本発明は、フィブリリン産生促進剤、化粧料組成物又は皮膚外用剤、経口投与剤、及びエラスチン線維産生促進剤に関する。
高齢化社会が進んだ現代においては、健康長寿だけでなく、QOL(Quality of Life)の向上が課題となっている。アンチエイジング分野における皮膚の美容は、高齢者のQOL向上に極めて重要であることから関心が高まっている。
肌の「ハリ」は、理想の肌の要素として挙げられることが多く、QOL向上のために改善要望の高い要素である。近年の研究によって、「ハリ」は「視覚的ハリ」と「触覚的ハリ」との2つの要素が存在することが明らかとなった(非特許文献1)。非特許文献1においては、「視覚的ハリ」と「触覚的ハリ」との関係について研究しており、「視覚的ハリ」は「視覚的うるおい感」と強い相関が見られる一方、「触覚的ハリ」は「視覚的うるおい感」と相関が見られなかった。非特許文献1には、角層水分量の増加とともに「視覚的ハリ」の有意な向上が観察されたが、一方で「触覚的ハリ」には有意な変化が見られなかったことが記載されている。
「触覚的ハリ」には、弾性線維(エラスチン線維)が重要な役割を果たしていることが知られている。皮膚真皮のエラスチン線維は、基底膜から皮下組織へ伸びる連続的なネットワークをつくっていて、皮膚の弾力性を保っている。老化に伴う皮膚の変化の中で、エラスチン線維の変性による変化は大きな位置を占めており、皮膚のタルミ及びシワの原因は、このエラスチン線維の変性が大きな要因であるといわれる。自然老化では、エラスチン線維は減少することが知られている。一方、光老化では、日光弾性線維症のように真皮乳頭層の線維は消失するが、網状層のエラスチン線維は量的にも増加し、かつ変性して無定型物質のような沈着が起こるとされている。これらが、弾力性がなく硬化したシワの多い光老化皮膚の原因と考えられている。
弾性線維は、皮膚の他では肺及び動脈に多く存在し、これらの組織の伸縮性を担っている。加齢に伴う弾性線維の劣化は、肺気腫性変化、動脈中膜硬化などの直接原因となる(非特許文献2)。
上記のように、弾性線維の劣化又は断裂は、肌の触覚的ハリの喪失、肺気腫性変化及び動脈中膜硬化といった症状を引き起こすため、弾性線維の維持、再生はQOL向上のため喫緊の課題である。しかし、弾性線維のターンオーバーは極めて遅く、加齢した組織では弾性線維再生能はほぼないとされる(非特許文献2)。弾性線維を再生できれば、皮膚の触覚的ハリ改善だけではなく、肺気腫及び動脈硬化の進展予防・治療など新たな予防・治療法が生まれると考えられる。
弾性線維の主要構成成分として、ミクロフィブリル(フィブリリン)及び無定型エラスチンが知られている。ミクロフィブリル上にエラスチンが沈着することにより、弾性線維が形成される。すなわち、ミクロフィブリルの産生を促進することにより、弾性線維の産生が促進すると考えられる。
シチジル酸、ウリジル酸はヌクレオチドの一種であり、生体内及び食品に広く含まれている物質であって、安全性が高い素材といえる。
特許文献1には、ウリジン又はその誘導体と、アセチルグルコサミンなどを含有する美容組成物が開示されている。特許文献1に記載の発明は、皮膚の保湿性を改善し角質層水分量を増大させるとともに、肌をハリのある状態に保つことが記載されている。
角層水分量の増加とともに「視覚的ハリ」の有意な向上が観察されたが、一方で「触覚的ハリ」には有意な変化が見られなかったとする非特許文献1の内容を参酌すると、特許文献1における肌の「ハリ」とは、いわゆる「視覚的ハリ」であると理解される。特許文献1には、ピリミジンヌクレオチド又はその前駆体がフィブリリンの産生を促進すること、及び肌の「触覚的ハリ」改善に寄与することは一切開示されていない。
特許文献2には、ウリジンのリン酸エステル又はその生理学的に許容される塩が、細胞のヒアルロン酸産生を促進し保湿などに寄与すること、及び細胞増殖を促進することが記載されている。また、特許文献2には、ヒアルロン酸産生促進及び細胞増殖促進に起因して、肌のシワ、タルミを改善する旨が開示されている。
特許文献2において開示されている肌のシワ、タルミの改善は、ヒアルロン酸産生促進による肌の保湿に起因する、いわゆる「視覚的ハリ」の改善と、細胞増殖に起因する量的変化から生じる外観上の変化であると考えられる。特許文献2には、ピリミジンヌクレオチド又はその前駆体がフィブリリンの産生を促進すること、及び肌の「触覚的ハリ」改善に寄与することは一切開示されていない。
特許文献3には、プリン系核酸関連物質及びピリミジン系核酸関連物質を含有する、コラーゲン産生促進用組成物が記載されている。しかし、特許文献3に実質的に開示された発明は、アデニル酸とウリジル酸との併用によるコラーゲン産生促進用組成物であり、ウリジル酸単体での使用及びシチジル酸の使用に関して実質的な開示はない。さらに、特許文献3には、ピリミジンヌクレオチド又はその前駆体がフィブリリンの産生を促進すること、及び肌の「触覚的ハリ」改善に寄与することは一切開示されていない。
従って、シチジル酸及びウリジル酸のような核酸関連物質が、フィブリリン産生促進作用を有するか否かについては、従来評価されていない。

すなわち、本発明は、ピリミジンヌクレオチド又はその前駆体を有効成分として含有する、フィブリリン産生促進剤である。
また、本発明は、ピリミジンヌクレオチド又はその前駆体を有効成分として含有する、フィブリリンの産生を促進するための化粧料組成物又は皮膚外用剤である。
また、本発明は、ピリミジンヌクレオチド又はその前駆体を有効成分として含有する、フィブリリンの産生を促進するため経口投与剤である。
さらに、本発明は、ピリミジンヌクレオチド又はその前駆体を有効成分として含有する、エラスチン線維産生促進剤である。

シチジル酸(シチジン一リン酸、cytidine 5'-phosphate、CMP)は、CAS登録番号63-37-6で示される化合物である。本明細書でシチジル酸と記載する場合、シチジル酸の塩も包含される。
本明細書において、シチジル酸の質量について記載する場合、シチジル酸2ナトリウム塩(CMP,2Na)に換算した際の質量を表す。シチジル酸溶液の濃度(%)について記載する場合、特に記載がなければ質量体積パーセント濃度(w/v%)であることとし、シチジル酸質量としてはCMP,2Na換算質量を用いる。2ナトリウム塩以外の塩を選択する場合、又は塩を形成しない遊離酸の場合には、シチジル酸の物質量を基準として、CMP,2Naに換算した場合の質量とする。
ウリジル酸(ウリジン一リン酸、uridine 5'-phosphate、UMP)は、CAS登録番号58-97-9で示される化合物である。本明細書でウリジル酸と記載する場合、ウリジル酸の塩も包含する概念である。

本明細書において、ピリミジンヌクレオチド前駆体とは、ピリミジンヌクレオチド、すなわちシチジル酸及び/又はウリジル酸へと代謝され得る化合物を意味する。化合物がピリミジンヌクレオチド前駆体に包含されるか否かは、その化合物がピリミジンヌクレオチドに変換されるという知見の有無によって判断される。具体的には、エクトヌクレオチダーゼなどの働きによりシチジル酸及び/又はウリジル酸へ分解されることが知られているシチジン二リン酸、シチジン三リン酸、ウリジン二リン酸、ウリジン三リン酸(松岡功,「神経系におけるエクトヌクレオチダーゼ」,臨床化学 33:11-18,2004)、キナーゼの働きによってシチジル酸及び/又はウリジル酸へとリン酸化されることが知られているシチジン、シトシン、ウリジン、ウラシル(A Orengo, 「Regulation of enzymic activity by metabolites. 本発明におけるシチジル酸の概念は、前述の通り塩を包含する。シチジル酸の塩としては、ナトリウム塩、カリウム塩などのアルカリ金属塩;カルシウム塩、マグネシウム塩、バリウム塩などのアルカリ土類金属塩;アルギニン、リジンなどの塩基性アミノ酸塩;アンモニウム塩、トリシクロヘキシルアンモニウム塩などのアンモニウム塩;モノエタノールアミン塩、ジエタノールアミン塩、トリエタノールアミン塩、モノイソプロパノールアミン塩、ジイソプロパノールアミン塩、トリイソプロパノールアミン塩などの各種のアルカノールアミン塩などを挙げることができる。これらの中でも、ナトリウム塩などのアルカリ金属塩が好ましい。アルカリ金属塩としては、具体的にはシチジル酸1ナトリウム、及びシチジル酸2ナトリウムを例示することができ、取り扱い性の面から、シチジル酸2ナトリウムが好ましい。
本発明におけるウリジル酸の概念は、前述の通り塩を包含する。ウリジル酸の塩としては、ナトリウム塩、カリウム塩などのアルカリ金属塩;カルシウム塩、マグネシウム塩、バリウム塩などのアルカリ土類金属塩;アルギニン、リジンなどの塩基性アミノ酸塩;アンモニウム塩、トリシクロヘキシルアンモニウム塩などのアンモニウム塩;モノエタノールアミン塩、ジエタノールアミン塩、トリエタノールアミン塩、モノイソプロパノールアミン塩、ジイソプロパノールアミン塩、トリイソプロパノールアミン塩などの各種のアルカノールアミン塩などを挙げることができる。これらの中でも、ナトリウム塩などのアルカリ金属塩が好ましい。アルカリ金属塩としては、具体的にはウリジル酸1ナトリウム、及びウリジル酸2ナトリウムを例示することができ、取り扱い性の面から、ウリジル酸2ナトリウムが好ましい。
本発明のフィブリリン産生促進剤、化粧料組成物又は皮膚外用剤、及び経口投与剤には、有効成分であるピリミジンヌクレオチド又はその前駆体を単独で用いてもよく、ピリミジンヌクレオチド又はその前駆体を2種以上同時に使用しても良く、ピリミジンヌクレオチド又はその前駆体とそれ以外の有効成分とを併用して用いても良い。
本発明におけるピリミジンヌクレオチド又はその前駆体の由来には格別の制限はなく、酵母、細菌、魚介類、動物、植物などの天然物由来のものが好適である。
本発明のフィブリリン産生促進剤は、経口投与剤に利用することができる。具体的には、飲食品、サプリメント、調製粉乳、経腸栄養剤、健康飲食品(特定保健用食品、機能性表示食品を含む)、動物飼餌料用添加物、ヒト又はヒト以外の動物用の医薬品などとして実用に供することができる。
本発明のフィブリリン産生促進剤を飲食品、健康飲食品又は調製粉乳などとして供する場合は、前記有効成分を公知の飲食品に適宜添加することにより、フィブリリン産生促進作用を有する飲食品とすることができる。対象の飲食品としては、乳・乳製品類、調味料類、飲料、菓子類、パン類、麺類、油脂類、畜肉加工品、水産加工品、農産加工品、冷凍食品、即席食品などを挙げることができる。
また、ピリミジンヌクレオチド又はその前駆体を、飲食品原料中に混合して、フィブリリン産生促進効果を有する新たな飲食品を製造することもできる。対象とする飲食品の形状としては、錠剤状、顆粒状、カプセル状、粉末状、溶液状、シロップ状、乳状、ペースト状など、各種形状のものを選ぶことができる。これらの飲食品の製造においては、本発明の有効成分のほかに、食品として使用可能な各種の賦形剤、調味成分などを適宜添加すれば良い。
前記飲食品は、フィブリリン産生促進効果との保健用途が表示された飲食品として提供・販売されていてもよい。「表示」行為には、需要者に対して前記用途を知らしめるための全ての行為が含まれ、前記用途を想起・類推させうるような表現であれば、表示の目的、表示の内容、表示する対象物・媒体などの如何に拘わらず、全て本技術の「表示」行為に該当する。
前記「表示」は、需要者が上記用途を直接的に認識できるような表現により行われることが好ましい。具体的には、飲食品に係る商品又は商品の包装に前記用途を記載したものを譲渡し、引き渡し、譲渡若しくは引き渡しのために展示し、輸入する行為、商品に関する広告、価格表若しくは取引書類に上記用途を記載して展示し、若しくは頒布し、又はこれらを内容とする情報に上記用途を記載して電磁気的(インターネットなど)方法により提供する行為などが挙げられる。
表示内容としては、行政などによって認可された表示(例えば、行政が定める各種制度に基づいて認可を受け、そのような認可に基づいた態様で行う表示など)であることが好ましい。また、そのような表示内容を、包装、容器、カタログ、パンフレット、POPなどの販売現場における宣伝材、その他の書類などへ付することが好ましい。
また、本発明のフィブリリン産生促進剤を経口投与に供する医薬品、サプリメント、経腸栄養剤などとして実用に供する場合、前記有効成分を単独、又は製剤補助剤などと合わせて製剤化することができる。
前記製剤の形状としては、錠剤、顆粒剤、カプセル剤、粒剤、散剤、溶液剤、シロップ剤、乳剤などとすることができる。
前記製剤においては、本発明の有効成分の他に、賦形剤、結合剤、崩壊剤、滑沢剤、矯味矯臭剤、溶解補助剤、懸濁剤、コーティング剤などの任意の製剤補助剤を、それぞれの供与形態に合わせて適宜組み合わせて用いれば良い。
本発明の剤における前記有効成分の配合量は、使用目的(予防、保健又は症状緩和など)、対象者の年令、投与又は摂取方法、剤型などに応じて、0.1~30%(W/W)の範囲から適宜選定すればよい。
本発明の経口投与剤の投与又は摂取量は、対象者の年令、体重、症状などの程度、投与又は摂取方法などに応じて変動するものの、1日当たり1mg~800g程度の範囲内から適宜選択すればよい。また、本発明の経口投与剤の投与又は摂取量は、1回あたりの投与量が1~5000mgであることが好ましく、1~1000mgであることがより好ましい。
本発明のフィブリリン産生促進剤は、化粧料組成物又は皮膚外用剤として実用に供することもできる。
本発明の化粧料組成物又は皮膚外用剤におけるピリミジンヌクレオチド又はその前駆体の含有量は、フィブリリン産生が促進されるという点で0.05(w/v%)以上であることが好ましい。特に、フィブリリン産生促進効果は濃度依存的に奏されることから、含有量は0.075(w/v%)以上であることがより好ましい。また、フィブリリン産生が有意に促進されるという点で0.092(w/v%)以上であることが好ましく、0.25(w/v%)以上であることがより好ましく、0.5(w/v%)以上であることが更に好ましく、1.0(w/v%)以上であることが特に好ましい。また、各種剤又は組成物として利用するときの扱い易さなどから、ピリミジンヌクレオチド又はその前駆体の含有量は10(w/v%)以下であることが好ましい。
本発明の化粧料組成物又は皮膚外用剤は、上記成分の他に、薬学的若しくは香粧的に許容される基剤又は担体を組み合わせて各種の形態に調製されても良い。薬学的若しくは香粧的に許容される基剤又は担体については、従来公知のものを用いることができる。また、本発明の化粧料組成物又は皮膚外用剤には、必要に応じて化粧料、外用の医薬品・医薬部外品などの皮膚又は粘膜に適用される外用組成物に配合される公知の各種成分を配合することができる。このような成分として、例えば、界面活性剤、色素(染料、顔料)、香料、防腐剤、殺菌剤(抗菌剤)、増粘剤、酸化防止剤、金属封鎖剤、清涼化剤、防臭剤、保湿剤、UV吸収剤、UV散乱剤、ビタミン類、植物エキス、皮膚収斂剤、抗炎症剤(消炎剤)、美白剤、細胞賦活剤、血管拡張剤、血行促進剤、及び皮膚機能亢進剤などを挙げることができる。
上記成分の内、界面活性剤の具体例としては、高級脂肪酸石けん、アルキル硫酸エステル塩、ポリオキシエチレンアルキルエーテル硫酸塩、アルキルエーテルリン酸エステル塩、N-アシルアミノ酸塩、アシルN-メチルタウリン塩などのアニオン界面活性剤;塩化アルキルトリメチルアンモニウム、塩化ジアルキルジメチルアンモニウムなどのカチオン界面活性剤;アルキルジメチルアミノ酢酸ベタイン、アルキルアミドジメチルアミノ酢酸ベタイン、2-アルキル-N-カルボキシ-N-ヒドロキシイミダゾリニウムベタインなどの両性界面活性剤;ポリオキシエチレン型、多価アルコールエステル型、エチレンオキシド・プロピレンオキシドブロック共重合体などの非イオン界面活性剤などを挙げることができる。また特に制限されることなく、高分子界面活性剤又は天然界面活性剤に属する界面活性剤も使用することができる。
防腐剤の具体例として、パラオキシ安息香酸エチル、サリチル酸、及びソルビン酸などを例示することができる。増粘剤の具体例として、キサンタンガム、カルボキシメチルセルロースナトリウム、カルボキシビニルポリマーなどを例示することができる。金属封鎖剤の具体例として、エチレンジアミン四酢酸のナトリウム塩、リン酸、クエン酸などを例示することができる。
保湿剤の具体例としては、ポリエチレングリコール、プロピレングリコール、グリセリン、1,3-ブチレングリコール、キシリトール、ソルビトール、マルチトール、コンドロイチン硫酸、ヒアルロン酸、ムコイチン硫酸、カロニン酸、アテロコラーゲン、乳酸ナトリウム、胆汁酸塩、dl-ピロリドンカルボン酸塩、短鎖可溶性コラーゲン、ジグリセリン(EO)PO付加物、イザヨイバラ抽出物、セイヨウノコギリソウ抽出物、メリロート抽出物などを例示することができる。
ビタミン類の具体例としては、ビタミンA油、レチノール、酢酸レチノール、パルミチン酸レチノールなどのビタミンA類;リボフラビン、酪酸リボフラビン、フラビンアデニンヌクレオチドなどのビタミンB2類;ピリドキシン塩酸塩、ピリドキシンジオクタノエートなどのビタミンB6類;L-アスコルビン酸、L-アスコルビン酸ジパルミチン酸エステル、L-アスコルビン酸-2-硫酸ナトリウム、dl-α-トコフェロール-L-アスコルビン酸リン酸ジエステルジカリウムなどのビタミンC類;パントテン酸カルシウム、D-パントテニルアルコール、パントテニルエチルエーテル、アセチルパントテニルエチルエーテルなどのパントテン酸類;エルゴカルシフェロール、コレカルシフェロールなどのビタミンD類;ニコチン酸、ニコチン酸ベンジル、ニコチン酸アミドなどのニコチン酸類;dl-α-トコフェロール、酢酸dl-α-トコフェロール、ニコチン酸dl-α-トコフェロール、コハク酸dl-α-トコフェロールなどのビタミンE類;ビタミンP;ビオチンなどを例示することができる。
本発明の化粧料組成物又は皮膚外用剤は、皮膚に適用(例えば、塗布又は噴霧など)される形態で使用される。具体的には、本発明の化粧料組成物又は皮膚外用剤は、化粧料、外用医薬品又は外用医薬部外品などとして使用される。これらの内、日常的にフィブリリンの産生を促進させることができるという観点から、化粧料が好ましい。なお、かかる化粧料又は外用剤には、養毛剤若しくは育毛剤、又は養毛若しくは育毛に効果のあるシャンプー、リンス、ヘアローション(トニック、リキッドを含む)などの各種のヘア化粧料が含まれる。
本発明の化粧料組成物又は皮膚外用剤の形態については、皮膚又は粘膜に適用可能なものであれば特に制限されず、例えば、ペースト状、ムース状、ジェル状、液状、乳液状、懸濁液状、クリーム状、軟膏状、シート状、エアゾール状、スプレー状、リニメント剤を例示することができる。特に化粧料とする場合であれば、ローション;エモリエント乳液、ミルキーローション、ナリシング乳液、クレンジング乳液などの乳液;エモリエントクリーム、マッサージクリーム、クレンジングクリーム、メイクアップクリームなどのクリームなどを例示することができる。また、特に養毛剤、育毛剤などのヘア製品とする場合であれば、トニック、ヘアクリーム、ヘアローション、エアゾール(噴霧剤)、ムース、シャンプー、リンス、リキッドなどを例示することができる。
本発明の化粧料組成物又は皮膚外用剤は、化粧料、外用医薬品又は外用医薬部外品として、皮膚又は粘膜に直接塗布、噴霧又は貼付して使用することができる。その使用割合は、使用者(ヒトの年齢、性別、用途、患部の症状の程度など)に応じて適宜選択可能であり、特に制限されるものではない。例えば、フィブリリンの産生を促進する有効量を、1回/日から5回/日の回数で皮膚に経皮的に投与することが例示される。
本発明の化粧料組成物又は皮膚外用剤を適用する際の適用量としては、ピリミジンヌクレオチド又はその前駆体の量にして1回あたり1mg以上が好ましく、1~1000mgがより好ましく、5~800mgがより好ましく、10~500mgがより好ましい。
これまでに記載した内容から明らかであるが、本発明は、ピリミジンヌクレオチドがフィブリリン産生を促進するという新規な属性を発見し、当該属性により、ピリミジンヌクレオチドが、単独でも、フィブリリン産生促進剤という新たな用途への使用に適することを見いだしたことに基づく発明である。
本発明の一態様は、エラスチン線維の産生を促進するエラスチン線維産生促進剤である。本発明のエラスチン線維産生促進剤は、フィブリリンの産生促進を通じて、エラスチン線維の産生を促進することができる。
エラスチン線維産生促進剤の適用量及び投与量は、上記の化粧料組成物又は皮膚外用剤を適用する際の適用量、及び経口投与剤の投与量と同様にすることができる。

本発明のエラスチン線維産生促進剤は、フィブリリンの産生促進を通じて皮膚の触覚的ハリ改善、肺気腫の治療、肺気腫の進展予防、動脈硬化の治療、動脈硬化の進展予防より選択される1つ以上の効果を奏することができる。

(実施例1)フィブリリン産生促進効果の評価
ピリミジンヌクレオチドのフィブリリン産生促進効果を評価した。
ピリミジンヌクレオチドのフィブリリン産生促進効果を評価した。
以下の手順に従って、実験を行った。
(正常ヒト表皮細胞の培養上清取得)
正常ヒト表皮細胞(NHEK;クラボウ製)を、96穴プレートに2.5×104cells/wellの密度で播種し、37℃で24時間培養した。100μLのサンプル含有培地(HuMedia-KG2;クラボウ製)に交換し、24時間培養した。培養後、培養上清を回収した。なお、サンプル溶液としては、CMP,2Na 0、2.5、5(mM)(すなわち、0、0.0918、0.184(w/v%))、及びUMP,2Na 0、2.5、5(mM)(すなわち、0、0.092、0.184(w/v%))をそれぞれ用いた。
正常ヒト表皮細胞(NHEK;クラボウ製)を、96穴プレートに2.5×104cells/wellの密度で播種し、37℃で24時間培養した。100μLのサンプル含有培地(HuMedia-KG2;クラボウ製)に交換し、24時間培養した。培養後、培養上清を回収した。なお、サンプル溶液としては、CMP,2Na 0、2.5、5(mM)(すなわち、0、0.0918、0.184(w/v%))、及びUMP,2Na 0、2.5、5(mM)(すなわち、0、0.092、0.184(w/v%))をそれぞれ用いた。
(正常ヒト線維芽細胞への培養上清処理)
正常ヒト線維芽細胞(NHDF;クラボウ製)を、96穴プレートに2.5×104cells/well/100μLの密度で播種し、37℃で24時間培養した。その後、上記の3次元培養表皮の培養上清取得工程で得た培養上清100μLに交換し、24時間培養した。
正常ヒト線維芽細胞(NHDF;クラボウ製)を、96穴プレートに2.5×104cells/well/100μLの密度で播種し、37℃で24時間培養した。その後、上記の3次元培養表皮の培養上清取得工程で得た培養上清100μLに交換し、24時間培養した。

正常ヒト線維芽細胞(NHDF;クラボウ製)を、96穴プレートに2.55×104cells/well/100μLの密度で播種し、37℃で24時間培養した。その後、CMP,2Na 0、2.5、5(mM)(すなわち、0、0.0918、0.184(w/v%))、及びUMP,2Na 0、2.5、5(mM)(すなわち、0、0.092、0.184(w/v%))に調整した培地(HuMedia-KG2;クラボウ製)100μLに交換し、24時間培養した。
正常ヒト線維芽細胞(NHDF;クラボウ製)を、96穴プレートに2.55×104cells/well/100μLの密度で播種し、37℃で24時間培養した。その後、CMP,2Na 0、2.5、5(mM)(すなわち、0、0.0918、0.184(w/v%))、及びUMP,2Na 0、2.5、5(mM)(すなわち、0、0.092、0.184(w/v%))に調整した培地(HuMedia-KG2;クラボウ製)100μLに交換し、24時間培養した。
(フィブリリン-1量の定量)
正常ヒト線維芽細胞の培養上清を回収し、ELISA法によりフィブリリン-1量を定量した。培養上清は、BCA protein assayにてタンパク質量も測定した。

シチジル酸処理を行ったときのフィブリリン-1量の定量結果を、図1に示す。ウリジル酸処理を行ったときのフィブリリン-1量の定量結果を、図2に示す。
ピリミジンヌクレオチド処理を行った3次元培養表皮の培養上清の添加によって、フィブリリンの産生が促進されていた。ピリミジンヌクレオチドの添加によっても、フィブリリンの産生が促進されていた。シチジル酸処理とウリジル酸処理とを比較したとき、フィブリリン産生促進効果は同程度であった。
(実施例2)3次元培養表皮培養上清を用いた線維芽細胞に対するコラーゲン合成関連因子のmRNA発現量の評価
エラスチン合成関連因子であるFibrillin1(FBN1)のmRNA発現量をReal time PCRにて定量した。

(3次元培養表皮の培養上清取得)
3次元培養表皮(LabCyte EPI MODEL 24 J TEC 製)を、付属のアッセイ培地を500μL添加した24穴プレートにセットし、37℃で一晩馴化した。サンプル含有PBS(-)を角層側から50μL適用し、24時間培養した。サンプル含有PBS(-)を除去後、PBS(-)にて洗浄した。新鮮培地に交換し、新たにサンプル含有PBS(-)を角層側から50μL適用し、さらに48時間培養した。培養後、培養上清を回収した。なお、各サンプル中のCMP,2Na濃度は0、1.0、2.0(w/v%)の各濃度で実施した。

(正常ヒト線維芽細胞への培養上清処理とCollagen 1A1のmRNA発現量の定量)
実施例1と同様の方法で、正常ヒト線維芽細胞への培養上清処理を行った。RNeasy Mini Kit(QIAGEN製)を用いてNHDFからRNAを回収し、Real time PCRによるFibrillin-1(FBN-1)のmRNA発現解析を行った(ΔΔCt法)。なお、内在性コントロールとしてはグリセルアルデヒド3-リン酸脱水素酵素(GAPDH)を用いた。
実施例1と同様の方法で、正常ヒト線維芽細胞への培養上清処理を行った。RNeasy Mini Kit(QIAGEN製)を用いてNHDFからRNAを回収し、Real time PCRによるFibrillin-1(FBN-1)のmRNA発現解析を行った(ΔΔCt法)。なお、内在性コントロールとしてはグリセルアルデヒド3-リン酸脱水素酵素(GAPDH)を用いた。
FBN1のmRNA発現解析結果を図3に示す。
処理したCMP,2Naの濃度依存的に、FBN-1の発現量が増大していることが明らかとなった。
(実施例3)新鮮ヒト摘出皮膚を用いたex vivo評価試験
実際の皮膚に対する作用をより高精度に評価するため、新鮮ヒト摘出皮膚を用いたex vivo評価試験を実施した。

研究用途としての利用についてインフォームドコンセントを得た39歳白人女性患者から美容整形手術時に採取された腹部皮膚組織を使用した。この摘出皮膚を、D-MEM/Ham´s F-12培地を1.3ml添加した培養プレートにセットし、5%CO2下、37℃で一晩馴化した。所定濃度のサンプルを含有したPBS(-)を角層側から適量適用し、さらに24時間培養した。サンプルと培地は24時間ごとに新しいものに交換しながら、合計72時間処理を行った。なお、各サンプル中のCMP,2Na濃度は0、1.0、2.0(w/v%)の各濃度で実施した。
定法に従って培養後の皮膚組織の固定化を行った後、皮膚組織切片を作成し、エラスチカ・ワンギーソン染色にてエラスチン線維を染色し、顕微鏡観察を行った。
エラスチカ・ワンギーソン染色による組織解析の結果を、図4に示す。
新鮮ヒト摘出皮膚においても、シチジル酸処理によってエラスチン線維が密になっている傾向が認められた。
本実施例の結果より、実際の皮膚においても、ピリミジンヌクレオチド又はその前駆体によってフィブリリン産生が促進され、エラスチン線維の産生が促進されることが強く示唆された。
Claims (22)
- ピリミジンヌクレオチド又はその前駆体を有効成分として含有する、フィブリリン産生促進剤。
- 前記ピリミジンヌクレオチド又はその前駆体の濃度が0.05~10.0(w/v%)である、請求項1に記載のフィブリリン産生促進剤。
- 皮膚の触覚的ハリ改善、肺気腫の治療、肺気腫の進展予防、動脈硬化の治療、及び動脈硬化の進展予防より選択される1つ以上の効果を奏する、請求項1又は2に記載のフィブリリン産生促進剤。
- ピリミジンヌクレオチド又はその前駆体を有効成分として含有する、フィブリリンの産生を促進するための化粧料組成物又は皮膚外用剤。
- 前記ピリミジンヌクレオチド又はその前駆体の濃度が0.05~10.0(w/v%)である、請求項4に記載の化粧料組成物又は皮膚外用剤。
- 皮膚の触覚的ハリ改善、肺気腫の治療、肺気腫の進展予防、動脈硬化の治療、及び動脈硬化の進展予防より選択される1つ以上の効果を奏する、請求項4又は5に記載の化粧料組成物又は皮膚外用剤。
- ピリミジンヌクレオチド又はその前駆体を有効成分として含有する、フィブリリンの産生を促進するため経口投与剤。
- 前記ピリミジンヌクレオチド又はその前駆体の濃度が0.05~10.0(w/v%)である、請求項7に記載の経口投与剤。
- 皮膚の触覚的ハリ改善、肺気腫の治療、肺気腫の進展予防、動脈硬化の治療、及び動脈硬化の進展予防より選択される1つ以上の効果を奏する、請求項7又は8に記載の経口投与剤。
- ピリミジンヌクレオチド又はその前駆体を有効成分として含有する、エラスチン線維産生促進剤。
- 前記ピリミジンヌクレオチド又はその前駆体の濃度が0.05~10.0(w/v%)である、請求項10に記載のエラスチン線維産生促進剤。
- 皮膚の触覚的ハリ改善、肺気腫の治療、肺気腫の進展予防、動脈硬化の治療、及び動脈硬化の進展予防より選択される1つ以上の効果を奏する、請求項10又は11に記載のエラスチン線維産生促進剤。
- 請求項4又は5に記載の化粧料組成物又は皮膚外用剤を皮膚に適用することによって、皮膚のフィブリリン産生を促進する方法。
- 前記化粧料組成物又は前記皮膚外用剤の1回あたりの適用量が1~1000mgである、請求項13に記載の方法。
- 請求項7又は8に記載の経口投与剤を経口投与することによって、フィブリリン産生を促進する方法。
- 前記経口投与剤の1回あたりの投与量が1~5000mgである、請求項15に記載の方法。
- 請求項10又は11に記載のエラスチン線維産生促進剤を皮膚に適用又は経口投与することによって、エラスチン線維産生を促進する方法。
- 前記エラスチン線維産生促進剤の適用量が1~1000mgであり、前記エラスチン線維産生促進剤の投与量が1~5000mgである、請求項17に記載の方法。
- フィブリリン産生の促進に用いるための、ピリミジンヌクレオチド又はその前駆体。
