본 출원은 2021년 5월 21일자 대한민국 특허출원 제10-2021-0065600호에 기초한 우선권의 이익을 주장하며, 해당 대한민국 특허출원의 문헌에 개시된 모든 내용은 본 명세서의 일부로서 포함된다.
본 출원은 암모니아의 지속 가능한 순환이 가능한 방향족 아미노산의 결정화 방법에 관한 것이다.
L-트립토판(tryptophan)은 필수아미노산의 하나로 대부분의 동물은 스스로 L-트립토판을 형성할 수 없기 때문에 섭취를 통해 공급되어야 한다. L-트립토판과 동일하게 방향족 아미노산으로서, 4종의 단백질원 아미노산 중에는 타이로신 (tyrosine), 페닐알라닌(phenylalanine), 히스티딘(histidine)이 해당한다. L-트립토판을 비롯한 아미노산들은 단백질 생합성 전구체로서의 역할을 가지며 L-트립토판의 경우 체내 대사를 거쳐 세로토닌 및 니아신 등 인간의 건강에 필수적인 물질로 변환된다.
방향족 아미노산은 순수한 물에서의 용해성이 다른 아미노산에 비해 낮은 편이다. 즉, 산업용 발효 공법으로 생산된 배양액(broth) 내의 L-트립토판을 포함하는 방향족 아미노산의 경우 용해도 이상의 발효 농도에서 결정의 형태로 존재한다. 이는 정제공정 중의 회수율을 감소시키는 문제를 발생할 수 있어 공정 중 결정 용해를 위한 첨가제 도입을 통한 용해도 증진으로 공정 개선이 필요한 실정이다.
L-트립토판을 포함하는 방향족 아미노산을 결정화하는 방법으로 사용되는 중화 결정화 방법 및 농축 결정화 방법은 불순물 증가에 따른 입도 하락과 불순물 이행에 따른 품질 하락 위험이 발생된다. 이에 따라, 결정화 과정 중 불순물 이행을 최소화하며 결정 입도를 향상시킬 수 있는 방법이 필요하다.
L-트립토판 정제방법으로 수지탑을 활용한 방법 및 중화 결정화 방법은 공정 중 첨가제를 사용함에 따른 비용상승이 발생되어 경제성이 떨어진다. 반면, 암모니아 처리를 통한 결정 용해 후 농축공정을 통한 결정회수 시 별도의 공정 추가 없이 고함량 제품 생산이 가능하며 농축과정에 회수된 암모니아는 결정 용해목적으로 재사용함에 따라 경제적이다.
이러한 기술적 배경 하에서, L-트립토판 정제를 위한 배양액(broth)내 L-트립토판을 용해한 후 재결정화를 통해 품질 및 수율에 유리한 공정 구축할 수 있음을 확인하고 이에 기초하여 본 출원을 완성하였다.

본 출원은 일 양상으로,
(a) 방향족 아미노산 결정을 포함하는 반응액과 암모니아를 혼합하여 상기 방향족 아미노산 결정이 용해된 용해액을 수득하는 단계 (결정 용해);
(b) 상기 수득된 용해액을 결정화하여 방향족 아미노산 결정을 포함하는 농축액을 수득하는 단계 (농축 결정화);
(c) 상기 결정화 과정에서 생성된, 암모니아를 포함하는 기체를 회수하는 단계; 및
(d) 상기 회수한 기체로부터 유래한 암모니아를 상기 (a) 단계의 암모니아로 재사용하는 단계;
를 포함하는 방향족 아미노산의 결정화 방법을 제공한다.

본 출원의 방향족 아미노산의 결정화 방법을 각각의 단계별로 상세하게 설명하면, 다음과 같다.
(a) 단계
우선, 본 출원의 방법은 (a) 단계, 방향족 아미노산 결정을 포함하는 반응액에 암모니아를 혼합(첨가)하여 방향족 아미노산 결정이 용해된 용해액을 수득하는 단계 (결정 용해)를 포함할 수 있다.
본 출원에서 용어, "방향족 아미노산"은 트립토판, 타이로신, 페닐알라닌, 및 히스티딘으로 이루어진 군에서 선택되는 1종 이상의 아미노산일 수 있고, L-트립토판, L-타이로신, L-페닐알라닌, 및 L-히스티딘으로 이루어지는 군에서 선택되는 1종 이상의 아미노산일 수 있으며, 일 예에서 L-트립토판일 수 있다.
상기 단계에서, 암모니아의 첨가는 방향족 아미노산 결정을 포함하는 반응액 및/또는 배양액의 pH를 증가시키고, 반응액 내 방향족 아미노산의 용해도를 증가시킬 수 있다. 상기 암모니아에 있어서, 최초의 공정에서는 결정화 피드(Crystallization Feed)를 생성하기 위하여, 별도의 암모니아, 예를 들어, 20 내지 30%(v/v), 20 내지 28%(v/v), 20 내지 26%(v/v), 20 내지 24%(v/v), 20 내지 22%(v/v), 22 내지 30%(v/v), 22 내지 28%(v/v), 22 내지 26%(v/v), 22 내지 24%(v/v), 24 내지 30%(v/v), 24 내지 28%(v/v), 24 내지 26%(v/v), 26 내지 30%(v/v), 26 내지 28%(v/v), 또는 28 내지 30%(v/v)의 암모니아수, 예컨대 26%(v/v)의 암모니아수를 첨가하는 것일 수 있으며, 이후의 공정에서는 (c) 단계로부터 회수한 기체로부터 유래한 암모니아를 첨가하는 것일 수 있다.
일 구현예에 있어서, 상기 (a) 단계에서 수득된 방향족 아미노산 결정이 용해된 용해액의 pH는 10 내지 12일 수 있다. 상기 용해액의 pH가 10 이하인 경우에는 용해액 내 방향족 아미노산 결정이 존재하여, 용해액의 결정화 효율을 감소시킬 수 있다. 상기 방향족 아미노산 용해액의 pH는 예를 들어, 10 내지 12, 10 내지 11.5, 10 내지 11, 10 내지 10.5, 10.5 내지 12, 10.5 내지 11.5, 10.5 내지 11, 11 내지 12, 11 내지 11.5, 또는 11.5 내지 12의 범위를 갖는 것일 수 있으나, 아미노산의 종류, 기타 반응 조건 등에 따라 적절하게 조절 가능하다. 상기 방향족 아미노산 결정을 포함하는 반응액 및/또는 배양액에 첨가되는 암모니아의 농도는 상기 반응액 및/또는 배양액의 pH를 상기 범위를 만족시키기 위한 정도로 사용되는 것일 수 있다.
일 구현예에 있어서, 상기 방향족 아미노산 결정이 용해된 용해액은 방향족 아미노산 결정이 용해됨에 따라 현탁액에서 투명한 용액의 형태로 변화된 것일 수 있다.
일 구현예에 있어서, 방향족 아미노산 결정을 포함하는 반응액은 방향족 아미노산이 과포화된 용액으로 현탁액 형태인 것일 수 있으며, 예를 들어, 방향족 아미노산 결정을 포함하는 용액 또는 발효액을 포함할 수 있다.
상기 방향족 아미노산 결정을 포함하는 용액은 방향족 아미노산 결정 및 증류수를 혼합한 용액일 수 있고, 상기 방향족 아미노산 결정을 포함하는 발효액은 방향족 아미노산을 생산하는 미생물을 배지에서 배양하여 수득되는 것일 수 있다.
상기 방향족 아미노산을 생산하는 미생물은 방향족 아미노산 생산능이 있는 미생물이면 특별히 제한이 없으며, 예를 들어, 코리네박테리움(Corynebacterium) 속 또는 에스케리키아(Escherichia) 속일 수 있고, 구체적으로, 코리네박테리움 글루타미쿰(Corynebacterium glutamicum) 균주 또는 이의 변이주일 수 있다.
본 출원에서 용어, “배양”은 미생물을 적당히 인공적으로 조절한 환경조건에서 생육시키는 것을 의미한다. 본 출원에서 방향족 아미노산의 생산능을 가지는 미생물을 이용한 방향족 아미노산 생산 방법은 당업계에 널리 알려져 있는 방법을 이용하여 수행할 수 있다. 구체적으로, 상기 배양은 배치 공정, 주입 배치 또는 반복 주입 배치 공정(fed batch or repeated fed batch process)에서 연속식으로 배양할 수 있으나, 이에 제한되는 것은 아니다.
배양에 사용되는 배지는 적절한 방식으로 특정 균주의 요건을 충족해야 한다. 예를 들어, 코리네박테리움 속 균주에 대한 배양 배지는 공지되어 있다 (예를 들면, Manual of Methods for General Bacteriology. American Society for Bacteriology. Washington D.C., USA, 1981). 사용될 수 있는 당원으로는 글루코즈, 사카로즈, 락토즈, 프락토즈, 말토즈, 전분, 셀룰로즈와 같은 당 및 탄수화물, 대두유, 해바라기유, 피마자유, 코코넛유 등 과 같은 오일 및 지방, 팔미트산, 스테아린산, 리놀레산과 같은 지방산, 글리세롤, 에탄올과 같은 알코올, 아세트산과 같은 유기산이 포함될 수 있다. 이들 물질은 개별적으로 또는 혼합물로서 사용될 수 있으며, 이에 제한되는 것은 아니다. 사용될 수 있는 질소원으로는 펩톤, 효모 추출물, 육즙, 맥아 추출물, 옥수수 침지액, 대두밀 및 요소 또는 무기화합물, 예를 들면 황산암모늄, 염화암모늄, 인산암모늄, 탄산 암모늄 및 질산암모늄이 포함될 수 있다. 질소원 또한 개별적으로 또는 혼합물로서 사용할 수 있으며, 이에 제한되는 것은 아니다. 사용될 수 있는 인원으로는 인산이수소칼륨 또는 인산수소이칼륨 또는 상응하는 나트륨-함유 염이 포함될 수 있다.
또한, 배양 배지는 성장에 필요한 황산마그네슘 또는 황산철과 같은 금속염을 함유할 수 있다. 추가적으로, 아미노산 및 비타민과 같은 필수 성장 물질이 사용될 수 있다. 또한, 배양 배지에 적절한 전구체들이 사용될 수 있다. 상기된 원료들은 배양과정에서 배양물에 적절한 방식에 의해 회분식으로 또는 연속식으로 첨가될 수 있다. 그러나, 이에 제한되는 것은 아니다.
(b) 단계
이후, 본 출원의 방법은 (b) 단계, 상기 수득된 방향족 아미노산 결정이 용해된 용해액을 결정화하여 방향족 아미노산 결정을 포함하는 농축액을 수득하는 단계 (농축 결정화)를 포함할 수 있다.

(c) 단계
이후, 본 출원의 방법은 (c) 단계, 상기 결정화 과정에서 생성된, 암모니아를 포함하는 기체를 회수(수득)하는 단계를 포함할 수 있다.
상기 (c) 단계의 암모니아를 포함하는 기체는 암모니아 및 수증기를 포함하는 혼합 기체일 수 있다.
일 구현예에 있어서, 상기 (c) 단계는 전술한 방향족 아미노산의 결정을 포함하는 농축액을 수득하는 (b) 단계와 동시 또는 순차적으로 실시되는 것일 수 있다.
일 구현예에 있어서, 상기 단계는 전술한 상기 (b) 단계의 방향족 아미노산 결정이 용해된 용해액의 pH가 5.5 내지 8이 될 때까지 실시하는 것일 수 있고, 구체적으로, 상기 용해액의 pH가 5.5 내지 8, 5.5 내지 7.5, 5.5 내지 7, 5.5 내지 6.5, 5.5 내지 6, 6 내지 8, 6 내지 7.5, 6 내지 7, 6 내지 6.5, 6.5 내지 8, 6.5 내지 7.5, 6.5 내지 7, 7 내지 8, 7 내지 7.5, 또는 7.5 내지 8이 될 때까지 실시하는 것일 수 있으나 이에 제한되는 것은 아니다. 상기 용해액의 pH가 상기 범위, 예컨대 8 미만인 경우에는 수득된 혼합 기체 내 암모니아의 농도가 낮으므로, 회수된 암모니아의 농도를 저하시켜 방향족 아미노산의 결정화 효율을 감소시킬 수 있다.
또한, 상기 암모니아를 포함하는 기체는 수증기와 암모니아가 혼합되어 있는 증기 상태의 기체일 수 있으며, 상기 혼합 기체의 조성비는 물-암모니아-방향족 아미노산의 기액상 평형계에 지배적인 영항을 받기 때문에, 결정화 장치의 내부 온도 및 압력 조건에 따라 다양한 분포를 가질 수 있다.
(d) 단계
이후, 본 출원의 방법은 상기 회수(수득)한 기체로부터 유래한 암모니아를 상기 (a) 단계의 암모니아로 재사용하여 방향족 아미노산 결정이 용해된 용해액을 다시 수득하는 단계를 포함할 수 있다.
일 구현예에 있어서, 상기 단계는 상기 암모니아를 증기 상태로 재사용하거나, 상기 증기가 압축 또는 응축된 액체 상태로 재사용하는 것일 수 있다.
상기 단계에 앞서서, 회수한 기체를 응축 또는 압축하여, 기체 내 암모니아를 액체 상태로 전환시키는 단계를 추가로 포함할 수 있으며, 예를 들어, 냉각기를 사용하여 혼합 기체를 응축하거나, 압축기를 사용하여 혼합 기체를 압축하거나, 상기 냉각기와 압축기를 동시에 사용하는 것일 수 있다.
일 구현예에 있어서, 상기 단계는 고농도의 방향족 아미노산 결정을 다량 포함하는 농축액을 수득할 수 있을 때까지 반복 실시될 수 있다. 예를 들어, 상기 단계는 1회 이상, 2회 이상, 3회 이상, 4회 이상, 5회 이상, 또는 2 내지 50회, 2 내지 45회, 2 내지 40회, 2 내지 35회, 2 내지 30회, 2 내지 25회, 2 내지 20회, 2 내지 15회, 2 내지 10회, 또는 2 내지 5회 반복 실시될 수 있으나, 이에 제한되는 것은 아니다.
본 출원의 방법은 (c) 단계에서 회수한 기체로부터 유래한 암모니아를 상기 (a) 단계의 암모니아로 재사용하는 단계를 포함함으로써, 다량의 pH 조절 물질을 필요로 하지 않아 경제적이고, 방향족 아미노산 결정화 효율을 향상시킬 수 있으며, 지속 가능한 순환이 어려운 염 폐기물 생성을 감소시켜 친환경적인 방법으로 활용될 수 있다.
또한, 본 출원의 방법은 (b) 단계 이후에, 상기 방향족 아미노산 결정이 용해된 용해액으로부터 원심분리 설비를 활용하여 상기 결정을 분리하는 단계를 포함할 수 있다.
일 실시예에 따르면, 다수 회의 결정화 과정에서 생성되는 암모니아를 높은 수준의 회수율로 수득할 수 있으며, 상기 회수된 암모니아, 예를 들어, 암모니아수는 반응액에 존재하는 방향족 아미노산의 결정을 용해시키는데 재사용 됨으로써 경제성을 증대시킬 수 있다. 따라서, 상기 방법은 별도의 pH 조절제 없이 결정화 공정에 기반하여, 지속적으로 방향족 아미노산의 결정을 생성할 수 있고, 이에 따라, 방향족 아미노산 생산 효율을 향상시킬 수 있다.
본 출원의 방법은 상기 (b) 단계의 방향족 아미노산 결정을 포함하는 농축액으로부터 방향족 아미노산 결정을 분리하는 단계를 추가로 포함할 수 있다. 상기 분리하는 단계는 원심분리 및/또는 여과 단계를 포함할 수 있다. 상기 방향족 아미노산 결정을 분리하는 단계는, 예를 들어, 상기 (b) 단계와 (c) 단계 사이에, 상기 (c) 단계와 (d) 단계 사이에, 또는 상기 (d) 단계 이후에 이루어질 수 있고, 상기 (c) 단계 또는 (d) 단계와 동시에 이루어질 수 있으나, 상기 (b) 단계 이후라면 (c) 단계 및 (d) 단계와의 순서와는 무관하게 어느 단계에나 이루어질 수 있다.
상기 단계에서, 농축액으로부터 방향족 아미노산 결정의 분리는 예를 들어, 상기 방향족 아미노산 결정을 포함하는 농축액을 고액 분리하여 방향족 아미노산 습정(Wet crystal)을 수득하는 단계; 및 상기 수득된 습정을 건조하여 방향족 아미노산 결정을 수득하는 단계를 포함할 수 있으나, 아미노산 결정의 분리 및 정제와 관련된, 당업계에 알려진 통상의 기술이 비제한적으로 적용될 수 있다.
이후, 본 출원의 방법은 상기 (d) 단계에서 수득된 용해액을 결정화하여 방향족 아미노산 결정을 포함하는 농축액을 수득하는 단계 (농축 재결정화)를 포함할 수 있다.
일 구현예에 있어서, 상기 (a) 단계 내지 (d) 단계는 1회 이상, 2회 이상, 3회 이상, 4회 이상, 5회 이상, 또는 또는 2 내지 50회, 2 내지 45회, 2 내지 40회, 2 내지 35회, 2 내지 30회, 2 내지 25회, 2 내지 20회, 2 내지 15회, 2 내지 10회, 또는 2 내지 5회 반복 수행될 수 있으나, 이에 제한되는 것은 아니다.
또한, 본 출원은 다른 양상으로, (a) 방향족 아미노산 결정을 포함하는 반응액과 암모니아를 혼합하여 상기 방향족 아미노산 결정이 용해된 용해액을 수득하는 단계 (결정 용해);
(b) 상기 수득된 용해액을 결정화하여 방향족 아미노산 결정을 포함하는 농축액을 수득하는 단계 (농축 결정화);
(c) 상기 결정화 과정에서 생성된, 암모니아를 포함하는 기체를 회수하는 단계; 및

상기 방향족 아미노산 및 방향족 아미노산의 결정화 방법은 상술한 바와 같다.
본 출원에 따른 방향족 아미노산의 결정화 방법은 방향족 아미노산의 용해도 증가를 위해 최초로 첨가된 암모니아를 결정화 과정에서 기체 상태로 회수하여, 이를 재사용함으로써, 방향족 아미노산 결정의 생산 효율을 향상시킬 수 있고, 다량의 pH 조절 물질 및 추가적인 중화 공정이 필요하지 않으므로 생산 비용이 절감될 수 있다.
또한 방향족 아미노산 결정을 암모니아 처리를 통한 용해 후 농축 결정화 진행을 통한 재결정화로 입도면에서 우수한 방향족 아미노산 결정을 형성할 수 있다. 이로서 형성된 방향족 아미노산 결정은 분리를 통해 습정의 형태로 회수 가능하다.

이하 본 발명을 실시예를 통하여 보다 상세하게 설명한다. 그러나, 이들 실시예는 본 발명을 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다. 또한, 본 명세서에 기재되지 않은 내용은 본 출원의 기술 분야 또는 유사 분야에서 숙련된 자이면 충분히 인식하고 유추할 수 있는 것이므로 그 설명을 생략한다.
실시예 1. 실험 재료 및 실험 방법
(1) 실험 재료
방향족 아미노산으로 L-트립토판은 순도 98% 이상의 제품을 사용하였으며, 모두 CJ 제일제당 제품을 사용하였다. 물은 직접 제조한 3차 증류수를 사용하였으며, 26%(v/v) 암모니아수는 대정화금社로부터 구매하여 사용하였다. 고성능 액체 크로마토그래피(high-performance liquid chromatograph; HPLC) 분석을 위한 0.1M 질산 수용액, 0.02M 디피콜린산 수용액은 Sigma-Aldrich (US)로부터 구매하여 사용하였다.
(2) L-트립토판 용해액의 농도 및 L-트립토판 결정의 순도 분석
L-트립토판 용해액의 농도 및 L-트립토판 결정의 순도 분석은 고성능 액체 크로마토그래피 (chromatography C18)를 이용하여 진행하였으며, 분석 조건은 다음과 같다:
컬럼: Hypersil gold HPLC column (Thermo Scientific, US)
컬럼 온도: 35℃
이동상: Potassium phosphate 7.35mM & Acetonitrile 용액
이동상 속도: 1.5 ml/분
감지기: UV detector.
(3) L-트립토판 용해액 내 암모니아 농도 분석
L-트립토판 용해액 내 암모니아 농도 분석은 이온 크로마토그래피 (model 930 compact IC Flex, Metrohm, Switzerland)를 이용하여 진행하였다.
컬럼: Metrosep C4-150 (Metrohm, Switzerland)
컬럼 온도: 25℃
이동상: 0.7 mM 질산 수용액+1.7 mM 디피콜린산 수용액
이동상 속도: 1.0 ml/분.
(4) L-트립토판 용해도 분석
L-트립토판 아미노산의 용해도 측정은 유리 재질의 1 L 자켓 반응기에서 진행하였다. 1 L 자켓 반응기에 증류수와 암모니아수를 다양한 비율로 섞은 후, 과량의 L-트립토판 결정을 첨가하고 이들을 교반함으로써, L-트립토판 결정을 포함하는 반응액을 제조하였다. 이후, 반응기의 내부 온도를 냉동/가열 순환 장치 (model F35, Julabo, Germany)를 사용하여 30℃로 일정하게 유지하면서, 이러한 조건을 교반과 함께 12시간 이상 동안 하였다. 이후, 교반을 멈추고 L-트립토판 결정이 모두 가라앉으면, 징명한 상태의 상등액 일부를 0.45μm 실린지 필터가 장착된 실린지 샘플러로 옮겨 담았다. 이때, 해당 용액의 pH를 pH 측정기 (model S220, Mettler Toledo, US)를 이용하여 측정하였다. 해당 샘플의 농도는 정량 플라스크를 사용하여 3차 증류수로 희석한 뒤, HPLC를 이용하여 측정하였다. 상기 상등액 샘플의 농도를 용해도로 상정하였으며, 용액의 pH에 따른 L-트립토판 용해도 변화를 도 2에 나타내었다. 즉, 용액의 pH가 10 이상이 되는 시점부터 L-트립토판의 용해도가 증가함을 확인할 수 있었다.
실시예 2. L-트립토판 결정을 포함하는 용액에서의 분리 및 정제
(1) L-트립토판의 결정화
L-트립토판의 결정화는 배양액 내에 암모니아를 처리한 반응액으로부터 진행하였다. L-트립토판 결정을 함유하는 배양액에 암모니아 처리를 통한 결정 용해를 진행한 후 농축 결정화를 통해 재결정화를 진행함으로서 입도 및 품질에 유리한 결정을 형성하는 단계를 거친다. 배양액 내에 처리한 암모니아는 26%(v/v)의 암모니아수이고, 암모니아를 처리함으로써 반응액의 pH를 10 내지 12 영역으로 조정하였다.
일 실시예에 따른 결정화 장치 및 암모니아 회수 장치의 개략도는 도 3에 나타낸 바와 같다. 구체적으로, 도 3에서 결정화 피드는 1회차 결정화 과정에서 수득된, L-트립토판 결정을 포함하는 농축액을 의미하며, 결정화는 유리 재질의 20 L 자켓 반응기에서 진행하였고, 열교환부에서의 스케일링 방지를 위하여, 자켓은 내부 액체의 부피가 10 L가 되는 지점의 높이까지만 설치하였다. 자켓 부위는 냉동/가열 순환 장치 (model F35, Julabo, Germany)로 온도가 제어되는 3차 증류수로 채워졌으며, L-트립토판 결정화 공정이 진행되는 동안 온도는 80℃로 유지하였다. 내부 반응액 등의 교반은 Teflon 재질의 4-blade impeller로 회전 속도 조절이 가능한 교반기 (model RW-20, IKA, Germany)를 이용하여 진행하였고, 결정화가 진행되는 동안 교반 속도는 200 rpm을 유지하였다. 반응기 내부의 압력 감소는 배관으로 연결된 압축기(Compressor)를 이용하여 진행하였으며, 압축기와 결정화기 사이에는 압력 조절을 위한 전자식 진공 제어기(model NVC 2300-A, Eyela, Japan)가 설치되어 이들의 압력을 조절하였다. 결정화기 내부의 압력은 100 mbar로 유지하였다. 암모니아 회수부는 20 bar까지 저장 가능한 스테인리스강 재질의 20 L 자켓 압력 용기로 구성하였으며, 자체 설비로 공급되는 4℃의 냉각수로 냉각을 진행하였다. 이때, 암모니아의 회수는 결정화기 내부의 pH가 8 미만이 될 때까지만 진행하였고, 이보다 낮은 pH에서는 회수 암모니아의 농도가 낮기 때문에 암모니아의 회수를 진행하지 않았다. 최종 농축액의 농도는 암모니아가 투입되기 전의 L-트립토판 결정을 포함하는 반응액의 농도 대비 3배가 되도록 조절하였다.
(2) L-트립토판 결정의 분리 및 건조
상기 최종 농축 농도에 이르면, 결정화 공정을 종료하고 반응기 내 농축액을 하부에 배치된 배출 배관을 통해 회수하였다. 이후, 고원심 데칸커를 사용하여 4000rpm의 속도로 고액 분리하였다. 필요에 따라, 증류수를 이용하여 분리 초기에 세척 공정을 진행하였다. 이후, 수득된 습정(wet crystal)을 80℃의 오븐 건조기에서 무게 변화가 없을 때까지 건조하여 L-트립토판의 결정을 수득하였다.
(3) 실험 결과: L-트립토판의 결정화 및 암모니아 회수
98% 이상 순도의 L-트립토판 결정으로 제조한 20 L 현탁액을 이용하여 L-트립토판의 결정화 및 암모니아 회수 실험을 진행하였다.
결정화는 각 조건당 동일한 공정 조건에서 총 5회 진행하였다. 단, 1회차 결정화 공정에서는 결정화 피드를 제조하기 위하여, L-트립토판 결정 및 증류수를 혼합하여 제조한 L-트립토판 결정을 포함하는 용액에 26%(v/v) 암모니아수를 첨가하여 반응액의 pH를 10 내지 12 영역으로 조정하고, L-트립토판 결정을 전량 용해하였다. 이후의 회차에서는 L-트립토판 결정을 포함하는 반응액에 회수한 암모니아수를 첨가하여 L-트립토판 결정을 용해하였다(실험군). 한편, 대조군으로는 암모니아수의 회수 과정없이 L-트립토판 결정을 포함하는 반응액에 26%(v/v)의 암모니아수를 첨가하고, 뒤이어 98%(v/v)의 황산으로 pH를 7로 재조정한 군을 사용하였다.
실험은 총 5회 진행하였으며, 해당 실험결과 중 2회차부터 5회차까지의 결정화 공정에 대한 평균 값을 아래 표 1에 나타내었다. 15 내지 80 g/L의 L-트립토판의 농도 범위를 가지는 반응액을 대상으로, 회수된 암모니아수를 첨가하여 상기 반응액의 pH를 9에서 12의 범위로 조정하여 결정의 용해를 진행하였으며, 이후 결정화를 진행하면서 다시 암모니아수를 회수하였다.
하기 표 1에 나타낸 바와 같이, 대조군에서는 회수된 암모니아수가 없었으나, 일 실시예에 따른 결정화 공정에서는 5 내지 23% 농도의 암모니아수를 90% 수준의 회수율로 확보할 수 있었으며, 상기 회수된 암모니아수는 반응액 내 L-트립토판의 결정을 용해하는데 재사용될 수 있었다.

실시예 3. L-트립토판 결정을 포함하는 발효액에서의 분리 및 정제
(1) L-트립토판을 포함하는 발효액의 제조
L-트립토판을 생산하는 균주인 코리네박테리움 KCCM12218P (대한민국 특허 출원번호 제10-2018-0022057호)를 이용하여 L-트립토판을 포함하는 발효액을 제조하였다.
구체적으로, 전배양 배지 40 mL를 500 mL 진탕용 삼각 플라스크에 분주하고, 121℃에서 15분간 가압 살균한 후, 균주를 식균하여 33℃에서 200 rpm으로 교반시키면서 회전 교반 배양기에서 24시간 동안 배양하였다. 이후, 5 L 발효조에 종배양 배지 3 L를 채우고 121℃에서 30분간 가압 살균한 후, pH를 7.0으로 조절하고, 상기 전배양물 4%를 식균하여 33℃에서 800 rpm 및 통기량 0.5 vvm 조건으로 OD 값이 20이 될 때까지 배양하여 종배양을 실시하였다. 이후, 5 L 발효조에 본배양 배지 2.1 L를 채우고 121℃에서 30분간 가압 살균한 후, 포도당을 0.6 L씩 첨가하고, 암모니아 가스를 이용하여 pH를 7.0으로 조절하였다. 상기 종배양액을 준비된 본배양조에 20%로 식균하여 배양온도 33℃ 및 통기량 1.0 vvm 조건에서 용존산소가 최소 30% 이상 유지되도록 400 내지 800 rpm까지 조절하면서 42시간 동안 배양하여 L-트립토판을 포함하는 각 발효액을 제조하였다. 상기 배양 과정에 사용된 전배양, 종배양 및 본배양 배지 조성은 하기 표 2와 같다.

(2) L-트립토판 결정의 분리 및 건조
L-트립토판 결정을 포함하는 용액 대신 상기 제조한 L-트립토판 결정을 포함하는 발효액을 이용하여 실시예 2의 (1) 및 (2)에 기재된 것과 동일한 방법으로 암모니아수(26%(v/v)) 첨가, 결정화, L-트립토판 결정의 분리 및 건조를 수행하였다. 결정의 분리 방법으로 원심력을 활용한 데칸터 장비가 사용되었으며 원심력은 4000rpm 수준으로 진행되었다. 필요 시 물 투입을 통해 세척을 진행하였다.
(3) 실험 결과: L-트립토판의 정제
상기 실시예 3의 (1)에서 제조한, L-트립토판 결정을 포함하는 L-트립토판 농도 60 g/L의 발효액으로부터 L-트립토판 정제를 5회 반복하여 시행하였다. L-트립토판 결정을 포함하는 발효액 20 L에 회수된 암모니아수(단, 최초에는 암모니아수 시약)를 첨가하여 pH를 10까지 조정하고, L-트립토판을 결정 전량 용해한 후, 농축 결정화를 180 g/L의 농도까지 진행하였으며, L-트립토판 결정을 포함하는 농축액은 원심분리 설비를 이용하여 고액 분리하였다. 이때, 세척은 필요시에 20 vol%의 3차 증류수를 이용하여 실시하였다.
이후 건조를 통해 최종 회수된 결정의 무게는 5회 평균 1.1 kg이었고, 결정의 순도는 5회 평균 98.2%였다. 일 실시예에 따른 결정화 공정에서는 13% 농도의 암모니아수를 90% 이상의 회수율로 확보할 수 있었으며, 상기 회수된 암모니아수는 발효액 내 L-트립토판의 결정을 용해하는데 재사용될 수 있었다.
전술한 본 발명의 설명은 예시를 위한 것이며, 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명의 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 쉽게 변형이 가능하다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해해야만 한다.
Claims (10)
- (a) 방향족 아미노산 결정을 포함하는 반응액과 암모니아를 혼합하여 상기 방향족 아미노산 결정이 용해된 용해액을 수득하는 단계;(b) 상기 수득된 용해액을 결정화하여 방향족 아미노산 결정을 포함하는 농축액을 수득하는 단계;(c) 상기 결정화 과정에서 생성된, 암모니아를 포함하는 기체를 회수하는 단계; 및(d) 상기 회수한 기체로부터 유래한 암모니아를 상기 (a) 단계의 암모니아로 재사용하는 단계;를 포함하고,상기 (b) 단계 및 (c) 단계는 동시 또는 순차적으로 실시되고,상기 (a) 단계 내지 (d) 단계는 반복 수행되는 것을 특징으로 하는,방향족 아미노산의 결정화 방법.
- 제1항에 있어서, 상기 방향족 아미노산은 L-트립토판, L-타이로신, L-페닐알라닌, 및 L-히스티딘으로 이루어지는 군에서 선택되는 1종 이상의 아미노산인, 방향족 아미노산의 결정화 방법.
- 제1항에 있어서, 상기 방향족 아미노산 결정을 포함하는 반응액은 방향족 아미노산 결정을 포함하는 용액 또는 발효액을 포함하는 것인, 방향족 아미노산의 결정화 방법.
- 제3항에 있어서, 상기 방향족 아미노산 결정을 포함하는 발효액은 방향족 아미노산을 생산하는 미생물을 배지에서 배양하여 수득되는 것인, 방향족 아미노산의 결정화 방법.
- 제1항에 있어서, 상기 (a) 단계에서, 상기 방향족 아미노산 결정이 용해된 용해액의 pH는 10 내지 12인 것인, 방향족 아미노산의 결정화 방법.
- 제1항에 있어서, 상기 (b) 단계의 방향족 아미노산의 결정을 포함하는 농축액의 방향족 아미노산의 농도는 (a) 단계의 방향족 아미노산 결정을 포함하는 반응액의 방향족 아미노산 농도 대비 1.3 내지 3배인, 방향족 아미노산의 결정화 방법.
- 제1항에 있어서, 상기 (c) 단계는 상기 (b) 단계의 방향족 아미노산 용해액의 pH가 8 미만이 될 때까지 실시하는 것인, 방향족 아미노산의 결정화 방법.
- 제1항에 있어서, 상기 (d) 단계는 상기 암모니아를 증기 상태로 재사용하거나, 상기 증기가 압축 또는 응축된 액체 상태로 재사용하는 것인, 방향족 아미노산의 결정화 방법.
- 제1항 내지 제8항 중 어느 한 항의 방향족 아미노산의 결정화 방법에 의해 생산된, 방향족 아미노산 결정.
- 제9항에 있어서, 상기 방향족 아미노산은 L-트립토판, L-타이로신, L-페닐알라닌, 및 L-히스티딘으로 이루어지는 군에서 선택되는 1종 이상의 아미노산인, 방향족 아미노산 결정.
