지방 생성(adipogenesis), 지방 축적은 대사증후군 또는 자가면역반응으로 인해 유발되며, 과도한 지방대사를 일으켜 지방조직이 증가하는 현상을 초래한다. 여러 질환에서의 지방이 축적되는 증상을 완화시키기 위한 치료법으로는 식이요법 또는 약물치료가 시행되고 있다. 그러나, 현재까지 지방축적을 근본적으로 치료할 수 있는 방법이 개발되지 않은 실정이다.
따라서, 지방 생성 및 지방 축적을 근본적으로 치료하기 위한 안전하고도 효과적인 새로운 치료제의 개발이 필요한 실정이다.
일 양상은 PRL-1(phosphatase of regenerating liver-1) 을 분비하거나 모세포에 비하여 PRL-1을 과발현하는 중간엽 줄기세포를 제공하는 것이다.
다른 양상은 PRL-1(phosphatase of regenerating liver -1)을 분비하거나 모세포에 비하여 PRL-1을 과발현하는 중간엽 줄기세포를 포함하는 비정상적인 지방 생성 및 대사 이상질환의 예방 또는 치료를 위한 약학적 조성물을 제공하는 것이다.
다른 양상은 PRL-1(phosphatase of regenerating liver -1)을 분비하거나 모세포에 비하여 PRL-1을 과발현하는 중간엽 줄기세포를 포함하는 비정상적인 지방 생성 및 대사 이상질환의 개선 또는 예방을 위한 건강기능식품을 제공하는 것이다.

본 명세서에서 용어 "중간엽 줄기세포(줄기세포(Mesenchymal Stem Cell: MSC)"는 자기재생능력(self-renewal)과 줄기세포능(stemness maintenance)을 유지하고 다양한 간엽 조직으로 분화할 수 있는 세포를 의미할 수 있고, 포유류, 예를 들면 인간을 포함한 동물의 중간엽 줄기세포를 포함할 수 있다. 또한, 상기 중간엽 줄기세포는 제대 유래, 제대혈 유래, 골수 유래, 태반(placenta) 유래, 또는 지방 유래 중간엽 줄기세포인 것일 수 있다. 상기 태반 유래 중간엽 줄기세포는 태반을 구성하는 다양한 조직, 예를 들어 양막 상피 세포, 양막, 영양막, 융모막등의 조직으로부터 유래될 수 있다. 바람직하게는 상기 태반 유래 중간엽 줄기세포는 태반의 융모막판(chorionic plate)으로부터 유래된 중간엽 줄기세포일 수 있으며, 더욱 바람직하게는 융모막판막(chorionic plate membrane)으로부터 유래된 중간엽 줄기세포일 수 있다. 중간엽 줄기세포의 분리는 통상의 당업자에게 자명한 방법으로 수행될 수 있으며, 예를 들면, Pittenger 등(Science 284: 143, 1997)와 van 등(J. Clin. Invest., 58: 699, 1976)의 문헌들에 개시되어 있다. 상기 중간엽 줄기세포는 PRL-1(phosphatase of regenerating liver -1) 또는 그의 활성 단편을 분비하거나, PRL-1 또는 그의 활성 단편을 발현하는 조작된 것일 수 있다.
상기 중간엽 줄기세포는 하기 a) 또는 b)의 특성을 가질 수 있다:
a) PRL-1(phosphatase of regenerating liver -1)을 발현하는 특성;
b) CD34, CD105, HLA-ABC, 및 HLA-G로 이루어진 군으로부터 선택되는 하나 이상의 표면 항원 특성.
상기 중간엽 줄기세포는 CD34, CD105, HLA-ABC, 또는 HLA-G를 발현하는 것일 수 있다. 상세하게는, 본 명세서에서 제공되는 중간엽 줄기세포는 세포 표면에 발현되는 세포 표지자에 대하여 CD34, CD105, HLA-ABC, 또는 HLA-G 양성 표면 마커를 적어도 약 20%, 25%, 30%, 35%, 40%, 45%, 50%, 55%, 60%, 65%, 70%, 75%, 80%, 85%, 90%, 95%, 98% 또는 약 99% 발현하는 것일 수 있다. 본 발명에서 용어, "양성"은 줄기세포 표지와 관련하여, 그 표지가 기준이 되는 다른 비줄기 세포와 비교하였을 때 더 많은 양, 또는 더 높은 농도로 존재하는 것을 의미할 수 있다. 즉, 세포는 어느 표지가 세포 내부 또는 표면에 존재하기 때문에 그 표지를 이용하여 그 세포를 하나 이상의 다른 세포 유형과 구별할 수 있으면 그 표지에 대하여 양성이 된다. 또한 세포가 배경 값보다 더 큰 값으로 신호, 예를 들어 세포 측정 장치의 신호를 낼 수 있는 만큼의 양으로 그 표지를 가지고 있다는 것을 의미할 수 있다. 예를 들어 세포를 CD90에 특이적인 항체로 검출 가능하게 표지할 수 있고, 이 항체로부터의 신호가 대조군(예를 들어 배경 값)보다 검출 가능하게 더 크면 그 세포는 "CD90+"이다. 본 명세서에서 용어, "음성"은 특정 세포 표면 표지에 특이적인 항체를 사용하여도 배경 값에 비교하여 그 표지를 검출할 수 없음을 뜻한다. 예를 들어 CD45에 특이적인 항체로 세포를 검출 가능하게 표지할 수 없으면 그 세포는 "CD45-"이다.
상기 중간엽 줄기세포는 대안적으로 그의 배양물, 용해물 또는 그의 추출물이 이용될 수 있다. 상기 배양물, 용해물 또는 추출물은 세포 그대로를 이용하기 어려운 경우 유용한 대안이 될 수 있으며, 단백질 등을 포함한 세포의 구성 성분을 포함하고 있으므로 본래 세포와 유사하거나 동등한 생물학적 활성을 나타낼 수 있다. 상기 용해물 또는 추출물은 상업적으로 이용가능한 세포 용해 키트, 또는 추출 키트 등을 이용하여 얻을 수 있다.
본 명세서에서 용어 "PRL-1(phosphatase of regenerating liver -1)"은 인간을 포함한 척추동물, 예를 들면, 포유류, 어류, 양서류, 조류, 또는 파충류 유래의 PRL-1을 포함할 수 있다. 또한, 상기 PRL-1은 PRL-1의 전구체도 포함하는 의미일 수 있다.

본 명세서에서 용어 "비정상적인 지방 생성 및 대사 이상질환"은 지방의 대사에 이상이 생겨 발생하는 질병을 의미할 수 있다. 상기 비정상적인 지방 생성 및 대사 이상질환의 예로는 비만, 당뇨, 이상지질혈증, 대사질환, 고혈압, 갑상선 안질환 또는 비정상적인 지방생성 및 대사질환성 퇴행성 질환을 포함할 수 있다.
본 명세서에서 용어 "갑상선 안질환(Thyroid associated ophthalmopathy: TAO)"은 갑상선 호르몬의 과다 분비로 인한 갑상선 기능 항진증으로 인해 발생하는 안와염증질환을 의미한다. 눈꺼풀 후퇴, 안구 돌출, 제한성 사기, 시력 저하, 복시, 시야 감소 등의 병증을 나타낼 수 있다.
용어 "치료"는 질환, 장애 또는 병태, 또는 그의 하나 이상의 증상의 경감, 진행 억제 또는 예방을 지칭하거나, 그를 포함하며, "유효성분" 또는 "약제학적 유효량"은 질환, 장애 또는 병태, 또는 그의 하나 이상의 증상의 경감, 진행 억제 또는 예방에 충분한 본원에서 제공되는 발명을 실시하는 과정에서 이용되는 조성물의 임의의 양을 의미할 수 있다.
용어, "투여하는," "도입하는" 및 "이식하는"은 상호교환적으로 사용되고 일 구체예에 따른 조성물의 원하는 부위로의 적어도 부분적 국소화를 초래하는 방법 또는 경로에 의한 개체내로의 일 구체예에 따른 조성물의 배치를 의미할 수 있다. 일 구체예에 따른 조성물의 세포 또는 세포 성분의 적어도 일부를 생존하는 개체내에서 원하는 위치로 전달하는 임의의 적절한 경로에 의해 투여될 수 있다. 개체 투여 후 세포의 생존 기간은 짧으면 수 시간, 예를 들면 24시간 내지 수일 내지 길면 수년일 수 있다.
상기 조성물은 조성물 총 중량에 대하여 0.001 중량% 내지 80 중량%의 PRL-1 또는 그의 활성 단편을 포함할 수 있다. 또한, PRL-1 또는 그의 활성 단편의 투여 용량은 0.01mg 내지 10,000mg, 0.1mg 내지 1000mg, 1mg 내지 100mg, 0.01mg 내지 1000mg, 0.01mg 내지 100mg, 0.01mg 내지 10mg, 또는 0.01mg 내지 1mg일 수 있다. 또한, 중간엽줄기세포의 투여 용량은 1.0Х 105 내지 1.0Х 108 세포/kg(체중)일 수 있다. 다만, 투여량은 제제화 방법, 투여 방식, 환자의 연령, 체중, 성별, 병적 상태, 음식, 투여 시간, 투여 경로, 배설 속도 및 반응 감응성과 같은 요인들에 의해 다양하게 처방될 수 있고, 당업자라면 이러한 요인들을 고려하여 투여량을 적절히 조절할 수 있다. 투여 횟수는 1회 또는 임상적으로 용인가능한 부작용의 범위 내에서 2회 이상이 가능하고, 투여 부위에 대해서도 1개소 또는 2개소 이상에 투여할 수 있다. 인간 이외의 동물에 대해서도, kg당 인간과 동일한 투여량으로 하거나, 또는 예를 들면 목적의 동물과 인간과의 기관(심장 등)의 용적비(예를 들면, 평균값) 등으로 상기의 투여량을 환산한 양을 투여할 수 있다. 가능한 투여 경로에는 경구, 설하, 비경구 (예를 들어, 피하, 근육내, 동맥내, 복강내, 경막내, 또는 정맥내), 직장, 국소 (경피 포함), 흡입, 및 주사, 또는 이식성 장치 또는 물질의 삽입을 포함할 수 있다. 일 구체예에 따른 치료의 대상동물로서는, 인간 및 그 밖의 목적으로 하는 포유동물을 예로 들 수 있고, 구체적으로는 인간, 원숭이, 마우스, 래트, 토끼, 양, 소, 개, 말, 돼지 등이 포함된다.
일 구체예에 따른 약학적 조성물은 약학적으로 허용가능한 담체 및/또는 첨가물을 포함할 수 있다. 예를 들어, 멸균수, 생리식염수, 관용의 완충제(인산, 구연산, 그 밖의 유기산 등), 안정제, 염, 산화방지제(아스코르브산 등), 계면활성제, 현탁제, 등장화제, 또는 보존제 등을 포함할 수 있다. 국소 투여를 위해, 생체고분자(biopolymer) 등의 유기물, 하이드록시아파타이트 등의 무기물, 구체적으로는 콜라겐 매트릭스, 폴리락트산중합체 또는 공중합체, 폴리에틸렌글리콜중합체 또는 공중합체 및 그의 화학적 유도체 등과 조합시키는 것도 포함할 수 있다. 일 구체예에 따른 약학적 조성물이 주사에 적당한 제형으로 조제되는 경우에는, 중간엽줄기세포 또는 PRL-1가 약학적으로 허용가능한담체 중에 용해되어 있거나 또는 용해되어 있는 용액상태로 동결된 것일 수 있다.
일 구체예에 따른 약학적 조성물은 그 투여방법이나 제형에 따라 필요한 경우, 현탁제, 용해보조제, 안정화제, 등장화제, 보존제, 흡착방지제, 계면활성화제, 희석제, 부형제, pH 조정제, 무통화제, 완충제, 환원제, 산화방지제 등을 적절히 포함할 수 있다. 상기에 예시된 것들을 비롯하여 본 발명에 적합한 약학적으로 허용되는 담체 및 제제는 문헌[Remington's Pharmaceutical Sciences, 19th ed., 1995]에 상세히 기재되어 있다. 일 구체예에 따른 약학적 조성물은 당해 발명이 속하는 기술 분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있는 방법에 따라, 약학적으로 허용되는 담체 및/또는 부형제를 이용하여 제제화함으로써 단위 용량 형태로 제조되거나 또는 다용량 용기 내에 내입시켜 제조될 수 있다. 이때 제형은 오일 또는 수성 매질 중의 용액, 현탁액 또는 유화액 형태이거나 분말, 과립, 정제 또는 캡슐 형태일 수 있다.
또한, 상기 건강기능식품은 상기 건강식품 조성물은 PRL-1 또는 그의 활성 단편 이외에 다른 식품 또는 식품 성분과 함께 사용될 수 있고, 통상적인 방법에 따라 적절하게 사용될 수 있다. 유효 성분의 혼합양은 사용 목적 (예방, 건강 또는 치료적 처치)에 따라 적합하게 결정될 수 있다. 일반적으로, 건강기능식품의 제조시에 본 명세서의 조성물은 원료에 대하여 15 중량부 이하의 양으로 첨가될 수 있다. 상기 건강기능식품의 종류에는 특별한 제한은 없다.
또 다른 양상은 PRL-1(phosphatase of regenerating liver-1)을 분비하거나 모세포에 비하여 PRL-1을 과발현하도록 배양 환경적 또는 유전적으로 조작된 중간엽 줄기세포를 인 비보(in vivo) 상에서 실험동물에 투여하거나, 또는 인 비트로(in vitro) 상에서 세포에 접촉시키는 단계를 포함하는 지방 축적을 억제시키는 방법을 제공한다.
상기 지방 축적을 억제시키는 방법에 있어서, 상기 접촉은 중간엽 줄기세포, PRL-1 또는 그의 활성 단편을 개체에 처리, 또는 접촉시키는 단계를 의미할 수 있다. 상기 처리 또는 접촉은 중간엽 줄기세포를 세포와 공배양하거나, 또는 중간엽 줄기세포, PRL-1 또는 그의 활성 단편을 실험 동물, 예를 들면 동물의 특정 장기 또는 안와조직 주변으로 국소이식 (Direct local injection), 정맥혈관을 통한 이식 (intravenous injection, i.v. injection) 등에 투여하는 것을 포함할 수 있다. 또한, 상기 처리 또는 접촉은 PRL-1 또는 그의 활성 단편을 코딩하는 뉴클레오티드 서열을 포함하는 숙주 세포와 세포와 접촉시키거나, 예를 들면, 공배양하거나, 또는 상기 숙주 세포를 실험 동물, 예를 들면, 동물의 특정 장기 또는 안와조직 주변으로 국소이식 (Direct local injection), 정맥혈관을 통한 이식 (intravenous injection, i.v. injection) 등에 투여하는 것을 포함할 수 있다. 안과질환 치료용으로 이상적인 투여 방법은 안와조직 주변으로의 국소이식 (local injection) 등이 효율적일 수 있다. 상기 공배양 및 투여는 통상의 당업자가 목적하는 효과, 예를 들면, 중간엽 줄기세포 또는 PRL-1 또는 그의 활성 단편이 개체에 영향을 미칠 수 있도록 하는 방법 및 기간 동안 수행될 수 있다.
일 양상에 따른 중간엽 줄기세포, 또는 이를 포함하는 조성물은 지방 축적 또는 지방 조직의 증가를 억제시켜 지방대사 이상과 관련된 질환의 예방, 치료 또는 개선에 유용하게 사용될 수 있다.

참고예 1. 안와섬유아세포(Orbital fibroblast) 배양 및 처리
사람으로부터 안와섬유아세포(정상 4명, 환자 4명)를 채취하여 DMEMF12(Gibco)(10% FBS, 및 1% 페니실린-스트렙토마이신)에 배양하였다. 섬유아세포가 배지에 분포되어 자라고 2일 후, DMEM(10% FBS)에서 5 μg/ml 인슐린, 1 mM 덱사메타손, 및 0.5 mM IBMX을 첨가하여 지방 세포로의 분화가 시작되게 하였다(day 0). 72시간 후(day 3), 배지를 10% FBS 및 5 μg/ml 인슐린이 보충된 DMEM 배지로 교체하고, 그 후 격일마다 10% FBS가 보충된 DMEM 배지를 공급하였다.
참고예 2. Real-time PCR
배양 8 일차(day 8)에, RPL-1이 과발현된 태반 유래 중간엽줄기세포(2 x 105)를 안와섬유아세포와 함께 48 시간 동안 공배양하였다. 세포 용해물은TRIzol(Invitrogen, Carlbad, CA, USA)에서 균질화하여, RNA를 추출하였다. 각각의 시료로부터의 1 μg의 총 RNA를 역전사하여 cDNA를 합성하였다. cDNA합성 조건은 아래와 같다: RNA 융해(65℃, 1분), 풀림 (25℃, 5분), 증폭(42℃, 60분) 및 효소 불활성 (85℃, 1분).
아래의 PCR 조건에 따라 각 유전자의 mRNA 발현을 증폭하여 표준화(normalization)하였다: 초기 융해(95℃, 2분), 증폭(95℃, 10초; 55℃, 20초; 및 72℃, 20초) 40 사이클. PPARγ, ADIPONECTIN 및 C/EBPα, 프라이머 세트는 하기 표 1과 같다.

각 유전자의 mRNA의 발현은 18s rRNA로 표준화하였다. 데이터는 정상 집단과 비교하여 지방분화 관련 인자의 배수(fold)(평균±SEM)로 표현하였다.
참고예 3. 웨스턴 블롯
RIPA 완충액을 이용하여 용해물을 준비하였다. 동량의 총 단백질을 SDS-PAGE로 분리하고 막에 옮겼다. 상기 막은 항-HAS1 및 HAS2(SantaCruz Biotechnology, SA, USA)로 1:1000 희석에서 면역블롯팅하였고, 같은 막을 GAPDH(SantaCruz)와 함께 배양하였다. 세척 후, 홀스래디쉬 과산화 효소-접합 항-염소 IgG(horseradish peroxidase-conjugated anti-goat IgG) 2차 항체와 함께 1:10000 희석에서 상온에서 3시간 배양하였다. 면역 반응 밴드는 향상된 화학 발광 솔루션(enhanced chemiluminescence solution)(Animal Genetics, Suwon, Korea)으로 이미지화하였고, ChemiDocTM XRS+ System Imager (Bio-Rad Laboratories, Hercules, CA, USA)로 검출하였다. 단백질 발현량은 GAPDH로 표준화(normalization)하였다. 데이터는 정상 집단과 비교하여 HAS2의 배수(fold)(평균±SEM)로 표현하였다.
참고예 4. FACS 분석
인간 섬유아세포(3 x 105)를 세포 해리 버퍼(Life Technologies)로 해리시키고 PBS(2%(v/v) FBS)로 세척하였다. 이를 아이소타입 대조군 IgG(isotype control IgG) 또는 항원-특이적 항체(BD Biosciences, CA, USA)와 함께 20분간 배양하고, 세포를 확인하는데 이용하였다. FACS 분류(sorting)는 FACS vantage Flow Cytometer (BD Biosciences, CA, USA)를 이용하여 수행하였다.
실시예 1. 기능 강화된 중간엽 줄기세포의 제작
1.1. 태반 유래 중간엽 줄기세포의 분리
정상적으로 분만한 건강한 산모로부터 사전에 충분한 설명에 근거한 동의(informed consent)를 받고, 정상 태반 분만 시에 수집된 태반조직으로부터 분리하였다. 분리한 조직 (chorioamniotic membranes) 을 50 ml tube에 넣고 DPBS를 첨가해 여분의 혈액을 제거 후 20 ml 효소 용액 I (1mg/ml collagenase type I, 2 mg/ml Trypsin, 20 mg/ml DNase I, 1.2 U/ml Dispase, x1 PS in HBSS) 에서 멸균된 슬라이드 글라스로 융모양막(chorioamniotic membranes)의 안쪽 부분을 긁어 회수되는 부유물들을 한쪽으로 모아주었다. 효소 용액 I 10ml을 넣고 고르게 섞어준 후 37℃ 효소반응을 15분간씩 2회 반복하면서, 조직으로부터 줄기세포를 분리하였다. 분리된 세포 현탁액을 원심분리하고, 분리된 세포는 10 %의 소 태아 혈청, 1 %의 페니실린-스트렙토마이신, 1 ug/ml의 헤파린, 및 25 ng/ml의 섬유아세포성장인자(Fibroblast Growth Factor-4, FGF-4)가 첨가된 DMEM/F12를 이용하여 배양하였으며, 이후 4일내지 5일 간격으로 배양배지를 교환하였으며, 첫 계대에서 Invitrogen사의 TrypLE를 37℃인큐베이터에서 단기간 (3분) 처리하여 계대 배양하였다.
1.2. PRL-1 유전자의 발현이 강화된 태반 유래 중간엽 줄기세포 제작
상기 1.1.에서 분리한 태반 유래 중간엽 줄기세포에서 PRL-1 유전자의 발현을 강화시키기 위해서, 전기천공법을 이용하였다.
구체적으로, 중간엽 줄기세포에 도 1의 구조를 갖는 유전자를 전기천공법을 이용해 도입하였다.
태반 융모막판막-유래 중간엽 줄기세포(CP-MSCs)를 10 % 우태아혈청(Fetal Bovine Serum, FBS), 1 % 페니실린-스트렙토마이신, 25 ng/ml 섬유아세포성장인자4 (Fibroblast Growth Factor-4, FGF-4), 1 ug/ml Heparin이 첨가된 MEM-alpha 배지 중에서 37℃, CO2 인큐베이터 중에서 배양하였다. 세포를 인산완충식염수(Phosphatase buffered saline, PBS)로 세척한 후, 37℃에서 2분간 트립신을 처리하여 세포를 떼어낸 후, PBS를 이용하여 떼어낸 세포를 모아 1200 rpm에서 5분간 원심분리하였다. 얻어진 펠렛에 배양액 1ml을 넣어 현탁시킨 후 혈색소계(hemocytometer)를 이용하여 계수하였다. 회수한 세포를 200 g에서 10 분간 원심분리하여 Nucleofector를 넣어 세포(5x105/100ul)를 현탁시킨 후 PRL-1을 포함하고 있는 DNA 플라스미드 2 ug을 넣었다. 세포를 큐벳에 옮겨 Nucelofection 기계에 넣어 'U-23' 프로그램을 작동시켰다. 프로그램이 종료되면 신속히 새로운 배지를 포함하고 있는 배양접시를 37℃, CO2 인큐베이터 중에서 안정화시켰다. 4시간 후, 배양 접시의 배지를 흡입기를 이용하여 제거한 후, 부착된 세포는 1.5 mg/ml 네오마이신(Neomycin)을 포함하는 배지(10 % 우태아혈청 (Fetal Bovine Serum, FBS), 1 % 페니실린-스트렙토마이신, 25 ng/ml 섬유아세포성장인자 4 (Fibroblast Growth Factor-4, FGF-4), 1 ug/ml Heparin이 첨가된 MEM-alpha 배지에서 배양하여 PRL-1 유전자가 형질도입된 태반 융모막판막-유래 중간엽 줄기세포를 얻었다.
도 2는 태반 유래 중간엽 줄기세포에 PRL-1 유전자가 도입되었는지 GFP를 이용해서 확인한 현미경 사진이다.
도 3은 PRL-1 유전자가 도입된 태반 유래 중간엽 줄기세포의 수(GFP+ cells)를 확인한 결과를 나타낸 그래프이다.
도 2 및 도 3에 나타낸 바와 같이, PRL-1 유전자를 도입하지 않은 대조군인 naive 태반 유래 중간엽 줄기세포에 비해서, 1.1.에서 제조된 강화 태반 유래 중간엽 줄기세포에서 인위적으로 도입된 PRL-1 유전자의 발현이 유의적으로 증가하였음을 확인하였다.
따라서, 상기 1.1.의 방법에 의해 PRL-1 유전자의 발현이 강화된 태반 유래 중간엽 줄기세포가 성공적으로 제조되었음을 확인하였다.
1.3. PRL-1 유전자의 발현이 강화된 태반 유래 중간엽 줄기세포의 특성 분석
1.1. 및 1.2.에서 확인한 중간엽 줄기세포의 특성을 분석하기 위해, 사이토카인 분비 특성 및 표면 항원 특성을 분석하였다. 구체적으로, RT-PCR 분석을 통해 상기 태반 유래 중간엽 줄기세포에서 PRL-1의 발현 수준을 확인하였고, 그 결과를 도 4에 나타내었다.
도 4는 세포가 발현하는 RPL-1의 mRNA 발현 수준을 나타낸다. 도 4에 나타낸 바와 같이, RT-PCR 분석 결과 1.1.에서 제조한 태반 유래 중간엽 줄기세포(p1, p6)가 naive 세포에 비해서 PRL-1의 mRNA 발현 수준이 유의적으로 높음을 확인하였다.
또한, RT-PCR 분석을 통해 태반 유래 중간엽 줄기세포에서 표면 항원(Oct4, Nanog, Sox2, HLA-G, TERT)의 발현 수준을 확인하였고, 그 결과를 도 5에 나타내었다.
또한, ELISA 분석을 통해 태반 유래 중간엽 줄기세포에서 표면 항원(Oct4, HLA-G)의 발현 수준을 확인하였고, 그 결과를 도 6에 나타내었다.
도 5 및 6에 나타낸 바와 같이, RT-PCR 및 ELISA 분석 결과 1.1.에서 제조한 태반 유래 중간엽 줄기세포(PRL1+)가 줄기세포 마커인 Oct4, Nanog, Sox2, HLA-G, TERT를 발현하는 것을 확인하였다.
또한, 세포배양과정 동안 세포수의 증가를 계수하여 세포의 수가 배가되는 시간(doubling time)을 확인하였고, 그 결과를 도 7에 나타내었다. 도 7에 나타낸 바와 같이, PRL-1 이 과발현된 세포의 계대배양과정 동안 배가 시간은 PRL-1의 발현을 유도하지 않은 대조군과 큰 차이가 없음을 확인 하였다.
또한, FACS 분석을 통해 상기 태반 유래 중간엽 줄기세포의 표면 항원(CD34, CD13, CD90, CD105, HLA-DR, HLA-ABC, 및 HLA-G) 특성을 분석하였고, 그 결과를 도 8 및 9에 나타내었다. 도 8 및 9에 나타낸 바와 같이, 중간엽 세포 마커인 CD34, CD105, HLA-ABC, 및 HLA-G가 양성으로 확인되었다.
따라서, PRL-1의 발현이 강화되어 PRL-1을 발현, 포함하는 태반 유래 중간엽 줄기세포가 줄기세포의 특성을 유지하는 것을 확인하였다.
또한, PRL-1의 별현이 강화된 태반 유래 중간엽 줄기세포를 골 분화 및 지방세포로의 분화를 유도했을 때, 표면 항원 마커 및 골세포, 지방세포 마커의 발현을 확인한 결과를 도 10에 나타내었다.
도 10에 나타낸 바와 같이, 골 분화 또는 지방세포로 분화를 유도하였을 때, 미분화 마커인 Oct4의 발현이 감소되면서, 골세포 및 지방세포의 마커인 osteocalsin (OC), type I collagen (Col 1), Adipsin, PPAR-r 등의 유전자의 발현이 증가됨을 확인함으로써, 골세포 및 지방세포로의 분화가 잘 이루어진다는 것을 확인하였다.
실시예 3. 지방 생성 억제의 확인
상기 제조한 PRL-1의 발현이 강화된 태반 유래 중간엽 줄기세포가 안와섬유아세포의 지방 생성 억제 효과가 있는지 평가하였다. 구체적으로, 정상인과 갑상선 안질환(Thyroid associated ophthalmopathy: TAO)을 갖는 개체로부터 분리한 안와섬유아세포(Orbital fibroblast)에 지방생성유도 분화 배지를 넣은 후 상기 제조한 PRL-1의 발현이 강화된 태반 유래 중간엽 줄기세포(CP-PRL-1)와 공배양 유무를 나눠 10일간 배양하였다 (초기 4 일동안 DMEM supplemented with 10% FBS, 33 uMbiotin, 17 uM pantothenic acid, 0.2 nM T3, 10 μg/mL transferrin, 0.2 uM prostaglandin I2, 0.1mM isobutylmethylxanthine (IBMX), 1 uM dexamethasone, 5 ug/ml insulin / 5-10일 동안 w/o IBMX, Dexamethaxone, Insulin). 10일 뒤 Oil-Red-O 염색을 통해 섬유아세포의 지질축적 변화를 관찰하였다. 관찰 결과를 도 11에 나타내었다. OF: 정상인; TAO: 갑상선 안질환을 갖는 개체; CP(-): 공배양 하지 않음; CP Naive (+): 태반 유래 중간엽 줄기세포와 공배양; CP-PRL-1 (+): CP-PRL-1과 공배양.
그 결과, 도 11에 나타낸 바와 같이 분화 배지에 유도된 TAO 환자의 섬유아세포의 지질 축적이 CP-PRL-1과의 공배양을 통해 유의적으로 감소됨을 확인하였다.
실시예 4. 지방 생성 관련 유전자의 조절 능력 확인
상기 제조한 PRL-1의 발현이 강화된 태반 유래 중간엽 줄기세포가 안와섬유아세포의 지방 생성 관련 유전자의 조절 능력이 있는지 평가하기 위해 CP-PRL-1와 공배양 하지 않거나(CP-), naive 상태의 중간엽 줄기세포와 공배양 하거나(CP Naive (+)), CP-PRL-1와 공배양 하고(CP-PRL-1(+)) 이들 섬유아세포에서 발현되는 지방 생성 관련 유전자(Adipsin, Adiponectin, PPARγ, Leptin, LPL, FABP4)의 mRNA 수준을 qRT-PCR로 분석하였다. 그 결과를 도 12에 나타내었다. OF: 정상인; TAO: 갑상선 안질환을 갖는 개체.
도 12에 나타낸 바와 같이, 중간엽 줄기세포와 공배양 한 안와섬유아세포에서 지방 생성 관련 유전자 모두의 발현 수준이 감소하였다. 특히, CP-PRL-1와 공배양 한 경우 naive 중간엽 줄기세포와 공배양한 경우에 비해서 지방 생성 관련 유전자의 발현 수준이 유의적으로 감소됨을 확인하였다.
추가적으로, 상기 실험군에서 지방 생성 관련 인자(Leptin, PPARγ) 및 염증 관련 인자 TNF-α의 유전자 발현 수준을 qRT-PCR로 확인하였고, 이들의 단백질 발현 수준을 웨스턴 블롯으로 확인하였다. 그 결과를 도 13에 나타내었다. TAO: 갑상선 안질환을 갖는 개체.
도 13에 나타낸 바와 같이, CP-PRL-1와 공배양 한 경우 naive 중간엽줄기세포와 공배양 한 경우에 비해서 지방 생성 관련 인자 및 염증 관련 인자의 유전자 및 단백질 발현 수준이 유의적으로 감소됨을 확인하였다.
따라서, PRL-1의 발현이 강화된 태반 유래 중간엽 줄기세포는 지방 생성을 억제하는 우수한 효과가 있으므로, 지방 생성 및/또는 지방 축적 관련 질환의 치료제로서 유용하게 사용될 수 있음을 알 수 있다.

- PRL-1(phosphatase of regenerating liver-1)을 분비하거나 모세포에 비하여 PRL-1을 과발현하는 중간엽 줄기세포.
- 청구항 1에 있어서, 상기 PRL-1은 미세주입법(microinjection), 전기천공법(electroporation), DEAE-덱스트란처리 형질감염법, 리포펙션(lipofection), 나노파티클-매개 형질주입법, 단백질 전달 도메인 매개 도입법, 인산칼슘(CaPO4) 형질 감염법, 바이러스-매개유전자전달법및 PEG-매개 형질주입법으로 이루어진 군에서 선택되는 방법에 의해 전달되어 발현되는 것인 중간엽 줄기세포.
- 청구항 1에 있어서, 상기 중간엽 줄기세포는 제대 유래, 제대혈 유래, 골수 유래, 태반 유래, 또는 지방 유래 중간엽 줄기세포인 것인 중간엽 줄기세포.

- 청구항 4에 있어서, 상기 중간엽 줄기세포는 제대 유래, 제대혈 유래, 골수 유래, 태반 유래, 또는 지방 유래 중간엽 줄기세포인 것인 약학적 조성물.
- PRL-1(phosphatase of regenerating liver-1을 분비하거나 모세포에 비하여 PRL-1을 과발현하는 중간엽 줄기세포를 포함하는 비정상적인 지방 생성 및 대사 이상질환의 개선 또는 예방을 위한 건강기능식품.
- PRL-1(phosphatase of regenerating liver-1)을 분비하거나 모세포에 비하여 PRL-1을 과발현하는 중간엽 줄기세포를 인 비보(in vivo) 상에서 실험동물에 투여하거나, 또는 인 비트로(in vitro) 상에서 세포에 접촉시키는 단계를 포함하는 지방 축적을 억제시키는 방법.
