Abstract
本発明は、溶出性に優れるゲルを形成でき、かつ、耐摩耗性にも優れるゲルを形成できる組成物および組成物の製造方法を提供する。本発明の組成物は、アルギン酸塩と、カルボキシビニルポリマーと、油性基材とを含む組成物であって、アルギン酸塩の含有量が、組成物の全質量に対して、5質量%超25質量%以下であり、カルボキシビニルポリマーの含有量が、組成物の全質量に対して、5質量%超25質量%以下であり、油性基材が、ゲル化炭化水素を含み、アルミニウム化合物を実質的に含まず、キシリトールを含まないか、または、キシリトールを含む場合、キシリトールの含有量が組成物の全質量に対して10質量%未満である。
Description
本発明は、組成物、および、組成物の製造方法に関する。
がん患者においては、がん治療が口の粘膜に影響して口内炎が起こりやすい。例えば、抗がん剤治療では口内炎を起こしやすい薬剤の投与を受けたとき、頭頸部がん(頭から首の範囲のがん)の放射線治療では口の粘膜に放射線が直接当たったとき、に口内炎が必発である。口内炎の痛みは強く、食事を口からとることもできないほどである。
口内炎の対症療法としては、患部に直接貼り付ける貼付剤(例えば、アフタシール(登録商標)25μg、有効成分:トリアムシノロンアセトニド、大正製薬社製等)、患部に塗り付ける軟膏剤(例えば、デキサルチン口腔用軟膏、有効成分:デキサメタゾン、日本化薬社製等)、および、患部に吹き付ける噴霧剤(例えば、サルコート(登録商標)カプセル外用50μg、有効成分:ベクロメタゾンプロピオン酸エステル、帝人ファーマ社製等)がある。
しかし、これらの治療剤は免疫抑制剤であるステロイドを有効成分とするため、がん患者にとって望ましいものとはいえない。
また、食事を口から摂る際に、咀嚼物の物理的刺激により患部に貼り付けた貼付剤が剥がれたり、患部に塗布した軟膏剤、または、噴霧剤が失われたりして、口内炎の痛みを抑制することができない場合がある。
このような食事中の口内炎の痛みを抑制でき、かつ、食事後はすぐに溶出して口腔内から消失する組成物が望まれている。

例えば、特許文献1には、「疎水性軟膏基材、多価アルコール、粘着性物質、および、薬剤を配合することを特徴とする徐放性口腔用軟膏。」が記載されている(請求項1)。
しかし、特許文献1に記載された組成物を用いて形成されるゲルの溶出性、および、耐摩耗性が十分ではなかった。
そこで、本発明は、溶出性に優れるゲルを形成でき、かつ、耐摩耗性にも優れるゲルを形成できる組成物を提供することを課題とする。また、本発明は、組成物の製造方法を提供することも課題とする。
本発明者らは、上記課題を解決すべく鋭意検討を重ねたところ、以下の構成の本発明を完成させた。

アルギン酸塩と、カルボキシビニルポリマーと、油性基材とを含む組成物であって、
上記アルギン酸塩の含有量が、組成物の全質量に対して、5質量%超25質量%以下であり、

本発明によれば、溶出性に優れるゲルを形成でき、かつ、耐摩耗性にも優れるゲルを形成できる組成物を提供できる。また、本発明によれば、組成物の製造方法も提供できる。
本発明において、「~」を用いて表される範囲には「~」の両端を含むものとする。例えば、「A~B」で表される範囲にはA、および、Bを含む。

組成物は、アルギン酸塩と、カルボキシビニルポリマーと、油性基材とを含む組成物であって、
アルギン酸塩の含有量が、組成物の全質量に対して、5質量%超25質量%以下であり、

「生体適合性材料」とは、生体表面(例えば、皮膚、粘膜(例えば、口腔内の粘膜等)、目、歯、舌、爪、および、毛髪等)に良好に付着する材を意味する。
組成物は、生体に対しての悪影響が小さく、生体によく馴染む。生体表面は、健常な状態であっても、創傷、または、潰瘍を有していてもよい。
「生体適合性材料」とは、生体表面(例えば、皮膚、粘膜(例えば、口腔内の粘膜等)、目、歯、舌、爪、および、毛髪等)に良好に付着する材を意味する。
組成物は、生体に対しての悪影響が小さく、生体によく馴染む。生体表面は、健常な状態であっても、創傷、または、潰瘍を有していてもよい。
組成物は、水と接触することでゲルを形成し得る。水は、生体表面(例えば、口腔内表面等)に存在するものを利用してもよく、付着性を促進する目的で添加してもよい。組成物が水と接触してゲルを形成した際には、形成されたゲルの生体表面に対する付着性は、ゲルを形成する前の組成物の生体表面に対する付着性よりもより強固となる。
更に、組成物より形成されるゲルは、被付着面である生体表面を、外部刺激から保護する機能を有することが好ましい。この目的を達するために、組成物より形成されるゲルは一定以上の物理的強度を有することが好ましい。また、組成物より形成されるゲルは、非接着面表面に潤滑性を有してもよい。
「生体」とは、ヒト、および、ヒト以外の動物(例えば、哺乳類等)を意味する。ヒト以外の動物としては、例えば、霊長類、齧歯類(例えば、マウス、および、ラット等)、ウサギ、イヌ、ネコ、ブタ、ウシ、ヒツジ、および、ウマが挙げられる。

アルギン酸塩はアルギン酸の塩であれば特に限定されないが、アルギン酸の1価金属塩、または、アルギン酸のアンモニウム塩が好ましく、アルギン酸ナトリウム、アルギン酸カリウム、および、アルギン酸アンモニウムからなる群から選択される少なくとも1種がより好ましく、アルギン酸ナトリウムが更に好ましい。

アルギン酸塩はアルギン酸の塩であれば特に限定されないが、アルギン酸の1価金属塩、または、アルギン酸のアンモニウム塩が好ましく、アルギン酸ナトリウム、アルギン酸カリウム、および、アルギン酸アンモニウムからなる群から選択される少なくとも1種がより好ましく、アルギン酸ナトリウムが更に好ましい。
アルギン酸塩を構成する1価カチオンの定量分析および定性分析は、例えば、以下の測定条件にてイオンクロマトグラフ法によって行うことができる。

アルギン酸塩の重量平均分子量は特に限定されないが、50万以上の場合が多く、100万以上が好ましく、200万以上がより好ましく、250万以上が更に好ましい。
アルギン酸塩の重量平均分子量の上限は特に限定されないが、1000万以下が好ましく、500万以下がより好ましく、450万以下が更に好ましく、300万以下が特に好ましい。
アルギン酸塩の重量平均分子量の上限は特に限定されないが、1000万以下が好ましく、500万以下がより好ましく、450万以下が更に好ましく、300万以下が特に好ましい。
アルギン酸塩の重量平均分子量は、ゲル浸透クロマトグラフィー(GPC:Gel Permeation Chromatography)を用いて測定できる。

アルギン酸塩の粘度は特に限定されないが、アルギン酸塩を水に1質量%溶解した水溶液の粘度(25℃)は、50mPa・s以上の場合が多く、100mPa・s以上が好ましく、200mPa・s以上がより好ましく、500mPa・s以上が更に好ましく、800mPa・s以上がより更に好ましく、1,000mPa・s以上が特に好ましい。
アルギン酸塩の粘度の上限は特に限定されないが、水に1質量%溶解した水溶液の粘度は、10,000mPa・s以下が好ましく、5,000mPa・s以下がより好ましく、4,500mPa・s以下が更に好ましく、3,000mPa・s以下が特に好ましい。
また、アルギン酸塩の粘度は、300~400mPa・sも好ましい。
アルギン酸塩の粘度の上限は特に限定されないが、水に1質量%溶解した水溶液の粘度は、10,000mPa・s以下が好ましく、5,000mPa・s以下がより好ましく、4,500mPa・s以下が更に好ましく、3,000mPa・s以下が特に好ましい。
また、アルギン酸塩の粘度は、300~400mPa・sも好ましい。
アルギン酸塩の粘度は、回転式粘度計装置や粘弾性測定装置(レオメータ)を用いて測定できる。
アルギン酸塩は、粒子状であってもよい。
アルギン酸塩の粒子の平均粒径は特に限定されないが、50μm以上300μm未満が好ましく、50μm以上200μm未満がより好ましく、50μm以上150μm未満が更に好ましい。
アルギン酸塩の粒子の平均粒径は特に限定されないが、50μm以上300μm未満が好ましく、50μm以上200μm未満がより好ましく、50μm以上150μm未満が更に好ましい。
アルギン酸塩の粒子の平均粒径は、湿式・乾式粒度分布測定装置(LS13320、ベックマン・コールター社製)を用いて測定できる。
アルギン酸塩の含有量は、組成物の全質量に対して、5質量%超25質量%以下であり、10~25質量%が好ましく、15~25質量%がより好ましく、20~25質量%が更に好ましい。
アルギン酸塩は、1種を単独で使用してもよいし、2種以上を組み合わせて使用してもよい。
組成物において、アルギン酸塩は、市販品を使用できる。アルギン酸塩の市販品としては、具体的には、キミカ社製の「キミカアルギンI-S」、「キミカアルギンI-8」、「キミカアルギンI-5」、「キミカアルギンI-3」、「キミカアルギンI-1」、富士化学工業社製の「スノーアルギンSSH」、「スノーアルギンSH」、「スノーアルギンH」、「スノーアルギンM」、および、「スノーアルギンL」が挙げられる。
〈カルボキシビニルポリマー〉
組成物は、カルボキシビニルポリマーを含む。
カルボキシビニルポリマーは、カルボキシ基を有する水溶性のビニルポリマーであり、具体的には、アクリル酸、および/または、メタクリル酸を主鎖として、架橋構造を有するポリマーである。架橋構造としては、アリルショ糖、および、ペンタエリスリトールのアリルエーテル等による架橋構造が挙げられる。
組成物は、カルボキシビニルポリマーを含む。
カルボキシビニルポリマーは、カルボキシ基を有する水溶性のビニルポリマーであり、具体的には、アクリル酸、および/または、メタクリル酸を主鎖として、架橋構造を有するポリマーである。架橋構造としては、アリルショ糖、および、ペンタエリスリトールのアリルエーテル等による架橋構造が挙げられる。
カルボキシビニルポリマーの粘度は、特に限定されないが、pH7.5に調整し、カルボキシビニルポリマーを水に0.2質量%溶解した水溶液(20℃)において、50,000mPa・s以下が好ましく、500~50,000mPa・sがより好ましく、1,000~10,000mPa・sが更に好ましい。上記水溶液(20℃)のpH7.5での粘度が50,000mPa・s以下であると、組成物をゲル化させた際の耐摩耗性、および、溶出性がより優れたものとなる。
カルボキシビニルポリマーの粘度は、カルボキシビニルポリマーのpH7.5に調整した0.2質量%水溶液を、粘弾性測定装置(MCR301、アントンパール社製)を用いて、shere rate 1(1/s)、GAP 0.05mm、20℃で測定した値である。
組成物において、カルボキシビニルポリマーは、市販品を使用することができる。カルボキシビニルポリマーの市販品としては、具体的には、Lubrizol社製の「カーボポール971」、「カーボポール974」、「カーボポール980」、および、「カーボポール981」;富士フイルム和光純薬社製の「ハイビスワコー103」、「ハイビスワコー104」、および、「ハイビスワコー105」;東亞合成社製の「ジュンロンPW-120」、「ジュンロンPW-121」、および、「ジュンロンPW-312S」;住友精化社製の「AQUPEC HV-501E」、「AQUPEC HV-505E」、および、「AQUPEC HV805」;3Vシグマ社製の「シンタレンK」、および、「シンタレンL」;ならびに、日光ケミカルズ製の「NTC-CARBOMER 380」、および、「NTC-CARBOMER 381」が挙げられる。
カルボキシビニルポリマーの含有量は、組成物の全質量に対して、5質量%超25質量%以下であり、10~25質量%が好ましく、10~20質量%がより好ましく、10~15質量%が更に好ましい。
カルボキシビニルポリマーは、1種を単独で使用してもよいし、2種以上を組み合わせて使用してもよい。
アルギン酸塩の含有量が、組成物の全質量に対して、15~25質量%であり、カルボキシビニルポリマーの含有量が、組成物の全質量に対して、10~15質量%であることも好ましい。
〈油性基材〉
組成物は、油性基材を含む。
油性基材は、ゲル化炭化水素を含む。
組成物が油性基材を含むことにより、組成物の展着性がより優れたものとなる。
「油性基材」とは、水と交じり合わない成分を意味する。
油性基材の粘度は特に限定されないが、本発明の組成物をゲル化させた際の密着性がより優れる点で、100~1,000,000mPa・sが好ましく、1000~500,000mPa・sがより好ましい。粘度は、粘弾性測定装置(MCR302、アントンパール社製)を用いて、測定温度25℃にて、せん断速度1(1/s)で測定する。
また、油性基材の稠度は、本発明の組成物を塗りやすい点で、200~400が好ましく、250~370がより好ましい。稠度は、25℃に温度調整した後、自動ちょう度計(RPM-201、離合社製)を用いて、針の貫入時間5秒で、測定する。
組成物は、油性基材を含む。
油性基材は、ゲル化炭化水素を含む。
組成物が油性基材を含むことにより、組成物の展着性がより優れたものとなる。

また、油性基材の稠度は、本発明の組成物を塗りやすい点で、200~400が好ましく、250~370がより好ましい。稠度は、25℃に温度調整した後、自動ちょう度計(RPM-201、離合社製)を用いて、針の貫入時間5秒で、測定する。
ゲル化炭化水素としては、医薬品添加物規格の「ゲル化炭化水素」に適合するものを使用でき、より具体的には、ゲル化炭化水素は、流動パラフィンを5~10質量%に相当するポリエチレンでゲル化したものを好適に使用できる。
ゲル化炭化水素としては、プラスチベース(大正製薬社製)、ハイコールジェル(カネダ社製)、または、Hydrocarbon Gel 55W(Contract Pharmaceuticals Limited社製)が好ましい。
ゲル化炭化水素としては、プラスチベース(大正製薬社製)、ハイコールジェル(カネダ社製)、または、Hydrocarbon Gel 55W(Contract Pharmaceuticals Limited社製)が好ましい。
油性基材は、ゲル化炭化水素以外に、その他油性基材を含んでいてもよい。
その他油性基材としては、通常の油性軟膏に用いる原料が挙げられ、例えば、炭化水素類、ワックス類、植物油、動物油、中性脂質、合成油脂、ステロール誘導体、モノアルコールカルボン酸エステル類、オキシ酸エステル類、多価アルコール脂肪酸エステル類、シリコーン類、高級アルコール類、高級脂肪酸類、および、フッ素系油剤類が挙げられる。
その他油性基材としては、通常の油性軟膏に用いる原料が挙げられ、例えば、炭化水素類、ワックス類、植物油、動物油、中性脂質、合成油脂、ステロール誘導体、モノアルコールカルボン酸エステル類、オキシ酸エステル類、多価アルコール脂肪酸エステル類、シリコーン類、高級アルコール類、高級脂肪酸類、および、フッ素系油剤類が挙げられる。
炭化水素類としては、例えば、流動パラフィン(ミネラルオイル)、重質流動イソパラフィン、軽質流動イソパラフィン、α-オレフィンオリゴマー、ポリイソブテン、水添ポリイソブテン、ポリブテン、スクワラン、オリーブ由来スクワラン、スクワレン、ワセリン、および、固形パラフィンが挙げられる。
なお、ワセリンとしては、日本薬局方、または、それに準ずる規格の「ワセリン」「白色ワセリン」、または、「黄色ワセリン」に適合するものから選択するのが好ましい。

ワックス類としては、例えば、キャンデリラワックス、カルナウバワックス、ライスワックス、木ろう、みつろう、モンタンワックス、オゾケライト、セレシン、マイクロクリスタリンワックス、ペトロラタム、フィッシャートロプシュワックス、ポリエチレンワックス、および、エチレン・プロピレンコポリマーが挙げられる。
植物油としては、大豆油、ゴマ油、オリーブ油、やし油、パーム油、こめ油、綿実油、ひまわり油、コメヌカ油、カカオ脂、コーン油、べに花油、および、なたね油が挙げられる。
動物油としては、ミンク油、タートル油、魚油、牛油、馬油、豚油、および、鮫スクワランが挙げられる。
中性脂質としては、トリオレイン、トリリノレイン、トリミリスチン、トリステアリン、および、トリアラキドニンが挙げられる。
合成油脂としては、例えば、リン脂質、および、アゾンが挙げられる。
ステロール誘導体としては、ジヒドロコレステロール、ラノステロール、ジヒドロラノステロール、フィトステロール、コール酸、および、コレステリルリノレートが挙げられる。
モノアルコールカルボン酸エステル類としては、ミリスチン酸オクチルドデシル、ミリスチン酸ヘキシルデシル、イソステアリン酸オクチルドデシル、および、パリミチン酸セチルが挙げられる。
オキシ酸エステル類としては、乳酸セチル、リンゴ酸ジイソステアリル、および、モノイソステアリン酸水添ヒマシ油が挙げられる。

油性基材の含有量は特に限定されないが、組成物の全質量に対して、40質量%以上が好ましく、50質量%以上がより好ましい。上限は特に限定されないが、組成物の全質量に対して、90質量%以下が好ましく、80質量%以下がより好ましく、75質量%以下が更に好ましい。油性基材の含有量が上記範囲内であると、組成物が油性基材を含むことによる効果がより発揮されることとなる。
油性基材は、1種を単独で使用してもよいし、2種以上を組み合わせて使用してもよい。
〈アルミニウム化合物〉
組成物は、アルミニウム化合物を実質的に含まない。
「実質的に含まない」とは、本発明の効果に影響を及ぼさない程度の少量のアルミニウム化合物(例えば、組成物の原料中に含まれる微量のアルミニウム等)を含むことは許容範囲にあるということを意味する。具体的には、「アルミニウム化合物を実質的に含まない」とは、組成物中のアルミニウム化合物の含有量が、組成物の全質量に対して、0.1質量%未満であることを意味する。なかでも、組成物の全質量に対して、0.01質量%以下であることが好ましい。下限は特に限定されないが、組成物の全質量に対して、0質量%以上が挙げられる。
組成物は、アルミニウム化合物を実質的に含まない。
「実質的に含まない」とは、本発明の効果に影響を及ぼさない程度の少量のアルミニウム化合物(例えば、組成物の原料中に含まれる微量のアルミニウム等)を含むことは許容範囲にあるということを意味する。具体的には、「アルミニウム化合物を実質的に含まない」とは、組成物中のアルミニウム化合物の含有量が、組成物の全質量に対して、0.1質量%未満であることを意味する。なかでも、組成物の全質量に対して、0.01質量%以下であることが好ましい。下限は特に限定されないが、組成物の全質量に対して、0質量%以上が挙げられる。
アルミニウム化合物は、アルミニウム(アルミニウム原子)を含む化合物であれば特に限定されない。具体的には、水溶性のアルミニウム化合物が挙げられる。
水溶性のアルミニウム化合物としては、例えば、塩化アルミニウム(AlCl3)、硫酸アルミニウム(Al2(SO4)3)、硝酸アルミニウム(Al(NO3)3)、アンモニウムミョウバン(AlNH4(SO4)2・12H2O)、カリウムミョウバン(AlK(SO4)2・12H2O)、酢酸アルミニウム、プロピオン酸アルミニウム、グリコール酸アルミニウム(ヒドロキシ酢酸アルミニウム)、乳酸アルミニウム、リンゴ酸アルミニウム、酒石酸アルミニウム、クエン酸アルミニウム、および、イソクエン酸アルミニウムが挙げられる。

〈キシリトール〉
組成物は、キシリトールを含まないか、または、キシリトールを含む場合、キシリトールの含有量が組成物の全質量に対して10質量%未満である。
キシリトールの含有量が上記態様である場合、組成物の溶出性および耐摩耗性が両立できる。
なお、組成物がキシリトールを含む場合、キシリトールの含有量は、組成物の全質量に対して、1質量%以下が好ましく、0.5質量%未満がより好ましく、0.1質量%以下が更に好ましい。
組成物は、キシリトールを含まないか、または、キシリトールを含む場合、キシリトールの含有量が組成物の全質量に対して10質量%未満である。
キシリトールの含有量が上記態様である場合、組成物の溶出性および耐摩耗性が両立できる。
なお、組成物がキシリトールを含む場合、キシリトールの含有量は、組成物の全質量に対して、1質量%以下が好ましく、0.5質量%未満がより好ましく、0.1質量%以下が更に好ましい。
組成物は、実質的に水を含まないことが好ましい。
「実質的に水を含まない」とは、本発明の効果に影響を及ぼさない程度の少量の水(例えば、組成物の原料中に含まれる微量の水分)を含むことは許容範囲にあるということを意味する。具体的には、「実質的に水を含まない」とは、組成物中の水の含有量が、組成物の全質量に対して、3質量%以下であることを意味する。なかでも、組成物の全質量に対して、1質量%以下であることが好ましい。下限は特に限定されないが、組成物の全質量に対して、0質量%以上が挙げられる。
組成物が水を実質的に含まない場合、組成物を生体に適用した際に、形成されるゲルの付着性がより向上し、保護性能も向上する。また、組成物の保存安定性もより向上する。
組成物中における水の含有量の測定方法としては、例えば、日本薬局方記載の水分測定法(カールフィッシャー法)が挙げられる。ただし、水分測定用メタノールに試料を分散させ水分を抽出する工程を追加し、抽出した液に対して電量滴定法による試験を行うことが好ましい。
「実質的に水を含まない」とは、本発明の効果に影響を及ぼさない程度の少量の水(例えば、組成物の原料中に含まれる微量の水分)を含むことは許容範囲にあるということを意味する。具体的には、「実質的に水を含まない」とは、組成物中の水の含有量が、組成物の全質量に対して、3質量%以下であることを意味する。なかでも、組成物の全質量に対して、1質量%以下であることが好ましい。下限は特に限定されないが、組成物の全質量に対して、0質量%以上が挙げられる。
組成物が水を実質的に含まない場合、組成物を生体に適用した際に、形成されるゲルの付着性がより向上し、保護性能も向上する。また、組成物の保存安定性もより向上する。

〈糖アルコール、および、糖〉
組成物は、糖アルコール、および、糖からなる群から選択される少なくとも1種を更に含んでもよい。組成物が糖アルコール、および、糖からなる群から選択される少なくとも1種を含むと、組成物をゲル化させた際の引っ掻き耐性がより優れたものとなる。
本明細書においてキシリトールは、糖アルコールには含まれない。
組成物は、糖アルコール、および、糖からなる群から選択される少なくとも1種を更に含んでもよい。組成物が糖アルコール、および、糖からなる群から選択される少なくとも1種を含むと、組成物をゲル化させた際の引っ掻き耐性がより優れたものとなる。
本明細書においてキシリトールは、糖アルコールには含まれない。
糖アルコールは、アルドース、または、ケトースのカルボニル基が還元された構造を有する有機化合物であり、具体的には、エリスリトール、マンニトール、および、ソルビトールが挙げられ、エリスリトール、マンニトール、および、ソルビトールからなる群から選択される少なくとも1種が好ましい。
糖は特に限定されないが、例えば、単糖類、または、二糖類であり、具体的には、グルコース、ガラクトース、スクロース、トレハロース、および、ラクトースを挙げることができ、グルコース、ガラクトース、および、トレハロースからなる群から選択される少なくとも1種が好ましく、トレハロースがより好ましい。
上記糖アルコール、および、糖からなる群から選択される少なくとも1種は、エリスリトール、マンニトール、ソルビトール、グルコース、ガラクトース、スクロース、トレハロース、および、ラクトースからなる群から選択される少なくとも1種であることが好ましい。
組成物が糖アルコール、および、糖からなる群から選択される少なくとも1種を含む場合、組成物中の糖アルコール、および、糖の合計含有量は、特に限定されないが、組成物の全質量に対して、0.5~20.0質量%が好ましく、5.0~15.0質量%がより好ましい。糖アルコール、および、糖の合計含有量が上記範囲内であると、組成物が糖アルコール、および、糖からなる群から選択される少なくとも1種を含むことによる効果がより発揮されることとなる。
[組成物の製造方法]
組成物の製造方法としては、アルギン酸塩、カルボキシビニルポリマー、および、油性基材を混合する工程を含むことが好ましい。また、必要に応じて、脱水処理を実施してもよい。

混合の方法は特に限定されず、粉末成分を混合する際に用いられる、従来公知の方法を用いることができる。
例えば、組成物を構成する成分のうち、一部の成分を事前に混合して、その後、残りの成分と混合するような、段階的な混合方法を実施してもよい。また、全ての成分を同時に混合してもよい。上記手順によれば、成分がより均一に分散された組成物を得ることができる。
上記混合物と油性基材とを混合する際には、混合物と油性基材とを一括で混合してもよいし、混合物を複数回に分けて油性基材に添加して混合してもよい。
なお、各成分を混合した後、得られた組成物に対して脱気処理を施して、組成物から水を除去することが好ましい。
例えば、組成物を構成する成分のうち、一部の成分を事前に混合して、その後、残りの成分と混合するような、段階的な混合方法を実施してもよい。また、全ての成分を同時に混合してもよい。上記手順によれば、成分がより均一に分散された組成物を得ることができる。
上記混合物と油性基材とを混合する際には、混合物と油性基材とを一括で混合してもよいし、混合物を複数回に分けて油性基材に添加して混合してもよい。
なお、各成分を混合した後、得られた組成物に対して脱気処理を施して、組成物から水を除去することが好ましい。
[組成物の機能]
組成物を水と接触させることにより、ゲルが形成される。換言すると、組成物が水を吸収することにより、ゲルが自発的に形成される。

組成物は、生体表面上に適用され、ゲルを形成し得る。ゲルを形成する方法としては、組成物を生体表面上に配置して、生体表面上に配置された組成物と水とを接触させることにより、生体表面上にゲルを形成する方法が挙げられる。
組成物を配置する生体表面としては、口腔内の粘膜表面が好適に挙げられる。

組成物の粘度は特に限定されないが、組成物の展着性がより優れる点で、100,000~1,000,000mPa・sが好ましい。粘度の測定は、粘弾性測定装置(MCR302、アントンパール社製)を使用して、測定温度25℃にて、せん断速度1(1/s)で測定する。
組成物の形態(性状)は特に限定されないが、例えば、軟膏状、クリーム状、および、半固体状が挙げられる。
[組成物の用途、および、使用方法]
組成物の用途としては、生体保護用途が挙げられる。
具体的には、組成物は、口腔粘膜保護材等の粘膜保護材、創傷被覆材、薬剤徐放基材、口腔内湿潤材、および、止血材として利用され得る。

組成物を粘膜に対して使用する場合、組成物を粘膜上に配置し、水、または、水を含む溶液を添加すれば、ゲル化して形成されるゲルが粘膜により強固に付着する。つまり、組成物の使用方法(または、ゲルの製造方法)として、組成物を粘膜上に配置して、粘膜上に配置された組成物と水とを接触させることにより、粘膜上にゲルを形成する方法が挙げられる。
特に、口腔粘膜に対して組成物を適用する場合、組成物を口腔粘膜に付着させれば、唾液中の水分によって、組成物がゲル化するため、取扱いが簡便である。また、仮に、唾液量が少ない場合には、組成物を口腔粘膜に付着させた後、水、または、人工唾液をスプレーする等して、水分を供給すればよい。
特に、口腔粘膜に対して組成物を適用する場合、組成物を口腔粘膜に付着させれば、唾液中の水分によって、組成物がゲル化するため、取扱いが簡便である。また、仮に、唾液量が少ない場合には、組成物を口腔粘膜に付着させた後、水、または、人工唾液をスプレーする等して、水分を供給すればよい。
組成物を口腔粘膜に付着させると、唾液中の水分により組成物の溶解や膨潤が始まり、上記ゲルの形成が開始すると同時に、口腔粘膜表面のムチンとアルギン酸塩またはカルボキシビニルポリマーとが水素結合により接着する。このようなメカニズムにより、本発明の組成物により形成されるゲルは、耐摩耗性に優れると推測される。
組成物が薬剤徐放基材として用いられる場合、徐放される薬剤の種類は特に限定されず、公知の薬剤が挙げられる。
以下、実施例により本発明をより具体的に説明するが、本発明はこれらの実施例に限定されるものではない。
[実施例C1~C16、比較例R1~R10]
〈組成物の調製〉
表1及び2に示す各成分および含有量で混合して、実施例C1~C16、および、比較例R1~R10の組成物を調製した。
実施例C1~C16の組成物中におけるアルミニウム化合物の含有量は、いずれの場合も、各組成物の全質量に対して、0.01質量%以下であった。つまり、いずれの実施例の組成物は、実質的にアルミニウム化合物を含んでいなかった。
なお、以下、代表的に実施例C1の組成物の製造手順を記載する。実施例C2~C16、および、比較例R1~R10においては、各成分の含有量を調整した以外は実施例C1と同様の手順で調製した。
〈組成物の調製〉
表1及び2に示す各成分および含有量で混合して、実施例C1~C16、および、比較例R1~R10の組成物を調製した。
実施例C1~C16の組成物中におけるアルミニウム化合物の含有量は、いずれの場合も、各組成物の全質量に対して、0.01質量%以下であった。つまり、いずれの実施例の組成物は、実質的にアルミニウム化合物を含んでいなかった。
なお、以下、代表的に実施例C1の組成物の製造手順を記載する。実施例C2~C16、および、比較例R1~R10においては、各成分の含有量を調整した以外は実施例C1と同様の手順で調製した。
ゲル化炭化水素(Hydrocarbon Gel 55W、Contract Pharmaceuticals Limited社製)(134.0g)、および、カルボキシビニルポリマー(Carbopol 971P NF、Lubrizol社製)(30.0g)を混合機(型式:HP-2P-03、Primix社製)に入れ、室温下、50rpmにて2分間撹拌した。
更に、上記混合機内にアルギン酸ナトリウム(キミカアルギンI-SF、キミカ社製)(36.0g)を入れ、室温下、50rpmにて10分間撹拌した。
最後に、アルミチューブ(φ12.8×86mm、武内プレス工業社製)に5g充填し、チューブ折曲機(中西製作所製)を用いて密封することで、実施例C1の組成物を得た。
更に、上記混合機内にアルギン酸ナトリウム(キミカアルギンI-SF、キミカ社製)(36.0g)を入れ、室温下、50rpmにて10分間撹拌した。
最後に、アルミチューブ(φ12.8×86mm、武内プレス工業社製)に5g充填し、チューブ折曲機(中西製作所製)を用いて密封することで、実施例C1の組成物を得た。
《高温保管》
上記で得られたアルミチューブに充填された実施例C1~C16、および、比較例R1~R10の各組成物を、表1及び2に示す高温保管条件に従って一定期間保管した。得られた高温保管後の組成物は、後述する溶出性および耐摩耗性の評価に用いた。

〈性能評価〉
《擬似生体膜の作製》
TDAB(50mg、テトラドデシルアンモニウムブロミド、富士フイルム和光純薬社製)、ポリ塩化ビニル(800mg、富士フイルム和光純薬社製)、および、DOPP(0.6mL、ジ-n-オクチルフェニルホスホナート、富士フイルム和光純薬社製)をTHF(10mL、テトラヒドロフラン、富士フイルム和光純薬社製)に溶解したものをシャーレで室温乾燥し、脂質膜(厚さ:200μm)を得た。
次に、寒天(0.5g、カリコリカン(登録商標)、伊那食品工業社製)、ジェランガム(0.1g、ケルコゲル(登録商標)、CPケルコ社製)および、蒸留水(49.4g)からなるハイドロゲルに、作製した脂質膜を貼り合わせた。
続いて、脂質膜表面をMPC(2-メタクリロイルオキシエチルホスホリルコリン)ポリマー(LIPIDURE(登録商標)-CM5206、日油社製)によりコートして、疑似生体膜を得た。
《擬似生体膜の作製》
TDAB(50mg、テトラドデシルアンモニウムブロミド、富士フイルム和光純薬社製)、ポリ塩化ビニル(800mg、富士フイルム和光純薬社製)、および、DOPP(0.6mL、ジ-n-オクチルフェニルホスホナート、富士フイルム和光純薬社製)をTHF(10mL、テトラヒドロフラン、富士フイルム和光純薬社製)に溶解したものをシャーレで室温乾燥し、脂質膜(厚さ:200μm)を得た。
次に、寒天(0.5g、カリコリカン(登録商標)、伊那食品工業社製)、ジェランガム(0.1g、ケルコゲル(登録商標)、CPケルコ社製)および、蒸留水(49.4g)からなるハイドロゲルに、作製した脂質膜を貼り合わせた。
続いて、脂質膜表面をMPC(2-メタクリロイルオキシエチルホスホリルコリン)ポリマー(LIPIDURE(登録商標)-CM5206、日油社製)によりコートして、疑似生体膜を得た。
(溶出性の評価)
溶出性の評価対象の組成物として、高温保管前の各組成物、および、高温保管後の各組成物を用いて、それぞれ溶出性を評価した。
作製した擬似生体膜上にワッシャー(内径10mm、厚さ0.5mm)を置き、ワッシャー内に調製した組成物を詰め、スパーテルヘラで押し付けることで組成物を塗布した。ワッシャーを剥がした後、人工唾液(サリベート(登録商標))を吹き付け1分間手で馴染ませ、組成物をゲル化させて溶出性評価用サンプルを作製した。
作製した溶出性評価用サンプル、および、人工唾液をタッパーに入れ、人工唾液の液面が、擬似生体膜とゲル化した組成物の間になるよう、人工唾液の液量を調整した。小型振蕩恒温器(型式:PIC-100S、アズワン社製)を用いて37℃で振とうし、ゲルが溶出して消失する時間(最長6時間まで)を評価した。
(溶出性の評価基準)
AA:1時間以下
A:1時間超2時間以下
B:2時間超4時間以下
C:4時間超6時間以下
溶出性の評価対象の組成物として、高温保管前の各組成物、および、高温保管後の各組成物を用いて、それぞれ溶出性を評価した。

作製した擬似生体膜の中央にワッシャー(内径12mm、厚さ0.5mm)を置き、ワッシャー内に調製した組成物を詰め、摩耗試験機(TRIBOGEARTYPE-40、新東科学社製)を低速300mm/分で単回稼働させることで、組成物を塗布した。ワッシャーを剥がした後、人工唾液(サリベート(登録商標))を吹き付け1分間手で馴染ませ、組成物をゲル化させて耐摩耗性評価用サンプルを作製した。
作製した耐摩耗性評価用サンプルを摩耗試験機(TRIBOGEARTYPE-40、新東科学社製)で繰り返し摩耗し、擬似生体膜からゲルが完全に消失するまでの回数(往復)を計測し、以下の基準により耐摩耗性を評価した。なお、摩耗試験機のヘッドにはアルミニウムヘッド(0.114mm研磨)をセットし、荷重0.3N、振幅30mm、速度6000mm/分の条件で試験を実施した。
(耐摩耗性の評価基準)
A:200回以上
B:100回以上200回未満
C:100回未満

「耐摩耗性」の欄の各数値は、耐摩耗性の評価におけるゲルが完全に消失までの回数(往復)を示す。

[結果の説明]
実施例C1~C16の組成物は、溶出性および耐摩耗性が優れることが確認された。更に、60℃1か月という高温条件で保管した後においても、溶出性および耐摩耗性が優れることが確認された。

アルギン酸塩の粘度(1質量%水溶液、25℃)が300~400mPa・sである場合、高温保管前の溶出性がより優れることが確認された(実施例C1およびC8~C10の比較等)。
アルギン酸塩の含有量が、組成物の全質量に対して、15~25質量%であり、カルボキシビニルポリマーの含有量が、組成物の全質量に対して、10~15質量%である場合、本発明の効果がより優れることが確認された(実施例C1、C5、C6、C11、C12、C14およびC2~C4、C7~C10、C13、C15の比較等)。
アルギン酸塩の含有量が、組成物の全質量に対して、15~25質量%であり、カルボキシビニルポリマーの含有量が、組成物の全質量に対して、10~15質量%である場合、本発明の効果がより優れることが確認された(実施例C1、C5、C6、C11、C12、C14およびC2~C4、C7~C10、C13、C15の比較等)。
[比較例R11~R13]
また、本発明の組成物に該当しない市販品を用いて、実施例1と同様に、高温保管前の溶出性および高温保管前の耐摩耗性の評価を実施した。
評価結果を表3に示す。表3に示すように、比較例R11~R13の組成物を用いた場合、形成されるゲルは溶出性および耐摩耗性の両立ができなかった。
また、本発明の組成物に該当しない市販品を用いて、実施例1と同様に、高温保管前の溶出性および高温保管前の耐摩耗性の評価を実施した。
評価結果を表3に示す。表3に示すように、比較例R11~R13の組成物を用いた場合、形成されるゲルは溶出性および耐摩耗性の両立ができなかった。

Claims (6)
- アルギン酸塩と、カルボキシビニルポリマーと、油性基材とを含む組成物であって、
前記アルギン酸塩の含有量が、組成物の全質量に対して、5質量%超25質量%以下であり、
前記カルボキシビニルポリマーの含有量が、組成物の全質量に対して、5質量%超25質量%以下であり、
前記油性基材が、ゲル化炭化水素を含み、
アルミニウム化合物を実質的に含まず、
キシリトールを含まないか、または、キシリトールを含む場合、キシリトールの含有量が組成物の全質量に対して10質量%未満である、組成物。 - 前記アルギン酸塩の含有量が、組成物の全質量に対して、10~25質量%である、請求項1に記載の組成物。
- 前記カルボキシビニルポリマーの含有量が、組成物の全質量に対して、10~25質量%である、請求項1または2に記載の組成物。
- 前記油性基材の含有量が、組成物の全質量に対して、50質量%以上である、請求項1~3のいずれか1項に記載の組成物。
- 口腔粘膜保護に用いられる、請求項1~4のいずれか1項に記載の組成物。
- 請求項1~5のいずれか1項に記載の組成物の製造方法であって、
前記アルギン酸塩、前記カルボキシビニルポリマー、および、前記油性基材を混合する工程を含む、組成物の製造方法。
