Abstract
본 발명은 신규 유산균 균주 및 이를 포함하는 조성물에 관한 것이다. 또한, 본 발명은 유산균 균주 또는 이것의 배양액을 포함하는 항균조성물, 화장용조성물, 및 기능성식품에 관한 것이다.
Description
본 발명은 신규 유산균 균주 및 이를 포함하는 조성물에 관한 것이다. 또한, 본 발명은 유산균 균주 또는 이것의 배양액을 포함하는 항균조성물, 화장용조성물, 및 기능성식품에 관한 것이다.
유산균 (Lactic acid bacteria) 또는 젖산균은 포도당 또는 유당과 같은 탄수화물을 분해하고 이를 이용하여 젖산을 만드는 박테리아로서, 현재 약 300 내지 400여 종의 유산균이 알려져 있다. 유산균은 사람 또는 동물의 소화관, 발효식품, 예를 들어, 우유, 요구르트, 치즈 및 김치 등, 식물 및 토양 등 자연계에 널리 분포하고 있으며, 락토바실러스 (Lactobacillus), 락토코커스 (Lactococcus), 스트렙토코커스 (Streptococcus), 류코노스록 (Leuconostoc), 페디오코커스 (Pediococcus) 및 비피더스 (Bifidobacterium) 등이 대표적인 유산균이다.
유산균이 생산하는 젖산 (lactic acid) 및 아세트산 (acetic acid) 등의 유기산은 주로 중성 및 염기성 환경에서 생육하는 유해균에 대해 우수한 항균작용을 나타내며, 이러한 항균 특성을 이용하여 유산균을 식품 등의 보존제로 이용하고자 하는 연구가 있어 왔다. 또한, 유산균이 생산하는 박테리오신은 펩타이드 또는 단백질 계열의 항균물질로서, 이들 박테리오신을 생산하는 균주는 주로 자신과 계통학적으로 유사한 균종에 대해 강력한 항균활성을 보인다. 이에 따라, 유산균으로부터 항균력을 갖는 펩타이드 또는 단백질을 분리하여 그 특성을 확인하고 이를 항균소재 또는 항염증 치료제로 개발하고자 하는 시도가 이뤄지고 있다. 특히, 박테리오신은 펩타이드 또는 단백질로 이루어져 있으므로 체내에서 쉽게 분해되어 인체 독성 및 잔류에 의한 문제가 없다는 점에서 이를 항균소재 또는 항염증 치료제로서 개발함에 있어 장점이 있다. 예를 들어, 나이신은 (nisin)은 광범위한 항균 활성을 갖는 박테리오신으로서 개발되어 널리 활용되고 있다.
장내 상피세포의 밀착연접 (tight junction)은 장내 면역기능의 항상성을 유지하는데 중요하다. 밀착연접을 구성하는 단백질로서 클라우딘-4 (claudin-4), 조-1 (Zo-1), 오클루딘 (Ocludin) 등이 알려져 있으며, 클라우딘-1 (claudin-4), 조-1 (Zo-1) 및 오클루딘 (Ocludin) 등은 피부 상피세포의 밀착연접에 관여한다. 식품에 의한 알러지, 장염, 자가면역질환, 및 Celiac disease and inflammatory bowel diseases 등에서 밀착연접 기능이 손상되는 것이 발견되었다 (Fasano, A., Pathological and therapeutical implications of macromolecule passage through the tight junction. In Tight Junctions, CRC Press, Inc., Boca Raton, Fla. 697-722 (2001)). 밀착연접은 장벽을 통과하여 유입되는 매크로분자 (macromolecule)에 대한 방어벽으로 작용한다. 건강한 상태에서는 적은 양의 면역적으로 활성인 항원들이 장 점막의 장벽을 통과하여 체내로 유입될 수 있을 뿐이다. 방사선, 화학요법 또는 독소 등에 의해 밀착연접 시스템이 손상되면, 유해한 자가면역질환 및 식품 알러지를 유발하는 유해한 면역반응이 발생하게 된다. 정상적인 장에서 장내 면역반응은 장의 항상성 유지를 위해 조절된다. 셀리악 병 (Celiac disease, CD)은 만성 자가면역질환의 일종이며, 밀의 주요 단백질 분획인 글루텐이 이 질환을 유발하는 인자로 알려져 있다. 소화된 글루텐 또는 이것의 유도체는 소장 상피 세포벽을 통과한 후 조직 트랜스글루타미나제 (tissue transglutaminase, tTG)에 탈아미드화를 수행과 class II MHC 분자에 관여하면서 인체에 해로운 T 세포-매개 면역반응을 유발한다. CD에서와 같이 밀착연접 시스템이 손상되면, paracellular leak (leaky gut) 및 주변환경적 항원에 대한 반응이 일어난다.
염증성장질환 (Inflammatory bowel disease, IBD)은 장내에서 발생하는 적절하지 못한 면역반응을 의미한다. 2종류의 IBD가 알려졌다: 크론병 (Crohn's disease) 및 궤양성 대장염 (ulcerative colitis, UC). 염증성 장질환의 이들 두가지 형태는 T 세포-매개 면역의 비정상적 프로파일을 보인다. 크론병 환자의 장에서는 강한 Th1 반응이 유도되고, UC 환자의 대장에서는 Th2 반응이 상승조절된다. IBD에서 장의 장벽기능이 손실된다. 예를 들어, 크론병은 장벽의 증가된 투과성과 관련된다 (Oshitani, et al., Int. J. Mol. Med. 15(3):407-10 (2005); Ye, et al., Am. J. Physiol.-Gastro. and Liver Physiol., 290(3):496-504 (2006), Willemsen, et al., Clin. Exp. Immunol. 142(2): 275-284 (2005)).
밀착연접(tight junction)은 상피세포에 존재하며 이웃한 세포막 측면을 서로 고정시키고 세포 측면 공간을 통한 수분과 체액의 흐름을 방지하는 장벽이다. 각질층이 손상되면서 각질층과 과립층 사이에 존재하는 체액의 경표피수분손실과 함께 칼슘이온 농도도 감소하면 바로 아래의 과립층 상부의 밀착연접이 열리고[21], 밀착연접 바로 아래까지 위치하고 있던 랑게르한스세포의 수지(dendrite)가 밀착연접 위로 올라오게 된다. 이때 손상된 각질층을 통과하여 들어온 분자량이 큰 단백질 항원들은 랑게르한스세포의 수지에 의해 포집되고 인식되어 국소 림프절로 이동하고, 이들 단백질 항원의 세린계단백분해효소 성분은 각질형성세포막에 존재하는 PAR-2를 활성화시킨다. 한편으로는 각질층을 침투한 균들은 toll-like receptor (TLR)를 활성화하게 된다. 이후 각질형성세포에서는 여러 케모카인과 TNF-α, IL-1, thymic stromal lymphopoietin (TSLP) 등의 사이토카인들이 분비되어 아토피피부염의 발병을 촉진하고 질환을 악화시킨다[22]. 또한 밀착연접 단백질의 발현이 아토피피부염 환자의 피부에서 정상인에 비하여 현저하게 감소하여 있으며, 최근에는 아토피피부염 환자에서 claudin 1 유전자 (CDN1) 변이가 보고되었고, 헤르페스 바이러스 감염이 쉽게 일어나는 원인이 밀폐연접 단백질 중 claudin-1의 감소 때문이라 설명하고 있다 (De Benedetto A, Rafaels NM, McGirt LY, Ivanov AI, Georas SN, Cheadle C, Berger AE, Zhang K, Vidyasagar S, Yoshida T, Boguniewicz M, Hata T, Schneider LC, Hanifin JM, Gallo RL, Novak N, Weidinger S, Beaty TH, Leung DY, Barnes KC, Beck LA. 본 발명자는 항균활성 뿐만 아니라 특히 항진균 활성이 우수한 유산균 균종을 분리하고자 부단히 노력하였으며, 그 결과 한국 고유의 전통 식품인 된장으로부터 분리된 유산균 균주가 광범위한 항균 스펙트럼 및 뛰어난 항진균을 활성을 나타내는 것을 확인하고, 본 발명을 완성하였다. 분리된 신규 유산균 균주를 동정한 결과, 페디오코커스 펜토사세우스 (Pediococcus pentosaceus) 속에 속하는 유산균임을 확인하고, Pediococcus pentosaceus AB160011로 명명하였다.
본 발명은 항균 및 항진균 활성을 지닌 신규한 유산균 균주를 제공하는 것을 목적으로 한다.
또한, 본 발명은 밀착연접 (tight junction)을 구성하는 단백질의 발현을 증가시키는 신규한 유산균 균주를 제공하는 것을 목적으로 한다.
또한, 본 발명은 유산균 균주를 포함하는 염증성 장질환 및 면역질환을 개선하기 위한 기능성식품을 제공하는 것을 목적으로 한다.
또한, 본 발명은 본 발명의 유산균 균주 배양액 및/또는 유산균 균주를 포함하는 항균 및 항진균 조성물을 제공하는 것을 목적으로 한다.
또한, 본 발명은 본 발명의 유산균 균주 배양액 및/또는 유산균 균주를 포함하는 화장료조성물을 제공하는 것을 목적으로 한다.
상기 목적을 달성하기 위해 본 발명은 신규한 페디오코커스 펜토사세우스 (Pediococcus pentosaceus) AB160011 균주를 제공한다.
본 발명에 따른 페디오코커스 펜토사세우스 (Pediococcus pentosaceus) AB160011은 우수한 내산성 및 내담즙성을 가지며, 항균 및 항진균 활성을 나타내었다. 또한, 본 발명의 균주 및 이것의 배양물은 피부 및 장의 상피세포의 밀착연접을 구성하는 단백질의 발현을 유의성 있게 증가시키는 것이 확인되었으며, 임상시험결과 피부보습 증진효과를 갖는 것으로 나타났다. 또한, 본 발명의 신규 유산균 균주 배양물은 항염증 및 항산화 효과를 나타내었다.

이하, 실시예에 의거하여 본 발명을 보다 상세히 설명한다. 하기 실시예는 본 발명을 설명하기 위한 예시에 불과한 것으로서, 본 발명의 권리범위가 하기 실시예에 제한되는 것이 아님은 물론이다.
균주의 분리 및 동정, 및 제조예
(1) 유산균 분리
항균 및 항진균 활성을 보유한 유산균 균주를 분리하기 위해 국내에서 제조된 김치, 간장, 및 된장을 포함하는 발효제품으로부터 다양한 유산균을 10배 희석법을 통해 2% (w/w) 한천 (agar)이 포함된 MRS (Difco 288110) 고체배지에서 분리하였다. 생성된 콜로니는 크기, 색상, 특성별로 구분하여 또 다른 MRS 배지에 획선 접종하여 스크리닝 대상균주 총 1,785개의 유산균을 분리하였다. 분리된 유산균은 각각의 콜로니를 MRS 아가 플레이트에 획선 접종하였고, 37℃에서 24시간 동안 배양하였다. 분리한 유산균은 멸균한 MRS 액체배지에 접종하여 37℃에서 24시간 정치배양 후 20%(v/v) 글리세롤을 첨가하여 -80℃ 초저온냉동고에 보관하였다.
(2) 균주 동정
형태학적 분석
본 발명의 균주를 동정하기 위해, MRS 평판배지에 배양한 후 콜로니의 형태를 현미경으로 관찰하였다. 콜로니 형태는 표 1과 같았으며, 도 1은 본 발명의 균주의 현미경 사진이다.

16s rRNA를 통한 동정
본 발명의 신규 균주는 서열목록 1 서열에 나타낸 것과 같은 16s rRNA 염기서열을 갖는 것으로 확인되었다. 이를 기초로 계통수 연구한 결과 (도 2 참조), 분리된 균주는 페디오코커스 펜토사세우스 (Pediococcus pentosaceus) AJ3053와 높은 유사도를 나타내어 페디오코커스 펜토사세우스에 속하는 페디오코커스 펜토사세우스 (Pediococcus pentosaceus) AB160011 균주로 명명하고, 2016년 12월 07일자로 한국미생물보존센터에 기탁번호 KCCM11954P로 기탁하였다.
DNA 염기서열 분석을 통한 동정
본 발명의 신규 균주와 높은 유사도를 나타내는 페디오코커스 펜토사세우스 (ATCC25745)와의 상동성을 DNA 염기서열 (Ggepard, Nucleotide sequences Dot plot)로 분석한 결과 98.97%의 상동성을 보였다 (도 3 참조).
배양특성 확인
본 발명의 신규 유산균 균주를 멸균 MRS 액체배지에 접종하고 37℃에서 정치배양하면서 탁도 (Ultrospec2100 pro, OD625nm)와 산도 (Orion star A211, pH)의 변화를 관찰하였다. 도 4 및 도 5는 본 발명의 균주의 성장곡선 및 산도 변화를 나타낸 것이다.
유산균 배양 및 배양액 동결건조
유산균 배양 배지조성은 표 2와 같고 제작 공정은 크게 종배양, 본배양, 균체/배양액 분리, 동결건조의 단계로 구분된다. 종배양은 최종 배양액의 규모에 따라 최종 배양액의 10%의 볼륨까지 배양하는 과정을 말한다. 최종 배양된 종배양을 멸균된 배지에 접종하여 배양한 것을 본배양이라 한다. 본배양이 완료된 배양액을 원심분리나 세라믹필터를 이용하여 유산균과 배양액을 분리한다. 분리된 유산균과 배양액은 예비동결과정을 거쳐 동결건조가 진행되고 최종적인 분말 형태로 가공된다. 수거된 최종 유산균 및 유산균 배양액은 클린벤치에서만 개봉하며, -20℃ 냉동 보관하여 사용한다.

실시예
실시예 1: 병원성 미생물의 배양
상기 분리한 유산균들 중에서 억제활성 세균 (bacteria)의 생육을 저해하는 유산균을 선별하기 위하여, 먼저 세균: 스타필로코커스 아우레우스구균 (Staphylococcus aureus, KCCM 11335), 대장균 (Escherichia coli, KCCM 11234), 바실러스 세레우스 (Bacillus cereus, KCCM 40935), 스타필로코쿠스 에피더미스 (Staphylococcus epidermidis, KCCM 40416), 슈도모나스 (Pseudomonas KCCM 11321), 프로피오니 박테리움 애크네스 (Propionicbacterium acenes, KCCM41747), 살모넬라 (Salmonella enterica subsp. enterica, KCCM 11806), 리스테리아 (Listeria monocytogenes, KCCM40307); 진균 (곰팡이균 포함): 크립토코커스 네오포만스 (Cryptococcus neoformnas, KCCM50785), 칸디다 알비칸스 (Candida albicans, KCCM 11282), 아스페르길루스 푸미가투스 (Aspergillus fumigatus, ATCC MYA-4609), 아스퍼길루스 나이거(Aspergillus niger, KCCM 60332), 푸사리움 옥시스포럼 (Fusarium oxysporum, KCCM 44187), 페니실리움 크리소게늄 (Penicillium chrysogenum, KCCM 60353), 말라세지아 푸르푸르 (Malassezia furfur, KCCM 12679)를 한국미생물보존센터로부터 분양받아, 아래 표 2의 고체영양배지에서 배양하였다. 다음, 생성된 각각의 콜로니를 액체영양배지에 접종하여 각 균의 생육 적정온도 (대장균, 스타필로코커스 아우레우스 및 바실러스의 경우 37℃; 크립토코커스 및 칸디다의 경우 30℃)에서 24시간 정치배양 후 20% (v/v) 글리세롤을 가하고 -80℃ 초저온냉동고에 보관하였다. 아스페르길루스 및 푸사리움 및 페니실리움의 경우, YPD 고체영양배지 (Sigma-Aldrich)에서 7일간 30℃ 배양한 후, PBS (Phosphate buffer saline)용액으로 포자를 수확하고, 20% (v/v) 글리세롤에 첨가한 후 -80℃ 초저온냉동고에서 보관하였다.

실시예 2: 항균 및 항진균 활성 확인
상기 분리한 균주들에 대해, 이들의 항균 및 항진균 활성을 확인하였다. 먼저, MRS 고체배지에 -80℃에서 보관 중인 분리 균주를 각각 접종하고, 37℃에서 24시간동안 배양한 후, 콜로니 (colony)를 취해 PBS 500 ㎕에 현탁하여 후속하는 항균 및 항진균 활성 실험을 위한 시료용액을 준비하였다. 다음, 분리 균주의 항균 및 항진균 활성을 위한 배지를 제조하였다. 이를 위해, 상기 실시예 1에서 배양하여 보관중인 세균 및 진균을 각각의 생육에 적합한 영양배지에 접종하고 24시간 동안 배양하였다. 다음, MRS가 함유된 소프트아가배지 (1%, w/w)를 121℃에서 15분 동안 멸균처리하고 55℃로 식힌 후, 상기 배양한 세균 또는 진균의 배양액을 각각 1% (v/v) 접종하였다. 다음, 상기 세균 또는 진균이 접종된 소프트아가배지를 일회용 페트리접시에 붓고 상온에 두어 항균 또는 항진균 활성 실험을 위한 중층배지를 제조하였다. 상기 수득한 중층배지 위에 멸균된 8 ㎜ 페이퍼디스크 (Advantec, 51020693)를 올려놓고, 상기 유산균 배양액을 50 ㎕씩 점적한 후, 각각의 세균 또는 진균의 최적 생육온도에서 24시간 동안 배양하였다. 배양이 종료된 후, 각 세균 또는 진균에 대한 생육억제환 (clear zone)의 지름을 측정하여, 분리된 유산균의 항균활성 및 항진균 활성을 측정하였다. 생육억제환의 크기가 8 mm 이상: +, 13 mm 이상: ++, 15 mm 이상: +++, 17 mm 이상: ++++로 표시하였다.
실시예 3: 본 발명 유산균의 내산성 및 내담즙성 실험
내산성은 소화관 중 위(stomach)에 유산균제제가 통과할 때 위산(gastric acid)에 노출되게 되는데 이러한 환경을 시험관 조건에서 실험하였다. Lactobacilli MRS broth에 순수 분리된 균주를 16시간 이상 배양시킨 후 균 배양액 100 μL를 96 well plate에 2가지 조건(pH 7.0, pH 2.5)을 지닌 MRS broth 100 μL에 각각 접종하여 37℃에서 24시간 배양하였다. Microplate reader를 이용하여 흡광도 620 nm에서 pH 7.0 배양액과 pH 2.5 배양액의 흡광도를 비교하여 생존률을 측정하였다.
내산성 확인 실험에 대한 생존률을 측정은 하기식으로 수행하였다.

내담즙성 확인을 위한 실험 방법은 Lactobacilli MRS broth에 순수 분리된 균주를 16시간 이상 배양시킨 후 균 배양액 100 μL를 96 well plate에 2가지 조건(MRS broth, MRS broth+0.3% oxgall)을 지닌 MRS broth 100 μL에 각각 접종하여 37℃에서 24시간 배양하였다. Microplate reader를 이용하여 흡광도 620 nm에서 흡광도를 비교하여 생존률을 측정하였다. 내담즙성 확인 실험에 대한 생존률을 측정은 하기 식으로 수행하였다.

본 발명의 신규 유산균의 내산성 및 내담즙성은 각각 86.87% 및 99.37%로서, 일반적인 기준인 70%를 넘는 우수한 특성을 갖는 것으로 확인되었다.
실시예 4: 장내 상피세포 (CaCO2 cell) 및 피부 각질세포 (HaCaT cell)를 이용한, 본 발명 유산균의 TJ (tight junction) 구성 단백질 발현 유도 확인
장내 상피세포인 CaCo2 세포 (한국세포주은행으로 부터 입수)를 이용하여 장내 TJ 구성 단백질의 발현 유도 여부를 확인하였다. 그 결과 도 6에 나타낸 것과 같이, 본 발명의 유산균 배양액이 ZO-1, Occludin, Claudin-4 단백질의 발현을 효과적으로 유도하는 것이 확인되었다. 다음 피부 각질세포 (HaCaT cell; 한국세포주은행으로부터 입수)을 사용하여 피부 각질의 TJ 구성 단백질의 발현 유도를 확인하였다. 그 결과 도 7에 나타낸 것과 같이, 본 발명의 유산균 배양액이 피부 각질세포에서 ZO-1, Occludin, Claudin-1, 및 Claudin-4 단백질의 발현을 효과적으로 유도하는 것이 확인되었다. 구체적으로, 먼저 유산균 배양액분말을 정량하여 PBS에 녹였다. 다음 미생물 오염을 방지하기 위해 0.2 ㎛ syringe filtration 진행하고, 6 well plate에 알맞은 농도로 처리하였다. 다음, 37℃ CO2 incubator에서 24시간 배양한 후, RIPA/PI buffer를 이용하여 cell lysis를 수행하였다. 다음, Bradford assay를 통하여 단백질을 정량한 후, SDS-PAGE를 수행하였다. SDS-PAGE는 각 레인당, 20 ㎍으로 정량하여 로딩하였으며, 150 V에서 1시간 30분 러닝 (running), 100V에서 1시간 트랜스퍼 (transfer) (NC membrane)한 후, 1시간 (4% skim milk) 동안 블로킹하였다. 다음, 1차 항체로서 Claudin-1 (1:1000)을 16시간 처리하고, β-actin (1:1000)을 1시간 처리하였다. 2차 항체로서 Claudin-1 (anti-rabbit, 1:2000)을 1시간 처리하고, β-actin (anti-mouse, 1:2000)을 1시간 처리 하였다. 끝으로, ECL kit를 이용하여 밴드를 검출하였다.
실시예 5: 본 발명의 유산균의 항균활성 시험 (MIC, Minimum Inhibitory Concentration)
최소저해농도 (MIC) 측정을 표준시험방법 (Wiegand et al. Nat Protoc. 2008)에 따라 수행하였다. 먼저, 멸균된 시험용 고체영양배지 (MHB, Muller Histone Broth, Difco)에 E.coli, S.aureus 및 B.cereus를 각각 접종하여 배양하였고; 영양배지 (YM, Difco)에는 C.neoformans 및 C.albicans; 영양배지 (YPD, Difco)에는 A.fumigatus를 각각 접종하고 37℃로 24시간 배양하였다. 다음, 세균은 멸균된 액체배지 (MHB)에, 진균은 RPMI1640 액체배지에 접종하였다. 다음, 상기 실시예에서 제조한 배양액을 가한 후, 37℃에서 24시간 배양하여 병원성 미생물의 혼탁도를 측정하였다. 상기 배양액은 동결건조 된 유산균 배양액 분말을 3차 증류수와 함께 용해시켰으며 테스트 농도는 농축 배양액을 연속해서 2배씩 희석하며 병원균의 가시적인 성장을 저지하는 가장 낮은 배양액의 농도를 측정하여 나타냈다. 항균 및 항진균 활성을 확인하기 위하여 3회 반복 실시하였으며, microplate spectrophotometer (Multiskan FC A28696)를 활용하여 배양액의 OD 값 (OD620nm)을 측정하였다. 표 3에 나타낸 것과 같이, 본 발명의 유산균 배양액은 그램양성 뿐만아니라 그램음성 세균, 및 진균에 대해 광범위한 억제 활성을 나타냈다.
실시예 6: 항곰팡이 활성 (MIC, Minimum Inhibitory Concentration)
항곰팡이 활성 및 MIC 시험을 표준시험방법 (CLSI document M38-A2, 2008)에 따라 수행하였다. 상기 실시예에서 제조한 배양액을 단계적으로 두 배씩 희석하였다. 각각의 96웰의 각 웰에 곰팡이 포자를 희석하여 2x103spore/ml 농도로 접종하였다. 플레이트는 35°C에서 48시간 배양하였다. MIC는 현미경 관찰을 통하여 시험 배양의 가시적인 성장을 저지하는 배양액의 최하 농도로 결정하였으며 항곰팡이 활성을 확인하기 위하여 실험은 3회 반복하였다. 표 3은 그 결과를 나타낸다.

실시예 7: 열 안정성
유산균 배양액에 의한 상기 병원균의 항균 및 항진균 활성이 열에 대한 안정성을 가지는지 알아보기 위한 실험을 수행하였다. 상기 실시예와 동일한 방법으로 제작한 농축 배양액을 각각 비 열처리, 열처리 그룹으로 나눠 항균 및 항진균 활성을 보이는 최소 저해농도를 측정하고자 하였다. 열처리 실험은 각각 80℃ 항온수조에 30분, 고압멸균기(바이오프리, BF60AC)에 121℃ 15분 열처리한 배양액을 이용하여 실험하였다. 실험 결과는 하기 표 4에 표시하였다.

실시예 8: pH 안정성
유산균 배양액에 의한 상기 병원균의 항균 및 항진균 활성이 pH에 대한 안정성을 가지는지 알아보기 위한 실험을 수행하였다. 상기 실시예와 동일한 방법으로 제작한 농축 배양액을 각각 pH 비 조절 및 조절 그룹으로 나눠 항균 및 항진균 활성을 보이는 최소 저해농도를 측정하고자 하였다.
pH 조절 실험은 latic acid와 NaHCO3를 이용하여 pH를 조절한 배양액을 이용하여 실험하였다. 실험 결과는 하기 표 5에 표시하였다.

실시예 9: 본 발명의 유산균의 억제활성의 선택성
본 발명의 유산균 (Pediococcus pentosaceus AB160011)과 장내 유익균인 락토바실러스 브레비스 (Lactobacillus brevis)와 유해균인 대장균 (Escherichia coli)를 각각 OD600 0.8이 되도록 배양하였다. 다음, Pediococcus pentosaceus AB160011은 0.5, 1 및 1.5 ㎖; Lactobacillus brevis와 Escherichia coli는 각각 1 ㎖씩을 윈심분리기를 이용하여 상등액을 제거하고, 침전된 균체를 DW로 풀어주면서 세척하였다. 다음, 다시 원심분리를 수행 한 후 상등액을 제거하고 Pediococcus pentosaceus AB160011은 DW 5 ㎕에 풀어주고 Lactobacillus brevis와 Escherichia coli는 DW 1 ㎖에 풀어주었다. 다음, DW 1 ㎖에 풀어준 Lactobacillus brevis와 Escherichia coli를 각각 MRS 고체배지에 골고루 펼쳐주고 완전히 마른 후에 각각 DW 5 ㎕에 Pediococcus pentosaceus AB160011을 일정거리를 두고 스폿팅하였다. 6 ~ 20시간 후에 결과를 확인하였다. 그 결과를 도 8에 나타냈다.
실시예 10: 본 발명의 유산균의 염증 유도 실험
염증반응지표인 Nitric Oxide을 양을 측정하였다. 먼저 RAW 264.7 세포를 12웰 플레이트에 각 웰당 1 X 10^5개의 세포로 시딩하였다. 하룻 밤 경과 후, 본 발명의 Pediococcus pentosaceus AB160011 배양액을 동결건조한 샘플 0.2 g을 1 ㎖ DMEM에 녹인 후, 각각 0.1, 0.25, 0.3, 0.35, 0.4, 0.45 및 0.5% (wt/v)의 농도로 처리하고 24시간 배양하였다. 다음, BioVisoin의 Ntric Oxide Colorimetric Assay kit를 이용하여 540 nm에서 흡광도를 측정함으로써 NO 함량을 확인하였다. 그 결과 본 발명의 유산균 배양액은 염증반응을 유도하지 않는 것으로 나타났다. 그 결과를 도 9에 나타냈다.
RAW 264.7 cell을 12well plate에 각 well당 1X10^5개의 세포를 시드 접종 후, 다음날 Pediococcus pentosaceus AB160011를 각각 5X10^7, 1X10^8, 2X10^8 cfu/ml의 농도로 2시간 처리 후 BioVisoin의 Ntric Oxide Colorimetric Assay kit를 이용하여 540nm의 흡광도에서 NO의 양을 측정하였다. 그 결과 본 발명의 유산균은 염증반응을 유도하지 않는 것으로 나타났다. 그 결과를 도 10에 나타냈다.
실시예 11: 본 발명의 유산균의 항염증 효과 실험
RAW 264.7 cell을 12well plate에 각 well당 1X10^5개의 세포를 시드 접종 후, 다음날 Pediococcus pentosaceus AB160011를 각각 1X10^8, 2X10^8 cfu/ml의 농도로 2시간 처리 후 LPS 1㎍/㎖을 24시간 처리하여 염증반응을 유도하였다. BioVisoin의 Ntric Oxide Colorimetric Assay kit를 이용하여 540nm의 흡광도에서 NO의 양을 측정하였다. 그 결과 본 발명의 유산균은 염증유도 반응을 억제하는 것으로 나타났다. 그 결과를 도 11에 나타냈다.
실시예 12: 항산화효과 측정
DPPH (1,1-diphenyl-2-picrylhydrazyl)은 화학적으로 안정화된 프리 라디칼을 가지고 있는 수용성 물질로서 515 ㎚ ~ 520 ㎚ 부근에서 최대 흡광도를 가지며, 항산화활성이 있는 물질과 만나면 전자를 내어주면서 라디칼 (DPPH)이 소멸되고 변색된다. 화학적으로 안정성 있는 1.1-diphenyl-2-picrylhydrazyl radical(DPPH)은 여러 종의 항산화 성분이 내재된 추출물, 음료와 오일, 순수 페놀화합물 등의 항산화 효과를 분석 할 수 있는 것으로 알려져 있다. 0.2 mM의 DPPH 500 ㎕와 배양액을 각각 0.1, 0.25, 0.3, 0.35, 0.4, 0.45 및 0.5 (wt/v)% 농도(0.1, 0.25, 0.5, 0.75, 1, 1.5 및 2 (wt/v)% 농도)로 DPPH와 섞어준 후, 최종 1 ㎖가 되도록 DW로 조절하였다. 다음, 암실에서 30분간 반응시킨 후 540 ㎚에서 흡광도를 측정하였다. 그 결과 도 12에 나타낸 것과 같이, 본 배양액은 농도 의존적으로 항산화 효과를 증가시키는 것으로 나타났다.
실시예 13: 피부자극대체시험
시험방법으로는 식품의약품안전처 민원인 안내서(안내서-0752-01) 화장품 독성시험 동물대체시험법 가이드라인(V)/ 피부자극시험법에 따라 SKINETHIC 인체피부모델 (EPISKIN)을 이용한 피부자극시험을 한국화학융합시험연구원에서 진행하였다. 시험물질을 인체피부모델에 16 ul 씩 조직 당 1분 간격으로 적용하였고, 클린벤치 안에서 42분 (± 1)간 노출시켰다. 이 후 시험 물질을 제거하고 PBS (1 mL/1회)로 총 25회 세척하였다. 37 ℃, 5 % CO2 세포배양기에서 42 시간 ± 1 시간 배양하여 시험물질이 인체피부모델에 미치는 가역적 손상을 평가하였다. 본시험의 구성은 음성대조군, 양성대조군 및 시험물질군 총 3군으로 각 군은 3개의 반복시료로 설정하였으며, 단회 시험으로 진행하였다. 시험물질의 세포생존율은 86.3 ± 2.9 % 으로 세포 생존율 기준치 50%를 초과하여 산출되어 피부 비자극성으로 판정되었다. 그 결과를 도 13에 나타냈다.
실시예 14: 유산균 배양액 함유 에센스의 피부보습 개선효과
유산균 배양액 함유 에센스의 피부보습 개선효과를 평가하기 위해 한국화학융합시험연구원에 위탁하여 유산균 배양액 함유 에센스의 24시간 수분지속력 및 피부보습 개선 효과에 대한 인체적용시험을 진행하였다. 만 21세에서 59세, 여성 22명을 대상으로 제품 사용 전과 후 Corneometer를 이용한 피부보습 평가, 이상반응 평가를 진행하였다. 피부보습을 확인한 결과, 제품 사용 전과 비교하여 사용 2주 및 4주 후에 이마, 양측 볼, 턱 부위에서 모두 통계학적으로 유의하게 증가하였다 (도 14 참고). 피부과 전문의에 의한 이상반응 평가는 시험 제품을 사용하는 기간 동안 시험대상자에게서 특별한 피부 이상반응에 대한 보고는 없었으며, 피부과 전문의에 의한 이학적 검사 상에서도 이상소견은 관찰되지 않았다.
본 발명은 한국 전통발효식품인 된장에서 분리한 유산균으로서, 우수한 항균 및 항진균 활성을 지니고 있으며, 피부 및 장내 상피세포의 TJ (tight junction)에 관여하는 단백질의 발현을 유도하는 것이 확인되었다. 또한, 항염 및 항산화 효과가 우수하고, 피부에 적용한 임상실험 결과 피부보습 증진 효과도 우수한 것으로 나타났다. 따라서 본 발명의 유산균은 건강기능식품, 화장품 및 보존제로서 광범위한 산업분야에 널리 이용될 수 있을 것이다.
