avec la condition supplémentaire que le composé contient au moins un atome de carbone de structure gem-difluoro.
Les composés ci-dessus seront dénommés sim plement Qem-difluorostéroïdes car, en vertu de la définition ci-dessus, ils contiennent tous un ou deux atomes de carbone portant deux atomes de fluor. Les atomes de carbone qui peuvent être substi tués de cette manière sont ceux dans les positions 3, 17 ou 20.
Les gem-difluorostéroïdes de formule générale I sont des agents thérapeutiques utiles grâce à leur activité hormonale androgène-antiandrogène. Ce type d'activité a été décrit plus en détail par Dorfman & Dorfman dans Acta Endocrinologica, volume 33, page 308 (1960), et peut être observé par des tests endocriniens sur des animaux à sang chaud.

dans un solvant non aqueux inerte, comme le chloro forme, et on le mélange avec du tétrafluorure de soufre en présence d'une quantité catalytique de trifluorure de bore ou d'acide fluorhydrique.
Le tri- fluorure de bore peut être remplacé par d'autres acides de Lewis donnant des résultats semblables. Lorsque le catalyseur est l'acide fluorhydrique, ce dernier peut être ajouté directement comme tel, ou il peut être formé in situ par addition au mélange réactionnel d'une faible quantité d'un composé hy- droxylé, par exemple eau, méthanol, éthanol, isopro- panol, phénol, etc.
Lorsque le catalyseur est le tri- fluorure de bore ou un autre acide de Lewis, il est ajouté directement au mélange réactionnel.
La réaction désirée se déroule lentement à basse température et rapidement aux températures plus hautes. On a constaté qu'une température d'environ 400 C convient particulièrement pour la formation des nouveaux gem-difluorostéroïdes, mais que la température peut être comprise entre 0 et 600 C environ. Dans ces conditions, le groupe céto en posi tion 11 reste sensiblement non affecté, alors que les groupes céto dans les positions 3, 17 ou 20 sont transformés en groupes gem-difluoro.
On entend ici par solvant non aqueux inerte un solvant qui ne réagit ni avec l'une ou l'autre des matières de départ, ni avec le gem-difluorostéroïde formé dans le mélange réactionnel. Comme on l'a expliqué, les composés contenant un groupe hy droxyle sont donc exclus par cette définition, car ces solvants réagissent avec le tétrafluorure de soufre. Parmi les solvants appropriés pour la présente réac tion, on peut citer le tétrachlorure de carbone, le chloroforme, l'éther, le chlorure de méthylène, le benzène et d'autres liquides saturés ou aromatiques exempts de groupes hydroxyles et céto.
Lorsque la matière de départ de formule II con tient des groupes céto dans les positions 2 et 17, ces groupes céto sont tous deux remplaçables par le substituant gem-difluoro sans que le groupe 11 céto soit affecté dans une mesure appréciable.
Il en est également ainsi lorsque le stéroïde comporte en posi tion 17 un substituant contenant un groupe céto, comme par exemple dans le cas des pregnane- 3,11,20-triones. Lorsque la matière de départ con tient seulement un groupe céto en plus de celui en position 11, par exemple dans le cas de la 17-acé- toxy-androstane-3,11-dione, de la 3-ac6toxy-andros- tane-11,17-dione ou de la 3-acétoxy-pregnane-11,20- dione,
un seul groupe céto subit la transformation ci-dessus, et là encore, la position 11 n'est le siège d'aucune réaction appréciable. <I>Exemple 1</I> On chauffe à 40() C pendant 15 h une, solution de 3 g de 5a-pregnane-3,11,20-trione dans 30 ml de chloroforme contenant 3 % d'éthanol, avec 14 g de tétrafluorure de soufre.
Après avoir chassé l'excès de tétrafluorure de soufre par évaporation, on extrait le produit au chloroforme, on chromatographie les extraits chloroformiques réunis sur de l'alumine neutre d'activité III, on élue la colonne d'alumine avec un mélange benzène/Skellysolve B (1 :
10) (le Skellysolve B est un éther de pétrole bouillant à 60-800 C) et on obtient 545 mg d'une matière cris talline dont on tire 290g de 3,3,20,20-tétrafluoro-5- a-pregnan-11-one pure fondant à 170-1710 C après recristallisation dans un mélange benzène/Skellysolve B (1 :
10). Ce nouveau composé titre 66,83 % C, 7,90 % H et 19,94 % F, ce .qui corespond à la valeur calculée pour C21H3oOF4.
Dans l'exemple ci-dessus, le chloroforme peut être remplacé par du chlorure de méthylène auquel 0,3 g de trifluorure de bore est ajouté comme cata lyseur. Le résultat obtenu après les traitements dé crits ci-dessus est sensiblement le même.
<I>Exemple 2:</I> En poursuivant l'élution de la colonne d'alumine susmentionnée avec un mélange benzène-Skellysolve B (1 : 5), on obtient 1,36 g d'une matière cristalline qui, après recristallisation dans un mélange benzène- Skellysolve B (1 : 5), fournit 890 g de 3,3-difluoro- 5a-pregnane-11,20-dione pure fondant à 155-156o C.
L'analyse de ce produit donne 71,81 % C, 8,36 % H et 10,53 % F, ce qui correspond avec les valeurs calculées pour C.,
1H3o02F@.
<I>Exemple 3:</I> On chauffe pendant 15 h à 400 C une solution de 2 g de 5(3-pregnane-3,11,20-trione dans 20 ml de chloroforme contenant 3 % d'éthanol, avec 10 g de tétrafluorure de soufre.
Après évaporation, on extrait le résidu au chloroforme et on l'adsorbe sur de l'alu- mine neutre dans une colonne chromatographique. Une élution avec un mélange benzène-Skellysolve B (1 : 10) fournit 168 mg d'une matière cristalline blanche fondant à 162-1641, C. Par recristallisation de cette matière dans un mélange benzène-Skelly- solve B (1 : 10), on obtient 122 mg de 3,3,20,20- tétrafluoro-5(3-pregnan-11-one pure, fondant à 167 1680 C.
Un échantillon analytique de ce produit titre 67,58 % C, 8,03 % H et 20,77 % F, ce qui corres- pond aux valeurs calculées pour C21H3oOF4.
Des résultats analogues sont obtenus en rem plaçant le Skellysolve B par de l'hexane dans cet exemple.
<I>Exemple 4:</I> Une nouvelle élution de la colonne d'alumine de l'exemple précédent avec un mélange benzène/Skelly solve B (1 : 1) fournit 603 mg d'un solide cristallin fondant à 133-1390 C. Par recristallisation de cette matière dans un mélange benzène-Skellysolve B (1 : 1), on obtient 391 g de 3,3-difluoro-5(3-pregnane- 11,20-dione pure.
Un échantillon analytique titre 71,84 % C, 8,61 % H et 10,87 % F, ce qui corres- pond aux valeurs calculées pour C21H3,02F2.
<I>Exemple 5:</I> On chauffe à 40o C pendant 15 :h, dans un auto clave en acier inoxydable, une solution de 2,1 g de 5a-androstane-3,11,17-trione dans 20 ml de chloro- forme contenant 1 % d'acide fluorhydrique, avec <B>10,6</B> g de tétrafluorure de soufre. Ensuite, on chasse les produits gazeux et on lave le résidu avec cinq portions de chloroforme de 50 ml dans un entonnoir de séparation de 500 ml. On lave la solution chloro- formique avec deux portions de 100 ml d'eau, avec 100 ml d'une solution de bicarbonate de sodium à 5<B>%</B> et finalement avec deux portions de 100 ml d'eau.
On sèche la solution chloroformique sur du sulfate de magnésium anhydre et on la concentre sous vide partiel au bain-marie, ce qui laisse un résidu brun foncé. On extrait cette matière avec deux portions de 200 m1 d'éthanol bouillant, puis on traite les extraits réunis par du noir et on les filtre à tra vers de la Celite (une silice de diatomées amorphe, hydratée). On lave le gâteau avec 100 ml d'éthanol chaud et on réunit les eaux de lavage à la solution éthanolique précédente. On chasse le solvant au bain-marie sous vide partiel et on chromatographie le résidu sur une colonne d'alumine neutre.
Une élution avec un mélange benzène/hexane (1 :10) fournit la 3,3,17,17-tétrafluoro-5a-androstan-11-one. Les résultats d'analyse confirment la formule empi rique C,gH-EOF.1. <I>Exemple 6:</I> Une nouvelle élution de la colonne d'alumine utilisée pour la chromatographie de l'exemple 5, avec du benzène fournit, après le traitement usuel de la solution, la 3,3-difluoro-5c,.-androstane-11,17-dione sous forme de cristaux blancs.
Les résultats d'analyse confirment la formule empirique C19jI,602F_ <I>Exemple 7:</I> En procédant comme décrit dans l'exemple 5, on fluore la 5p-androstane-3,11,17-trione et on obtient la 3,3,17,17-tétrafluoro-5p-androstan-11-one par élu- tion de la colonne d'alumine avec un mélange ben- zène/hexane (1 : 10).
<I>Exemple 8:</I> Après élution de la 3,3,17,17-tétrafluoro-5(0-an- drostan-11-one dans l'exemple 7, on élue à nouveau la colonne d'alumine avec du benzène, dont on re tire la 3,3-difluoro-5p-androstane-11,17-dione après évaporation du solvant. <I>Exemple 9:</I> On chauffe dans un autoclave pendant 10h à 400 C un mélange de 2,2 g d7andros-4-ène-3,11,17- trione, 8,0 g de tétrafluorure de soufre, 0,4 g de tri- fluorure de bore et 20 ml de chloroforme exempt d'éthanol.
On laisse le mélange réactionnel se refroi dir jusqu'à la température ordinaire et on chasse les produits gazeux. On lave le résidu avec six portions de 50 ml de chloroforme dans un entonnoir de sépa ration de 500 ml. On abandonne le sous-produit rési neux noir. On lave la solution chloroformique avec deux portions de 100 ml d'eau, puis avec 100 ml d'une solution de bicarbonate de sodium à 5 Vo et finalement avec deux portions de 100 ml d'eau. On sèche la solution chloroformique sur du sulfate de magnésium anhydre et on chasse le chloroforme sous vide partiel au bain-marie, ce qui laisse un résidu cristallin foncé.
On chauffe cette matière avec 80 ml d'éthanol bouillant, on décolore la solution au noir et on la filtre à travers de la Celite. On lave la couche de noir et de Celite avec trois portions de 25 ml d'éthanol bouillant et on ajoute les eaux de lavage au filtrat éthanolique. On chasse l'éthanol sous vide partiel au bain-marie et il reste un précipité brun. On chromatographie cette matière sur 70 g d'alumine, on l'élue avec un mélange benzène/hexane (1 : 1) et on obtient un solide cristallin après éva poration de la solution.
Une recristallisation dans un mélange éthanol/eau fournit la 17,17-difluoro-andros- 4-ène-3,11.-dione. L'analyse est en accord avec les valeurs calculées pour CIq#H,,O?F2.
<I>Exemple 10:</I> Par hydrogénation de la 17,17-difluoro-andros-4- ène-3,11-dione de l'exemple 9 en présence de palla dium suivant la méthode décrite par Steiger et Reich- stein dans Helv. Chem. Acta, volume 20, pages 822 ss. (1937), on obtient la 17,17-difluoro-5a-andros- tane-3,11-dione en bon rendement.
<I>Exemple 11</I> En procédant comme décrit dans l'exemple 9, mais en remplaçant l'androst-4-ène-3,11,17-trion e par la pregn-4-ène-3,11,20-trione, on obtient la 20, 20-difluoro-pregne-4-ène-3,11-dione. L'analyse est en accord avec les valeurs calculées pour la formule brute C"H,,#O,F.2. <I>Exemple 12</I> En hydrogénant la 20,20-difluoro-pregn-4-ène- 3,11-dione en présence de palladium conformément à l'exemple 10, on obtient la 20,20-difluoro-5a-pre- anane-3,11-dione en bon rendement.
L'analyse est en accord avec les valeurs calculées pour G,H3o@,,F@ . <I>Exemple 13:</I> En faisant réagir de la 17(3-acétoxy-5(3-andros- tane-3,11-dione avec du tétrafluorure de soufre dans les conditions indiquées dans l'exemple 3, en éluant la colonne d'alumine avec du benzène-Skellylsolve B (1 :
1) et en évaporant l'éluat, on obtient la 17(3- acétoxy-3,3-difluoro-5(3-androstan-11-one. L'analyse est en accord avec les valeurs calculées pour la for mule brute C.;,H.,flO.;F@. <I>Exemple 14:</I> En remplaçant l'androst-4-ène-3,11,17-trione par la 3a-acétoxy-5(3-pregnane-11,20-dione dans l'exem ple 9, on obtient la 3a-acétoxy-20,20-difluoro-5(3- pregnan-11-one de formule C.,,3H,1Fz03.
<I>Exemple 15:</I> On hydrolyse la 3a-acétoxy-20,20-difluoro-5p- pregnan-11-one de l'exemple 14 avec de l'hydroxyde de potassium à 2 % dans du méChanol, on évapore le solvant et on lave le résidu à l'eau, obtenant ainsi la 3a-hydroxy 20,20-difluoro-5p-pregnan-11-one. Par oxydation à l'acide chromique en solution dans l'acide acétique à la température ordinaire pendant 4 h, et addition d'eau pour précipiter le produit,
on obtient la 20,20-difluoro-5(3-pregnane-3,11-dione de formule C2lHaoOgF2.
<I>Exemple 16:</I> En mélangeant une partie de 3a-acétoxy-5p-an drostane-11,17-dione avec 10 parties de chloroforme exempt d'éthanol, 4 parties de tétrafluorure de soufre et 0,2 partie de trifluorure de bore, et en traitant le mélange comme décrit dans l'exemple 9, on obtient la 3a-acétoxy-17,17-difluoro-5[3-androstan-11-one de formule brute q1H,O,,F2.
Dans les exemples ci-dessus, les structures attri buées concordent avec les spectres dans l'ultraviolet et dans l'infrarouge escomptés pour ces composés. Les points de fusion ont été déterminés dans un appareil à point de fusion Fisiher-Johns.

Gem-difluorosteroids of general formula I are useful therapeutic agents due to their hormonal androgen-antiandrogen activity. The desired reaction proceeds slowly at low temperature and rapidly at higher temperatures. It has been found that a temperature of about 400 ° C. is particularly suitable for the formation of new gem-difluorosteroids, but that the temperature may be between 0 and 600 ° C. approximately.
