Le procédé de l'invention s'applique en particu lier à la préparation des dérivés Ai de la cortisone et de l'hydrocortisone et des 21-esters de ceux-ci. Ces dérivés sont les 1,4-pregnadiène-17a,21-diol-3, 11,20-trione et 1,4-pregnadiène-11a,17a,21-triol- 3,20-dione. Ces dérivés et leurs 21-esters se sont montrés doués de propriétés hormonales cortico- surrénales inattendues.
Ils sont en effet plus actifs que la cortisone, l'hydrocortisone (composés E et F de Kendall) et les esters de celles-ci, tout en étant exempts, tout au moins dans une large mesure, des phénomènes secondaires indésirables qui accompa gnent fréquemment l'usage de la cortisone, de l'hy drocortisone et de leurs esters.

Le procédé de l'invention permet de préparer les composés I et II à partir de dérivés halogénés de pregnane-17a,21-diol-3,20-diones saturées, portant en position 11 un groupe céto ou hydroxyle a ou (3, lesquels dérivés sont facilement obtenables, ainsi qu'à partir des 21-esters desdits dérivés halogénés. Le noyau de ces dérivés présente soit la configura tion normale, soit la configuration allo.
Lorsque le procédé est appliqué à un 21-ester, on peut hydrolyser de manière connue le 21-ester de diène obtenu en alcool primaire correspondant (la et Ha), à l'aide d'un acide ou d'une base.
Dans le cas des composés 11-hydroxylés, et par ticulièrement des composés 11a-hydroxylés, on peut oxyder le groupe hydroxyle en groupe céto, avant ou après l'hydrolyse. Bien que l'on puisse utiliser des agents oxydants divers, on a constaté que l'acide chromique constitue un agent oxydant efficace et peu coûteux.
Les composés 2,4-dihalogénés préférés de départ du procédé de l'invention peuvent être préparés par halogénation en positions 2 et 4 des 21-esters de pregnane-17a,21-diol-3,11,20-trione et de pregnane- 11,17a,21-triol-3,20-dione, par traitement par un ou plusieurs halogènes de poids atomique supérieur à celui du fluor, et avec un rendement satisfaisant. L'halogénation dans les deux positions peut être effectuée en un ou deux temps et, dans ce dernier cas, on peut employer des halogènes différents tels que le chlore et le brome.
On traite ensuite le di- halogénure, conformément au procédé de l'invention, avec un agent de déshalohydratation pour introduire deux doubles liaisons. dans le noyau A, dans les posi tions 1(2) et 4(5), obtenant ainsi le composé lb ou IIb.
On préfère utiliser du brome comme agent ha- logénant. On peut ainsi obtenir facilement et avec un bon rendement les 21-esters de 2,4-dibromo- pregnane-17a,21-diol-3,11,20-trione et de 2,4-di- bromo-pregnane-11,17a,21-triol-3,20-dione.
On effectue la bromuration du dérivé du pre- gnane, qui est de préférence un 21 -ester, dans l'acide acétique ou dans un autre solvant sensiblement an- hydre et inerte à l'égard du brome. Il n'est pas néces saire de purifier le dibromodérivé ainsi obtenu avant de le faire réagir avec l'agent de débromhydrata- tion.
On préfère effectuer la débromhydratation par ébullition à reflux avec de la collidine, suivie de séparation chromatographique du 21-ester de 1,4- diène. On peut également utiliser en lieu et place de collidine d'autres bases organiques à point d'ébulli tion élevé, telles que la quinaldine et la diméthylani- line. On peut alors hydrolyser l'ester à deux doubles liaisons ainsi obtenu,
au moyen d'une base de métal alcalin, telle qu'un carbonate, un bicarbonate, un hydroxyde, de manière connue, dans un solvant neutre convenable tel que l'alcool méthylique ou éthylique.
La bromuration s'applique à la série de l'allo- pregnane aussi bien qu'à celle du pregnane normal. Ainsi, la dibromuration du 21-acétate d'allopre- gnane-17a,21-,diol-3,11,20-trione et du 21-acétate d'allopregnane-11(3,17a,21-triol-3,20-dione conduit aux dérivés dibromés de départ correspondants qui, par débromhydratation conformément au procédé de l'invention,
donnent les 21-acétates des diènes I et II, respectivement.
On peut préparer les diènes de stéroïdes lla- hydroxylés de la même façon. La dibromuration du 21-acétate de pregnane-11a,17a,21-triol-3,20-dione conduit au dérivé 2,4-dibromé de départ correspon- dant. Celui-ci donne par débromhydratation, con formément au procédé de l'invention, le 21-acétate de 1,4-pregnadiène-lla,17a,21-triol-3,
20-dione. On peut oxyder ce composé et obtenir ainsi le composé lb (R = acétyle) avec un bon rendement, ou l'hy drolyser en 1,4-pregnadiène-11a,17a,21-triol-3,20- dione qui, par oxydation, donne le composé la.
Comme mentionné, le procédé selon l'invention s'applique en particulier aux dérivés 2,4-dihalogénés de 21-esters (de préférence de l'acétate) d'allopre- gnane-17a,21-diol-3,11,20-trione.
Les exemples A et B ci-dessous montrent com ment on peut préparer ces esters de départ. <I>Exemple A</I> 21-acétate <I>de</I> 2,4-dibromo-allopregnane- 17a-21-diol-3,11,20-trione On hydrogène une solution de 2,00 g d'acétate de cortisone (21-acétate de 4-pregnène-17a,21-diol- 3,11,20-trione) dans 200 ml d'acétate d'éthyle, avec de l'hydrogène à pression atmosphérique et à tem pérature ordinaire, en utilisant comme catalyseur 0,20 g de palladium à 5 % sur du charbon de bois.
On laisse la réaction se poursuivre pendant la nuit, puis on sépare le catalyseur par filtration et l'on évapore le filtrat sous pression réduite. On cristal lise le résidu dans l'acétone et l'on obtient le 21- acétate d'allopregnane-17a,21-diol-3,11,20-trione. On bromure une solution de 1 g de 21-acétate d'allopregnane-17a, 21-diol-3,11, 20-trione dans 15 ml d'acide acétique par addition de 0,80 g de brome dans 10 ml d'acide acétique.
Une addition d'eau précipite le 21-acétate de 2,4-dibromo-allo- pregnane-17a,21-diol-3,11,20-trione.
<I>Exemple B</I> 21-acétate <I>de</I> 2,4-dibromo-allopregnane- 11(3,17a,21-triol-3,20-dione De manière semblable, on hydrogène 5,0g de 21-acétate du composé F de Kendall (21-acétate de 4-pregnène-11(3,17a,21-triol-3,20-dione) dans 750 ml d'acétate d'éthyle avec de l'hydrogène, en présence de 0,5 g de catalyseur à 5 % de palladium sur du charbon de bois, et l'on obtient le 21-acétate d'allo- pregnane-11(3,17a,21-triol-3,20-dione.
On bromure ce composé en procédant de la ma nière décrite dans l'exemple A.
Les exemples 1 à 5 ci-dessous montrent com ment le procédé de l'invention peut être mis en aeuvre <I>Exemple 1</I> Sans purification, on fait bouillir sous reflux le dibromure obtenu à l'exemple A (1,5 g) dans 50 ml de S-collidine symétrique pendant une heure. On ajoute du chloroforme et de l'eau, on lave l'extrait organique avec de l'acide sulfurique dilué, puis avec de l'eau, ensuite on sèche et l'on évapore.
On chro matographie le produit brut sur Florisil pour avoir le 21-acétate de 1,4-pregnadiène-17a,21-diol- 3,11, 20-trione <B>(lb,</B> R = acétyle), P. F. 232 - 235 C (déc.).
<I>Exemple 2</I> En procédant comme décrit dans l'exemple 1, on débromhydrate au moyen de collidine le dérivé 2,4Jdibromé obtenu par bromuration du 21-acétate de pregnane-17a,21-diol-3,11,20-trione avec 2 équi valents de brome, ce qui conduit au composé lb (R = acétyle).
<I>Exemple 3</I> De manière semblable, on débromhydrate avec de la collidine le dérivé 2,4-dibromé obtenu par bromuration du 21-acétate d'allo-pregnane-11p,17a, 21-triol-3,20-dione, pour obtenir le 21-acétate de 1,4-pregnadiène-11p,17a,21-triol-3,20-dione (com posé Ilb, R =acétyle) ; P.

<I>Exemple 5:</I> On prépare comme suit le composé de départ On bromure en position 2 et 4 une solution de 2,0g de 21-acétate de pregnane-11a,17a,21-triol- 3,20-dione dans 25 ml d'acide acétique par addition de 1,60 g de brome dans 15 ml d'acide acétique. On précipite par une addition d'eau le 21-acétate de 2,4 - dibromo - pregnane -11 a,17a,21-triol- 3,20-dione brut.
La déshydrobromuration avec 100 ml de colli- dine pendant 1 heure donne, après chromatogra phie, du 21-acétate de 1,4,pregrnadiène-11a,17a,21- triol-3,20Jdione.
L'hydrolyse avec du bicarbonate de potassium dans du méthanol aqueux pendant 10 minutes sous reflux donne la 1,4-pregnadiène-lla,17a,21-triol- 3,20-dione.
L'oxydation de l'acétate susmentionné avec Cr03,, la N-bromo-acétamide ou la N-bromosucci- nimide donne le composé<B>lb</B> (R =acétyle), alors que l'oxydation de la 1,4 pregnadiène-11a,17a,21- triol-3,20-dione avec de la N-bromoacétamide ou de la N-bromosucçinimide donne le composé la.
On donne ci-dessous encore deux exemples montrant comment on peut hydrolyser les 21-esters résultant du procédé selon l'invention en alcools correspondants.
<I>Exemple 1:</I> On fait bouillir pendant 10 min. à reflux un mélange de 1,0g de 21-acétate de 1,4-pregnadiène- 17a-21-diol-3,11,20-trione, 20 ml de méthanol, 5 ml d'eau et 0,3 g de bicarbonate de potassium, on neu tralise l'excès d'alcali avec de l'acide acétique et l'on chasse le méthanol sous pression réduite pour préci piter la 1,4-pregnadiène-17a,21-diol-3,11,20-trione (composé la) ; P.

The process of the invention applies in particular to the preparation of the Ai derivatives of cortisone and of hydrocortisone and of the 21-esters thereof. These derivatives are 1,4-pregnadiene-17a, 21-diol-3, 11,20-trione and 1,4-pregnadiene-11a, 17a, 21-triol-3,20-dione. These derivatives and their 21-esters have been shown to have unexpected cortico-adrenal hormonal properties.
They are in fact more active than cortisone, hydrocortisone (compounds E and F from Kendall) and the esters thereof, while being free, at least to a large extent, from the undesirable secondary phenomena which frequently accompany the use of cortisone, hy drocortisone and their esters.

The process of the invention makes it possible to prepare compounds I and II from halogenated derivatives of saturated pregnane-17a, 21-diol-3,20-diones, bearing in position 11 a keto or hydroxyl group a or (3, which derivatives are easily obtainable, as well as from the 21-esters of said halogenated derivatives.The nucleus of these derivatives has either the normal configuration or the allo configuration.
When the process is applied to a 21-ester, the obtained 21-ester diene can be hydrolyzed in a known manner to the corresponding primary alcohol (Ia and Ha), using an acid or a base.
In the case of 11-hydroxy compounds, and in particular 11α-hydroxy compounds, the hydroxyl group can be oxidized to a keto group, before or after hydrolysis. Although a variety of oxidizing agents can be used, chromic acid has been found to be an effective and inexpensive oxidizing agent.
Preferred 2,4-dihalogenated compounds starting from the process of the invention can be prepared by halogenation in positions 2 and 4 of the 21-esters of pregnane-17a, 21-diol-3,11,20-trione and pregnane-. 11,17a, 21-triol-3,20-dione, by treatment with one or more halogens of atomic weight greater than that of fluorine, and with a satisfactory yield. The halogenation in the two positions can be carried out in one or two stages and, in the latter case, different halogens such as chlorine and bromine can be employed.
The dihalide is then treated, in accordance with the process of the invention, with a dehalohydrating agent to introduce two double bonds. in ring A, in positions 1 (2) and 4 (5), thus obtaining compound lb or IIb.
It is preferred to use bromine as the halogenating agent. It is thus possible to obtain easily and with a good yield the 21-esters of 2,4-dibromopregane-17a, 21-diol-3,11,20-trione and of 2,4-di-bromo-pregnane-11, 17a, 21-triol-3,20-dione.
The bromination of the pregnan derivative, which is preferably an ester, is carried out in acetic acid or other solvent which is substantially anhydrous and inert to bromine. It is not necessary to purify the dibromoderate thus obtained before reacting it with the dehydrating agent.
It is preferred to carry out the dehydration by boiling under reflux with collidine, followed by chromatographic separation of the 21-1,4-diene ester. Other high boiling organic bases, such as quinaldine and dimethylanilin, can also be used instead of collidine. The ester with two double bonds thus obtained can then be hydrolyzed,
by means of an alkali metal base, such as a carbonate, a bicarbonate, a hydroxide, in known manner, in a suitable neutral solvent such as methyl or ethyl alcohol.
Bromination applies to the allopregane series as well as that of normal pregnane. Thus, the dibromination of 21-acetate of allopregnane-17a, 21-, diol-3,11,20-trione and of 21-acetate of allopregnane-11 (3,17a, 21-triol-3,20 -dione leads to the corresponding starting dibromo derivatives which, by debromhydration in accordance with the process of the invention,
give the 21-acetates of dienes I and II, respectively.
The lla-hydroxylated steroid dienes can be prepared in the same way. Dibromination of pregnane-11a, 17a, 21-triol-3,20-dione 21-acetate leads to the corresponding starting 2,4-dibromo derivative. This gives by dehydration, in accordance with the process of the invention, the 21-acetate of 1,4-pregnadiene-11a, 17a, 21-triol-3,
20-dione. This compound can be oxidized and thus obtain the compound Ib (R = acetyl) with a good yield, or hydrolyze it to 1,4-pregnadiene-11a, 17a, 21-triol-3,20-dione which, by oxidation , gives the compound la.
As mentioned, the process according to the invention applies in particular to 2,4-dihalogenated derivatives of 21-esters (preferably acetate) of allopregnane-17a, 21-diol-3,11,20 -trione.
Examples A and B below show how these starting esters can be prepared. <I> Example A </I> 21-acetate <I> de </I> 2,4-dibromo-allopregnane-17a-21-diol-3,11,20-trione A solution of 2.00 g is hydrogenated cortisone acetate (4-pregnene-17a 21-acetate, 21-diol- 3,11,20-trione) in 200 ml of ethyl acetate, with hydrogen at atmospheric pressure and temperature ordinary, using 0.20 g of 5% palladium on charcoal as a catalyst.
The reaction is allowed to continue overnight, then the catalyst is separated by filtration and the filtrate is evaporated under reduced pressure. The residue is crystallized from acetone to give 21-acetate of allopregnane-17a, 21-diol-3,11,20-trione. A solution of 1 g of 21-acetate of allopregnane-17a, 21-diol-3,11, 20-trione in 15 ml of acetic acid is bromined by adding 0.80 g of bromine in 10 ml of acid acetic.
Addition of water precipitates 2,4-dibromo-allopregane-17a, 21-diol-3,11,20-trione 21-acetate.

Examples 1 to 5 below show how the process of the invention can be carried out <I> Example 1 </I> Without purification, the dibromide obtained in Example A is boiled under reflux (1, 5 g) in 50 ml of symmetrical S-collidine for one hour. Chloroform and water are added, the organic extract is washed with dilute sulfuric acid, then with water, then dried and evaporated.
The crude product is chromatographed on Florisil to obtain 1,4-pregnadiene-17a 21-acetate, 21-diol-3,11, 20-trione <B> (lb, </B> R = acetyl), PF 232 - 235 C (dec.).
<I> Example 2 </I> By proceeding as described in Example 1, the 2,4Jdibromo derivative obtained by bromination of 21-acetate of pregnane-17a, 21-diol-3,11 is dehydrated by means of collidine, 20-trione with 2 equivalents of bromine, which leads to compound lb (R = acetyl).
<I> Example 3 </I> In a similar manner, the 2,4-dibromo derivative obtained by bromination of 21-acetate of allo-pregnane-11p, 17a, 21-triol-3,20 is dehydrated with collidine. -dione, to obtain 1,4-pregnadiene-11p, 17a, 21-triol-3,20-dione 21-acetate (compound Ilb, R = acetyl); P.

<I> Example 5: </I> The starting compound is prepared as follows: A solution of 2.0 g of pregnane 21-acetate-11a, 17a, 21-triol-3,20- in position 2 and 4 is bromined. dione in 25 ml of acetic acid by adding 1.60 g of bromine in 15 ml of acetic acid. The crude 2,4 - dibromo - pregnane -11α, 17a, 21-triol-3,20-dione 21-acetate is precipitated by adding water.
Dehydrobromination with 100 ml of collidine for 1 hour gives, after chromatography, 1,4, pregrnadiene-11a, 17a, 21-triol-3,20Jdione 21-acetate.
Hydrolysis with potassium bicarbonate in aqueous methanol for 10 minutes under reflux gives 1,4-pregnadiene-11a, 17a, 21-triol-3,20-dione.
Oxidation of the above-mentioned acetate with Cr03, N-bromo-acetamide or N-bromosuccinimide gives the compound <B> lb </B> (R = acetyl), while the oxidation of 1 , 4 pregnadiene-11a, 17a, 21-triol-3,20-dione with N-bromoacetamide or N-bromosuccinimide gives compound la.
Two more examples are given below showing how the 21-esters resulting from the process according to the invention can be hydrolyzed into corresponding alcohols.
