ニコチンアミドモノヌクレオチド(Nicotinamide MonoNucleotide、NMN)は、ニコチンアミドアデニンジヌクレオチド(NAD)の前駆体であって、近年特に注目されている物質である。
例えば、老化の制御には長寿遺伝子と呼ばれるサーチュインの活性維持が重要とされるが、年を重ねること(加齢)に伴ってその活性に必要なNADの組織中濃度が低下することが老化の一因とされるため、NADの前駆体であるNMNの投与又は摂取は、抗老化効果をもたらすといわれている。

ヒトを含む動物の腸内(特に大腸内)には数多くの細菌が生息し、その集合体は腸内細菌叢と呼ばれている。腸内細菌叢は、消化補助、免疫反応調節、抗炎症等の多様な役割を担っていると考えられており、また、腸内細菌叢が産生する代謝物を宿主が吸収することで、宿主に様々な健康効果をもたらすことも知られている。本発明者らは、上記従来技術の有する課題に鑑みて、腸内細菌叢がNMNを産生する場合には、それを宿主が吸収することで、宿主にNMNを投与又は摂取せしめた場合と同等の効果をもたらすことができると考えた。本発明は、このような課題及び知見に鑑みてなされたものであり、腸内細菌叢によるニコチンアミドモノヌクレオチド(NMN)産生向上作用を有する組成物を提供することを目的とする。
本発明者らは、上記目的を達成すべく鋭意研究を重ね、先ず、宿主の個体差による違いはあるものの、腸内細菌叢がNMNを産生することを見出した。そこで、本発明者らは、従来より知られている様々なプレバイオティクス(すなわち、消化管上部で分解・吸収されず、大腸において腸内細菌の選択的な栄養源となり、腸内細菌叢の構成を健康的なバランスに改善し維持して宿主の健康の増進維持に役立つ食品成分(Gibson et al.,J Nutr.,1995,125,p.1401-1412等))の中から、腸内細菌叢によるNMN産生量を向上させることが可能なプレバイオティクスを探索した。その結果、特定の炭水化物、すなわち、単糖(ただし、グルコース及びキシロースを除く)、二糖、三糖、四糖、五糖、糖アルコール、及び食物繊維からなる群から選択される少なくとも1種の炭水化物には、in vitroにおいてもin vivoにおいても、腸内細菌叢によるNMN産生量を高める作用があることを新たに見出した。よって、前記炭水化物を有効成分とする組成物を宿主に投与又は摂取せしめることにより、その腸内細菌叢によるNMN産生量を向上できることが明らかとなった。また、これによって宿主に大腸を経由してNMNをより多く摂取せしめることができるため、当該NMNに加えて、NMNを前駆体とするNADやその代謝物の生体内濃度を高めることができることも明らかとなった。さらに、NMNの摂取によって加齢による諸疾患を改善できること(非特許文献1~4)、便秘を改善できること(非特許文献5)、及びNMNの代謝物であるNADが炎症性腸疾患の改善に有効であること(Navarro et al.,Br J Pharmacol.,2022,179,p.1839-1856)から、前記炭水化物を有効成分とする組成物を投与又は摂取せしめることにより、加齢による疾患の予防又は改善や、便秘及び炎症性腸疾患の予防又は改善も可能となることが明らかとなり、本発明を完成するに至った。かかる知見により得られた本発明の態様は次のとおりである。

本発明によれば、腸内細菌叢によるニコチンアミドモノヌクレオチド(NMN)産生向上作用を有する組成物を提供することが可能となる。また、本発明の組成物は、飲食組成物としても提供することができるため、摂取しやすく、摂取を習慣化しやすい。さらに、本発明の組成物は、腸内細菌叢によるNMN産生量を高めることで、生体内におけるNAD濃度の向上や、加齢による疾患の予防又は改善、並びに、便秘や炎症性腸疾患の予防又は改善のために用いることができる。
以下、本発明をその好適な実施形態に即して詳細に説明する。
<NMN産生向上用組成物>
本発明は、単糖(ただし、グルコース及びキシロースを除く)、二糖、三糖、四糖、五糖、糖アルコール、及び食物繊維からなる群から選択される少なくとも1種の炭水化物を有効成分として含有する、ニコチンアミドモノヌクレオチド(NMN)産生向上のための組成物(本明細書中、場合により「NMN産生向上用組成物」又は単に「本発明の組成物」という)を提供する。
本発明は、単糖(ただし、グルコース及びキシロースを除く)、二糖、三糖、四糖、五糖、糖アルコール、及び食物繊維からなる群から選択される少なくとも1種の炭水化物を有効成分として含有する、ニコチンアミドモノヌクレオチド(NMN)産生向上のための組成物(本明細書中、場合により「NMN産生向上用組成物」又は単に「本発明の組成物」という)を提供する。
本発明において、ニコチンアミドモノヌクレオチドとは、β-ニコチンアミドモノヌクレオチドのことを示し、NMNとも略す。β-ニコチンアミドモノヌクレオチドは、ニコチンアミド及びニコチンアミドリボシドが生体内でニコチンアミドアデニンジヌクレオチド(NADH、NAD+、本明細書中、場合により「NAD」と略す)に変換される際の中間代謝物である。ニコチンアミドアデニンジヌクレオチドは、酸化還元補酵素として生体内の脱水素反応で中心的な役割を果たすことが知られている。本発明者らは、腸内細菌叢によってニコチンアミドモノヌクレオチドが産生されること、及び、本発明に係る炭水化物が、かかるニコチンアミドモノヌクレオチドの産生量を高め、優れたニコチンアミドモノヌクレオチド産生向上作用を有することを見出した。
本発明において、腸内細菌叢とは、動物の腸管(特に大腸)内に存在する細菌の集団を示し、腸内フローラ、腸管微生物叢ともいわれる。本発明に係る腸内細菌叢に含まれる微生物には、その大部分を成す腸内細菌に加えて、他の腸管微生物(ファージ、ウイルス、古細菌、真菌、寄生虫)も含む。例えば、ヒトの大腸内には、一般に、前記腸内細菌が、600~1000兆個、1000種類以上存在するといわれている。腸内細菌叢の構成は個体間で大きく異なり、宿主の加齢や健康状態によっても変化する。そのため、一概にいえるものではないが、例えば、ヒトや動物の糞便を、グルコース0.5%含有GAM培地において37℃、嫌気条件で12時間培養して得られる微生物群を、本発明に係る腸内細菌叢としてもよい。
本発明のNMN産生向上用組成物に含有される有効成分は、単糖(ただし、グルコース及びキシロースを除く)、二糖、三糖、四糖、五糖、糖アルコール、及び食物繊維からなる群から選択される少なくとも1種の炭水化物である。本発明に係る炭水化物としては、糖質類であることが好ましく、糖類であることがより好ましく、二糖、三糖、四糖、及び五糖からなる群から選択される少なくとも1種であることがさらに好ましく、三糖及び四糖からなる群から選択される少なくとも1種であることがさらにより好ましく、また、三糖、四糖の順に好ましい。また、本発明に係る炭水化物としては、フコースを構成糖として含まないものであることが好ましく、グルコース、ガラクトース、及びフルクトースからなる群から選択される1~3種を構成糖としてなるものであることがより好ましい。
(単糖)
本発明に係る単糖としては、直鎖状であっても環状であってもよく、アルデヒド基を含むアルドースであってもケトン基を含むケトースであってもよく、また、D体及びL体がいずれも存在する場合にはそのどちらであってもよい。本発明に係る単糖の炭素数としては、4~7であることが好ましく、6である(すなわち、単糖が六炭糖である)ことがより好ましい。
本発明に係る単糖としては、直鎖状であっても環状であってもよく、アルデヒド基を含むアルドースであってもケトン基を含むケトースであってもよく、また、D体及びL体がいずれも存在する場合にはそのどちらであってもよい。本発明に係る単糖の炭素数としては、4~7であることが好ましく、6である(すなわち、単糖が六炭糖である)ことがより好ましい。
ただし、本発明に係る単糖からは、グルコース及びキシロースを除く。グルコース及びキシロースは小腸で体内への吸収が100%完了してしまうために大腸まで到達せず(山田和彦ら,栄養学雑誌,2001年,Vol.59,No.4,p.169-176;慶應義塾大学病院,「D-キシローステスト」,[on line],2018年2月22日,[2023年8月3日検索],インターネット<https://kompas.hosp.keio.ac.jp/contents/000347.html>)、大腸に存在する腸内細菌のNMN産生向上には寄与しない。
本発明に係る単糖として、より具体的には、例えば、タガトース、アラビノース、アロース、プシコース、ガラクトース、フルクトース、マンノースが挙げられ、これらのうちの1種であっても2種以上の組み合わせであってもよい。中でも、本発明に係る単糖としては、タガトース、アラビノース、アロース、及びプシコースからなる群から選択される少なくとも1種が好ましく、生体内においてNMNを産生する腸内細菌により到達しやすい、すなわち、プレバイオティクスとして消化管上部で分解・吸収されず大腸までより到達しやすい観点から、タガトースであることが好ましい。
(二糖)
本発明に係る二糖とは、前記単糖2分子が脱水縮合により結合した糖を示す。また、D体及びL体がいずれも存在する場合にはそのどちらであってもよい。また、本発明に係る二糖には、二糖が修飾された修飾糖も含む。前記二糖の修飾としては、例えばシアル化が挙げられ、その修飾箇所は1箇所であっても複数個所であってもよく、複数個所である場合には1種の修飾であっても2種以上の修飾の組み合わせであってもよい。
本発明に係る二糖とは、前記単糖2分子が脱水縮合により結合した糖を示す。また、D体及びL体がいずれも存在する場合にはそのどちらであってもよい。また、本発明に係る二糖には、二糖が修飾された修飾糖も含む。前記二糖の修飾としては、例えばシアル化が挙げられ、その修飾箇所は1箇所であっても複数個所であってもよく、複数個所である場合には1種の修飾であっても2種以上の修飾の組み合わせであってもよい。
前記二糖を構成する単糖としては、互いに同じであっても異なっていてもよく、例えば、タガトース、グルコース、ガラクトース、フルクトース、マンノース、キシロース、アラビノース、アロース、プシコースが挙げられ、タガトース、グルコース、ガラクトース、フルクトース、マンノース、キシロースが好ましい。中でも、本発明に係る二糖としては、グルコース、ガラクトース、及びフルクトースからなる群から選択される1種又は2種を構成糖としてなる二糖であることが好ましい。
本発明に係る二糖として、より具体的には、例えば、セロビオース、ラクチュロース、トレハロース、ラクトース、イソマルツロースが挙げられ、これらのうちの1種であっても2種の組み合わせであってもよい。中でも、本発明に係る二糖としては、生体内においてNMNを産生する腸内細菌により到達しやすい、すなわち、プレバイオティクスとして消化管上部で分解・吸収されず大腸までより到達しやすい観点から、セロビオース、ラクチュロース、トレハロース、及びシアリルラクトース(3’-シアリルラクトース等)からなる群から選択される少なくとも1種であることが好ましく、前記修飾がなされていない二糖であることがより好ましく、セロビオース、ラクチュロース、及びトレハロースからなる群から選択される少なくとも1種であることがさらに好ましく、セロビオースが特に好ましい。
(三糖)
本発明に係る三糖とは、前記単糖3分子が脱水縮合により結合した糖を示す。また、D体及びL体がいずれも存在する場合にはそのどちらであってもよい。また、本発明に係る三糖には、三糖が修飾された修飾糖も含む。前記三糖の修飾としては、前記二糖の修飾と同じものが挙げられ、その修飾箇所は1箇所であっても複数個所であってもよく、複数個所である場合には1種の修飾であっても2種以上の修飾の組み合わせであってもよい。
本発明に係る三糖とは、前記単糖3分子が脱水縮合により結合した糖を示す。また、D体及びL体がいずれも存在する場合にはそのどちらであってもよい。また、本発明に係る三糖には、三糖が修飾された修飾糖も含む。前記三糖の修飾としては、前記二糖の修飾と同じものが挙げられ、その修飾箇所は1箇所であっても複数個所であってもよく、複数個所である場合には1種の修飾であっても2種以上の修飾の組み合わせであってもよい。
前記三糖を構成する単糖としては、全て同じであっても、一部が同じであっても、全て異なっていてもよく、例えば、タガトース、グルコース、ガラクトース、フルクトース、マンノース、キシロース、アラビノース、アロース、プシコース、フコースが挙げられ、タガトース、グルコース、ガラクトース、フルクトース、マンノース、キシロース、フコースが好ましい。中でも、本発明に係る三糖としては、グルコース、ガラクトース、フルクトース、及びフコースからなる群から選択される1種~3種を構成糖としてなる三糖であることが好ましく、グルコース、ガラクトース、及びフルクトースからなる群から選択される1種~3種を構成糖としてなる三糖であることがより好ましい。
本発明に係る三糖として、より具体的には、例えば、ケストース(1-ケストース等)、ラクトスクロース、ラフィノース、フコシルラクトース(2’-フコシルラクトース等)、ガラクトシルラクトース(4’-ガラクトシルラクトース等)が挙げられ、これらのうちの1種であっても2種以上の組み合わせであってもよい。中でも、本発明に係る三糖としては、生体内においてNMNを産生する腸内細菌により到達しやすい、すなわち、プレバイオティクスとして消化管上部で分解・吸収されず大腸までより到達しやすい観点から、ケストース、ラクトスクロース、ラフィノース、フコシルラクトース、及びガラクトシルラクトースからなる群から選択される少なくとも1種であることが好ましく、ケストース、ラクトスクロース、及びラフィノースからなる群から選択される少なくとも1種であることがさらに好ましい。
(四糖)

前記四糖を構成する単糖としては、全て同じであっても、一部が同じであっても、全て異なっていてもよく、例えば、タガトース、グルコース、ガラクトース、フルクトース、マンノース、キシロース、アラビノース、アロース、プシコースが挙げられ、タガトース、グルコース、ガラクトース、フルクトース、マンノース、キシロースが好ましい。中でも、本発明に係る四糖としては、グルコース、ガラクトース、及びフルクトースからなる群から選択される1種~3種を構成糖としてなる四糖であることが好ましい。
本発明に係る四糖としては、1種であっても2種以上の組み合わせであってもよい。中でも、本発明に係る四糖としては、生体内においてNMNを産生する腸内細菌により到達しやすい、すなわち、プレバイオティクスとして消化管上部で分解・吸収されず大腸までより到達しやすい観点から、スタキオース及びニストースからなる群から選択される少なくとも1種であることが好ましい。
(五糖)
本発明に係る五糖とは、前記単糖5分子が脱水縮合により結合した糖を示す。また、D体及びL体がいずれも存在する場合にはそのどちらであってもよい。また、本発明に係る五糖には、五糖が修飾された修飾糖も含む。前記五糖の修飾としては、前記二糖の修飾と同じものが挙げられ、その修飾箇所は1箇所であっても複数個所であってもよく、複数個所である場合には1種の修飾であっても2種以上の修飾の組み合わせであってもよい。
本発明に係る五糖とは、前記単糖5分子が脱水縮合により結合した糖を示す。また、D体及びL体がいずれも存在する場合にはそのどちらであってもよい。また、本発明に係る五糖には、五糖が修飾された修飾糖も含む。前記五糖の修飾としては、前記二糖の修飾と同じものが挙げられ、その修飾箇所は1箇所であっても複数個所であってもよく、複数個所である場合には1種の修飾であっても2種以上の修飾の組み合わせであってもよい。
前記五糖を構成する単糖としては、全て同じであっても、一部が同じであっても、全て異なっていてもよく、例えば、タガトース、グルコース、ガラクトース、フルクトース、マンノース、キシロース、アラビノース、アロース、プシコースが挙げられる。中でも、本発明に係る五糖としては、グルコース、ガラクトース、及びフルクトースからなる群から選択される1種~3種を構成糖としてなる五糖であることが好ましい。
本発明に係る五糖としては、1種であっても2種以上の組み合わせであってもよい。中でも、本発明に係る五糖としては、生体内においてNMNを産生する腸内細菌により到達しやすい、すなわち、プレバイオティクスとして消化管上部で分解・吸収されず大腸までより到達しやすい観点から、フラクトフラノシルニストース(1-フラクトフラノシル-D-ニストース等)が好ましい。
(糖アルコール)
本発明に係る糖アルコールとは、糖化合物のカルボニル基が還元された化合物である。前記糖化合物としては、前記単糖、二糖、三糖、四糖、及び五糖が挙げられる。
本発明に係る糖アルコールとは、糖化合物のカルボニル基が還元された化合物である。前記糖化合物としては、前記単糖、二糖、三糖、四糖、及び五糖が挙げられる。
本発明に係る糖アルコールとして、より具体的には、例えば、ラクチトール、ソルビトール、マンニトール、キシリトール、還元イソマルツロースが挙げられ、これらのうちの1種であっても2種以上の組み合わせであってもよい。中でも、本発明に係る糖アルコールとしては、前記単糖又は二糖が還元されてなる糖アルコールであることが好ましく、生体内においてNMNを産生する腸内細菌により到達しやすい、すなわち、プレバイオティクスとして消化管上部で分解・吸収されず大腸までより到達しやすい観点から、ラクチトール、ソルビトール、マンニトール、キシリトール、及び還元イソマルツロースからなる群から選択される少なくとも1種であることがより好ましく、ラクチトール及びソルビトールからなる群から選択される少なくとも1種であることがさらに好ましい。
(食物繊維)
本発明に係る食物繊維とは、ヒトの消化酵素(アミラーゼ、プロテアーゼ、マルターゼ等)に分解されない炭水化物であって、前記単糖6分子以上が脱水縮合により結合した糖を示す。前記食物繊維としては、直鎖状であっても分岐鎖状であってもよい。また、本発明に係る食物繊維の分子量としては、500~50000であることが好ましく、1000~30000であることがより好ましい。
本発明に係る食物繊維とは、ヒトの消化酵素(アミラーゼ、プロテアーゼ、マルターゼ等)に分解されない炭水化物であって、前記単糖6分子以上が脱水縮合により結合した糖を示す。前記食物繊維としては、直鎖状であっても分岐鎖状であってもよい。また、本発明に係る食物繊維の分子量としては、500~50000であることが好ましく、1000~30000であることがより好ましい。
前記食物繊維を構成する単糖としては、全て同じであっても、一部が同じであっても、全て異なっていてもよく、例えば、タガトース、グルコース、ガラクトース、フルクトース、マンノース、キシロース、アラビノース、アロース、プシコースが挙げられる。中でも、本発明に係る食物繊維としては、グルコース、ガラクトース、及びフルクトースからなる群から選択される1種~3種を構成糖としてなる食物繊維であることが好ましい。
本発明に係る食物繊維として、より具体的には、例えば、イヌリン、難消化性デキストリン、グアーガム分解物が挙げられ、これらのうちの1種であっても2種以上の組み合わせであってもよい。中でも、本発明に係る食物繊維としてはイヌリンが好ましい。
本発明に係る炭水化物としては、上述のように、単糖、二糖、三糖、四糖、五糖、糖アルコール、及び食物繊維からなる群から選択される2種以上の組み合わせであってもよい。この場合、本発明に係る炭水化物の組み合わせとしては特に制限されないが、二糖、三糖、四糖、及び五糖、より好ましくは三糖、四糖、及び五糖からなる群から選択される2種以上の糖質類の組み合わせ(本明細書中、場合により「混合糖」という)であることがより好ましい。このような組み合わせとしては、例えば、ケストース(三糖)、ニストース(四糖)、及びフラクトフラノシルニストース(五糖)からなるフラクトオリゴ糖;ガラクトシルラクトース(三糖)を含むガラクトオリゴ糖;ラフィノース(三糖)及びスタキオース(四糖)を含む大豆オリゴ糖;ラクトスクロース(三糖)を含む乳果オリゴ糖;キシロビオース(二糖)を含むキシロオリゴ糖;イソマルトース(二糖)、イソマルトトリオース(三糖)、バノース(三糖)を含むイソマルトオリゴ糖;マンノースが直鎖状にβ1,4マンノシド結合した形態をした二糖から十糖の混合物であるコーヒー豆マンノオリゴ糖が挙げられる。
これらの中でも、本発明に係る混合糖としては、生体内においてNMNを産生する腸内細菌により到達しやすい、すなわち、プレバイオティクスとして消化管上部で分解・吸収されず大腸までより到達しやすい観点から、フラクトオリゴ糖及びガラクトオリゴ糖からなる群から選択される少なくとも1種であることが好ましく、フラクトオリゴ糖であることがより好ましい。
本発明の組成物において、前記炭水化物の含有量(前記炭水化物が2種以上の組み合わせである場合にはそれらの合計量、以下同じ)としては、組成物の形態、投与量又は摂取量、投与又は摂取の目的や方法等に応じて適宜調整されるものであるため、特に限定されず、組成物の全質量に対して0.001~100質量%とすることができるが、例えば、0.001~99質量%の範囲が挙げられ、0.005~95質量%、0.01~80質量%、0.1~70質量%、1~60質量%、5~50質量%、又は10~50質量%とすることができる。
本発明の組成物の投与量又は摂取量(以下、場合により「用量」ともいう)としては、組成物の形態;投与又は摂取の目的や方法;対象の種、年齢、体重、性別、症状の程度等を考慮して、個々の場合に応じて適宜決定することができる。よって、特に限定されるものではないが、例えば、ヒト(好ましくは、成人)に対する用量としては、前記炭水化物の量(前記炭水化物が2種以上の組み合わせである場合にはそれらの合計量、以下同じ)で、対象の体重1kgあたり、かつ、1日あたり、1~1000mgであることが好ましく、10~1000mgであることがより好ましく、例えば、20~800mg、30~700mg、50~500mgとすることができる。
本発明の組成物は、本発明の効果が阻害されない限り、前記炭水化物の他に、さらに、食品又は医薬品上許容可能な他の成分を含有していてもよい。前記他の成分としては、特に限定されるものではないが、例えば、水、脂質、ミネラル類、ビタミン類、タンパク質、ペプチド、アミノ酸、有機酸、乳酸菌(ラクトバチルス属、ビフィドバクテリウム属等)、酵母、前記乳酸菌及び/又は酵母の処理物(破砕処理物、加熱処理物等)、前記乳酸菌及び/又は酵母の発酵物、製剤化補助剤が挙げられる。また、前記他の成分としては、前記炭水化物以外の他の糖類や他の有効成分であってもよい。
前記脂質としては、特に限定されないが、より具体的には、例えば、大豆油、コーン油、パーム油、エゴマ油、キャノーラ油、サフラワー油、ひまわり油、ごま油、米油、ぶどう種子油、魚油、べに花油、なたね油、及び落花生油等の天然油脂;炭素数6~12程度の中鎖脂肪酸トリグリセリド(MCT)等の合成油脂が挙げられる。
前記ミネラル類としては、特に限定されないが、より具体的には、例えば、カルシウム、マグネシウム、ナトリウム、カリウム、リン、鉄、マンガン、銅、亜鉛、ヨウ素、亜鉛、セレン、クロム、及びモリブデンが挙げられる。
前記ビタミン類としては、特に限定されないが、より具体的には、例えば、ビタミンA、B1、B2、B5、B6、B7、B9、B12、C、D、E、Kが挙げられる。
前記製剤化補助剤としては、特に限定されないが、より具体的には、例えば、溶剤、分散剤、乳化剤、増粘剤、増粘安定剤、ゲル化剤、界面活性剤、緩衝剤、安定剤、賦形剤、結合剤、崩壊剤、滑沢剤、矯味剤、溶解助剤、懸濁剤、コーティング剤、担体(固体担体、水等の液体担体)、保存剤、香料、着色剤、pH調整剤が挙げられる。
前記炭水化物以外の他の糖類としては、特に限定されないが、より具体的には、例えば、デンプン、マルトデキストリンが挙げられる。
前記炭水化物以外の他の有効成分としては、特に限定されないが、より具体的には、例えば、果実・野菜及びその加工品、動物及び植物抽出エキス、天然由来高分子(コラーゲン、ヒアルロン酸、コンドロイチン等)等が挙げられる。
上記の他の成分としては、1種のみであっても2種以上の組み合わせで含有されていてもよい。また、これら他の成分を含有する場合、当該他の成分の含有量は特に限定されず、組成物の形態、用量、投与又は摂取の目的や方法等に応じて適宜調整することができる。
本発明の組成物の投与形態としては、特に限定されず、経口投与、経腸投与、腹腔内投与等が挙げられ、本発明の組成物は、各投与形態に応じた組成物とすることができる。中でも、本発明の組成物としては、経口又は経腸によって投与又は摂取することが好ましく、それに応じて、経口用組成物又は経腸用組成物とすることが好ましい。特に、その容易さの観点から、経口によって投与又は摂取することがより好ましく、経口用組成物とすることがより好ましい。また、本発明の組成物は、組成物を投与又は摂取する目的、対象、方法、用量等に応じて、例えば、医薬組成物、医薬部外組成物、飲食組成物、又は飼料組成物とすることができる。
本発明に係る医薬組成物及び医薬部外組成物としては、例えば、製剤とすることができ、その形態は特に限定されないが、例えば、錠剤、丸剤、顆粒剤、散剤、粉末剤、カプセル剤等の固形剤;一般液剤、懸濁剤、乳剤、シロップ剤等の液剤;ゼリー剤;経腸投与剤;坐剤等の外用剤等が挙げられる。前記製剤は、例えば、前記炭水化物に前記製剤化補助剤のうちの1種又は2種以上、さらに必要に応じて前記他の成分のうちの1種又は2種以上を加えて、公知の方法又はそれに準じた方法によって製造することができる。
本発明に係る飲食組成物の形態としては、特に限定されず、例えば、バーのような固形状、飲料や流動食のような液状、ペースト状、半液体状、ゲル状(ゼリー状)、ゲル状油脂(半固形状油脂)、粉末状等の形態が挙げられる。また、前記飲食組成物としては、流動食、粉末流動食、栄養ペースト、経口・経管栄養剤、飲料、ゲル状食品などとして、経口・経腸栄養患者や高齢者、乳幼児等に摂取させることもできる。
本発明に係る飲食組成物の例としては、特に限定されないが、例えば、飲料(茶類、炭酸飲料、ココア、コーヒー、乳酸菌飲料、豆乳飲料、果汁・野菜汁飲料、清涼飲料、栄養飲料、アルコール飲料等)、加工食品(チョコレート、ガム、グミ、ゼリー、焼菓子(パン、ケーキ、クッキー、ビスケット等)、キャンデー等)、乳製品(調製粉乳(粉末ミルク等)、調整乳、乳飲料、発酵乳、ヨーグルト、アイスクリーム、チーズ、クリーム、バター、マーガリン、練乳等)、調味料(ソース、スープ、ドレッシング、マヨネーズ、マヨネーズタイプ調味料、クリーム等)、サプリメント、食用油、機能性食用油脂が挙げられる。このような飲食組成物は、例えば、既存の飲食品又はその原料若しくは製造過程の中間産物に、本発明に係る炭水化物を配合する方法等によって製造することができる。この場合に配合する炭水化物は、前記医薬組成物又は医薬部外組成物の形態であってもよい。
また、本発明に係る飲食組成物としては、例えば、一般食品、健康食品、機能性食品、保健機能食品(例えば、特定保健用食品、栄養機能食品、栄養補助食品、機能性表示食品等)、特別用途食品(例えば、幼児用食品、妊産婦用食品、病者用食品等)、メディカルフード(米国食品医薬品局(FDA)及びオーファンドラッグ法により定義された医師の管理において処方される食品)、治療食(治療の目的を果たすものであり、医師による食事箋に従って栄養士等が作成した献立に基づいて調理されたもの)、食事療法食としてもよい。また、前記飲食組成物には、その製品において本発明に係る炭水化物によりもたらされる作用・効能(例えば、加齢による疾患の予防又は改善、便秘の予防又は改善や炎症性腸疾患の予防又は改善)が表示されていてもよい。
本発明に係る飼料組成物としては、飼料組成物を投与又は摂取する目的、対象、方法、用量等に応じて、上記飲食組成物を適宜改変したものが挙げられる。
本発明の組成物としては、製造後から投与又は摂取までの間、包装容器内に包装(好ましくは封入)されていることが好ましい。前記包装容器としては、特に限定されるものではないが、例えば、包装紙、包装袋、ソフトバック、チューブ、チアパック、紙容器、缶、ボトル、カプセルが挙げられる。
本発明の組成物は、上記の炭水化物を有効成分として含有することで、腸内細菌叢によるNMN産生向上、より好ましくは、生体内における腸内細菌叢によるNMN産生向上のために用いることができる。そのため、本発明は、NMN産生向上のための、前記炭水化物の使用、並びに、NMN産生向上用組成物の製造のための、前記炭水化物の使用も提供する。
また、腸内細菌叢によるNMN産生が向上し、産生されたNMNを宿主(対象)が吸収することで、その生体内において、NMN濃度を向上させ、かかるNMNを前駆体とするNAD、さらにはその代謝物(ニコチンアミド、ニコチン酸、NAMN、NAAD等)の産生も向上させ、生体内濃度を向上させることが可能である。そのため、本発明の組成物は、生体内におけるNAD濃度向上のために用いることができ、本発明は、生体内NAD濃度向上のための、前記炭水化物の使用、並びに、生体内NAD濃度向上用組成物の製造のための、前記炭水化物の使用も提供する。
さらに、本発明の組成物は、生体内のNMN濃度及び/又はNAD濃度を向上させることで、NMNやその代謝産物であるNADの投与又は摂取によって予防又は改善されることが報告されている各種疾患や症状である、加齢による疾患(非特許文献1~4)、便秘(非特許文献5)、炎症性腸疾患(Navarro et al.,Br J Pharmacol.,2022,179,p.1839-1856)などの予防又は改善のために用いることもでき、加齢による疾患の予防又は改善のため、便秘の予防又は改善のため、又は、炎症性腸疾患の予防又は改善のために用いることが好ましい。本発明は、これらの疾患の予防又は改善のための、前記炭水化物の使用、並びに、これらの疾患の予防又は改善用組成物の製造のための、前記炭水化物の使用も提供する。
前記加齢による疾患としては、加齢(ヒトにおいて好ましくは、40歳以上、より好ましくは50歳以上、さらに好ましくは60歳以上)によって惹き起こされることが報告されている各疾患が挙げられ、例えば、肥満、インスリン抵抗性低下、脂質代謝能低下、眼機能低下、骨密度低下、免疫機能低下、耐糖能低下、酸化ストレス上昇、炎症、概日リズムの乱れ、肝機能低下、腎機能低下、骨格筋機能低下、心機能低下、アルツハイマー型認知症、中枢神経機能低下が挙げられる。
本発明において、生体内における腸内細菌叢によるNMN産生が向上することは、例えば、糞便を採取して、その糞便中のNMN濃度を測定し、本発明の組成物又は本発明に係る炭水化物を投与前の対象又は投与しなかった群を基準として、前記NMN濃度が高くなったこと等によって確認することができる。前記NMN濃度は、適宜公知の方法又はそれに準じた方法で測定することができ、例えば、下記の実施例に記載の液体クロマトグラフ質量分析計(LC-MS/MS)を用いた方法で測定することができる。
<NMN産生向上方法>
上記の炭水化物を対象に投与することで、同対象の腸内細菌叢によるNMN産生を向上させることができる。また、それにより、生体内NAD濃度を向上させることや、前記各種疾患や症状(好ましくは、加齢による疾患、便秘、炎症性腸疾患)を予防又は改善させることができる。そのため、本発明は、前記炭水化物の有効量を対象に投与する、NMN産生向上方法、生体内NAD濃度向上方法、前記各種疾患(好ましくは、加齢による疾患、便秘、炎症性腸疾患)の予防又は改善方法(以下、場合により「本発明の方法」と総称する)も提供する。
上記の炭水化物を対象に投与することで、同対象の腸内細菌叢によるNMN産生を向上させることができる。また、それにより、生体内NAD濃度を向上させることや、前記各種疾患や症状(好ましくは、加齢による疾患、便秘、炎症性腸疾患)を予防又は改善させることができる。そのため、本発明は、前記炭水化物の有効量を対象に投与する、NMN産生向上方法、生体内NAD濃度向上方法、前記各種疾患(好ましくは、加齢による疾患、便秘、炎症性腸疾患)の予防又は改善方法(以下、場合により「本発明の方法」と総称する)も提供する。
本発明の方法は、本発明に係る炭水化物の有効量を対象に投与する工程を含む。本発明の方法に係る対象としては、ヒト又は非ヒト哺乳動物が挙げられ、前記非ヒト哺乳動物としては、マウス、ヒツジ、ウシ、ブタ、ウマ、サル、イヌ、ネコ、ウサギ等が挙げられる。上述のように腸内細菌叢には個体差があり、NMNを産生しないものも存在し得るため、前記対象としては、その腸内細菌叢がNMNを産生するものであることが好ましい。なお、腸内細菌叢がNMNを産生することは、個体差があるため一概にいえるものではないが、例えば、対象の糞便(好ましくは、1mg)をグルコース0.5%含有GAM培地(好ましくは、1mL)で37℃において嫌気条件で12時間培養した培地においてNMNが検出される(好ましくは、液体クロマトグラフ質量分析計(LC-MS/MS)を用いた方法(例えば、下記の実施例に記載の方法)で測定したときに定量限界値(例えば、1ng/mL)以上が検出される)ことで判断することができる。また、前記対象としては、既に上記各種疾患(例えば、加齢による疾患、便秘、炎症性腸疾患)に罹患している者であっても、その発症の予防を目的とする者であってもよい。
本発明の方法において、前記炭水化物は、そのまま、より好ましくは本発明の組成物として投与することができ、前記対象に経口又は経腸で投与することが好ましく、経口で投与することがより好ましい。なお、本発明において、経口投与には、前記飲食組成物や前記飼料組成物等の摂取も含む。
本発明の方法において、前記炭水化物の有効量としては、投与の目的や方法;対象の種、年齢、体重、性別、疾患、症状の程度等を考慮して、個々の場合に応じて適宜決定されるものであるため、特に限定されないが、上述の本発明の組成物の用量と同じく、例えば、ヒト(好ましくは、成人)に対しては、前記炭水化物の量で、対象の体重1kgあたり、かつ、1日あたり、1~1000mgであることが好ましく、10~1000mgであることがより好ましく、例えば、20~800mg、30~700mg、50~500mgとすることができる。本発明の方法において、投与回数は、1日1回~適当な回数に分けることができる。
以下、試験例に基づいて本発明をより具体的に説明するが、本発明は以下の例に限定されるものではない。なお、下記の各試験例において、「%」の表示は、特に記載のない場合、重量/容量(w/v:g/mL)パーセントを示す。
<試験例1> ヒト腸内細菌叢のNMN産生能
それぞれ別人から採取したヒト糞便(#1~3)1mgを、終濃度が0.5%となるようにグルコースを添加したGAM培地1mLに添加し、37℃、嫌気グローブボックス内において12時間培養した後、培養液中のNMN濃度を測定した。前記GAM培地は、GAM糖分解用半流動培地(日水製薬製)を所定濃度となるように水に溶解し、0.8μmフィルターでろ過した後に115℃において15分間オートクレーブ滅菌したものを用いた。また、前記NMN濃度は、次の方法で測定した。すなわち、先ず、培養液を遠心分離して得られた上清を0.22μmフィルターでろ過した後、ろ液をメタノール(終濃度80v/v%)と混合して30分間氷上に静置した。次いで、これを遠心分離して得られた上清を遠心エバポレーターで乾固した。次いで、得られた乾固物を酢酸アンモニウム溶液で溶解し、液体クロマトグラフ質量分析計(LC-MS/MS)に供して下記の条件で分析し、NMN濃度を測定した。各ヒト糞便(#1~3)におけるNMN濃度(ng/mL)を図1に示す。

図1に示したように、#1~3のヒト糞便でNMNが検出され、ヒト腸内細菌叢がNMNを産生することが確認された。また、前記GAM培地におけるヒト糞便培養系により、当該ヒト糞便由来の腸内細菌叢によるNMN産生能の評価が可能であることも確認された。
<試験例2> 炭水化物によるNMN産生向上評価
先ず、前記GAM培地(ただし、グルコース不添加)に、下記の表1に示す各炭水化物のいずれかを終濃度が0.5%となるように添加して、各評価培地をそれぞれ調製した。また、比較例として、炭水化物に代えて蒸留水(DW)を添加した評価培地も調製した。次いで、試験例1と同じ#1~3のヒト糞便1mgを、前記評価培地1mLにそれぞれ添加し、37℃、嫌気グローブボックス内において12時間培養した。培養後、培養液中のNMN濃度を試験例1に記載の方法で液体クロマトグラフ質量分析計(LC-MS/MS)により測定した。#1~3のヒト糞便をそれぞれ培養したときのNMN濃度の平均値を図2に示す。また、前記平均値についてさらに、各炭水化物分類ごとの平均値(ただしグルコース及びスクロースを除く)を算出した結果を下記の表2に示す。

図2に示したように、各炭水化物を添加した評価培地では、添加しない評価培地(DW)とは異なり、NMNが検出された。特に、ケストース、セロビオース、フラクトオリゴ糖、ラクトスクロース、及びラフィノースは、培養に用いられる一般的な糖であるグルコースやスクロース(ただし、これらは生体内において消化管上部で分解・吸収される)よりもNMN濃度が高値となった。また、表2に示したように、各炭水化物分類ごとのNMN濃度の平均値は、三糖で最も高く、次いで、混合糖(三~五糖の混合糖)、四糖、二糖の順に高値となった。ただし、図2に示したように、二糖であっても、シアル化された修飾糖であるシアリルラクトースでは、NMN濃度が比較的低値となった。さらに、三糖であっても、構成糖にフコースを含むフコシルラクトースでは、NMN濃度が比較的低値となった。
<試験例3> マウス腸管内におけるNMN産生向上評価
試験例2で優れたNMN産生向上効果が確認された炭水化物のうち、フラクトオリゴ糖(FOS、1-ケストース、ニストース、及び1-フラクトフラノシル-D-ニストースの混合物、明治フードマテリア株式会社製、製品名:メイオリゴP)を用いて、マウス腸管内(大腸内)におけるNMN産生向上効果を評価した。先ず、7週齢の雄ICRマウス(日本エスエルシー株式会社製)を8個体ずつ2群に分け、一方の群には5% FOS水溶液を自由摂取させてFOS群とし、他方の群には水を自由摂取させてControl群とし、それぞれ10日間飼育した。飼育期間経過後、3時間絶食させた後に盲腸内容物を採取し、その重量を測定した。また、採種した盲腸内容物をマイクロチューブに20mg秤量し、メタノールを終濃度80v/v%となるように混合して30分間氷上に静置した。次いで、これを遠心分離して得られた上清を別のマイクロチューブに移した後、ペレットに80v/v%メタノールを添加して混合し、再度遠心分離して得た上清を先に別のマイクロチューブに移したものと混合した。次いで、この混合した上清を遠心エバポレーターで乾固し、得られた乾固物を酢酸アンモニウム溶液で溶解し、0.22μmフィルターでろ過した後、ろ液中のNMN濃度を試験例1に記載の方法で液体クロマトグラフ質量分析計(LC-MS/MS)により測定し、当該盲腸内容物中のNMN濃度を算出した。さらに、測定されたNMN濃度に盲腸内容物重量を乗じた量(total(μg))を算出し、totalの各群における平均値を算出した。結果を図3に示す。
試験例2で優れたNMN産生向上効果が確認された炭水化物のうち、フラクトオリゴ糖(FOS、1-ケストース、ニストース、及び1-フラクトフラノシル-D-ニストースの混合物、明治フードマテリア株式会社製、製品名:メイオリゴP)を用いて、マウス腸管内(大腸内)におけるNMN産生向上効果を評価した。先ず、7週齢の雄ICRマウス(日本エスエルシー株式会社製)を8個体ずつ2群に分け、一方の群には5% FOS水溶液を自由摂取させてFOS群とし、他方の群には水を自由摂取させてControl群とし、それぞれ10日間飼育した。飼育期間経過後、3時間絶食させた後に盲腸内容物を採取し、その重量を測定した。また、採種した盲腸内容物をマイクロチューブに20mg秤量し、メタノールを終濃度80v/v%となるように混合して30分間氷上に静置した。次いで、これを遠心分離して得られた上清を別のマイクロチューブに移した後、ペレットに80v/v%メタノールを添加して混合し、再度遠心分離して得た上清を先に別のマイクロチューブに移したものと混合した。次いで、この混合した上清を遠心エバポレーターで乾固し、得られた乾固物を酢酸アンモニウム溶液で溶解し、0.22μmフィルターでろ過した後、ろ液中のNMN濃度を試験例1に記載の方法で液体クロマトグラフ質量分析計(LC-MS/MS)により測定し、当該盲腸内容物中のNMN濃度を算出した。さらに、測定されたNMN濃度に盲腸内容物重量を乗じた量(total(μg))を算出し、totalの各群における平均値を算出した。結果を図3に示す。
図3に示したように、水を摂取させたControl群に比べて、フラクトオリゴ糖を摂取させたFOS群では盲腸内容物中のNMN量が多く、Control群とFOS群との間の有意差検定をt-testにより行ったところ、p<0.01となり、両群の間には有意差があることが確認された。したがって、フラクトオリゴ糖は生体内(in vivo)においても腸内細菌叢によるNMN産生量を高めることが確認され、また、これに含有されるケストース、ニストース、及びフラクトフラノシルニストースからなる群から選択される少なくとも1種も同様の効果を奏することが示唆された。
<試験例4> ヒトフローラマウス腸管内におけるNMN産生向上評価
ケストース(明治フードマテリア株式会社製)を用いて、無菌マウスにヒト腸内細菌叢を移植したヒトフローラマウスの腸管内(大腸内)におけるNMN産生向上効果を評価した。先ず、8週齢の雄無菌マウス(C57BL/6N、日本クレア株式会社製、3個体)にヒト糞便を経口投与し、3週間馴化させた後(Day 0)に採糞した。この間、水を自由摂取させた。採糞後、飲水を水から5%ケストース水溶液に交換して自由摂取させ、14日間飼育した。ケストース投与開始から1、3、9、14日後(Day 1、3、9、14)に採糞した。また、各採糞日(Day)に採糞した糞便はそれぞれマイクロチューブに20mg秤量し、メタノールを終濃度80v/v%となるように混合して30分間氷上に静置した。次いで、これを遠心分離して得られた上清を別のマイクロチューブに移した後、ペレットに80v/v%メタノールを添加して混合し、再度遠心分離して得た上清を先に別のマイクロチューブに移したものと混合した。次いで、この混合した上清を遠心エバポレーターで乾固し、得られた乾固物を酢酸アンモニウム溶液で溶解し、0.22μmフィルターでろ過した後、ろ液中のNMN濃度を試験例1に記載の方法で液体クロマトグラフ質量分析計(LC-MS/MS)により測定し、当該糞便中のNMN濃度(ng/mL)を算出した。各採糞日におけるNMN濃度の3個体の平均値を図4に示す。
ケストース(明治フードマテリア株式会社製)を用いて、無菌マウスにヒト腸内細菌叢を移植したヒトフローラマウスの腸管内(大腸内)におけるNMN産生向上効果を評価した。先ず、8週齢の雄無菌マウス(C57BL/6N、日本クレア株式会社製、3個体)にヒト糞便を経口投与し、3週間馴化させた後(Day 0)に採糞した。この間、水を自由摂取させた。採糞後、飲水を水から5%ケストース水溶液に交換して自由摂取させ、14日間飼育した。ケストース投与開始から1、3、9、14日後(Day 1、3、9、14)に採糞した。また、各採糞日(Day)に採糞した糞便はそれぞれマイクロチューブに20mg秤量し、メタノールを終濃度80v/v%となるように混合して30分間氷上に静置した。次いで、これを遠心分離して得られた上清を別のマイクロチューブに移した後、ペレットに80v/v%メタノールを添加して混合し、再度遠心分離して得た上清を先に別のマイクロチューブに移したものと混合した。次いで、この混合した上清を遠心エバポレーターで乾固し、得られた乾固物を酢酸アンモニウム溶液で溶解し、0.22μmフィルターでろ過した後、ろ液中のNMN濃度を試験例1に記載の方法で液体クロマトグラフ質量分析計(LC-MS/MS)により測定し、当該糞便中のNMN濃度(ng/mL)を算出した。各採糞日におけるNMN濃度の3個体の平均値を図4に示す。
図4に示したように、ケストース投与期間中のDay1、3、9においては、ケストース投与期間前であるDay 0に比べて糞便中のNMN濃度が高くなり、Day1、9においては、Dunnett’s testで、p<0.10(図4中、#)又はp<0.05(図4中、*)となった。したがって、ケストースは生体内(in vivo)におけるヒト腸内細菌叢によるNMN産生量を高めることが確認された。
以上説明したように、本発明によれば、腸内細菌叢によるニコチンアミドモノヌクレオチド(NMN)産生向上作用を有する組成物を提供することが可能となる。また、本発明の組成物は、飲食組成物としても提供することができるため、摂取しやすく、摂取を習慣化しやすい。さらに、本発明の組成物は、腸内細菌叢によるNMN産生量を高めることで、生体内におけるNAD濃度の向上や、加齢による疾患の予防又は改善、並びに、便秘や炎症性腸疾患の予防又は改善のために用いることができる。

- 単糖(ただし、グルコース及びキシロースを除く)、二糖、三糖、四糖、五糖、糖アルコール、及び食物繊維からなる群から選択される少なくとも1種の炭水化物を有効成分として含有することを特徴とする、ニコチンアミドモノヌクレオチド(NMN)産生向上のための組成物。
- 前記炭水化物として単糖を含有し、前記単糖がタガトースであることを特徴とする、請求項1に記載の組成物。
- 前記炭水化物として二糖を含有し、前記二糖が、セロビオース、ラクチュロース、トレハロース、及びシアリルラクトースからなる群から選択される少なくとも1種であることを特徴とする、請求項1に記載の組成物。
- 前記炭水化物として三糖を含有し、前記三糖が、ケストース、ラクトスクロース、ラフィノース、及びフコシルラクトースからなる群から選択される少なくとも1種であることを特徴とする、請求項1に記載の組成物。
- 前記炭水化物として四糖を含有し、前記四糖が、スタキオース及びニストースからなる群から選択される少なくとも1種であることを特徴とする、請求項1に記載の組成物。
- 前記炭水化物として五糖を含有し、前記五糖が、フラクトフラノシルニストースであることを特徴とする、請求項1に記載の組成物。
- 前記炭水化物として糖アルコールを含有し、前記糖アルコールが、還元イソマルツロース、ラクチトール、ソルビトール、マンニトール、及びキシリトールからなる群から選択される少なくとも1種であることを特徴とする、請求項1に記載の組成物。
- 前記炭水化物として食物繊維を含有し、前記食物繊維が、イヌリンであることを特徴とする、請求項1に記載の組成物。
- 経口用組成物又は経腸用組成物であることを特徴とする、請求項1~8のうちのいずれか一項に記載の組成物。
- 飲食組成物、医薬組成物、又は医薬部外組成物であることを特徴とする、請求項1~8のうちのいずれか一項に記載の組成物。
- 便秘の予防又は改善のための組成物であることを特徴とする、請求項1~8のうちのいずれか一項に記載の組成物。
- ヒトに対する投与量が、体重1kgあたり、かつ、1日あたり、前記炭水化物の量で10~1000mgであることを特徴とする、請求項11に記載の組成物。
- 炎症性腸疾患の予防又は改善のための組成物であることを特徴とする、請求項1~8のうちのいずれか一項に記載の組成物。
- ヒトに対する投与量が、体重1kgあたり、かつ、1日あたり、前記炭水化物の量で10~1000mgであることを特徴とする、請求項13に記載の組成物。
- 加齢による疾患の予防又は改善のための組成物であることを特徴とする、請求項1~8のうちのいずれか一項に記載の組成物。
- ヒトに対する投与量が、体重1kgあたり、かつ、1日あたり、前記炭水化物の量で10~1000mgであることを特徴とする、請求項15に記載の組成物。
