Claims (5)

1. Postupak za istovremeno utvrđivanje i/ili tipizaciju i/ili kvantifikaciju prisutnosti HPV 18, 52, 59, 39, 51, 56, 66, 16, 45, 58, 31, 33 i 35 u biološkom uzorku, naznačen time da postupak sadrži sljedeće četiri paralelne reakcije: Reakcija 1- a) umnažanje fragmenata nukleinskih kiselina od HPV 18, 52 i 59 u prisutnosti (i) polimeraze nukleinske kiseline (ii) nizvodne PCR početnice s oligonukleotidnom sekvencom od SEQ ID NO: 1, (iii) uzvodne PCR početnice s oligonukleotidnom sekvencom od SEQ ID NO: 2, (iv) probe s oligonukleotidnom sekvencom od SEQ ID NO: 7,8 i 9, označene s fluoroforom i molekulom za gašenje i b) otkrivanje promjene fluorescencije njihove valne duljine koja određuje HPV tip 18, 52 i 59; Reakcija 2- a) umnažanje fragmenata nukleinskih kiselina od HPV 39,51,56 i 66 u prisutnosti (i) polimeraze nukleinske kiseline (ii) nizvodne PCR početnice s oligonukleotidnom sekvencom od SEQ ID NO: 1, (iii) uzvodne PCR početnice s oligonukleotidnom sekvencom od SEQ ID NO: 3, (iv) probe s oligonukleotidnom sekvencom od SEQ ID NO: 10, 11, 12 i 13, označene s fluoroforom i molekulom za gašenje i b) otkrivanje promjene fluorescencije njihove valne duljine koja određuje HPV tip 39, 51, 56 i 66; Reakcija 3- a) umnažanje fragmenata nukleinskih kiselina od HPV 16,45 i 58 u prisutnosti (i) polimeraze nukleinske kiseline (ii) nizvodne PCR početnice s oligonukleotidnom sekvencom od SEQ ID NO: 1, (iii) uzvodne PCR početnice s oligonukleotidnom sekvencom od SEQ ID NO: 4, (iv) probe s oligonukleotidnom sekvencom od SEQ ID NO: 14, 15 i 16, označene s fluoroforom i molekulom za gašenje i b) otkrivanje promjene fluorescencije njihove valne duljine koja određuje HPV tip 16, 45 i 58; Reakcija 4- a) umnažanje fragmenata nukleinskih kiselina od HPV 31, 33, 35 i unutrašnje kontrole u prisutnosti (i) polimeraze nukleinske kiseline (ii) nizvodne PCR početnice s oligonukleotidnom sekvencom od SEQ ID NO: 1, (iii) uzvodne PCR početnice s oligonukleotidnom sekvencom od SEQ ID NO: 5, (iv) probe s oligonukleotidnom sekvencom od SEQ ID NO: 17, 18, 19 i 20, označene s fluoroforom i molekulom za gašenje i (v) unutrašnje kontrole, te b) otkrivanje promjene fluorescencije njihove valne duljine koja određuje HPV tip 31, 33, 35 i unutrašnju kontrolu.
2. Postupak prema zahtjevu 1, naznačen time da se navedene reakcije provode paralelno i u zasebnim bočicama.
3. Postupak prema zahtjevima 1-2, naznačen time da su četiri fluorofore FAM, VIC, NED i ROX.
4. Postupak prema zahtjevima 1-2, naznačen time da unutrašnja kontrola je HPV6.
5. Komplet za istovremeno utvrđivanje i/ili tipizaciju i/ili kvantifikaciju HPV 16, 18, 31, 33, 35, 39, 45, 51, 52, 56, 58, 59 i 66 u biološkom uzorku naznačen time da sadrži četiri kombinacije početnica-proba koje se koriste u skladu s patentnim zahtjevom 1.
