PROCEDE POUR LA PREPARATION DU CHLOR AMPHENYCOL RACEMIQUE ET LEVOGYRE, A PARTIR DU 1-ACETOXY-3-OXO-3-PHENYLPROPANE.
On connait par la littérature et par des brevets antérieurs de la demanderesse des procédés pour la préparation du chloramphénycol racémi- que et lévogyre.
Suivant le procédé faisant l'objet de la présente invention, on peut préparer ces produits à partir du produit dénommé 1-acétoxy-3-oxo- 3-phényl-propane. On nitrose ce produit au moyen du nitrite sodique en so- lution acétique ou au moyen du nitrite alkylique en solution éthérée en pré- sence de HCL. On soumet l'isonitrose produit sans même l'avoir isolé, à une réduction sélective et à l'acylation, ou bien à.une acylation réductrice pour obtenir le l-acéthoxy-2-acétamino-3-oxo-3-phényl-propane.
On soumet ce dernier produit à une réduction en présence de ca- talyseurs, et l'on obtient un mélange de l-acétyl-2-acétamino-3-phényl-1-3- propanediol de forme tréo et de forme érythro. Sans séparer les constituants, on dissout ce mélange dans l'acide sulfurique concentré et l'on obtient, l'a- cétoxy-oxazolidine correspondante en solution sulfurique. On soumet cette dernière, sans qu'il soit nécessaire de l'isoler, à la nitrition au moyen de l'acide nitrique concentré et l'on obtient, en définitive, à partir du mé- lange de nitration, une solution du chlorhydrate de 1-p-nitro-phényl-2-ami- no-1-3-propanediol de forme thréo.
On peut isoler à partir de ce chlorhydra- te la base et la séparer en ses antipodes optiques, suivant le procédé fai- sant l'objet de notre demande de brevet ? 393.398. La solution du chlorhy- drate de racémate ci-dessus mentionnée ou bien la solution du chlorhydrate de la base lévogyre, séparée comme il est dit ci-dessus,peuvent être conver- ties, respectivement, en chloramphénycol racémique ou'bien en chloramphény- col lévogyre en substituant l'hydrogène dans le groupe aminé par le radical dichlor-acétique.

EXEMPLE:
On traite 1113 grammes de 1-acétoxy-3-oxo-phényl-propane (ZB1.1933-II.1186) en solution dans 840 centimètres cubes d'acide acétique à 30/35 par 450 gr. de nitrite sodique très finement divisé. A la fin de la réaction l'on ajoute'1,450 kilos de glace et 1,160 kilos d'anhydride acé- tique. En remuant activement on ajoute encore 1,450 kilos de glace, 0,580 kilos d'anhydride acétique et 1 kilo de zinc, en maintenant la température au dessous de 20 .
A la fin de la réaction on filtre le mélange.
On dilue le résidu sur le filtre plusieurs fois par de lacé- tone et on concentre les extraits acétoniques jusqu'à consistence pâteuse.
On concentre la solution filtrée sous vide jusqu'à également consistence pâteuse, on réunit les deux résidus, on dilue plusieurs fois par de l'eau et on fait recristalliser à partir de l'alcohol la partie insoluble dans l'eau. On obtient ainsi le l-acétoxy-2-acétamino-3-oxo-3-phényl-propane.
Point de fusion 84 -85 - C.
On dissout le produit désigné ci-dessus dans 10 parties d'al- cool méthylique, on ajoute 2 parties de catalyseur palladium-carbone à 5% de palladium et on réduit à la pression et la température ordinaires jusqu'à l'absorption de 10% en plus de la quantité d'hydrogène théoriquement néces- saire. On filtre pour séparer le catalyseur et on distille sous vide l'al- cool méthylique. Le résidu obtenu est un mélange de 1-acétoxy-2-acétamino- 3-phényl-l- 3-propanédiol de forme tréo et érytro; on le dissout conformé- ment à la demande dans trois fois son poids d'acide sulfurique concentré, et on laisse la solution reposer à la température normale pendant 12 heures.
On verse cette solution à une température de -15 C environ dans trois parties d'acide nitrique concentré, densité 1,42. On, la laisse pendant une heure environ à cette température et pendant une demie-heure à + 15 C.
On verse ensuite sur la glace. On extrait complètement par l'acétate d'éthyle après avoir neutralisé par du carbonate sodique. On évapore à sec et on dis- tille l'extrait acétique. On dilue le résidu avec 10 parties d'acide chlorhy- drique à 5% et on chauffe au bain marie pendant deux heures. On concentre sous vide à un faible volume, on ajoute une partie égale d'acide chlorhydri- que concentré et on laisse cristalliser le chlorhydrate du 1-nitro-phényl-2- aminol-1-3-propanédiol. Point de fusion 177-180 .
On peut isoler de ce chlorhydrate la base, que l'on peut sépa- rer en ses deux antipodes suivant le procédé faisant l'objet de la demande.
On suspend dans l'eau le chlorhydrate de la base racémique ou bien le chlorhydrate de la nitrobase lévogyre; on les traite en remuant éner- giquement par un faible excès de bicarbonate de sodium et par la quantité stoichométrique de chlorure de l'acide dichoracétique.
A l'issue de la réaction on sépare respectivement le chloramphé- nycol racémique ou le chloramphénycol lévogyre mélangés à des sels inorgani- ques.
On filtre et on fait recristalliser à partir de l'eau les pro- duits séparés, que l'on obtient ainsi à un état de pureté chimique suffisan- te.

Processes for the preparation of racemic and levorotatory chloramphenycol are known from the literature and from the prior patents of the applicant.
According to the process forming the subject of the present invention, these products can be prepared from the product called 1-acetoxy-3-oxo-3-phenyl-propane. This product is nitrided by means of sodium nitrite in acetic solution or by means of alkyl nitrite in ethereal solution in the presence of HCL. The isonitrosis produced without even having isolated it is subjected to a selective reduction and to acylation, or else to a reductive acylation to obtain 1-acethoxy-2-acetamino-3-oxo-3-phenyl-propane .
The latter product is subjected to reduction in the presence of catalysts, and a mixture of 1-acetyl-2-acetamino-3-phenyl-1-3-propanediol in the treo form and in the erythro form is obtained. Without separating the constituents, this mixture is dissolved in concentrated sulfuric acid and the corresponding acetoxy-oxazolidine is obtained in sulfuric solution. The latter is subjected, without it being necessary to isolate it, to nitration by means of concentrated nitric acid and, finally, from the nitration mixture, a solution of hydrochloride is obtained. 1-p-nitro-phenyl-2-amino-1-3-propanediol in threo form.
We can isolate the base from this hydrochloride and separate it into its optical antipodes, according to the process which is the subject of our patent application? 393,398. The hydrochloride of the racemic base or the hydrochloride of the levorotatory nitrobase is suspended in water; they are treated with vigorous stirring with a slight excess of sodium bicarbonate and with the stoichometric amount of dichoracetic acid chloride.
At the end of the reaction, the racemic chloramphenycol or the levorotatory chloramphenycol, mixed with inorganic salts, are respectively separated.
The separated products are filtered and recrystallized from water, thereby obtaining a sufficient chemical purity.
Levorotatory chloramphenycol - white crystals melting point 150/151.
