La présente invention se rapporte à des esters stéroïdes.
Les esters d'acides carboxyliques de la 19-nortestostérone sont des produits connus ; indépendamment de leur activité androgèn ils exercent un effet anabolique puissant. On connaît également les 17-alcoyl-19-nor-testostérones possédant un groupe hydroxyle libre en position 17 (brevet allemand n 931.769 en date du 20 novembre 1952 et.brevet américain n 2.721.871 en date du 13 septembre 1954)et manifestant également des effets thérapeutiques intéressants. Ainsi,les composés alcoyliques saturés se distin-

guent par de bons effets anaboliques alors que la 17- éthinyl- 19-nor-tes/terone est douée de propriétés stimulant de façon prononcée la gestation et plus intenses que celles de la 17- éthinyl-testostérone.
Mais on n'avait pas jusqu'ici produit d'esters d'acides carboxyliques des 17-acoyl-19-nor-testostéro- nes.
La présente invention permet d'obtenir des esters car- boxyliques de 17-alcoyl-19-nor-testostérones. Ces composés nou- veaux manifestent des propriétés thérapeutiques très intéres- santes et se distinguent particulièrement par un effet stimu- lant la gestation qui s'exerce à la fois par administration par voie buccale ou par voie hypodermique. Ces esters ayant une bonne solubilité dans les solvants ordinaires des hormones stéroïdes, pan/exemple les huiles végétales comme l'huile de sésame', l'huile de ricin, l'huile de coton, l'huile de tournesol, l'huile d'olive, été,.ainsi que dans les solvants synthétiques comme les glycols, les esters de l'acide lactique, etc, on peut injecter leurs solutions et ainsi disposer d'hormones-retard et obtenir des effets prolongés.
L'invention fournit également un procédé de préparation de ces esters, procédé de type en lui-même connu. L'estérifica- tion peut être effectuée soit sur les 17-alcoyl-19-nor-testosté- rones elles-mêmes,soit sur des produits intermédiaires contenant en position 17 le groupe

dans ce dernier cas avec formation d'autres produits intermédi- aires qui peuvent alors être convertis en esters de 17-alcoyl- 19-nor-testostérones.
Si ce dernier mode,de préparation ne donne pas;lui non plus,satisfaction, on peut opérer de la façon indiquée ci-avant en second lieu, à savoir estérifier des produits intermédiaires

Cette méthode offre l'avantage de permettre l'estérification de matières premières qui ne possèdent qu'à un moindre degré la tendance indésirable du groupe 17ss -OH de donner lieu à une élimination d'eau, comme par exemple les composés l7-éthiny liques fortement non saturés. Ainsi,il est possible par exemple, pour produire les esters de la 17-vinyl-19-nor-testostérone, d'estérifier la 17-éthinyl-19-nor-testostérone de la manière désirée,,puis de convertir le groupe éthinylique par hydrogéna- tion partielle en groupe plus saturé. On l'estérifie d'a- bord de la manière désirée puis on l'hydrolyse partiellement pour enlever le groupe acylique en position 3, on le soumet ensuite à une hydrogénation partielle et,,enfin)on l'oxyde en ester de la 17-éthyl-19-nor-testostérone au moyen d'un oxydant propre à convertir en groupe cétonique le groupe hydroxylique secondaire du noyau. (voir schéma n 4).
Il est à noter que dans ce cas une hydrogénation du groupe éthinylique en groupe éthylique est possible sans hydro- génation simultanée de la double liaison A 4 ainsi que cela se produit, comme on le sait, avec les A -3-cétones.
On peut obtenir une protection similaire de la double liaison 4 4 contre une hydrogénation indésirable par conver- sion intermédiaire du groupe 3-cétonique en cétal. En fait, si on utilise comme produit intermédiaire le 17-éthinyl-19-nor- testostérone-3-cétal ou son 17-ester, on obtient facilement par hydrogénation partielle les composés 17-éthyliques corres- -nor- pondants, qui donnent les esters de 17-éthyl-19/testostérone cherchés par une réaction facile de scission du groupe cétal en milieu acide. (voir schéma n 5).
Comme composés alcoyliques,on doit considérer à la fois les composés alcoyliques aliphatiques inférieurs saturés conte- nant jusqu'à 8 atomes de carbone, en particulier les composés méthylique et éthylique, et les composés non saturés,'en par- ticulier le composé éthinylique.
Les acides carboxyliques à envisager sont principale- ment les acides carboxyliques aliphatiques inférieurs ou cyclo- aliphatiques contenant jusqu'à 11 atomes de carbone, que l'on peut utiliser sous forme d'anhydrides, de chlorures ou d'esters,

butyrique, l'acide valérique, l'acide eaproique, l'acide ipso- caproique, l'acide oenanthylique, l'acide undécylénique, l'aci- de caprique, l'acide undécylique, l'acide malonique, l'acide caprylique, l'acide malique, l'acide cyclopentyl- ou cyclohexyl- propionique, etc. Le type de ces acides peut varier entre de larges limites ; comprennent par exemple les acides céto- carboxyliques.

EXEMPLE 1
Acétate de 17-méthyl-19-nor-testostérone.

On chauffe 2 g de 17-méthyl-19-nor-testostéroneapendant cinq heures, au reflux,sous atmosphère d'azote,avec 7 cm3 de pyridine et 7 cm3 d'anhydride acétique. Après refroidissement, on décompose soigneusement l'excès d'anhydride acétique par l'eau et on épuise le tout à l'éther. On lave la solution éthé- rée au moyen d'acide chlorhydrique 2N, de bicarbonate de sodium et d'eau, on la sèche sur sulfate de sodium,puis on l'évapore à siccité sous atmosphère d'azote. On soumet le résidu huileux à une chromatographie sur 150 g d'alumine (pH environ 4) à l'aide d'un mélange de benzène et de chlorure de méthylène 1/1.
On fait recristalliser au sein d'hexane les fractions cris- tallines ainsi obtenues. Point de fusion 91-93,5 .
EXEMPLE 2
Acétate de 17-éthinyl-19-testostérone. a) on met en suspension 2.98 g de 17-éthinyl-19-nor- testostérone dans 30 cm3 d'anhydride acétique et on traite pro- gressivement, en refroidissant et en agitant, avec une solution de 1,9 g d'acide p-toluène-sulfonique dans 19 cm3 d'anhydride acétique. La dissolution est complète en une heure. Après encore

trente à soixante minutes-de plus, il se dépose une pâte épais- se..Après une durée totale de réaction de cinq-heures, on traite le mélange réactionnel à l'eau, -on agite pendant une à deux heures le 3-énol-17-diacétate précipité,puis.on le sépare par filtration, on le lave à neutralité et on le sèche sous vide à la température ambiante sur du chlorure de calcium.
Pour ef- fectuer l'hydrolyse,on met le diacétate brut en suspension dans ;150 cm3 de méthanol, on ajoute 1,5 cm3 d'acide chlorhydri- que concentré et ,on chauffe à l'ébùllition pendant quinze minu- tes sous atmosphère d'azote. Après refroidissement à la tempé- rature ambiante,,on traite par l'eau et on sépare par filtra- tion le monoacétate brut, on le lave et on le sèche à la tempé- rature ambiante sous vide, sur du chlorure de calcium.
Pour purifier le produite on le fait recristalliser à plusieurs re - prises au sein d'un mélange de chlorure de méthylène et d'hexane Le 17=acétate pur fond à 161-162 . b) on chauffe,pendant dix heures) au bain d'huile (tempé- rature du bain 150 ), un mélange de 5 g de 17 Ó -éthinyl-19-nor- testostérone, 15 cm3 de pyridine et 10 cm3 d'anhydride acétique sous' atmosphère d'azote. Après refroidissement à la température ambiante, on effectue la décomposition par l'eau sous agitation; au bout de deux à trois heures on sépare le 3-énol-17ss -diacéta te brut par filtration, on le lave à l'eau et on le sèche dans le vide sur du chlorure de calcium. On effectue l'hydrolyse du groupe 3-énol-ester comme au paragraphe a ci-avant.
On purifie le 17-acétate brut par chromatographie et recristallisation ou simplement par recristallisation; il fond alors à 161-162 ,

Oenanthylate de 17-éthinyl-19-nor-testostérone.
On chauffe au reflux,pendant dix-sept heures,à une tem- pérature du bain d'huile de 180 , sous atmosphère d'azote, 1 g de 17-éthinyl-19-nor-testostérone avec 5 cm3 de pyridine et 5 cm3 d'anhydride oenanthylique fraîchement distillé. On soumet ensuite le mélange réactionnel à un entraînement à la vapeur d'eau jusqu'à ce que l'odeur de l'acide oenanthylique ait dispa- ru. On extrait le résidu à l'éther, et on lave l'extrait éthéré au moyen d'acide sulfurique 2 N, d'une solution de soude caus- tique 2 N, enfin à l'eau jusqu'à neutralité. Après séchage sur du sulfate de sodium,on concentre la solution et on triture le résidu huileux avec quelques gouttes de méthanol. Le rendement en cristaux est amélioré par vingt-quatre heures de repos au ré- frigérateur à -8¯ Point de fusion 82-84 .

On dissout 2 g de ce 3-énol-diester dans 120 cm3 de méthanol et on fait bouillir au reflux, pendant une demi-heure, avec 1,2 cm3 d'acide chlorhydrique concentré. On.soumet ensuite le produit partiellement hydrolysé à un entraînement par la va- peur d'eau jusqu'à ce qu'on ne perçoive plus l'odeur de l'aci- de oenanthylique. On reprend le résidu par l'éther, on lave au moyen d'acide sulfurique 2 N, d'une solution de soude caustique 2 N, enfin à l'eau jusqu'à neutralité, on sèche sur du sulfate de sodium et on évapore. On provoque la cristallisation progres- sive du résidu huileux par agitation avec du pentane à froid.
Après recristallisation au sein de pentane, l'oenanhylate pur fond à 68-71 .
On obtient par une réaction analogue le 17-caprylate fondant à 43-46 et le 17-undécylate fondant à 61-62 . On peut, si l'on veut,, débarrasser d'une manière plus ou moins complète des impuretés très colorées les 3-énol-17-diesters bruts inter- médiaires par filtration sur alumine (Brockmann), ce qui pro- voque en outre une certaine hydrolyse des groupes 3-énol-ester On n'a pas tentée dans le cas de deux derniers esters mentionnés, d'effectuer la purification du 3-énol-17-diester par recristal- lisation.

On débarras. se la solution hydrogénée du catalyseur par filtration, on l'é- vapore sous vide, on reprend le résidu dans de l'éther, on lave successivement la solution éthérée avec àe l'acide sulfurique étendu, ensuite avec une solution étendue de bicarbonate de sodium puis à l'eau et on la sèche sur du sulfate de sodium.
Après évaporation de la solution éthérée il reste une huile qui cristallise au bout d'un certain temps. On effectue la purifica- tion par recristallisation répétée au sein d'hexane. L'ester pur fond à 9-100 .
On peut obtenir, à partir du composé 17-vinylique libre, l'ester correspondant par acétylation comme il/a été décrit ci- avant.
Par hydrogénation du n-butyrate de 17Ó -éthinyl-19-nor- testostérone dans des conditous analogues on obtient le compo- sé 17-vinylique correspondant, qui se présente à l'état brut sous forme d'une huile visqueuse.

The present invention makes it possible to obtain carboxylic esters of 17-alkyl-19-nor-testosterones. These new compounds show very interesting therapeutic properties and are particularly distinguished by an effect stimulating gestation which is exerted both by administration by the buccal route or by the hypodermic route. These esters having good solubility in ordinary solvents for steroid hormones, eg vegetable oils such as sesame oil, castor oil, cottonseed oil, sunflower oil, olive oil, 'olive, summer, .as well as in synthetic solvents such as glycols, esters of lactic acid, etc., their solutions can be injected and thus have depot hormones and obtain prolonged effects.
