Abstract

"PROCESSO PARA A TIPIFICAçãO RáPIDA DE MICROORGANISMOS, E, CONJUNTO DE REATIVOS". Proporciona-se um processo para tipificar rapidamente leveduras, parasitas e bactérias. Compreende as seguintes etapas: a) Preparar, em de 5 a 60 minutos, DNA intacto e imobilizado mediante um método que emprega um conjunto de reativos que apenas contém solução tampão, um detergente, um agente quelante e um agente que rompe as pontes de hidrogênio. b) Separar moléculas de DNA intactas ou seus fragmentos de restrição utilizando-se n-úni-equipamentos de eletroforese de campos pulsantes dos sistemas CHEF (Contour Clamped Homogeneous Electric Field) e TAFE (Transversal Altemating Field Electrophoresis) em tempos compreendidos entre 2,5 e 7 horas. c) Selecionar as condições ótimas que serão aplicadas no miniCHEF empregando-se um método que simula a priori os padrões eletroforéticos que se obteria nos referidos géis. d) Proporcionar os tempos de reorientação, velocidades de migração e tamanhos das moléculas sem empregar marcadores de tamanhos, porém valendo-se de um método que analisa distâncias migradas.
