Abstract
Description
ここで、生体試料を測定対象とすることから試料に由来するさまざまな干渉物質の影響による測定誤差が問題となる。特に静注用脂肪乳剤成分の投与を受けている患者や高脂血症の患者の血液試料中には脂肪成分が含まれるため、これらの影響により測定値に誤差が生じる。
不溶性担体粒子を用いた血液試料中の成分の測定方法において、脂肪成分による影響を回避する方法として例えば特許文献1~3が知られている。
特許文献2には、磁性粒子を用いたテストステロンおよびエストラジオールの測定において、血清試料中に脂質が存在することによって生じる誤差を、リパーゼを検体中に添加することにより解消する方法が開示されている。
しかし、特許文献1と特許文献2は、いずれも試料中に存在し測定誤差の原因となる脂質をリパーゼにより分解する方法であり、構成試薬中においてリパーゼ活性を安定に維持する必要がある。
特許文献3には、血清や血漿等の血液試料の脂質成分による濁りを除去するために非イオン界面活性剤である2級直鎖アルコールエトキシレートとともに多核フェノールエトキシレートを用いる方法が開示されている。しかし、あくまでも試料の脂肪成分による濁り除去についての記載にとどまり、ラテックスなどの不溶性担体粒子を用いた免疫凝集測定法に関する具体的な記載はない。
すなわち、本発明は以下の構成を有する。

ラテックス免疫凝集法における血液試料に由来する測定誤差を低減する方法であって、液相中で、イミダゾール又はその誘導体あるいはこれらの塩の存在下、当該試料と、測定対象物質に対する特異的親和性物質を担持するラテックス粒子を接触させる工程を含む、前記低減する方法。

血液試料が血清又は血漿である、<1>に記載の方法。
<3>
凝集反応を測定するときのイミダゾール又はその誘導体あるいはこれらの塩の濃度が37.5mM以上375mM以下である、<1>または<2>に記載の方法。

LTIA(ラテックス免疫凝集法)は、抗原あるいは抗体等の測定対象物質に対する特異的親和性物質を固定化したラテックス粒子を用いて、測定対象物質を測定する方法であり、臨床検査の分野で広く使用されている。LTIAにより測定対象物質である抗原を測定する方法としては、測定対象物質に対する抗体を固定化したラテックス粒子と、測定対象物質である抗原とを反応させ、サンドイッチ型の免疫複合体を形成させ、免疫複合体形成に伴う当該ラテックス粒子の凝集の程度から測定対象物質(抗原)を測定する方法と、抗原を固定化したラテックス粒子と試料中の抗原(測定対象物質)とを競合させて、当該ラテックス粒子と抗体との免疫複合体の形成を阻害し、免疫複合体の形成阻害に伴う当該ラテックス粒子の凝集阻害の程度から測定対象物質(抗原)を測定する方法に大別することができる。
本発明における試料は、血液試料であり、例えば全血、血清、血漿が挙げられる。
LTIAにおいては、血液試料中に含まれる何らかの成分により、測定対象物質に対する特異的親和性物質を固定化したラテックス粒子に生じるべきでない凝集が生じたり(正の測定誤差)、あるいは生じるべき凝集が生じなかったり(負の測定誤差)することがしばしば発生する。これらは非特異的反応と呼ばれ、様々な測定誤差の原因となることが知られている。
本発明において、「測定誤差を低減する」とは、上記測定値の正又は負の測定誤差を、本来の測定値(真値)に近づけることをさす。
本発明において血液試料に由来する測定誤差とは、血液試料中の成分に由来して生じるなんらかの非特異反応による測定誤差をいい、非特異反応を引き起こす血液試料中の成分としては、乳びや、単純脂質、中性脂肪などの脂質が挙げられる。
本発明は、特に、乳び所見の個体(高脂状態の個体)や脂肪乳剤を点滴された個体等に由来する血液試料において生じる測定誤差を低減する。
脂肪乳剤としては、油脂を乳化剤で乳化して脂肪粒子の平均粒子径を1μm以下、好ましくは0.5μm以下、より好ましくは0.3μm以下に乳化させた水中油型脂肪乳剤が挙げられる。輸液用(静注用)として用いられている各種の脂肪乳剤が使用でき、市販品としては、例えば、イントラリポス(登録商標、大塚製薬工場社製)、イントラファット(登録商標、日本製薬社製)、イントラリピッド(登録商標、フレゼニウスカービジャパン社製)などが挙げられる。いずれも添付文書に記載された成分は、精製大豆油、精製卵黄レシチン、濃グリセリン、水酸化ナトリウム(pH調整剤)である。
LTIAの測定方法としては、生じた凝集の程度を光学的あるいは電気化学的に観察することにより被検物質を測定できる。光学的に観察する方法としては、散乱光強度、吸光度、又は透過光強度を光学機器で測定する方法(エンドポイント法、レート法等)が挙げられる。試料を測定して得た吸光度等の測定値を、標準物質(測定対象物質の濃度が既知の試料)を測定して得た吸光度等の測定値と比較して、試料中に含まれていた測定対象物質の濃度(定量値)を算出する。なお、透過光又は散乱光などの吸光度等の測定は、1波長測定であっても、又は2波長測定(2つの波長による差又は比)であってもよい。測定波長は、500nmから800nmの中から選ばれるのが一般的である。
本発明における測定対象物質としては、免疫学測定方法で測定される物質であれば特に限定されず種々の物質、例えば、タンパク質(抗原、ハプテン、抗体)、糖質、脂質、核酸、化学物質(ホルモン、薬剤)等などを挙げることができる。中でも、抗原を測定対象物質とするものが好ましく、蛋白抗原がより好ましい。具体的には、例えば、可溶性IL-2レセプター(sIL-2R)、CRP、フィブリン及びフィブリノーゲン分解産物、Dダイマー、可溶性フィブリン(SF)、リポ蛋白(a)(Lp(a))、マトリックスメタロプロテアーゼ-3(MMP-3)、前立腺特異抗原(PSA)、IgG、IgA、IgM、IgE、IgD、抗ストレプトリジンO、リウマチ因子、トランスフェリン、ハプトグロビン、α1-アンチトリプシン、α1-アシドグリコプロテイン、α2-マクログロブリン、ヘモペキシン、アンチトロンビン-III、α-フェトプロテイン、CEA(カルシノエンブリオニツク抗原)、フェリチン、HBs-Ag(B型肝炎外被抗原)、Anti-HBs(抗B型肝炎外被抗体)、HBe-Ag(B型肝炎e抗原)、Anti-HBe(抗B型肝炎e抗体)、Anti-HBc(抗B型肝炎コア抗体)などを挙げることができるが、これらに限定されるものではない。
本発明において、ラテックスに担持される測定対象物質に対する特異的親和性物質としては、タンパク質、ペプチド、アミノ酸、脂質、糖質、核酸、ハプテンなどが挙げられ、分子量の高低および天然、合成といった由来に特に制限はないが、免疫反応を利用する免疫学的測定法に使用され得る抗体または抗原が挙げられる。
本発明に用いる抗体は、ポリクローナル抗体でもモノクローナル抗体でもよい。より好ましくは、モノクローナル抗体である。
本発明の抗体としては、抗体分子全体のほかに抗原抗体反応活性を有する抗体の機能性断片を使用することも可能である。一般的な動物(マウス、ヤギ、ヒツジなど)への免疫工程を経て得られたもののほか、遺伝子組み換え技術等により免疫原(測定対象物質)を免疫する動物とは異なる動物種のアミノ酸配列に変化させた抗体(キメラ抗体、ヒト化抗体、又は完全ヒト化抗体等)であってもよい。抗体の機能性断片としては抗原抗体反応活性を有する断片であるF(ab')2、Fab'や一本鎖抗体(scFv)などが挙げられる。これらの抗体の機能性断片は前記のようにして得られた抗体をタンパク質分解酵素(例えば、ペプシンやパパインなど)で処理することにより製造できる。
本発明に用いるラテックス粒子としては、免疫学的測定試薬として一般的に用いられているラテックス粒子であれば特に制限されない。ラテックス粒子は、種々のモノマーを重合又は共重合させることによって得ることができる。ここにモノマーとしては、例えば、スチレン、α-メチルスチレン,o-メチルスチレン、p-メチルスチレン、p-クロロスチレン、4-ビニル安息香酸、ジビニルベンゼン、ビニルトルエン等のフェニル基を有する重合性単量体、スチレンスルホン酸塩、ジビニルベンゼンスルホン酸塩、o-メチルスチレンスルホン酸塩、p-メチルスチレンスルホン酸塩等のフェニル基及びスルホン酸塩を有する重合性単量体、1-ビニルナフタレン、2-ビニルナフタレン、(メタ)アクリル酸α-ナフチル、(メタ)アクリル酸β-ナフチル等のナフチル基を有する重合性単量体などの重合性不飽和芳香族類、例えば(メタ)アクリル酸、イタコン酸、マレイン酸、フマール酸等の重合性不飽和カルボン酸類、例えば(メタ)アクリル酸メチル、(メタ)アクリル酸エチル、(メタ)アクリル酸-n-ブチル、(メタ)アクリル酸-2-ヒドロキシエチル、(メタ)アクリル酸グリシジル、エチレングリコール-ジ-(メタ)アクリル酸エステル、(メタ)アクリル酸トリブロモフェニル等の重合性不飽和カルボン酸エステル類、(メタ)アクリロニトリル、(メタ)アクロレイン、(メタ)アクリルアミド、N-メチロール-(メタ)アクリルアミド、メチレンビス(メタ)アクリルアミド、ブタジエン、イソプレン、酢酸ビニル、ビニルピリジン、N -ビニルピロリドン、塩化ビニル、塩化ビニリデン、臭化ビニル等の不飽和カルボン酸アミド類、重合性不飽和ニトリル類、ハロゲン化ビニル類、共役ジエン類等を挙げることができる。これらのモノマーは、要求される表面特性、比重等によって適宜選択され、1種を単独で又は2種以上を混合して使用することができる。
ラテックス粒子の平均粒径は、測定対象物質の試料中での濃度あるいは測定機器の検出感度などを考慮し、好ましくは0.02~1.6μm、より好ましくは0.1μm~0.4μmのものが適宜選択される。使用されるラテックス粒子は、感度向上等の所望の性能を得るため、材質や粒子径を適宜選択することができ、材質や粒子径が異なるものを組み合わせて使用することもできる。 また、本発明における凝集反応測定時のラテックス粒子の濃度は特に制限がなく、所望の感度や性能に応じて適宜設定することができる。
測定対象物質に対する特異的親和性物質は、物理的吸着(疎水結合)法、化学的結合法又はこれらの併用等の公知の方法によりラテックス粒子に固定化して担持させることができる。
本発明においては、イミダゾール又はその誘導体あるいはこれらの塩を、測定反応液中又は測定試薬中に存在又は含有させることにより、試料中の測定対象物質の測定を行う。
ここで、イミダゾール又はその誘導体としては、イミダゾール骨格を持つ化合物であればよく、例えば、イミダゾール、1-メチルイミダゾール、1-エチルイミダゾール、1-プロピルイミダゾール、1-ブチルイミダゾール、1-フェニルイミダゾール、1-ビニルイミダゾール、1-アリルイミダゾール、1-ベンジル-2-メチルイミダゾール、1-ベンジル-2-フォルミルイミダゾール、1-ベンジル-4-ヒドロキシメチルイミダゾール、1-ベンジル-5-ヒドロキシメチルイミダゾール、1-(2-ヒドロキシエチル)-イミダゾール、1-(2-ヒドロキシエチル)-2-メチルイミダゾール、2-メチルイミダゾール、2-エチルイミダゾール、2-プロピルイミダゾール、2-ブチルイミダゾール、2-フェニルイミダゾール、2-フォルミルイミダゾール、2-ヒドロキシメチルイミダゾール、2-メチル-1-ビニルイミダゾール、2-ブチル-4-フォルミルイミダゾール、2-ブチル-4-ヒドロキシメチルイミダゾール、2-ブチル-4-クロロ-5-フォルミルイミダゾール、2-ヒドロキシメチル-1-ベンジルイミダゾール、2-ヒドロキシメチル-2-メチルイミダゾール、2-エチル-4-メチルイミダゾール、4-ブチルイミダゾール、4-フォルミルイミダゾール、4-フォルミル-1-メチルイミダゾール、4-フォルミル-1-トリシルイミダゾール、5-フォルミル-1-メチルイミダゾール、4-フォルミル-5-メチルイミダゾール、4-ヒドロキシメチルイミダゾールヒドロクロライド、メチルイミダゾール-4-カルボキシレート、エチルイミダゾール-4-カルボキシレート、1,2-ジメチルイミダゾール、又は1,2,4-トリメチルイミダゾール等の公知のものを挙げることができる。
イミダゾール又はその誘導体の塩とは、化学的に許容される塩であり、カリウム塩、マグネシウム塩、リチウム塩、カルシウム塩または亜鉛塩のほか、アジピン酸塩、アルギン酸塩、クエン酸塩、アスパラギン酸塩、安息香酸塩、ベンゼンスルホン酸塩、重硫酸塩、酪酸塩、樟脳酸塩、カンファースルホン酸塩、ジグルコン酸塩、グリセロリン酸塩、ヘミ硫酸塩、ヘプタン酸塩、ヘキサン酸塩、フマル酸塩、セスキ(フマル酸塩)、塩化水素酸塩、二塩化水素酸塩、三塩化水素酸塩、臭化水素酸塩、ヨウ化水素酸塩、2-ヒドロキシエタンスルホン酸塩(イセチオン酸塩)、乳酸塩、マレイン酸塩、メタンスルホン酸塩、ニコチン酸塩、2-ナフタレンスルホン酸塩、シュウ酸塩、パモ酸塩、ペクチニン酸塩、過硫酸塩、3-フェニルプロピオン酸塩、ピクリン酸塩、ピバル酸塩、プロピオン酸塩、コハク酸塩、硫酸塩、ビス(酒石酸塩)、酒石酸塩、(L)酒石酸塩、ビス((L)酒石酸塩)、(D)酒石酸塩、ビス((L)酒石酸塩)、(DL)酒石酸塩、ビス((DL)酒石酸塩)、メソ酒石酸塩、ビス(メソ酒石酸塩)、チオシアン酸塩、リン酸塩、グルタミン酸塩、炭酸水素塩、ビス((D)酒石酸塩)、ビス(臭化物)、ビス(硫酸塩)、ビス(リン酸塩)、トリス(塩化水素酸塩)、p-トルエンスルホン酸塩、およびウンデカン酸塩からなる群から選択され塩が挙げられる。
好ましい濃度範囲としては、37.5~750mMの範囲のほかに、上記上限と下限の組み合わせが挙げられ、たとえば、37.5~675mM、37.5~600mM、37.5~525mM、37.5~450mM、37.5~375500mM、37.5~300mMにあることが好ましく、より好ましい範囲は37.5~225mM、45~225mM、52.5~225mM、60~225mM、67.5~225mMであり、さらに75~225mMの範囲が特に好ましい。
また、イミダゾール等の濃度は、375mM以上含有させても問題はないが、375mMまででも充分な効果が得られる。更に、前記のイミダゾール等は単独で用いてもよいし、複数のイミダゾール等を併用してもよい。また、複数のイミダゾール等を併用する場合には、両者を併せた濃度が上記の濃度範囲であればよい。
本発明の測定用試薬に含まれる「イミダゾールを含有する緩衝液」は、イミダゾール緩衝液でもよいし、イミダゾール等を含むイミダゾール緩衝液以外の緩衝液でもよい。また、イミダゾール緩衝液以外の緩衝液とイミダゾール緩衝液を組み合わせて使用することも可能である。
イミダゾール等の存在下で測定対象物質を含む試料と、測定対象物質に対する特異的親和性物質を担持するラテックス粒子とを接触させるとは、当該試料と、測定対象物質に対する特異的親和性物質を担持するラテックス粒子とを反応させる反応液中にイミダゾール等が含まれていればよく、典型的には、以下の(1)~(4)が挙げられる。
(1)当該試料とイミダゾール等を含む緩衝液を混合した後、この混合液に測定対象物質に対する特異的親和性物質を担持するラテックス粒子を含む試薬とを混合する態様、
(2)当該試料と、イミダゾール等を含む緩衝液と、測定対象物質に対する特異的親和性物質を担持するラテックス粒子を含む試薬と、を同時に混合する態様、
(3)当該試料と、測定対象物質に対する特異的親和性物質を担持するラテックス粒子を含む試薬とを混合した後に、この混合液にイミダゾール等を含む緩衝液を添加して混合する態様
(4)測定対象物質に対する特異的親和性物質を担持するラテックス粒子を含む試薬と、イミダゾール等を含む緩衝液とを混合した後に、当該試料を添加して混合する対象
本発明の測定用試薬は、2つ以上の構成試薬により構成され、少なくとも1つの構成試薬は測定対象物質に対する特異的親和性物質を担持するラテックス粒子を含み、当該構成試薬と同一の構成試薬及び/または異なる構成試薬はイミダゾール等を含む。また、本発明の測定用試薬は、液状の試薬である。
したがって、2試薬型、3試薬型において、イミダゾール等は全ての構成試薬に含まれていてもよいし、ラテックス粒子を含まない構成試薬の全てに含まれていてもよい。
イミダゾール等の各構成試薬中における濃度は、測定時である、試薬と試料の混合状態において37.5mM以上375mM以下に調整しうるような形態で含まれていればよく、各試薬型により異なる。
本発明の試薬キットは、少なくとも下記(1)および(2)
(1)イミダゾール等を含有する緩衝液を含む第1試薬
(2)測定対象物質に対する特異的親和性物質を担持するラテックス粒子を含む第2試薬
の要素を含むことを特徴とする。イミダゾール等の緩衝液中における濃度は、測定時である、試薬と試料の混合状態において37.5mM以上375mM以下に調整しうるような形態で含まれていればよく、各試薬型により異なるが、例えば、第1試薬と第2試薬が3:1で、採取試料が微量である場合は、第1試薬である緩衝液中のイミダゾール等の濃度は、50mM以上500mM以下が好ましい。
また、本発明の試薬キットには、上記測定試薬のほかに、使用説明書、試料採取用具(採取ピペット、シリンジ、綿棒、ろ過フィルターなど)、試料希釈液、試料抽出液を含むことができる。
本発明の測定誤差低減剤は、ラテックス免疫凝集法における血液試料に由来する測定誤差を低減するための薬剤であり、少なくともイミダゾール等を有効成分として含む。上記測定試薬中のイミダゾール等を含む試薬をそのまま使用することができる。
本発明においてホモジーニアス法とは、試料と試薬液の混和溶液(反応液)中で測定対象物質により進行する反応をB/F(結合/非結合)分離を行うことなく特異的に検出する測定法を指し、B/F分離操作によって測定反応に関与しなかった余剰成分を完全に洗浄・除去した後、主反応を進行させて検出するヘテロジーニアス測定法と対比して呼称される測定法のことをいう。したがって、本発明でいう「ラテックス免疫凝集法が、ホモジーニアス法にもとづく方法である」とは、典型的な下記(1)~(3)の工程において、
(1)液相中で、試料とイミダゾール等とを接触させる工程
(2)(1)工程の後に、当該測定対象物質に対する特異的親和性物質を担持するラテックス粒子を液相中に添加する工程
(3)(2)の工程の後に、当該測定対象物質と当該ラテックス粒子の凝集反応を測定する工程
(3)の工程は、「(2)の工程の途中、あるいは(2)の工程の後に、洗浄・分離工程を経ることなく当該測定対象物質と当該ラテックス粒子の凝集反応を測定する工程」であることを意味する。
本発明の試薬は、不溶性担体粒子の凝集形成を増強する成分としてポリエチレングリコール、ポリビニルピロリドン、リン脂質ポリマーなどの高分子を含んでもよい。また、凝集の形成をコントロールする成分として、タンパク質、アミノ酸、糖類、金属塩類、界面活性剤類、還元性物質やカオトロピック物質など汎用される成分を1種類、または複数の成分を組み合わせて含んでもよい。
血液試料中の脂質による測定誤差の影響を確認し、緩衝液種類、本発明のイミダゾール等の添加による測定誤差低減効果の検討をおこなった。
1.試薬
(1)第1試薬
以下に示す成分(濃度は、試薬中の濃度を示す。以下同じ。)を含む。
・表1に示す緩衝剤
・500mM NaCl
・1.0% BSA
・0.05% Proclin300
(2)第2試薬
下記の2種類の抗体感作ラテックス粒子溶液を等量混合し、5mM MOPS緩衝液(pH7.0)で波長600nmの吸光度が5.0Abs.となるように希釈して第2試薬とした。
(i)92212抗体感作ラテックス粒子溶液
平均粒子径0.3μmの1.0%ラテックス溶液(5mM トリス緩衝液(以下、Tris-HCl又は単にTrisという)(pH8.5)に、等量の5mM Tris-HCl(pH8.5)で0.36mg/mLに希釈した92212抗体溶液を添加して4℃2時間攪拌した。その後、等量の0.5% BSA含有5mM Tris-HCl(pH8.5)を添加して4℃、1時間攪拌し、92212抗体感作ラテックス粒子溶液を作製した。
(ii)92204R抗体感作ラテックス粒子溶液
平均粒子径0.3μmのラテックスを用いて上記(i)と同じ方法により92204R抗体感作ラテックス粒子溶液を作製した。
無作為抽出し混合した血清A(プール血清A)
(イントラリポスの濃度は、試薬と混合する前の試料中の濃度を示す。以下同じ。)
被検試料9/10量に、生理食塩液(大塚製薬)を等量混合した静脈用脂肪乳剤(イントラリポス輸液10%(大塚製薬))を1/10量添加して、0.5%イントラリポス試料を調整した。
被検試料9/10量に、生理食塩液を1/10量添加し、0%イントラリポス試料とした。
被検試料4/5量に、0.5%イントラリポス1/5量を添加し、0.1%イントラリポス試料とした。
第1試薬と第2試薬を組み合わせ、日立7180形自動分析装置を用いて、静脈用脂肪乳剤を含む試料中の可溶性インターロイキン2レセプター濃度を測定した。具体的には、試料5μLに第1試薬150μLを加えて37℃で5分間保温した後、第2試薬50μLを加えて攪拌した。凝集形成に伴う吸光度変化を、その後5分間にわたり、主波長570nm、副波長800nmで測定し、その吸光度変化量を濃度既知の標準物質を測定して得られる検量線にあてはめ、可溶性インターロイキン2レセプター濃度を算出した(試験例1~3において共通)。
0%イントラリポス試料の測定値を100%とし、各濃度のイントラリポス試料の測定値の相対値を算出した(試験例1,2において共通)。
日立7180形自動分析装置のパラメータ条件を以下に示す。
(1)液量 検体―第1試薬―第2試薬;5μL-150μL-50μL
(2)分析法 2ポイントエンド法(測光ポイント19-34)
(3)測定波長 主波長570nm/副波長800nm
(4)キャリブレーション スプライン
0%イントラリポス試料の測定値を100%とした場合に、0.1%イントラリポス試料の測定値が85%未満または115%を超える場合は、イントラリポス共存による影響が起きていると判断した。結果を表1に示す。
第1試薬に緩衝剤としてTris又はHEPESのみを用いた場合には、イントラリポス共存による影響が起きていたが、イミダゾールバッファーのみを用いた場合、あるいはTris又はHEPESとイミダゾールバッファーを組み合わせた場合には、イントラリポス共存の影響は起こらなかった。
免疫凝集反応液中のイミダゾール濃度を変更し、測定誤差低減効果との関係を検討した。
1.試薬
(1)第1試薬
表1に示す緩衝剤を表2に示す緩衝剤に置き換えた以外は試験例1に同じである。
(2)第2試薬
試験例1に同じである。
(1)無作為抽出し混合した血清B(プール血清B)
(2)無作為抽出し混合した血清C(プール血清C)
(3)無作為抽出し混合した血清D(プール血清D)
0.5%イントラリポス試料、0%イントラリポス試料、0.1%イントラリポス試料の調整方法は、試験例1と同じである。0.05%イントラリポス試料は、被検試料9/10量に、0.5%イントラリポス試料1/10量を添加して調整した。
試験例1に同じである。
0% イントラリポス試料の測定値を100%とした場合に、0.05%又は0.1%イントラリポス試料の測定値が、85%以上115%以下場合はイントラリポス共存による影響が起きていない(+)と判断した。
プール血清B~Dのいずれにおいても影響が起きていない場合を+++、いずれか2つにおいて影響が起きていない場合を++、いずれか1つにおいて影響が起きていない場合を+とした。結果を表2に示す。
イミダゾールバッファーの濃度が、50mM~300mMの範囲において、イントラリポスの影響を回避することが可能だった。
測定誤差が、脂質を含む血液試料のみに基づくものか、ラテックスを含む試薬に基づくものか確認するために、第2試薬にラテックスを含まない試薬を用いて検討を行った。
1.試薬
(1)第1試薬
表1に緩衝剤を100mM Tris(pH7.0)に置き換えた以外は試験例1の第1試薬に同じ
(2)第2試薬
生理食塩液
試験例2のプール血清B、プール血清C、プール血清D
試験例2に同じである。
試験例1に同じである。
測定吸光度は0~1.2mAbs.であった。イントラリポス濃度依存的な吸光度の上昇が観察されなかったことから、測定値への影響とはイントラリポスの添加による血液試料の濁りではなく、イントラポリスを含む血液試料との接触によるラテックス粒子の非特異的な凝集によるものであると推測される。
Claims (19)
- ラテックス免疫凝集法における血液試料に由来する測定誤差を低減する方法であって、液相中で、イミダゾール又はその誘導体あるいはこれらの塩の存在下、当該試料と、測定対象物質に対する特異的親和性物質を担持するラテックス粒子を接触させる工程を含む、前記測定誤差を低減する方法。
- 血液試料が血清又は血漿である、請求項1に記載の方法。
- 凝集反応を測定するときのイミダゾール又はその誘導体あるいはこれらの塩の濃度が37.5mM以上375mM以下である、請求項1または2に記載の方法。
- ラテックス免疫凝集法が、ホモジーニアス法にもとづく方法である請求項1~3のいずれかに記載の方法。
- 血液試料中の測定対象物質をラテックス免疫凝集法により測定する方法であって、液相中で、イミダゾール又はその誘導体あるいはこれらの塩の存在下、当該試料と、測定対象物質に対する特異的親和性物質を担持するラテックス粒子を接触させる工程を含む、前記測定方法。
- 血液試料が血清又は血漿である、請求項5に記載の方法。
- 凝集反応を測定するときのイミダゾール又はその誘導体あるいはこれらの塩の濃度が37.5mM以上375mM以下である、請求項5または6に記載の方法。
- ラテックス免疫凝集法が、ホモジーニアス法にもとづく方法である請求項5~7のいずれかに記載の方法。
- 血液試料中の測定対象物質をラテックス免疫凝集法により測定する方法であって、以下の工程を含む、前記測定方法。
(1)液相中で、当該試料とイミダゾール又はその誘導体あるいはこれらの塩とを接触させる工程
(2)(1)工程の後に、当該測定対象物質に対する特異的親和性物質を担持するラテックス粒子を液相中に添加する工程
(3)(2)の工程の後に、当該測定対象物質と当該ラテックス粒子の凝集反応を測定する工程 - 血液試料が血清又は血漿である、請求項9に記載の方法。
- (3)のラテックス粒子の凝集反応を測定する工程のイミダゾール又はその誘導体あるいはこれらの塩の濃度が37.5mM以上375mM以下である、請求項9または10に記載の方法。
- ラテックス免疫凝集法が、ホモジーニアス法にもとづく方法である請求項9~11のいずれかに記載の方法。
- 血液試料中の測定対象物質をラテックス免疫凝集法により測定するための試薬キットであって、以下を含む試薬キット。
(1)イミダゾール又はその誘導体あるいはこれらの塩を含有する緩衝液を含む第1試薬
(2)測定対象物質に対する特異的親和性物質を担持するラテックス粒子を含む第2試薬 - 血液試料中の測定対象物質をラテックス免疫凝集法により測定するときのイミダゾール又はその誘導体あるいはこれらの塩の濃度が37.5mM以上375mM以下に調整しうるような形態で含まれている、請求項13に記載の試薬キット。
- ラテックス免疫凝集法が、ホモジーニアス法にもとづく方法である請求項13または14に記載の試薬キット。
- 血液試料中の測定対象物質をラテックス免疫凝集法により測定するための液状試薬であって、イミダゾール又はその誘導体あるいはこれらの塩を含む液状試薬。
- 血液試料中の測定対象物質をラテックス免疫凝集法により測定するときのイミダゾール又はその誘導体あるいはこれらの塩の濃度が37.5mM以上375mM以下に調整しうるような形態で含まれている、請求項16に記載の液状試薬。
- ラテックス免疫凝集法が、ホモジーニアス法にもとづく方法である請求項16または17に記載の液状試薬。
- イミダゾール又はその誘導体あるいはこれらの塩を有効成分として含む、ラテックス免疫凝集法における血液試料に由来する測定誤差の低減剤。
