Abstract
novo método de sequencimanto de pcr e uso do mesmo na genotipagem hla - a invenção proporciona um método de sequencimento de pcr, onde a combinação de índices primários, uma estratégia de cisalhamento de dna incompleto e uma técnica de sequenciamento de segunda geração (técnica de sequenciamento de extremidade em pares) pode fazer com que o comprimento dos produtos de pcr que podem ser sequenciados por um sequenciador maiores do que o comprimento do sequenciamento máximo do sequenciador enquanto faz o uso completo das características da técnica de sequenciamento de segunda geração tal como um alto rendimento e baixo custo, desse modo, ampliando muito seu escopo aplicável. além disso, a presente invenção também proporciona índices primários para o método de sequencimanto de pcr e o uso do método em genotipagem, particularmente na analisa de hla, e também proporciona os pcr primários usados, particularmente os pcr primários para os genes hla-a,b, hla-c e hla-dqb1.
