본 발명은 발모 촉진 또는 탈모증의 방지 또는 억제 활성을 갖는 펩타이드에 관한 것이다. 본 발명의 펩타이드는 모낭 모유두세포(HFDPC) 및 인간외모근초세포(HHORSC)의 증식을 촉진하고, 모낭 모유두세포(HFDPC)에서 베타-카테닌 하위인자의 발현을 상향 조절하고, 성장인자의 발현을 상향조절한다.
Description
본 발명은 발모 촉진 및 탈모 억제 활성을 갖는 펩타이드 및 이의 용도에 관한 것이다.
탈모증이란 모발 성장 주기에서 정상보다 휴지기가 길어지거나 휴지기에 있는 모낭의 수가 증가하여 모발 탈락이 과도하게 일어나는 현상으로 스트레스, 영양부족, 국소 혈류 장애, 호르몬 및 유전적 요인 등에 의해 모발이 과도하게 탈락되어 복구되지 않는 것을 지칭한다.
탈모 치료제로는 남성호르몬인 테스토스테론을 안드로겐 성호르몬인 디하이드로테스토스테론(DHT)으로 변환시키는 작용을 하는 5α-환원효소(reductase) 억제제, 예를 들어, 피나스테리드(finasteride) 또는 두타스테리드(dutasteride)와 같은 약물이 개발되어 시판이 허가되어 있으며, 모낭을 자극하고 혈류를 증가시켜 발모를 촉진하는 작용을 갖는 약물인 미녹시딜(minoxidil)도 허가되어 사용되고 있다.
그러나, 5α-환원효소 억제제의 경우 성기능 장애, 우울증, 불안, 가슴비대와 같은 부작용이 비교적 빈번하게 발생된다고 보고되어 있고, 미녹시딜의 경우도 심전도 이상, 빈맥, 심내막염과 같은 심혈관계 부작용이 발생되는 것으로 보고되어 있으며, 또한 미녹시딜은 약물 복용을 중단하면 다시 탈모가 시작되는 것으로 알려져 있다.
펩타이드는 생체친화성이 높으며, 약물 또는 화장료에서 단일의 활성성분으로 사용되면 다양한 성분이 함유된 추출물 등에 비해 부작용이 적고, 국소적으로 탈모 억제 작용을 한 이후 체내의 분해 단백질에 의해 쉽게 분해되어 제거될 수 있으므로 기존 치료제와는 달리 부작용에 대한 우려가 적다.
PCT 국제특허 WO2005-082395에는 발모 주기에서 휴지 단계로부터 성장 단계로의 진행을 촉진시켜 모발 성장을 촉진시키는 효과를 가진 펩타이드가 개시되어 있고, 대한민국 등록특허 제10-2387764호에는 모낭세포의 증식을 촉진시키는 작용을 갖는 이합체 형태의 펩타이드 유도체가 개시되어 있다.

본 발명자들은 발모 촉진에 효과적이면서 부작용이 적은 발모 촉진 및 탈모 억제 활성을 갖는 보다 개선된 활성의 펩타이드를 개발하기 위해 연구 노력하였다. 그 결과, 본 발명자들이 개발한 신규 펩타이드가 인간 모낭모유두 세포(HHFDPC)와 외모근초세포(HHORSC)의 증식을 촉진하고, 모유두 세포에서 베타-카테닌 하위인자 LEF-1 및 Cyclin D1의 발현수준을 향상시키고, 성장인자인 bFGF, HGF, VEGF의 발현 수준을 증가시키는 것을 실험적으로 증명하여 본 발명을 완성하게 되었다.
따라서, 본 발명의 목적은 발모 촉진 및 탈모 억제 활성을 갖는 신규 펩타이드를 제공하는 것이다.
본 발명의 다른 목적은 상술한 활성을 갖는 펩타이드를 유효성분으로 포함하는 발모 촉진 또는 탈모증의 억제, 예방 또는 개선용 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 탈모증의 예방 또는 치료용 약학적 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 발모 촉진 또는 탈모증의 예방 또는 개선용 화장료 조성물을 제공하는 것이다.
상기의 목적을 달성하기 위하여,
본 발명의 일 측면은 서열번호 1의 아미노산 서열을 포함하는 펩타이드를 제공한다.

이하에서 본 발명을 상세하게 설명한다.
1. 펩타이드 및 이의 활성
본 발명의 일 측면에 따르면, 서열번호 1에 개시된 아미노산 서열을 포함하는 펩타이드를 제공한다.
본 명세서 용어 "펩타이드"는 펩타이드 결합에 의해 아미노산 잔기들이 서로 결합되어 형성된 선형의 분자를 의미한다.
본 발명의 서열번호 1의 아미노산 서열을 포함하는 펩타이드는 변형 없이 사용될 수 있으나, 상기 펩타이드가 가지는 본래의 활성, 예컨대 발모 촉진 또는 탈모증의 억제 활성에 영향을 미치지 않는 범위 내에서, 아미노산 잔기의 결실, 삽입, 치환, 또는 이들의 조합에 의해서 상이한 서열을 가지는 아미노산의 변이체 또는 단편을 사용할 수 있다.
본 발명의 펩타이드는 그 활성을 변화시키지 않는 범위내에서 인산화(phosphorylation), 황화(sulfation), 아크릴화(acrylation), 당화(glycosylation), 메틸화(methylation), 파네실화(farnesylation) 등으로 변형될 수 있다.
본 발명의 펩타이드는 서열번호 1의 아미노산 서열을 포함하는 펩타이드와 실질적으로 동일한 아미노산 서열을 포함하는 펩타이드 및 이의 변이체 또는 이의 활성 단편을 포함한다. 상기 실질적으로 동일한 아미노산 서열이란 상기 서열번호 1의 아미노산 서열과 각각 75% 이상, 예컨대 80% 이상, 85% 이상, 90% 이상, 95% 이상, 97% 이상의 서열 동일성을 가지는 아미노산 서열을 의미한다. 또한, 상기 펩타이드에는 표적화 서열, 태그(tag), 표지된 잔기, 반감기 또는 펩타이드 안정성을 증가시키기 위한 특정 목적으로 제조된 아미노산 서열을 추가적으로 포함할 수 있다.
본 발명의 펩타이드는 아미노산 서열의 일부 부위를 선정하고 그 활성을 증가시키기 위해 N-말단 및/또는 C-말단 변형이 유도된 것일 수 있다. 이러한 N-말단 및/또는 C-말단 변형을 통해 본 발명의 펩타이드의 안정성을 현저하게 향상시킬 수 있으며, 예를 들어, 펩타이드의 생체 내 투여 시 반감기를 증가시킬 수 있다. 상기 용어 "안정성"은 생체 내 단백질 절단 효소의 공격으로부터 본 발명의 펩타이드를 보호하는 인 비보에서의 안정성 뿐만 아니라, 저장안정성(예컨대, 상온 저장 안정성)도 포함하는 의미이다.

본 발명에서 탈모는 유전성인 안드로겐성 탈모(남성형 탈모), 여성형 탈모, 휴지기 탈모(telogen effluvium), 성장기 탈모(anagen effluvium), 원형 탈모증(alopecia areata), 곰팡이 감염에 의한 두부 백선, 갑상선 기능 저하증 또는 갑상선 기능 항진에 의한 탈모, 염증(지루성 두피염)에 의한 탈모, 영양실조에 의한 탈모, 당뇨에 의한 탈모, 루푸스에 의한 탈모, 모발 생성 장애 질환에 의한 탈모, 약물(항응고제, 항우울제, 경구 피임약의 중단, 화학요법)에 의한 탈모, 독발성 모낭염, 모공성 편평 태선, 화상 또는 외상에 의한 탈모, 자외선에 의한 탈모 또는 공기 중의 미세먼지 접촉에 의한 탈모을 포함하나, 이에 한정되는 것은 아니다.

본 발명의 약학적 조성물은 약학적으로 허용되는 담체를 포함할 수 있다.

본 발명의 약학적 조성물은 상기 성분들 이외에 윤활제, 습윤제, 감미제, 향미제, 유화제, 현탁제, 보존제 등을 추가로 포함할 수 있으나, 이에 한정되는 것은 아니다.
적합한 약학적으로 허용되는 담체 및 제제는 Remington: The Science and Practice of Pharmacy, (19th ed., 1995, Williams & Wilkins)에 상세히 기재되어 있다.
본 발명의 약학적 조성물은 탈모증을 치료하기 위한 적합한 모든 경로로 투여될 수 있으며, 예를 들어, 경구 또는 비경구로 투여할 수 있고, 비경구 투여인 경우에는 정맥내 주입, 피하 주입, 근육내 주입, 복강내 주입, 국소 투여, 경피 투여 등으로 투여할 수 있다. 본 발명의 약학적 조성물이 탈모증의 예방 또는 치료 활성을 갖는 것이므로, 두피에 적용하는 것과 같은 국소 투여로 적용될 수 있다.
상기 약학적 조성물의 투여량은 1일당 0.0001 μg 내지 100 mg, 0.001 μg 내지 100 mg, 0.01 μg 내지 100 mg, 0.1 μg 내지 100 mg, 또는 1.0 μg 내지 1000 mg 일 수 있으나, 이에 한정되는 것은 아니며, 제제화 방법, 투여 방식, 환자의 연령, 체중, 성, 병적 상태, 음식, 투여 시간, 투여 경로, 배설 속도 및 반응 감응성과 같은 요인들에 의해 다양하게 처방될 수 있다.
본 발명의 약학적 조성물은 당해 발명이 속하는 기술분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있는 방법에 따라, 약제학적으로 허용되는 담체 및/또는 부형제를 이용하여 제제화함으로써 단위 용량 형태로 제조되거나 또는 다용량 용기 내에 내입시켜 제조될 수 있다. 이 때 제형은 오일 또는 수성 매질중의 용액, 현탁액 또는 유화액 형태이거나 엑스제, 분말제, 과립제, 정제 또는 캅셀제 형태일 수도 있으며, 분산제 또는 안정화제를 추가적으로 포함할 수 있다.
본 발명의 약학적 조성물은 피부외용제일 수 있다. 상기 피부외용제는 피부의 외부에 도포되어 이용될 수 있는 형태의 제제로, 본 발명의 약학적 조성물이 피부외용제로 이용될 경우 이는 두피, 구체적으로 탈모가 발생한 부위의 두피나 발모를 촉진하고자 하는 부위의 두피에 적용되는 것일 수 있다. 상기 피부외용제는 크림, 겔, 연고, 피부 유화제, 피부 현탁액, 경피전달성 패치, 약물 함유 붕대, 로션, 또는 그 조합일 수 있고. 상기 피부외용제는 통상 화장품이나 의약품 등의 피부외용제에 사용되는 성분, 예를 들어 수성성분, 유성성분, 분말성분, 알코올류, 보습제, 중점제, 자외선흡수제, 미백제, 방부제, 산화방지제, 계면활성제, 향료, 색제, 각종 피부 영양제, 또는 이들의 조합과 필요에 따라서 적절하게 배합될 수 있다. 상기 피부외용제는, 에데트산이나트륨, 에데트산삼나트륨, 시트르산나트륨, 폴리인산나트륨, 메타인산나트륨, 글루콘산 등의 금속봉쇄제, 카페인, 탄닌, 벨라파밀, 감초추출물, 글라블리딘, 칼린의 과실의 열수추출물, 각종 생약, 아세트산토코페롤, 글리틸리틴산, 트라넥삼산 및 그 유도체 또는 그 염등의 약제, 비타민 C, 아스코르브산 인산 마그네슘, 아스코르브산글루코시드, 알부틴, 코지산, 글루코스, 프룩토스, 트레할로스 등의 당류 등도 적절하게 배합할 수 있다.
본 발명의 다른 측면에서, 본 발명의 조성물은 서열번호 1의 아미노산 서열을 포함하는 펩타이드를 유효성분으로 포함하는 발모 촉진 또는 탈모증의 예방 또는 개선용 화장료 조성물일 수 있다.

상기 화장료 조성물은 유연 화장수, 영양 화장수, 영양 크림, 마사지 크림, 에센스, 아이 크림, 클렌징 크림, 클렌징 폼, 클렌징 워터, 팩, 스프레이, 파우더, 헤어토닉, 헤어크림, 헤어로션, 헤어샴푸, 헤어린스, 헤어컨디셔너, 헤어스프레이, 헤어 에어졸, 포마드, 젤 등과 같은 용액, 솔젤, 에멀젼, 오일, 왁스, 에어졸 등의 다양한 형태로 제조될 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 화장료 조성물은 부형제, 담체 등 기타 첨가제를 포함할 수 있으며 일반 피부 화장료에 배합되는 보통의 성분을 필요한 만큼 적용 배합하는 것이 가능하다.
상기 화장료 조성물의 제형의 형태가 페이스트, 크림 또는 겔인 경우에는 담체 성분으로서 동물성유, 식물성유, 왁스, 파라핀, 전분, 트라칸트, 셀루로오스 유도체, 폴리에틸렌글리콜, 실리콘, 벤토나이트, 실리카, 탈크 또는 산화아연 등이 이용될 수 있다.
상기 화장료 조성물의 제형의 형태가 파우더 또는 스프레이인 경우에는 담체 성분으로서 락토오스, 탈크, 실리카, 알루미늄 히드록시드, 칼슘 실리케이트 또는 폴리아미드 파우더가 사용될 수 있고, 특히 스프레이인 경우에는 추가적으로 클로로플루오로히드로카본, 프로판/부탄 또는 디메틸 에테르와 같은 추진체가 포함될 수 있으나 이에 제한되는 것은 아니다.
상기 화장료 조성물의 제형의 형태가 용액 또는 유탁액인 경우에는 담체 성분으로서 용매, 용해화제 또는 유탁화제가 이용될 수 있고, 예를 들면 물, 에탄올, 이소프로판올, 에틸 카보네이트, 에틸 아세테이트, 벤질 알코올, 벤질 벤조에이트, 프로필렌글리콜, 1,3-부틸글리콜 오일, 글리세롤 지방족 에스테르, 폴리에틸렌글리콜 또는 소르비탄의 지방산 에스테르 등이 이용될 수 있다.
상기 화장료 조성물의 제형의 형태가 현탁액인 경우에는 담체 성분으로서 물, 에탄올 또는 프로필렌 글리콜과 같은 액상의 희석제, 에톡실화 이소스테아릴 알코올, 폴리옥시에틸 소르비톨 에스테르 및 폴리옥시에틸렌 소르비탄 에스테르 등과 같은 현탁제, 미소결정성 셀룰로오스, 알루미늄 메타히드록시드, 벤토나이트, 아가 또는 트라칸트 등이 이용될 수 있다.
상기 화장료 조성물의 제형이 계면-활성제 함유 클렌징인 경우에는 담체 성분으로서 지방족 알코올 설페이트, 지방족 알코올 에테르 설페이트, 설포숙신산 모노에스테르, 이세티오네이트, 이미다졸리늄 유도체, 메틸타우레이트, 사르코시네이트, 지방산 아미드 에테르 설페이트, 알킬아미도베타인, 지방족 알코올, 지방산 글리세리드, 지방산 디에탄올아미드, 식물성유, 라놀린 유도체 또는 에톡실화 글리세롤 지방산 에스테르 등이 이용될 수 있다.
상기 화장료 조성물의 제형이 헤어샴푸인 경우에는 본 발명의 펩타이드에 증점제, 계면활성제, 점도 조절제, 보습제, pH 조절제, 방부제, 에센셜 오일 등과 같이 샴푸를 조성하기 위한 베이스 성분들이 혼합될 수 있다. 상기 증점제로는 CDE가 사용될 수 있고, 상기 계면활성제로는 음이온 계면활성제인 LES와, 양쪽성 계면활성제인 코코 베타인이 사용될 수 있고, 상기 점도조절제로는 폴리 쿼터가 사용될 수 있으며, 상기 보습제로는 글리세린이 사용될 수 있고, 상기 pH 조절제로는 구연산, 수산화나트륨이 사용될 수 있다. 상기 방부제로는 자몽 추출물 등이 사용될 수 있고, 이 외에도 시더우드, 페퍼민트, 로즈마리 등의 에센셜 오일과 실크 아미노산, 펜타올 또는 비타민 E가 첨가될 수 있다.
상기 화장료 조성물에 포함되는 성분은 유효 성분으로서 본 발명의 펩타이드와 담체 성분 이외에 화장료 조성물에 통상적으로 사용되는 성분들, 예를 들면 항산화제, 안정화제, 용해화제, 비타민, 안료 및 향료와 같은 통상적인 보조제 등을 더 포함할 수 있으나, 이에 제한되는 것은 아니다.

3. 본 발명의 펩타이드의 용도

본 발명의 펩타이드는 발모 촉진 또는 탈모증의 방지 또는 억제 활성을 갖는다. 본 발명의 펩타이드는 모낭 모유두세포(HFDPC) 및 인간외모근초세포(HHORSC)의 증식을 촉진하고, 모낭 모유두세포(HFDPC)에서 베타-카테닌 하위 유전자인 LEF-1 및 Cyclin D1의 발현을 상향 조절하고, bFGF, HGF, 및 VEGF와 같은 성장인자의 발현을 상향조절한다.
다만, 본 발명의 효과는 상기에서 언급한 효과로 제한되지 아니하며, 언급되지 않은 또 다른 효과들은 하기의 기재로부터 당업자에게 명확히 이해될 수 있을 것이다.
도 1은 본 발명의 펩타이드를 HHFDPC 세포에 10μM 및 100μM로 처리하였을 때 HHFDPC의 증식을 촉진하는 것을 보여주는 실험결과이다.

도 4a은 HHFDPC 세포에 본 발명의 펩타이드를 10μM, 50μM, 및 100μM으로 처리하였을 때 HHFDPC의 활성이 증가하여 bFGF, KGF, VEGF와 같은 성장인자의 발현이 증가되는 것을 보여주는 실험결과이다.

이하, 본 발명을 실시예에 의하여 상세히 설명한다. 단, 하기 실시예는 본 발명을 구체적으로 예시하는 것이며, 본 발명의 내용이 하기 실시예에 의해 한정되지 아니한다.
제조예 1: 펩타이드 및 펩타이드 복합체의 제조
자동 펩타이드 합성기(Milligen 9050, Millipore, 미국)를 이용하여 하기의 표 1에 기재된 서열번호 1의 아미노산 서열을 갖는 펩타이드를 합성하고, C18 역상 고성능액체크로마토그래피(HPLC)(Waters Associates, 미국)를 이용하여 이들 합성된 펩타이드를 순수 분리하였다. 컬럼은 ACQUITY UPLC BEH300 C18 (2.1 ㎜ Х 100 ㎜, 1.7 ㎛, Waters Co, 미국)을 이용하였다.

HHFDPC를 4 x 103 cells/well의 밀도로 96-웰 세포배양 플레이트에 접종(seeding)하여 중간엽줄기세포 완전배지(mesenchymal stem cell complete media)조건에서 24시간 동안 배양하였다. 이어서, 무혈청(serum free)중간엽줄기세포 배지로 교체한 후, 24시간 동안 더 배양하였다. 배양한 세포 배양물에 상기 제조예 1에서 제조한 펩타이드를 각각 10 μM, 100 μM 처리하고, 양성 대조군으로는 미녹시딜(monoxidil, MNX)을 1 μM 처리하고, 아무것도 처리하지 않은 것을 음성 대조군(con)으로하여, 37℃에서 72시간 더 배양하였다. 이어서, 여기에 5 mg/ml MTT 용액 10 μL를 첨가하여, 37℃ CO2 인큐베이터에서 4시간 동안 인큐베이션하고, 배지를 제거한 후, DMSO를 100 μL 처리하여, 10분간 진탕(shaking) 해주었다. 마지막으로 분광기(Spectrophotometer, Molecular Devices (USA, CA), SpectraMax iD3)를 이용하여 540 nm 파장에서의 흡광도를 측정하였다.
실험결과, 도 1에서 보여지는 바와 같이, 제조예 1의 펩타이드를 HHFDPC에 10μM 및 100μM로 처리하였을 때 음성대조군(con) 대비 HHFDPC의 증식을 촉진함을 확인하였다.
실험예 2: 인간외모근초세포(HHORSC)의 증식 촉진
상기 제조예 1에서 제조한 펩타이드가 인간외모근초세포(human hair outer root sheath cell, HHORSC)의 증식에 미치는 영향에 대해 실험하였다.
HHORSC를 4 x 103 cells/well의 밀도로 96-웰 세포배양 플레이트에 접종(seeding)하여 중간엽줄기세포 완전배지(mesenchymal stem cell complete media)조건에서 24시간 동안 배양하였다. 이어서, 무혈청(serum free)중간엽줄기세포 배지로 교체한 후, 24시간 동안 더 배양하였다. 배양한 세포 배양물에 상기 제조예 1에서 제조한 펩타이드를 각각 50 μM, 100 μM 처리하고, 양성 대조군으로 EGF(epidermal growth factor)를 100 nM 처리하고, 아무것도 처리하지 않은 것을 음성대조군(con)으로 하여, 37℃에서 72시간 더 배양하였다. 이어서, 여기에 5 mg/ml MTT 용액 10 μL를 첨가하여, 37℃ CO2 인큐베이터에서 4시간 동안 인큐베이션하고, 배지를 제거한 후, DMSO를 100 μL 처리하여, 10분간 진탕(shaking) 해주었다. 마지막으로 분광기(Spectrophotometer, Molecular Devices (USA, CA), SpectraMax iD3)를 이용하여 540 nm 파장에서의 흡광도를 측정하였다.
실험결과, 도 2에서 보여지는 바와 같이, 제조예 1의 펩타이드를 HHORSC에 50μM 및 100μM으로 처리하였을 때 음성대조군(con)과 비교하여 HHORSC의 증식을 촉진함을 확인하였다.
실험예 3: 인간 모유두세포에서 베타-카테닌 하위 유전자의 발현 촉진
상기 제조예 1에서 제조한 펩타이드가 인간 모낭 모유두세포(HHFDPC)에서 베타-카테닌(β-catenin)의 하위 유전자인 LEF-1(Lymphoid enhancer-binding factor 1) 및 Cyclin D1의 발현 수준에 미치는 영향을 RT-PCR 방법으로 확인하였다.
HHFDPC를 4 x 105 cells/well의 밀도로 6-웰 세포배양 플레이트에 접종(seeding)하여 중간엽줄기세포 완전배지(mesenchymal stem cell complete media)조건에서 24시간 동안 배양하였다. 이어서, 무혈청(serum free)중간엽줄기세포 배지로 교체한 후, 24시간 동안 더 배양하였다. 배양한 세포 배양물에 상기 제조예 1에서 제조한 펩타이드를 각각 10 μM, 50 μM , 100 μM 처리하고, 양성대조군으로는 10 ng/mL의 rhWnt-3a(recombinant human Wnt-3a protein, R&D systems (USA, MN))를 처리하였고, 아무것도 처리하지 않은 것을 음성대조군(con)으로 하여, 각각을 37℃에서 24시간 더 배양하였다. 배양물을 PBS로 세척한 후, eaay blue (Intron, South Korea)를300 μL 처리하여 RNA를 분리하였다. 분리한 RNA를 Nano drop으로 정량한 후 cDNA synthesis kit (Enzynomics, South Korea)를 사용하여 cDNA 합성을 수행하였다. 이어서, 아래 표 2의 프라이머와 PCR 프리믹스(permix)(Enzynomics, South Korea)를 이용하여 PCR 반응을 진행하였다. PCR 반응 생성물을 1.5% 아가로스겔에 전기영동하고, Bio-Rad Gel Image System으로 밴드를 검출하였다.

실험결과, 도 3a 및 도 3b에서 보여지는 바와 같이, HHFDPC에 제조예 1의 펩타이드를 10μM, 50μM, 및 100μM으로 처리하였을 때 세포의 증식과 분화에 영향을 주는 베타-카테닌이 활성화되어 하위 인자인 LEF-1 및 Cyclin D1의 유전자 발현이 증가된 것을 확인할 수 있었다.
실험예 4: 인간모유두세포에서 성장인자의 발현 촉진
상기 제조예 1에서 제조한 펩타이드가 인간 모낭 모유두세포(HHFDPC)에서 성장인자(growth factor)의 발현 수준에 미치는 영향을 RT-PCR 방법으로 확인하였다.
HHFDPC를 4 x 105 cells/well의 밀도로 6-웰 세포배양 플레이트에 접종(seeding)하여 중간엽줄기세포 완전배지(mesenchymal stem cell complete media)조건에서 24시간 동안 배양하였다. 이어서, 무혈청(serum free) 중간엽줄기세포 배지로 교체한 후, 24시간 동안 더 배양하였다. 배양한 세포 배양물에 상기 제조예 1에서 제조한 펩타이드를 각각 10 μM, 50 μM , 100 μM 처리하고, 양성대조군으로 미녹시딜(MNX)를 처리하고, 아무것도 처리하지 않은 것을 음성대조군(con)으로 하여, 37℃ 에서 24시간 더 배양하였다. 배양물을 PBS로 세척한 후, eaay blue (Intron, South Korea)를300 μL 처리하여 RNA를 분리하였다. 분리한 RNA를 Nano drop으로 정량한 후 cDNA synthesis kit (Enzynomics, South Korea)를 사용하여 cDNA 합성을 수행하였다. 이어서, 아래 표 3의 프라이머와 PCR 프리믹스(permix)(Enzynomics, South Korea)를 이용하여 PCR 반응을 진행하였다. PCR 반응 생성물을 1.5% 아가로스겔에 전기영동하고, Bio-Rad Gel Image System으로 밴드를 검출하였다.

실험결과, 도 4a 및 도 4b에서 보여지는 바와 같이, HHFDPC에 제조예 1의 펩타이드를 10μM, 50μM, 및 100μM으로 처리하였을 때 HHFDPC의 활성이 증가하여 bFGF, HGF, VEGF와 같은 성장인자의 발현이 증가되는 것을 확인하였다.
제조예 2: 약학적 조성물의 제조
2-1. 산제의 제조
본 발명의 펩타이드 2 g
유당 1 g
상기의 성분을 혼합하고 기밀포에 충진하여 산제를 제조하였다.
2-2. 정제의 제조
본 발명의 펩타이드 100 ㎎
옥수수전분 100 ㎎
유당 100 ㎎
스테아린산 마그네슘 2 ㎎
상기의 성분을 혼합한 후, 통상의 정제의 제조 방법에 따라서 타정하여 정제를 제조하였다.
2-3. 캡슐제의 제조
본 발명의 펩타이드 100 ㎎
옥수수전분 100 ㎎
유당 100 ㎎
스테아린산 마그네슘 2 ㎎
상기의 성분을 혼합한 후, 통상의 캡슐제의 제조방법에 따라서 젤라틴 캡슐에 충전하여 캡슐제를 제조하였다.
2-4. 환의 제조
본 발명의 펩타이드 1 g
유당 1.5 g
글리세린 1 g
자일리톨 0.5 g
상기의 성분을 혼합한 후, 통상의 방법에 따라 1환 당 4g이 되도록 제조하였다.
2-5. 과립의 제조
본 발명의 펩타이드 150 mg
대두추출물 50 mg
포도당 200 mg
전분 600 mg
상기의 성분을 혼합한 후, 30% 에탄올 100 mg을 첨가하여 섭씨 60℃에서 건조하여 과립을 형성한 후 포에 충진하였다.
제조예 3: 화장료 조성물의 제조

Claims (12)
- 서열번호 1의 아미노산 서열을 포함하는 펩타이드.
- 청구항 1의 펩타이드를 유효성분으로 포함하는 발모 촉진 또는 탈모증의 억제, 예방 또는 개선용 조성물.
- 청구항 1의 펩타이드를 유효성분으로 포함하는 탈모증의 예방 또는 치료용 약학적 조성물.
- 청구항 3에 있어서,상기 펩타이드는 모낭 모유두세포(HFDPC) 또는 외모근초세포(HORSC)의 증식을 촉진하는 것인, 탈모증의 예방 또는 치료용 약학적 조성물.
- 청구항 3에 있어서,상기 펩타이드는 모낭 모유두세포(HFDPC)에서(i) 베타-카테닌의 하위인자인 LEF-1 및 Cyclin D1으로 이루어지는 군에서 선택되는 하나 이상의 인자의 유전자 발현을 상향조절하거나, 또는(ii) bFGF, HGF, 및 VEGF로 이루어지는 군에서 선택되는 하나 이상의 성장인자의 발현을 상향조절하는 것인, 탈모증의 예방 또는 치료용 약학적 조성물.
- 청구항 3에 있어서,상기 약학적 조성물은 피부 외용제인 것인, 탈모증의 예방 또는 치료용 약학적 조성물.
- 청구항 1의 펩타이드를 유효성분으로 포함하는 발모 촉진 또는 탈모증의 예방 또는 개선용 화장료 조성물.
- 청구항 7에 있어서,상기 펩타이드는 모낭 모유두세포(HFDPC) 또는 외모근초세포(HORSC)의 증식을 촉진하는 것인, 발모 촉진 또는 탈모증의 예방 또는 개선용 화장료 조성물.
- 청구항 7에 있어서,상기 펩타이드는 모낭 모유두세포(HFDPC)에서(i) 베타-카테닌의 하위인자LEF-1 및 Cyclin D1으로 이루어지는 군에서 선택되는 하나 이상의 인자의 유전자 발현을 상향조절하거나, 또는(ii) bFGF, HGF, 및 VEGF로 이루어지는 군에서 선택되는 하나 이상의 성장인자의 발현을 상향조절하는 것인, 발모 촉진 또는 탈모증의 예방 또는 개선용 화장료 조성물.
- 청구항 7에 있어서,상기 화장료 조성물은 피부 외용제인 것인, 발모 촉진 또는 탈모증의 예방 또는 개선용 화장료 조성물.
- 청구항 7에 있어서,상기 화장료 조성물은 용액, 현탁액, 유탁액, 페이스트, 겔, 크림, 로션, 파우더, 비누, 계면활성제-함유 클린싱, 오일, 분말 파운데이션, 유탁액 파운데이션, 왁스 파운데이션 및 스프레이로 이루어지는 군에서 선택되는 하나 이상의 제형인 것인, 발모 촉진 또는 탈모증의 예방 또는 개선용 화장료 조성물.
- 청구항 7에 있어서,상기 화장료 조성물은 헤어토닉, 헤어크림, 헤어로션, 헤어샴푸, 헤어린스, 헤어컨디셔너, 헤어스프레이, 헤어 에어졸, 포마드 및 젤로 이루어지는 군에서 선택되는 하나 이상의 제형인 것인, 발모 촉진 또는 탈모증의 예방 또는 개선용 화장료 조성물.
