Abstract
본 발명은 개선된 생체 내 리프로그래밍 시스템 및 이를 이용한 세포 전환 방법에 관한 것이다. 본 발명의 리프로그래밍 시스템은 출발세포 마커 프로모터, 유전자 다능성 유지 단백질, 아미노산 분리 펩타이드, Cre 재조합효소, 지향세포 마커 프로모터, LoxP, 및 형광단백질을 코딩하는 유전자를 포함하고, 세포의 전환을 확인하기 위해 세포의 고정을 필요치 않고, 세포가 살아있는 상태에서 실시간으로 모니터링이 가능하며, 생체 외(in vitro) 또는 생체 내(in vivo)에서 모두 이용이 가능하다. 따라서 본 발명은 바이오 및 의학 분야에서 크게 이용될 것으로 기대된다.
Description
본 발명은 개선된 생체 내 리프로그래밍 시스템 및 이를 이용한 세포 전환 방법에 관한 것이다.
최근 줄기세포 연구 분야에서는 SOX2(Sex determining region Y-box 2)와 같은 유전자 다능성 유지 단백질이 규명됨에 따라, 이미 분화가 완료된 세포에서 유전자 다능성 유지 단백질을 과발현시켜 다른 세포로 전환시키고자 하는 노력이 이루어지고 있다. 예를 들면, Huang 등은 일반 체세포에서 Foxa3, Hnf1a, 및 Gata4 유전자를 이용하여 간세포로 전환시키는 연구를 수행하였고(Huang et al., Nature, vol.475, pp.386-389, 2011), 한국 등록특허 제 10-1702629 호에는 일반 체세포를 FLI1, 또는 ETV2 유전자를 이용하여 혈관전구세포로 전환시키는 방법이 개시되어 있다. 이와 같이 분화가 이미 완료된 세포를 다른 세포로 직접 전환시키는 방법은 기존의 유도만능줄기세포(Induced Pluripotent Stem Cells, iPSCs)로 리프로그래밍 하고 이를 재분화하여 목적하는 세포로 만들어야 하는 과정과 달리, 유도만능줄기세포단계를 거치지 않고 바로 목적하는 세포로의 전환을 유도한다는 점에서 질병모델링과 신약 발굴 등에 이용될 가능성을 인정받고 있으며, 재생의학 분야에서도 활용성이 높을 것으로 주목받고 있다.
그러나 종래의 공지된 세포 전환 방법들은 전환된 세포의 효율이 상당히 낮고, 세포의 전환을 실시간으로 모니터링할 수 있는 기술이 부재한 문제점이 있었다. 세포의 상태를 실시간으로 측정이 불가하므로, 세포의 전환이 완료되기까지의 정확한 시간을 측정할 수 없고, 또한 세포의 전환을 확인하기 위해서는 세포를 고정(fixation)한 후 면역학적 방법으로 확인해야했다. 따라서 전환된 세포를 2차적으로 이용하는 것이 불가하고, 오직 생체 외(in vitro) 세포에서만 실시 가능한 문제점이 파생하였다.
따라서 본 발명은 개선된 생체 내 리프로그래밍 시스템 및 이를 이용한 세포 전환 방법에 관한 것으로, 본 발명의 리프로그래밍 시스템은 세포의 전환을 확인하기 위해 세포의 고정을 필요치 않고, 세포가 살아있는 상태에서 실시간으로 모니터링이 가능하며, 생체 외(in vitro) 또는 생체 내(in vivo)에서 모두 이용이 가능하므로, 바이오 및 의학 분야에서 크게 이용될 것으로 기대된다.
본 발명은 상기와 같은 종래의 기술상의 문제점을 해결하기 위해 안출된 것으로, 개선된 생체 내 리프로그래밍 시스템 및 이를 이용한 세포 전환 방법에 관한 것이다.
그러나 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 당 업계에서 통상의 지식을 가진 자에게 명확하게 이해될 수 있을 것이다.

본 발명은 개선된 생체 내 리프로그래밍 시스템을 제공한다. 본 발명의 생체 내 리프로그래밍 시스템은 HEK293FT(human embryonic kidney 293FT cells)세포에 G glycoprotein이 융합된 수포성 구내염바이러스 유전자(fusiogenic envelope G glycoprotein of the vesicular stomatitis virus, VSV-G), GAG/Pol 유전자, 및 발현벡터(expression vector)를 Viafect transfection reagent(E4981, Promega, USA)를 사용하여 도입시켜 제조한 것이다. HEK293FT 세포는 인간 태아 신장세포에서 유래된 세포로, 성장 속도가 빠르고 형질전환(transfection) 효율이 높으므로, 유전자 연구 분야에서 활발하게 이용되는 세포이다.
상기의 발현벡터는 출발세포 마커 프로모터, 유전자 다능성 유지 단백질, 아미노산 분리 펩타이드, Cre 재조합효소, 지향세포 마커 프로모터, LoxP, 및 형광단백질을 코딩하는 유전자를 순차적으로 전사하도록 설계하였다. 형광단백질은 역전사 방향으로 디자인하여 Cre 재조합효소의 발현에 의해 LoxP를 인지하여 전사 방향으로 복위하도록 설계하였다.
본 명세서에 있어서, 상기의 출발세포 마커 프로모터란 엄밀하게는 출발세포에서 세포 특이적으로 발현하는 마커 단백질을 코딩하는 유전자의 프로모터인 것이고, 지향세포 마커 프로모터란 엄밀하게는 지향세포에서 세포 특이적으로 발현하는 마커 단백질을 코딩하는 유전자의 프로모터인 것이다.
상기의 출발세포는 분화가 이미 완료된 세포로서 발현벡터가 도입되는 세포이고, 발현벡터의 전사방향에서 첫번째 위치하는 마커 프로모터를 발현하는 세포이다. 상기 출발세포는 세포 종류에 제한 없이 선택 가능하고, 분화가 완료된 모든 체세포 또는 생식세포가 출발세포로 선택될 수 있다.
상기의 지향세포는 출발세포가 전환을 목표로 하는 세포이고, 발현벡터의 전사방향에서 두번째 위치하는 마커 프로모터를 발현하는 세포이다. 상기 지향세포 또한 세포 종류에 제한 없이 선택 가능하며, 분화가 완료되거나, 또는 완료되지 않은 모든 체세포 또는 생식세포가 지향세포로 선택될 수 있다.
구체적으로 한 양태에서, 상기 출발세포와 지향세포는 상피세포, 근육세포, 신경세포, 기질세포, 및 골세포의 하위 분류로부터 선택될 수 있다.
상기의 상피세포는 세포의 층을 이루고, 조직 또는 샘의 안과 밖을 덮는 막조직을 구성하는 세포로서, 단층편평상피(simple squamous epithelium), 단층입방상피(simple cuboidal epithelium), 단층원주상피(simple columnar epithelium), 중층편평상피(stratified squamous epithelium), 중층입방상피(stratified cuboidal epithelium), 중층원주상피(stratified columnar epithelium), 거짓중층상피(pseudostratified epithelium), 요로상피(transitional epithelium), 및 샘상피(glandular epithelium)를 포괄하는 의미이다.

상기 A세포가 기질세포인 경우에 A세포 마커는 Col1A2(Collagen Type I Alpha 2 Chain), FAP(Fibroblast-activation protein), FSP1(Fibroblast-specific protein 1), 비멘틴(Vimentin), ACTA(alpha smooth muscle Actin), Hsp47(Heat shock protein 47), 아그레칸(Aggrecan), CD44, CD45, CD73, Calcitonin, OSCAR(osteoclast-associated receptor), RANK(Receptor activator of nuclear factor κ B), GFAP(Glial fibrillary acidic protein), TREM2(Triggering receptor expressed on myeloid cells 2), HexB(Beta-hexosaminidase subunit beta), S100(calcium-binding protein), CD69, 및/또는 Gpr34(Probable G-protein coupled receptor 34) 등을 선택할 수 있으나, 이에 제한되는 것은 아니다.
상기 B세포가 신경세포인 경우에 B세포 마커는 SYN(Synapsin), Tuj1(Neuron-specific class III beta-tubulin), MAP2(Microtubule-associated protein 2), 및/또는 신경미세섬유(Neurofilament) 등을 선택할 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 일 구체예에서와 같이, A세포 마커 프로모터로 GFAP(Glial fibrillary acidic protein) promotor, 유전자 다능성 유지 단백질로 SOX2(Sex determining region Y-box 2), 아미노산 분리 펩타이드로 T2A(2A peptide), B세포 마커 프로모터로 SYN(Synapsin), 형광단백질로 eGFP(enhanced green fluorescent protein)를 대응시킬 수 있다. 이러한 경우, A세포 별아교세포이고, 본 발현벡터에 의해 별아교세포로부터 전환되는 B세포는 신경세포가 된다. 상기의 예시 벡터를 별아교세포에 도입하면, 별아교세포 마커인 GFAP 프로모터에 의해 SOX2와 Cre 재조합효소가 발현되고, 유전자 다능성 유지 단백질인 SOX2의 발현은 별아교세포를 신경세포로 전환시킬 수 있으며 Cre 재조합효소에 의해 LoxP site가 인지돼 Loxp site 사이의 역전사 방향 형광단백질을 전사방향으로 복위시킨다. 별아교세포가 신경세포로 전환되면, 신경세포 마커인 SYN 프로모터가 작동하고, SYN 프로모터에 의해 전사방향으로 복위된 eGFP가 발현되어, 세포가 녹색 형광을 발현한다. 이로서, A세포(별아교세포)로부터 B세포(신경세포)로 전환된 세포를 생체 내에서 세포가 살아있는 상태로 확인 가능하다.
만약, 상기의 발현벡터가 A세포(별아교세포) 이외의 다른 세포로 도입된다면 A세포 마커 프로모터(GFAP promotor)가 작동하지 않으므로, 형광이 발현하지 않는다. 만약, 상기의 발현벡터가 A세포(별아교세포)에 도입되더라도 유전자 다능성 유지 단백질(SOX2)에 의해 B세포(신경세포) 이외의 다른 세포로 전환된다면, B세포 마커 프로모터(SYN promotor)가 작동하지 않으므로 형광이 발현하지 않는다. 만약, 상기의 발현벡터가 처음부터 B세포(신경세포)에 도입된다면 Cre 재조합효소가 발현되지 않으므로 Cre-loxP 작동의 실패에 의해 형광이 발현하지 않는다. 오직 A세포 (별아교세포)가 B세포(신경세포)로 전환된 경우에만, 형광이 발현된다.
상기의 T2A 펩타이드는 18개 정도의 비교적 짧은 아미노산 서열을 갖는 self-processing viral peptide로서, 두개의 서로 다른 단백질 사이에 2A서열을 넣고 fusion하여 세포에서 발현시키면 2A의 마지막 아미노산인 Pro앞에서 아미노산 결합이 분리되므로, 마치 두개의 단백질을 따로 발현한 것과 같은 효과를 낼 수 있다. 2A의 일종으로 F2A, 또는 E2A로 대체 이용이 가능하다.
상기의 Cre-loxP 시스템은 특정 유전자를 선택적으로 제거하거나(Deletion), 방향을 회전시키거나(Inversion), 또는 위치를 변경(Translication)시킬 수 있는 특정 사이트 재조합(site-specific recombination) 시스템이다. 형질전환 순간부터 유전자가 발현되는 다른 벡터와 달리, 필요한 시기에 필요한 조직에서만 삽입된 유전자의 발현을 유도할 수 있다. 조작하고자 하는 유전자 앞뒤로 loxP를 위치시키고, 이에 앞서 Cre 재조합효소를 발현시키면, Cre-loxP 결합에 의해 loxP 사이에 위치한 타겟 유전자가 조작된다(https://www.addgene.org/cre-lox/).
본 발명의 개선된 생체 내 리프로그래밍 시스템은 질병모델링과 신약 발굴 등에 이용될 가능성을 인정받고 있으며, 재생의학 분야에서도 활용성이 높을 것으로 주목받고 있다.
예를 들면, 특정세포를 A세포로 설정하여 B세포로 전환시킴으로서 A세포를 제거하면, A세포의 부재에 의해서 어떤 질병이 발생하는지 모델링이 가능하다.
예를 들면, 특정 질병이 있는 개체에서 A세포를 B세포로 전환시켜 B세포의 수를 증가시킴으로서 질병이 치료되는 경우, 상기 질병에 대해 B세포를 약학적 용도로 이용할 수 있다.
또한 예를 들면, 분화가 완료된 세포로서 재생이 거의 이루어지지 않은 세포인 경우, 손상 후 기능의 상실을 기타세포(A세포)를 해당세포(B세포)로 전환시킴으로서 기능의 재생을 유도할 수 있다. 보다 구체적으로 설명하면, 신경세포는 손상 후 재생이 거의 이루어지지 않으므로 신경 손상시 감각적, 운동적 장애를 유발할 수 있다. 그러나 신경세포 이외의 기타세포(A세포)를 신경세포(B세포)로 전환시킴으로서, 기능의 상실을 회복하는 것이 가능하다.
본 발명의 개선된 생체 내 리프로그래밍 시스템은 특정세포를 A세포로 설정하여 B세포로 전환시키는 과정을 세포가 살아있는 상태에서 생세포 이미징이 가능하다. 이는 세포를 고정(fixation)하지 않으므로 전환된 세포를 2차적으로 이용하는 것이 가능하고, 세포가 A세포로부터 B세포로 전환되는 과정을 타임랩스(time-lapse 시간차) 없이 실시간으로 확인 가능하다. 상기 생세포 이미징은 형광 현미경을 이용하는 것이 바람직하다.
본 발명의 일 구체예에서 "진단"이란, 병리 상태의 존재 또는 특징을 확인하는 것을 의미하며, 질환의 발생을 확인하고, 질환의 심도를 판단하는 것이다. 이를 위해 질환 의심 개체로부터의 조직의 육안적 또는 세포학적 확인으로 의심 질환을 진단할 수 있으며, 질환의 발병 의심 개체의 검체(임상적으로는 세포, 혈액, 수액, 흉수, 복수, 관절액, 농(膿), 분비액, 담,인두점액, 요(尿), 담즙, 대변등) 내에 포함되어 있는 지질의 농도를 측정하는 방법, 질환 관련 단백질을 직접 검출하는 방법, 상기 단백질에 대한 항체의 농도를 측정하는 방법, 또는 상기 단백질 및/또는 상기 항체를 코딩하는 핵산을 직접 검출하는 방법으로 질환을 진단할 수 있다. 지질을 측정하는 방법으로는 지질에 특이적으로 반응하는 염색액을 이용한 염색이 있을 수 있으나, 이에 제한되는 것은 아니며, 항원-항체 결합 또는 질병 관련 단백질을 직접 검출하는 방법을 이용한 진단적 수단으로는 웨스턴 블랏, ELISA(enzyme linked immunosorbent assay), 방사선면역분석(RIA: Radioimmunoassay), 방사 면역 확산법(radioimmunodiffusion), 오우크테로니(Ouchterlony) 면역 확산법, 로케트(rocket) 면역전기영동, 조직면역 염색, 면역침전 분석법(Immunoprecipitation Assay), 보체 고정 분석법(Complement Fixation Assay), 유세포분석(Fluorescence Activated Cell Sorter, FACS), 단백질 칩(protein chip) 등이 있으나, 이에 제한되는 것은 아니며, 질환 관련 단백질을 코딩하는 핵산을 직접 검출하는 방법으로는 역전사 중합효소반응(RT-PCR), 경쟁적 역전사 중합효소반응(Competitive RT-PCR), 실시간 역전사 중합효소반응(Real-time RT-PCR), RNase 보호 분석법(RPA; RNase protection assay), 노던 블랏팅(Northern blotting) 또는 DNA 칩 등이 있으나, 이에 제한되는 것은 아니다.
본 발명의 일 구체예에서 "스크리닝"이란, 여러 물질로 이루어진 후보군으로부터 목적으로 하는 어떤 특정한 성질을 갖는 물질을 특정한 조작 또는 평가 방법으로 선별하는 것이다. 본 발명의 목적상, 본 발명의 스크리닝은, 특정 질병이 있는 개체에서 A세포를 B세포로 전환시켜 B세포의 수를 증가시킴으로서 질병이 치료되는 경우가 발생하는 B세포를 추적하는 과정이고, 또한 상기의 B세포로 전환 효과가 가장 현저한 A세포를 추적하는 과정이나, 이에 한정하는 것은 아니다.
본 발명의 일 구체예에서 "투여"란, 어떠한 적절한 방법으로 개체에게 특정한 조성물을 도입하는 것을 의미하며, 조성물의 투여경로는 목적 조직에 도달할 수 있는 한 어떠한 일반적인 경로를 통하여 투여될 수 있다. 경구 투여, 복강 내 투여, 혈관 내 투여, 근육 내 투여, 피하 투여, 피내 투여, 비내 투여, 폐내 투여, 직장내 투여, 강내 투여, 복강 내 투여, 경막 내 투여가 이루어질 수 있으나, 이에 제한되지는 않는다. 본 발명에서 유효량은 질환의 종류, 질환의 중증도, 조성물에 함유된 유효 성분 및 다른 성분의 종류 및 함량, 제형의 종류 및 개체의 연령, 체중, 일반 건강 상태, 성별 및 식이, 투여 시간, 투여 경로 및 조성물의 분비율, 치료 기간, 동시 사용되는 약물을 비롯한 다양한 인자에 따라 조절될 수 있다. 성인의 경우, 상기 조성물을 1회 50ml~500ml의 양으로 체내에 투여 가능하며, 화합물일 경우 0.1ng/kg-10㎎/kg, 단백질에 대한 모노클로날 항체일 경우 0.1ng/kg-10㎎/kg의 용량으로 투여될 수 있다. 투여간격은 1일 1회 내지 12회일 수 있으며, 1일 12회 투여할 경우에는 2시간마다 1회씩 투여할 수 있다. 또한 펩타이드들 및 핵산들은 면역 반응을 증진하기 위하여 고안된 다른 치료, 예를 들어 당업계에 주지된 것과 같은 어쥬번트 또는 사이토카인(또는 사이토카인을 코딩하는 핵산)과 혼합하여 투여될 수 있다. 바이오리스틱(biolistic) 전달 또는 생체 외(ex vivo) 처리와 같은 다른 표준 전달 방법들이 사용될 수도 있다. 생체 외 처리에서 예를 들어 항원제시 세포들(APCs), 수지상세포들, 말초혈액 단핵구 세포들, 또는 골수세포들을 환자 또는 적당한 공여자로부터 얻어서 면역 조성물로 생체 외에서 활성화된 후 그 환자에게 투여될 수 있다.

이하 상기 본 발명을 단계별로 상세히 설명한다.
종래의 공지된 세포 전환 방법들은 세포의 전환을 실시간으로 모니터링할 수 있는 기술이 부재하여, 세포의 상태를 실시간으로 측정이 불가능하고, 오직 생체 외(in vitro) 세포에서만 실시 가능한 문제점이 있었다. 따라서 본 발명은 개선된 생체 내 리프로그래밍 시스템 및 이를 이용한 세포 전환 방법에 관한 것으로, 본 발명의 리프로그래밍 시스템은 세포가 살아있는 상태에서 실시간으로 모니터링이 가능하며, 생체 외(in vitro) 또는 생체 내(in vivo)에서 모두 이용이 가능하므로, 바이오 및 의학 분야에서 크게 이용될 것으로 기대된다.

본 발명의 한 양태에서, 출발세포로 신경아교세포(neuroglial cell), 지향세포로 신경세포(neuron)를 선택하여 신경아교세포를 신경세포로 전환시키는 경우에, 상기 출발세포 마커 프로모터는 GFAP(Glial fibrillary acidic protein) 프로모터를, 지향세포 마커 프로모터는 SYN(Synapsin) 프로모터를 선택한다. 본 발명의 다른 양태에서, 출발세포로 섬유아세포(fibroblast), 지향세포로 신경세포(neuron)를 선택하여 섬유아세포를 신경세포로 전환시키는 경우에는, 출발세포 마커 프로모터로 Col1A2(Collagen Type I Alpha 2 Chain) 프로모터를 선택한다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로서, 본 발명의 요지에 따라 본 발명의 범위가 이들 실시예에 의해 제한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에게 있어서 자명할 것이다.

HEK293FT(human embryonic kidney 293FT cells)세포에 G glycoprotein이 융합된 수포성 구내염바이러스(fusiogenic envelope G glycoprotein of the vesicular stomatitis virus, VSV-G), GAG/Pol 유전자, 및 발현벡터(expression vector)를 Viafect transfection reagent(E4981, Promega, USA)를 사용하여 도입시켰다. 발현벡터는 출발세포 마커 프로모터, SOX2(Sex determining region Y-box 2), T2A(2A peptide), Cre 재조합효소, 지향세포 마커 프로모터, LoxP, 및 eGFP(enhanced green fluorescent protein)를 순차적으로 전사되도록 배열하였고, eGFP는 역전사 방향으로 디자인하여 LoxP 발현에 의해 전사 방향으로 복위하도록 설계하였다. 상기 발현벡터의 유전자도를 도 1에 나타내었다.
예를 들어 본 발명의 한 양태에서, 출발세포로 신경아교세포(neuroglial cell), 지향세포로 신경세포(neuron)를 선택하여 신경아교세포를 신경세포로 전환시키는 경우에, 상기 출발세포 마커 프로모터는 GFAP(Glial fibrillary acidic protein) 프로모터를, 지향세포 마커 프로모터는 SYN(Synapsin) 프로모터를 선택한다. 본 발명의 다른 양태에서, 출발세포로 섬유아세포(fibroblast), 지향세포로 신경세포(neuron)를 선택하여 섬유아세포를 신경세포로 전환시키는 경우에는, 출발세포 마커 프로모터로 Col1A2(Collagen Type I Alpha 2 Chain) 프로모터를 선택한다.
72시간 후 배지를 걷어 렌티바이랄 벡터를 농축할 수 있는 PEG-it Virus Precipitation Solution(LV810A-1, System Biosciences, USA)으로 72시간 동안 농축한 뒤, EMEM(Eagle's minimum essential medium) 배지에 재부유하여, 본 발명의 직접교차 분화 확인용 벡터인 Advanced In vivo Reprogramming system(A-IVR) 렌티바이랄 벡터를 수득하였다.
실시예 2. 신경아교세포로부터 전환된 신경세포 제조 및 확인
6 well plate에 신경아교세포를 1x105/well 로 EMEM, 10%FBS, 1% 페니실린-스트렙토마이신(PS) 배지로 배양하고, 상기 실시예 1의 방법으로 출발세포를 신경아교세포, 지향세포를 신경세포로 선택하여 제작한 렌티바이랄 벡터(A-IVR)를 10ug/ml의 Polybrene Transfection Reagent(TR-1003-G, Merck Millipore, USA)와 함께 도입하였다. Polybrene은 바이러스와 표적세포의 막사이의 전기적 반발을 중화시켜, 바이러스와 표적세포의 결합력을 증가시킨다. 3일 후 EMEM, 10%FBS, 1ug 퓨로마이신 배지로 교체하고, 일주일간 유전자가 도입되지 않은 세포를 퓨로마이신을 이용하여 선별하였다. 일주일 후 DMEM/F12:neurobasal(2:1), 0.8%N2, 0.4% B27, 1% PS 배지로 교체 후 2일에 한번씩 배지를 교체해 주면서 5주간 신경세포로의 전환을 유도하였다.
5주 후, 세포를 PBS를 사용하여 3번 세척하고, 4% 파라포름알데하이드로 30분간 고정하였다. 이후 0.3% Tween20으로 세 차례 세척 후 10% Normal Donkey Serum을 사용하여 blocking하고, 신경아교세포 마커(GFAP), 및 신경세포 마커(Tuj1, MAP2, Neurofilament)로 면역염색을 진행하였다. 면역염색의 구체적인 프로세스는 선행문헌(Lee HY et al. Tissue Eng Part A. 2015 Jul;21(13-14):2044-52)과 동일하게 진행하였다. 상기 면역염색 결과를 도 2에 나타내었다.
실험 결과, In vitro에서 A-IVR을 사용하여 신경아교세포, 보다 구체적으로는 별아교세포를 신경세포로 전환을 유도할 경우, 신경아교세포 표지 단백질인 GFAP의 발현은 감소되고 신경세포 표지 단백질인 Tuj1, Map2, Neurofilament가 GFP와 함께 발현되는 것으로 나타났다. 이를 통하여 A-IVR에 의하여 신경아교세포가 신경세포로 전환되었으며, GFP역시 신경세포의 분화와 함께 발현됨을 확인할 수 있었다.
실시예 3. 섬유아세포로부터 전환된 신경세포 제조 및 확인
0.1% 젤라틴(gelatin) 코팅된 12 well plate에 미토마이신-C(mitomycin C)를 처리해 성장을 억제시킨 STO feeder cell(세포 성장을 위한 지지체로 사용)을 2.5x105/well 로 DMEM 10% FBS 1% 페니실린-스트렙토마이신(PS) 배지로 하루 배양하고, 다음날 마우스 배아 섬유아세포(Mouse Embryonic Fibroblast; MEF), 또는 인간 섬유아세포(Human Fibroblast)를 4x105/well 로 feeder cell 위에 파종하여, DMEM 혹은 IMDM 10% FBS, 1% P/S로 하루 배양하였다. 이후, 상기 실시예 1의 방법으로 출발세포를 피부세포, 지향세포를 신경세포로 선택하여 제작한 렌티바이랄 벡터(A-IVR)를 10ug/ml의 Polybrene Transfection Reagent(TR-1003-G, Merck Millipore, USA)와 함께 도입하였다. 24시간 후 DMDM/F12, 1% B27, 2mM L-Glutamate, 1% P/S, 20ng/ml FGF, 20ng/ml EGF, 및 2ug/ml 헤파린을 포함하는 배지로 교체 후 6-7일 추가 배양하였다. 이후 작은 집락(colony)이 생기면 accutase(cell detachment solution)를 사용해 세포를 떼어낸 후 6 well plate에서 3일간 추가 배양하고, 생성된 구체(sphere)를 0.1% 젤라틴 코팅된 12 well plate에 이전하여 14일간 배양하며 신경세포로의 전환을 유도하였다.
면역 염색은 실시예 2에 기재된 방법과 동일하게 진행하되, 면역 염색 시의 마커는 섬유아세포 마커(Col1A2), 및 신경세포 마커(Tuj1)를 사용하였다. 상기 면역염색 결과를 세포의 광학 촬영 사진과 함께 도 3과 도 4에 나타내었다.
실험 결과, In vitro에서 A-IVR을 사용하여 섬유아세포를 신경세포로 전환을 유도할 경우, 섬유아세포 표지 단백질인 Col1A2의 발현은 감소되고 신경세포 표지 단백질인 Tuj1이 GFP와 함께 발현되는 것으로 나타났다. 상기 결과는 출발세포로서 섬유아세포를 마우스 배아 섬유아세포를 이용하거나 인간 섬유아세포를 이용하는 경우 모두 동일하였다. 이를 통하여 A-IVR에 의하여 섬유아세포가 신경세포로 전환되었으며, GFP역시 신경세포의 분화와 함께 발현됨을 확인할 수 있었다.
실시예 4. A-IVR 이식된 척수손상 동물모델의 신경 재생 효과 확인
척수손상 동물 모델은 체중 20g 내외의 수컷 C57BL/6마우스를 사용하였다. 복강주사로 케타민과 럼푼을 혼합하여 마취하고, 후궁절제술로 Thoracic Vertebrae 10(흉추10번, T10)부분을 절개하여 척수를 노출시킨 후, 전용 포셉(self-closed forceps, Germany)을 사용하여 3초간 척수를 압박 시킴으로서 척수 손상을 주었다. 이후 근육과 피부를 봉합하였다. 척수손상 동물 모델 제작 2주 후 손상부위의 피부와 근육을 다시 열어, 손상부위에 실시예 1의 방법으로 출발세포를 신경아교세포(보다 구체적으로는 별아교세포), 지향세포를 신경세포로 선택하여 제작한 렌티바이랄 벡터(A-IVR) 4㎕를 33G 헤밀턴 주사기로 이식하였다(n=10). 대조군 마우스에는 A-IVR 대신 EMEM을 이식하였다(n=10). 손상 일주일 후부터 운동기능 검사를 위해 매주 Basso-Mouse-Scale(BMS)을 측정하였다. BMS는 총 9점으로 나뉘어져 있으며 크게 3단계로 구분하는데, 첫번째 단계에서 발목의 움직임을 측정하고, 두번째 단계에서 체중을 싣는 보행 및 걸음의 회복을 측정하며, 마지막 단계에서 걸음의 안정과 꼬리의 회복상태를 측정한다. BMS의 구체적인 프로세스는 선행문헌(Basso DM et al. J Neurotrauma. 2006 May;23(5):635-59)과 동일하게 진행하였다. 상기 BMS 측정 결과를 도 5에 나타내었다. 실험결과, A-IVR를 이식한 2주 후부터 대조군과 비교하여 A-IVR 이식된 마우스의 운동기능이 유의미하게 회복되는 것을 알 수 있었다.
척수손상 후 8주째(A-IVR이식 6주째) 마우스를 희생시키고, 척수 조직을 수득하여 4% 파라포름알데하이드(paraformaldehyde)로 고정한 뒤, 탈수하여 동결 포매를 시행하였다. 포매를 시행한 조직 중 손상부위(흉추10번)을 신경(MAP2), 활성형신경아교세포(GFAP), 신경모세포(SOX2), 초기신경세포(Tuj1), 성숙한 신경세포(neurofilament) 마커로 면역염색하고, 모든 유핵세포(nucleated cell)의 핵은 4'6'diamidino-2- phenylindole(DAPI, 1 ug/ml)(vector,CA,USA)로 대조염색하였다. 면역염색의 구체적인 프로세스는 선행문헌(Lee HL et al. J Control Release. 2016 Mar 28;226:21-34)과 동일하게 진행하였고, 염색 결과는 레이져 스캐닝 공초점 현미경(laser scanning confocal microscopy, LSCM)으로 검경하였다. 상기 염색 결과를 도 6에 나타내었다. 실험 결과, 손상부위를 중심으로 신경아교세포 표지단백질인 GFAP의 발현이 확인되었다. 또한 신경모세포 표지단백질인 Sox2는 GFP와 함께 발현되지 않으나, 초기 신경세포 표지 단백질인 Tuj1과 성숙한 신경세포 표지 단백질인 Neurofilament은 GFP와 함께 발현되는 것으로 나타났다. GFAP는 함께 발현되지 않았다. 이를 통하여 A-IVR에 의해 신경아교세포가 신경세포로 전환(reprogramming)되며, 벡터 도입 6주 후에는 신경모세포를 지나 신경세포로 분화됨을 알 수 있었다.
상기 실시예 1 내지 4의 결과로부터 본 발명의 개선된 생체 내 리프로그래밍 시스템이 세포를 출발세포로부터 지향세포로 전환(reprogramming)시킬 수 있다는 것과, 이를 세포가 살아있는 상태에서 확인 가능하다는 것을 알 수 있었다. 이는 생체 내에서 기타세포로부터 필요세포를 생산 가능하다는 것을 의미한다.
이상으로 본 발명의 특정한 부분을 상세히 기술하였는바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구체적인 기술은 단지 바람직한 구현예일 뿐이며, 이에 본 발명의 범위가 제한되는 것이 아닌 점은 명백하다. 따라서 본 발명의 실질적인 범위는 첨부된 청구항과 그의 등가물에 의하여 정의된다고 할 것이다.
본 발명은 개선된 생체 내 리프로그래밍 시스템 및 이를 이용한 세포 전환 방법에 관한 것으로, 본 발명의 리프로그래밍 시스템은 세포의 전환을 확인하기 위해 세포의 고정을 필요치 않고, 세포가 살아있는 상태에서 실시간으로 모니터링이 가능하며, 생체 외(in vitro) 또는 생체 내(in vivo)에서 모두 이용이 가능하므로, 바이오 및 의학 분야에서 크게 이용될 것으로 기대된다.
Claims (26)
- A세포 마커 프로모터, 유전자 다능성 유지 단백질, Cre 재조합효소, B세포 마커 프로모터, LoxP, 및 형광단백질을 코딩하는 유전자를 순차적으로 포함하는, A세포의 B세포 전환용 발현벡터.
- 제 1항에 있어서,상기 발현벡터는 세포가 살아있는 상태에서 A세포가 B세포로 전환되는 것을 확인 가능한 것인, A세포의 B세포 전환용 발현벡터.
- 제 1항에 있어서,상기 형광단백질을 코딩하는 유전자는 역전사 방향으로 포함되는 것인, A세포의 B세포 전환용 발현벡터.
- 제 1항에 있어서,상기 발현벡터는 아미노산 분리 펩타이드를 추가로 포함하는 것인, A세포의 B세포 전환용 발현벡터.
- 제 4항에 있어서,상기 아미노산 분리 펩타이드는 T2A, F2A, 및 E2A로 구성된 그룹으로부터 선택된 어느 하나 이상인 것인, A세포의 B세포 전환용 발현벡터.
- 제 1항에 있어서,상기 유전자 다능성 유지 단백질은 SOX 유전자족(Sex determining region gene faimly), Myc 유전자족(proto-oncogene gene family), Klf 유전자족(Kruppel-like factors gene family), 및 Oct 유전자족(octamer-binding transcription factor gene family)으로 구성된 그룹으로부터 선택된 어느 하나 이상인 것인, A세포의 B세포 전환용 발현벡터.
- 제 1항에 있어서,상기 A세포는 기질세포이고,상기 B세포는 신경세포인 것인, A세포의 B세포 전환용 발현벡터.
- 제 7항에 있어서,상기 기질세포는 섬유아세포(fibroblast), 연골아세포(chondroblast), 골아세포(osteoblast), 신경아교세포(neuroglial cell), 지방세포(adipocyte), 대식세포(macrophage), 및 플라즈마세포(plasma cell)로 구성된 그룹으로부터 선택되는 어느 하나인 것인, A세포의 B세포 전환용 발현벡터.
- 제 7항에 있어서,상기 A세포 마커 프로모터는 Col1A2(Collagen Type I Alpha 2 Chain), FAP(Fibroblast-activation protein), FSP1(Fibroblast-specific protein 1), 비멘틴(Vimentin), ACTA(alpha smooth muscle Actin), Hsp47(Heat shock protein 47), 아그레칸(Aggrecan), CD44, CD45, CD73, Calcitonin, OSCAR(osteoclast-associated receptor), RANK(Receptor activator of nuclear factor κ B), GFAP(Glial fibrillary acidic protein), TREM2(Triggering receptor expressed on myeloid cells 2), HexB(Beta-hexosaminidase subunit beta), S100(calcium-binding protein), CD69, 및 Gpr34(Probable G-protein coupled receptor 34)로 구성된 그룹으로부터 선택되는 어느 하나의 프로모터인 것인, A세포의 B세포 전환용 발현벡터.
- 제 7항에 있어서,상기 B세포 마커 프로모터는 SYN(Synapsin), Tuj1(Neuron-specific class III beta-tubulin), MAP2(Microtubule-associated protein 2), 및 신경미세섬유(Neurofilament)으로 구성된 그룹으로부터 선택되는 어느 하나의 프로모터인 것인, A세포의 B세포 전환용 발현벡터.
- 제 1항의 발현벡터를 유효성분으로 포함하는, 신경 손상 치료용 약학조성물.
- 제 1항의 발현벡터, G glycoprotein이 융합된 수포성 구내염바이러스(fusiogenic envelope G glycoprotein of the vesicular stomatitis virus, VSV-G), 및 GAG/Pol 유전자를 포함하는, 렌티바이럴 벡터.
- 제 1항의 발현벡터를 포함하는 인간을 제외한 형질전환체.
- (a) 제 1항의 발현벡터를 제조하는 단계; 및,(b) 상기 발현벡터를 A세포에 도입하는 단계;를 포함하는, A세포를 B세포로 전환시키는 방법.
- 제 14항에 있어서,상기 (b)단계 이후에 세포를 B세포 배양용 배지로 배양하는 단계를 추가로 포함하는, A세포를 B세포로 전환시키는 방법.
- 제 14항에 있어서,상기 (b)단계 이후에 세포 내 형광 발현을 확인하는 단계를 추가로 포함하는, A세포를 B세포로 전환시키는 방법.
- 세포가 살아있는 상태에서 A세포가 B세포로 전환되는 것을 확인하는 방법으로서,(a) 제 1항의 발현벡터를 제조하는 단계; 및,(b) 상기 발현벡터를 A세포에 도입하는 단계;를 포함하는, 생세포 이미징(live cell imaging) 방법.
- 제 17항에 있어서,상기 이미징은 형광 현미경을 이용하는 것을 특징으로 하는, 생세포 이미징 방법.
- (a) 제 1항의 발현벡터를 제조하는 단계; 및,(b) 인간을 제외한 개체에 상기 발현벡터를 도입하는 단계;를 포함하는, A세포가 B세포로 전환된 동물모델의 제조방법.
- (a) 인간을 제외한 질병 동물 모델 제 1 개체와 제 2개체를 준비하는 단계;(b) (가)세포를 A세포, (다)세포를 B세포로 하는 제 1항의 발현벡터 Ⅰ을 제조하는 단계;(c) (나)세포를 A세포, (다)세포를 B세포로 하는 제 1항의 발현벡터 Ⅱ를 제조하는 단계;(d) 상기 제 1개체에 발현벡터 Ⅰ을 도입하고, 상기 제 2 개체에 발현벡터 Ⅱ를 도입하는 단계; 및,(e) 상기 제 1개체와 제 2 개체의 질병 치료 효과를 비교하여 제 1개체의 치료 효과가 더 좋을 경우에 상기 (가)세포를 (다)세포 전환용 세포로 선별하는 단계;를 포함하는, B세포 전환용 A세포의 스크리닝 방법.
