Abstract
Description
また、本発明は、密着連接(tight junction)を構成するタンパク質の発現を増加させる新規な乳酸菌菌株を提供することを目的とする。
また、本発明は、乳酸菌菌株を含む炎症性腸疾患と自己免疫疾患を改善するための機能性食品を提供することを目的とする。
また、本発明は、本発明の乳酸菌菌株培養液および/または乳酸菌菌株を含む抗菌剤および抗真菌組成物を提供することを目的とする。
また、本発明は、本発明の乳酸菌菌株培養液および/または乳酸菌菌株を含む化粧料組成物を提供することを目的とする。
(1)乳酸菌分離
抗菌および抗真菌活性を有する乳酸菌菌株を分離するために、国内で製造されたキムチ、醤油、および味噌を含む発酵製品から様々な乳酸菌を10倍希釈法を使用して2%(w/w)寒天(agar)が含まれているMRS(Difco 288110)固体培地で分離した。生成されたコロニーは、サイズ、色、特性別に区分して、別のMRS培地に画線法で接種し、スクリーニング対象菌株合計1,785個の乳酸菌を分離した。分離された乳酸菌は、それぞれのコロニーをMRSアガプレートに画線法で接種し、37℃で24時間培養した。分離した乳酸菌は、滅菌したMRS液体培地に接種して37℃で24時間静置培養した後、20%(v/v)グリセロールを添加して-80℃超低温冷凍庫に保管した。
形態学的分析
本発明の菌株を同定するために、MRS平板培地に培養した後、コロニーの形態を顕微鏡で観察した。コロニーの形態は、表1のようであった。図1は、本発明の菌株の顕微鏡写真である。
本発明の新規菌株は、配列のリスト1の配列に示すような16s rRNAの塩基配列を有することが確認された。これを基礎に系統樹研究した結果(図2参照)、分離された菌株は、ペディオコッカスペントサセウス(Pediococcus pentosaceus)AJ3053と高い類似度を示しペディオコッカスペン土砂歳ウスに属するペディオコッカスペントサセウス(Pediococcus pentosaceus)AB160011菌株と命名し、2016年12月07日付で韓国微生物保存センターに寄託番号KCCM11954Pで寄託した。
本発明の新規菌株と高い類似度を示すペディオコッカスペン土砂歳ウス(ATCC25745)との相同性をDNA塩基配列(Gepard、Nucleotide sequences Dot plot)で分析した結果、98.97%の相同性を示した(図3参照)。
本発明の新規乳酸菌菌株を滅菌MRS液体培地に接種し、37℃で、政治培養しながら濁度(Ultrospec2100 pro、OD625nm)と酸度(Orion star A211、pH)の変化を観察した。図4及び図5は、本発明の菌株の成長曲線とpHの変化を示したものである。
表2は乳酸菌培養培地組成を示す。大きく種培養、本培養、菌体/培養液分離、凍結乾燥の段階に区分される。種培養は、最終的な培養液の規模に応じて、最終培養液の10%のボリュームまで培養する過程をいう。最終的な培養された種培養を滅菌された培地に接種して培養したものを、本培養とする。本培養が完了した培養液を遠心分離やセラミックフィルターを利用して、乳酸菌と培養液を分離する。分離された乳酸菌と培養液は、予備凍結過程を経て凍結乾燥が進行して、最終的な粉末状に加工される。回収された最終的な乳酸菌と乳酸菌培養液は、クリーンベンチでのみ開封し、-20℃冷凍保存して使用する。
実施例1:病原性微生物の培養
前記分離した乳酸菌の中から阻害活性細菌(bacteria)の生育を阻害する乳酸菌を選別するためには、まず細菌:スタフィロコッカスアウレウス球菌(Staphylococcus aureus、KCCM 11335)、大腸菌(Escherichia coli、KCCM 11234)、バチルスセレウス菌(Bacillus cereus、KCCM 40935)、スタフィロコッカスエピデルミジス(Staphylococcus epidermidis、KCCM 40416)、シュードモナス(Pseudomonas , KCCM 11321)、プロピオニバクテリウム アクネス(Propionicbacterium acenes、KCCM41747)、サルモネラ(Salmonella enterica subsp。enterica、 KCCM11806)、リステリアモノサイトゲネス(Listeria monocytogenes、KCCM40307); 及び 真菌(カビ菌を含む):クリプトコックスネオフォルマンス(Cryptococcus neoformnas、KCCM50785)、カンジダアルビカンス(Candida albicans、KCCM11282)、アスペルギルスフミガタス(Aspergillus fumigatus、ATCCMYA-4609)、アスペルギルスニガ(Aspergillus niger、KCCM60332)、フザリウムオキシスポーラム(Fusarium oxysporum、KCCM44187)、ペニシリウムクリソゲナム(Penicillium chrysogenum、KCCM60353)、マラセチアフルフル(Malassezia furfur、KCCM12679)を韓国微生物保存センターから分譲受け、下記の表3の固体栄養培地で培養した。その後、生成された各コロニーを液体栄養培地に接種し、各菌の生育適温(大腸菌、スタフィロコッカスアウレウス, バチルスの場合 、37℃; クリプトコックス, カンジダアルビカンスの場合、30℃)で24時間静置培養後20%(v / v)グリセロールを加え-80℃超低温冷凍庫に保管した。アスペルギルス、フザリウム、ペニシリウムの場合、YPD固形栄養培地(Sigma-Aldrich)で7日間、30℃培養した後、PBS(Phosphate buffer saline)溶液に胞子を収穫し、20%(v/v)グリセロールに添加した後-80℃超低温冷凍庫で保管した。
前記分離した菌株に対して、これらの抗菌及び抗真菌活性を確認した。まず、MRS固体培地に-80℃で保管されている分離菌株をそれぞれ接種し、37℃で24時間培養した後、コロニー(colony)を取ってPBS 500μlに懸濁して後続する抗菌及び抗真菌活性実験のための試料溶液を準備した。その後、分離菌株の抗菌及び抗真菌活性のためのバッジを製造した。このため、上記実施例1で培養し、保管中の細菌や真菌をそれぞれの生育に適した栄養培地に接種し、24時間培養した。その後、MRSが含有されたソフトアガ培地(1%、w/w)を121℃で15分間滅菌処理して55℃で冷やされた後、上記培養した細菌や真菌の培養液をそれぞれ1%(v/v)接種した。その後、上記の細菌または真菌が接種されたソフトアガバッジを使い捨てペトリ皿に入れ、室温に置いて抗菌や抗真菌活性実験のための重層培地を製造した。上記得られた重層培地上に滅菌された8mmペーパーディスク(Advantec、51020693)を乗せて、上記乳酸菌培養液を50μlずつ点滴した後、それぞれの細菌または真菌の最適生育温度で24時間培養した。培養が終了した後、各細菌や真菌の生育抑制環(clear zone)の直径を測定し、分離された乳酸菌の抗菌活性と抗真菌活性を測定した。生育抑制環サイズが8 mm以上:+、13 mm以上:++、15 mm以上:+++、17 mm以上:++++で表示した。
耐酸性は、消化管の中で、上記(stomach)に乳酸菌製剤が通過するときに胃酸(gastric acid)にさらされることになるが、これらの環境を試験管条件で実験した。Lactobacilli MRSブロス(broth)に純粋分離された菌株を16時間以上培養した後、菌培養液100μLを96 well plateに2つの条件(pH 7.0、pH 2.5)を持ったMRS broth 100μLにそれぞれ接種し、37℃で24時間培養した。Microplate readerを利用して吸光度620 nmでpH 7.0培養液とpH 2.5培養液の吸光度を比較して生存率を測定した。耐酸性確認実験の生存率を測定は、下記式で実施した。

本発明の新規乳酸菌の耐酸性及び耐胆汁性それぞれ86.87%と99.37%であり、一般的な基準である70%を超える優れた特性を有することが確認された。
腸内上皮細胞であるCaCo2細胞(韓国細胞株銀行から入手)を用いて、腸内TJ構成タンパク質の発現誘導するかどうかを確認した。その結果、図6に示すように、本発明の乳酸菌培養液がZO-1、Occludin、Claudin-4タンパク質の発現を効果的に誘導することが確認された。次の皮膚の角質細胞(HaCaT cell;韓国細胞株銀行から入手)を使用して、皮膚の角質のTJ構成タンパク質の発現誘導を確認した。その結果、図7に示すように、本発明の乳酸菌培養液が皮膚の角質細胞でZO-1、Occludin、Claudin-1、およびClaudin-4タンパク質の発現を効果的に誘導することが確認された。具体的には、まず、乳酸菌培養液粉末を定量してPBSに溶かした。次の微生物汚染を防止するために0.2μmシリンジろ過(syringe filtration)行い、6 well plateに適切な濃度で処理した。その後、37℃ CO2インキュベータ(incubator)で24時間培養した後、RIPA/PIバッファ (buffer)を用いて 細胞溶解(cell lysis)を行った。その後、Bradford assayを介してタンパク質を定量した後、SDS-PAGEを行った。SDS-PAGEは、各レーンごとに、20μgで定量しロードし、150 Vで1時間30分ランニング(running)、100Vで1時間トランスファー(transfer)(NC membrane)した後、1時間(4% skim milk )の間にブロッキングした。その後、1次抗体としてClaudin-1(1:1000)を16時間処理し、β-actin(1:1000)を1時間処理した。2次抗体としてClaudin-1(anti-rabbit、1:2000)を1時間処理し、β-actin(anti-mouse、1:2000)を1時間処理した。最後に、ECL kitを用いてバンドを検出した。
最小阻害濃度(MIC)の測定を標準試験方法(Wiegand et al。Nat Protoc 2008)に基づいて行った。まず、滅菌された試験用の固体栄養培地(MHB、Muller Histone Broth、Difco)に E.coli、S.aureusおよび B.cereusをそれぞれ接種して培養し;栄養培地(YM、Difco)にC.neoformansとC.albicans;栄養培地(YPD、Difco)にA.fumigatusをそれぞれ接種し、37℃で24時間培養した。その後、細菌は、滅菌された液体培地(MHB)に、真菌は、RPMI1640液体培地に接種した。その後、前記実施例で製造した培養液を加えた後、37℃で24時間培養して病原性微生物の濁度を測定した。上記培養液は、凍結乾燥された乳酸菌培養液粉末を3次蒸留水と一緒に溶解させたテスト濃度は、濃縮培養液を連続して2倍希釈し、病原菌の具体的な成長を阻止するための最も低い培養液の濃度を測定して示した。抗菌および抗真菌活性を確認するために、3回繰り返し実施し、マイクロプレート分光光度計 (microplate spectrophotometer)(Multiskan FC A28696)を活用して、培養液のOD値(OD620nm)を測定した。表3に示したように、本発明の乳酸菌培養液は、グラム陽性だけでなく、グラム陰性細菌、および真菌の幅広い抑制活性を示した。
抗カビ活性とMIC試験を標準的な試験方法(CLSI document M38-A2、2008年)に基づいて行った。上記実施例で製造した培養液を段階的に二倍希釈した。それぞれの96ウェルの各ウェルにカビ胞子を希釈して2x103spore/mlの濃度で接種した。プレートは、35°Cで48時間培養した。MICは、顕微鏡観察を通じて試験培養の可視的な成長を阻止する培養液の最低濃度を決定しており 抗カビ活性を確認するために実験は3回繰り返した。表4は、その結果を示す。
乳酸菌培養液による上記病原菌の抗菌及び抗真菌活性が熱に対する安定性を持つことを確認するための実験を行った。上記実施例と同様の方法で作製した濃縮培養液をそれぞれ非熱処理、熱処理のグループに分け抗菌および抗真菌活性を示す最小阻害濃度を測定したいた。熱処理実験は、それぞれ80℃の恒温水槽に30分、高圧滅菌(バイオフリー、BF60AC)に121℃、15分熱処理した培養液を用いて実験した。実験結果は、下記表5に示す。
乳酸菌培養液による上記病原菌の抗菌及び抗真菌活性がpHに対する安定性を持つことを確認するための実験を行った。上記実施例と同様の方法で製作した濃縮培養液をそれぞれpH調整グループと非調整グループに分けた後、抗菌及び抗真菌活性を示す最小阻害濃度をそれぞれ測定した。
このために、latic acidとNaHCO3を利用してpHを調整した培養液を利用して実験した。実験結果は、下記表6に示す。
本発明の乳酸菌(Pediococcus pentosaceus AB160011)と腸内有益菌であるラクトバチラスブレビス(Lactobacillus brevis)と有害菌である大腸菌(Escherichia coli)をそれぞれOD600 0.8になるように培養した。その後、Pediococcus pentosaceus AB160011は0.5、1および1.5ml; Lactobacillus brevisとEscherichia coliは、それぞれ1mlずつ遠心分離機を利用して上澄み液を除去し、沈殿した菌体をDWに入れて洗浄した。その後、再び遠心分離を行った後、上澄み液を除去し、Pediococcus pentosaceus AB160011はDW 5μlに入れて、Lactobacillus brevisと Escherichia coliは、DW 1mlに 入れた。その後、DW 1mlに入れたLactobacillus brevisとEscherichia coliをそれぞれMRS固体培地に均一に広げてくれて、完全に乾いた後、それぞれDW5μlに Pediococcus pentosaceus AB160011を一定の距離を置いてスポッティングした。6〜20時間後に結果を確認した。その結果を図8に示した。
炎症反応の指標であるNitric Oxideを量を測定した。まず、RAW 264.7細胞を12ウェルプレートに、各ウェル当り1x105個の細胞に播種した。一夜経過後、本発明の Pediococcus pentosaceus AB160011培養液を凍結乾燥したサンプル0.2 gを1mlDMEMに溶かした後、それぞれ0.1、0.25、0.3、0.35、0.4、0.45、および0.5%(wt/v)の濃度で処理し、24時間培養した。その後、BioVisoinのNitric Oxide Colorimetric Assay kitを用いて540 nmで吸光度を測定することにより、NOの含有量を確認した。その結果、本発明の乳酸菌培養液は、炎症反応を誘導していないことが分かった。その結果を図9に示した。
RAW 264.7 cellを12well plateに各well当たり1X105個の細胞をシード接種後、翌日 Pediococcus pentosaceus AB160011をそれぞれ5X107、1x108、2x108cfu/mlの濃度で2時間処理した後、BioVisoinのNtric Oxide Colorimetric Assay kitを用いて540nmの吸光度でNOの量を測定した。その結果、本発明の乳酸菌は、炎症反応を誘導していないことが分かった。その結果を図10に示した。
RAW 264.7 cellを12wellプレートに各well当たり1x105個の細胞をシード接種後、翌日 Pediococcus pentosaceus AB160011をそれぞれ1x108、2x108cfu/mlの濃度で2時間処理した後、LPS1μg/mlを24時間処理して炎症反応を誘導した。BioVisoinのNtric Oxide Colorimetric Assay kitを用いて540nmの吸光度でNOの量を測定した。その結果、本発明の乳酸菌は、炎症誘導反応を抑制することが分かった。その結果を図11に示した。
DPPH(1,1-diphenyl-2-picrylhydrazyl)は、化学的に安定したフリーラジカルを持っている水溶性物質として515nm〜520nm付近で最大吸光度を有し、抗酸化活性のある物質と出会うと、電子を出し、ラジカル(DPPH)が消滅して変色する。化学的に安定性のある1.1-diphenyl-2-picrylhydrazyl radical(DPPH)はいくつかの種の抗酸化成分が内在された抽出物、飲み物と油、純粋なフェノール化合物などの抗酸化効果を分析することができることが知られている。0.2 mMのDPPH 500μlと培養液をそれぞれ0.1、0.25、0.3、0.35、0.4、0.45、および0.5(wt/v)%の濃度(0.1、0.25、0.5、0.75、1、1.5、および2(wt/v)%濃度)でDPPHと混ぜた後、最終的な1mlになるようにDWに調節した。その後、暗室で30分間反応させた後、540?で吸光度を測定した。その結果、図12に示すように、本培養液は、濃度依存的に抗酸化効果を増加させることが分かった。
試験方法としては、食品医薬品安全処請願ガイド(ガイド-0752-01)化粧品毒性試験動物代替試験法ガイドライン(V)/皮膚刺激試験法に基づいてSKINETHIC人体の皮膚モデル(EPISKIN)を用いた皮膚刺激性試験を韓国化学融合試験研究院で行った。試験物質を人体の皮膚モデルに16 ulずつ組織ごとに1分間隔で適用し、クリーンベンチ内で42分(±1)の間の露出させた。この後、試験物質を除去し、PBS(1 mL/1回)で、合計25回洗浄した。37℃、5%CO2細胞培養器で42時間±1時間培養し、試験物質が人体の皮膚モデルに及ぼす可逆損傷を評価した。本試験群陰性対照群、陽性対照群及び試験物質群の計3群で構成した. 各群は3つの反復試料で設定し、単回試験を行った。試験物質の細胞生存率は86.3±2.9%で、細胞生存率基準値50%を超えて算出され、皮膚非刺激性と判定された。その結果を図13に示した。
乳酸菌培養液含有エッセンスの皮膚保湿改善効果を評価するため、韓国化学融合試験研究院に委託し、乳酸菌培養液含有エッセンスの24時間水分持続力および皮膚保湿改善効果に対する人体適用試験を行った。満21歳から59歳、女性22人を対象に製品の使用前と後、Corneometerを利用した皮膚保湿評価及び異常反応評価を実施した。皮膚保湿を確認した結果、製品使用前と比較して使用2週及び4週後に額、両頬、あご の部位において、水分含有量が統計学的に有意に増加した(図14参照)。 皮膚科専門医による異常反応評価の結果、試験品を使用する期間中、試験対象者において特別な皮膚異常反応についての報告はなかった。また、皮膚科専門医による理学的検査上でも異常所見は観察されなかった。
