la N- éthyl-N-P-hydroxyéthyl-amine pour obtenir 1 - (N- éthy l - N -P-hy droxyéthyl) amino-penta- none-4, qui, par a.mination réductive, donne-la 5-(N-éthyl-N f-hydroxyéthyl)amino-2-pentyl- amine.
Le produit intermédiaire, la 1-(N-étliyl-N- P-hydroxyéthyl)amino-pentanone--I, peut être préparé en chauffant, sous agitation, un mé lange de 1-chloro-pentanone-4, de N-éthyl-N-pJ- hydroxyéthylamine et de chlorure de sodium (pour aider la filtration ultérieure) dans du xylène pendant 2 heures sur un bain (le va peur, puis au reflux pendant 3 heures. lie produit obtenu distille entre 89 et 90 C sous 0,35 mm (nD =1,4600) ;
il peut être soumis directement à l'amination réductive. Un échantillon d'une pureté analytique peut. être obtenu par redistillation dans une colonne- point d'ébullition: 85-87 C sous 0,4 mm, nD =1,4583.
La réduction catalytique de la 1-(N-éthyl- N-P-hy droxyéthyl) amino-pentanone-4 dans une solution de méthanol ammoniacal en présence de nickel de Raney à la température ambiante donne la 5-(.LI#-éthyl-N-P-hydroxyétliyl)-a.mino- 2-pentylamine, qui distille entre 88,5 et 96 C sous 0,3 mm. Un échantillon d'une pureté analytique de ce produit distille à 93 C sous 0,6 mm; nD =1,4703.
Le nouveau dérivé de quinoléine, obtenu sous forme de base libre, peut être employé sous forme de phosphate, qui est soluble dans l'eau. Il v a de soi que d'autres sels qui sont solubles dans l'eau et qui sont dérivés d'acides inorganiques non toxiques, tels que l'acide chlorhydrique, bromhydrique, sulfurique, sulf- amique, ete. ou d'acides organiques non toxi ques tels que l'acide tartrique, citrique, succi nique, acétique, benzoïque, oléique, etc. peu vent servir dans le même but.
<I>Exemple:</I> Un mélange de 90 g de 4-, 7-dieliloi-oquino- léine, 90 g de phénol, 1 g de iodure de potassium et 132 g de 5- (N-étliv T-N-fl-hydr oxyéthyl) amino- 2-pentylamine est chauffé sous agitation pen dant 18 heures à l25-130 C.
Le mélange réactionnel obtenu contient, la 7-chloro-4-[5'- (N-éthy 1-N-P-hydroxyéthyl) amino-pentyl-2']- aminoquinoléine principalement sous forme de la. base libre (environ 70%), mais aussi en moindre quantité (environ 30 l0) sous forme de chlorhydrate. Si l'on emploie une propor tion plus grande de 5-(N-éthyl-N-P-hydroxy- éthyl)amino-\'-pentylamine par rapport.
à la 4,7-diehloroquinoléine, la proportion de la 7 chloro-4- [ 5'-(N-éthy 1-N-P-hydroxy éthy 1) amino- pentyl-2']-amino-quiiioléine sous forme de base libre sera plus grande.
Le mélange contenant la quinoléine désirée peut être purifiée et celle-ci mise complètement sous sa forme de base libre, en la transformant d'abord en sel d'acide phosphorique et en le purifiant comme suit: On ajoute 1.,9 1 d'alcool méthylique et on filtre le mélange contenant la. 7-chloro-4-[5'-(N- éthy 1-N -P - hydroxyéthyl) amino - pentyl - 2'] - amino-qninoléine, sous forme de base libre et de chlorhydrate, additionné de charbon.
On traite le filtrat avec 270 ems d'une solution de 1.00 g d'acide phosphorique dans 300 cm d'alcool méthylique. On gratte les parois du récipient avec une baguette de verre et on laisse reposer le mélange pendant 2 jours. Le produit eristallisé est filtré, lavé à l'alcool méthylique et, séché. On obtient ainsi 101 g de disphosphate brut de 7-cliloro-4-[5'-(N-éthyl- N -P-hydroxyéth@#1)amino-pentyl- 2'] - amino- quinoléine fondant à l55-156 C.
On peut obtenir une quantité supplémentaire de di- phosphate sous forme d'une masse gommeuse en concentrant les eaux-mères jusqu'à environ la moitié de leur volume et en ajoutant de l'acétone. Le diphosphate obtenu est dissous dans l'eau, la solution est. rendue basique à l'aide d'hydroxyde d'ammonium et la base libre est extraite à l'aide de chloroforme. Après avoir évaporé le chloroforme, le résidu est dissous dans de l'éther et la cristallisa tion amorcée en grattant les parois du réci pient avec une baguette de verre. Il se sépare la base brute, fondant à 77-82 C.
Par re- cristallisation de ce produit dans du dichlo- rure d'éthylène ou de l'acétate d'éthyle, on obtient la 7-chloro-4-[5'-(N-éthyl-N-f-hydroxy- éthyl) amino-pentyl-2'] -amino-quinoléine puri fiée, fondant à 89-9l C.
Avant la libération de la quinoléine basi que, le diphosphate susmentionné peut aussi être purifié par dissolution de 100 g de ce produit dans 200 cm3 d'eau en ajoutant 200 cm-' d'a.lcool éthylique et, en filtrant la solution avec du charbon. Au filtrat chaud, on ajoute encore 400 cms d'éthanol et la cris tallisation est amorcée comme auparavant, en. grattant les parois du récipient avec une ba- ",mette de verre. Le produit séparé est séché pendant trois heures à 100 C.
Le diphosphate de 7-chloro-4-[5'-(N-éthyl-N-fl-hydroxyéthyl)- amino-2-pentyl-2']-amino-qizinoléine purifié a un point de fusion de 162-164 C. Du filtrat, on obtient encore 10 g de quinoléine basique en se servant de la méthode déjà décrite ci- avant.
Au lieu de la 4,7-dichloroquinoléine, on peut se servir de la 4-bromo-7-chloroquino- léine ou de la 4-iodo-7-chloroquinoléine comme produit de départ, mais leur utilisation n'offre aucun avantage qui puisse compenser le prix de ces matières premières plus diffi ciles à obtenir.
