本発明は、ペット又はヒトの血糖値スパイクを抑制するための血糖値スパイク抑制剤、食品及び血糖値スパイク抑制剤の製造方法に関するものである。
血糖値スパイクは肥満等の生活習慣病の基本的な要因のひとつであるであるとされ、これを抑制することが肥満の予防につながる。また、血糖値スパイクを抑制できれば、高血糖を特徴とする糖尿病及びそれらの合併症に対して顕著な治療効果または予防効果があるということが知られている(例えば、非特許文献1及び2を参照)。

非特許文献1に記載されているように、肥満したペットは重大な慢性疾患(糖尿病、認知症や腎不全等)になりやすく、飼い主に大きな精神的・経済的な負担を強いることになる。さらに、ペットを失うことによる精神的なダメージも大きい。このように、ペットの問題は、もはやペットの飼い主の問題を越えて、社会全体の問題となった。特に多くの生活習慣病の原因となる肥満の問題は、急激に大きくなっていると考えられる。そこで、ペットが肥満状態になりにくくするサプリメントが求められている。
肥満を引き起こすホルモンとしてインスリンが知られている。インスリンは血糖値のスパイク状の増加によって膵臓のランゲルハンス島のベータ細胞から遊離される。インスリンは糖質から脂肪への変換を促進することによって肥満を引き起こす「肥満ホルモン」としての役割がある。血糖値スパイクを日常的にできるだけ回数を少なくすることと、血糖値スパイクをできるだけゆるやかな増加にすることが、インスリンの恒常的な異常(インスリン耐性)の発生を予防することにつながる。この意味で、肥満を予防するためには、血糖値スパイクを抑制することが重要である。
また、日常の運動量の低下等の生活習慣要因により、ヒトにおいても糖尿病を発症したりインスリン耐性を示したりする人が増えている。老齢化社会への移行に伴い、糖尿病患者が年々爆発的に増加する傾向にあり、わが国の糖尿病患者の増加は深刻である。
糖尿病は根本的な治癒がないから、インスリン耐性を示すヒトやペットに対して、糖尿病を発症させない取組みを実行することが重要である。特に、血糖値スパイクをできるだけ小さくすることが高血糖の改善には重要である。
ペットやヒトにとって高血糖の状態を改善しやすくするために、経口投与が可能な薬剤やサプリメントが求められている。しかしながら、従来、経口投与が可能であり、かつ血糖値スパイクを抑制するための、特にペット用の経口剤が見出されていなかった。
そこで、本発明はこれらの点に鑑みてなされたものであり、経口投与が可能であり、かつ血糖値スパイクを抑制するための血糖値スパイク抑制剤を提供することを目的とする。
本発明の第1の態様においては、ポリ(R)-3-ヒドロキシ酪酸の粉末を含む血糖値スパイク抑制剤を提供する。上記のポリ(R)-3-ヒドロキシ酪酸の割合は、例えば60%以上100%以下である。上記のポリ(R)-3-ヒドロキシ酪酸の割合は、70%以上90%以下であってもよい。
本発明の第2の態様においては、上記のポリ(R)-3-ヒドロキシ酪酸の粉末を含む血糖値スパイク抑制剤が混合された食品を提供する。
本発明の第3の態様においては、ポリ(R)-3-ヒドロキシ酪酸の顆粒を内部に蓄積することができる細菌を含む培養液を準備する準備工程と、前記培養液を固液分離することにより、ポリ(R)-3-ヒドロキシ酪酸の顆粒を含む残渣を回収する回収工程と、前記残渣を乾燥させることにより、ポリ(R)-3-ヒドロキシ酪酸の粉末を生成する生成工程と、を有する血糖値スパイク抑制剤の製造方法を提供する。前記準備工程と前記回収工程との間に、前記培養液を加圧する工程をさらに有してもよい。
本発明によれば、経口投与が可能であり、かつ血糖値スパイクを抑制するための血糖値スパイク抑制剤を提供することができるという効果を奏する。
[本実施の形態の概要]
地球環境問題への世界的関心の高まりとともに、自然界で完全に分解する生分解性プラスチックスへの関心が高まっている。本願の発明者は、一部の微生物が、生分解性プラスチックスとして用いられるポリ(R)-3-ヒドロキシ酪酸(以下、PHBという)の顆粒を蓄積するという性質に着目し、血糖値スパイクを抑制するという目的にPHB粉末を利用することを検討した。本明細書におけるPHB粉末は、本発明で述べるように、例えば化学合成等によって得られるPHBをペレット状にした態様のPHB、菌体内で生合成されたPHBで結晶構造を維持している態様のPHB、又は生合成された菌体内の顆粒状のPHBとは明らかに様態が異なる。

粉末化されていないPHBにも血糖値スパイクを抑制する作用があることは容易に類推ができる。すなわち、粉末化されていないPHBは小腸では生理作用が生じることなく小腸を通過し、大腸において腸内細菌によりHBに加水分解され、動物に吸収され、動物のHB濃度を増加させることができる。HB濃度が持続的に増加すれば、インスリン耐性を改善することは今や定説である(Cell Metabolism 2016年8月9日、第256頁-第268頁)。この作用の発現には、PHBを分解する腸内細菌が十分に生育するまでの時間が必要であり、腸内細菌が十分に生育していなければPHBは殆ど分解されずに排泄されることになる。すなわちPHBは小腸内を通過して大腸内で分解されて吸収されることで効果が発現するので、経口投与をしてから効果が発現するまでに長時間を要するという課題があった。
この課題を解決するために、発明者は、PHBの顆粒を蓄積した微生物から製造したPHB粉末を配合した食品を摂取することにより、PHB粉末を配合していない同じ食品を摂取した場合に比べて、血糖値スパイクの大きさを抑制できるということが確認できた。そして、発明者は、PHB粉末は、粉末化されていないPHBと異なる原理で血糖値スパイクを抑制する作用が生じることから、経口投与をしてから短時間で効果が発現することを発見した。すなわち発明者は、PHBを粉末化することによって、「生理作用の発現に少なくとも数日間必要である」という課題を解決した。

化学合成法によってPHBを生産する場合、触媒を用いて(R)-3-ヒドロキシ酪酸(HB:ケトン体)を化学的に結合させることでPHBを生産することができる。カルボン酸残基を減らして酸度を低くするため、またHBとは異なるPHBとしての性質を持たせるため、重量平均分子量1,000以上のPHBが望ましい。
[血糖値スパイク抑制作用]
図2は、粉末化されていないPHBの血糖値スパイク抑制作用の原理について説明するための図である。従来技術では、動物の血液中のHB濃度を増加させる機能にのみ注意が注がれていた。経口投与されたPHBは胃や腸で分解されることなく大腸まで送達され、大腸において腸内細菌により分解される。そして、数日という時間をかけてHB濃度を非常に緩やかに増加させる。体内のHB濃度が増加することにより、ケトン体濃度が増加することにより血糖値スパイクが抑制されると考えられる。したがって、PHBを数日以上にわたって投与し続けることがなければ、PHBを分解する腸内細菌は十分に生育しないと考えられ、動物のケトン体濃度を有意に増加させるには数日程度必要である。
図2は、粉末化されていないPHBの血糖値スパイク抑制作用の原理について説明するための図である。従来技術では、動物の血液中のHB濃度を増加させる機能にのみ注意が注がれていた。経口投与されたPHBは胃や腸で分解されることなく大腸まで送達され、大腸において腸内細菌により分解される。そして、数日という時間をかけてHB濃度を非常に緩やかに増加させる。体内のHB濃度が増加することにより、ケトン体濃度が増加することにより血糖値スパイクが抑制されると考えられる。したがって、PHBを数日以上にわたって投与し続けることがなければ、PHBを分解する腸内細菌は十分に生育しないと考えられ、動物のケトン体濃度を有意に増加させるには数日程度必要である。
ここで、HB濃度を増加させるためには、HB自体を経口投与することが考えられる。この場合、HB自体は酸性なので、HBのNa塩等が用いられることになる。
図3は、HBを経口投与することにより体内のHB濃度が増加する原理を示す図である。HB塩は胃の酸性環境の中でフリーの酸になり、小腸上皮にある特異的な輸送体(モノカルボン酸トランスポーター)で体内に輸送され、摂取後数分で体内のHB濃度が増加する。その機能発現に必要な時間は数分で十分である。
ところが、HBのNa塩ではNa負荷が問題になる。そこで、ケトンエステル(KE)を用いることも考えられる(Cell Metabolism 2016年8月9日、第256頁-第268頁)。KEは、HBと1,3-ブタンジオールがエステル結合した化合物である。
図4は、KEを経口投与することにより体内のHB濃度が増加する原理を示す図である。KEのエステル結合を加水分解する酵素は哺乳動物も持っており、哺乳動物は、速やかに(数分以内)HBを高濃度に産生することができる。図4に示すように、KEは小腸内の消化酵素で分解されて、2分子のHBを生じる。HBは小腸上皮の特異的な輸送体(モノカルボン酸トランスポーター)で吸収され、速やかにHB濃度を増加させる。したがって、KEもHB塩同様に作用は即効的であり、数分で数mMまで血中のHB濃度が増加するが、効果が発現される期間が2~3時間と短い(Cell Metabolism 2016年8月9日、第256頁-第268頁)。
このように、HBを経口投与する方法、又はKEを経口投与する方法により、HB濃度を増加させ、血糖値スパイクを抑制できる。しかしながら、HB又はKEを経口投与した場合、HB濃度が増加してから数時間でHB濃度が低下してしまう(Cell Metabolism 2016年8月9日、第256頁-第268頁)。
図5は、PHB粉末を経口投与することにより血糖値スパイクが抑制される原理を示す図である。PHB粉末は少なくとも二つの生理作用を生じさせる。一つ目の作用は、PHB重合体のまま小腸でブドウ糖の分解または吸収を抑制する作用である。もう一つの作用は、PHBが大腸の腸内細菌の酵素でHBに分解され、動物の大腸上皮から吸収されて、HB濃度の増加に寄与する作用である。前者の作用は、PHBを粉末化したことによって生じる作用である。PHB粉末を経口投与した場合、小腸でのブドウ糖の吸収又はデンプン(又は2糖類)からブドウ糖への変換を抑制する作用が短時間で生じる。したがって、PHB粉末を経口投与した場合は、血糖値スパイク抑制に対して即効性がある。さらに、PHB粉末が大腸の腸内細菌により分解されてHBが生成される過程に長時間を要するので、HB濃度が高い状態に維持される時間が、HB又はKEを経口投与した場合に比べて長いと考えられる。その結果、PHB粉末を摂取した後に速やかに血糖値スパイクを抑制するとともに、継続的にHB濃度を高い状態に維持することができるので、HB、KE、非粉末のPHBを経口投与する場合の課題を解決することができる。
図6は、HB、KE、非粉末のPHB及びPHB粉末の比較表である。この比較表から明らかなように、PHB粉末を経口投与することにより、短時間で血糖値スパイクを抑制する効果が現れるとともに、効果が長時間にわたって継続し、PHB粉末が血糖値スパイク抑制剤として好適であることがわかる。
[血糖値スパイク抑制剤としてのPHB粉末の製造方法]
PHBは、薬学上許容される溶媒和物、又は懸濁物として、例えば、アルコール懸濁(例えば、メタノール懸濁物、エタノール懸濁物)、エーテル懸濁物とすることができる。

本実施の形態に係る血糖値スパイク抑制剤は、有効成分であるPHB粉末を、生理学的に許容されうる担体、賦形剤、結合剤、希釈剤等と混合することにより製造することができる。血糖値スパイク抑制剤は、経口摂取することができる形態に製造される。経口摂取可能な形態としては、食品、顆粒剤、散剤、錠剤(糖衣錠を含む)、丸剤、カプセル剤、シロップ剤、乳剤、又は懸濁剤等が挙げられる。
血糖値スパイク抑制剤は、薬学的に許容される担体(例えば、賦形剤、添加剤)とともに製剤化することができる。薬学的に許容される賦形剤や添加剤としては、担体、結合剤、香料、緩衝剤、増粘剤、着色剤、安定剤、乳化剤、分散剤、懸濁化剤、防腐剤等が挙げられる。薬学的に許容される担体としては、例えば、炭酸マグネシウム、ステアリン酸マグネシウム、タルク、砂糖、ラクトース、ペクチン、デキストリン、澱粉、ゼラチン、トラガント、メチルセルロース、ナトリウムカルボキシメチルセルロース、低融点ワックス、カカオバター等が挙げられる。
経口剤を製造するためには、まず、有効成分として、例えば賦形剤(乳糖、白糖、デンプン、マンニトール等)、崩壊剤(炭酸カルシウム、カルボキシメチルセルロースカルシウム等)、結合剤(α化デンプン、アラビアゴム、カルボキシメチルセルロース、ポリビニールピロリドン、ヒドロキシプロピルセルロース等)又は滑沢剤(タルク、ステアリン酸マグネシウム、ポリエチレングリコール6000等)を添加して圧縮成形する。
続いて、必要により、味をマスキングすることを目的として、又は腸溶性若しくは持続性を確保することを目的として、圧縮成形した有効成分をコーティングする。コーティング剤としては、例えばエチルセルロース、ヒドロキシメチルセルロース、ポリオキシエチレングリコール、セルロースアセテートフタレート、ヒドロキシプロピルメチルセルロースフタレートおよびオイドラギット(メタアクリル酸・アクリル酸共重合物)等を用いることができる。
本実施の形態に係る血糖値スパイク抑制剤としてのPHB粉末は、PHB粉末を含む組成物または活性化剤の形態で、食品に配合することができる。より具体的には、本実施の形態に係る食品は、本実施の形態に係るPHB粉末を調整することにより製造してもよく、製造された食品に各種タンパク質、糖類、脂肪、微量元素、ビタミン類等をさらに配合して調製したもの、液状、半液体状若しくは固体状にしたもの、ペースト状にしたもの、又は本発明に係る有効成分を含まない飲食品にPHB粉末を添加したものであってもよい。
本発明において「食品」には、健康食品、機能性食品、特定保健用食品、機能性表示食品、又は病者用食品が包含される。さらに「食品」という用語は、ヒト以外の哺乳動物を対象として使用される場合には、飼料やペットフードを包含する。「食品」は、通常の食品の形状であっても、サプリメントのような栄養補助食品の形状であってもよい。
本発明の別の態様によれば、PHB粉末を有効成分として含む食品であって、HBの濃度を増加させることにより治療、予防または改善しうる疾患または状態の治療、予防または改善する機能が表示された食品が提供される。本発明のさらに別の態様によれば、PHB粉末を有効成分として含む食品であって、抗酸化能、解毒能、または、抗炎症能が表示された食品が提供される。
本発明において提供される飲料(飲料形態の健康食品や機能性食品を含む)の製造に当たっては、通常の飲料の処方設計に用いられている糖類、香料、果汁、食品添加剤等を適宜添加することができる。飲料の製造に当たってはまた、当業界に公知の製造技術を参照することができる。
本発明による食品は様々な形態を取ることができ、公知の医薬品の製造技術に準じて本発明による食品を製造してもよい。その場合には、本発明による治療剤の製造の項目において述べたような担体や製剤用添加剤を用いて製造することができ、具体的には、経口剤の欄に記載された担体や製造用添加剤を用いて製造することができる。
血糖値スパイク抑制剤の組成物および食品を投与または摂取する場合、本発明によるPHB粉末の投与量または摂取量は、受容者、受容者の年齢および体重、症状、投与時間、剤形、投与方法、薬剤の組み合わせ等に依存して決定できる。例えば、本発明によるPHB粉末を健康食品として投与する場合、PHB粉末の有効量として、成人1人当たり10~2000mg/kg体重(好ましくは100~1000mg/kg体重)の範囲で一日1回または数回の投与単位に分割して投与することができる。なお、これらの投与量または摂取量は、成人の体重を60kgと仮定して、体重60kgの成人1人1日あたりのPHBの摂取量もしくは投与量として、必要により計算することによって表すことができる。
[実施例]
(PHB粉末の製造)
図7は、血糖値スパイク抑制剤として製造したPHB粉末の外観を示す写真である。PHBは、ハロモナス・エスピー(Halomonas sp.)OITC1261株(NITE P-02027)を用いて発酵させることにより製造した。PHB粉末の形状(特に粒子の大きさ)は、製造工程の最後の粉砕する過程において微調整することができる。

図7は、血糖値スパイク抑制剤として製造したPHB粉末の外観を示す写真である。PHBは、ハロモナス・エスピー(Halomonas sp.)OITC1261株(NITE P-02027)を用いて発酵させることにより製造した。PHB粉末の形状(特に粒子の大きさ)は、製造工程の最後の粉砕する過程において微調整することができる。
まず、原料となるスクロース(蔗糖)と、PHBを内部に蓄積することができる菌体とを含む培養液を準備した。続いて、培養液で菌体を培養した。この間に、菌体はPHBを体内に蓄積する。好気培養後の培養液には、菌体内にPHB顆粒が蓄積されたハロモナス菌、水、無機イオン(硝酸塩、ナトリウム等)が含まれる。OITC1261株はPHBと同時にHBを生産するが、HBは菌体外(培養液中)へ放出されるため、後の固液分離の工程で除去される。PHBは、非常に長い鎖上の構造であるため、菌体内では高度に折りたたまれて、非常に大きな構造(数十から数百ナノメートルの顆粒状の構造)として存在する。図8は、PHB顆粒を蓄積した菌の模式図である。培養条件によっては菌体内の体積の70%がPHB顆粒で占められる。培養液の組成は、例えば、1リットルの蒸留水に対して、炭酸水素ナトリウム12.6g、炭酸ナトリウム5.3g、リン酸水素カリウム2.0g、塩1.0g、硝酸ナトリウム12.5g、硫酸カリウム1.0g、硫酸マグネシウム七水和物40mg、塩化カルシウム二水和物、硫酸鉄(II)七水和物10mg、エデト酸二ナトリウム80mg、である。必要に応じて、5w/v%のグルコースを含んでもよい。必要に応じて、菌を培養中に培養液を追加してもよい。ハロモナス菌を加え、30℃に保温しながら、3~4日間好気培養してもよい。
続いて、菌体を含む培養液を加圧滅菌した。具体的には、1%~2%の界面活性剤を培養液に添加した後にオートクレーブ(1.2気圧、120℃、20分、湿度100%)を数回施した。オートクレーブを施すことにより、菌体内で分子間力又は水素結合で会合することにより顆粒状に存在しているPHBの会合状態が壊れて粉末状になると推定される。その結果、菌体内のPHB顆粒を、数千~数万重合度のPHB粉末にすることができる。オートクレーブを施すことにより、PHB直鎖が会合した高次構造が壊れて、PHB直鎖が会合せずに存在する状態になると推定される。「PHB粉末」は、「PHB直鎖が弱い分子間力や水素結合により会合して高次構造を取る菌体内の顆粒」とは対照的に、PHB直鎖が会合せず存在する状態の物である。このときPHB粉末の重合度は分子量の実測値から推定すると、数千から数万である。
続いて、培養液を加圧滅菌した後に固液分離した。具体的には、菌体を含む発酵液に10000Gの負荷をかけた状態で遠心分離機を5分間回転させた。その後、上澄みを廃棄し、菌体の残渣を得た。得られた残渣は水を加えて懸濁した後、遠心分離を行うという工程を3回繰り返して洗浄することにより固液分離した。
続いて、水溶液を廃棄して、PHB顆粒、小顆粒、菌体(タンパク質、脂肪、炭水化物、水分)を含む残渣を回収した。回収した残渣を100℃で2時間乾燥した後に乳鉢で砕いて、図7に示すPHB粉末を製造した。製造したPHB粉末の約70%の成分がPHBであった。オートクレーブや熱乾燥処理を施すことにより、菌体の細胞膜が破壊されるとともに、弱い分子間力で高次構造を形成していた菌体内のPHB顆粒(数平均重合度1万以上)を、平均重合度数千程度のPHB粉末にすることができる。
上述した濃縮処理は、ハロモナス属のように、PHBを大量に蓄積する細菌を用いて初めて食品で用いることが可能になる。ハロモナス属等の細菌は、体重量の70%までPHBを蓄積することができる。したがって、PHBを蓄積した細菌を用いることで、上述のように70%か以上ら90%以下にまで精製できることを確認できた。濃度の低いPHBを精製するには、クロロホルム等が必要であるが、ハロモナス属等の細菌を用いることで、上述のように70%以上90%以下の範囲で精製できることを確認できた。製造したPHB粉末の分子量分布をGPC法により測定したところ、重量平均分子量は59万であり、数平均重合度は1,939であった。この数値は、201,671(数平均分子量)÷104(HBの分子量)=1,939(数平均重合度)により算出された。

製造したPHB粉末40g(含有されるPHBは28g=70%ポリケトン)をプレーンヨーグルト(250g)に均一になるように混合し、人に食べさせた。平均体重を70Kgとして、PHBの摂取量は体重1kg当たり約400mgである。

(血糖値スパイク抑制効果の確認:図10)
製造したPHB粉末40g(含有されるPHBは36g=90%ポリケトン)をプレーンヨーグルト(250g)に均一になるように混合し、人に食べさせた。平均体重を70Kgとして、PHBの摂取量は体重1kg当たり約514mgである。

続いて、プレシジョンエクシード(アボット)を用いて、1時間おきに血糖値電極(FSプレシジョン血糖測定電極)を用いて血糖値を測定した。PHBとコントロール(プレーンヨーグルトのみ)を食べる前に安定していることを確認するために最低3回の測定を行い、測定結果が略一定であることを確かめた。
図9及び10は、プレーンヨーグルトを摂取した人の体内の血糖値の変化を示すグラフである。図9及び10に示すグラフの横軸は時間である。血糖値が安定しているのを確認して、2時間後にプレーンヨーグルトを摂取した。縦軸は血糖値である。図9または10における白丸は、PHB粉末を混合していないプレーンヨーグルトを摂取した場合の血糖値変化を示しており、黒丸は、PHB粉末(図9及び10においてPHB粉末の純度はそれぞれ70%及び90%である)を混合したプレーンヨーグルトを摂取した場合の血糖値変化を示している。図9及び10に示すように、PHB粉末を混合したプレーンヨーグルトを摂取した場合、摂取してから短時間で血糖値スパイクが小さくなっていることがわかる。さらに、少なくともPHB粉末を混合したプレーンヨーグルトを摂取した場合、長時間にわたって、PHB粉末を混合していないプレーンヨーグルトを摂取した場合に比べて血糖値が低く抑えられることが確認できた。人がヨーグルトを食べると、ヨーグルトに含まれる糖質がすみやかに分解され、血糖値を速やかに増加させる。この血糖値の速やかな増加のためにインスリンスパイクが誘導される。しかしながら、プレーンヨーグルトとともにPHB粉末40gを摂取すると、血糖値スパイクは有意に抑制された。
[本実施の形態の血糖値スパイク抑制剤による効果]
本実施の形態に係る血糖値スパイク抑制剤としてのPHB粉末により、飲食物にPHB粉末を混合することで、短時間で血糖値スパイクを抑制するとともに、長時間にわたって血糖値スパイクを抑制する効果を維持することができる。PHB粉末は無味無臭なので、ペット用食品に混ぜることも容易であるから、ペットに恒常的に毎日摂取することも可能である。小腸での生理作用と想定される血糖値スパイクの抑制作用は、PHB粉末を用いたことによって得られる、本発明が初めて提示した新規性である。図9及び図10に示すように、純度70%のPHB粉末、及び純度90%PHB粉末が、血糖値スパイクを有意に抑制することを確認できた。図10では図9よりも有意に強く血糖値スパイクを抑制していることから考えれば、純度90%以上のPHB粉末がさらに強く抑制することは妥当な推測である。すなわち例えば純度を99%以上の純度に精製すればさらに強い血糖値スパイク抑制剤となることは容易に推察できる。また、図9及び図10に示した結果から、純度60%以上のPHB粉末であれば効果が生じると考えられる。
本実施の形態に係る血糖値スパイク抑制剤としてのPHB粉末により、飲食物にPHB粉末を混合することで、短時間で血糖値スパイクを抑制するとともに、長時間にわたって血糖値スパイクを抑制する効果を維持することができる。PHB粉末は無味無臭なので、ペット用食品に混ぜることも容易であるから、ペットに恒常的に毎日摂取することも可能である。小腸での生理作用と想定される血糖値スパイクの抑制作用は、PHB粉末を用いたことによって得られる、本発明が初めて提示した新規性である。図9及び図10に示すように、純度70%のPHB粉末、及び純度90%PHB粉末が、血糖値スパイクを有意に抑制することを確認できた。図10では図9よりも有意に強く血糖値スパイクを抑制していることから考えれば、純度90%以上のPHB粉末がさらに強く抑制することは妥当な推測である。すなわち例えば純度を99%以上の純度に精製すればさらに強い血糖値スパイク抑制剤となることは容易に推察できる。また、図9及び図10に示した結果から、純度60%以上のPHB粉末であれば効果が生じると考えられる。
また、PHB粉末は微生物を使って大量に生産できるとともに、糖蜜等の安価な原料を利用することができるため低コストで製造することができる。
以上、本発明を実施の形態を用いて説明したが、本発明の技術的範囲は上記実施の形態に記載の範囲には限定されず、その要旨の範囲内で種々の変形及び変更が可能である。例えば、装置の分散・統合の具体的な実施の形態は、以上の実施の形態に限られず、その全部又は一部について、任意の単位で機能的又は物理的に分散・統合して構成することができる。また、複数の実施の形態の任意の組み合わせによって生じる新たな実施の形態も、本発明の実施の形態に含まれる。組み合わせによって生じる新たな実施の形態の効果は、もとの実施の形態の効果を併せ持つ。
Claims (6)
- ポリ(R)-3-ヒドロキシ酪酸の粉末を含む血糖値スパイク抑制剤。
- ポリ(R)-3-ヒドロキシ酪酸の濃度が60%以上100%以下であることを特徴とする、請求項1に記載の血糖値スパイク抑制剤。
- ポリ(R)-3-ヒドロキシ酪酸の濃度が70%以上90%以下であることを特徴とする、請求項1又は2に記載の血糖値スパイク抑制剤。
- 請求項1から3のいずれか一項に記載のポリ(R)-3-ヒドロキシ酪酸の粉末を含む血糖値スパイク抑制剤が混合された食品。
- ポリ(R)-3-ヒドロキシ酪酸の顆粒を内部に蓄積することができる細菌を含む培養液を準備する準備工程と、
前記培養液を固液分離することにより、ポリ(R)-3-ヒドロキシ酪酸の顆粒を含む残渣を回収する回収工程と、
前記残渣を乾燥させることにより、ポリ(R)-3-ヒドロキシ酪酸の粉末を生成する生成工程と、
を有する血糖値スパイク抑制剤の製造方法。 - 前記準備工程と前記回収工程との間に、前記培養液を加圧する工程をさらに有する、
請求項5に記載の糖値スパイク抑制剤の製造方法。
