Die Erfindung betrifft ein Verfahren zur Herstellung von neuen Piperazinderivaten der allgemeinen Formel

Die Niedrigalkyl-, Niedrigalkoxy- und Niedrigalkenylgruppen der oben angegebenen allgemeinen Formel (1) können verzweigt oder nicht-verzweigt sein und bis zu 6 Kohlenstoffatome enthalten. Die Cycloalkylgruppen enthalten 3 bis 7 Kohlenstoffatome im Ring, der auch einen Niedrigalkylsubstituenten aufweisen kann. Die Carboalkoxygruppen enthalten Alkylgruppen mit 1 bis 5 Kohlenstoffatomen und Beispiele dafür sind Carbomethoxy, Carboäthoxy, Carbopropoxy, Carbobutoxy u. dgl.
Zweckmässig bedeuten R und R'Niedrigalkyl, vorzugsweise Methyl, oder Halogen, vorzugsweise Chlor, R kann ein Wasserstoffatom bedeuten und R'bedeutet dann vorzugsweise Chlor, Niedrigalkyl, vorzugsweise Methyl, oder Trifluormethyl, R" bedeutet vorzugsweise Niedrigalkyl, insbesondere Isobutyl, und R"' bedeutet zweckmässig Carbäthoxy.
Nach dem erfindungsgemässen Verfahren werden die neuen Piperazine der oben angegebenen allgemeinen Formel (1) dadurch hergestellt, dass man ein substituiertes Benzimidoylchlorid der allgemeinen Formel

Salze von Verbindungen der allgemeinen Formel (I) fallen gelegentlich unmittelbar an und können in die freien Basen übergeführt werden, die in einem inerten Lösungsmittel mit der gewünschten Menge einer an-

stellung dieser Salze geeignete Säuren sind Chlorwasserstoffsäure, Bromwasserstoffsäure, Phosphorsäure, Schwefelsäure, Benzoesäure, Mandelsäure, Zimtsäure, Essigsäure, Propionsäure, Milchsäure, Citronensäure, Weinsäure, Apfelsäure, Malonsäure, Bernsteinsäure, Maleinsäure und Fumarsäure.
Die Erfindung wird durch die folgenden Beispiele näher erläutert, ohne jedoch darauf beschränkt zu sein.
Beispiel1 :N1-Methyl-N4-[3-methyl-(N-isobutyliminobenzyl)]-piperazindihydroclorid
Zu einer Lösung von 5,0 g (0,05 Mol) N-Methylpiperazin und 5,1 g (0,05 Mol) Triäthylamin in 100 ml trockenem Benzol wurden über einen Zeitraum von 15 min 10, 4 g (0, 05 Mol) 3-Methyl-N-isobutylbenzimidoyl- chlorid gegeben. Die Reaktionsmischung wurde dann weitere 2 h bei Raumtemperatur gerührt. Nachdem das Triäthylaminhydrochlorid durch Filtrieren entfernt worden war, erhielt man nach dem Einengen der Lösung einen gelben öligen Rückstand, der anschliessend mit einer verdünnten wässerigen Natriumhydroxydlösung behandelt und dann mit 250 ml Äther extrahiert wurde.
Nach dem Einengen der getrockneten Ätherschicht und nach der Destillation des Rückstandes erhielt man N1-Methyl-N4.. [3-methyl (N-isobutyliminobenzyl) ]piperazin mit Kp. =112 bis 114 C (0,4 mm) in 50%iger Ausbeute (8, 2 g). Nach dem Einleiten von Hel-Gas in eine ätherische Lösung der Base erhielt man das entsprechende Dihydrochloridsalz. Nach dem Umkristallisieren aus Acetonitril erhielt man 8, 9 g (57%) Produkt, Fp. = 235 bis 2400C.
Beispiel2 :N1-Carboäthoxy-N4-[3-methyl-(N-isobutyliminobenzyl)]-piperazinhydrochlorid
Zu einer Lösung von 7,5 g (0, 05 Mol) Carbäthoxypiperazin in 100 ml trockenem Benzol wurden über einen Zeitraum von 15 min 11,5 g (0, 05 Mol) 3-Methyl-N-isobutylbenzimidoylchlorid zugegeben. Die Reaktionsmischung wurde 1 h unter Rückfluss erhitzt und dann gekühlt. Nach dem Filtrieren erhielt man 14,3 g

Nach zweimaligem Umkristallisieren ausAceton/ÄtherZu einer Lösung von 6, 1 g (0, 024 Mol) N-Benzhydrylpiperazin in 120 ml trockenem Benzol wurden über einen Zeitraum von 15 min 5, 6 g (0,024 Mol) N-Cyclohexylbenzimidoylchlorid zugegeben. Die Reaktionsmi- schung wurde dann weitere 8 h unter Rückfluss erhitzt.
Nach dem Einengen des Lösungsmittels erhielt man einen gelben öligen Rückstand, der mit einer verdünnten Natriumhydroxydlösung behandelt und dann mit
300 ml Chloroform extrahiert wurde. Nach dem Einengen der getrockneten Chloroformschicht erhielt man einen gelben Feststoff. Nach zweimaligem Umkristallisieren des Feststoffes aus Acetonitril erhielt man 5, 0 g (48%) eines weissen Stoffes mit Fp. = 155 bis 1560C.
Beispiel4 :N1-(n-Hexyl)-N4-(N-isobutyliminobenzyl)piperazin-dimaleat
Zu einer Lösung von 13, 6 g (0, 08 Mol) n-Hexylpiperazin und 8, 1 g (11, 2 ml) (0,08 Mol) Triäthylamin in
120ml Toluol wurden über einen Zeitraum von 15 min 15, 6 g (0, 08 Mol) N-Isobutylbenzimidoylchlorid zuge- geben. Die Reaktionsmischung wurde 2 h bei Raumtemperatur gerührt. Das Triäthylaminhydrochlorid wurde abfiltriert und das Filtrat wurde unter Vakuum eingeengt. Die zurückbleibende ölige Base wurde mit 20 ml
Wasser gewaschen, mit 150 ml Äther extrahiert und über wasserfreiem Magnesiumsulfat getrocknet. Die trockene ätherische Lösung wurde dann zu einer Lösung von 18, 6 g (0, 16 Mol) Maleinsäure in 800 ml Äther gegeben, um das Dimaleat herzustellen.
Nach zweimaligem Umkristallisieren aus Äthanol erhielt man 22, 5 g (50, 2%) Produkt, Fp. 175 bis 176 C.
Nach den oben und in den Beispielen beschriebenen Verfahren wurden die in der folgenden Tabelle I an- gegebenen Verbindungen hergestellt.

Die erfindungsgemäss herstellbaren Verbindungen stellen starke hypoglykämische Mittel dar, die bei mit Glucose vorbehandelten Ratten bei ihrer Verabreichung in einer Dosis von 100 mg/kg p. o. eine Senkung des Blutglucose-Spiegels um bis zu 48% bewirken. Sie eignen sich daher zur Behandlung von Diabetes. In der folgenden Tabelle n sind die Aktivitäten von bevorzugten, erfindungsgemäss herstellbaren Verbindungen angegeben.
