近年、老化および老化関連疾患は、ニコチンアミドアデニンジヌクレオチド(nicotinamide adenine dinucleotide:NAD+)量低下及びNAD+依存性脱アセチル化酵素サーチュインの活性低下と密接な関わりを持つことが示されている(非特許文献1、2)。また、サーチュインの活性化は、カロリー制限における寿命延長又は健康増進に関わる効果の多くを説明すると考えられている(非特許文献2)。
NAD+は、古くから酸化還元反応の補酵素として知られている一方、近年では、ポリADPリボースポリメラーゼ、CD38/CD157、サーチュインの基質等としての役割も知られるようになった(非特許文献1)。特に、サーチュインによるNAD+からニコチンアミドへの分解反応は、それと共役するサーチュインによるリシン残基脱アセチル化反応を促進することで、健康、長寿に関わるさまざまな生命現象に関与している(非特許文献1、2)。老化に伴いNAD+量およびサーチュイン活性は低下する一方で、ニコチンアミドからNAD+を再合成する律速酵素ニコチンアミドホスホリボシルトランスフェラーゼ(NAMPT)の酵素反応産物、具体的には、ニコチンアミドモノヌクレオチド(nicotinamide mononucleotide:NMN)及びニコチンアミドリボシド(nicotinamide riboside:NR)といったNAD+中間代謝産物の補充は、サーチュインを効果的に再活性化する(非特許文献1)ことが知られており、更に、サーチュインの活性化を介して幅広い抗酸化作用を発現することが知られている。このため、ニコチンアミドモノヌクレオチドは、カロリー制限における寿命延長及び健康増進等に関わる多くの生体現象に寄与することが知られている。
従って、ニコチンアミドモノヌクレオチドを効率的に生産する方法が望まれている。これまで、トルラ酵母等の食経験のある酵母からニコチンアミドモノヌクレオチドが取得できること(特許文献1)、及び、大腸菌(E.coli)、B.サブチリス(B.subtilis)、C.グルタミカム(C.glutamicum)、A.バイリイ(A.baylyi)、及びR.オイトロファ(R.eutropha)からなる群から選択される細菌の遺伝修飾細菌からニコチンアミドリボシドを取得できること(特許文献2)が報告されている。
また、ニコチンアミドモノヌクレオチドの製造方法として、β―ニコチンアミドアデニンジヌクレオチドを基質として、Aspergillus 属に属する微生物の代謝組成物を用いて反応させる工程含むβ―ニコチンアミドモノヌクレオチドの製造方法(特許文献3)が知られている。より具体的には、培養した酵母中に含まれるβ―ニコチンアミドアデニンジヌクレオチドを酵素反応により、ニコチンアミドモノヌクレオチドを生成させている。特許の方法では、反応に酵素を用いることを必須としており、生成したニコチンアミドモノヌクレオチドを食用とする際に、安全性の観点も懸念が残る。また、ニコチンアミドモノヌクレオチドの生産量も十分なものとは言えない。

ニコチンアミドモノヌクレオチドを産生するこれまでの微生物学的方法では、依然として生産効率に改善の余地がある。
そこで、本発明は、ニコチンアミドモノヌクレオチドの産生効率の高い製造方法を提供することを目的とする。
本発明者は鋭意検討の結果、ニコチンアミドモノヌクレオチド生産能を有する微生物をpH4.0~10.0の反応液(例えば、緩衝液でpHを調整した反応液等)中で反応させることで、ニコチンアミドモノヌクレオチドの産生量が増加することを見出した。本発明は、この知見に基づいてさらに検討を重ねることにより完成したものである。

本発明によれば、ニコチンアミドモノヌクレオチド生産能を有する微生物をpH4~10の反応液中で反応させることで、非常に温和な条件下、常温常圧下であっても、容易にかつ効率よく目的とするニコチンアミドモノヌクレオチドを高濃度で得ることができる。
以下、本発明を詳細に説明する。
本発明はニコチンアミドモノヌクレオチド生産能を有する微生物をpH4~10の反応液(例えば、緩衝液でpHを調整した反応液等)中で反応させるニコチンアミドモノヌクレオチド富化反応工程を含む、ニコチンアミドモノヌクレオチドの製造方法である。

ニコチンアミドモノヌクレオチド生産能を有する微生物
本発明に用いるニコチンアミドモノヌクレオチド生産能を有する微生物としては、特に限定されないが、酵母、乳酸菌等を例示することができる。
本発明に用いるニコチンアミドモノヌクレオチド生産能を有する微生物としては、特に限定されないが、酵母、乳酸菌等を例示することができる。
酵母として食用酵母が使用できる。例えばSaccharomyces属に属する酵母、Kluyveromyces属、Candida属、Pichia属等が挙げられ、中でも、Candida属のCandida utilisが好ましい。より具体的には、Candida utilis IAM4264、Candida utilis ATCC9950、Candida utilis ATCC9550、Candida utilis IAM4233、Candida utilisAHU3259等を例示することができる。
乳酸菌としては、例えば、ラクトバチルス(Lactobacillus)属、ロイコノストック(Leuconostoc)属、ストレプトコッカス(Streptococcus)属、ペディオコッカス(Pediococcus)属、メリソコッカス(Melissococcus)属、エンテロコッカス(Enterococcus)属、トリココッカス(Trichococcus)属、ラクトコッカス(Lactococcus)属、カルノバクテリウム(Carnobacterium)属、バゴコッカス(Vagococcus)属、テトラゲノコッカス(Tetragenococcus)属、アトポビウム(Atopobium)属、ワイセラ(Weissella)属、オエノコッカス(Oenococcus)属、アビオトロフィア(Abiotrophia)属、デスメジア(Desemzia)属、パララクトバチルス(Paralactobacillus)属、グラヌリカテラ(Granulicatella)属、アルカリバクテリウム(Alkalibacterium)属、オルセネラ(Olsenella)属、イソバキュラム(Isobaculum)属、マリニラクチバチルス(Marinilactibacillus)属、アトポスタイペス(Atopostipes)属、ラクトバム(Lactovum)属、ピリバクター(Pilibacter)属、フルクトバチルス(Fructobacillus)属、ラクチシゲミウム(Lacticigemium)属、ババリコッカス(Bavariicoccus)属、ビフィドバクテリウム(Bifidobacterium)属等を例示することができ、フルクトバシラス(Fructobacillus)属が好ましい。
中でも、フルクトバシラス(Fructobacillus)属としては、フルクトバシラス・ドゥリオニス(Fructobacillus durionis)、フルクトバシラス・トロパエオイル(Fructobacillus tropaeoil)、及びフルクトバシラス・フルクトサス(Fructobacillus fructosus)が挙げられ、より好ましくは、フルクトバシラス・トロパエオイル(Fructobacillus tropaeoil)、及びフルクトバシラス・フルクトサス(Fructobacillus fructosus)が挙げられる。
より好ましい乳酸菌の例としては、Fructobacillus durionis RD011727株(寄託番号NITE BP-02764)、Fructobacillus tropaeoil RD012353株(寄託番号NITE BP-02765)、Fructobacillus tropaeoil RD012354株(寄託番号NITE BP-02766)、Fructobacillus fructosus NBRC3516株、及びFructobacillus durionis NBRC113239株が挙げられ、更に好ましくは、Fructobacillus durionis RD011727株(寄託番号NITE BP-02764)、Fructobacillus tropaeoil RD012353株(寄託番号NITE BP-02765)、Fructobacillus tropaeoil RD012354株(寄託番号NITE BP-02766)、及びFructobacillus fructosus NBRC3516株が挙げられる。
本発明においては、上記のニコチンアミドモノヌクレオチド生産能を有する微生物から1種を単独で、又は複数種を組み合わせて用いることができる。
本発明に用いるニコチンアミドモノヌクレオチド生産能を有する微生物は、別途培養工程で培養した微生物の菌体を用いることができる。
微生物を培養する際の培地には、炭素源、窒素源、ミネラルを含むものであれば、特に制限なく用いることができる。
炭素源としては、糖質及び糖質材料が挙げられる。糖質としては、糖類(単糖、二糖、オリゴ糖)、多糖類、及び糖アルコールが挙げられる。糖質としては、乳糖、ショ糖、グルコース、デンプン、キシリトール、デキストロース等が挙げられる。糖質材料は、糖質を含む有機組成物であればよく、例えば、乳及びその加工品(脱脂粉乳、ホエイ、ミルクパウダー、練乳等)、豆乳及びその加工品(豆乳加水分解物等)、穀類、果実、野菜等の食品が挙げられる。乳としては、ウシ、ヤギ、ヒツジ、水牛、ラクダ、ラマ、ロバ、ヤク、ウマ、トナカイ等の任意の哺乳動物に由来するものが挙げられる。なお糖質は、単離されたものであってもよいし、糖質材料に含まれているものであってもよい。例えば、フルクトース(糖質)は、果実(糖質材料)に含まれる形態のものを用いてもよい。これらの炭素源は、1種を単独で用いてもよいし、複数種を組み合わせて用いてもよい。
培地中の炭素源の濃度については特に限定されず、培地の種類や培養方式等に応じて適宜設定すればよいが、例えば0.5~15w/w%、好ましくは1~10w/w%、より好ましくは1.5~8.5w/w%が挙げられる。
窒素源としては、任意の無機窒素源又は有機窒素源を使用することができる。例えば、酵母エキス(ビール酵母等)、肉エキス、カゼイン等のタンパク質;ペプトン(プロテアーゼペプトン等)等のタンパク質加水分解物、ペプチド等のペプチド類;アンモニウム塩(クエン酸アンモニウム等)、硝酸塩等の含窒素塩等が挙げられる。これらの窒素源は、1種を単独で用いてもよいし、複数種を組み合わせて用いてもよい。
培地中の窒素源の濃度については特に限定されず、培地の種類や培養方式等に応じて適宜設定すればよいが、タンパク質の場合、例えば0.3~4w/w%、好ましくは0.5~3w/w%、より好ましくは1~2w/w%が挙げられ;ペプチド類の場合、例えば0.1~2w/w%、好ましくは0.3~1.8w/w%、より好ましくは0.5~1.5w/w%が挙げられ;含窒素塩の場合、例えば0.03~1.5w/w%、好ましくは0.05~1w/w%、より好ましくは0.1~0.5w/w%が挙げられる。
ミネラルとしては、例えば、マンガン(硫酸マンガン等)、亜鉛、鉄、ナトリウム(酢酸ナトリウム等)、カリウム(硫酸水素二カリウム、リン酸カリウム等)、マグネシウム(硫酸マグネシウム等)、カルシウム、リン(リン酸カリウム等)、硫黄(硫酸マンガン、硫酸水素カリウム、硫酸マグネシウム等)、微量元素等が挙げられる。これらのミネラルは、1種を単独で用いてもよいし、複数種を組み合わせて用いてもよい。これらのミネラルの中でも、好ましくは、マンガン、ナトリウム、マグネシウム、カリウムが挙げられる。
培地中のミネラルの濃度については特に限定されず、培地の種類や培養方式等に応じて適宜設定すればよいが、マンガンの場合、例えば0.001~0.01w/w%、好ましくは0.003~0.008w/w%が挙げられ;ナトリウムの場合、例えば0.05~1.5w/w%、好ましくは0.1~1w/w%が挙げられ;マグネシウムの場合、例えば0.001~0.02w/w%、好ましくは0.005~0.015w/w%が挙げられ;カリウムの場合、例えば0.05~1w/w%、好ましくは0.1~0.5w/w%が挙げられる。
培地は、上記成分以外に、ビタミン(ビタミンB群等)、界面活性剤(非イオン性界面活剤(Tween等)、陰イオン性界面活性剤(SDS等)、抗菌剤(トリクロサン等)、抗生物質(モネシン等)等の他の成分を含んでもよい。これらの他の成分は、1種を単独で用いてもよいし、複数種を組み合わせて用いてもよい。これらの他の成分の中でも、好ましくは界面活性剤、より好ましくは非イオン性界面活性剤が挙げられる。
培地中の他の成分の濃度については特に限定されず、他の成分の種類、培地の種類、培養方式等に応じて適宜設定すればよいが、界面活性剤を含む場合、界面活性剤の濃度として、例えば0.01~0.5w/w%、好ましくは0.05~0.3w/w%が挙げられる。
培養温度やpH等の培養条件は、特に制限なく適用でき、使用する微生物に合わせて設定すれば良い。一般的には、培養温度は21~37℃、好ましくは25~34℃が良く、pHは3.0~8.0、特に3.5~7.0が好ましい。
培養形式としては、バッチ培養、流加培養、あるいは連続培養のいずれでも良いが、工業的には後者が望ましい。培養時の撹拌、通気等の条件は特に制限なく、一般的な方法でよい。
培養によって得られた微生物は、そのまま、あるいはろ紙を用いたろ過、遠心分離、デカンテーション、スクリュープレス、ローラープレス、ロータリードラムスクリーン、ベルトスクリーン、振動スクリーン、多重板振動フィルター、真空脱水、加圧脱水、ベルトプレス、遠心濃縮脱水、多重円板脱水等によって固液分離を行い、培養液から菌体を回収し、又はさらに回収した菌体を洗浄したものをニコチンアミドモノヌクレオチド富化反応に用いることができる。好ましくは、培養液から回収した菌体、より好ましくは培養液から回収し且つ洗浄した菌体を、ニコチンアミドモノヌクレオチド富化反応に用いることができる。
培養工程で得られたニコチンアミドモノヌクレオチド生産能を有する微生物をニコチンアミドモノヌクレオチド富化反応工程に用いることで、微生物中のニコチンアミドモノヌクレオチド含有量を向上させることができる。
ニコチンアミドモノヌクレオチド富化反応工程では、具体的には、ニコチンアミドモノヌクレオチド生産能を有する微生物と、適当な反応用液体とを含み、且つpH4~10に調整された反応液を調整し、当該反応液を適当な温度で維持することで、ニコチンアミドモノヌクレオチドを富化する反応を進行させる。なお、本発明において、反応液のpHは、ニコチンアミドモノヌクレオチド富化反応時に供される温度でのpHを意味する。
ニコチンアミドモノヌクレオチド富化反応工程における反応液のpHとしては、好ましくは4.8~9.2、より好ましくは5.8~9.2、さらに好ましくは6.8~9.2、6.8~8.2、又は7.8~9.2が挙げられる。
また、ニコチンアミドモノヌクレオチド生産能を有する微生物としてFructobacillus tropaeoil RD012353株(寄託番号NITE BP-02765)を用いる場合、ニコチンアミドモノヌクレオチド富化反応工程における反応液のpHとしては、好ましくは4.8~9.2、より好ましくは5.8~8.2、さらに好ましくは6.8~8.2、一層好ましくは7.8~8.2が挙げられる。
ニコチンアミドモノヌクレオチド生産能を有する微生物としてFructobacillus durionis RD011727株(寄託番号NITE BP-02764)を用いる場合、ニコチンアミドモノヌクレオチド富化反応工程における反応液のpHとしては、好ましくは5.8~8.2、より好ましくは5.8~8.2、さらに好ましくは6.8~8.2が挙げられる。
ニコチンアミドモノヌクレオチド生産能を有する微生物としてFructobacillus fructosus NBRC3516株)を用いる場合、ニコチンアミドモノヌクレオチド富化反応工程における反応液のpHとしては、好ましくは4.8~9.2、より好ましくは5.8~9.2、さらに好ましくは6.8~9.2、一層好ましくは7.8~9.2、より一層好ましくは7.8~8.2が挙げられる。
上記反応用液体としては、反応液のpHを上記範囲内に調整できる限りにおいて特に制限なく用いることができる。例えば、培養によって得られた微生物をそのままニコチンアミドモノヌクレオチド富化反応に用いる場合は、上記反応用液体としては、ニコチンアミドモノヌクレオチド生産能を有する微生物の培養に用いられた培地が挙げられる。培養によって得られた微生物を培養液から回収し、より好ましくはさらに洗浄した後にニコチンアミドモノヌクレオチド富化反応に用いる場合は、上記反応用液体としては、水(反応中にpH調整剤を用い、至適なpH範囲に調整することを含む)、緩衝液、有機溶剤、及びこれらの2種以上の混合液等が挙げられる。
緩衝液としては、例えば、酢酸緩衝液、リン酸緩衝液、ホウ酸緩衝液、炭酸緩衝液、クエン酸緩衝液、トリス緩衝液、HEPES衝液、MES緩衝液を例示することができ、これらの緩衝液から1種を単独で、又は複数種を組み合わせて用いることができる。
より具体的には、KHC8H4O4-NaOH (pH4.0)、CH3COOH-CH3COONa(pH4.0)、MES-NaOH (pH5.0)、CH3COOH-CH3COONa(pH5.0)、KH2PO4-K2HPO4(pH6.0)、MES-NaOH(pH6.0)、 KH2PO4-K2HPO4(pH7.0)、PIPES-NaOH (pH7.0) 、HEPES-NaOH(pH8.0)、H3BO4-NaOH(pH8.0)、CHES-NaOH(pH9.0)、H3BO4-NaOH(pH9.0)、H2CO3-NaHCO3(pH10.0)、CHES-NaOH(pH10.0)を例示することができる。中でも、CH3COOH-CH3COONa、KH2PO4-K2HPO4、H3BO4-NaOHが好ましく、KH2PO4-K2HPO4がより好ましい。
有機溶剤としては、ベンゼン、ベンゾニトリル等の芳香族化合物、アセトン、アセチルアセトン、メチルエチルケトン等のケトン類、酢酸エチル、酢酸ブチル、酪酸エチル、蟻酸エチル等の脂肪酸エステル類、ジエチルエーテル、ジプロピルエーテル、ジブチルエーテル、テトラヒドロフラン、1,4-ジオキサン等のエーテル類、ジクロロメタン、クロロホルム、ジクロロエタン等のハロゲン化炭化水素類、1,2-プロパンジオール、1,2-ブタンジオール、1,3-ブタンジオール、1,4-ブタンジオール、2,3-ブタンジオール、1,2-ヘキサンジオール、1,6-ヘキサンジオール、1,2-ペンタンジオール、1,5-ペンタンジオール、2-メチル-2,4-ペンタンジオール、3-メチル-1,5-ペンタンジオール等のジオール類、炭素数1~7の直鎖又は分岐アルキルを有するアルコール、シクロヘキサノール、3-メトキシ-3-メチル-1-ブタノール、3-メトキシ-1-ブタノール等のアルコール類を例示することができ、これらの有機溶剤から1種を単独で、又は複数種を組み合わせて用いることができる。
pH調整剤としては、反応中に至適pHに調整することのできるものを適宜選択すればよいが、例えば、塩酸等の無機酸、クエン酸等の有機酸、水酸化ナトリウム、水酸化カリウム等の水酸化物等の無機塩基、有機アミン等の有機塩基が挙げられ、これらの化合物から1種を単独で、又は複数種を組み合わせて用いることができる。
本発明のニコチンアミドモノヌクレオチド富化反応工程における反応温度は、例えば10~55℃、好ましくは15~45℃、より好ましくは20~37℃、さらに好ましくは20~35℃である。反応時間は、例えば0.1~48時間、好ましくは1~24時間、より好ましくは3~20時間である。
ニコチンアミドモノヌクレオチド富化反応は、ニコチンアミドモノヌクレオチド生産能を有する微生物を上記の反応用液体に懸濁し、静置、攪拌または振盪することで行うことができる。
ニコチンアミドモノヌクレオチド富化反応によって得られたニコチンアミドモノヌクレオチドを含む微生物は、そのまま凍結乾燥、棚乾燥、スプレードライ等により乾燥することで、菌体粉末とすることで、食品への添加物若しくは化粧品又は医薬品の有効成分として配合することができる。
また、ニコチンアミドモノヌクレオチド富化反応によって得られたニコチンアミドモノヌクレオチドを含む微生物は、遠心分離法、膜ろ過法等によって、緩衝液と固液分離して回収することもできる。回収された菌体は、そのまま棚乾燥や凍結乾燥等により乾燥することで、菌体粉末とすることで、食品への添加物若しくは化粧品又は医薬品の有効成分として配合することができる。
以下、本発明を実施例により具体的に説明するが、本発明はこれら実施例によって限定されるものではない。
試験例1
[1-1]Fructobacillus tropaeoil RD012353株の培養
ニコチンアミドモノヌクレオチド生産能を有するフルクトバシラス属乳酸菌であるFructobacillus tropaeoil RD012353株(寄託番号NITE BP-02765)を、Difco社製のMRS培地(前培養培地)3mlに植菌し、30℃で24時間、静置で拡大培養した。得られた培養液をOD660が0.02となるように、MRS培地(本培養培地)100mlに植菌し、30℃で12時間、振とう培養した。
得られた培養液を遠心分離に供して菌体を回収した。回収した菌体を100mlの0.85w/w%KCl水溶液で洗浄した。洗浄した菌体を再度遠心分離に供し、菌体を回収した。
[1-1]Fructobacillus tropaeoil RD012353株の培養
ニコチンアミドモノヌクレオチド生産能を有するフルクトバシラス属乳酸菌であるFructobacillus tropaeoil RD012353株(寄託番号NITE BP-02765)を、Difco社製のMRS培地(前培養培地)3mlに植菌し、30℃で24時間、静置で拡大培養した。得られた培養液をOD660が0.02となるように、MRS培地(本培養培地)100mlに植菌し、30℃で12時間、振とう培養した。
得られた培養液を遠心分離に供して菌体を回収した。回収した菌体を100mlの0.85w/w%KCl水溶液で洗浄した。洗浄した菌体を再度遠心分離に供し、菌体を回収した。
[1-2]ニコチンアミドモノヌクレオチド富化反応工程(NMN富化反応)
上記[1-1]で回収した菌体をそれぞれ20mlの下記反応用液体中に懸濁して反応液を調製し、25℃で8時間静置するNMN富化反応に付した。NMN富化反応中の反応液のpHは、表1に示す通りである。

反応終了後、遠心分離にて菌体を回収した。回収した菌体を100mlの0.85w/w%KCl水溶液で洗浄した。洗浄した菌体を再度遠心分離に供し、菌体を回収した。
[1-3]ニコチンアミドモノヌクレオチド(NMN)量測定
上記[1-1]で回収した菌体と、上記[1-2]で回収した菌体を20mlのイオン交換水に懸濁し、ガラスビーズを等量添加した後、ビーズ破砕機にて菌体を破砕した。破砕した菌体を遠心分離にて分離し、上清(菌体破砕抽出液)を回収した。
回収した上清を下記分析条件のHPLC分析で分析し、回収菌体中のニコチンアミドモノヌクレオチド(NMN)生産量を測定した。上記[1-1]で回収した菌体(NMN富化反応を行っていない)におけるNMN生産量を100とした場合の、上記[1-2]で回収した菌体におけるNMN生産量の相対値を導出した。結果を表1に示す。
上記[1-1]で回収した菌体と、上記[1-2]で回収した菌体を20mlのイオン交換水に懸濁し、ガラスビーズを等量添加した後、ビーズ破砕機にて菌体を破砕した。破砕した菌体を遠心分離にて分離し、上清(菌体破砕抽出液)を回収した。
回収した上清を下記分析条件のHPLC分析で分析し、回収菌体中のニコチンアミドモノヌクレオチド(NMN)生産量を測定した。上記[1-1]で回収した菌体(NMN富化反応を行っていない)におけるNMN生産量を100とした場合の、上記[1-2]で回収した菌体におけるNMN生産量の相対値を導出した。結果を表1に示す。

表1に示すように、培養終了後の菌体をpH4からpH10の溶液中で反応することにより、NMNの生産性を著しく向上させることができる。

ニコチンアミドモノヌクレオチド生産能を有するフルクトバシラス属乳酸菌であるFructobacillus durionis RD011727株(寄託番号NITEBP-02764)を、Difco社製のMRS培地(前培養培地)3mlに植菌し、30℃で24時間、静置で拡大培養した。得られた培養液をOD660が0.02となるように、MRS培地(本培養培地)100mlに植菌し、30℃で12時間、振とう培養した。
得られた培養液を遠心分離に供して菌体を回収した。回収した菌体を100mlの0.85w/w%KCl水溶液で洗浄した。洗浄した菌体を再度遠心分離に供し、菌体を回収した。
[2-1]Fructobacillus durionis RD011727株の培養
ニコチンアミドモノヌクレオチド生産能を有するフルクトバシラス属乳酸菌であるFructobacillus durionis RD011727株(寄託番号NITEBP-02764)を、Difco社製のMRS培地(前培養培地)3mlに植菌し、30℃で24時間、静置で拡大培養した。得られた培養液をOD660が0.02となるように、MRS培地(本培養培地)100mlに植菌し、30℃で12時間、振とう培養した。

[2-2]ニコチンアミドモノヌクレオチド富化反応工程(NMN富化反応)
上記[2-1]で回収した菌体と、試験例1の実施例1~7で用いた反応用液体とを用いたことを除き、試験例1の[1-2]と同様にしてNMN富化反応を行った。NMN富化反応中の反応液のpHは、反応用液体と同程度であり、表2に示す通りである。

[2-3]ニコチンアミドモノヌクレオチド(NMN)量測定
上記[2-1]で回収した菌体と、上記[2-2]で回収した菌体を用いたことを除いて、試験例1の[1-3]と同様にしてNMN量を測定した。上記[2-1]で回収した菌体(NMN富化反応を行っていない)におけるNMN生産量を100とした場合の、上記[2-2]で回収した菌体におけるNMN生産量の相対値を導出した。結果を表2に示す。
上記[2-1]で回収した菌体と、上記[2-2]で回収した菌体を用いたことを除いて、試験例1の[1-3]と同様にしてNMN量を測定した。上記[2-1]で回収した菌体(NMN富化反応を行っていない)におけるNMN生産量を100とした場合の、上記[2-2]で回収した菌体におけるNMN生産量の相対値を導出した。結果を表2に示す。
表2に示すように、培養終了後の菌体をpH4からpH10の溶液中で反応することにより、NMNの生産性を著しく向上させることができる。
試験例3
[3-1]Fructobacillus fructosus NBRC3516株の培養
ニコチンアミドモノヌクレオチド生産能を有するフルクトバシラス属乳酸菌であるFructobacillus fructosus NBRC3516株を、Difco社製のMRS培地(前培養培地)3mlに植菌し、30℃で24時間、静置で拡大培養した。得られた培養液をOD660が0.02となるように、MRS培地(本培養培地)100mlに植菌し、30℃で12時間、振とう培養した。
得られた培養液を遠心分離に供して菌体を回収した。
[3-1]Fructobacillus fructosus NBRC3516株の培養
ニコチンアミドモノヌクレオチド生産能を有するフルクトバシラス属乳酸菌であるFructobacillus fructosus NBRC3516株を、Difco社製のMRS培地(前培養培地)3mlに植菌し、30℃で24時間、静置で拡大培養した。得られた培養液をOD660が0.02となるように、MRS培地(本培養培地)100mlに植菌し、30℃で12時間、振とう培養した。
得られた培養液を遠心分離に供して菌体を回収した。
[3-2]ニコチンアミドモノヌクレオチド富化反応工程(NMN富化反応)
上記[3-1]で回収した菌体と、試験例1の実施例1~6で用いた反応用液体とを用いたことを除き、試験例1の[1-2]と同様にしてNMN富化反応を行った。NMN富化反応中の反応液のpHは、反応用液体と同程度であり、表3に示す通りである。

[3-3]ニコチンアミドモノヌクレオチド(NMN)量測定
上記[3-1]で回収した菌体と、上記[3-2]で回収した菌体を用いたことを除いて、試験例1の[1-3]と同様にしてNMN量を測定した。上記[3-1]で回収した菌体(NMN富化反応を行っていない)におけるNMN生産量を100とした場合の、上記[3-2]で回収した菌体におけるNMN生産量の相対値を導出した。結果を表3に示す。

表3に示すように、培養終了後の菌体をpH4からpH10の溶液中で反応することにより、NMNの生産性を著しく向上させることができる。
試験例4
NMN富化反応に用いた反応用液体を、下記に記載した緩衝液に変更した(NMN富化反応中の反応液のpHは、反応用液体と同程度であり、表4に示す通りである。)以外は、試験例1~3と同様の操作を行い、NMN生産量の相対値を導出した。結果を表4に示す。

・実施例21,24,27:0.1M MES緩衝液(pH5.0)

・実施例23,26,29:0.1M Tris緩衝液(pH9.0)
・実施例21,24,27:0.1M MES緩衝液(pH5.0)
・実施例22,25,28:0.1M HEPES緩衝液(pH7.0)
・実施例23,26,29:0.1M Tris緩衝液(pH9.0)
表4に示すように、培養終了後の菌体をpH4からpH10の溶液中で反応することにより、緩衝液の種類に因らずNMNの生産性を著しく向上させることができる。
試験例5
NMN富化反応に用いた反応用液体を、イオン交換水を用い、24重量%NaOH水溶液を用いて、反応中のpHを7に調整しながら反応を行った(NMN富化反応中の反応液のpHは、表5に示す通りである。)以外は、試験例3と同様の操作を行い、NMN生産量の相対値を導出した。結果を表5に示す。

Claims (5)
- ニコチンアミドモノヌクレオチド生産能を有する微生物を、pH4~10の反応液中で反応させるニコチンアミドモノヌクレオチド富化反応工程を含む、ニコチンアミドモノヌクレオチドの製造方法。
- ニコチンアミドモノヌクレオチド生産能を有する微生物が、乳酸菌である請求項1に記載の製造方法。
- pH4~10の反応液が、酢酸緩衝液、リン酸緩衝液、ホウ酸緩衝液、炭酸緩衝液、トリス緩衝液、HEPES緩衝液、MES緩衝液からなる群より選択される緩衝液を含む、請求項1または2に記載の製造方法。
- 乳酸菌が、フルクトバシラス(Fructobacillus)属である請求項1~3のいずれかに記載の製造方法。
- 乳酸菌が、Fructobacillus durionis RD011727株(寄託番号NITE BP-02764)、Fructobacillus tropaeoil RD012353株(寄託番号NITE BP-02765)、Fructobacillus tropaeoil RD012354株(寄託番号NITE BP-02766)、及びFructobacillus fructosus NBRC3516株からなる群より選ばれる、請求項1~4のいずれかに記載の製造方法。
