Abstract
Patente de Invenção: "MéTODO E áCIDOS NUCLéICOS PARA A DETECçãO DE MICROORGANISMOS PERTINENTES A FERMENTAçãO". A invenção refere-se à um método para a detecção de microorganismo pertinente à fermentação, assim como à ácidos nucléicos e combinações destes os quais podem ser empregados neste método. A invenção também refere-se ao emprego dos ácidos nucléicos de acordo com a invenção ou combinação para detecção e/ou para a identificação e /ou caracterização de diferentes gêneros ou espécies de microorganismos pertinentes à fermentação. Em conseq³ência, o problema a ser resolvido pela presente invenção era fornecer um método e recursos que tornem possível um teste rápido de cerveja e materiais brutos de fermentação por contaminação com microorganismos, o teste sendo requerido para detectar a faixa total de possíveis microorganismos de contaminação de cerveja. Este problema é resolVido de acordo com a invenção por um método que compreende as seguintes etapas: (a) trazer a amostra em contato com uma combinação de pelo menos duas primeiras moléculas de ácido nucléico (iniciadores), que hibridizam com uma região de um ácido nucléico microbiano conservado em microorganismos pertinentes à fermentação; (b) amplificação do ácido nucléico microbiano ou uma porção deste para produzir pelo menos um fragmento de amplificação; (c) trazer os fragmentos de amplificação obtido na etapa (b) em contato com pelo menos uma segunda molécula de ácido nucléico (sonda) que especificamente hibridiza com pelo menos um fragmento de amplificação o qual compreende uma seq³ência do ácido nucléico microbiano específico para todos os microorganismos pertinentes à fermentação ou para uma ou diversas famílias, gêneros ou espécies de microorganismos pertinentes à fermentação; e (d) detectar pelo menos um ácido nucléico híbrido que consista em um fragmento de amplificação e uma segunda molécula de ácido nucléico introduzida na etapa (c). Além disso são fornecidos ácidos nucléicos que podem ser empregados no método de acordo com a invenção.
