Abstract
本発明は、動物細胞内のNAD+量を増加する作用を有し、かつ動物細胞に対する細胞毒性が低い低分子化合物を提供することを課題とする。 以下の式(A-1)で表される化合物又はその生理的に許容される塩を用いる。
Description
本発明は、4-(2,4-ジフルオロフェニル)-2-(1H-インドール-3-イル)-4-オキソ-ブタン酸(以下、「MA5」ということがある)のエナンチオマーであるR体又はその生理的に許容される塩;MA5のR体又はその生理的に許容される塩を含む、哺乳動物細胞における酸化型ニコチンアミドアデニンジヌクレオチドの生成促進剤(以下、「本件生成促進剤」ということがある);MA5のR体又はその生理的に許容される塩を含む、ミトコンドリア病の治療剤若しくは予防剤(以下、「本件ミトコンドリア病治療/予防剤」ということがある);MA5のR体又はその生理的に許容される塩を含む、加齢に伴う症状若しくは疾患の治療剤又は予防剤(以下、「本件抗加齢剤」ということがある);等に関する。
本発明者らは、4-(2,4-ジフルオロフェニル)-2-(1H-インドール-3-イル)-4-オキソ-ブタン酸(すなわち、MA5)のラセミ体が、エリスロポエチン発現増強効果及びミトコンドリア病の治療効果(特許文献1)、臓器線維化抑制効果(特許文献2)、難聴の予防又は改善効果(特許文献3)を有することを報告している。しかしながら、MA5のエナンチオマーであるR体が、動物細胞内の酸化型ニコチンアミドアデニンジヌクレオチド(NAD;nicotinamide adenine dinucleotide)(NAD+)量を増加する作用を有することはこれまで知られていなかった。
本発明の課題は、動物細胞内のNAD+量を増加する作用を有し、かつ動物細胞に対する細胞毒性が低い低分子化合物を提供することにある。
本発明は、上記課題を解決すべく鋭意研究を続けている。その過程において、動物細胞を、MA5のラセミ体の存在下で培養しても、MA5のラセミ体非存在下で培養した場合と比べ、細胞内のNAD+量はほとんど変わらなかったのに対して、MA5のラセミ体を、MA5のR体と、MA5のS体に分離し、それぞれの存在下で動物細胞を培養したところ、MA5のS体は、動物細胞におけるNAD+量を減少させる作用を有するのに対して、MA5のR体は、逆に、動物細胞におけるNAD+量を増加させる作用を有することを見いだした。すなわち、MA5が、見かけ上、動物細胞におけるNAD+の生成を促進する作用を有さないように見えたのは、MA5のR体とMA5のS体が、驚いたことに互いに相反する作用を有していた結果、作用効果が相殺されたためであり、今回、光学分割することにより、その事実が初めて明らかとなった。
また、MA5のR体は、MA5のS体と比べ、生体内において毒性物質(グルクロン酸抱合体)に代謝されにくく、血中で保持されやすく、かつ動物細胞に対する細胞毒性が低いことも確認した。また、MA5のR体は、MA5のS体とは異なり、動物細胞におけるSIRT(Sirtuin)の発現量を増加させる作用を有することも確認した。
さらに、細胞におけるミトコンドリア機能を改善する作用を有するものは、MA5のS体ではなく、MA5のR体であることも確認した。本発明は、これらの知見に基づいて完成するに至ったものである。
すなわち、本発明は、以下のとおりである。

本発明の化合物であるMA5のR体は、MA5のラセミ体やMA5のS体とは異なり、動物細胞におけるNAD+量を増加させる作用を有する。また、MA5のR体は、MA5のS体とは異なり、動物細胞におけるSIRTの発現量を増加させる作用を有する。さらに、MA5のR体は、MA5のS体と比べ、生体内において毒性物質(グルクロン酸抱合体)に代謝されにくく、血中で保持されやすく、かつ動物細胞に対する細胞毒性が低い。
一方、動物細胞内のNAD+レベルは加齢とともに低下することや、加齢に関連する疾患の発症に重要な役割を担っていることが知られている。また、NAD+量を増加させると、線虫(C. elegans)においては寿命を延長し、マウスにおいては代謝性疾患に対する保護作用をもたらすことが報告されている(文献「Nature. 2018 563(7731) 354-359.」参照)。また、線虫は、ヒト老化のモデル生物として利用されている(文献「東海大学先進生命科学研究所紀要 第1巻 2017年3月」参照)。
このため、MA5のR体を動物に投与すると、副作用などの悪影響を及ぼすことなく、動物細胞におけるNAD+量を増加させた結果、寿命を延長したり、加齢に伴う症状又は疾患や代謝性疾患などの症状若しくは疾患を治療又は予防できることが十分期待できる。
さらに、本発明の化合物であるMA5のR体は、細胞におけるミトコンドリア機能を改善する作用を有することから、ミトコンドリア機能賦活剤やミトコンドリア病の治療剤又は予防剤として有用である。
本発明の化合物は、以下の式(A-1)で表される化合物(すなわち、以下の式(A)で表される化合物(4-(2,4-ジフルオロフェニル)-2-(1H-インドール-3-イル)-4-オキソ-ブタン酸のエナンチオマーであるR体)又はその生理的に許容される塩である。
本明細書において、「生理的に許容される塩」とは、妥当な医学的、薬学的、又は生物学的判断の範囲内で、動物の組織と接触して用いるのに、過度の毒性、刺激性、アレルギー応答、及びその他の問題や合併症を伴うことなく、適度な受益性/危険性比率に相応して適している塩を意味する。生理的に許容される塩としては、例えば、アンモニウム塩;ナトリウム塩、リチウム塩、カリウム塩等のアルカリ金属塩;アルミニウム塩、カルシウム塩、マグネシウム塩等のアルカリ土類金属塩;ジシクロヘキシルアミン塩、N-メチル-D-グルカミン等の有機塩基との塩;アルギニン、リシン、オルニチン等のアミノ酸との塩;塩基性窒素含有基により生じる塩;などを挙げることができる。
本発明における式(A-1)で表される化合物は、公知の有機化学反応を用いる有機合成手法、例えば、後述する実施例における「1.MA5のR体の生成」の項目に記載の方法に従って得ることができる。
本件生成促進剤は、「動物細胞におけるNAD+の生成を促進するため」という用途に特定された、MA5のR体又はその生理的に許容される塩を含有する剤である。また、本件ミトコンドリア病治療/予防剤は、「ミトコンドリア病を治療又は予防するため」という用途に特定された、MA5のR体又はその生理的に許容される塩を含有する剤である。また、本件抗加齢剤は、「加齢に伴う症状又は疾患を治療若しくは予防するため」という用途に特定された、MA5のR体又はその生理的に許容される塩を含有する剤である。また、また、本件ミトコンドリア機能賦活剤は、「動物におけるミトコンドリア機能を賦活(化)するため」という用途に特定された、MA5のR体又はその生理的に許容される塩を含有する剤である。本件剤は、有効成分であるMA5のR体又はその生理的に許容される塩を、単独で飲食品又は医薬品(製剤)として使用してもよいし、さらに添加剤を混合し、組成物の形態(飲食品組成物又は医薬組成物)として使用してもよい。例えば、本件剤は、実質的にMA5のS体又はその生理的に許容される塩を含まない、MA5のR体又はその生理的に許容される塩を有効成分としてなる、医薬組成物、又は飲食品組成物であり、実質的にMA5のS体又はその生理的に許容される塩を含まない、MA5のR体又はその生理的に許容される塩を含有する医薬組成物、又は飲食品組成物である。かかる飲食品としては、例えば、健康食品(機能性食品、栄養補助食品、健康補助食品、栄養強化食品、栄養調整食品、サプリメント等)、保健機能食品(特定保健用食品、栄養機能食品、機能性表示食品等)を挙げることができる。
本明細書において、動物としては、哺乳類(ヒト又は非ヒト哺乳動物)、鳥類、爬虫類、両生類、魚類、無脊椎動物等を挙げることができる。また、動物の他の態様として、ヒト、家畜を挙げることができる。ここで「家畜」とは、ヒトが飼育し、繁殖させた動物を意味する。かかる家畜としては、非ヒト哺乳動物(例えば、マウス、ラット、ハムスター、モルモット等のげっ歯類;ウサギ等のウサギ目;ブタ、ウシ、ヤギ、ウマ、ヒツジ等の有蹄目;イヌ、ネコ等のネコ目など)、鳥類(例えば、ニワトリ、ウズラ、シチメンチョウ、ハト、アヒル、ガチョウ等)、魚類(例えば、コイ、キンギョ等)、無脊椎動物(例えば、カイコ、ミツバチ等)などを例示することができる。
本件剤の添加剤としては、生理的に許容される通常の担体、結合剤、安定化剤、賦形剤、希釈剤、pH緩衝剤、崩壊剤、等張剤、添加剤、被覆剤、可溶化剤、潤滑剤、滑走剤、溶解補助剤、滑沢剤、風味剤、甘味剤、溶剤、ゲル化剤、栄養剤等の配合成分を例示することができる。かかる配合成分としては、具体的に、水、生理食塩水、動物性脂肪及び油、植物油、乳糖、デンプン、ゼラチン、結晶性セルロース、ガム、タルク、ステアリン酸マグネシウム、ヒドロキシプロピルセルロース、ポリアルキレングリコール、ポリビニルアルコール、グリセリンを例示することができる。
本件生成促進剤の投与対象としては、動物細胞におけるNAD+の生成を促進する必要がある動物(対象)であればよく、また、本件ミトコンドリア病治療/予防剤の投与対象としては、ミトコンドリア病を治療若しくは予防する必要がある動物(対象)であればよく、また、本件抗加齢剤の投与対象としては、加齢に伴う症状若しくは疾患を治療又は予防する必要がある動物(対象)であればよく、また、本件ミトコンドリア機能賦活剤の投与対象としては、ミトコンドリア機能を賦活する必要がある動物(対象)であればよい。
上記「ミトコンドリア病」としては、細胞核DNAやミトコンドリアDNAにおける遺伝子変異等により、ATP産生、アポトーシスの調節、カルシウムイオンや鉄の細胞内濃度の調節等のミトコンドリア機能が低下することが原因の症状であればよく、具体的にはCPEO(慢性進行性外眼麻痺症候群)、MELAS(メラス:脳筋症・乳酸アシドーシス・脳卒中様症候群)、MERRF(マーフ:赤色ぼろ線維・ミオクローヌスてんかん症候群)、Leigh脳症(リー脳症:亜急性壊死性脳脊髄症)、Leber病(レーバー病:レーバー遺伝性視神経症[LHON])、カーンズ・セイヤー症候群(KSS)、バース症候群、Pearson病(ピアソン病)、フリードライヒ失調症(FRDA)を挙げることができる。
上記「加齢に伴う症状若又は疾患」としては、例えば、薄毛又は脱毛(より具体的には、加齢性薄毛又は脱毛)、難聴(より具体的には、加齢性難聴)、視力障害(より具体的には、加齢性視力障害)、貧血(より具体的には、加齢性貧血)、筋力低下(より具体的には、加齢性筋力低下)、寿命短縮(より具体的には、加齢性寿命短縮)等を挙げることができる。
本明細書において、「動物細胞におけるNAD+の生成を促進する必要がある動物(対象)」としては、例えば、寿命を延長する必要がある対象(長寿を必要とする対象);神経疾患(パーキンソン、うつ、アルツハイマー病、筋萎縮性側索硬化症[ALS]など)患者;循環器疾患(心不全、不整脈など)患者;筋肉疾患(サルコペニア、封入体筋炎、筋ジストロフィーなど)患者;腎疾患(慢性腎臓病、腎不全、糖尿病性腎症、腎炎など)患者;代謝性疾患(糖尿病、肝機能障害、アルコール性肝障害、非アルコール性脂肪性肝疾患[NAFLD;non-alcoholic fatty liver disease])、非アルコール性脂肪肝炎[NASH;non-alcoholic steatohepatitis]、甲状腺・副腎疾患など)患者;消化器系疾患(炎症性腸疾患など)患者;癌患者;加齢に伴う症状又は疾患(薄毛又は脱毛、難聴、視力障害、貧血、筋力低下、寿命短縮など)患者;放射線障害患者;等を挙げることができる。本件生成促進剤をこれら対象に投与すると、これら対象の細胞におけるNAD+の生成が促進した結果、寿命を延長したり、上記疾患や障害を予防又は治療することができる。
本明細書において、「ミトコンドリア機能を賦活する必要がある動物(対象)」としては、例えば、ミトコンドリア機能の減弱又は不全に関連する若しくは起因する疾患に罹患した患者を挙げることができ、具体的には、糖尿病、ミトコンドリア病、脳疾患などに罹患した患者を挙げることができる。
本件剤の投与形態としては、粉末、顆粒、錠剤、カプセル剤、シロップ剤、懸濁液などの剤型で投与する経口投与や、溶液、乳剤、懸濁液などの剤型を注射(例えば、皮下注射、静脈内注射、筋肉内注射)、又はスプレー剤の型で鼻孔内投与する非経口投与を挙げることができる。
本件剤におけるMA5のR体又はその生理的に許容される塩の投与量は、年齢、体重、性別、症状、薬剤への感受性等に応じて適宜決定され、例えば、1μg~200mg/kg(体重)/日の投与量の範囲である。なお、本件剤は、一日あたり単回又は複数回(例えば、2~4回)に分けて投与することができる。本件剤には、MA5のS体又はその生理的に許容される塩をさらに含むものであってもよいが、その効果を効果的に発揮する観点から、含まないものが好ましい。
本件促進方法における動物細胞の培養温度は、通常30~40℃の範囲内であり、好ましくは約37℃(36~38℃)である。また、培養時のCO2濃度は、通常約1~10%の範囲内であり、好ましくは約5%(4~6%)である。また、培養時のO2濃度は、通常約10~40%の範囲内であり、好ましくは約21%(20~22%)である。
本件促進方法において、動物細胞を培養するときに使用する培養液としては、例えば、血清含有又は無血清培養液、生理食塩水、リン酸緩衝化生理食塩水、トリス緩衝化生理食塩水、HEPES緩衝化生理食塩水、リンゲル液(乳酸リンゲル液、酢酸リンゲル液、重炭酸リンゲル液等)、5%グルコース水溶液等の生理的水溶液を挙げることができ、ここで血清含有培養液としては、0.1~30(v/v)%の血清(ウシ胎児血清[Fetal bovine serum;FBS]、子牛血清[Calf bovine serum;CS]等)を含有する哺乳動物細胞培養用培養液(DMEM、EMEM、IMDM、RPMI-1640、αMEM、F-12、F-10、M-199、AIM-V等)を挙げることができ、また、上記無血清培養液としては、市販のB27サプリメント(-インスリン)(Life Technologies社製)、N2サプリメント(Life Technologies社製)、B27サプリメント(Life Technologies社製)、Knockout Serum Replacement(Invitrogen社製)等の血清代替物を適量(例えば、1~30%)添加した上記哺乳動物細胞培養用培養液などを挙げることができる。
以下、実施例により本発明をより具体的に説明するが、本発明の技術的範囲はこれらの例示に限定されるものではない。なお、INS-1E細胞株は、11.1mM グルコース、10% FBS、1mM ピルビン酸ナトリウム、10mM HEPES、2mM グルタミン、50μM βメルカプトエタノール、100U/mL ペニシリン、及び100μg/mL ストレプトマイシを含むRPMI-1640培養液(GIBCO社製)(以下、単に「RPMI-1640培養液」という)の存在下、インキュベーター(37℃、21%O2、5%CO2)内で培養した。また、リー脳症患者由来線維芽細胞株(MT-ND3変異細胞株[Leigh synd., m.10191 T>C〔p.Ser45Pro〕])は、1% FBS、100U/mLペニシリン、及び100μg/mLストレプトマイシンを含むDMEM低グルコース(GIBCO社製)培養液(以下、「DMEM低グルコース培養液」という)の存在下、上記インキュベーター内で培養した。また、HBlEpC細胞株は、10% FBS、100U/mL ペニシリン、100μg/mL ストレプトマイシ、及びHBlEpC Growth Supplement(Cell Application社製)を含むHuman Bladder Epithelial Cell Basal medium(Cell Application社製)培養液(以下、「HBlEpC培養液」という)の存在下、上記インキュベーター内で培養した。
1.MA5のR体の生成

合成したMA5のラセミ体からMA5のR体の分取は、分取装置LC-Forte(ワイエムシィ社製)を用いて行った。また、分取カラ厶は、CHIRALPAK AD-H(カラム内径2cm、カラム長さ25cm、ダイセル社製)を用いた。A液(0.05%TFA/ヘキサン)、B液(0.05%TFA/2-プロパノール)、A液/B液=60/40の単一組成の移動相によって、流速10mL/分で40分かけて溶出させた。なお、移動相の流速は20mL/分で行うこともでき、溶出時間は半分の20分にすることも可能である。MA5の検出波長は220nmに設定した。MA5のR体及びS体(表1参照)の保持時間は、それぞれ17.3分及び29.8分(流速10mL/分)であった。カラム分離により、MA5のR体を光学純度100%で得た。

2.動物細胞内のNAD+量の評価
動物細胞内のNAD+量に及ぼすMA5のR体の影響を調べるために、以下の「2-1」の項目に記載の方法に従って解析した。

Pharmacokinet. 25 (3): 274-282 (2010).」参照)を播種し、RPMI-1640培養液の存在下で48時間培養後、培養液を除去し、化合物の溶媒である0.1%のDMSOを含むRPMI-1640培養液、又は各種濃度(3pM、10pM、30pM、100pM、300pM、1nM、若しくは3nM)の3種類の化合物(MA5のラセミ体、MA5のR体、若しくはMA5のS体)を含むRPMI-1640培養液を、それぞれ100μLずつ添加し、6時間培養した。その後、NAD/NADH-Glo Assay(G9071、Promega社製)を用いて、細胞内のNAD+量を測定した。

2-2 結果
INS-1E細胞株を、MA5のラセミ体の存在下で培養しても、MA5のラセミ体非存在下で培養した場合と比べ、細胞内のNAD+量はほとんど変わらなかった(図1A参照)。一方、INS-1E細胞株を、MA5のS体の存在下で培養すると、MA5のS体非存在下で培養した場合と比べ、細胞内のNAD+量が培養時間依存的に減少した(図1C参照)。また、INS-1E細胞株を、MA5のR体の存在下で培養すると、MA5のR体非存在下で培養した場合と比べ、細胞内のNAD+量が培養時間依存的に増加した(図1B参照)。
INS-1E細胞株を、MA5のラセミ体の存在下で培養しても、MA5のラセミ体非存在下で培養した場合と比べ、細胞内のNAD+量はほとんど変わらなかった(図1A参照)。一方、INS-1E細胞株を、MA5のS体の存在下で培養すると、MA5のS体非存在下で培養した場合と比べ、細胞内のNAD+量が培養時間依存的に減少した(図1C参照)。また、INS-1E細胞株を、MA5のR体の存在下で培養すると、MA5のR体非存在下で培養した場合と比べ、細胞内のNAD+量が培養時間依存的に増加した(図1B参照)。
これらの結果は、MA5のS体は、動物細胞内のNAD+量を減少させる作用を有するのに対して、MA5のR体は、逆に、動物細胞内のNAD+量を増加させる作用(すなわち、動物細胞におけるNAD+の生成を促進する作用)を有することを示している。
3.生体内への投与試験1
MA5のR体を生体内に投与したとき、キラル反転(コンバージョン)が起こるかどうかと、血漿中のMA5のR体の濃度の推移について、以下の「3-1」の項目に記載の方法に従って解析した。

MA5の各エナンチオマー(R体又はS体)を、生理食塩水と0.1Nの水酸化ナトリウムの混合液(9:1)に溶解させ、0.22μmのフィルターを用いてろ過滅菌を行い、2mL/kgのMA5の各エナンチオマーを含む液を調製した。次いで、4~5歳齢の雄性カニクイザル3匹に、それぞれ、MA5の各エナンチオマーを含む液を、1mg/kgの用量で単回静脈内投与した。なお、コントロールとしてMA5の各エナンチオマーを含まない液も同様に投与した。投与後5分、15分、30分、1時間、2時間、4時間、6時間、及び24時間に採決を行い、血漿を採取し、血漿における、MA5の各エナンチオマーの濃度を、LC―MS/MS(液体クロマトグラフィー/タンデム質量分析法 (liquid chromatography/tandem mass spectrometry)により測定し、血中濃度-時間曲線下面積(AUC)(μg・hr/mL)を算出した。LC―MS/MSの分析条件は、以下のとおりである。

イオン化部として、ESIプローブを装着した三連四重極型質量分析(TSQ Quantum Ultra、Thermo Fisher Scientific社製)に高速液体クロマトグラフとしてNANOSPACE SI-2(Osaka soda社製)を接続した装置を使用した。分析対象物質であるMA5のR体((R)-MA5)及びMA5のS体((S)-MA5)に対し、それぞれの安定同位体である(R)-MA5-d6及び(S)-MA5-d6用いた内部標準法で定量解析を行った。分析カラムにはCHIRALPAK AD-3R(150 x 2.1 mm i.d., 3.0 μm particle size、ダイセル社製)を、ガードカラムにはCAPCELL C8 UG120(10 x 2.0 mm i.d., 5.0 μm particlesize、Osaka soda社製)を使用した。カラムオーブン温度は35℃とした。移動相は、ギ酸/水(0.1/100[v/v])(A液)/アセトニトリル(B液)を用い、流速は200μL/分とした。A液/B液(57/43[v/v])の単一組成により溶出させるアイソクラティック条件で測定を行った。分析時間9.5分までA液/B液(57/43[v/v])とし、分析時間9.7分でA液/B液(10/90[v/v])とした。また、分析時間9.7分~11.7分は、A液/B液(10/90[v/v])にてカラムの洗浄を行う系を組み込み、11.8分~15分は、A液/B液(57/43[v/v])でカラムの平衡化を行った。保持時間は、(R)-MA5が6.95分、(R)-MA5-d6が6.80分、(S)-MA5が8.69分、(S)-MA5-d6が8.56分であった。
MS条件は、ネガティブイオンモードで検出し、spray voltage、vaporizer temperature、及びion transfer tube temperatureを、それぞれ2.5kV、250℃、及び250℃に設定した。プリカーサーイオン>プロダクトイオンのSRMチャネルは、MA5については「m/z 328.0>116.2」に設定し、MA5-d6については、「m/z 334.0>121.1」に設定した。プロダクトイオンを生成させるためのcollision energyは、MA5については12eVに設定し、MA5-d6については17eVに設定した。波形解析及び内部標準法による定量計算は、Xcalibur(Thermo Fisher Scientific社製)により行った。
3-2 結果
MA5のR体を投与直後(投与後5分)から、血液中のMA5のR体が消失する投与後24時間までの間、MA5のS体は検出されず、MA5のR体のみが検出された。この結果は、MA5のR体は、生体内においてキラルコンバージョンを起こさないことを示している。

また、MA5のS体を投与したときのAUC値は、12(μg・hr/mL)であったのに対して、MA5のR体を投与したときのAUC値は、30(μg・hr/mL)であった(表2参照)。この結果は、MA5のR体が、MA5のS体よりも2.5倍以上血中で保持されやすいことを示しており、MA5のR体の投与量を、MA5のS体よりも少なくできることを示唆している。
4.ヒト肝ミクロソームを用いた代謝試験
生体内において、MA5のR体が、MA5のS体よりも血中で保持されやすかったことから、MA5の各エナンチオマーについて、代謝率に違いがあるかどうかを、以下の「4-1」の項目に記載の方法に従って解析した。

MA5のR体又はS体を、アセトニトリル(LC-MS用、関東化学社製)に溶解し、それぞれ10mMに調整した。スクロース(富士フイルム和光純薬社製)855mgを、水10mLに溶解し、250mMのスクロース溶液を調整した後、かかる溶液を用いて、20mg/mLのヒト肝ミクロソーム(8-donor-pool、Sekisui XenoTech社製)含有液を4倍希釈し、5mg/mLのヒト肝ミクロソーム懸濁液を調整した。1mLチューブにて、ヒト肝ミクロソーム懸濁液50μLに、5μLの3種類の被験液(10mMのMA5のR体を含むスクロース溶液、10mMのMA5のS体を含むスクロース溶液、又はコントロールであるVehicle[スクロース溶液])と、5μLのNADPH Regenerating System Solution B(Corning社製)、100μLのUGT Reaction Mix Solution B(Corning社製)、及び275μLの水とを加え、30秒震盪し、37℃で5分間プレインキュベートした。その後、25μLのNADPH Regenerating System Solution A(Corning社製)及び40μLのUGT Reaction Mix Solution A(Corning社製)を加え、30秒震盪し、37℃で1時間インキュベートした。0.1%のギ酸を含む氷冷アセトニトリル溶液500μLを加えて反応を停止し、30秒震盪、10分超音波処理した後、20,000×g、4℃で10分間遠心処理した。新しい1mLチューブに上清50μLを回収し、2,000rpm、室温(25~35℃)で30分間遠心濃縮し、-80℃で凍結した。これを凍結乾燥機で30分間乾燥し、0.1%のギ酸を含む20%メタノール(LC-MS用、関東化学社製)25μLにて再溶解の後、そのうち2μLをLC/HRMSに付した。なお、サンプル数はn=3で実施し、可能な限り氷上で操作を行った。

LC/HRMS測定
LC装置としてNanospace si-2(Osaka soda社製)を用いた。カラムとしてDevelosil C30-ug-3(2.1 x 150 mm, 3.0 μm particle size、Nomura Chemical社製)を用い、移動相A液に10mMのNH5CO3/H2O、移動相B液にメタノールを用い、流速200μL/分でリニアグラジエント溶出をおこなった(15-65%B液(0.0-22.0分)、65-90%B液(22.0-24.4分)、100%B液(24.5-29.0分)、15%B液(30.0-40.0分))。カラムオーブンは40℃、サンプル注入量は2μLに設定し、Wash液に10%メタノール、Wash port液に50%アセトニトリルを選択した。

HRMS装置としてQ-Exactive(Thermo Fisher Scientific社製)を用いた。ESI条件として、スプレー電圧;3000V、シースガス圧;40psi、オグジュアリーガス圧;10psi、キャピラリー温度・ヴェイポライザー温度;300°Cに設定した。検出のため、正負両イオンのFull scan-ddMS2(topN)モードを選択した。各種パラメータは以下のとおりである:Full scan; Resolution 70,000, AGC target 3e6, Maximum IT 100 ms, Scan range 80-1,200, ddMS2; Resolution 17,500, AGC target 5e4, Maximum IT50 ms, topN N=20, Isolation window 4.0 m/z, NCE 20(50% stepped NCE)。追加のMS2データは、正負両イオンのtarget MS2モードで測定した。各種パラメータは以下のとおりである:Resolution 35,000, AGC target 6e5, Maximum IT 200 ms, Isolation window 4.0 m/z, NCE 20(50% stepped NCE)。
データ解析
データの解析は、Compound Discoverer(Thermo Fisher Scientific社製)及びXcalibur(Thermo Fisher Scientific社製)を用いて行った。ピーク抽出には、Metabolism w/ FlSh ワークフローを選択し、「Compound」にMA5の構造情報を登録した。下限ピーク強度は各サンプル中のMA5平均ピーク強度の1万分の1に設定した。代謝物予測ノードのイオンアダクトに、[M+Na]+、[M+NH4]+、[M-H2O+H]+、[M+Cl]-を追加した。また代謝経路に、トリプトファン>キヌレニン経路[-C+O]、AMP抱合[+C10H12N5O6P]、CoA抱合[+C21H34N7O15P3S]を追加した。

4-2 結果
ヒト肝ミクロソームを、MA5のラセミ体又はMA5のS体の存在下でインキュベートした場合、グルクロン酸抱合体(+C6H8O6[+1.7min、+2.6min])が検出されたのに対して、ヒト肝ミクロソームを、MA5のR体の存在下でインキュベートした場合、かかるグルクロン酸抱合体はほとんど又はコントロールと変わらないレベルでしか検出されなかった(図2参照)。
この結果は、MA5のS体の方がMA5のR体よりも、10倍以上グルクロン酸抱合されやすいことを示しており、上記「4.ヒト肝ミクロソームを用いた代謝試験」の項目の結果、すなわち、生体内において、MA5のR体が、MA5のS体よりも血中で保持されやすかったという結果を支持している。
ヒト肝ミクロソームを、MA5のラセミ体又はMA5のS体の存在下でインキュベートした場合、グルクロン酸抱合体(+C6H8O6[+1.7min、+2.6min])が検出されたのに対して、ヒト肝ミクロソームを、MA5のR体の存在下でインキュベートした場合、かかるグルクロン酸抱合体はほとんど又はコントロールと変わらないレベルでしか検出されなかった(図2参照)。
この結果は、MA5のS体の方がMA5のR体よりも、10倍以上グルクロン酸抱合されやすいことを示しており、上記「4.ヒト肝ミクロソームを用いた代謝試験」の項目の結果、すなわち、生体内において、MA5のR体が、MA5のS体よりも血中で保持されやすかったという結果を支持している。
さらに、GLP準拠非臨床試験からラット4週間反復投与毒性試験及びサル4週間反復投与毒性試験を行った結果、高用量投与群で尿路上皮に対する毒性所見が認められた。その原因を明らかにするため、尿中代謝物について評価したところ、サルでは尿中MA5のラセミ体のグルクロン酸抱合体、ラットではMA5のラセミ体の硫酸抱合体の尿中濃度が毒性発現と相関し、毒性の本体であると推定された。
肝臓の代謝酵素や腎臓トランスポーターはサルとラットの間で種差があり、ヒトの肝代謝はサルに近いことが明らかになったことから、ヒトにおける副作用の本体もグルクロン酸抱合体であると推定される。MA5のR体は、上記ヒト肝ミクロソームを用いた代謝試験から、S体に比べてグルクロン酸抱合体がほとんど(1/10以下)産生されないことから、MA5のR体は、MA5のS体に比べてより副作用がない安全な化合物と言える。
5.細胞毒性評価1
動物細胞に対するMA5のR体の毒性を評価するために、以下の「5-1」の項目に記載の方法に従って解析した。

HBlEpC細胞株を、MA5のラセミ体あるいはエナンチオマー(R体又はS体)の存在下で培養すると、MA5のラセミ体の存在下で培養した場合と比べ、細胞の生存率が高く、特に、MA5のR体の方が、MA5のS体よりも細胞の生存率が高かった(図3参照)。この結果は、MA5のR体は、MA5のラセミ体やMA5のS体よりも動物細胞に対する細胞毒性が低いことを示している。
HBlEpC細胞株を、MA5のラセミ体あるいはエナンチオマー(R体又はS体)の存在下で培養すると、MA5のラセミ体の存在下で培養した場合と比べ、細胞の生存率が高く、特に、MA5のR体の方が、MA5のS体よりも細胞の生存率が高かった(図3参照)。この結果は、MA5のR体は、MA5のラセミ体やMA5のS体よりも動物細胞に対する細胞毒性が低いことを示している。
6.細胞毒性評価2

96ウェルプレートに、1ウェルあたり2×103個のMT-ND3変異細胞株を播種し、DMEM低グルコース培養液の存在下で2日間培養後、化合物の溶媒である0.1%のDMSO、又は各種濃度(0.003μM、0.01μM、0.03μM、0.1μM、0.3μM、1μM、3μM、10μM、30μM、又は100μM)のMA5の各エナンチオマー(R体又はS体)を添加し、さらに5日間培養後、細胞生存レベルを測定した。具体的には、細胞生存レベルは、Cell Counting Kit-8(同仁化学研究所社製)を用いたMTTアッセイにより測定した。すなわち、各ウェルにCell Count Reagent SFを100μLずつ添加し、2時間インキュベートし、マイクロプレートリーダーで3秒間撹拌した後、吸光度450nm(リファレンス750nm)を測定した(図4参照)。なお、コントロールとして何も添加しなかった場合の実験も行った(図4中の「-」)。

6-2 結果
MT-ND3変異細胞株を、高濃度(30μMや100μM)のMA5のR体の存在下で培養しても、生細胞レベルの低下は認められなかった(図4A参照)。一方、MT-ND3変異細胞株を、高濃度(30μMや100μM)のMA5のS体の存在下で培養すると、生細胞レベルの有意な低下が認められた(図4B参照)。この結果は、MA5のR体は、MA5のS体よりも動物細胞に対する細胞毒性が低いことを示しており、上記「5.細胞毒性評価1」の項目の結果を支持している。
MT-ND3変異細胞株を、高濃度(30μMや100μM)のMA5のR体の存在下で培養しても、生細胞レベルの低下は認められなかった(図4A参照)。一方、MT-ND3変異細胞株を、高濃度(30μMや100μM)のMA5のS体の存在下で培養すると、生細胞レベルの有意な低下が認められた(図4B参照)。この結果は、MA5のR体は、MA5のS体よりも動物細胞に対する細胞毒性が低いことを示しており、上記「5.細胞毒性評価1」の項目の結果を支持している。
7.動物細胞におけるATP産生能の評価
MA5のラセミ体は、動物細胞におけるATP産生能を増強する作用を有することを報告している(例えば、国際公開第2014/080640号パンフレット参照)。そこで、MA5のR体及びMA5のS体の間で、ATP産生能の増強作用に違いがあるかどうかを確認するために、以下の「7-1」の項目に記載の方法に従って解析した。
MA5のラセミ体は、動物細胞におけるATP産生能を増強する作用を有することを報告している(例えば、国際公開第2014/080640号パンフレット参照)。そこで、MA5のR体及びMA5のS体の間で、ATP産生能の増強作用に違いがあるかどうかを確認するために、以下の「7-1」の項目に記載の方法に従って解析した。

96ウェルプレートに、1ウェルあたり2×103個のMT-ND3変異細胞株を播種し、DMEM低グルコース培養液の存在下で24時間培養後、化合物の溶媒である0.1%のDMSO、又は各種濃度(0.1μM又は1μM)の3種類の化合物(MA5のラセミ体、MA5のR体、若しくはMA5のS体)を添加し、6時間培養後、培養液中に産生されたATPの濃度を、『細胞の』ATP測定試薬(東洋ビーネット社製)を用いてGloMa 96 Microplate Luminometer(Promega社製)により測定した。
96ウェルプレートに、1ウェルあたり2×103個のMT-ND3変異細胞株を播種し、DMEM低グルコース培養液の存在下で24時間培養後、化合物の溶媒である0.1%のDMSO、又は各種濃度(0.1μM又は1μM)の3種類の化合物(MA5のラセミ体、MA5のR体、若しくはMA5のS体)を添加し、6時間培養後、培養液中に産生されたATPの濃度を、『細胞の』ATP測定試薬(東洋ビーネット社製)を用いてGloMa 96 Microplate Luminometer(Promega社製)により測定した。

MA5のR体及びMA5のS体の間で、動物細胞におけるATP産生レベルに違いは認められなかった(図5参照)。

8.寿命の評価
MA5のR体が、生物の寿命を延長する効果を有することを確認するために、例えば、以下の[方法1]や[方法2]の項目に記載の方法に従って解析する。

〔3〕5%のスクロース溶液、又は各種濃度(100nM若しくは1000nM)の3種類の化合物(MA5のラセミ体、MA5のR体、若しくはMA5のS体)を含む5%のスクロース溶液を入れたバイアルを用意し、それぞれに20匹のオス(♂)ハエ及びメス(♀)ハエを移し、それぞれの溶液を摂取させる(遮光して29℃で20時間)。
〔4〕通常のエサを入れた別のバイアルにハエを移し、通常のエサを摂取させる。(1日間)
〔5〕翌日以降、上記手順〔3〕~〔4〕を繰り返す。
〔6〕上記手順〔3〕~〔4〕の際に、死んだハエを取り除き、その数を記録するとともに、死んだハエの寿命を記録する。バイアルが空になったら(すなわち、バイアルの中に生きたハエがいなくなったら)、記録した死ハエの数の総計を元の数として生存数を計算し、死んだ個々ハエの寿命と、生存したハエの総数を基に、ハエの平均寿命を算出する。なお、18℃から29℃に移す理由は、温度感受性の遺伝子発現システムを使っているためであり、発生過程では神経変性タンパク質を発現せず、成虫に羽化してから神経変性タンパク質を発現させているからである(発生過程で全ての神経細胞に神経変性タンパク質を発現させると致死のケースが多い)。
ハエは、ヒト疾患のモデル生物として利用されたり(文献「Hum Mol Genet. 2019. PMID: 31227826」参照)、ヒト老化のモデル生物として利用されている(文献「FEBS Open Bio. 2022. PMID: 34854258」参照)。このため、ハエにおいて、MA5のR体による寿命延長効果が認められれば、その効果はヒトにも適用できると言える。

〔2〕生後30日目に、水で懸濁した3種類の化合物(MA5のラセミ体、MA5のR体、又はMA5のS体)を、10mg/kg体重/100μL/日の用量で上記KOマウスに経口投与する。
〔3〕1週間に一度、KOマウスの体重を測定し、死亡が確認されるまで記録する。なお、コントロールとして、上記3種類の化合物を経口投与しなかったKOマウスについても同様に、死亡が確認されるまで体重を測定する。

9.ミトコンドリア機能の評価
ミトコンドリア内膜タンパク質(ミトフィリン[Mitofilin])の線虫オーソログであるimmt-1欠損変異型線虫(以下、「immt-1/mitofilin変異体」という)では、筋細胞におけるミトコンドリアがswellingすることが報告されている(文献「Journal of Cellular Physiology. 224 (2010) 748-756」、文献「Molecular Biology of the Cell. 22 (2011) 831-841.」参照)。そこで、immt-1/mitofilin変異体に対して、3種類の化合物(MA5のラセミ体、MA5のR体、又はMA5のS体)を投与した場合、ミトコンドリアのswellingが緩和されるかどうかを解析した。
ミトコンドリア内膜タンパク質(ミトフィリン[Mitofilin])の線虫オーソログであるimmt-1欠損変異型線虫(以下、「immt-1/mitofilin変異体」という)では、筋細胞におけるミトコンドリアがswellingすることが報告されている(文献「Journal of Cellular Physiology. 224 (2010) 748-756」、文献「Molecular Biology of the Cell. 22 (2011) 831-841.」参照)。そこで、immt-1/mitofilin変異体に対して、3種類の化合物(MA5のラセミ体、MA5のR体、又はMA5のS体)を投与した場合、ミトコンドリアのswellingが緩和されるかどうかを解析した。
具体的には、ミトコンドリアDNA及びゲノムDNAをGFPで可視化できるように遺伝子操作したccIs4251(Pmyo-3::GFP::LacZ::NLS, Pmyo-3::mitochondrial-GFP+dpy-20(+)、文献「Cell 139 (2009) 623-633」)のimmt-1/mitofilin変異体(NBRP[National BioResource Project] 線虫[東京女子医科大学])を、コントロールであるDMSO(溶媒)を散布した培養プレート(E. coli OP-50 NGM寒天培地)、又は上記3種類の化合物(MA5のラセミ体、MA5のR体、又はMA5のS体)を、終濃度が10μMとなるように散布した上記培養プレート上で、L4幼虫期から5日間(すなわち、4日齢成虫まで)飼育し、4日齢成虫において体壁筋細胞を観察し(図6参照)、異常なミトコンドリア(swellingしたミトコンドリア)の数と、異常なミトコンドリアが認められた体壁筋細胞の割合を測定した(図7及び8参照)。なお、比較対照として、ccIs4251の野生型線虫(CGC[Caenorhabditis Genetics Center]より入手)を培養プレート上で飼育後、同様に解析した。
その結果、immt-1/mitofilin変異体を、MA5のR体の存在下で飼育すると、コントロールであるDMSOの存在下で飼育した場合や、MA5のS体の存在下で飼育した場合と比べ、体壁筋細胞当たり5個以上のswellingミトコンドリアが認められた体壁筋細胞の割合が大幅に減少するとともに(図7参照)、体壁筋細胞当たりのswellingミトコンドリアの数も大幅に減少した(図8参照)。また、immt-1/mitofilin変異体を、MA5のS体の存在下で飼育すると、コントロールであるDMSOの存在下で飼育した場合と比べ、体壁筋細胞当たり5個以上のswellingミトコンドリアが認められた体壁筋細胞の割合や、体壁筋細胞当たりのswellingミトコンドリアの数は、ほとんど変わらないかわずかの減少にとどまった(図7及び8参照)。
この結果は、MA5のS体には、ミトコンドリアのswellingを緩和する作用はほとんどなく、MA5のR体がミトコンドリアのswellingを緩和する作用を有することを示している。すなわち、MA5のR体が、線虫におけるミトコンドリア機能を改善する作用を有することを示している。また、線虫は、ヒトのミトコンドリア機能を解析するモデル生物として利用されていることから(文献「Nature. 2018. PMID: 30356218」参照)、MA5のR体は、ヒトに対するミトコンドリア機能賦活剤やミトコンドリア病の治療剤又は予防剤に有用であることが示されたと言える。
10.生体内への投与試験2
上記「3.生体内への投与試験1」において、静脈内投与したMA5のR体が、静脈内投与したMA5のS体よりも血中で保持されやすいことが確認されたが、経口投与した場合も同様の結果となるかどうかを確認した。具体的には、上記「3-1 方法」に記載の方法に従って、MA5の各エナンチオマー(R体又はS体)を含む液を、100mg/kgの用量で雄性カニクイザルに単回経口投与し、血漿における、MA5の各エナンチオマーの濃度と、MA5の各エナンチオマーの還元代謝物(式(B)で表される化合物[すなわち、Red-MA5])の濃度を測定した。
上記「3.生体内への投与試験1」において、静脈内投与したMA5のR体が、静脈内投与したMA5のS体よりも血中で保持されやすいことが確認されたが、経口投与した場合も同様の結果となるかどうかを確認した。具体的には、上記「3-1 方法」に記載の方法に従って、MA5の各エナンチオマー(R体又はS体)を含む液を、100mg/kgの用量で雄性カニクイザルに単回経口投与し、血漿における、MA5の各エナンチオマーの濃度と、MA5の各エナンチオマーの還元代謝物(式(B)で表される化合物[すなわち、Red-MA5])の濃度を測定した。
その結果、経口投与後、血漿におけるMA5のR体の濃度は、血漿におけるMA5のS体の濃度よりも高かった(図9参照)。また、血漿における還元代謝物(式(B)で表される化合物、すなわち、Red-MA5)は、MA5のS体を経口投与した場合、検出されたのに対して、MA5のR体を経口投与した場合は、ほとんど検出されなかった(図10参照)。
これらの結果は、MA5のR体を経口投与した場合も、MA5のS体を経口投与した場合よりも血中で保持されやすく、また、MA5のR体の方がMA5のS体よりも代謝されにくいことから、MA5のR体は、MA5のS体よりも、その効果を生体内で持続的に発揮できることを示している。
11.細胞毒性評価3
動物細胞に対するRed-MA5の毒性を評価するために、以下の「11-1」の項目に記載の方法に従って解析した。

11-1 方法
96ウェルプレートに、1ウェルあたり3×103個のKCMC10細胞株を播種し、化合物の溶媒である0.1%のDMSO、又は各種濃度(3nM、10nM、30nM、100nM、300nM、1μM、3μM、10μM、又は30μM)の2種類の化合物(MA5のラセミ体又はRed-MA5)の存在下で24時間培養後、細胞生存レベルを測定した。具体的には、細胞生存レベルは、Cell Counting Kit-8(同仁化学研究所社製)を用いたMTTアッセイにより測定した。すなわち、各ウェルにCell Count Reagent SFを100μLずつ添加し、2時間インキュベートし、マイクロプレートリーダーで3秒間撹拌した後、吸光度450nm(リファレンス750nm)を測定した(図11参照)。なお、コントロールとして何も添加しなかった場合の実験も行った(図11中の「-」)。

KCMC10細胞株を、30μMのMA5のラセミ体の存在下で培養しても、生細胞レベルの低下は認められなかったのに対して(図11A参照)、30μMのRed-MA5の存在下で培養すると、生細胞レベルの有意な低下が認められた(図11B参照)。この結果は。Red-MA5よりもMA5の方が、細胞毒性が低いことを示している。すなわち、上記「10.生体内への投与試験2」の結果と総合すると、生体内に投与したMA5のR体の方が、生体内に投与したMA5のS体よりも細胞毒性が低いことを示している。
KCMC10細胞株を、30μMのMA5のラセミ体の存在下で培養しても、生細胞レベルの低下は認められなかったのに対して(図11A参照)、30μMのRed-MA5の存在下で培養すると、生細胞レベルの有意な低下が認められた(図11B参照)。この結果は。Red-MA5よりもMA5の方が、細胞毒性が低いことを示している。すなわち、上記「10.生体内への投与試験2」の結果と総合すると、生体内に投与したMA5のR体の方が、生体内に投与したMA5のS体よりも細胞毒性が低いことを示している。
12.動物細胞内のSIRT発現量の評価
細胞内において、NAD+量とSIRT発現量との間には、正の相関関係があることが知られている(文献「Nat Rev Nephrol. 2017. PMID: 28163307」参照)。また、内耳細胞におけるSIRTの発現増加と、難聴の改善との関連性が報告されている(文献「Front Cell Dev Biol. 2021. PMID: 34869361」、文献「Exp Cell Res. 2022 Jul 11:113280.」、文献「Hindawi Neural Plasticity Volume 2021, Article ID 5520794」参照)。そこで、MA5のR体が、細胞内のNAD+量を増加させることによって、内耳細胞内のSIRTの発現量が増加し、難聴の改善に有用であるかどうかを調べるために、以下の「12-1」の項目に記載の方法に従って解析した。

12-1 方法
10mLの培養液(10重量%のFBSを添加したグルコース低含有DMEM培地[グルコース濃度1.0g/dL])を含むディッシュに、1×106cell/dishのマウス内耳細胞株(HEI-OC1細胞株)を播種して培養した。1日後に、化合物の溶媒である0.1%のDMSO、又は各種濃度(1μM、3μM、10μM、又は30μM)の3種類の化合物(MA5のラセミ体、MA5のR体、又はMA5のS体)を添加し、24時間培養した。培養終了後、細胞を回収し、常法によって細胞を破壊して細胞内の各種SIRT(SIRT1、SIRT2、SIRT3、SIRT5、SIRT6、及びSIRT7)の発現レベルをウエスタンブロット法により解析した(図12~14[図12-1~図14-2]参照)。

12-2 結果
HEI-OC1細胞株を、MA5のS体の存在下で培養しても、MA5のS体の非存在下で培養した場合と比べ、細胞内の各種SIRT(SIRT1、SIRT2、SIRT3、SIRT5、SIRT6、及びSIRT7)の発現量は、ほとんど変わらなかったのに対して(図14[図14-1及び図14-2]参照)、HEI-OC1細胞株を、MA5のR体の存在下で培養すると、MA5のR体の非存在下で培養した場合と比べ、細胞内の各種SIRTの発現量が増加した(図13[図13-1及び図13-2]参照)。
この結果は、上記「2.動物細胞内のNAD+量の評価」の結果と総合すると、MA5のR体が、細胞内のNAD+量を増加させた結果、細胞内のSIRTの発現量が増加し、難聴の改善に有用であることを示している。

この結果は、上記「2.動物細胞内のNAD+量の評価」の結果と総合すると、MA5のR体が、細胞内のNAD+量を増加させた結果、細胞内のSIRTの発現量が増加し、難聴の改善に有用であることを示している。
13.動物の運動機能の評価
MA5のR体が、動物の運動機能障害を改善する効果を有することを確認するために、例えば、以下の[老化マウス]や[幼若マウス]の項目に記載のマウスを用い、以下の[方法1]~[方法3]の項目に記載の方法に従って解析する。

[老化マウス]
3種類の化合物(MA5のラセミ体、MA5のR体、若しくはMA5のS体)を含む水を、それぞれ50mg/kg体重/日の用量で老化マウス(92週)(5匹)に9週間経口投与する。また、コントロールとして、水を老化マウス(6匹)に9週間経口投与する。

[幼若マウス]
3種類の化合物(MA5のラセミ体、MA5のR体、若しくはMA5のS体)を含む水を、それぞれ10mg/kg体重/日(低用量)又は50mg/kg体重(高用量)の用量で幼若マウス(4か月)(6匹)に9週間経口投与する。また、コントロールとして、水を幼若マウス(6匹)に9週間経口投与する。
3種類の化合物(MA5のラセミ体、MA5のR体、若しくはMA5のS体)を含む水を、それぞれ10mg/kg体重/日(低用量)又は50mg/kg体重(高用量)の用量で幼若マウス(4か月)(6匹)に9週間経口投与する。また、コントロールとして、水を幼若マウス(6匹)に9週間経口投与する。
[方法1:トレッドミル検査]
〔1〕マウス用のトレッドミル装置(MK-690、室町機械社製、以下同じ)を用いて、トレッドミル検査の練習を行う。老化マウスの場合、1日目;8m/分の速度で10分間×2回、2日目;10m/分の速度で10分間×2回、及び3日目;12m/分の速度で10分間×2回の計3日間行う。一方、幼若マウスの場合、1日目;8m/分の速度で10分間×2回、2日目;9m/分の速度で10分間×2回、3日目;10m/分の速度で10分間×2回、4日目;11m/分の速度で10分間×2回、及び5日目;12m/分の速度で10分間×2回の計5日間行う。
〔2〕マウス用のトレッドミル装置を用いて、トレッドミル検査の本番を、以下の条件で最大2時間行う。5m/分の速度で開始し、3分毎に5m/分ずつ速度を上げて、最終的には28m/分の速度まで上げる(すなわち、5m/分→10m/分→15m/分→20m/分→25m/分→28m/分)。
〔3〕刷毛で尾部を刺激しても前進せず、通電された棒の上で10秒以上移動しなかった場合に力尽きたと判定する。2時間以上走行した場合は、打ち切りとし2時間と記録する。

マウスの逆格子吊り下げ試験は、直径37cmで8mm四方の金網を使用して行う。金網の周囲には、マウス脱出を防ぐ目的で12cmのプラスチック製の壁を作製する。マウス順応のために、1分間金網の上に置く。金網の中央にマウスがいる状態で金網をひっくり返し、マウスが落下するまでの時間(分)を測定する。地面から60cmの高さで金網を保持する。落下した時のマウスの怪我の防止のため、下には飼育用の床敷を敷く。30秒未満で落下した場合は、事故による落下と判断し30秒以上になるまで追加で測定する。別日に2回測定し、その平均値を使用する。また、マウス体重を測定し、「Hanging Impulse Score」(捕まった時間[分]×体重[分])を算出する。

[方法3:握力]
マウスの握力測定は、マウス用の握力測定装置(GPM-101、Melquest社製)を使用して行う。前脚の握力測定には、前脚用のグリップを使用し、四肢の握力測定には、四肢用の金網をセットして使用する。前脚の握力測定から行い、横方向と縦方向で測定する。横方向の測定のために、マウスの両前脚がグリップを把持した状態で水平方向に引っ張り、手が離れるまで引っ張る。縦方向の測定のために、マウスの両前脚がグリップを把持した状態で垂直方向に引っ張り、手が離れるまで引っ張る。5回連続で行った最大値を計測する。四肢用の測定は、マウスの四肢が金網にのった状態で、手が離れるまで尾を水平方向に引っ張り、5回連続で行った最大値を計測する。

本発明は、加齢に伴う症状又は疾患、代謝性疾患、ミトコンドリア病などの症状若しくは疾患の予防又は治療や、寿命の延長に資するものである。
