Description
본 발명은 신규 바이페닐 유도체 화합물 및 이의 용도에 관한 것으로, 보다 구체적으로 본 발명은 신규 바이페닐 유도체 화합물, 상기 신규 바이페닐 유도체 화합물 또는 이의 약학적으로 허용가능한 염을 포함하는 암의 치료 또는 예방용 약학 조성물, 상기 약학 조성물을 투여하는 단계를 포함하는 암을 치료 또는 예방하는 방법, 상기 신규 바이페닐 유도체 화합물 또는 이의 약학적으로 허용가능한 염을 포함하는 암의 전이 억제용 약학 조성물, 상기 약학 조성물을 투여하는 단계를 포함하는 암의 전이를 억제하는 방법, 상기 신규 바이페닐 유도체 화합물 또는 이의 약학적으로 허용가능한 염을 포함하는 암의 예방 또는 개선용 식품 조성물 및 암의 치료용 약제의 제조를 위한 상기 바이페닐 유도체 화합물 또는 이의 약제학적으로 허용가능한 염의 용도에 관한 것이다.
암의 전이 (tumor metastasis)는 암 환자의 예후를 결정하는 가장 중요한 인자 중 하나로 암으로 인한 사망을 결정하는 주요 과정이다. 암의 치료에서 수술과 방사선 요법, 화학 요법 등 환자의 생존을 위해 많은 노력이 이루어지고 있지만 여전히 암 환자의 생존을 높이기 위한 노력은 계속되고 있다. 암 전이를 연구하는 분야는 암을 극복하는 마지막 전략 중 하나이고 암 전이 억제 단백질 (tumor metastasis suppressor)을 연구하는 것은 전이 억제 약물을 개발하는데 있어 필수적이다.
Nm23은 정상 조직의 발달 및 분화에 관여하는 단백질을 암호화하는 유전자로, 다양한 전이성 세포주에서 Nm23의 발현 감소가 보고되고 있다. 일반적으로 150 내지 180 개의 아미노산으로 구성되는 Nm23 단백질은 류우신 지퍼 모티프 (leucine zipper motif)를 포함하고 이인산 뉴클레오사이드 키나제 (nucleoside diphosphate kinase, NDPK) 활성을 갖는다. 특히, Nm23-H1은 암 전이 및 다른 다양한 세포 기작들, 예컨대 세포 증식, 배아 발달, 분화, 종양 형성 등에 중요한 역할을 담당하는 것으로 규명되었다. 암의 전이는 암 세포가 일차로 생성된 종양 조직 (primary tumor)에서 혈관을 침투하는 것을 시작으로 혈관을 통해 이동하고 생존하여 이차 지점 (secondary site)에서 새로운 콜로니 (colony)를 형성하는 여러 단계의 과정을 거쳐 일어난다. Nm23은 NDPK (Nucleotide diphosphate kinase)로서, ATP 를 이용하여 NDP (UDP, GDP, CDP)를 NTP (UTP, GTP, CTP)로 전환하는 단백질로 세포 내 NTP의 양을 조절하고 있는 효소로 밝혀졌다. 또한, Nm23-H1의 과발현이 암세포의 침윤 감소와 밀접한 연관을 갖는다고 밝혀진바 있다. 예를 들어, WO1997-035024에는 NDPK를 포함한 효소 및 뉴클레오사이드 동족체의 배합물을 암세포에 투여하여 암을 치료하는 방법이 개시되어 있다.
이러한 발견을 토대로 Nm23의 발현을 증가시키거나, 세포 침투성 (cell permeable) Nm23-H1을 처리하는 방향으로 연구가 진행되어 왔다. 구체적으로, MPA (Medroxyprogesterone acetate)를 처리하면 Nm23-H1의 발현 양이 증가하는 것을 확인하였고 이러한 현상은 MPA 처리에 의해 암 전이가 억제되는 메커니즘으로 이해되고 있다. 그러나, MPA의 처리는 Nm23-H1의 수준을 높이는 것 이외에도 예상치 못한 세포 내 반응을 일으키기 때문에, MPA는 약물로서 사용되지 못하고 있다.
또한, 최근에는 세포 침투성 Nm23-H1을 이용하여 암의 전이를 억제하는 방법이 제안되었다. 세포 침투성 Nm23-H1의 경우에는 원형질막을 통과할 수 있는 운반자 펩타이드 (transporter peptide)를 융합하는 방식으로 세포에 투입시켰다. 그 결과, 세포 침투성 Nm23-H1 처리에 의해 암 전이 억제 활성이 나타나는 것을 확인하였다. 다만, 이러한 세포 침투성 Nm23-H1이 단백질 의약품으로 적용되기까지는 생체 내 안정성을 극복해야 하는 과제가 남아있고, 암 전이 억제 약물은 단기간 처리로 큰 효과를 볼 수 없기 때문에 단백질 의약품과 같은 고비용의 약제를 선택할 수 없다는 현실적인 문제를 가지고 있다.
본 발명자들은 보다 효과적으로 암의 발병 및 전이를 억제할 수 있는 제제를 개발하고자 예의 연구노력한 결과, 새로이 개발된 바이페닐 유도체들을 사용할 경우, 암의 발병을 예방하고, 발병된 암을 치료할 수 있을 뿐만 아니라, 발병된 암의 전이를 억제할 수 있음을 확인하고, 본 발명을 완성하였다.
본 발명의 한 가지 목적은 신규 바이페닐 유도체 화합물을 제공하는 것이다.
본 발명의 다른 목적은 상기 신규 바이페닐 유도체 화합물 또는 이의 약학적으로 허용가능한 염을 포함하는, 암의 치료 또는 예방용 약학 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 상기 약학 조성물을 투여하는 단계를 포함하는 암을 치료 또는 예방하는 방법을 제공하는 것이다.
본 발명의 또 다른 목적은 상기 신규 바이페닐 유도체 화합물 또는 이의 약학적으로 허용가능한 염을 포함하는, 암의 전이 억제용 약학 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 상기 약학 조성물을 투여하는 단계를 포함하는 암의 전이를 억제하는 방법을 제공하는 것이다.
본 발명의 또 다른 목적은 상기 신규 바이페닐 유도체 화합물 또는 이의 약학적으로 허용가능한 염을 포함하는, 암의 예방 또는 개선용 식품 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 암의 치료용 약제의 제조를 위한 상기 바이페닐 유도체 화합물 또는 이의 약제학적으로 허용가능한 염의 용도를 제공하는 것이다.
본 발명에 따른 화학식 1의 신규 바이페닐 유도체 화합물, 또는 이의 약학적으로 허용가능한 염은 Nm23-H1/NDPK 활성 증진 물질로 암의 전이 및 성장을 억제할 수 있다. 이에 따라, 본 발명에 따른 조성물은 암의 예방, 개선 및 치료 뿐만 아니라 암의 전이 억제 용도로 우수한 효과를 나타낸다.

R1 내지 R4는 각각 독립적으로 수소, 히드록시 또는 C1 내지 C3 알콕시 그룹이다. 여기서 C1 내지 C3 알콕시 그룹은 메톡시, 에톡시 및 프로폭시 그룹으로 이루어진 군으로부터 선택되는 어느 하나이다.

상기 바이페닐 유도체는 하기 반응식 1, 2 또는 3의 반응경로를 통한 순차적 또는 수렴적 합성경로에 의해 제조될 수 있다.

Triphenylphosphonium bromide 유도체를 2-bromobenzaldehyde와 반응시키고 platinum oxide을 첨가하여 수소로 환원 반응을 수행한 후, boronic pinacol ester 유도체, potassium carbonate과 tetrakis (triphenylphosphine) palladium와 반응을 수행하여 화학식 1의 생성물을 생성한다.

본 발명의 약학 조성물은 투여를 위해서 상기 화학식 1의 바이페닐 유도체 화합물, 또는 이의 약제학적으로 허용가능한 염 외에 추가로 약제학적으로 허용가능한 담체를 1 종 이상 더 포함할 수 있다. 약제학적으로 허용 가능한 담체는 식염수, 멸균수, 링거액, 완충 식염수, 덱스트로즈 용액, 말토 덱스트린 용액, 글리세롤, 에탄올 및 이들 성분 중 1 성분 이상을 혼합하여 사용할 수 있으며, 필요에 따라 항산화제, 완충액, 정균제 등 다른 통상의 첨가제를 첨가할 수 있다. 또한 희석제, 분산제, 계면활성제, 결합제 및 윤활제를 부가적으로 첨가하여 수용액, 현탁액, 유탁액 등과 같은 주사용 제형, 환약, 캡슐, 과립 또는 정제로 제제화할 수 있다. 따라서, 본 발명의 약학 조성물은 패치제, 액제, 환약, 캡슐, 과립, 정제, 좌제 등일 수 있다. 이들 제제는 당 분야에서 제제화에 사용되는 통상의 방법 또는 Remington's Pharmaceutical Science(최근판), Mack Publishing Company, Easton PA 에 개시되어 있는 방법으로 제조될 수 있으며 각 질환에 따라 또는 성분에 따라 다양한 제제로 제제화될 수 있다.
본 발명의 약학 조성물은 목적하는 방법에 따라 경구 투여하거나 비경구 투여 (예를 들어, 정맥 내, 피하, 복강 내 또는 국소에 적용)할 수 있으며, 투여량은 환자의 체중, 연령, 성별, 건강상태, 식이, 투여시간, 투여방법, 배설율 및 질환의 중증도 등에 따라 그 범위가 다양하다. 본 발명의 화학식 1의 화합물의 일일 투여량은 약 1 내지 1000 ㎎/㎏ 이고, 바람직하게는 5 내지 100 ㎎/㎏ 이며, 하루 일회 내지 수회에 나누어 투여할 수 있다.
본 발명의 약학 조성물은 상기 화학식 1의 바이페닐 유도체 화합물, 또는 이의 약학적으로 허용가능한 염 외에 동일 또는 유사한 약효를 나타내는 유효성분을 1 종 이상 더 포함할 수 있다.
본 발명은 상기 바이페닐 유도체 화합물, 이의 약제학적으로 허용가능한 염 또는 상기 암의 치료 또는 예방용 약학 조성물을 암의 발병이 우려되거나 또는 암이 발병된 개체에 투여하는 단계를 포함하는, 암을 치료 또는 예방하는 방법을 제공한다.
이때, 상기 용어 "바이페닐 유도체 화합물", "약제학적으로 허용가능한 염", "치료" 및 "예방"의 정의는 상기에서 서술한 바와 같다.
본 발명의 용어 "개체"란, 암이 발병하였거나 발병할 수 있는 인간을 포함한 쥐, 생쥐, 가축 등의 모든 동물을 의미한다. 구체적인 예로, 인간을 포함한 포유동물일 수 있다.
본 발명의 약학 조성물은 약학적으로 유효한 양으로 투여한다. 상기 용어, "약학적으로 유효한 양"은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효 용량 수준은 개체 종류 및 중증도, 연령, 성별, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료 기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 예를 들면, 상기 상기 바이페닐 유도체 화합물, 이의 약제학적으로 허용가능한 염은 1일 0.01 내지 500 mg/kg으로, 구체적으로 10 내지 100 mg/kg의 용량으로 투여할 수 있으며, 상기 투여는 하루에 한 번 또는 수회 나누어 투여할 수도 있다.
본 발명의 치료방법은 상기 화학식 1의 화합물을 투여함으로써, 징후의 발현 전에 질병 그 자체를 다룰 뿐만 아니라, 이의 징후를 저해하거나 피하는 것을 또한 포함한다. 질환의 관리에 있어서, 특정 활성 성분의 예방적 또는 치료학적 용량은 질병 또는 상태의 본성(nature)과 심각도, 그리고 활성 성분이 투여되는 경로에 따라 다양할 것이다. 용량 및 용량의 빈도는 개별 환자의 연령, 체중 및 반응에 따라 다양할 것이다. 적합한 용량 용법은 이러한 인자를 당연히 고려하는 이 분야의 통상의 지식을 가진 자에 의해 쉽게 선택될 수 있다. 또한, 본 발명의 치료방법은 상기 화학식 1의 화합물과 함께 질환 치료에 도움이 되는 추가적인 활성 제제의 치료학적으로 유효한 양의 투여를 더 포함할 수 있으며, 추가적인 활성제제는 상기 화학식 1의 화합물과 함께 시너지 효과 또는 보조적 효과를 나타낼 수 있다.
본 발명은 상기 화학식 1의 바이페닐 유도체 화합물, 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 암의 전이 억제용 약학 조성물 및 상기 암의 전이 억제용 조성물을, 암의 전이가 우려되거나 또는 암이 전이된 개체에 투여하는 단계를 포함하는 암의 전이를 억제하는 방법을 제공한다.
이때, 상기 용어 "바이페닐 유도체 화합물", "약제학적으로 허용가능한 염", "치료", "예방" 및 "개체"의 정의는 상기에서 서술한 바와 같다.
본 발명에 따른 조성물은 Nm23-H1/NDPK 활성 증진을 통해 암의 전이를 억제함으로써 암의 전이 억제에 우수한 효과를 나타낸다.
본 발명은 상기 화학식 1의 바이페닐 유도체 화합물, 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 암의 예방 또는 개선용 식품 조성물을 제공한다.
본 발명의 식품 조성물은 건강기능식품으로서 사용될 수 있다. 상기 "건강기능식품"이라 함은 건강기능식품에 관한 법률 제6727호에 따른 인체에 유용한 기능성을 가진 원료나 성분을 사용하여 제조 및 가공한 식품을 의미하며, "기능성"이라 함은 인체의 구조 및 기능에 대하여 영양소를 조절하거나 생리학적 작용 등과 같은 보건 용도에 유용한 효과를 얻을 목적으로 섭취하는 것을 의미한다.
본 발명의 식품 조성물은 통상의 식품 첨가물을 포함할 수 있으며, 상기 "식품 첨가물"로서의 적합 여부는 다른 규정이 없는 한, 식품의약품안정청에 승인된 식품 첨가물 공전의 총칙 및 일반시험법 등에 따라 해당 품목에 관한 규격 및 기준에 의하여 판정한다.
본 발명의 식품 조성물은 암의 예방 및/또는 개선을 목적으로, 조성물 총 중량에 대하여 상기 화학식 1의 화합물을 0.01 내지 95 %, 바람직하게는 1 내지 80 % 중량백분율로 포함할 수 있다. 또한, 암의 예방 및/또는 개선을 목적으로, 정제, 캅셀, 분말, 과립, 액상, 환, 음료 등의 형태로 제조 및 가공할 수 있다.
본 발명은 암의 치료용 약제의 제조를 위한 상기 화학식 1의 바이페닐 유도체 화합물, 또는 이의 약제학적으로 허용가능한 염의 용도를 제공한다.
약제의 제조를 위한 상기 화학식 1의 화합물은 허용되는 보조제, 희석제, 담체 등을 혼합할 수 있으며, 기타 활성제제와 함께 복합 제제로 제조되어 활성 성분들의 상승 작용을 가질 수 있다.
본 발명의 조성물, 용도, 치료방법에서 언급된 사항은 서로 모순되지 않는 한 동일하게 적용된다.
이하, 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있도록 본 발명의 실시예에 대하여 상세히 설명한다. 그러나 본 발명은 여러 가지 상위한 형태로 구현될 수 있으며 여기에서 설명하는 실시예에 한정되지 않는다.
이하 본 발명에 있어서, 이하에서 언급된 시약 및 용매는 특별한 언급이 없는 한 Sigma-Aldrich, TCI로부터 구입한 것이며, HPLC는 chiral IB column, Hex/iPA = 80/20, 0.5 mL/min 를 사용하였으며, 컬럼크로마토그래피용 실리카겔은 60, 230-400 mesh ASTM을 사용하였다. 1HNMR 데이터는 Bruker Fourier Transform AV300 (300 MHz) spectrometers, Bruker Fourier Transform AV400 (400 MHz) spectrometers or Agilent Technologies DD2 (600 MHz)를 사용하여 측정하였다.
실시예 1. 2-(3,4-디메톡시펜에틸))-3',4'-디메톡시-1,1'-바이페닐 합성 (HYL-NM-038)
o-Bromobenzaldehyde (1.0 당량)을 DMF에 녹인 후 3,4-dimethoxyphenylboronic acid (1.0 당량)과 aq. 2 M Na2CO3 (3.0 당량) 과 Pd(PPh3)4 (0.01 당량)을 실온에서 첨가해 주었다. 혼합물을 80℃에서 밤새 섞어 준 후 아세트산 에틸로 희석시킨 후, aq. 염화암모늄과 물을 넣어 반응을 종료한 후 아세트산 에틸로 추출해 준 후 유기층을 MgSO4로 건조시켰다. 이 혼합물을 필터해서 모아진 유기층을 농축시킨 후 컬럼 크로마토그라피를 사용하여 실리카겔에서 분리하여, 화합물을 합성하였다.

위 합성 화합물을 dialkyl benzylphosphonate (2.0 당량)가 녹아있는 테트라히드로퓨란에 n-BuLi (2.48 M in Hx) (1.95 당량)과 함께 0 ℃에서 첨가하였다. 30분 동안 이 혼합물을 실온에서 섞어준 후 다시 0℃로 온도를 낮추어 cis-aldehyde (1.0 당량)와 DMPU (5.0 당량)을 THF에 녹여 캐뉼라를 사용해 첨가해 주었다. 이 혼합물을 밤새 실온에서 섞어준 후 aq. 염화암모늄과 물을 사용해 반응을 종료시켰다. 아세트산 에틸로 추출한 후 MgSO4로 건조한 후, 농축시켜 컬럼 크로마토그라피를 사용하여 실리카겔에서 분리하였다.

위 합성된 화합물을 메탄올과 디클로로메탄 1/1 용매에 녹였다. 이곳에 Pd/C (10 wt%) (0.1 당량)을 첨가한 후 수소로 치환된 상태에서 3시간동안 섞어주었다. 이후 셀라이트와 실리카로 필터를 한 후 유기 용매를 증발시킨 후 농축된 유기 혼합물을 컬럼 크로마토그라피를 사용하여 실리카겔에서 생성물[2-(3,4-디메톡시펜에틸))-3',4'-디메톡시 -1,1'-바이페닐]을 분리하였다.

실시예 2. 2-(3',4'-디메톡시-[1,1'-바이페닐]-2-일)-3-(3,4-디메톡시페닐)옥시란의 합성 (HYL-NM-039)
아세톤과 아세토나이트릴에 녹아있는 2-(3,4-디메톡시스티릴)-3'4'-디메톡시바이페닐 (1.0 당량)에 aq. NaHCO3 를 넣어주고 0℃로 온도를 낮추었다. Oxone (2.2 당량)을 첨가해 준 후 4시간을 섞어주었다. 반응을 증류수를 넣어 종료시켜준 후 아세트산 에틸로 추출한 후 MgSO4로 건조시킨 후 농축시켰다. 이 혼합물을 컬럼 크로마토그라피를 사용해 실리카겔에서 분리하였다. 이를 통해 생성물[2-(3',4'-디메톡시 -[1,1'-바이페닐]-2-일)-3-(3,4-디메톡시페닐)옥시란]을 제조하였다.

실시예 3. N-(3,4-디메톡시페닐)-3',4'-디메톡시-[1,1'-바이페닐]-2-카복스아미드의 합성 (HYL-NM-041)
디클로로메탄에 녹아 있는 0℃의 2-iodobenzoic acid (1.0 당량)에 DMF 2 ml를 첨가하였다. 이후 oxalyl chloride (2.0 당량)을 한 방울씩 천천히 첨가한 후 한시간 동안 섞어주었다. 이후 3,4-dimethoxyaniline을 혼합물에 첨가한 후 온도를 실온으로 올려준 후 두시간 반 동안 섞어주었다. 디클로로메탄으로 희석한 후 물을 넣음으로 반응을 종료시켰다. 혼합물을 디클로로메탄으로 추출한 후 MgSO4 로 건조시킨 후 진공상태에서 농축시켰다. 이후 컬럼 크로마토그래피를 사용하여 실리카겔에서 원하는 생성물을 분리하였다.
DMF(1 ml) 와 증류수(0.1 ml)에 녹아있는 N-(3,4-dimethoxyphenyl)-2-iodobenzamide (1.0 당량) 과 3,4-dimethoxyphenylboronic acid (2.0 당량)에 tetrakis(triphenylphosphine) palladium (0.01 당량) 과 K2CO3 (3.0 당량)를 실온에서 첨가한다. 이후 이 혼합물을 110℃에서 16시간동안 섞어주었다. 이후 aq.염화암모늄을 넣어 반응을 종료시킨 후 아세트산에틸로 추출한 후 MgSO4 로 건조시킨 후 진공상태에서 농축시켰다. 이후 컬럼 크로마토그래피를 사용하여 실리카겔에서 원하는 생성물[N-(3,4-디메톡시페닐)-3',4'-디메톡시-[1,1'-바이페닐]-2-카복스아미드]을 분리하였다.

실시예 4. N-(3',4'-디메톡시-[1,1'-바이페닐]-2-일)-3,4-디메톡시벤즈아미드의 합성 (HYL-NM-042)
0℃의 디클로로메탄에 녹아있는 3,4-dimethoxybenzoic acid (1.0 당량)에 디클로로메탄의 10분의 1 양의 DMF를 넣어주었다. 여기에 oxalyl chloride (2.0 당량)을 한방울씩 첨가한 후 한시간 동안 섞어주었다. 혼합물의 온도를 실온으로 올린 후 30분을 더 섞어주었다. 이후에 2-iodoaniline (5.0 당량)을 첨가한 후 두시간을 더 섞어주었다. 테트라히드로퓨란을 5 mL 더 첨가하고 30분을 더 섞은 후 증류수로 반응을 종료시켰다. 혼합물을 디클로로메탄으로 추출한 후 MgSO4로 건조시킨 후 진공에서 농축시킨다. 이후 컬럼 크로마토그래피를 사용하여 실리카겔에서 원하는 생성물을 분리하였다.

Dioxane과 증류수 (10:1)에 N-(2-iodophenyl)-3,4-dimethoxybenzamide (1.0 당량), 3,4-dimethoxyboronic acid (2.0 당량), Pd catalyst (0.01 당량) 그리고 potassium carbonate (3.0 당량)을 녹였다. 이 혼합물을 100℃에서 13시간동안 섞어준 후 aq. 염화암모늄을 넣어 반응을 종료시켰다. 유기혼합물을 아세트산에틸로 추출한 후 MgSO4로 건조시킨 후 진공에서 농축시켰다. 이후 컬럼 크로마토그래피를 사용하여 실리카겔에서 원하는 생성물[N-(3',4'-디메톡시 -[1,1'-바이페닐]-2-일)-3,4-디메톡시벤즈아미드]을 분리하였다.

실시예 5 내지 11. 화합물의 합성 (HYL-NM-049 - 55)
General Procedure A
0℃의 테트라히드로퓨란에 triphenylphosphonium bromide 유도체 (1.2 당량)를 섞어 준 후, t-BuOK (1.4 당량)을 넣어주었다. 30분 후에 테트라히드로퓨란에 녹아있는 2-bromobenzaldehyde (1.0 당량)을 10분에 걸쳐 첨가해주었다. 반응 혼합물을 실온으로 올린 후 16시간동안 섞어주었다. 이후에 증류수를 넣어 반응을 종료시킨 후 아세트산에틸로 추출하였다. 추출물을 MgSO4를 넣어 건조시킨 후 농축시켜 컬럼 크로마토그래피를 사용하여 실리카겔에서 원하는 생성물을 분리하였다
생성된 bromide (1 당량)을 디클로로메탄에 녹인 후 수소로 치환시킨 조건에서 Platinum oxide (0.73 당량)을 첨가하였다. 이 혼합물을 6시간동안 섞어준 후 실리카와 셀라이트로 필터한 후 진공상태에서 농축시켰다. 컬럼 크로마토그래피를 사용하여 실리카겔에서 원하는 생성물을 분리하였다
General Procedure B
DME/증류수 (2/1)에 boronic pinacol ester 유도체(1.5 당량), potassium carbonate (3.0 당량)과 tetrakis(triphenylphosphine) palladium (0.05 당량)을 녹인다. 이 혼합물에 bromide (1.0 당량)을 DME에 녹여 캐뉼라를 사용해 한방울씩 천천히 첨가하였다. 시작 물질이 다 사라질때까지 혼합물을 90℃에서 섞어주었다. 반응이 끝난 후 실온으로 다시 식혀준 후 혼합물에 증류수를 넣어 반응을 종료시키고 아세트산 에틸로 추출한 혼합물을 brine으로 씻어주었다. 유기혼합물을 MgSO4로 건조시키고 농축시켜서 컬럼 크로마토그래피를 사용하여 실리카겔에서 원하는 생성물을 분리하였다.
General Procedure C (General Procedure A의 반응물 제조단계)
아세토나이트릴에 녹아 있는 benzyl alcohol 유도체(1.0 당량)와 triphenylphosphine hydrogen bromide (1.0 당량)을 85 ℃에서 두시간 동안 섞어준다. 생성물이 염으로 생기게 되면 실온으로 식혀준 후 필터로 걸러주었다. 생성물을 아세토나이트릴로 씻어주었다.
아래 일곱 개의 화합물을 general procedure A,B 와 C에 따라 각각 R위치를 H나 Me로 바르게 치환시킨 반응물을 사용해 합성하였다.
실시예 5. 2'-(3,4-디메톡시펜에틸)-3-메톡시 -[1,1'-바이페닐]-4-올

general procedure B 에서 pinacol boronic ester대신 boronic acid를 사용하였다. 용매의 비율은 DME/H2O 2:1에서 5:1로 변경해 사용하였다.

실시예 10. 4-(2-(3',4'-디메톡시-[1,1'-바이페닐]-2-일)에틸)-2-메톡시페놀
General procedure B 에서 pinacol boronic ester대신 boronic acid를 사용하였다. 용매의 비율은 DME/H2O 2:1에서 5:1로 변경해 사용하였다.
1H NMR (400 MHz, Chloroform-d) δ 7.36 - 7.25 (m, 2H), 7.24 - 7.14 (m, 2H), 6.93 - 6.85 (m, 1H), 6.82 - 6.75 (m, 2H), 6.72 (d, J = 8.0 Hz, 1H), 6.44 (dd, J = 8.0, 1.9 Hz, 1H), 6.31 (d, J = 1.9 Hz, 1H), 5.40 (s, 1H), 3.90 (s, 3H), 3.84 (s, 3H), 3.73 (s, 3H), 2.90 - 2.79 (m, 2H), 2.70 - 2.55 (m, 2H).
실시예 11. 2'-(4-히드록시-3-메톡시펜에틸)-3-메톡시 -[1,1'-바이페닐]-4-올
1H NMR (400 MHz, Chloroform-d) δ 7.26 - 7.24 (m, 2H), 7.23 - 7.13 (m, 2H), 6.89 - 6.82 (m, 2H), 6.78 - 6.62 (m, 2H), 6.48 (dd, J = 8.0, 2.0 Hz, 1H), 6.38 (d, J = 1.9 Hz, 1H), 5.66 (s, 1H), 5.44 (s, 1H), 3.91 (s, 3H), 3.75 (s, 3H), 2.91 - 2.79 (m, 2H), 2.71 - 2.57 (m, 2H).
실시예 1 내지 11에서 제조한 본 발명의 화합물들의 구조는 다음 표 1에 정리된 바와 같다.
실시예 12. NDPK 의 활성 증진 확인
5 ng의 재조합 Nm23-H1을 상온에서 10 분 동안 시험물질 (실시예 1 내지 3)과 함께 NDPK 분석 완충액 (20 mM HEPES, 3 mM MgCl2)에서 배양하였고, 이를 1 분 동안 5 μM ADP와 반응시켜 NDPK 분석을 수행하였다.
세포기반 NDPK 분석을 통해 동일한 효과를 확인하였고, 세포기반 NDPK 분석은 다음과 같이 수행하였다. 프로테아제 억제제 칵테일(protease inhibitor cocktail)과 NDPK 분석 완충액 (NDPK assay buffer)으로 용해시킨 5,000K MDA-MB-231 세포에서 얻은 세포 용해물을 4℃에서 10 분 동안 8,000 rpm으로 원심분리하였다. 40 μL의 용해물을 시험물질과 함께 5 분간 배양한 다음, 50 μM UDP를 첨가하여 NDPK와 반응시켰다. ATP 소비는 ATP 결정 키트 (Molecular probe, USA)에 의해 평가되었다.
그 결과를 표 2에 나타내었다.

위 확인되는 바와 같이, 실시예 화합물들이 우수한 NDPK 활성 증진(Nm23-H1에 대한 활성 증가)을 나타내는 것을 확인하였다.
실시예 13. 실시예 1 화합물의 농도별 NDPK 활성 증진 확인
실시예 2와 동일한 방법을 이용하여, 실시예 1의 화합물의 농도별 NDPK 활성 증진을 확인하여, 그 결과를 도 1에 나타내었다.
도 1에서 확인되는 바와 같이, 실시예 1의 화합물 처리는 낮은 농도에서부터 우수한 NDPK 활성 증진 효과를 나타냄을 확인하였다.
실시예 14. Matrigel invasion assay를 통한 암 전이 억제 확인
Invasion assay를 폴리카보네이트 멤브레인 (pore size, 8 μm)을 가진 Transwell® unit(Corning, USA)를 이용하여 Matrigel coating 또는 이것이 없이 수행하였다. 멤브레인을 Matrigel™ 기반 멤브레인 메트릭스 50 μg (BD Bioscience, USA)로 코팅하였다. 5 Х 104 MDA-MB-231 세포를 무혈청 배지에 실시예 1 화합물과 함께 상부 챔버에 배양하고, 아래 챔버를 10% FBS를 함유하는 배지로 채웠다. 24시간 동안 37℃에서 배앙하고, 상부 멤브레인의 세포를 제거하였다. 막의 아래 부분으로 세포의 침투를 25% 메탄올 하 0.5% 크리스탈 바이올렛으로 염색하고 100 배 비율에서 현미경으로 관찰하였다.
그 결과를 도 2에 나타내었다.
도 2의 (A)는 현미경 사진을 나타내며, (B)는 이를 수치화 한 결과를 나타낸다. 도 2에서 확인되는 바와 같이, 실시예 1의 화합물의 처리가 세포의 침투를 크게 억제하는 것으로 확인되었다.
실시예 15. 면역 형관 염색 분석을 통한 암 전이 억제 확인
MDA-MB-231 세포를 Secureslip™ (Sigma) 세포 배양 유리 커버 슬립 상에서 50 내지 70% 수준까지 배양한 뒤, 실시예 1 (B038)의 존재 또는 부재 하에 다양한 시간 동안 처리하였다. 세포를 차가운 HBSS로 부드럽게 세척한 후, RBS에서 10 분 동안 상온에서 4% 파라포름알데히드 함유 HBSS로 고정시켰다. HBSS로 세척한 후, 10 분간 상온에서 0.1 % Triton X-100에 의한 침투를 수행하였고, HBSS로 2회 세척한 후, 상온에서 1 시간 동안 3% BSA, 0.2% 트윈 20 및 0.2% 젤라틴 함유 HBSS로 블로킹을 하고, 37℃에서 2 시간 동안 1 차 항체 배양을 하였다. F-actin을 2시간 동안 37℃에서 Rhodamine-Phalloidin (Thermo Fisher Scientific)에 의하여 염색하고 이어서 20분 동안 HBSS로 3회 워싱하였다. 커버 슬립 (Cover slips)은 안티 페이딩 용액 (anti-fading solution)으로 고정시키고, LSM510 META (Zeiss) 레이저 스캐닝 공초점 현미경 (confocal microscope)의 x63 대물렌즈를 사용하여 관찰하였다.
그 결과를 도 3에 나타내었다.
도 3에서 확인되는 바와 같이, MDA-MB-231 세포의 F-actin의 변화를 보면, 러플 (Ruffle)이 줄고, 세포 간 접촉이 증가하는 것을 확인할 수 있었다(B038). 이는 Nm23-H1의 세포 이동에 관여 하는 Rac1의 활성을 억제하고, 세포의 이동성을 감소시켜 암세포의 전이를 억제하는 것임을 보여주는 것이다.
상기 결과들을 종합하여 볼 때, 본 발명에 따른 신규 바이페닐 유도체 화합물들은 NDPK 활성을 증진시켜 암의 전이를 억제하였다. 즉, 본 발명에 따른 신규 화합물들은 암의 전이 억제 에 우수한 효과를 나타내여 항암제 및 항암 보조제로써 우수한 예방 및 치료 효과를 나타낸다.
이상으로 본 발명 내용의 특정한 부분을 상세히 기술하였는바, 당업계의 통상의 지식을 가진 자에게 있어서, 이러한 구체적 기술은 단지 바람직한 실시양태일 뿐이며, 이에 의해 본 발명의 범위가 제한되는 것이 아닌 점은 명백할 것이다. 따라서 본 발명의 실질적인 범위는 첨부된 청구항들과 그것들의 등가물에 의하여 정의된다고 할 것이다.
Claims (13)
- 제1항에 있어서,C1 내지 C3 알콕시 그룹은 메톡시, 에톡시 및 프로폭시 그룹으로 이루어진 군으로부터 선택되는 어느 하나인, 바이페닐 유도체 화합물, 또는 이의 약학적으로 허용가능한 염.
- 제1항에 있어서,R1 내지 R4는 각각 독립적으로 히드록시 또는 메톡시인, 바이페닐 유도체 화합물, 또는 이의 약학적으로 허용가능한 염.
- 제3항에 있어서,R1 내지 R4는 각각 독립적으로 히드록시 또는 메톡시이며, 적어도 2개 이상의 메톡시 그룹을 가지는, 바이페닐 유도체 화합물, 또는 이의 약학적으로 허용가능한 염.
- 제1항에 있어서,L은 -(C-C)-이며,R1 내지 R4는 메톡시인, 바이페닐 유도체 화합물, 또는 이의 약학적으로 허용가능한 염.
- 제1항 내지 제6항 중 어느 한 항에 따른 바이페닐 유도체 화합물, 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 암의 치료 또는 예방용 약학 조성물.
- 제7항에 있어서,상기 암은 유방암, 폐암, 흑색종, 전립선암, 대장암, 방광암, 뼈암, 혈액암, 갑상선암, 부갑상선암, 골수암, 직장암, 인후암, 후두암, 식도암, 췌장암, 위암, 설암, 피부암, 뇌종양, 자궁암, 두부 또는 경부암, 담낭암, 구강암, 결장암, 항문부근암, 중추신경계 종양, 간암, 대장암 및 이들의 조합으로 구성된 군으로부터 선택되는 암인 것인, 암의 치료 또는 예방용 약학 조성물.
- 제7항에 있어서,상기 암은 유방암, 폐암, 대장암 및 피부암으로 이루어진 군으로부터 선택되는 암인 것인, 암의 치료 또는 예방용 약학 조성물.
- 제7항의 암의 치료 또는 예방용 약학 조성물을, 암의 발병이 우려되거나 또는 암이 발병된 개체에 투여하는 단계를 포함하는, 암을 치료 또는 예방하는 방법.
- 제1항 내지 제6항 중 어느 한 항에 따른 바이페닐 유도체 화합물, 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 암의 전이 억제용 조성물.
- 제11항의 암의 전이 억제용 조성물을, 암의 전이가 우려되거나 또는 암이 전이된 개체에 투여하는 단계를 포함하는, 암의 전이를 억제하는 방법.
- 제1항 내지 제6항 중 어느 한 항에 따른 바이페닐 유도체 화합물, 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 암의 예방 또는 개선용 식품 조성물.
