PROCEDE DE PREPARATION DE NOUVELLES DIPHENYLTHIOUREES SUBSTITUEES.
La présente invention concerne un procédé de préparation de bis-phényl-thiourée, dont les restes phényles sont substitués en position para, de formule:

Dans cette formule. les substituants R et R1. qui représentent chacun un reste n-batylique ou amylique, ou un reste alcoyl-oxy-alcoyle ayant en tout 4 ou 5 atomes de carbone et d'oxygènesont liés aux noyaux phényliques par un groupe méthylénique qui leur appartient.
Ces nouvelles thiourées sont obtenues par les procédés en euxmêmes connus de préparation de thiourées 1,3-bis-substituées.
C'est ainsi que, dans les composés des formules :

On peut donc faire réagir une phénylamine substituée en position para de manière appropriée, avec un phényl-isothiocyanate, un halogénure d'acide phényl-thiocarbamique, comme le chlorure, ou un ester d'un acide phénylthio- ou phényldithio-carbamique.

On peut aussi obtenir les thiourées substituées de façon symétrique, en faisant réagir les amines portant des substituants correspondants, avec le sulfure de carbones le thiophosgène ou la thiourée.
La transformation suivant l'invention des substituants transformables en R ou R1 en ces substituants, à lieu de manière en elle-même connue. Par exemple, un composé ayant un groupe hydroxy-alcoyle libre, peut être transformé en composé voulu, ayant un groupe alcoyloxy-alcoyle, par réaction avec un ester pouvant réagir de l'alcool correspondant, notamment d'acides organiques ou inorganiques forts, par exemple d'hydracides halogénés, tels que l'iodure d'éthyle ou de propyle, en présence d'alcalis.
Les produits initiaux sont connus ou peuvent être obtenus par des procédés en eux-mêmes connus. Ainsi, par exemple, les phényl-iso- thiocyanates substitués peuvent être préparés de manière connue par réaction de phénylamines avec le thiophosgène, l'isolement des isothiocyana- tes formés n'étant pas absolument nécessaire ; peut faire réagir ceux-ci directement avec les phénylamines correspondantes, pour obtenir les thiourées voulues.
Ces réactions peuvent être effectuées en présence ou en l'absence de diluants et/ou de catalyseurs, à température ordinaire ou à chaud.
Pour la réaction des phényliso-thiocyanates avec les anilines, des solvants neutres, comme par exemple l'éthanol, le benzène, le toluène ou le propanol sont appropriés.
Les nouveaux composés obtenus suivant la présente invention possèdent de précieuses propriétés thérapeutiques et sont actifs en particulier contre la tuberculose. G'est ainsi que, quelques-uns d'entre eux,

sont plus actifs et beaucoup moins toxiques que des agents antituberculeux connus, par exemple que les thiosemicarbazones.
Leur activité peut être déterminée, par exemple, par des essais sur des animaux,comme suit:
On inocule à des souris le myco-bactérium tuberculosis humanum (souche H 37 RV), de manière que 50% des animaux meurent en 20 jours après inoculation. On donne à de tels souris une nourriture à laquelle on a mélangé des quantités déterminées de la substance à essayer et cela pendant 30 jours,-après quoi on leur donne une nourriture normale. Si, par ce traitement, 50% ou plus des animaux sont encore en vie au bout de 35 jours.. on considère l'activité du produit utilisé comme bonne.
De tels résultats sont obtenus, par exemple, avec 1 à 2 mg de streptomycine par voie souscutanée pour 20 g de poids de l'animal, ou avec une nourriture contenant 0.5% d'acide p-aminosalicylique. Des résultats de tests repré-

La présente invention concerne également, à titre de produits industriels nouveaux, les produits conformes à ceux obtenus par le procédé défini ci-dessus.
Les exemples suivants illustrent l'invention sans la limiter aucunement. Entre chaque partie en poids et chaque partie en volume, il y a le même rapport qu'entre le gramme et le centimètre cube. Les températures sont indiquées en degrés centigrades.
Exemple 1.
On fait bouillir au réfrigérant à reflux, pendant 4 heures, 20 parties en poids de p-n-butyl-aniline dans 100 parties en volume d'éthanol, avec 16 parties en volume de sulfure de carbone et 2 parties en poids de xantogénate d'éthyle et de potassium. En refroidissant la masse réactionnelle, il cristalise la 1.3-bis-(p-n-butyl-phényl)- thiourée, de formule :

sous forme de lamelles qu'on filtre,, lave avec de l'alcool puis de l'eau., et qu'on recristallise dans de l'éthanol.
Elle fondent à 149 - 150 .

Dans la réaction décrite ci-dessus, au lieu d'employer 2 parties en poids de xantogénate d'éthyle et de potassium. on peut aussi employer 1 partie en poids d'hydroxyde de potassium.
Exemple 2.
On maintient pendant 4 heures à l'ébullition au réfrigérant à reflux, 22 parties en poids de p-isoamyl-aniline dans 100 parties en volume d'éthanol avec 16 parties en volume de sulfure de carbone et 2 parties en poids de xantogénate d'éthyl et de potassium. Après refroidissement,.on filtre les cristaux lammellaires qui se sont formés. on les lave avec de 1' alcool puis avec de l'eau et les recristallise dans de l'éthanol. On obtient ainsi la 1,3-bis-(p-isoamyl-phényl)- thiourée, de formule :

On maintient à l'ébullition pendant 4 heures, au réfrigérant à reflux, un mélange de p-amino-(ss-méthoxyéthyl)-benzéne (obtenu par hydrogénation de 27 parties en poids de p-nitro-(ss-méthoxyéthyl)-benzène dans 100 parties en volume d'éthanol avec de l'hydrogène. en présence de nickel-Raney, jusqu'à absorbtion de 3 mol-équivalents d'hydrogène et élimination du catalyseur par filtration). avec 2 parties en poids de xanthogénate d'éthyle et de potassium et 15,5 parties en volume de sulfure de carbone. On concentre la solution réactionnelle et laisse refroidir; la 1,3-bis-[ss-méthoxy-éthyl)phényl)-thiourée, de formule:

Le p-nitro- (P-méthoxyéthyl)-benzène employé dans cet exem- ple peut être préparé de la manière suivante :
A un mélange bien agité de 60 parties en volume d'acide sul- furique ( d =1,84) et de 60 parties en volume d'acide azotique (d = 1,42), refroidi à - 30 par l'extérieur, on ajoute lentement 45 parties en poids d' éther -phényléthyl-méthylique. Lorsque la réaction est terminée, on laisse la température monter à - 10 et ajoute un grand excès de glace. On essore ensuite le mélange partiellement cristallin, l'isomère ortho huileux étant séparé du p-nitro-(ss -méthoxy-éthyl)-benzéne cristallin. Ce dernier, après recristallisation dans de l'éther, fond à 60 - 61 .
On obtient de manière analogue le p-nitro-(Ó-méthoxy-propyl)benzène, fondant à 42 - 49 , en partant deÓ -méthoxy-propylbenzéne, bouillant à 205 - 210 , obtenu en faisant réagir du bromure dex-phénylpropyle avec du méthylate de sodium dans du méthanol.
Exemple 4.
A une suspension de 10,2 parties en poids de thiophosgène dans 100 parties en volume d'eau, on ajoute lentement, à 15 , en remuant soigneu- sement. 20 parties en poids de p-n-butyl-aniline dans 65 parties en volume de chloroforme. Lorsque l'addition-est terminée, on remue encore pendant 1/4 d'heure, sépare là couche chloroformique., extrait., puis distille le p-n-butyl-phényl-isothiocyanate brut qui passe à 150 - 154,5 sous une pres- sion de 14mm de mercure.
On dissout 2 parties en poids de cet isothiocyanate dans 5 parties en volume d'éthanol, puis on ajoute 1,7 partie en poids de p-n-amyl-aniline en solution dans 5 parties en volume d'éthanol. Lorsque la chaleur réaction- nelle qui se dégage spontanément commence à diminuer, on refroidit le mélan- ge réactionnel à température ordinaire, puis on essore la 1-(p-n-amyl-phényl)- 3-(p-n-butyl=phényl)-thiourée formée, de formule :

Exemple 11.
A 3,6 parties en volume d'éthyl-mercaptan, on ajoute 10 parties en poids de p-n-butyl-phényl-isothiocyanate et 50 parties en volume d'éthanol, on maintient la solution 1 heure à l'ébullition au réfrigérant à reflux, puis on isole, si on le désire., l'ester éthylique de l'acide p-n-butyl-phényl-dithiocarbamique formé, ou on le maintient 1 heure à l'ébullition, au réfrigérant à reflux, avec 7,8 parties en poids de p-n-butyl-anilineo On évapore ensuite à sec la solution réactionnelle puis cristallise dans de l'éthanol la 1.3-bis-(p-n-butyl-phényl)thiourée déjà décrite à l'exemple 1; elle fond 149 - 150 .
