ピリミジンヌクレオチド又はその前駆体を有効成分として含有する、コラーゲン産生促進剤である。
Description
本発明は、コラーゲン産生を促進する、コラーゲン産生促進剤、化粧料組成物又は皮膚外用剤、及び経口投与剤に関する。
高齢化社会が進んだ現代においては、健康長寿だけでなく、QOL(Quality of Life)の向上が課題となっている。アンチエイジング分野における皮膚の美容は、高齢者のQOL向上に極めて重要であることから関心が高まっている。
老化に伴い、皮膚はシミ、シワ、タルミといった外見的変化を生じる。これらの外見的変化は、高齢者のQOLを顕著に低下させる。これらの外見的変化の主要な要因の一つとして、加齢に伴うコラーゲンの減少が注目されている。
皮膚は、表皮、真皮、皮下組織の3層からなっており、真皮の大部分はコラーゲンから構成されている。コラーゲン分子は、分子量約10万のポリペプチド鎖が3本絡み合った3重らせん構造を有している。コラーゲン分子は、真皮細胞に弾力性、吸水性、膨潤性をもたらすと考えられている(非特許文献1)。
コラーゲンは老化に伴って減少することが知られている。その原因としては、コラーゲンと糖とが結合する糖化、表皮細胞への紫外線刺激を端緒とするコラーゲン分解酵素の産生などが知られている。コラーゲンが減少することにより、細胞は弾力性、吸水性、膨潤性を失う。すなわち、肌のハリがなくなる、保水性が失われる、肌が弛むといった変化が生じる。
コラーゲンの減少によってシミ、シワ、タルミといった外見的変化が生じることから、それらの老化現象を防止するために、皮膚中のコラーゲン量を高める検討がなされてきた。その一例としては、コラーゲンの経口摂取、及びコラーゲンの化粧料組成物又は皮膚外用剤としての使用が挙げられる。しかし、コラーゲンの経口摂取については、コラーゲンが消化管で分解されてから吸収されることから、直接的に皮膚中のコラーゲン量を増やすものではないと考えられている。コラーゲンの化粧料組成物又は皮膚外用剤としての使用については、高分子であるコラーゲンは皮膚中に浸透することは出来ないことから、皮下のコラーゲン量の増加効果はないと考えられている。
そこで近年では、線維芽細胞のコラーゲン産生能に注目し、線維芽細胞のコラーゲン産生を促進させることで、皮膚のコラーゲン量を増加させる方法に注目が集まっている。
特許文献1には、プリン系核酸関連物質及びピリミジン系核酸関連物質を含有する、コラーゲン産生促進用組成物が記載されている。しかし、特許文献1に実質的に開示された発明は、アデニル酸とウリジル酸との併用によるコラーゲン産生促進用組成物であり、活性の主体はアデニル酸である。特許文献1には、ウリジル酸単体での使用及びシチジル酸の使用に関して実質的な開示はない。さらに、特許文献1には、ウリジル酸単体ではコラーゲン産生促進作用がないことが記載されている。
加えて、特許文献1にて示された実施例では、培養ヒト皮膚線維芽細胞に試料を直接適用することで試験を行っている。しかし、線維芽細胞は真皮に存在しているため、外用組成物を線維芽細胞に直接届けることはできない。すなわち、特許文献1に記載の実験系は、試料を皮膚外用剤として用いた際、皮膚で起きる現象を精確に評価することのできない系である。

本発明の課題は、コラーゲン産生促進効果を有する、新規かつ安全性の高いコラーゲン産生促進剤、化粧料組成物又は皮膚外用剤、及び経口投与剤を提供することにある。
本発明者は、上記目的を達成するため鋭意検討を行った結果、驚くべきことに、これまでコラーゲン産生促進作用が全くないと考えられていたピリミジンヌクレオチド又はその前駆体が、表皮細胞を介することで、単独でも線維芽細胞におけるコラーゲン産生を促進するという属性を有することを初めて見出し、本発明をなすに至った。
すなわち、本発明は、ピリミジンヌクレオチド又はその前駆体を有効成分として含有する、コラーゲン産生促進剤である。
また、本発明は、ピリミジンヌクレオチド又はその前駆体を有効成分として含有する、コラーゲンの産生を促進するための化粧料組成物又は皮膚外用剤である。
さらに、本発明は、ピリミジンヌクレオチド又はその前駆体を有効成分として含有する、コラーゲンの産生を促進するための経口投与剤である。
また、本発明は、ピリミジンヌクレオチド又はその前駆体を有効成分として含有する、コラーゲンの産生を促進するための化粧料組成物又は皮膚外用剤である。
さらに、本発明は、ピリミジンヌクレオチド又はその前駆体を有効成分として含有する、コラーゲンの産生を促進するための経口投与剤である。
本発明のコラーゲン産生促進剤、化粧料組成物又は皮膚外用剤、及び経口投与剤は、コラーゲン産生促進効果を有し、新規かつ安全性が高い。特に、本発明の化粧料組成物又は皮膚外用剤、及び経口投与剤は、表皮細胞を介して、線維芽細胞におけるコラーゲンの産生を促進させることができる。
本発明は、ピリミジンヌクレオチド又はその前駆体を有効成分として含有する、コラーゲン産生促進剤、コラーゲンの産生を促進するための化粧料組成物又は皮膚外用剤、及び経口投与剤に関する。
ここで、本明細書において「コラーゲン産生促進剤」とは、ピリミジンヌクレオチド又はその前駆体を有効成分として含有するものであり、その他の成分を更に含有する組成物の形態を包含する概念である。同様に、本明細書において「皮膚外用剤」とは、ピリミジンヌクレオチド又はその前駆体を有効成分として含有するものであり、その他の成分を更に含有する組成物の形態を包含する概念である。
ここで、本明細書において「コラーゲン産生促進剤」とは、ピリミジンヌクレオチド又はその前駆体を有効成分として含有するものであり、その他の成分を更に含有する組成物の形態を包含する概念である。同様に、本明細書において「皮膚外用剤」とは、ピリミジンヌクレオチド又はその前駆体を有効成分として含有するものであり、その他の成分を更に含有する組成物の形態を包含する概念である。
本明細書において、ピリミジンヌクレオチドとは、シチジル酸及び/又はウリジル酸を意味する。
シチジル酸(シチジン一リン酸、cytidine 5'-phosphate、CMP)は、CAS登録番号63-37-6で示される化合物である。本明細書でシチジル酸と記載する場合、シチジル酸の塩も包含される。
本明細書において、シチジル酸の質量について記載する場合、シチジル酸2ナトリウム塩(CMP,2Na)に換算した際の質量を表す。シチジル酸溶液の濃度(%)について記載する場合、特に記載がなければ質量体積パーセント濃度(w/v%)であることとし、シチジル酸質量としてはCMP,2Na換算質量を用いる。2ナトリウム塩以外の塩を選択する場合、又は塩を形成しない遊離酸の場合には、シチジル酸の物質量を基準として、CMP,2Naに換算した場合の質量とする。
ウリジル酸(ウリジン一リン酸、uridine 5'-phosphate、UMP)は、CAS登録番号58-97-9で示される化合物である。本明細書でウリジル酸と記載する場合、ウリジル酸の塩も包含する概念である。
本明細書において、ウリジル酸の質量について記載する場合、ウリジル酸2ナトリウム塩(UMP,2Na)に換算した際の質量を表す。ウリジル酸の濃度(%)について記載する場合、特に記載がなければ質量体積パーセント濃度(w/v%)であることとし、ウリジル酸質量としてはUMP,2Na換算質量を用いる。2ナトリウム塩以外の塩を選択する場合、及び塩を形成しない遊離酸の場合には、ウリジル酸の物質量を基準として、UMP,2Naに換算した場合の質量とする。
本明細書において、ピリミジンヌクレオチド前駆体とは、ピリミジンヌクレオチド、すなわちシチジル酸及び/又はウリジル酸へと代謝され得る化合物を意味する。化合物がピリミジンヌクレオチド前駆体に包含されるか否かは、その化合物がピリミジンヌクレオチドに変換されるという知見の有無によって判断される。具体的には、エクトヌクレオチダーゼなどの働きによりシチジル酸及び/又はウリジル酸へ分解されることが知られているシチジン二リン酸、シチジン三リン酸、ウリジン二リン酸、ウリジン三リン酸(松岡功,「神経系におけるエクトヌクレオチダーゼ」,臨床化学 33:11-18,2004)、キナーゼの働きによってシチジル酸及び/又はウリジル酸へとリン酸化されることが知られているシチジン、シトシン、ウリジン、ウラシル(A Orengo, 「Regulation of enzymic activity by metabolites. I. Uridine-cytidine kinase of Novikoff ascites rat tumor」, J Biol Chem. 1969 Apr 25;244(8):2204-9.)が、本明細書中のピリミジンヌクレオチド前駆体として例示される。
本明細書において、本発明のコラーゲン産生促進作用は、I型コラーゲン量を定量することによって評価する。具体的な定量方法については、後述の実施例に記載する。
本発明におけるピリミジンヌクレオチド又はその前駆体としては、前述の通り、シチジン、シトシン、シチジル酸、シチジン二リン酸、シチジン三リン酸、ウリジン、ウラシル、ウリジル酸、ウリジル二リン酸、ウリジル三リン酸が例示される。これらの中でも、シチジル酸、ウリジル酸が好ましい。
本発明におけるシチジル酸の概念は、前述の通り塩を包含する。シチジル酸の塩としては、ナトリウム塩、カリウム塩などのアルカリ金属塩;カルシウム塩、マグネシウム塩、バリウム塩などのアルカリ土類金属塩;アルギニン、リジンなどの塩基性アミノ酸塩;アンモニウム塩、トリシクロヘキシルアンモニウム塩などのアンモニウム塩;モノエタノールアミン塩、ジエタノールアミン塩、トリエタノールアミン塩、モノイソプロパノールアミン塩、ジイソプロパノールアミン塩、トリイソプロパノールアミン塩などの各種のアルカノールアミン塩などを挙げることができる。これらの中でも、ナトリウム塩などのアルカリ金属塩が好ましい。アルカリ金属塩としては、具体的にはシチジル酸1ナトリウム、及びシチジル酸2ナトリウムを例示することができ、取り扱い性の面から、シチジル酸2ナトリウムが好ましい。
本発明におけるウリジル酸の概念は、前述の通り塩を包含する。ウリジル酸の塩としては、ナトリウム塩、カリウム塩などのアルカリ金属塩;カルシウム塩、マグネシウム塩、バリウム塩などのアルカリ土類金属塩;アルギニン、リジンなどの塩基性アミノ酸塩;アンモニウム塩、トリシクロヘキシルアンモニウム塩などのアンモニウム塩;モノエタノールアミン塩、ジエタノールアミン塩、トリエタノールアミン塩、モノイソプロパノールアミン塩、ジイソプロパノールアミン塩、トリイソプロパノールアミン塩などの各種のアルカノールアミン塩などを挙げることができる。これらの中でも、ナトリウム塩などのアルカリが好ましい。アルカリ金属塩としては、具体的にはウリジル酸1ナトリウム、及びウリジル酸2ナトリウムを例示することができ、取り扱い性の面から、ウリジル酸2ナトリウムが好ましい。
本発明のコラーゲン産生促進剤、化粧料組成物又皮膚外用剤、及び経口投与剤中におけるピリミジンヌクレオチド又はその前駆体の含有量は0.025(w/v%)以上であることが好ましい。特に、コラーゲン産生が有意に促進されるという点で、含有量は0.046(w/v%)以上であることが好ましい。また、コラーゲン産生促進効果は濃度依存的に奏されることから、含有量は0.075(w/v%)以上であることが好ましく、0.1(w/v%)以上であることがより好ましく、0.125(w/v%)以上であることが更に好ましく、0.25(w/v%)以上であることがまた更に好ましく、0.5(w/v%)以上であることがまた更に好ましく、1.0(w/v%)以上であることが特に好ましい。また各種剤又は組成物として利用するときの扱い易さなどから、ピリミジンヌクレオチド又はその前駆体の含有量は10(w/v%)以下であることが好ましい。
本発明のコラーゲン産生促進剤、化粧料組成物又皮膚外用剤、及び経口投与剤には、有効成分であるピリミジンヌクレオチド又はその前駆体を単独で用いてもよく、ピリミジンヌクレオチド又はその前駆体を2種以上同時に使用しても良く、ピリミジンヌクレオチド又はその前駆体とそれ以外の有効成分とを併用して用いても良い。
本発明におけるピリミジンヌクレオチド又はその前駆体の由来には格別の制限はなく、酵母、細菌、魚介類、動物、植物などの天然物由来のものが好適である。
本発明のコラーゲン産生促進剤、化粧料組成物又は皮膚外用剤、及び経口投与剤は、上記成分の他に、薬学的若しくは香粧的に許容される基剤又は担体を組み合わせて各種の形態に調製されても良い。薬学的若しくは香粧的に許容される基剤又は担体については、従来公知のものを用いることができる。特に、化粧料組成物又は皮膚外用剤の場合、必要に応じて化粧料、外用の医薬品・医薬部外品などの皮膚又は粘膜に適用される外用組成物に配合される公知の各種成分を配合することができる。このような成分として、例えば、界面活性剤、色素(染料、顔料)、香料、防腐剤、殺菌剤(抗菌剤)、増粘剤、酸化防止剤、金属封鎖剤、清涼化剤、防臭剤、保湿剤、UV吸収剤、UV散乱剤、ビタミン類、植物エキス、皮膚収斂剤、抗炎症剤(消炎剤)、美白剤、細胞賦活剤、血管拡張剤、血行促進剤、及び皮膚機能亢進剤などを挙げることができる。
上記成分の内、界面活性剤の具体例としては、高級脂肪酸石けん、アルキル硫酸エステル塩、ポリオキシエチレンアルキルエーテル硫酸塩、アルキルエーテルリン酸エステル塩、N-アシルアミノ酸塩、アシルN-メチルタウリン塩などのアニオン界面活性剤;塩化アルキルトリメチルアンモニウム、塩化ジアルキルジメチルアンモニウムなどのカチオン界面活性剤;アルキルジメチルアミノ酢酸ベタイン、アルキルアミドジメチルアミノ酢酸ベタイン、2-アルキル-N-カルボキシ-N-ヒドロキシイミダゾリニウムベタインなどの両性界面活性剤;ポリオキシエチレン型、多価アルコールエステル型、エチレンオキシド・プロピレンオキシドブロック共重合体などの非イオン界面活性剤などを挙げることができる。また特に制限されることなく、高分子界面活性剤又は天然界面活性剤に属する界面活性剤も使用することができる。
防腐剤の具体例として、パラオキシ安息香酸エチル、サリチル酸、及びソルビン酸などを例示することができる。増粘剤の具体例として、キサンタンガム、カルボキシメチルセルロースナトリウム、カルボキシビニルポリマーなどを例示することができる。金属封鎖剤の具体例として、エチレンジアミン四酢酸のナトリウム塩、リン酸、クエン酸などを例示することができる。
保湿剤の具体例としては、ポリエチレングリコール、プロピレングリコール、グリセリン、1,3-ブチレングリコール、キシリトール、ソルビトール、マルチトール、コンドロイチン硫酸、ヒアルロン酸、ムコイチン硫酸、カロニン酸、アテロコラーゲン、乳酸ナトリウム、胆汁酸塩、dl-ピロリドンカルボン酸塩、短鎖可溶性コラーゲン、ジグリセリン(EO)PO付加物、イザヨイバラ抽出物、セイヨウノコギリソウ抽出物、メリロート抽出物などを例示することができる。
ビタミン類の具体例としては、ビタミンA油、レチノール、酢酸レチノール、パルミチン酸レチノールなどのビタミンA類;リボフラビン、酪酸リボフラビン、フラビンアデニンヌクレオチドなどのビタミンB2類;ピリドキシン塩酸塩、ピリドキシンジオクタノエートなどのビタミンB6類;L-アスコルビン酸、L-アスコルビン酸ジパルミチン酸エステル、L-アスコルビン酸-2-硫酸ナトリウム、dl-α-トコフェロール-L-アスコルビン酸リン酸ジエステルジカリウムなどのビタミンC類;パントテン酸カルシウム、D-パントテニルアルコール、パントテニルエチルエーテル、アセチルパントテニルエチルエーテルなどのパントテン酸類;エルゴカルシフェロール、コレカルシフェロールなどのビタミンD類;ニコチン酸、ニコチン酸ベンジル、ニコチン酸アミドなどのニコチン酸類;dl-α-トコフェロール、酢酸dl-α-トコフェロール、ニコチン酸dl-α-トコフェロール、コハク酸dl-α-トコフェロールなどのビタミンE類;ビタミンP、ビオチンなどを例示することができる。
本発明のコラーゲン産生促進剤は、化粧料、外用医薬品又は外用医薬部外品などに用いられる添加剤として使用することができる。また、本発明の化粧料組成物又は皮膚外用剤は、皮膚に適用(例えば、塗布又は噴霧など)される形態で使用される。具体的には、本発明の化粧料組成物又は皮膚外用剤は、化粧料、外用医薬品又は外用医薬部外品などとして使用される。これらの内、日常的にコラーゲンの産生を促進させることができるという観点から、化粧料が好ましい。なお、かかる化粧料又は外用剤には、養毛剤若しくは育毛剤、又は養毛若しくは育毛に効果のあるシャンプー、リンス、ヘアローション(トニック、リキッドを含む)などの各種のヘア化粧料が含まれる。
本発明の化粧料組成物又は皮膚外用剤の形態については、皮膚又は粘膜に適用可能なものであれば特に制限されず、例えば、ペースト状、ムース状、ジェル状、液状、乳液状、懸濁液状、クリーム状、軟膏状、シート状、エアゾール状、スプレー状、リニメント剤を例示することができる。特に化粧料とする場合であれば、ローション;エモリエント乳液、ミルキーローション、ナリシング乳液、クレンジング乳液などの乳液;エモリエントクリーム、マッサージクリーム、クレンジングクリーム、メイクアップクリームなどのクリームなどを例示することができる。また、特に養毛剤、育毛剤などのヘア製品とする場合であれば、トニック、ヘアクリーム、ヘアローション、エアゾール(噴霧剤)、ムース、シャンプー、リンス、リキッドなどを例示することができる。
本発明の化粧料組成物又は皮膚外用剤は、化粧料、外用医薬品又は外用医薬部外品として、皮膚又は粘膜に直接塗布、噴霧又は貼付して使用することができる。その使用割合は、使用者(ヒトの年齢、性別、用途、患部の症状の程度など)に応じて適宜選択可能であり、特に制限されるものではない。例えば、コラーゲンの産生を促進する有効量を、1回/日から5回/日の回数で皮膚に経皮的に投与することが例示される。
本発明の化粧料組成物又は皮膚外用剤を適用する際の適用量としては、ピリミジンヌクレオチド又はその前駆体の量に換算して1回あたり1mg以上が好ましく、1~1000mgがより好ましく、5~800mgがより好ましく、10~500mgがより好ましい。
本発明の経口投与剤は、医薬品、サプリメント、経腸栄養剤などとして実用に供することができる。この場合、ピリミジンヌクレオチド又はその前駆体を単独、又は製剤補助剤などと合わせて製剤化することができる。
前記製剤の形状としては、錠剤、顆粒剤、カプセル剤、粒剤、散剤、溶液剤、シロップ剤、乳剤などとすることができる。
前記製剤においては、本発明の有効成分の他に、賦形剤、結合剤、崩壊剤、滑沢剤、矯味矯臭剤、溶解補助剤、懸濁剤、コーティング剤などの任意の製剤補助剤を、それぞれの供与形態に合わせて適宜組み合わせて用いることができる。
本発明の経口投与剤の投与又は摂取量は、対象者の年令、体重、症状などの程度、投与又は摂取方法などに応じて変動するものの、1回あたりの投与量が1~5000mgであることが好ましく、1~1000mgであることがより好ましい。
本発明は、後述する実施例に示すように、表皮細胞を介することで線維芽細胞のコラーゲン産生を促進する。これは、これまでに知られていたピリミジンヌクレオチドの属性、及び適した用途とは全く異なっている。例えば、特許文献1(WO2005/034902号)には、「UMP2Na自体にはコラーゲン産生促進作用がない」と記載されている。コラーゲン産生促進作用がないピリミジンヌクレオチドは、当然ながらコラーゲン産生促進用途には好適ではないと考えられてきた。
これまでの知見とは正反対の、ピリミジンヌクレオチドがコラーゲン産生促進作用を奏するという結果が得られた理由としては、これまでの実験系とは異なる実験系を用いることで、新規の作用機序が発見されたことに起因すると考えられる。すなわち、特許文献1に代表されるこれまでの知見においては、線維芽細胞に直接UMPなどを処理していたが、この場合、コラーゲン産生促進効果は全く発揮されない。一方、本願実施例においては、表皮細胞にピリミジンヌクレオチドを処理し、その培養上清を線維芽細胞に処理することにより、顕著なコラーゲン産生促進効果が発揮されることを見出した。皮膚外用剤として使用されるときには、当然表皮に使用されることから、本発明に関する知見は、ピリミジンヌクレオチドの皮膚外用剤としての真価を発見したものである。
これまでに記載した内容から明らかであるが、本発明は、ピリミジンヌクレオチドが表皮細胞を介して線維芽細胞のコラーゲン産生を促進するという新規な属性を発見し、当該属性により、ピリミジンヌクレオチドが、単独でも、コラーゲン産生促進剤という新たな用途への使用に適することを見いだしたことに基づく発明である。したがって、本発明は、以下のように例示される。
[1] ピリミジンヌクレオチド又はその前駆体を有効成分として含有する、コラーゲン産生促進剤。
[2] 前記ピリミジンヌクレオチド又はその前駆体が、アデニン、アデノシン、アデノシンのリン酸エステル、ヒポキサンチン、イノシン、イノシン酸、及びこれらの塩よりなる群から選択される少なくとも1種のプリン系核酸関連物質ではない、[1]に記載のコラーゲン産生促進剤。

以下、本発明を実施例により説明するが、本発明はこれらによって何ら制限されるものではない。
(実施例1)3次元培養表皮の培養上清を用いた線維芽細胞に対するコラーゲン産生促進効果の評価
実際の表皮にて起こる現象を再現するため、3次元培養表皮の培養上清と線維芽細胞を用いて、ピリミジンヌクレオチドのコラーゲン産生促進効果を評価した。

以下の手順に従って、実験を行った。
(3次元培養表皮の培養上清取得)
3次元培養表皮(LabCyte EPI MODEL 24 J TEC 製)を、付属のアッセイ培地を500μL添加した24穴プレートにセットし、37℃で一晩馴化した。サンプル含有PBS(-)を角層側から50μL適用し、24時間培養した。サンプル含有PBS(-)を除去後、PBS(-)にて洗浄した。UVB処理を行う試料については、UVBランプ(TL20W/12RS;PHILIPS 製)にて、600mJ/cm2を照射した。UVB処理を行う試料、UVB処理を行わない試料ともに、新鮮培地に交換し、新たにサンプル含有PBS(-)を角層側から50μL適用し、さらに48時間培養した。培養後、培養上清を回収した。なお、各サンプル中のCMP,2Na濃度は0、0.125、0.25、0.5、1.0、2.0(w/v%)の各濃度で実施した。

正常ヒト線維芽細胞(NHDF;クラボウ製)を、96穴プレートに2.5×104cells/well/100μLの密度で播種し、37℃で24時間培養した。その後、上記の3次元培養表皮の培養上清取得工程で得た培養上清100μLに交換し、24時間培養した。

(I型コラーゲン量の定量)
正常ヒト線維芽細胞の培養上清を回収し、ELISA法によりI型コラーゲン量を定量した。培養上清は、BCA protein assayにてタンパク質量も測定した。

I型コラーゲン量の定量結果を、図1に示す。
UVB照射、非照射に関わらず、3次元培養表皮の培養上清にて処理することで、3次元培養表皮を処理したCMP,2Naの濃度に依存して、正常ヒト線維芽細胞の培養上清中のコラーゲン量が増大していた(図1)。
本実施例の結果から、CMP,2Naにて角層側から3次元培養表皮を処理し、その培養上清を線維芽細胞に処理することで、線維芽細胞によるコラーゲンの産生が促進されることが明らかとなった。
(実施例2)3次元培養表皮培養上清を用いた線維芽細胞に対するコラーゲン合成関連因子のmRNA発現量の評価
同様に、I型コラーゲン合成関連因子であるCollagen 1A1(COL1A1)のmRNA発現量をReal time PCRにて定量した。

実施例1と同様の方法で、正常ヒト線維芽細胞への培養上清処理を行った。RNeasy Mini Kit(QIAGEN製)を用いてNHDFからRNAを回収し、Real time PCRによるCollagen 1A1(COL1A1)のmRNA発現解析を行った(ΔΔCt法)。なお、内在性コントロールとしてはグリセルアルデヒド3-リン酸脱水素酵素(GAPDH)を用いた。
実施例1と同様の方法で、正常ヒト線維芽細胞への培養上清処理を行った。RNeasy Mini Kit(QIAGEN製)を用いてNHDFからRNAを回収し、Real time PCRによるCollagen 1A1(COL1A1)のmRNA発現解析を行った(ΔΔCt法)。なお、内在性コントロールとしてはグリセルアルデヒド3-リン酸脱水素酵素(GAPDH)を用いた。
COL1A1のmRNA発現解析結果を図2に示す。
3次元培養表皮の培養上清にて処理することで、3次元培養表皮を処理したCMP,2Naの濃度に依存して、COL1A1の発現量が増大していることが明らかとなった。
本実施例の結果から、CMP,2Naにて角層側から3次元培養表皮を処理し、その培養上清を線維芽細胞に処理することで、線維芽細胞のコラーゲン合成関連因子の発現量が高まることが明らかとなった。
(実施例3)正常ヒト表皮細胞培養上清を用いた線維芽細胞に対するコラーゲン産生促進効果の評価
続いて、正常ヒト表皮細胞の培養上清と線維芽細胞を用いて、ピリミジンヌクレオチドのコラーゲン産生促進効果を評価した。この評価では、ピリミジンヌクレオチドにて直接的に線維芽細胞を処理した場合と、ピリミジンヌクレオチドで処理した正常ヒト表皮細胞培養上清にて線維芽細胞を処理した場合とで効果を比較した。
続いて、正常ヒト表皮細胞の培養上清と線維芽細胞を用いて、ピリミジンヌクレオチドのコラーゲン産生促進効果を評価した。この評価では、ピリミジンヌクレオチドにて直接的に線維芽細胞を処理した場合と、ピリミジンヌクレオチドで処理した正常ヒト表皮細胞培養上清にて線維芽細胞を処理した場合とで効果を比較した。
(正常ヒト表皮細胞の培養上清取得)
正常ヒト表皮細胞(NHEK;クラボウ製)を、96穴プレートに2.5×104cells/wellの密度で播種し、37℃で24時間培養した。100μLのサンプル含有培地(HuMedia-KG2;クラボウ製)に交換し、24時間培養した。培養後、培養上清を回収した。なお、サンプル溶液としては、CMP,2Na 0、1.25、2.5、5(mM)(すなわち、0、0.0459、0.0918、0.184(w/v%))、及びUMP,2Na 0、1.25、2.5、5(mM)(すなわち、0、0.046、0.092、0.184(w/v%))をそれぞれ用いた。
正常ヒト表皮細胞(NHEK;クラボウ製)を、96穴プレートに2.5×104cells/wellの密度で播種し、37℃で24時間培養した。100μLのサンプル含有培地(HuMedia-KG2;クラボウ製)に交換し、24時間培養した。培養後、培養上清を回収した。なお、サンプル溶液としては、CMP,2Na 0、1.25、2.5、5(mM)(すなわち、0、0.0459、0.0918、0.184(w/v%))、及びUMP,2Na 0、1.25、2.5、5(mM)(すなわち、0、0.046、0.092、0.184(w/v%))をそれぞれ用いた。
(正常ヒト線維芽細胞への培養上清処理)

(I型コラーゲン量の定量)

I型コラーゲン量の定量結果を、図3及び図4に示す。
正常ヒト表皮細胞培地に添加されたCMP,2Naの濃度に依存して、正常ヒト線維芽細胞の培養上清中のコラーゲン量が増大していた(図3)。一方、CMP,2Naを直接正常ヒト線維芽細胞に処理した群では、コラーゲン量の増大は見られなかった。
正常ヒト表皮細胞培地に添加されたUMP,2Naの濃度に依存して、正常ヒト線維芽細胞の培養上清中のコラーゲン量が増大していた(図4)。一方、UMP,2Naを直接正常ヒト線維芽細胞に処理した群では、コラーゲン量の増大は見られなかった。
本実施例の結果から、ピリミジンヌクレオチドを含む皮膚外用剤を皮膚に処置した場合、表皮細胞を経由して線維芽細胞に作用することによって、線維芽細胞におけるコラーゲン産生が促進されることが強く示唆された。
シチジル酸とウリジル酸のコラーゲンの産生促進効果を比較すると、シチジル酸の方が、より高いコラーゲン産生促進効果を奏した。
一方、ピリミジンヌクレオチドを線維芽細胞に直接処理した場合には、WO2005/034902号などの既報と同じく、コラーゲン産生促進作用は示されないことが確認された。
(実施例4)新鮮ヒト摘出皮膚を用いたex vivo評価試験
実際の皮膚に対する作用をより高精度に評価するため、新鮮ヒト摘出皮膚を用いたex vivo評価試験を実施した。

研究用途としての利用についてインフォームドコンセントを得た39歳白人女性患者から美容整形手術時に採取された腹部皮膚組織を使用した。この摘出皮膚を、D-MEM/Ham´s F-12培地を1.3ml添加した培養プレートにセットし、5%CO2下、37℃で一晩馴化した。所定濃度のサンプルを含有したPBS(-)を角層側から適量適用し、さらに24時間培養した。サンプルと培地は24時間ごとに新しいものに交換しながら、合計72時間処理を行った。なお、各サンプル中のCMP,2Na濃度は0、1.0、2.0(w/v%)の各濃度で実施した。
定法に従って培養後の皮膚組織の固定化を行った後、皮膚組織切片を作成し、エラスチカ・ワンギーソン染色にてコラーゲン線維を染色し、顕微鏡観察を行った。
エラスチカ・ワンギーソン染色による組織解析の結果を、図5に示す。
新鮮ヒト摘出皮膚においても、シチジル酸処理によってコラーゲン線維が密になっている傾向が認められた。
本実施例の結果より、実際の皮膚においても、ピリミジンヌクレオチド又はその前駆体によってコラーゲン産生が促進されることが強く示唆された。
