図1は、本発明の第1実施形態の模擬血管を示す図である。本実施形態においては、模擬血管1は、基材層2と、導電層3とを備える。
基材層は、0.01~50MPa以下の引張弾性率及び50~200℃の融点を有する熱可塑性樹脂組成物から成形された管状の構造体となっている。基材層は、1種類の熱可塑性樹脂組成物もしくは2種類又は3種類以上の熱可塑性樹脂組成物をアロイした熱可塑性樹脂組成物から成形された単層構造であってもよく、2種類又は3種類以上の熱可塑性樹脂組成物を用いて成形された多層構造であってもよい。
基材層の内径は、0.3~9.8mmであることが好ましく、0.3~3.6mmであることがより好ましく、0.4~2.0mmであることがさらに好ましい。
JIS K 6768に従い、英弘精機株式会社製の自動接触角測定装置OCA20を用いて、プライマー層表面に水(純水)を1μL滴下し、30秒後に測定した場合の接触角を本実施形態における接触角とした。なお、接触角の測定は、基材層の材料である熱可塑性樹脂組成物を160~200℃にてプレス成型した厚さ1.0mm厚の樹脂シートに加工した検体を用いて測定することができる。
引張弾性率は、JIS K7127のプラスチックの引張試験方法に従って、1.0mm厚シートを試験片タイプ5の形状に打ち抜き、23±1℃の環境下にて、引張試験機(島津製作所製、オートグラフAG-Xplus)を用い、引張速度50.0mm/minの条件にて測定し、応力-ひずみ曲線における初期の直線の傾きから算出される。
本実施形態においては、融点は、示差走査熱量計(DSC;METTLER TOLEDO社製、DSC 3+)を用い、窒素雰囲気下、昇温速度10℃/minにて測定される。
導電層は、基材層の外側表面の少なくとも一部に配置された層であって、導電性ポリマーを含有する。本実施形態において「少なくとも一部に配置されている」とは、導電層が、少なくともエネルギーデバイスによる熱凝固法に用いるために必要な面積を有するように配置されていることを意味している。本発明の一実施形態において、導電層は、基材層の外側表面の5%未満、5%以上、10%以上、30%以上、50%以上、70%以上もしくは90%以上の面積を有している。
本発明の一実施形態において、導電性ポリマーは、π共役系導電性高分子である。π共役系導電性高分子としては、主鎖がπ共役系で構成されている有機高分子であれば本発明の効果を有する限り特に制限されず、例えば、ポリチオフェン系導電性高分子、ポリピロール系導電性高分子、ポリアセチレン系導電性高分子、ポリフェニレン系導電性高分子、ポリフェニレンビニレン系導電性高分子、ポリアニリン系導電性高分子、ポリアセン系導電性高分子、ポリチオフェンビニレン系導電性高分子、及びこれらの共重合体等が挙げられる。空気中での安定性の点からは、ポリピロール系導電性高分子、ポリチオフェン系導電性高分子及びポリアニリン系導電性高分子が好ましく、透明性の面から、ポリチオフェン系導電性高分子がより好ましい。
ポリピロール系導電性高分子としては、ポリピロール、ポリ(N-メチルピロール)、ポリ(3-メチルピロール)、ポリ(3-エチルピロール)、ポリ(3-n-プロピルピロール)、ポリ(3-ブチルピロール)、ポリ(3-オクチルピロール)、ポリ(3-デシルピロール)、ポリ(3-ドデシルピロール)、ポリ(3,4-ジメチルピロール)、ポリ(3,4-ジブチルピロール)、ポリ(3-カルボキシピロール)、ポリ(3-メチル-4-カルボキシピロール)、ポリ(3-メチル-4-カルボキシエチルピロール)、ポリ(3-メチル-4-カルボキシブチルピロール)、ポリ(3-ヒドロキシピロール)、ポリ(3-メトキシピロール)、ポリ(3-エトキシピロール)、ポリ(3-ブトキシピロール)、ポリ(3-ヘキシルオキシピロール)、ポリ(3-メチル-4-ヘキシルオキシピロール)が挙げられる。
ポリアニリン系導電性高分子としては、ポリアニリン、ポリ(2-メチルアニリン)、ポリ(3-イソブチルアニリン)、ポリ(2-アニリンスルホン酸)、ポリ(3-アニリンスルホン酸)が挙げられる。
上記π共役系導電性高分子の中でも、導電性、透明性、耐熱性の点から、ポリ(3,4-エチレンジオキシチオフェン)が特に好ましい。
前記π共役系導電性高分子は1種を単独で使用してもよいし、2種以上を併用してもよい。
ポリアニオンとは、アニオン基を有するモノマー単位を、分子内に2つ以上有する重合体である。このポリアニオンのアニオン基は、π共役系導電性高分子に対するドーパントとして機能して、π共役系導電性高分子の導電性を向上させる。
ポリアニオンのアニオン基としては、スルホ基、またはカルボキシ基であることが好ましい。
このようなポリアニオンの具体例としては、ポリスチレンスルホン酸、ポリビニルスルホン酸、ポリアリルスルホン酸、ポリアクリルスルホン酸、ポリメタクリルスルホン酸、ポリ(2-アクリルアミド-2-メチルプロパンスルホン酸)、ポリイソプレンスルホン酸、ポリスルホエチルメタクリレート、ポリ(4-スルホブチルメタクリレート)、ポリメタクリルオキシベンゼンスルホン酸等のスルホン酸基を有する高分子や、ポリビニルカルボン酸、ポリスチレンカルボン酸、ポリアリルカルボン酸、ポリアクリルカルボン酸、ポリメタクリルカルボン酸、ポリ(2-アクリルアミド-2-メチルプロパンカルボン酸)、ポリイソプレンカルボン酸、ポリアクリル酸等のカルボン酸基を有する高分子が挙げられる。これらの単独重合体であってもよいし、2種以上の共重合体であってもよい。
これらポリアニオンのなかでも、帯電防止性をより高くできることから、スルホン酸基を有する高分子が好ましく、ポリスチレンスルホン酸がより好ましい。
前記ポリアニオンは1種を単独で使用してもよいし、2種以上を併用してもよい。
ポリアニオンの質量平均分子量は2万以上100万以下であることが好ましく、10万以上50万以下であることがより好ましい。
本明細書における質量平均分子量は、ゲルパーミエーションクロマトグラフィで測定し、標準物質をポリスチレンとして求めた値である。
ただし、本態様におけるポリアニオンにおいては、全てのアニオン基がπ共役系導電性高分子にドープすることはなく、ドープに寄与しない余剰のアニオン基を有するようになっている。
本発明の一実施形態において使用される分散媒としては、水、有機溶剤、水と有機溶剤との混合液が挙げられる。
有機溶剤としては、例えば、エステル系溶剤、エーテル系溶剤、炭化水素系溶剤、窒素原子含有溶剤、アルコール系溶剤、ケトン系溶剤等が挙げられる。
エステル系溶剤としては、例えば、酢酸エチル、酢酸プロピル、酢酸ブチル等が挙げられる。
エーテル系溶剤としては、例えば、ジエチルエーテル、ジメチルエーテル、エチレングリコール、プロピレングリコール、プロピレングリコールジアルキルエーテル、ジエチレングリコールジエチルエーテル等が挙げられる。
炭化水素系溶剤としては、例えば、ヘキサン、シクロヘキサン、ペンタン、オクタン、デカン、ドデカン、ベンゼン、トルエン、キシレン、エチルベンゼン、プロピルベンゼン、イソプロピルベンゼン等が挙げられる。
窒素原子含有溶剤としては、例えば、N-メチルピロリドン、ジメチルアセトアミド、ジメチルホルムアミド等が挙げられる。
アルコール系溶剤としては、例えば、メタノール、エタノール、1-プロパノール、2-プロパノール、2-メチル-2-プロパノール、1-ブタノール、2-ブタノール、2-メチル-1-プロパノール、アリルアルコール、プロピレングリコールモノメチルエーテル、エチレングリコールモノメチルエーテル等が挙げられる。
ケトン系溶剤としては、例えば、ジエチルケトン、メチルプロピルケトン、メチルブチルケトン、メチルイソプロピルケトン、メチルイソブチルケトン、メチルアミルケトン、ジイソプロピルケトン、メチルエチルケトン、アセトン、ジアセトンアルコール等が挙げられる。
前記有機溶剤は1種を単独で使用してもよいし、2種以上を併用してもよい。
前記分散媒のなかでも、エステル系溶剤、炭化水素系溶剤、エーテル系溶剤、N-メチルピロリドンよりなる群から選ばれる少なくとも1種の溶剤が好ましい。さらには、前記分散媒のなかでも、酢酸エチル、酢酸ブチル、ヘプタン、トルエン、ジエチレングリコールジエチルエーテルよりなる群から選ばれる少なくとも1種がより好ましい。
本発明の一実施形態における分散液は、前記導電性ポリマーと共に透明バインダ成分を含有する。透明バインダ成分は、熱可塑性樹脂、熱硬化性化合物、光硬化性化合物等であって、透明な化合物が挙げられる。熱可塑性樹脂はそのままで後述のバインダ樹脂となる。熱硬化性化合物及び光硬化性化合物は、各々、モノマー又はオリゴマーであり、熱硬化性化合物は熱硬化することで後述のバインダ樹脂となり、光硬化性化合物は光硬化することで後述のバインダ樹脂となる。
導電性ポリマーと共に透明バインダ成分を含有する分散液であれば、透明性、導電性及び反射防止性を有する導電層を容易に形成できる。
前記透明バインダ成分のうち、熱可塑性樹脂としては、アクリル樹脂、ポリスチレン、スチレン-アクリル共重合体、ポリカーボネート、ポリエチレンテレフタレート、ポリブチレンテレフタレート、ポリエチレンナフタレート、環状ポリオレフィンが挙げられる。
前記透明バインダ成分のうち、熱硬化性化合物としては、アクリル化合物、エポキシ化合物が挙げられる。
光硬化性化合物としては、アクリル化合物が挙げられる。
前記透明バインダ成分のなかでも、透明性が高く、容易に硬化でき、低コストであることから、光硬化性のアクリル化合物が好ましい。
透明バインダ成分は、1種を単独で使用してもよいし、2種以上を併用してもよい。
透明バインダ成分が熱硬化性化合物である場合、導電性粒子分散液に熱重合開始剤を含有させることが好ましく、透明バインダ成分が光硬化性化合物である場合、分散液に光重合開始剤を含有させることが好ましい。
導電性ポリマー分散液を塗工するに前には、前記基材層にコロナ放電処理、プラズマ放電処理、火炎処理等を施し、基材層表面の親水基(ヒドロキシル基、カルボニル基、カルボキシル基等)を形成させることが好ましい。基材層が親水化処理されていると、導電層の接着性をさらに向上させることができる。
本実施形態において、模擬血管は、管状の基材層と、基材層の外側表面に配置された導電層とを有する。本発明の一実施形態において、模擬血管の直径、つまり外径は0.5~10.0mmである。本発明の別の実施形態において、模擬血管の直径は、0.7~5.0mmであることが好ましく、1.0~3.0mmであることがより好ましい。基材層の直径を0.5mm以上とすることで成形時の加工安定性及び潰瘍モデル作製時のハンドリング性が高くなり、模擬血管の直径を10.0mm以下とすることで人体血管の再現性が高くなり、エネルギーデバイスによる通電不良を抑制できる。
模擬血管の内径は、0.3~8.0mmであることが好ましく、0.3~3.6mmであることがより好ましく、0.4~2.0mmであることがさらに好ましい。
本実施形態において、表面抵抗値は、JIS C2139に従い、株式会社三菱ケミカルアナリテック社製ハイレスタUXMCP-HT800及び、ロレスタ-GP(MCP-T610)を用いて、23±1℃の条件にて測定される。測定に際しては、熱可塑性樹脂組成物を160~200℃にてプレス成型し、2.5cm×2.5cm、厚さ1.0mmの樹脂シートに加工し、表面に導電層を形成した検体を用いて評価される。
本実施形態において、全光線透過率は、JIS K 7136-1に従い、日本電飾工業社製 ヘイズ Meter「NDH7000」を使用して測定することができる。測定に際しては、熱可塑性樹脂組成物を1.0mm厚の樹脂シートに加工し、表面に導電層を形成した検体を用いて評価される。
本実施形態の模擬血管は、熱可塑性樹脂組成物を押出成形することによって基材層を製造し、基材層の外側表面に導電性ポリマーが分散媒中に分散された分散液を塗工することによって製造される。別の実施形態にいては、模擬血管は導電性ポリマー分散液に浸漬させる方法等することによって形成される。
導電性ポリマー分散液を塗工するに前には、前記基材層にコロナ放電処理、プラズマ放電処理、火炎処理等を施し、基材層表面の親水基(ヒドロキシル基、カルボニル基等)を形成させることが好ましい。基材層が親水化処理されていると、導電層の接着性をさらに向上させることができる。
本発明の第2実施形態にかかる潰瘍モデル101は、対向する上面S1及び下面S2と、前記上面S1及び前記下面S2の間に設けられた第1実施形態の模擬血管5とを有する。図2は、本実施形態の潰瘍モデルの断面を示す図であり、図3は、潰瘍モデル101の一実施形態を示す上面図である。図2及び図3に示す潰瘍モデル101は、対向する略円形の上面S1及び下面S2と、上面S1及び下面S2の外縁同士を連続的に連結する外周側面S11と、上面S1及び下面S2の間に設けられた、入口開口部5a及び出口開口部5bを有する管状の模擬血管5とを有するモデル本体部10を備える。上面S1が、モデル本体部10の厚み方向に隆起した環状の隆起面S10と、隆起面S10に囲まれた凹部の底面である模擬潰瘍面S20とを含む。ここで、環状の隆起面S10の形状は、図1のように上面S1側から見たときに円形に沿う形状であってもよいが、模擬潰瘍面S20を取り囲む形状であれば、特に制限されない。例えば、隆起面S10を上面S1側から見たときに、隆起面S10の外周及び内周が、それぞれ独立に、円形、楕円形、多角形又は不定形から選ばれる任意の形状を有することができる。同様に、模擬潰瘍面S20を上面S1側から見たときに、模擬潰瘍面S20の外周が、例えば円形、楕円形、多角形又は不定形から選ばれる任意の形状を有することができる。模擬血管5が、モデル本体部10の外周側面S11から模擬潰瘍面S20にかけて、出口開口部5bが模擬潰瘍面S20側に位置する向きでモデル本体部10の内部に延在している。図2に示される潰瘍モデル102のように、模擬血管5が1箇所以上で屈曲していてもよい。

(模擬血管の製造)
熱可塑性樹脂組成物としてエチレン-酢酸ビニル系樹脂(エバフレックスEV170、三井・ダウポリケミカル社製)を用いて、押出成形により直径1.0mm、厚み0.25mmの管状の構造体を作製し、基材層とした。基材層表面はコロナ処理による表面改質を行った。春日電機株式会社製のコロナ表面改質評価装置(TEC-4AX)を用いて、出力80W、速度0.5m/min、電極と基材とのクリアランスを5mmの条件で行った。
次いで、導電層としてポリチオフェン系導電性高分子分散液デナトロンPT-436(ナガセケムテックス株式会社、純粋/アルコール分散液)を準備し、基材層の外側表面に塗工し、室温環境下で24時間乾燥させ、厚み180nmの導電層を有する模擬血管とした。
水添スチレン系熱可塑性エラストマー100質量部に対し、300質量部のオイルを滴下し、十分に染みこませた。24時間23℃±1℃環境下で静置後、ブラベンダープラスチコーダー(ブラベンダー社製PL2000型)を使用し、180℃、回転速度100回/分、10分間混練した。
金型に水添スチレン系熱可塑性エラストマーとオイルとを混合させた熱可塑性樹脂組成物を充填させ、160℃で5分間、圧力10Mpaで溶融成形した。
成形した潰瘍モデルに模擬血管を図2及び図3、図4のように設置した。
次いで、模擬血管の端部に疑似血液が充填された注射器をつなぎ合わせた。
基材層の直径及び厚みを変更した以外は、実施例1と同様に模擬血管及び潰瘍モデルを作製した。
基材層の熱可塑性樹脂組成物をエチレン-酢酸ビニル系樹脂(ウルトラセン640)に変更した以外は、実施例1と同様に模擬血管及び潰瘍モデルを作製した。
基材層の熱可塑性樹脂組成物を熱可塑性ポリウレタンエラストマー樹脂(エラストランET880)に変更した以外は、実施例1と同様に模擬血管及び潰瘍モデルを作製した。
基材層の熱可塑性樹脂組成物を、塩化ビニル樹脂(TH-1000)100質量部に対して、アジピン酸系ポリエステル化合物(アデカサイザーPN-9302)を45部配合した樹脂組成物に変更した以外は、実施例1と同様に模擬血管及び潰瘍モデルを作製した。
基材層の熱可塑性樹脂組成物を水添スチレン-エチレンブタジエンブロック共重合体(DYNARON1320P)に変更した以外は、実施例1と同様に模擬血管及び潰瘍モデルを作製した。
基材層の熱可塑性樹脂組成物を1,2―ポリブタジエンエラストマー樹脂(RB810)に変更した以外は、実施例1と同様に模擬血管及び潰瘍モデルを作製した。
導電層を形成しなかった以外は、実施例1と同様に模擬血管及び潰瘍モデルを作製した。
基材層の熱可塑性樹脂組成物を低密度ポリエチレン樹脂(B128)に変更した以外は、実施例1と同様に模擬血管及び潰瘍モデルを作製した。
基材層の熱可塑性樹脂組成物を水添スチレン-ブタジエンブロック共重合体(DYNARON8300P)に変更した以外は、実施例1と同様に模擬血管及び潰瘍モデルを作製した。

表面抵抗率1.0×100以上、1.0×103Ω/□以下の場合を優、表面抵抗率1.0×103より大きく、1.0×106Ω/□以下の場合を良、表面抵抗率1.0×106Ω/□より大きい場合を不可とした。
JIS K 7136-1に従い、23±1℃の条件にて日本電色工業社製 ヘイズ Meter「NDH7000」を使用し、測定した全光線透過率をもとに、以下の基準に従い判定した。なお、測定に際しては、実施例等の熱可塑性樹脂組成物を1.0mm厚の樹脂シートに加工し、表面に導電層を形成した検体を評価に用いた。比較例1については、導電層を形成していない検体を用いた。
モデル本体部から突き出た模擬血管を電気メス(エルベ社製 高周波手術装置:VIO100C、条件:バイポーラー、凝固モード、60W)でつまみ把持した状態で、模擬血液を溶出するか否かを目視で評価した。溶出しない場合を優、溶出する場合を不可とした。
模擬血液が溶出している模擬血管に対して電気メス(条件:バイポーラー、凝固モード、60W)を用いて止血操作を行い、止血操作における模擬血液の溶出度合いを目視で評価した。止血操作による止血が再現されている場合は優、再現されていない場合は不可とした。
