Abstract
본 발명은 생약재 추출물을 유효성분으로 포함하는 폐경기 증후군의 예방 또는 치료용 조성물에 관한 것으로, 상기 조성물은 폐경에 따른 심장기능 저하 개선 및 우울증 개선 효과가 있고 생체안정성을 가지므로, 여성 폐경기 증후군의 예방 또는 치료용 조성물로 유용하게 사용될 수 있다.
Description
본 발명은 생약재 추출물을 유효성분으로 포함하는 폐경기 증후군의 예방 또는 치료 용도에 관한 것이다.
폐경은 모든 여성에게서 나타나는 필연적 현상이기는 하나, 그로 인한 여성호르몬의 결핍은 장기간에 걸쳐 신체적, 정신적 질환을 유발하는 중요한 인자로 작용하게 된다. 폐경에 따른 급성 증상 중 대표적인 것으로는 열성홍조, 발한 등의 혈관운동계 증상과 우울, 불안, 신경예민 및 불면 등의 정신적, 정서적 증상 등이 있다. 또한, 만성 증상은 폐경이 발행한 후 수년 후에 나타나는 증상으로써, 이상성감증, 위축성질염 및 요도염 등의 비뇨생식기계 증상, 동맥경화성 순환기 계통의 질환, 골다공증 등이 있다.
갱년기 및 폐경 후 증상의 치료에 가장 보편적으로 사용되고 있는 호르몬 대체요법에 대해 2002년 미국의 WHI(Women's Health Initiative)는 이 요법에 의해 유방암, 관상동맥질환 및 뇌졸중 등의 발병 위험성이 증가하는 경향을 보인다고 발표하였다. 이에 따라 국내에서도 여성호르몬 치료에 의해 심각한 부작용이 초래될 수 있으므로 호르몬 요법은 각 개개인의 위험성과 유익성을 고려하여 개별화된 치료와 저용량의 단기 사용을 권고하며, 치료 목적과 위험성이 시간의 경과에 따라 변하므로 주기적인 재평가가 필요하다고 인식하고 있다.
우리나라에서는 전통적으로 침과 한약 등의 치료방법을 통해 갱년기와 폐경 후의 여러 가지 증상들을 치료해왔으며, 최근에는 호르몬 대체요법의 부작용으로 인해 더 많은 여성들이 한의학적인 치료방법으로 폐경 이후의 임상적 문제들을 해결하고자 한다.
한편, 토사자(cuscutae semen)는 갯실새삼의 씨로써, 주로 간과 신장을 보호하며 눈을 밝게 해주고, 양기를 도우며 신장 기능을 튼튼하게 해주는 약재로 알려져 있다. 오수유(Evodia fructus)는 석호 또는 소모오수유 등의 열매로써, 휘발성 정유를 함유하고 있으며, 소화가 허약하고 차서 발병한 복통에 효과가 있고, 위장기능을 조절하며 소화력을 증진해 준다고 알려져 있다. 속단(Dipsaci radix)은 천속단의 뿌리로써, 간과 신장을 보호하며 근골을 튼튼하게 하는 효과가 있다고 알려져 있다.
최근까지 폐경을 연구하기 위한 동물 모델은 수술적인 방법으로 난소를 적출하는 ovariectomized(OVX) rats 또는 mouse가 이용되었으나, 폐경기 여성의 거의 90%는 난소를 가지고 있기 때문에 OVX 모델은 한계가 있다. 최근에 이런 한계를 극복하기 위한 모델로 VCD에 의해 유도된 폐경 동물 모델이 개발되었는데, VCD 유도 폐경 모델은 자궁의 다른 조직에는 영향을 주지 않고, 선택적으로 원시 여포(primordial follicles)와 일차 난포(primary follicles)의 자연적인 소멸을 촉진 시키기 때문에 사람의 폐경 과정과 매우 유사하며, 또한 폐경 후기(post-menopause) 과정만을 보이는 OVX 모델과는 다르게 폐경기 전후(peri-menopause) 및 폐경 후기 과정을 모두 관찰할 수 있다는 장점이 있다.
이에 본 발명자들은 여성 폐경기 증후군의 예방 또는 치료 효과는 높고, 부작용이 적은 새로운 약물을 개발하기 위해 노력한 결과, 토사자, 오수유 및 속단의 혼합 추출물이 VCD 폐경 동물모델에서 폐경에 따른 심장기능 저하 개선 및 우울증 개선 효과가 있음을 확인함으로써 본 발명을 완성하였다.
본 발명의 하나의 목적은 토사자, 오수유 및 속단의 혼합 추출물을 유효성분으로 포함하는 여성 폐경기 증후군의 예방 또는 치료용 조성물을 제공하는 것이다.
본 발명의 일 양상은 토사자, 오수유 및 속단의 혼합 추출물을 유효성분으로 포함하는 여성 폐경기 증후군의 예방 또는 치료용 조성물을 제공한다.
본 발명의 일 구체예에 따르면, 상기 혼합 추출물은 토사자, 오수유 및 속단의 혼합분말을 물, 탄소수 1 내지 4의 알코올 및 이들의 혼합 용매로 이루어진 군에서 선택된 어느 하나 이상의 용매로 추출한 것일 수 있다.
본 발명의 일 구체예에 따르면, 상기 토사자, 오수유 및 속단은 각각 15 내지 60중량% : 5 내지 50중량% : 15 내지 60중량%의 비율로 포함되는 것일 수 있으며, 바람직하게는, 20 내지 30중량% : 10 내지 15중량% : 20 내지 30중량% 의 비율로 포함될 수 있다.
본 발명의 일 구체예에 따르면, 상기 폐경기 증후군은 에스트로겐 분비 감소에 의해 유발되는 것일 수 있다.
본 발명의 일 구체예에 따르면, 상기 폐경기 증후군은 동맥경화성 심혈관병, 빈맥, 안면홍조, 가슴 두근거림, 발한, 골다공증, 우울증, 요실금, 배뇨곤란, 오줌 소태, 재발성 비뇨기계 염증 및 탈모증으로 이루어진 군에서 선택되는 것일 수 있다.
본 발명의 일 구체예에 따르면, 상기 조성물은 약학적 조성물일 수 있다.

본 발명에 따른 조성물은 하기와 같이 수득될 수 있다. 토사자, 오수유 및 속단을 물로 세척하여 이물질을 제거한 후 그늘에서 건조하고 분쇄한다. 토사자, 오수유 및 속단은 재배한 것 또는 시판되는 것 등 제한 없이 사용할 수 있다. 분쇄된 토사자, 오수유 및 속단의 분말에 적당한 양의 용매를 첨가하여 완전히 침지되도록 한다. 토사자, 오수유 및 속단의 혼합 분말은 통상의 추출용매를 이용하여 추출할 수 있으며, 바람직하게는, (a) 탄소수 1-4의 무수 또는 함수 저급 알코올 (예: 메탄올, 에탄올, 프로판올, 부탄올, 노말-프로판올, 이소-프로판올 및 노말-부탄올 등), (b) 상기 저급 알코올과 물과의 혼합용매, (c) 아세톤, (d) 에틸 아세테이트, (e) 클로로포름, (f) 1,3-부틸렌글리콜, (g) 헥산, (h) 디에틸에테르, (i) 부틸아세테이트 또는 (j) 물을 이용하여 추출할 수 있으며, 가장 바람직하게는 물 또는 에탄올로 추출할 수 있다. 추출시 실온에서 함침하거나 가온할 수 있다. 상기 추출액을 여과하고, 농축하여 최종 추출물을 수득할 수 있다.
본 발명의 일 구체예에 따르면, 상기 혼합 추출물은 토사자, 오수유 및 속단의 혼합분말을 물, 탄소수 1 내지 4의 알코올 및 이들의 혼합 용매로 이루어진 군에서 선택된 어느 하나 이상의 용매로 추출한 것일 수 있다.
본 발명의 일 구체예에 따르면, 상기 토사자, 오수유 및 속단은 각각 15 내지 60중량% : 5 내지 50중량% : 15 내지 60중량%의 비율로 포함되는 것일 수 있으며, 바람직하게는, 20 내지 30중량% : 10 내지 15중량% : 20 내지 30중량% 의 비율로 포함될 수 있다.
본 발명의 일 구체예에 따르면, 상기 폐경기 증후군은 에스트로겐 분비 감소에 의해 유발되는 것일 수 있다.
본 발명의 일 구체예에 따르면, 상기 폐경기 증후군은 동맥경화성 심혈관병, 빈맥, 안면홍조, 가슴 두근거림, 발한, 골다공증, 우울증, 요실금, 배뇨곤란, 오줌 소태, 재발성 비뇨기계 염증 및 탈모증으로 이루어진 군에서 선택되는 것일 수 있다.
본 발명의 일 구체예에 따르면, 상기 조성물은 약학적 조성물일 수 있다.
본 발명의 조성물이 약학적 조성물로 제조되는 경우, 본 발명의 약학적 조성물은 약학적으로 허용되는 담체를 포함한다. 본 발명의 약학적 조성물에 포함되는 약학적으로 허용되는 담체는 제제시에 통상적으로 이용되는 것으로써, 락토스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 전분, 아카시아 고무, 인산 칼슘, 알기네이트, 젤라틴, 규산 칼슘, 미세결정성 셀룰로스, 폴리비닐피롤리돈, 셀룰로스, 물, 시럽, 메틸 셀룰로스, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 활석, 스테아르산 마그네슘 및 미네랄 오일 등을 포함하나, 이에 한정되는 것은 아니다. 본 발명의 약학적 조성물은 상기 성분들 이외에 윤활제, 습윤제, 감미제, 향미제, 유화제, 현탁제, 보존제 등을 추가로 포함할 수 있다. 적합한 약학적으로 허용되는 담체 및 제제는 Remington's
Pharmaceutical Sciences (22th ed., 2013)에 상세히 기재되어 있다.
본 발명의 약학적 조성물은 경구 또는 비경구 투여할 수 있으며, 바람직하게는 경구 투여 방식으로 적용된다.
본 발명의 약학적 조성물의 적합한 투여량은 제제화 방법, 투여 방식, 환자의 연령, 체중, 성, 병적 상태, 음식, 투여 시간, 투여 경로, 배설 속도 및 반응 감응성과 같은 요인들에 의해 다양하게 처방될 수 있다. 본 발명의 약학적 조성물의 바람직한 투여량은 성인 기준으로 0.001-100㎎/kg 범위 내이다.
본 발명의 약학적 조성물은 당해 발명이 속하는 기술분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있는 방법에 따라, 약학적으로 허용되는 담체 및/또는 부형제를 이용하여 제제화함으로써 단위 용량 형태로 제조되거나 또는 다용량 용기 내에 내입시켜 제조될 수 있다. 이때 제형은 오일 또는 수성 매질중의 용액, 현탁액, 시럽제 또는 유화액 형태이거나 엑스제, 산제, 분말제, 과립제, 정제 또는 캅셀제 형태일 수도 있으며, 분산제 또는 안정화제를 추가적으로 포함할 수 있다.
본 발명의 일 구체예에 따르면, 상기 조성물은 식품 조성물일 수 있다.
본 발명의 조성물이 식품 조성물로 제조되는 경우, 유효성분으로서 토사자, 오수유 및 속단의 혼합 추출물뿐만 아니라, 식품 제조 시에 통상적으로 첨가되는 성분을 포함할 수 있으며, 예를 들어, 단백질, 탄수화물, 지방, 영양소, 조미제 및 향미제를 포함할 수 있다. 탄수화물은 모노사카라이드(예를 들어, 포도당, 과당 등), 디사카라이드(예를 들어, 말토스, 슈크로스, 올리고당 등) 및/또는 폴리사카라이드(예를 들어, 덱스트린, 사이클로덱스트린 등)과 같은 통상적인 당 및/또는 자일리톨, 소르비톨, 에리트리톨 등의 당알콜을 사용할 수 있다. 향미제는 천연 향미제(타우마틴, 스테비아 추출물(예를 들어, 레바우디오시드 A, 글리시르히진 등)) 및/또는 합성 향미제(사카린, 아스파르탐 등)를 사용할 수 있다.
또한, 본 발명의 식품 조성물은 여러 가지 영양제, 비타민, 광물(전해질), 합성 풍미제 및 천연 풍미제 등의 풍미제, 착색제 및 중진제(치즈, 초콜릿 등), 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알콜, 탄산음료에 사용되는 탄산화제 등을 함유할 수 있다. 이러한 성분은 독립적으로 또는 조합하여 사용할 수 있으며, 이러한 첨가제의 비율은 본 발명의 식품 조성물 100 중량부 당 0 내지 약 20 중량부의 범위에서 선택될 수 있으나 이에 한정되는 것은 아니다.
본 발명의 토사자, 오수유 및 속단의 혼합 추출물은 당업계에서 일반적으로 사용하는 제형의 제조방법에 의하여 다양한 폐경기 증후군 예방 또는 개선용 식품 조성물로 제조될 수 있다. 예를 들어, 상기의 추출물은 음료나 환, 분말 등과 같은 통상의 건강기능식품 제형으로 제조될 수 있다. 본 발명의 식품 조성물이 드링크제로 제조되는 경우에는 본 발명의 토사자, 오수유 및 속단의 혼합 추출물 이외에 구연산, 액상과당, 설탕, 포도당, 초산, 사과산, 과즙, 두충 추출액, 대추 추출액, 감초 추출액 등을 추가로 포함할 수 있다.
본 발명의 다른 양상은 본 발명의 토사자, 오수유 및 속단의 혼합 추출물을 폐경기 증후군 환자에 투여하는 단계를 포함하는 폐경기 증후군의 치료방법을 제공한다.
이하 하나 이상의 구체예를 실시예를 통하여 보다 상세하게 설명한다. 그러나, 이들 실시예는 하나 이상의 구체예를 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
실시예
1: 폐경기 증후군에 대한
생약재
혼합 추출물의 개선 효과 분석 방법
1-1: 동결건조법 또는 분무건조법에 따른 생약재 혼합 추출물의 제조
정선한 토사자, 오수유 및 속단을 하기 표 1에 기재되어 있는 중량으로 추출포에 넣어 혼합한 것을 물 16L와 함께 80℃에서 4시간 동안 가열하여 1차 추출하고, 이를 다시 80℃에서 4시간 동안 가열하여 2차 추출하였다. 120mesh 필터로 1차 여과한 추출물을 5㎛ 필터로 2차 여과한 후 버퍼탱크로 옮겨 15Bx까지 농축하였다. 이렇게 농축한 추출물은 동결건조법 또는 분무건조법에 따라 최종 추출물로 제조된다.
동결건조법에 의할 경우, 15Bx로 농축한 상기 추출물을 85℃에서 1시간 동안 살균한 후 냉동하여 5일 동안 동결건조하였으며, 최종적으로 총 2kg의 원료에서 198.8g의 추출물을 얻었다(수율: 19.8%).
분무건조법에 의할 경우, 15Bx로 농축한 상기 추출물을 12.98Bx로 희석하여 85℃에서 1시간 동안 살균한 후, 주입구 온도는 190℃, 배출구 온도는 90℃, 타공은 10sec/1회 및 유속은 1.0L/hr의 조건으로 분무건조하였으며, 최종적으로 총 2kg의 원료에서 170.2g의 추출물을 얻었다(수율: 17%).

1-2: 에탄올 추출법에 따른
생약재
혼합 추출물의 제조
정선한 토사자, 오수유 및 속단을 상기 표 1에 기재되어 있는 중량으로 추출포에 넣어 혼합한 것을 30% 에탄올 16L와 함께 80℃에서 4시간 동안 가열하여 1차 추출하고, 이를 다시 80℃에서 4시간 동안 가열하여 2차 추출하였다. 120mesh 필터로 1차 여과한 추출물을 5㎛ 필터로 2차 여과한 후 버퍼탱크로 옮겨 15Bx까지 농축하였다. 이렇게 농축한 추출물은 동결건조법 또는 분무건조법에 따라 최종 추출물로 제조된다.
이후의 동결건조법 또는 분무건조법은 상기 실시예 1-1의 과정과 동일하며, 동결건조법에 의할 경우, 총 2kg의 원료에서 185.2g의 추출물을 얻었으며(수율: 18.5%), 분무건조법에 의할 경우, 총 2kg의 원료에서 168.2g의 추출물을 얻었다(수율: 16.8%).
1-3: VCD 폐경 동물모델의 설계
4주령 C57BL/6 암컷 쥐를 사용하였으며, 1주일간 사육환경에 적응시킨 후 실험에 사용하였다. 실험군은 다음과 같이 5개의 그룹으로 나누고, 각 그룹별로 하기와 같이 처리하였다.
- 그룹 1(Sham group) : sesame oil을 17일간 복강 투여한 후 일반 식이 사료를 15주간 먹인 군,
- 그룹 2(VCD group) : 쥐의 복강에 sesame oil(2.5mg/kg) 및 VCD(160mg/kg)를 17일간 투여하여 VCD 폐경을 유도한 후 일반 식이 사료를 15주간 먹인 군,
- 그룹 3(저농도 group) : 그룹 2와 동일한 방법으로 VCD 폐경을 유도한 후 상기 실시예 1-1의 추출물(100mg/kg)을 함유하는 사료를 15주간 먹인 군,
- 그룹 4(고농도 group) : 그룹 2와 동일한 방법으로 VCD 폐경을 유도한 후 상기 실시예 1-1의 추출물(300mg/kg)을 함유하는 사료를 15주간 먹인 군,
- 그룹 5(Estradiol group) : 그룹 2와 동일한 방법으로 VCD 폐경을 유도한 후 estradiol(2mg/kg)을 함유하는 사료를 15주간 먹인 군.
1-4: 몸무게, 사료 섭취량 측정 및 식이 효율(food efficiency ratio,
FER
) 분석
몸무게 및 사료 섭취량은 1주일마다 측정하며, 실험 종료 후 실험기간 동안의 식이 효율을 분석하였다. 식이 효율은 (체중증가량/총사료섭취랑)×100으로 계산하여 분석하였다.
1-5: 심장, 난소 및 자궁의 무게 및 지수 측정
실험 종료 후 각 실험쥐의 심장, 난소, 및 자궁의 무게를 측정하였고, 각 장기의 지수는 실험동물의 몸무게에 대한 각 장기 무게의 백분율로 산출하였다.
1-6: 행동 실험(Open field test)
실험쥐를 40cm×40cm×40cm의 하얀색 상자에서 10분간 자유롭게 움직이게 하였다. 상자의 바닥은 10cm 간격으로 선을 그었으며, 정중앙에 가로, 세로 10cm 박스를 그렸다. 실험쥐의 움직임은 캠코더로 측정하고, 행동 분석 프로그램인 ANY-maze를 이용하여 중심부에서의 시간(time in the center) 및 총 이동 거리(travelled distance)를 분석하였다.
1-7: 혈액 분석
실험쥐를 avertin tribromoethanol로 마취 후, 복대정맥으로부터 혈액을 채취하여, 4℃에서 2000g로 15분간 원심분리하고, 상층액을 조심스럽게 분리한 후 -80℃에 보관하였다. 이후, 시판되는 kit를 사용하여 -80℃에 보관 중인 혈청에서 혈중 총 콜레스테롤(total cholesterol), 고밀도지단백질(HDL), 중성지방(TG), GOT(glutamic oxalacetic transaminase)/GPT(glutamic pyruvate transaminase), 및 글루코오스(glucose) 농도를 측정하였다.
1-8: 통계 처리
모든 분석 자료는 결과는 SPSS 23(SPSS Inc., Chicago, IL,USA)를 이용하였으며, 통계적 유의성 검정은 p<0.05 수준일 때를 기준으로 하고, 평균±표준오차(mean±SE)로 나타내었다. 일원배치 분산분석(one-way ANOVA)을 한 후, 각 시료간의 통계적 유의성에 대하여 Tukey'smultiplecomparison test 또는 Dunnett`stest를 이용하여 사후검증하였다.
실시예 2: 체중, 식이섭취량 및 식이효율 확인
상기 실시예 1-1의 추출물을 포함한 사료를 식이한 군과 포함하지 않은 사료를 식이한 군에서 체중, 식이섭취량 및 식이효율에 차이가 있는지를 확인하였다.
구체적으로, 상기 실시예 1-4의 방법을 통하여 5개의 실험군에서 각각 체중 변화여부, 식이섭취량 및 식이효율을 분석하였다.
그 결과, 체중 증가량과 식이섭취량은 Sham군이 가장 높고, 그 외 군은 비슷한 경향을 보였으며(도 2 및 도 3), 식이효율은 고농도 군에서 가장 낮게 나왔으나, 유의성은 없었다(도 4). 상기 결과를 통하여 본 발명의 혼합 추출물이 폐경 후 체중증가를 유발하지 않음을 확인하였다.
실시예 3: 심장, 난소, 및 자궁 무게 및 지수의 정상화 확인
상기 실시예 1-1의 추출물을 포함한 사료를 식이한 군에서 장기의 무게 및 지수가 정상적으로 회복되는지를 확인하였다.
구체적으로, 폐경 후에 나타나는 자궁의 위축 및 심장 비대증 증상(heart hypertrophy)이 유발되는지를 확인하기 위하여 상기 실시예 1-5에 따른 분석을 수행하였다.
그 결과, 심장 지수는 VCD 군이 가장 높게 나와서, 폐경 후 심장 비대증과 같은 질병이 유발되는 것을 알 수 있으며, 저농도 군, 고농도 군 및 Estradiol 군의 경우 Sham 군 수준으로 회복되는 확인하였다(도 5).
자궁 지수는 VCD 군이 가장 낮았으나, 저농도 군 및 고농도 군의 경우 Sham 군 수준으로 유의성 있게 회복되었으며(p<0.01)(도 6), 난소 지수 역시 VCD 군에서 가장 낮은 수치를 보였으나(p<0.01), 저농도 군 및 Estradiol 군에서 유의성 있게 회복되는 것을 확인하였다(p<0.01)(도 7).
실시예 4: 갱년기 우울 증상의 개선 효과 확인
상기 실시예 1-1의 추출물을 포함한 사료를 식이한 군에서 갱년기 우울 증상이 개선되는지를 확인하였다.
구체적으로, 폐경 후 우울증 유발에 따른 불안감과 활동성 저하에 대한 본 발명의 혼합 추출물의 효능을 확인하기 위하여 상기 실시예 1-6 및 상기 실시예 1-7에 따른 분석을 수행하였다.
그 결과, VCD 군의 경우 시험상자의 중심부에서 보내는 시간이 Sham 군에 비해 현저히 감소한 것을 볼 수 있으며(p<0.05), 저농도 군 및 고농도 군의 경우 Sham 군 수준으로 유의성 있게 회복되었다(p<0.05)(도 8). 전체적인 이동 거리도 VCD 군이 Sham 군에 비해 확연히 감소하였으나(p<0.001), 저농도 군 및 고농도 군에서 유의성 있게 증가하였다(각각 p<0.05, p<0.01)(도 9).
또한, 혈중 코티솔(cortisol) 수치를 측정한 결과 VCD 군에서 급격히 증가하였으나(p<0.001), 본 발명의 혼합 추출물을 섭취한 군 모두에서 혈중 코티솔 수치가 감소한 것을 확인하였으며, 특히 저농도 군에서 유의하게 감소하였다(p<0.05)(도 10).
실시예 5: 심혈관계 인자 개선 효과 확인
상기 실시예 1-1의 추출물을 포함한 사료를 식이한 군에서 심혈관계 인자가 개선되는지를 확인하였다.
구체적으로, 폐경 후의 총콜레스테롤, HDL 콜레스테롤, 중성지방(TG), 및 혈중 글루코오스의 농도를 확인하기 위하여 상기 실시예 1-7에 따른 분석을 수행하였다.
그 결과, VCD 군에서는 총콜레스테롤 농도가 Sham 군에 비해 현저히 증가하였으나(p<0.001), 고농도 군에서 Sham 군 수준으로 낮아졌으며(p<0.01)(도 11), 반대로 HDL 콜레스테롤의 경우 VCD 군에서 유의성있게 감소하였으나(p<0.05), 약재 섭취군에서는 회복되는 경향을 보였다(도 12).
또한, 중성지방은 VCD 군에서 현저히 증가하였으나(p<0.001), 저농도 군, 고농도 군, 및 Estradioil 군에서 유의성있게 감소하였다(각각 p<0.05, p<0.01 및 p<0.001)(도 13). 글루코오스는 각군이 유의성있는 차이를 보이지는 않았지만, 약재 섭취군이 VCD 군에 비해서는 낮게 측정되었다(도 14).
실시예 6: 간독성 검사
상기 실시예 1-1의 추출물을 포함한 사료를 식이한 군에서 간독성이 나타나는지를 확인하였다.
구체적으로, 상기 실시예 1-7에 따라 혈중 GOT/GPT의 수치를 측정하여 본 발명의 혼합 추출물의 섭취에 의한 간독성이 나타나는지를 분석하였다.
그 결과, 모든 군의 GOT/GPT 수치는 유사한 경향을 보였다는 점에서(도 15 및 도 16), 본 발명의 혼합 추출물이 실험동물에 대해 간독성이 없음을 확인하였다.
이제까지 본 발명에 대하여 그 바람직한 실시예들을 중심으로 살펴보았다. 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자는 본 발명이 본 발명의 본질적인 특성에서 벗어나지 않는 범위에서 변형된 형태로 구현될 수 있음을 이해할 수 있을 것이다. 그러므로 개시된 실시예들은 한정적인 관점이 아니라 설명적인 관점에서 고려되어야 한다. 본 발명의 범위는 전술한 설명이 아니라 특허청구범위에 나타나 있으며, 그와 동등한 범위 내에 있는 모든 차이점은 본 발명에 포함된 것으로 해석되어야 할 것이다.
Claims (7)
- 토사자, 오수유 및 속단의 혼합 추출물을 유효성분으로 포함하는 여성 폐경기 증후군의 예방 또는 치료용 조성물.
- 제 1항에 있어서, 상기 혼합 추출물은 토사자, 오수유 및 속단의 혼합분말을 물, 탄소수 1 내지 4의 알코올 및 이들의 혼합 용매로 이루어진 군에서 선택된 어느 하나 이상의 용매로 추출한 것인 여성 폐경기 증후군의 예방 또는 치료용 조성물.
- 제 1항에 있어서, 상기 토사자, 오수유 및 속단은 각각 15 내지 60중량% : 5 내지 50중량% : 15 내지 60중량%의 비율로 혼합된 것인 여성 폐경기 증후군의 예방 또는 치료용 조성물.
- 제1항에 있어서, 상기 폐경기 증후군은 에스트로겐 분비 감소에 의해 유발되는 것인 여성 폐경기 증후군의 예방 또는 치료용 조성물.
- 제1항에 있어서, 상기 폐경기 증후군은 동맥경화성 심혈관병, 빈맥, 안면홍조, 가슴 두근거림, 발한, 골다공증, 우울증, 요실금, 배뇨곤란, 오줌 소태, 재발성 비뇨기계 염증 및 탈모증으로 이루어진 군에서 선택되는 것인 여성 폐경기 증후군의 예방 또는 치료용 조성물.
- 제 1항에 있어서, 상기 조성물은 약학적 조성물인 것인 여성 폐경기 증후군의 예방 또는 치료용 조성물.
- 제 1항에 있어서, 상기 조성물은 식품 조성물인 것인 여성 폐경기 증후군의 예방 또는 치료용 조성물.
