resumo patente de invenção: "detecção de sequências de ácido nucleico alvo por clivagem e hibridização de po". a presente invenção refere-se à detecção de uma sequência de ácido nucleico alvo por um ensaio de poch (clivagem e hibridização de po) em um substrato sólido. a presente invenção detecta a sequência de ácido nucleico alvo pelo uso de em que o po (oligonucleotídeo de sondagem) hibridizado com a sequência de ácido nucleico alvo é clivado e a clivagem do po é detectada por hibridização com o co (oligonucleotídeo de captura). na presente invenção, um po não clivado é hibridizado com o co imobilizado sobre o substrato sólido. os projetos do po e o co são convenientes e a otimização das condições de reação é rotineiramente fácil na presente invenção. onde a detecção do sinal no substrato sólido é continuamente realizada junto com a repetição da clivagem dos pos na presente invenção, o número dos pos clivados é aumentado no número de repetição da reação de clivagem e o sinal é mudado em paralelo com o número dos pos clivados. depois, a sequência de ácido nucleico alvo pode ser detectada em uma maneira em tempo real. ao contrário, a mudança do sinal não é observada na ausência da sequência de ácido nucleico alvo.
