Abstract
본 발명은 암 유래 AMF를 유효성분으로 함유하는 항암용 조성물에 관한 것으로, 더욱 상세하게는 본 발명의 암 유래 AMF는 췌장암, 폐암, 뇌암, 자궁경부암, 후두암, 간암, 백혈암, 전립선암, 유방암 및 대장암으로 이루어진 군으로부터 선택된 하나 이상의 암의 증식을 억제하고, 세포사멸을 일으키는 효과가 우수하므로, 암 유래 AMF를 유효성분으로 함유하는 본 발명의 조성물은 항암치료제로 사용될 수 있다.
Description
본 발명은 암 유래 AMF를 유효성분으로 함유하는 항암용 조성물에 관한 것으로, 더욱 상세하게는 간암, 유방암, 췌장암, 폐암, 난소암 또는 전립선암 유래 AMF를 유효성분으로 함유하는 항암용 약학 조성물에 관한 것이다.
암은 세계적으로 감염성 질환 및 심혈관계 질환과 더불어 3대 사망원인 중 하나로, 환경문제, 수명연장, 식문화의 서구화 등으로 인해 향후 암 발생인구가 급격하게 증가할 것으로 예상되는 주요 질환 중 하나이다. 보다 더욱 효과적인 항암제의 개발을 위해 암에 대한 많은 연구가 진행되고 있음에도 불구하고 암에 대한 발병기전의 다양화로 인해 부작용이 적고 내성을 극복할 수 있는 새로운 항암제의 개발이 여전히 절실한 상황이다.
한편, 세포경쟁은 세포간 생장 적응 및 생존의 비교 과정으로, 경쟁에서 밀리는 세포는 사멸될 수 있다. 세포경쟁은 암 발생과 밀접한 연관성을 지니는데 암세포에 비해 생장 적응력이 열세인 주변의 세포는 사라질 수 있다. 현재까지 경쟁에서 우위를 점하는 세포(winner cell)가 세포경쟁 초기에 세포사멸신호(death signal)를 분비할 것으로 가정되어 왔으나 규명된 바 없다.
한편, AMF(autocrine motility factor)는 하우스키핑(housekeeping) 단백질로서 세포 내에서 에너지 대사와 관련하여 해당작용(glycolysis)의 두 번째 과정의 글루코오스-6-포스페이트(glucose-6-phosphate)와 프락토오스-6-포스페이트(fructose-6-phosphate)를 상호전환(interconversion)하는 역할을 하는 것으로 알려졌으며, 이러한 기능으로 인해 글루코오스-6-포스페이트 이성질화제(glucose-6-phosphate isomerase, GPI)라고 불리기도 한다. 종양에서 분비되는 사이토카인인 AMF는 종양 부위에 풍부하며 종양의 증식, 분화 및 생존에 중요 역할을 지닌다.
한편, 한국공개특허 제2016-0050773호에는 AMFR-Fc 융합 단백질 및 이의 항암용도를 개시하고 있지만, 본 발명의 암 유래 AMF를 유효성분으로 함유하는 항암용 조성물은 개시된 바 없다.
본 발명은 상기와 같은 요구에 의해 도출된 것으로서, 암 유래 AMF를 유효성분으로 함유하는 항암용 조성물을 제공하고, 상기 조성물이 암의 증식을 억제하고, 세포사멸을 일으킴을 확인함으로써, 본 발명을 완성하였다.
상기 과제를 해결하기 위해, 본 발명은 암 유래 AMF(autocrine motility factor)를 유효성분으로 함유하는 항암용 약학 조성물을 제공한다.
또한, 본 발명은 상기 항암용 약학 조성물을 개체에게 투여하는 단계를 포함하는 암세포의 증식억제 또는 세포사멸 증진방법을 제공한다.
본 발명은 암 유래 AMF를 유효성분으로 함유하는 항암용 조성물에 관한 것으로, 본 발명의 암 유래 AMF는 췌장암, 폐암, 뇌암, 자궁경부암, 후두암, 간암, 백혈암, 전립선암, 유방암 및 대장암으로 이루어진 군으로부터 선택된 하나 이상의 암의 증식을 억제하고, 세포사멸을 일으키는 효과가 우수하므로, 암 유래 AMF를 유효성분으로 함유하는 본 발명의 조성물은 암 치료용 소재로 사용될 수 있다.
도 1은 간암세포 조정배지(conditioned medium, CM)를 타 암세포주에 처리하였을 때, 암세포의 증식을 MTT 분석으로 확인한 결과이다. DU145는 전립선암 세포주, MCF-7은 유방암 세포주, SNU 484는 위암 세포주, HeLa는 자궁경부암 세포주, HT-29는 대장암 세포주, A549는 폐암 세포주, SKOV3는 난소암 세포주, AsPC-1은 췌장암 세포주, U-87 MG 및 A172는 뇌암 세포주이다.
도 2는 AsPC-1 췌장암 세포에 대한 간암세포 조정배지(CM)의 세포 증식 억제효과가 AMF에 의한 것임을 확인한 결과이다. Con은 음성대조군이고, CM은 간암세포 조정배지 처리군이고, E4P는 AMF의 활성억제제인 E4P(erythrose 4-phosphate)를 0.5mM 처리한 군이고, E4P+CM은 간암세포 조정배지에 E4P 0.5mM을 함께 처리한 군이며, IP-CM은 AMF 항체를 이용하여 AMF가 제거된 간암세포 조정배지 처리군이다.

A549는 폐암 세포주, DU145는 전립선암 세포주, HeLa는 자궁경부암 세포주, Hep 2는 후두암 세포주, Hep G2는 간암 세포주, HL-60는 백혈암 세포주, HT-29는 대장암 세포주, MCF-7은 유방암 세포주, SKOV3는 난소암 세포주, SNU 484는 위암 세포주이고, 각각의 세포주는 CM이 처리되지 않은 무처리군과 비교하여 세포성장율을 확인하였다.
도 10은 DU145 전립선암세포 및 A549 폐암세포에 대한 췌장암세포 조정배지(CM)의 세포 증식 억제효과가 AMF에 의한 것임을 확인한 결과이다. Con은 음성대조군이고, AsPC-1 CM은 췌장암세포 조정배지 처리군이고, AsPC-1 CM+E4P는 췌장암세포 조정배지에 AMF의 활성억제제인 E4P(erythrose 4-phosphate)를 0.5mM 처리한 군이고, IP-AsPC-1 CM은 AMF 항체를 이용하여 AMF가 제거된 췌장암세포 조정배지 처리군이다.

도 14는 폐암세포 조정배지(CM)의 세포 증식 억제효과가 AMF에 의한 것임을 확인한 결과이다. Con은 음성대조군이고, E4P는 AMF의 활성억제제인 E4P(erythrose 4-phosphate)를 0.5mM 처리한 군이고, A549 CM은 폐암세포 조정배지 처리군이고, A549 CM+E4P는 폐암세포 조정배지에 E4P를 0.5mM 처리한 군이고, IP-A549 CM은 AMF 항체를 이용하여 AMF가 제거된 폐암세포 조정배지 처리군이다.
도 15는 정제한 폐암 유래 재조합 AMF의 암세포 사멸효과를 확인한 결과이다. rAMF는 폐암 유래 재조합 AMF이고, AsPC-1은 췌장암세포, MCF-7은 유방암세포, Hep G2는 간암세포, HT-29는 대장암세포 그리고 HeLa는 자궁경부암세포이다.
도 16은 폐암 유래 재조합 AMF가 HER2의 길항제(antagonist)로 작용하여 세포사멸을 일으키는 것을 AsPC-1 췌장암세포를 이용하여 확인한 결과이다. rAMF는 폐암 유래 재조합 AMF이다.
도 17은 난소암세포 조정배지(conditioned medium, CM)를 타 암세포주에 처리하였을 때, 암세포의 증식을 MTT 분석으로 확인한 결과이다. A549는 폐암 세포주, AsPC-1은 췌장암 세포주, DU145는 전립선암 세포주, HeLa는 자궁경부암 세포주, Hep G2는 간암 세포주, HL60은 백혈암 세포주, HT-29는 대장암 세포주, MCF-7는 유방암 세포주이다.
도 18은 MCF-7 유방암세포에 대한 난소암세포 조정배지(CM)의 세포 증식 억제효과가 AMF에 의한 것임을 확인한 결과이다. Con은 음성대조군이고, CM은 난소암세포 조정배지 처리군이고, E4P는 AMF의 활성억제제인 E4P(erythrose 4-phosphate)를 0.5mM 처리한 군이고, CM+E4P는 난소암세포 조정배지에 E4P 0.5mM을 함께 처리한 군이며, IP-CM은 AMF 항체를 이용하여 AMF가 제거된 난소암세포 조정배지 처리군이다.

도 21은 전립선암세포 조정배지(conditioned medium, CM)를 타 암세포주에 처리하였을 때, 암세포의 증식을 MTT 분석으로 확인한 결과이다. AsPC-1은 췌장암 세포주, Hep 3B는 간암 세포주, HL60은 백혈암 세포주, MCF-7은 유방암 세포주, PC3은 전립선암 세포주, SNU 484는 위암 세포주, U-87 MG는 뇌암 세포주이다.
도 22는 HL60 백혈암세포에 대한 전립선암세포 조정배지(CM)의 세포 증식 억제효과가 AMF에 의한 것임을 확인한 결과이다. 5%(v/v) CM은 배양배지 대비 5%(v/v) 전립선암세포 조정배지를 처리하는 것이고, E4P는 AMF의 활성억제제인 E4P(erythrose 4-phosphate)를 농도별로 처리하는 것이다. 5%(v/v) IP-CM은 AMF 항체를 이용하여 AMF가 제거된 전립선암세포 조정배지를 5%(v/v) 처리한 군이다.

상기 서열번호 1의 아미노산 서열로 구성된 AMF는 간암 세포주 유래 AMF이고, 바람직하게는 인간 유래의 간암 세포주인 Hep G2 유래 AMF인 것이지만, 이에 제한되는 것은 아니다.
상기 서열번호 2의 아미노산 서열로 구성된 AMF는 유방암 세포주 유래 AMF이고, 바람직하게는 인간 유래의 유방암 세포주인 MCF-7 유래 AMF인 것이지만, 이에 제한되는 것은 아니다.
상기 서열번호 3의 아미노산 서열로 구성된 AMF는 췌장암 세포주 유래 AMF이고, 바람직하게는 인간 유래의 췌장암 세포주인 AsPC-1 유래 AMF인 것이지만, 이에 제한되는 것은 아니다.
상기 서열번호 4의 아미노산 서열로 구성된 AMF는 폐암 세포주 유래 AMF이고, 바람직하게는 인간 유래의 폐암 세포주인 A549 유래 AMF인 것이지만, 이에 제한되는 것은 아니다.
상기 서열번호 5의 아미노산 서열로 구성된 AMF는 난소암 세포주 유래 AMF이고, 바람직하게는 인간 유래의 난소암 세포주인 SKOV3 유래 AMF인 것이지만, 이에 제한되는 것은 아니다.
상기 서열번호 6의 아미노산 서열로 구성된 AMF는 전립선암 세포주 유래 AMF이고, 바람직하게는 인간 유래의 전립선암 세포주인 DU145 유래 AMF인 것이지만, 이에 제한되는 것은 아니다.
본 발명의 암 유래 AMF의 범위는 서열번호 1 내지 서열번호 6으로 표시되는 아미노산 서열을 갖는 단백질의 기능적 동등물을 포함한다. "기능적 동등물"이란 아미노산의 부가, 치환 또는 결실의 결과, 상기 서열번호 1 내지 서열번호 6으로 표시되는 아미노산 서열과 적어도 70% 이상, 바람직하게는 80% 이상, 더 바람직하게는 90% 이상, 더욱 바람직하게는 95% 이상의 서열 상동성을 갖는 것으로, 서열번호 1 내지 6으로 표시되는 단백질과 실질적으로 동질의 생리활성을 나타내는 단백질을 말한다. 또한, 암 유래 AMF와 실질적으로 동질의 생리활성을 나타내는 펩타이드를 포함한다. "실질적으로 동질의 생리활성"이란 암 예방 또는 치료에 대한 활성을 의미한다.
본 발명에서 항암이란 암세포의 증식을 억제하거나 암세포를 죽이는 것을 의미한다. 본 발명의 조성물은 췌장암, 폐암, 뇌암, 자궁경부암, 후두암, 간암, 백혈암, 전립선암, 유방암 및 대장암으로 이루어진 군으로부터 선택된 하나 이상에서 항암 효과를 나타내는 것일 수 있고, 바람직하게는 서열번호 1의 간암 세포주 유래 AMF를 포함하는 조성물은 췌장암, 폐암 및 뇌암으로 이루어진 군으로부터 선택된 하나 이상에서 항암효과를 나타내는 것일 수 있고, 서열번호 2의 유방암 세포주 유래 AMF를 포함하는 조성물은 자궁경부암, 폐암, 췌장암, 후두암, 간암 및 백혈암으로 이루어진 군으로부터 선택된 하나 이상에서 항암효과를 나타내는 것일 수 있고, 서열번호 3의 췌장암 세포주 유래 AMF를 포함하는 조성물은 폐암, 전립선암, 후두암, 간암, 백혈암 및 유방암으로 이루어진 군으로부터 선택된 하나 이상에서 항암효과를 나타내는 것일 수 있고, 서열번호 4의 폐암 세포주 유래 AMF를 포함하는 조성물은 유방암, 췌장암, 간암 및 대장암으로 이루어진 군으로부터 선택된 하나 이상에서 항암효과를 나타내는 것일 수 있고, 서열번호 5의 난소암 세포주 유래 AMF를 포함하는 조성물은 췌장암, 백혈암 및 유방암으로 이루어진 군으로부터 선택된 하나 이상에서 항암효과를 나타내는 것일 수 있으며, 서열번호 6의 전립선암 세포주 유래 AMF를 포함하는 조성물은 췌장암, 백혈암 및 뇌암으로 이루어진 군으로부터 선택된 하나 이상에서 항암효과를 나타내는 것일 수 있으나, 이에 한정하는 것은 아니다.
본 발명의 일 구현 예에 따른 항암용 약학 조성물에 있어서, 상기 조성물은 암의 증식을 억제하고, 세포사멸을 유도할 수 있으나, 이에 제한되지 않는다.
본 발명에 따른 상기 약학 조성물은 각각 통상의 방법에 따라 캡슐제, 산제, 과립제, 정제, 현탁액, 에멀젼, 시럽, 에어로졸 등의 경구형 제형, 외용제, 좌제 및 멸균 주사용액의 형태로 제형화하여 사용될 수 있다.
본 발명의 약학 조성물에 포함될 수 있는 담체, 부형제 및 희석제로는 락토즈, 덱스트로즈, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로오스, 메틸 셀룰로오스, 미정질 셀룰로오스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유 등을 포함한 다양한 화합물 혹은 혼합물을 들 수 있다.
제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제된다. 경구 투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형제제는 상기 약학 조성물에 적어도 하나 이상의 부형제 예를 들면, 전분, 칼슘카보네이트, 수크로오스 또는 락토오스, 젤라틴 등을 섞어 조제된다. 또한, 단순한 부형제 이외에 마그네슘 스테아레이트, 탈크 같은 윤활제들도 사용된다. 경구를 위한 액상 제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당하는데 흔히 사용되는 단순 희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조 제제, 좌제가 포함된다. 비수성용제, 현탁제로는 프로필렌글리콜, 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔, 마크로골, 트윈 61, 카카오지, 라우린지, 글리세로젤라틴 등이 사용될 수 있다.
본 발명의 약학 조성물의 적합한 투여량은 제제화 방법, 투여 방식, 환자의 연령, 체중, 성, 병적 상태, 음식, 투여 시간, 투여 경로, 배설 속도 및 반응 감응성과 같은 요인들에 의해 다양하게 처방될 수 있다.
본 발명의 약학 조성물은 경구 또는 비경구로 투여할 수 있으며, 비경구 투여의 경우, 피부에 국소적으로 도포, 정맥 내 주입, 피하 주입, 근육 주입, 복강 주입, 경피 투여 등으로 투여할 수 있다.
또한, 본 발명은 상기 항암용 약학 조성물을 개체에게 투여하는 단계를 포함하는, 암세포의 증식억제 또는 세포사멸 증진 방법을 제공한다. 상기 개체는 암, 바람직하게는 췌장암, 폐암, 뇌암, 자궁경부암, 후두암, 간암, 백혈암, 전립선암, 유방암 및 대장암으로 이루어진 군으로부터 선택된 하나 이상의 암에 걸린 개체일 수 있으나, 이에 제한되지 않는다.
이하, 실시예를 이용하여 본 발명을 더욱 상세하게 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로 본 발명의 범위가 이들에 의해 제한되지 않는다는 것은 당해 기술분야에서 통상의 지식을 가진 자에게 있어 자명한 것이다.
재료 및 방법
1. 세포배양
본 실험에 사용된 인간 암 세포주는 한국세포주은행(Seoul, Korea)에서 구매하였다. 폐암 세포인 A549, 췌장암 세포인 AsPC-1, 전립선암 세포인 DU145 및 PC3, 자궁경부암 세포인 HeLa, 후두암 세포인 Hep 2, 간암 세포인 Hep G2 및 Hep 3B, 백혈암 세포인 HL60, 대장암 세포인 HT-29, 유방암 세포인 MCF-7, 난소암 세포인 SKOV3, 위암 세포 SNU 484, 뇌암 세포인 U-87 MG 및 A172를 10%(v/v) 소태아혈청(fetal bovine serum , FBS), 100U/㎖의 페니실린 G 및 100㎍/㎖의 스트렙토마이신이 포함된 RPMI 또는 DMEM 배지에서 37℃, 5% CO
2 조건하에 배양하였다.
2. 무혈청 조정배지(serum-free conditioned medium, CM)의 준비
각각의 암 세포주가 75㎠ 배양 플라스크(culture flask)에 대략 80% 비율로 자랐을 때, 인산완충생리식염수(phosphate-buffered saline, PBS)로 3번 세척한 후, 무혈청 배지로 교체하였다. 무혈청 배지는 24시간 후에 수득하여 수크로오즈에 대한 투석에 의해 투석 튜브(7000 MWCO)에서 1/5 부피로 농축시키고, 0.22㎛로 필터한 후 영하 70℃에서 보관하였다.
유사한 방법으로 대량의 무혈청 조정배지를 획득하기 위해, 175㎠ 배양 플라스크(culture flask)에 각각의 암 세포주를 배양하여 준비하고, 대략 80% 비율로 자랐을 때 PBS로 세척한 후, 신선한 무혈청 배지로 교체하여 24시간 동안 세포를 배양하여 무혈청 조정배지(이하 CM)를 획득하였으며, 그 후 24시간 간격을 두고 최종 72시간 동안 수득하여 총 3~5ℓ의 각각의 암 세포주에 대한 CM을 얻었다. CM은 0.22㎛로 필터한 후 동결 건조하여 영하 70℃에서 보관하였다.
3. 세포 성장 분석
세포는 24 웰 배양 접시에서 24시간 동안 배양한 후, 배양한 세포에 각각의 암세포주 유래 CM을 배양 배지에 대하여 5%(v/v) 농도로 처리하여 48시간에서 72시간 동안 재배양하였다.
세포 성장은 MTT(3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide; thiazolyl blue) 분석을 통해 570㎚의 파장에서 관찰하였다.
또한, 생존 세포 수 측정을 위해 트립판 블루 배제 분석(Trypan blue exclusion assay)을 수행하였다.
또한, 집략형성 분석법(Clonogenic assay)을 위해 24 웰 배양 접시에 세포를 접종한 후 37℃에서 12시간 동안 배양하고, 배양된 세포에 5%(v/v) 각각의 암세포주 유래 CM을 처리하고 콜로니가 형성될 때까지 배양하였다. 콜로니가 형성되면 PBS로 세척하고, 100% 메탄올로 5분 동안 고정한 후, 0.5%(w/v) 크리스탈 바이올렛으로 2시간 동안 염색한 다음 증류수에 침지시켜 세척 후 관찰하였다.
4. 단백질 정제(purification) 및 동정(identification)
동결건조된 각각의 암세포주 유래 CM은 트리스 완충액(50mM Tris-HCl, pH 7.5)에 용해시키고, 4℃에서 같은 완충액에 대해 투석하였다. 투석 후 각각의 암세포주 유래 CM 샘플을 트리스 완충액에서 예비 평형된 DEAE 세파로오스(diethylaminoethyl group sepharose) 30㎖를 함유하는 칼럼에 로딩하였다. 결합 단백질(bound protein)은 1분당 0.7㎖의 유속에서 트리스 완충액 중 0 내지 1.0 M 염화나트륨(NaCl)의 선형 구배를 사용하여 칼럼으로부터 용리되었다. 생체 활성을 갖는 주요 피크를 포함하는 용출 분획을 모아 수크로오즈에 대한 투석에 의해 투석 튜브(7000 MWCO)에서 농축시켰다. 농축된 샘플을 트리스 완충액에서 미리 평형화된 0.8×30㎝ Sephacryl S-200이 충진된 칼럼에서 재분획하였다. 1분당 0.1㎖의 유속에서 생리활성 분획을 선택하고, 상기 기재된 바와 같이 투석에 의해 농축시켰다. 생리활성은 트립판 블루 배제 분석(Trypan blue exclusion assay) 또는 MTT 분석을 이용하여 암세포주의 세포 성장을 확인하여 평가하였고, 생리활성을 나타낸 샘플을 12% SDS-PAGE 젤에서 크기를 확인한 후 단백질 동정을 위해 말디토프(MALDI-TOF) 질량 분석을 실시하였다.
5. AMF 차단 분석(blocking assay)
각각의 암세포주 유래 CM, 0.5 mM E4P(AMF 활성 억제제)가 포함된 각각의 암세포주 유래 CM 또는 면역침강법(immunoprecipitation, IP)으로 AMF가 제거된 IP-CM의 처리 또는 무처리 조건 하에서 세포 배양 2~3일 후에 생존 세포수를 측정하였다. IP-CM을 준비하기 위해 1㎖의 각각의 암세포주 유래 CM을 1㎍의 항-GPI/AMF 항체와 1시간 동안 배양한 후, 1㎕의 단백질 A/G 아가로오스 비드(agarose bead)를 1시간 동안 처리했다. 그 후 항원-항체 복합체가 결합된 비드를 제거하기 위해 3000rpm에서 2분 동안 원심분리를 수행하였다.
6. DNA 클로닝 및 재조합 단백질 발현
각각의 암세포주로부터 RNA를 추출하여 역전사한 후, 이를 PCR(95℃에서 30초, 58℃에서 30초 및 72℃에서 100초를 30회 반복)에 의해 AMF cDNA를 증폭시키기 위한 주형으로 사용하였다.
PCR에 사용한
NdeⅠ 제한효소 사이트가 포함된 정방향 프라이머와
XhoⅠ 제한효소 사이트가 포함된 역방향 프라이머는 GPI(genbank accession number: AK301103.1)를 암호화하는 DNA 서열을 기초로 제작되었다.
정방향 프라이머: 5'-TACATATGGCCGCTCTCACCCGGGACCCCCAGTTCCAGAA-3'(서열번호 7)
역방향 프라이머: 5'-ATCTCGAGTTATTGGACTCTGGCCTCGCGCTGCT-3'(서열번호 8)
1.7kb 길이의 PCR 산물은 DNA 서열을 확인하였고, pCold DNA1 벡터에 클로닝하여 pHep G2-AMF, pMCF-7-AMF, pAsPC-1-AMF, pA549-AMF, pSKOV3-AMF 및 pDU145-AMF를 구축하였다. pHep G2-AMF, pMCF-7-AMF, pAsPC-1-AMF, pA549-AMF, pSKOV3-AMF 또는 pDU145-AMF를 함유한 대장균 BL21을 100㎍/㎖의 암피실린이 함유된 LB배지에서 200rpm, 37℃의 조건하에서 16시간 동안 배양한 후 새 배지에서 1:100으로 희석하고, 200rpm, 15℃의 조건하에서 2시간 동안 재배양하였다. 그 후 세포에 0.5mM의 IPTG를 처리하고 100rpm, 20℃의 조건하에서 16시간 동안 배양하였다. 배양 후 수확된 세포를 완충용액(20mM Tris-HCl pH 8.0, 200mM NaCl, 1mM DTT 및 0.5mM PMSF)에 재현탁시키고, 그 후 French pressure cell press (Model FA-078A, Thermo IEC, Milford, MA, USA)를 사용하여 세포를 파쇄하고, 4℃에서 12000rpm으로 20분 동안 원심분리하여 용해된 분획물을 획득하였다. 획득한 분획물은 0.22㎛ 주사기 필터를 통과하였다. 재조합 AMF(rAMF)는 공급업체의 방법에 따라 His60 Ni 수지 친화성 크로마토그래피(Promega, Madison, WI, USA)로 정제하였으며, 정제된 단백질의 함량은 Bio-Rad 단백질 분석 시약을 사용하여 정량하였다.
7. 단백질 추출 및 웨스턴블랏(Western blot)
세포를 고무 스크레이퍼를 사용하여 배양 접시로부터 분리하고, 원심분리(3000rpm, 5분)에 의해 수집한 후, PBS로 2회 세척하였다. 세포 펠렛은 단백질 분해효소 억제 칵테일을 함유하는 차가운 RIPA 완충액(50mM Tris-HCl pH7.4, 1% NP-40, 0.5% Na-deoxycholate, 0.1% SDS, 150mM NaCl, 2mM EDTA, 50mM NaF)에 부유시키고, 세포 용해를 위해 30분 동안 얼음 위에서 반응하였다. 반응 후 상기 혼합물을 원심분리(13000rpm, 20분, 4℃)하여 맑은 용해물을 얻었다. 이후 샘플별 같은 양의 단백질을 넣고 12% SDS-PAGE로 분리하여 PVDF 막으로 옮겼다. 막에 결합된 단백질은 ECL 시스템을 이용하여 웨스턴 블랏(Western blot) 분석을 진행하였다.
실시예 1. 간암 세포주 유래 AMF의 항암효과
1) Hep G2 CM의 암세포 증식 억제효과
Hep G2 CM의 암세포 증식 억제효과를 확인하기 위해 간암을 제외한 다양한 암세포에 Hep G2 CM을 처리하여, MTT 분석을 실시하였다. 그 결과, 도 1에 나타난 바와 같이 간암 유래 세포인 Hep G2의 CM을 처리한 폐암(A549), 췌장암(AsPC-1) 및 뇌암(U-87 MG 및 A172) 세포의 증식은 억제되었고, 전립선암(DU145), 유방암(MCF-7), 위암(SNU 484), 자궁경부암(HeLa), 대장암(HT-29) 및 난소암(SKOV3) 세포에서는 증식 억제 효과가 나타나지 않았다.
이를 통해 Hep G2 CM은 폐암(A549), 췌장암(AsPC-1) 및 뇌암(U-87 MG 및 A172) 세포의 증식을 특이적으로 억제하는 것을 확인하였다.
2) Hep G2 분비 단백질의 분석
Hep G2 CM에 포함된 암세포 증식 억제효과를 나타내는 단백질을 분석하기 위해 단백질을 정제하고 동정하였다. DEAE 세파로오스(sepharose) 분획물 및 DEAE 세파로오스(sepharose) 분획물 중 생리활성을 나타내는 분획물을 Sephacryl S-200이 충진된 칼럼을 이용하여 재분획하고, 상기 재분획물 중 우수한 생리활성을 가지는 분획을 확인하였다. 생리활성은 분획물을 AsPC-1 세포에 처리하여 암세포의 증식을 억제하는 것을 통해 확인하였다.
우수한 생리활성을 나타내는 분획을 말디토프를 이용하여 질량 분석한 결과, 암세포의 이동성 및 증식과 관련되어 있는 단백질인 AMF를 확인하였으며, 이후 AMF를 이용하여 실험을 진행하였다.
3) AMF 활성 억제효과
Hep G2 CM 내에 존재하는 AMF가 세포의 증식을 억제하는지 확인하기 위해 AMF 차단 분석을 시행하였다. 그 결과 도 2에 나타난 바와 같이, Hep G2 CM에 AMF의 활성억제물질인 E4P를 처리하거나(CM+E4P), AMF 항체를 이용하여 사전에 Hep G2 CM 내의 AMF를 제거하였을 경우(IP-CM)에는 Hep G2 CM에 의해 증식이 억제되었던 췌장암(AsPC-1) 세포에 대한 효과가 회복되는 것을 확인하였다. 이를 통해 Hep G2 CM 내의 AMF가 특정 암세포의 증식을 억제하는 효과를 나타낸다는 것을 확인하였다.
4) Hep G2 유래 AMF 유전자 구조 분석
Hep G2 RNA를 이용한 역전사 및 PCR을 통하여 Hep G2 AMF cDNA를 증폭시킨 후 pCold DNA1 벡터에 클로닝하여 pHep G2-AMF를 구축하였다. 그 결과 서열번호 9와 같이 DNA 서열이 분석되었으며, 예측되는 재조합 AMF의 아미노산 서열을 분석한 결과, 도 3에서 나타난 바와 같이 GPI 동형단백질(Genbank accession number: BC004982)과 99%의 상동성을 나타내며, 몇 가지 돌연변이가 나타나는 것을 확인하였다.
5) Hep G2 유래 재조합 AMF에 의한 세포 증식 억제 효과
상기 재조합 AMF를 정제한 후 암세포에 처리하여 세포 증식에 미치는 효과를 확인하였다. 그 결과, 도 4에 나타난 바와 같이 다양한 농도(2, 20 및 100ng/㎖)의 재조합 AMF를 처리하였을 경우 무처리 대조군에 비하여 농도의존적으로 췌장암(AsPC-1) 세포의 증식이 억제되는 것을 현미경으로 관찰하였으며, 세포수를 측정한 결과, 재조합된 Hep G2 유래 AMF를 처리하였을 경우 췌장암(AsPC-1), 뇌암(A172 및 U-87MG) 및 폐암(A549) 세포의 증식이 억제되는 것을 확인하였다.
실시예 2. 유방암 세포주 유래 AMF의 항암효과
1) MCF-7 CM의 암세포 증식 억제효과
MCF-7 CM의 암세포 증식 억제효과를 확인하기 위해 유방암을 제외한 다양한 암세포에 MCF-7 CM을 처리하여, MTT 분석을 실시하였다. 그 결과, 도 5A에 나타난 바와 같이 유방암 유래 세포인 MCF-7의 CM을 처리한 폐암(A549), 췌장암(AsPC-1), 후두암(Hep 2), 자궁경부암(HeLa), 간암(Hep G2) 및 백혈암(HL60) 세포의 증식은 억제되었고, 전립선암(DU145), 대장암(HT-29), 난소암(SKOV3) 및 위암(SNU 484) 세포에서는 증식 억제 효과가 나타나지 않거나, 오히려 증식을 촉진하는 효과가 나타났다.
이를 통해 MCF-7 CM은 폐암(A549), 췌장암(AsPC-1), 자궁경부암(HeLa), 후두암(Hep 2), 간암(Hep G2) 및 백혈암(HL60) 세포의 증식을 특이적으로 억제하는 것을 확인하였다.
또한, 도 5B에 나타낸 바와 같이 집락형성 분석법(Clonogenic assay)을 통하여 MCF-7 CM에 의하여 췌장암(AsPC-1) 세포의 집락형성이 억제되는 것을 확인하였다.
2) MCF-7 분비 단백질의 분석
MCF-7 CM에 포함된 암세포 증식 억제효과를 나타내는 단백질을 분석하기 위해 단백질을 정제하고 동정하였다. DEAE 세파로오스(sepharose) 분획물 및 DEAE 세파로오스(sepharose) 분획물 중 생리활성을 나타내는 분획물을 Sephacryl S-200이 충진된 칼럼을 이용하여 재분획하고, 상기 재분획물 중 우수한 생리활성을 가지는 분획을 확인하였다. 생리활성은 분획물을 AsPC-1 세포에 처리하여 암세포의 증식을 억제하는 것을 통해 확인하였다.
우수한 생리활성을 나타내는 분획을 말디토프를 이용하여 질량 분석한 결과, 암세포의 이동성 및 증식과 관련되어 있는 단백질인 AMF를 확인하였으며, 이후 AMF를 이용하여 실험을 진행하였다.
3) AMF 활성 억제효과
MCF-7 CM 내에 존재하는 AMF가 세포의 증식을 억제하는지 확인하기 위해 AMF 차단 분석을 시행하였다. 그 결과 도 6에 나타난 바와 같이, MCF-7 CM에 AMF의 활성억제물질인 E4P를 처리하거나(CM+E4P), AMF 항체를 이용하여 사전에 MCF-7 CM 내의 AMF를 제거하였을 경우(IP-CM)에는 MCF-7 CM에 의해 증식이 억제되었던 췌장암(AsPC-1) 세포에 대한 효과가 회복되는 것을 확인하였다. 이를 통해 MCF-7 CM 내의 AMF가 특정 암세포의 증식을 억제하는 효과를 나타낸다는 것을 확인하였다.
4) MCF-7 유래 AMF 유전자 구조 분석
MCF-7 RNA를 이용한 역전사 및 PCR을 통하여 MCF-7 AMF cDNA를 증폭시킨 후 pCold DNA1 벡터에 클로닝하여 pMCF-7-AMF를 구축하였다. 그 결과 서열번호 10과 같이 DNA 서열이 분석되었으며, 예측되는 재조합 AMF의 아미노산 서열을 분석한 결과, 도 7에서 나타난 바와 같이 GPI 동형단백질(Genbank accession number: BC004982)과 99%의 상동성을 나타내며, 몇 가지 돌연변이가 나타나는 것을 확인하였다.
5) MCF-7 유래 재조합 AMF에 의한 세포 증식 억제 효과
상기 재조합된 AMF를 정제한 후 암세포에 처리하여 세포 증식에 미치는 효과를 확인하였다. 그 결과, 도 8A에 나타난 바와 같이 다양한 농도(2, 10 및 50ng/㎖)의 재조합 AMF를 처리하였을 경우 무처리 대조군에 비하여 췌장암(AsPC-1) 세포의 증식이 억제되는 것을 현미경을 통해 관찰하였고, 도 8B에 나타난 바와 같이 무처리 대조군에 비해 농도의존적으로 폐암(A549), 췌장암(AsPC-1), 자궁경부암(HeLa), 간암(Hep G2), 후두암(Hep 2) 및 백혈암(HL60) 세포의 생존율이 저하되는 것을 확인하였다.
실시예 3. 췌장암 세포주 유래 AMF의 항암효과
1) AsPC-1 CM의 암세포 증식 억제효과
AsPC-1 CM의 암세포 증식 억제효과를 확인하기 위해 췌장암을 제외한 다양한 암세포에 AsPC-1 CM을 처리한 후, 트립판 블루 배제분석을 실시하였다. 그 결과, 도 9에 나타난 바와 같이 췌장암 유래 세포인 AsPC-1의 CM을 처리한 폐암(A549), 전립선암(DU145), 후두암(Hep 2), 간암(Hep G2), 백혈암(HL-60) 및 유방암(MCF-7) 세포의 세포증식이 억제되는 것을 확인하였고, 자궁경부암(HeLa), 대장암(HT-29), 난소암(SKOV3) 및 위암(SNU484) 세포의 증식에는 영향을 미치지 않거나, 오히려 촉진하는 효과가 나타나는 것을 확인하였다.
이를 통해 AsPC-1 CM은 폐암(A549), 전립선암(DU145), 후두암(Hep 2), 간암(Hep G2), 백혈암(HL-60) 및 유방암(MCF-7) 세포의 증식을 특이적으로 억제하는 것을 확인하였다.
2) AsPC-1 분비 단백질의 분석
AsPC-1 CM에 포함된 암세포 증식 억제효과를 나타내는 단백질을 분석하기 위해 단백질을 정제하고 동정하였다. DEAE 세파로오스(sepharose) 분획물 및 DEAE 세파로오스(sepharose) 분획물 중 생리활성을 나타내는 분획물을 Sephacryl S-200이 충진된 칼럼을 이용하여 재분획하고, 상기 재분획물 중 우수한 생리활성을 가지는 분획을 확인하였다. 생리활성은 분획물을 A549 세포에 처리하여 암세포의 증식을 억제하는 것을 통해 확인하였다.
우수한 생리활성을 나타내는 분획을 말디토프를 이용하여 질량 분석한 결과, 암세포의 이동성 및 증식과 관련되어 있는 단백질인 AMF를 확인하였으며, 이후 AMF를 이용하여 실험을 진행하였다.
3) AMF 활성 억제효과
AsPC-1 CM 내에 존재하는 AMF가 세포의 증식을 억제하는지 확인하기 위해 AMF 차단 분석을 시행하였다. 그 결과, 도 10에 나타난 바와 같이 AsPC-1 CM에 AMF의 활성억제물질인 E4P를 처리하거나(AsPC-1 CM+E4P), AMF 항체를 이용하여 사전에 AsPC-1 CM 내의 AMF를 제거하였을 경우(IP-AsPC-1 CM)에는 AsPC-1 CM에 의해 증식이 억제되었던 전립선암(DU145) 및 폐암(A549) 세포에 대한 증식억제효과가 회복되는 것을 확인하였다. 이를 통해 AsPC-1 CM 내의 AMF가 특정 암세포의 증식을 억제하는 효과를 나타낸다는 것을 확인하였다.
4) AsPC-1 유래 AMF 유전자 구조 분석
AsPC-1 RNA를 이용한 역전사 및 PCR을 통하여 AsPC-1 AMF cDNA를 증폭시킨 후 pCold DNA1 벡터에 클로닝하여 pAsPC-1-AMF를 구축하였다. 그 결과 서열번호 11과 같이 DNA 서열이 분석되었으며, 예측되는 재조합 AMF의 아미노산 서열을 분석한 결과, 도 11에서 나타난 바와 같이 GPI 동형단백질(Genbank accession number: BC004982)과 99%의 상동성을 나타내며, 몇 가지 돌연변이가 나타나는 것을 확인하였다.
5) AsPC-1 유래 재조합 AMF에 의한 세포 증식 억제 효과
상기 재조합된 AMF를 정제한 후 암세포에 처리하여 세포 증식에 미치는 효과를 확인하였다. 그 결과, 도 12에 나타난 바와 같이 다양한 농도(1, 5, 25 및 100ng/㎖)의 재조합 AMF를 처리하였을 경우 무처리 대조군에 비하여 농도의존적으로 폐암(A549), 전립선암(DU145), 후두암(Hep 2), 간암(Hep G2), 백혈암(HL-60) 및 유방암(MCF-7) 세포의 증식이 억제되는 것을 확인하였으며, 대조군인 자궁경부암(HeLa) 세포는 AsPC-1 유래 재조합 AMF 처리에 의한 증식억제효과가 나타나지 않는 것을 확인하였다.
실시예 4. 폐암 세포주 유래 AMF의 항암효과
1) A549 CM의 암세포 증식 억제효과
A549 CM의 암세포 증식 억제효과를 확인하기 위해 폐암을 제외한 다양한 암세포에 A549 CM을 처리한 후, MTT 분석을 실시하였다. 그 결과, 도 13에 나타난 바와 같이 폐암 유래 세포인 A549 CM을 처리한 유방암(MCF-7), 간암(Hep G2), 대장암(HT-29) 및 췌장암(AsPC-1) 세포의 증식이 억제되는 것을 확인하였고, 자궁경부암(HeLa), 전립선암(DU145), 백혈암(HL60), 난소암(SKOV3) 및 위암(SNU 484) 세포의 증식에는 영향을 미치지 않거나 오히려 증식을 촉진하는 효과가 나타난 것을 확인하였다.
이를 통해 A549 CM은 유방암(MCF-7), 간암(Hep G2), 췌장암(AsPC-1) 및 대장암(HT-29) 세포의 증식을 특이적으로 억제하는 것을 확인하였다.
2) A549 분비 단백질의 분석
A549 CM에 포함된 암세포 증식 억제효과를 나타내는 단백질을 분석하기 위해 단백질을 정제하고 동정하였다. DEAE 세파로오스(sepharose) 분획물 및 DEAE 세파로오스(sepharose) 분획물 중 생리활성을 나타내는 분획물을 Sephacryl S-200이 충진된 칼럼을 이용하여 재분획하고, 상기 재분획물 중 우수한 생리활성을 가지는 분획을 확인하였다. 생리활성은 분획물을 AsPC-1 세포에 처리하여 암세포의 증식을 억제하는 것을 통해 확인하였다.
우수한 생리활성을 나타내는 분획을 말디토프를 이용하여 질량 분석한 결과, 암세포의 이동성 및 증식과 관련되어 있는 단백질인 AMF를 확인하였으며, 이후 AMF를 이용하여 실험을 진행하였다.
3) AMF 활성 억제효과
A549 CM 내에 존재하는 AMF가 세포의 증식을 억제하는지 확인하기 위해 AMF 차단 분석을 시행하였다. 그 결과, 도 14에 나타난 바와 같이 A549 CM에 AMF의 활성억제물질인 E4P를 처리하거나(A549 CM+E4P), AMF 항체를 이용하여 사전에 A549 CM 내의 AMF를 제거하였을 경우(IP-A549 CM)에는 A549 CM에 의해 증식이 억제되었던 AsPC-1 세포에 대한 증식억제효과가 회복되는 것을 확인하였다. 이를 통해 A549 CM 내의 AMF가 특정 암세포의 증식을 억제하는 효과를 나타낸다는 것을 확인하였다.
4) A549 유래 AMF 유전자 구조 분석
A549 RNA를 이용한 역전사 및 PCR을 통하여 A549 AMF cDNA를 증폭시킨 후 pCold DNA1 벡터에 클로닝하여 pA549-AMF를 구축하였다. 그 결과 서열번호 12와 같이 DNA 서열이 분석되었으며, 예측되는 재조합 AMF의 아미노산 서열을 분석한 결과, GPI 동형단백질(Genbank accession number: BC004982)과 100%의 상동성을 나타내는 것을 확인하였다.
5) A549 유래 재조합 AMF에 의한 세포 증식 억제 효과
상기 재조합된 AMF를 정제한 후 암세포에 처리하여 세포 증식에 미치는 효과를 확인하였다. 그 결과, 도 15에 나타난 바와 같이 다양한 농도(2, 10, 25 및 100ng/㎖)의 재조합 AMF를 3일 동안 처리하였을 경우 무처리 대조군에 비하여 농도의존적으로 췌장암(AsPC-1), 유방암(MCF-7), 간암(Hep G2) 및 대장암(HT-29) 세포의 증식이 억제되는 것을 확인하였으며, 대조군인 자궁경부암(HeLa) 세포는 A549 유래 재조합 AMF 처리에 의한 증식억제효과가 나타나지 않는 것을 확인하였다.
6) A549 유래 재조합 AMF에 의한 세포사멸관련 인자 발현변화
A549 세포에 재조합 AMF를 10ng/㎖의 농도로 처리하고, 48시간 동안 처리한 후 세포사멸과 관련한 인자의 발현변화를 확인하였다. AMF는 AMF 수용기와 결합하여 세포내 PI3K/AKT 신호전달에 관여하여 암세포의 증식과 혈관형성을 조절하고, HER2와 결합하여 종양세포의 성장을 촉진시킨다는 것이 공지되어 있으므로, 재조합 AMF가 HER2 및 AKT의 인산화에 미치는 영향을 확인하였다.
그 결과, 도 16에 나타낸 바와 같이 재조합 AMF를 처리하였을 때 HER2의 발현 및 AKT의 인산화가 감소하는 것을 확인하였으며, 이를 통해 자신이 분비하는 AMF와는 암세포의 유래가 다른 타 암세포 유래의 재조합 AMF를 처리하였을 경우 암세포의 HER2 발현을 억제시키며, AMF가 HER2의 길항제로 작용할 수 있다는 것을 확인하였다.
실시예 5. 난소암 세포주 유래 AMF의 항암효과
1) SKOV3 CM의 암세포 증식 억제효과
SKOV3 CM의 암세포 증식 억제효과를 확인하기 위해 난소암을 제외한 다양한 암세포에 SKOV3 CM을 처리하여, MTT 분석을 실시하였다. 그 결과, 도 17에 나타난 바와 같이 난소암 유래 세포인 SKOV3 CM을 처리한 췌장암(AsPC-1), 백혈암(HL60) 및 유방암(MCF-7) 세포의 증식이 억제되는 것을 확인하였고, 폐암(A549), 전립선암(DU145), 자궁경부암(HeLa), 간암(Hep G2) 및 대장암(HT-29) 세포의 증식에는 영향을 미치치 않았다.
이를 통해 SKOV3 CM은 췌장암(AsPC-1), 백혈암(HL60) 및 유방암(MCF-7) 세포의 증식을 특이적으로 억제하는 것을 확인하였다.
2) SKOV3 분비 단백질의 분석
SKOV3 CM에 포함된 암세포 증식 억제효과를 나타내는 단백질을 분석하기 위해 단백질을 정제하고 동정하였다. DEAE 세파로오스(sepharose) 분획물 및 DEAE 세파로오스(sepharose) 분획물 중 생리활성을 나타내는 분획물을 Sephacryl S-200이 충진된 칼럼을 이용하여 재분획하고, 상기 재분획물 중 우수한 생리활성을 가지는 분획을 확인하였다. 생리활성은 분획물을 AsPC-1 세포에 처리하여 암세포의 증식을 억제하는 것을 통해 확인하였다.

SKOV3 CM 내에 존재하는 AMF가 세포의 증식을 억제하는지 확인하기 위해 AMF 차단 분석을 시행하였다. 그 결과 도 18에 나타난 바와 같이, SKOV3 CM에 AMF의 활성억제물질인 E4P를 처리하거나(CM+E4P), AMF 항체를 이용하여 사전에 SKOV3 CM 내의 AMF를 제거하였을 경우(IP-CM)에는 SKOV3 CM에 의한 유방암(MCF-7) 세포의 증식억제효과가 회복되는 것을 확인하였다. 이를 통해 SKOV3 CM 내의 AMF가 특정 암세포의 증식을 억제하는 효과를 나타낸다는 것을 확인하였다.
4) SKOV3 유래 AMF 유전자 구조 분석
SKOV3 RNA를 이용한 역전사 및 PCR을 통하여 SKOV3 AMF cDNA를 증폭시킨 후 pCold DNA1 벡터에 클로닝하여 pSKOV3-AMF를 구축하였다. 그 결과 서열번호 13과 같이 DNA 서열이 분석되었으며, 예측되는 재조합 AMF의 아미노산 서열을 분석한 결과, 도 19에서 나타난 바와 같이 GPI 동형단백질(Genbank accession number: BC004982)과 99%의 상동성을 나타내며, 몇 가지 돌연변이가 나타나는 것을 확인하였다.
5) SKOV3 유래 재조합 AMF에 의한 세포 증식 억제 효과
상기 재조합된 AMF를 정제한 후 암세포에 처리하여 세포 증식에 미치는 효과를 확인하였다. 그 결과, 도 20에 나타난 바와 같이 다양한 농도(2, 20 및 100ng/㎖)의 재조합 AMF를 처리하였을 경우 무처리 대조군에 비하여 농도의존적으로 췌장암(AsPC-1), 백혈암(HL60) 및 유방암(MCF-7) 세포의 증식이 억제되는 것을 확인하였다.
실시예 6. 전립선암 세포주 유래 AMF의 항암효과
1) DU145 CM의 암세포 증식 억제효과
DU145 CM의 암세포 증식 억제효과를 확인하기 위해 다양한 암세포에 DU145 CM을 처리하여, MTT 분석을 실시하였다. 그 결과, 도 21에 나타난 바와 같이 DU145 CM은 췌장암(AsPC-1), 백혈암(HL60) 및 뇌암(U-87 MG) 세포의 증식을 억제하였고, 간암(Hep 3B), 유방암(MCF-7), 전립선암(PC3) 및 위암(SNU 484) 세포의 증식에는 영향이 없거나 오히려 증식을 촉진하였다.
이를 통해 DU145 CM은 췌장암(AsPC-1), 백혈암(HL60) 및 뇌암(U-87 MG) 세포의 증식을 특이적으로 억제하는 것을 확인하였다.
2) DU145 분비 단백질의 분석

DU145 CM 내에 존재하는 AMF가 세포의 증식을 억제하는지 확인하기 위해 AMF 차단 분석을 시행하였다. 그 결과 도 22에 나타난 바와 같이, DU145 CM에 AMF의 활성억제물질인 E4P를 처리하거나, AMF 항체를 이용하여 사전에 DU145 CM 내의 AMF를 제거하였을 경우(IP-CM)에는 DU145 CM에 의한 백혈암(HL60) 세포의 증식억제효과가 회복되는 것을 확인하였다. 이를 통해 DU145 CM 내의 AMF가 특정 암세포의 증식을 억제하는 효과를 나타낸다는 것을 확인하였다.
4) DU145 유래 AMF 유전자 구조 분석
DU145 RNA를 이용한 역전사 및 PCR을 통하여 DU145 AMF cDNA를 증폭시킨 후 pCold DNA1 벡터에 클로닝하여 pDU145-AMF를 구축하였다. 그 결과 서열번호 14와 같이 DNA 서열이 분석되었으며, 예측되는 재조합 AMF의 아미노산 서열을 분석한 결과, 도 23에서 나타난 바와 같이 GPI 동형단백질(Genbank accession number: BC004982)과 99%의 상동성을 나타내며, 몇 가지 돌연변이가 나타나는 것을 확인하였다.
5) DU145 유래 재조합 AMF에 의한 세포 증식 억제 효과
상기 재조합된 AMF를 정제한 후 암세포에 처리하여 세포 증식에 미치는 효과를 확인하였다. 그 결과, 도 24A에 나타난 바와 같이 재조합 AMF 처리시 뇌암(U-87 MG) 세포는 무처리군에 비해 작은 입자 형태로 변하며, 사멸하는 모습이 현미경으로 관찰되었으며, 도 24B에 나타난 바와 같이 다양한 농도(2, 10, 50 및 100ng/㎖)의 재조합 AMF를 3일 동안 처리하였을 경우 무처리 대조군에 비하여 농도의존적으로 췌장암(AsPC-1), 백혈암(HL60) 및 뇌암(U-87 MG) 세포의 증식이 억제되며 사멸하는 것을 확인하였다.
Claims (7)
- 암 유래 AMF(autocrine motility factor)를 유효성분으로 함유하는 항암용 약학 조성물.
- 제1항에 있어서, 상기 암 유래 AMF는 서열번호 1 내지 서열번호 6의 아미노산 서열로 이루어진 군으로부터 선택된 어느 하나인 것을 특징으로 하는 항암용 약학 조성물.
- 제1항에 있어서, 상기 항암은 췌장암, 폐암, 뇌암, 자궁경부암, 후두암, 간암, 백혈암, 전립선암, 유방암 및 대장암으로 이루어진 군으로부터 선택된 하나 이상의 암종에 대한 항암인 것을 특징으로 하는 항암용 약학 조성물.
- 제1항에 있어서, 상기 암 유래 AMF는 암세포의 증식을 억제하는 것을 특징으로 하는 항암용 약학 조성물.
- 제1항에 있어서, 상기 암 유래 AMF는 암세포의 세포사멸을 유도하는 것을 특징으로 하는 항암용 약학 조성물.
- 제1항에 있어서, 상기 약학조성물은 캡슐제, 산제, 과립제, 정제, 현탁액, 에멀젼, 시럽, 에어로졸, 외용제, 좌제 또는 멸균 주사용액의 제형으로 제조하는 것을 특징으로 하는 항암용 약학 조성물.
- 제1항 내지 제6항 중 어느 한 항에 따른 항암용 약학 조성물을 개체에게 투여하는 단계를 포함하는 암세포의 증식억제 또는 세포사멸 증진방법.
