s'est révélée faible. dans laquelle R2, R3 et R4 ont la signification donnée précédem- ment, et un tel acide de départ est soit alkylé directement en un # position 4 par/halogénure d'alkényle, d'alkynyle ou de cyclo- alkényle, soit estérifié, alkylé en position 4 et rehydrolysé en acide.
L'acide de départ'peut lui-même être obtenu pour ce procédé I par diverses voies de synthèse résumées brièvement ci- après et reprises dans les schémas de réactions donnés par la suite.

(A) Le phénol substitué en position 3 ou 3 et 5 par alkényle # des groupements halogène, alkoxy ou alkyle est substitué sur la fraction hydroxyle par un groupe protecteur, tel qu'alkyle, benzyle, benzoyle, etc. Une réaction du type Friedel-Craft sur ce composé donne l'acétophénone correspondante, laquelle par réaction de
Willegerodt-Kindler, puis traitement par un agent désalkylant, donne l'acide p-hydroxyphénylacétique désiré, (B) Le phénol substitué en position 3 ou 3 et 5 par alkényle, est ,des groupements halogène, alkoxy ou alkyle/substitué sur la fraction hydroxyle par un groupe protecteur, tel qu'alkyle, benzyle, benzoyle, etc.
Une réaction de chlorométhylation sur ce composé, suivie d'une cyanuration et d'un traitement par un agent désalkylant, donne l'acide p-hydroxyphénylacétique désiré.
(C) Cette méthode est particulière aux dérivés halo- halogéné génés l'acide p-hydroxyphénylacétique ou son dérivé/portant un groupe protecteur sur la fraction hydroxyle est traité par un agent d'halogénation pour donner selon les conditions expérimentales les acides p-hydroxyphénylacétiques .halogénés sur le cycle.
On peut représenter comme suit les diverses réactions citées.

Suivant un autre procédé (Procédé II), les'acides substitués de la formule I sont préparés en 'portant d'un p-alkényl- oxy-, p-alkynyloxy- ou p-cycloalkényloxyphénylacétonitrile, de la formule :

Divers exemples de préparation des acides substitués suivant l'invention sont donnas ci-après, ces exemples n'étant ce- pendant nullement limitatifs.Dans ces divers exemples, les étapes sont numérotées de manière à permettre une référence aux schémas dé- veloppés précédemment.

Acide 3-chloro-4-(2-.butyloxhylacétigue

On prépare cet acide en suivant la méthode B du pro- cédé I, de la façon suivante., (1) On mélange 1285 gr de o-chlorophénol, 400 gr de soude et 4 litres d'eau. On refroidit par un bain de glace à une température d'environ la et on ajoute goutte à goutte en agitant vigoureusement 1260 gr (940 cc) de diméthylsufate On agite pen- dant 2 heures au reflux, on refroidit, on dilue par de l'eau, on sépare la phase organique inférieure et on extrait la phase aqueuse à l'éther. La phase organique est lavée à l'eau, à l'acide sulfurique dilué, puis à l'eau, on sèche sur du sulfate de magnésium et on distille.
On obtient 1324 gr de 3-chloro-4-méthoxybenzène Eb: 59-61 /0,8 mm; n25D = 1,5423 Rendement - 93% (2) On mélange 142 gr de 3-chloro-4-méthoxybenzène, 150 gr de formaldéhyde à 40% et 400 ce d'acide acétique. La solution est portée à 90 C et maintenue pendant 4 heures et demie à cette température avec agitation et passage d'un courant d'acide chlorhy- drique gazeux sec. Après refroidissement, on sépare la phase orga- nique et l'on extrait plusieurs fois la phase aqueuse au benzène.
Les extraits organiques combinés sont lavés jusqu'à neutralité par une solution aqueuse glacée saturée de sel; on sèche sut sulfate de magnésium, on évapore et on rectifie sous vide. On obtient 118 gr

(3) 190 gr de 3-chloro-4-méthoxychlorométhylbenzène (1 mole) sont ajoutés goutte à goutte avec agitation à une solution de 53 gr de cyanure de sodium dans 300 cc de diméthylsulfoxyde.
Le mélange est chauffé au bain-marie à 35-40 C pendant 2 heures et demie avec agitation. Après refroidissement, on dilue par de l'eau et on extrait à l'éther. La phase organique est lavée-par du HCl 6N et à l'eau, séchée sur sulfate de Mg, évaporée, et le résidu cris- tallise après peu de temps. On obtient 165 gr de 3-chloro-4-méthoxy- phénylacétonitrile. (Rendement : 91%; F :56 C; Ebt 148-154 C/2mm), utilisé brut pour l'opération suivante.
(4) 690 gr de 3-chloro-4-méthoxyphénylacétonitrile et 2500cc d'acide bromhydrique aqueux à 48% (d = 1,5) sont portés au reflux pendant 15 heures. On évapore l'acide bromhydrique sous vide et le résidu solide est filtré et lavé à l'éther de pétrole, On obtient 670 gr d'acide 3-chloro-4-hydroxyphénylacétique.
(Rendement 95%; F : 104-106 C), utilisé brut pour l'opération suivante.
(5) 670 gr d'acide 3-chloro-4-hydroxyphénylacétique, 2000 ce de méthanol et 0,8 gr d'acide p-toluènesulfonique sont por- tés au reflux une nuit. On évapore le maximum d'alcool, on dilue par -de l'eau, on extrait à l'éther, on sèche la phase éthérée sur sul- fate de magnésium et on évapore. Le résidu rectifié sous vide donne
325 gr de 3-chloro-4-hydroxyphénylacétate de méthyle.
Eb : 124-130 /1 mm; n25D = 1,5482; Rendement : 45%.
(6) 100 gr de 3-chloro-4-hydroxyphénylacétate de mé- thyle (0, 5 mole), 67,5gr de bromure de 2-butényle (O, 5 mole) ,- 69 gr de carbonate de potassium anhydre et 150 cc d'acétone sont portési

au reflux pendant 6 heures avec agitation vigoureuse. Après refroi- dissement, on dilue avec de l'eau et on extrait à l'éther. La phase éthérée est lavée à l'eau, au carbonate, puis à l'eau, séchée sur sulfate de magnésium et évaporée. La distillation du résidu donne 91,4 gr de 3-chloro-4-(2-butényloxy)phénylacétate de méthyle.
Eb : 160-164 /2 mm; n25D = 1,5300; Rendement =- 72% (7).50,8 gr de 3-chloro-4-(2-butényloxy)phénylacétate de méthyle (0,2 mole), 22 gr de potasse caustique, 50 cc d'eau et 150 cc de méthanol sont portés au reflux pendant 2 heures. On évapore le méthanol, on dilue le résidu par de l'eau et de la glace et on acidifie par HCl à 20%. Le solide est filtré, lavé à l'éther de pé- trole et séché. Le produit brut, 39,8 gr (83%), est recristallisé dans un mélange benzène-éther de pétrole.
F: 82-83 (benzène- éther de pétrole)
C H Analyse centésimale % calc, 59,88 5,44 % trouvé 59,75 5,26' (8) A 8,85 gr d'acide 3-chloro-4-hydroxyphénylacétique (obtenu lors de l'étape (4) et recristallisé dans l'eau, F = 107- 108 C), 5,6 gr de potasse caustique et 100 cc d'éthanol, on ajoute goutte à goutte 12,1 gr de bromure d'allyle à la température ordi- naire. On porte au reflux pendant 2 heures avec agitation. On refroi- dit, on filtre le sel formé et on évapore l'alcool sous vide.
Le résidu est dilué par H20 et acidifié par HCl à 20%. On filtre et on sèche le précipité. on obtient 6 gr d'acide 3-chloro-4-allyloxyphé- nylacétique. F : 92-93 (éthanol-H20) ou (éther-éther de pétrole 35-40).

On prépare cet acide par le procédé I-A. Les phases suivantes se référant à nouveau au schéma de réactions donné anté- rieurement pour cette méthode,certaines phases ayant été inversées ou groupées, (2) On mélange 124 gr de gaiacol (1 mole), 108 cc d'anhydride acétique et on ajoute rapidement 149 gr de chlorure de zinc. On porte au reflux pendant 3 minutes avec agitation et on verse le mélange encore chaud dans 800 cc d'eau.
On extrait à l'éther,/ on lave la couche éthérée à l'eau, au carbonate à 5% et à l'eau, on 1 sèche, on évapore, Le résidu est rectifié sous vide et donne 93 gr de 3-méthoxy-4-hydroxyacétophénone qui se solidifie rapidement, i Eb :122-132/0,6 mm ; F 115-116,5 (benzène) s Renderrint - 28%.
(1) A 160 gr de 3-méthoxy-4-bydroxyacétophénone dis- sous dans 250 cc d'éthanol, on ajoute goutte à goutte en agitant, 59 gr de potasse dans 48 cc d'eau, puis goutte à goutte 148 gr de chlorure de benzyle. On porte au reflux pendant 3 heures en poursui- vant l'agitation. On évapore le maximum d'alcool et on dilue le ré- sidu par de l'eau. On extrait à l'acétate d'éthyle et on lave la phase organique à l'eau, à la soude à 10%, puis à l'eau. On sèche sur sulfate de magnésium et on évapore le solvant. On obtient 219 gr

On ajoute ensuite une solution de 40 gr de aoude dans 70 cc d'eau et on reflue une nuit. La solution est acidifiée par du HC1 concentré jusqu'à pH 1-2, puis extraite à l'éther et à la méthyl-

éthyl cétone. La phase organique est extraite au carbonate à 10% et la phase aqueuse basique est acidifiée par du HCl à 50%, On fil- tre le solide et on lave à l'éther de pétrole. On obtient 28,5 gr d'acide 3-méthoxy-4-benzyloxyphénylacétique. Rendement 62%; F : : 97-99 (éthanol-eau).
(4 et,5) 100 cc de méthanol anhydre sont saturés par de l'acide chlorhydrique gazeux sec. On ajoute 10 gr d'acide 3-méthoxy- 4-benzyloxyphénylacétique et on porte au reflux une nuit. On éva- pore le maximum d'alcool, on dilue lerésidu avec de l'eau et on ex- trait à l'éther. La phase éthérée est séchée sur du sulfate de ma- gnésium et évaporée. Le résidu, rectifié sous vide, donne 4,4 gr de 3-méthoxy-4-hydroxyphénylacétate de méthyle. Eb : 118 /0,5 mm; n25D = 1,5330; Rendement : 61,1% (6) 10 gr de 3-méthoxy-4-hydroxyphénylacétate de mé- thyle (0,051 mole), 7 gr de carbonate de potassium anhydre, 6,2 gr de bromure d'allyle et 20 cc d'acétone sont portés au reflux pendant 6 heures avec agitation vigoureuse.
On dilue avec de l'eau, on ex- trait à l'éther, on lave la phase éthérée à l'eau, au carbonate, puis à l'eau, on sèche sur du sulfate de sodium et on évapore. Le résidu rectifié sous vide donne 9,4 gr de 3-méthoxy-4-allyloxyphé- nylacétate de méthyle. Eb : 160-161/2,5 mm; n25D = 1,5264; Rendement : 78%.
(7) 4,7 gr de 3-méthoxy-4-allyloxyphénylacétate de méthyle, 1,2 gr da potasse, 20 cc de méthanol et 5 ce d'eau sont portés au reflux pendant 2 heures. (4) 170 gr d'acide 3-méthyl-4-méthoxyphénylacétique ( 1 mole) et 500 ce d'acide bromhydrique aqueux à 48% (d - 1,5) sont portés au reflux pendant 15 heures. On évapore l'acide bromhydrique sous vide et on laisse recristalliser le résidu. On filtre et on lave à léther de pétrole; on obtient 126 gr de l'acide 3-méthyl-4-

hydroxyphénylacétique (Rendement : 81%). Ce produit peut être uti- lisé brut pour l'estérification ou être recristallisé dans l'eau (F : 119-121 ) (5) 95 gr de l'acide 3-méthyl-4-hydroxyphénylacétique (0,67 mole), 0,12 gr d'acide paratoluènesulfonique et 280 gr de méthanol sont portés au reflux pendant une nuit. On évapore le ; maximum d'alcool et on dilue le résidu avec de l'eau.
On extrait à l'éther, on sèche la phase éthérée sur du sulfate de magnésium, on évapore le solvant et on rectifie le résidu sous vide. On obtient 84,5 gr de 3-méthyl-4-hydroxyphénylacétate de méthyle.

On évapore le maxi- mum d'alcool, on dilue le résidu avec de l'eau et de la glace, on acidifie par du HCl à 20% et on filtre la solide. On obtient 10,5 gr de produit brut qui, recristallisé deux fois dans un mélange benzène- éther de pétrole, donne 6,5 gr d'acide 3-méthyl-4-allyloxyphényl- acétique. Rendement 63,1%; F ; 65-67 (éther de pétrole-benzène).

On porte le mélange au reflux pendant 6 heures avec agitation vigoureuse. Après refroidis- sement, on ajoute 200 cc d'eau et on extrait & l'éther. La phase éthérée est lavée à l'eau, au carbonate, puis à l'eau, séchée sur du sulfate de magnésium et évaporée. La distillation du résidu donne 152,3 gr de 3-chloro-4-allyloxybenzène.

On évapore le maximum d'alcool, on on dilue le résidu avec de l'eau et de la glace et/acidifie par du HCl à 20%. On filtre le solide et on lave à l'éthor de pétrole.
On obtient 91,5 gr (Rendement : 81%) de l'acide, recristallisé dans léthanol aqueux ! F 92-93 .

The present invention relates to the synthesis and to the applications to the pharmaceutical industry of p-alkenylo-xy-, p-alkynyloxy- and p-cycloalkenyloxyphenylacetic acids and their salts, in particular their sodium and magnesium salts.
