Abstract
MéTODO PARA INTENSIFICAçãO DA PRODUçãO DE COMPOSTOS DE ISOPRENóIDE. A presente invenção refere-se a métodos de produção de um isoprenóide ou precursor de isoprenóide em uma célula hospedeira geneticamente modificada. Os métodos geralmente envolvem modulação do nível de hidróximetilglutaril-CoA (HMG-CoA) na célula, de modo que o nível de HMG-CoA não é tóxico para a célula e/ou não inibe substancialmente o crescimento celular, mas é mantido em um nível que proporciona produção em alto nível de mevalonato, IPP e outros produtos a jusante de um isoprenóide ou via de isoprenóide (por exemplo, poliprenila) difosfatos e compostos de isoprenóide. A presente invenção ainda proporciona células hospedeiras geneticamente modificadas que são adequadas para uso no método em questão. A presente invenção ainda proporciona estruturas de ácido nucléico recambinantes para uso na geração de uma célula hospedeira geneticamente modificada em questão, incluindo estruturas de ácido nucléico recombinantes compreendendo seqüências de nucleotídeo que codificam uma ou mais enzimas da via de mevalanato e vetores recombinantes (por exemplo, vetores de expressão recombinantes) compreendendo as mesmas. A presente invenção ainda proporciona métodos para a identificação de ácidos nucléicos que codificam variantes de reductase de HMG-CoA (HMGR) que proporcionam alívio da toxicidade acúmulo de HMG-CoA-induzida.
