Abstract
[要約]アテローム型の動脈硬化を防ぐために、食事によるRLP-C濃度の上昇を抑制することを目的とする。 [解決手段]食後における血中レムナント様リポ蛋白コレステロールの濃度上昇を抑制する、サイリウム種皮を有効成分とする食品組成物である。また、食後における血中レムナント様リポ蛋白コレステロールの濃度上昇を抑制する方法であって、食前または食事とともにサイリウム種皮を摂取する方法である。これにより、食事由来のRLP-C濃度の上昇を抑制し、アテローム型の動脈硬化を予防することができる。
Description
本発明は、食後における血中レムナント様リポ蛋白コレステロールの濃度上昇を抑制する食品組成物に関する。より詳しくは、サイリウム種皮を有効成分とする血中レムナント様リポ蛋白コレステロールの濃度上昇抑制組成物に関する。
食生活、運動習慣、喫煙、飲酒などの生活習慣により引き起こされる生活習慣病が問題となっている。例えば、食習慣が理由で発症する生活習慣病としては、糖尿病、肥満症、高脂血症、高血圧症、大腸がん、歯周病などがある。なかでも、糖尿病、肥満症、高脂血症、高血圧症と言った生活習慣病は、患者数が多いことに加えて、脳梗塞や心筋梗塞の原因となる動脈硬化症の発症と密接に関連していることがわかっている。
動脈硬化は、主に三つの型に分類することができる。第一に、動脈の中膜にカルシウムが溜まって石灰化する中膜硬化型、第二に、動脈の内壁にコレステロールなどの脂質が溜まり、アテロームと呼ばれる膨らみを形成するアテローム型、第三に、細動脈が硬化する細動脈硬化型である。このうち、アテローム型が脳梗塞や心筋梗塞を引き起こすと考えられている。
近年、アテローム型動脈硬化を惹起する蛋白として、レムナント様リポ蛋白(RLP-C)が注目されている。RLP-Cとは、カイロミクロン(CM)やVLDLなどの中間代謝産物であるCMレムナントやVLDLレムナントなどの総称である。通常、CMやVLDLはサイズが大きいため血管内皮に侵入できないが、小粒子化したRLP-Cは血管内皮に侵入することができる。そして、血管内皮に侵入したRLP-Cは、マクロファージ上にあるアポB48受容体を介してマクロファージに取り込まれ、マクロファージの泡沫化を促進し、動脈硬化巣を形成することが報告されている(非特許文献1参照)。

アテローム型の動脈硬化を防ぐためには、血中RLP-C濃度管理が重要となる。しかし、血中のRLP-C濃度は、他のコレステロールと異なり食事による影響を受けやすく、管理が難しいという問題がある。そのため、食事によるRLP-C濃度の上昇抑制について検討されていなかった。
本発明者らは、食後の血中RLP-C濃度上昇を抑制できる物質について検討を行った。そして、サイリウム種皮を摂取するにより、食後の血中RLP-C濃度の上昇を抑制できることを新たに見出し、本発明を完成するに至った。
上記課題解決のため、本発明は、食後における血中レムナント様リポ蛋白コレステロールの濃度上昇を抑制する、サイリウム種皮を有効成分とする食品組成物、およびこれを用いた飲食品を提供する。また、上記課題解決のため、本発明は、食後における血中レムナント様リポ蛋白コレステロールの濃度上昇を抑制する方法であって、食前または食事とともにサイリウム種皮を摂取する方法を提供する。
かかる構成によれば、サイリウム種皮を有効成分とする食品組成物を摂取することにより、食後における血中RLP-C濃度の上昇を抑制することができる。
本発明によれば、食後における血中RLP-C濃度の上昇を抑制することができる。これにより、アテローム型の動脈硬化を予防することができる。
以下、本発明を実施するための好適な形態について説明する。
<サイリウム種皮>
サイリウム種皮は、高度に分岐した構造を有する多糖類をその主成分とした食物繊維素材である。本発明で用いられるサイリウム種皮としては、オオバコ科の植物プランタゴオバタ(Plantago ovata)の種子から得られる種皮(ハスク)またはその粉砕物が挙げられる。ここで、サイリウム種皮またはその粉砕物としては、サイリウム、サイリウムハスク、サイリウム種皮末、サイリウムシードガム、イサゴールなど(以下、「サイリウム種皮末」という。)として市販されているものが挙げられる。本発明において、サイリウム種皮末は、いかなる粒度、グレードのものを用いても構わないが、夾雑物が少なく、純度が高いものが好ましい。
サイリウム種皮は、高度に分岐した構造を有する多糖類をその主成分とした食物繊維素材である。本発明で用いられるサイリウム種皮としては、オオバコ科の植物プランタゴオバタ(Plantago ovata)の種子から得られる種皮(ハスク)またはその粉砕物が挙げられる。ここで、サイリウム種皮またはその粉砕物としては、サイリウム、サイリウムハスク、サイリウム種皮末、サイリウムシードガム、イサゴールなど(以下、「サイリウム種皮末」という。)として市販されているものが挙げられる。本発明において、サイリウム種皮末は、いかなる粒度、グレードのものを用いても構わないが、夾雑物が少なく、純度が高いものが好ましい。
本発明の食品組成物の形状としては特に限定されない。上記サイリウム種皮は安全性が高く、無味無臭であることから、長期間の継続的摂取が容易である。摂取方法としては、例えば、錠剤、カプセル剤、顆粒剤、散剤、シロップ剤、ドライシロップ剤、液剤、懸濁剤、ゼリー剤、粉末飲料などが挙げられる。
飲食品としては特に制限されず、例えば、飲料、スプレッド類、ドレッシング類、パン類、米飯類、麺類、ソース類、菓子などが挙げられる。
本発明の飲食品は、各種栄養素、各種ビタミン、ミネラル、食物繊維、種々の添加剤をさらに配合することができる。
以下、実施例に基づいて本発明を具体的に説明する。本実施例では、健常な成人76名を対象とした摂取試験を行い、本発明の効果を調べた。
<試験食品の作成>
試験食品は、水150mlにサイリウムを3.6g溶解させた飲料とした。また、対照食品(プラセボ食品)は、サイリウムを含有しない飲料とした。なお、盲検性を保つため、いずれの飲料も外観および風味による区別がつかないようにした。

<負荷食品>
負荷食品はハンバーグ180g、バターロール2個、シューストリングポテト30g(総計:731kcal、タンパク質24.4g、脂肪43.2g、炭水化物61.3g)とした。

<試験>
本実施例では、試験食品とプラセボ食品ランダム化二重盲検クロスオーバー試験を行った。試験食品とプラセボ食品間のウォッシュアウト期間は22週間とした。まず、被験者76名を、スクリーニング時に性別、年齢、空腹時血中脂肪値を割付因子としたブロックランダム法により、2群にランダム割付した。被験者のスクリーニング時における一般背景は表1の通りであった。

被験者は各試験日前日の夕食を21時までに済ませ、その後は絶食(水のみ摂取可)とした。翌朝空腹状態における採血を行ってから、試験食品またはプラセボ食品を摂取させた。試験食品またはプラセボ食品摂取15分後、高脂肪負荷食を20分以内に摂取させ、高脂肪食摂取開始2,3,4,6時間経過後に採血を行った。また、摂取6時間後の採決が終了するまでの間は絶食とし、坐位安静とした。
<血液検査及び解析>
血中に含まれるRLP-C濃度は、臨床試験機関で既存の方法を用いて分析を行った。主要評価項目はRLP-C濃度の経時変化及び台形法により算出した血中濃度曲線下面積とした。なお、試験結果は、平均値±標準誤差(SE)で示した。

次に、得られた分析結果を統計解析し、試験食品摂取時の結果とプラセボ食品摂取時の結果とを比較した。統計解析は両側検定で行い、優位水準は5%に設定した。解析ソフトウェアはWindows版SPSS(Ver.24.0、日本アイ・ビー・エム株式会社製)を用い、統計解析方法として一般化線形モデル(General Liner Model:GLM)を選択した。
食後血中RLP-C濃度の経時的変化についての統計解析結果を図1に示す。
図1から明らかなように、試験食品における食後血中RLP-C濃度は、プラセボ食品における食後血中RLP-C濃度と比べて、低く抑えられていることがわかる。特に、食後2,3,4時間においては、優位に低い値となった(P<0.01)。また、食後血中RLP-C濃度の上昇抑制は食直後から始まり、食後4時間でプラセボ食品との差が最大となっていた。
次に、図1のデータを用いて、食後6時間までの血中濃度曲線下面積(AUC)を台形法により算出した。AUCは、食後に血液中に吸収されるRLP-Cの総量を意味する。結果を表2に示す。
表2から明らかなように、試験食品のAUCはプラセボ食品のAUCよりも優位に低い値を示した(P<0.003)。具体的には、試験食品を摂取したことにより、血中のRLP-C量がプラセボ食品を摂取した場合よりも約15%も減少していることがわかる。
ここで、試験食品であるサイリウム種皮が食後血中RLP-C濃度の上昇を抑制する理由としては、次のように考えられる。サイリウム種皮は保水性があり、水に溶解させるとゲルを形成する。このサイリウムゲルは、消化管内で脂質とリパーゼとの接触を妨げることで、血中への食事由来の脂質の吸収を抑制するものと考えられる。これにより、血中の脂質量が減るため、結果として食後血中RLP-C濃度が低下するものと考えられる。
以上説明したように、本発明によれば、サイリウム種皮の摂取により、食後血中RLP-C濃度の上昇を抑制することができる。RLP-Cはアテローム型動脈硬化を惹起する蛋白と考えられているため、RLP-C濃度上昇の抑制により、アテローム型動脈硬化の発生を効果的に抑制することができるものと考えられる。

- 食後における血中レムナント様リポ蛋白コレステロールの濃度上昇を抑制する、サイリウム種皮を有効成分とする食品組成物。
- 請求項1に記載のレムナント様リポ蛋白コレステロールの濃度上昇抑制用食品組成物を含有する飲食品。
- 食後における血中レムナント様リポ蛋白コレステロールの濃度上昇を抑制する方法であって、
食前または食事とともにサイリウム種皮を摂取することを含む前記方法。 - サイリウム種皮を水で膨潤させてから摂取する、請求項3記載の方法。
