PROCEDE DE PREPARATION DE NOUVELLES DIPHENYL-THIOUREES SUBSTITUEES.
La présente invention concerne un procédé de préparation de bis- phényl-thiourées dans lesquelles les restes phényles sont substitués en po- sition para, de formule :

Dans cette formule R représente un reste alcoyle ayant 2 à 5 atomes de carbo- ne et R1 le reste d'un hydrocarbure aliphatique ayant 3 à 6 atomes de carbone, ou un reste correspondant interrompu par de l'oxygène, ayant 3 à 6 ato- mes de carbone.
On obtient ces nouvelles thiourées par des procédés en eux-mêmes connus de préparation .n de thiourées 1,3-bis-substituées.

En conséquences on peut faire réagir, par exemple, une phénylami- ne para-substituée de manière appropriée avec un phénylisothiocyanate, un halogénure d'acide phéhylthiocarbamique, comme le chlorure, ou un ester d'un acide phénylthio- ou phényldithocarbmique, par exemple un ester alcoyli- que, entre autres un ester éthylique.
La transformation, suivant le procédé, de substituants transfor- mables en un groupe hydroxyle substitué, comme le groupe hydroxyle, en res- tes définis ci-dessusp a lieu d'après des méthodes en elles-mêmes connues.
C'est ainsi, par exemple? qu'un composé avec des groupes hydroxyles libres, se laisse transformer en composé souhaité, ayant des groupes hydroxyles sub- stitués, par réaction avec un ester réactif de l'alcool correspondant, en' particulier un ester diacides minéraux ou organiques farta, comme les hydra- cides halogénés, par exemple avec l'iodure d'éthyle, de propyle ou de butyle, en présence d'alcalis.
Les matières de départ sont connues ou peuvent être obtenues par des procédés en eux-mêmes connus. Ainsi, par exemple,, les phénylisothiocya- nates substitués se laissent préparer, d'une manière connue, par réaction de phénylamines sur le thiophosgéne, sans qu'il soit absolument nécessaire d'i- soler l'isothiocyanate formé; on peut faire réagir celui-ci directement avec les phénylamines correspondantes pour obtenir les thiourées souhaitées.
On peut effectuer ces réactions en présence ou en l'absence d'a- gents de dilution et/ou de catalyseurs, à la température ordinaire ou à chaudo Pour la réaction des phénylisothiocyanates sur les anilines, des sol- vants neutres, par exemple l'éthanol, le benzène, le toluène ou le propanol sont appropriés
Les nouveaux composés obtenus suivant la présente invention pré- sentent de précieuses propriétés thérapeutiques et sont actifs en particulier contre la tuberculoseo C'est ainsi que quelques-uns d'entre eux, en particu- lier la 1-(par-éthoxyphényl)-3-(para-buthl-phényl)-thiourée, de formule:

Leur activité peut être déterminée par des essais sur des ani- maux effectués comme suit :
On inocule à des souris le myco-bacterium tuberculosis hamanum (souche H 37 RV), de manière que 50 % des animaux meurent en 20 jours après inoculation. On donne à de telles souris une nourriture à laquelle on a in- corporé des quantités déterminées de la substance à essayer, et celà pen- dant 30 jours, après quoi l'on utilise une nourriture normale. Si, par ce traitement, 50 % ou plus des animaux sontencore en vie après 35 jours, on considère comme bonne l'activité du produit utilisé.
On obtient, par exem- ple, de tels résultats avec 1 à 2 mg de streptomycine, administrée par voie sous-cutanée, pour 20 grammes de poids de l'animal, ou avec une nourriture contenant 0.5% d'acide para-amino-salicylique.
Des résultats de test représentatifs faits avec les nouveaux composés, dont quelques-uns ne provoquent aucun phénomène toxique chez les animaux, même à une-concentration de 3 % dans la nourrirure, sont les sui- vants :
