Abstract

BACTÉRIAS E MÉTODOS DE PRODUÇÃO DE ÁCIDO SUCCÍNICO E OUTROS PRODUTOS QUÍMICOS UTILIZANDO DIFUSÃO FACILITADA PARA IMPORTAÇÃO DE AÇÚCAR. A presente invenção refere-se à produção de ácido succínico e outros produtos químicos derivados de fosfoenolpiruvato (PEP) por fermentação com um micro-organismo em meio de fermentação contendo um ou mais açúcares importados para a célula por difusão facilitada. Preferencialmente, tais micro-organismos têm sua capacidade de utilizar glicose como uma fonte de carbono e energia aumentada e por meio de manipulação genética de cepas progenitoras que foram originalmente construídas com o objetivo de produzir um ou mais produtos químicos (por exemplo, ácido succínico e/ou um sal de ácido succínico) em uma escala comercial utilizando matérias-primas que incluem, por exemplo, glicose, frutose, ou sacarose. As manipulações genéticas da presente invenção envolvem a introdução de genes exógenos envolvidos no transporte e metabolismo de glicose ou frutose nas cepas progenitoras. Os genes envolvidos no transporte e metabolismo de glicose ou frutose também podem ser introduzidos em um micro-organismo antes de desenvolver o organismo para produzir um produto químico particular. Os genes envolvidos no transporte e metabolismo de sacarose também podem ser usados para aumentar ou aprimorar a eficiência de transporte e metabolismo de açúcar por cepas já conhecidas por terem alguma capacidade de utilização de glicose em fermentações biológicas.
