加齢や疲労により低下した運動調節機能を向上させる運動調節機能向上剤、当該運動調節機能向上剤の評価又は選択方法の提供。 GIP機能阻害剤を有効成分とする運動調節機能向上剤。
Description
本発明は、運動調節機能向上剤に関する。
ヒトは、前庭器官や四肢の筋肉などが身体の状態(体勢、姿勢)を感知し、その刺激が神経を介して脊髄や小脳、大脳皮質などに伝達されることで、身体の平衡性を保つために必要な動作調節を行う能力を有している(非特許文献1及び2)。このような体の動きや重力の方向の変化を感じる感覚は、一般に平衡感覚と称され、運動を調節する重要な能力の1つとされている。
運動調節機能は、加齢や疲労により能力低下が起こり、その原因としては、三半規管や前庭の感度の低下、視覚など感覚器における識別能力の低下、筋肉・骨格など運動を制御する部位の能力低下、神経や脊髄、脳の機能低下もしくは障害などが考えられている。
このような運動調節機能低下は、転倒事故や転落事故をはじめとする多くの事故の要因ともなっており、捻挫・骨折などの障害に至る場合もある。また、立位を維持したり、そのまま動作を行ったりする際のふらつきや、座位からの立ち上がり時のふらつきなどが増加し、日常生活を送る上で支障をきたす場合もある。
従来、この運動調節機能、特に平衡感覚を維持又は改善する方法として、主に薬物療法が適用される。運動調節機能を改善する薬剤としては、精神身体神経安定剤(精神安定剤、自律神経安定剤、精神神経用剤、催眠鎮静剤など)、循環改善剤(血管拡張剤、 血流改善剤、血管収縮剤、血管強固剤、循環器官用剤、動脈硬化用剤など)、代謝改善剤(脳、組織、細胞賦活剤、代謝賦活剤など)及びビタミン剤などが用いられる(非特許文献3)が、運動調節機能を根本的に治療するものではなく、十分な効果を上げているとは未だ言い難い。また、その他にはリハビリテーションの観点から、運動療法も用いられる(非特許文献4)が、モチベーションの維持の困難さや怪我の恐れもあり、現実的にはなかなか難しく、より効果的な方法が望まれている。
一方、GIP(gastric inhibitory polypeptide又はglucose-dependent insulinotropic polypeptide)は、グルカゴン・セクレチンファミリーに属する消化管ホルモンの1つである。GIPはGLP-1(グルカゴン様ペプチド1)と共にインクレチンと称され、脂質や糖質の摂食時に小腸に存在するK細胞より分泌される。
GIPは、膵β細胞からのインスリン分泌を促進し、インスリン存在下でのグルコースの脂肪細胞への取り込みを亢進することが知られている。そのため、GIPの作用が肥満の一要因になっているとも考えられ、実際、GIPの機能を阻害すると、肥満が抑制されるとの報告がある(非特許文献5)。
GIPは、膵β細胞からのインスリン分泌を促進し、インスリン存在下でのグルコースの脂肪細胞への取り込みを亢進することが知られている。そのため、GIPの作用が肥満の一要因になっているとも考えられ、実際、GIPの機能を阻害すると、肥満が抑制されるとの報告がある(非特許文献5)。
さらに、GIPはインスリン抵抗性の一因となることが報告されている(非特許文献1)。インスリン抵抗性を発症すると、インスリンによる糖の吸収作用が低下し、その結果、高インスリン血症を引き起こす。高インスリン血症は、肥満をはじめとする様々な生活習慣病の発症につながる根本的な原因であるとも言われており、インスリン抵抗性の予防・改善は生活習慣病のリスク軽減の面からも重要である。
しかしながら、GIPと運動調節機能が関係するという報告はなく、GIP機能を抑制することにより、運動調節機能が向上できることは全く知られていない。

本発明は、加齢や疲労により低下した運動調節機能を向上させる運動調節機能向上剤、及び当該運動調節機能向上剤の評価又は選択方法を提供することに関する。
本発明者らは、GIPと運動調節機能との関係について検討したところ、GIPを投与した場合に運動調節機能が有意に低下すること、そしてGIP機能を抑制することによって、運動調節機能を向上できること、またGIP機能阻害活性を評価することにより運動調節機能向上剤をスクリーニングできることを見出した。
本発明の運動調節機能向上剤は、運動調節機能、例えば加齢や疲労により低下した運動調節機能を向上させる効果を発揮する。したがって、三半規管や前庭の感度の低下、視覚など感覚器における識別能力の低下、筋肉・骨格など運動を制御する部位の能力低下、神経や脊髄、脳の機能低下もしくは障害などに有用である。
本発明において、GIP(gastric inhibitory polypeptide又はglucose-dependent insulinotropic polypeptide)は、42個のアミノ酸からなるポリペプチドである。GIP(1-42)は生理活性を有するが(活性型GIP)、生体内に存在するdipeptidyl peptidase-4(DPP-4)によってN末端の2アミノ酸が切断されて不活性型のGIP(3-42)となる。
本発明において、「GIP機能阻害剤」とは、GIPが有する消化管ホルモンとしての機能を阻害又は抑制する物質、すなわちGIP遺伝子又はGIP受容体遺伝子レベルで、あるいはGIP自体又はGIP受容体レベルで機能を阻害する物質を意味し、具体的には抗GIP抗体、GIP受容体拮抗剤、GIP分泌又は上昇抑制剤が挙げられる。
本発明において、「抗GIP抗体」とは、少なくとも活性型GIPの機能を阻害する抗体であればよく、ポリクローナル抗体、モノクローナル抗体の何れであっても良いが、国際公開第2016/104439号、特開2013-138638号公報に記載の、非活性型GIPとは実質的に結合しない抗体(「抗活性型GIP抗体」と称する)であるのが好ましい。また、活性型GIPとの結合定数(Ka)が107M-1以上であるのが好ましく、より好ましくは108M-1以上であり、より好ましくは109M-1以上である。
抗活性型GIP抗体としては、被験抗体の非活性型GIPに対する結合量が、活性型GIPに対する結合量を100%とした場合、多くとも10%以下、好ましくは5%以下、更に好ましくは1%以下、更により好ましくは0.1%であるものが挙げられる。活性型又は非活性型GIPとの結合量は、被験抗体と活性型又は非活性型GIPとの結合を、ウエスタンブロッティング、免疫沈降、免疫組織化学染色、及びELISA等の方法を用いて測定することにより判断することができる。
抗活性型GIP抗体としては、被験抗体の非活性型GIPに対する結合量が、活性型GIPに対する結合量を100%とした場合、多くとも10%以下、好ましくは5%以下、更に好ましくは1%以下、更により好ましくは0.1%であるものが挙げられる。活性型又は非活性型GIPとの結合量は、被験抗体と活性型又は非活性型GIPとの結合を、ウエスタンブロッティング、免疫沈降、免疫組織化学染色、及びELISA等の方法を用いて測定することにより判断することができる。
抗活性型GIP抗体としては、例えば、活性型GIP(配列番号5)のN末端から8番目以降のアミノ酸を認識する抗体であり、少なくも8~10番目(SDY)から選ばれる1以上のアミノ酸を認識する抗体が好適に挙げられる。
当該抗活性型GIP抗体としては、更に、H鎖に、下記(1)で表されるアミノ酸配列又はその保存的配列改変からなる領域を含むものが好適である。

本明細書において、「保存的配列改変」とは、抗原決定に関与する相補性決定領域(CDR)以外のアミノ酸改変であって、改変前のアミノ酸配列からなる抗体の反応性に有意な影響を及ぼさない又は変化させないアミノ酸改変を意味する。そのような保存的配列改変には、1~数個、好ましくは1~3個、より好ましくは1個のアミノ酸の置換、付加及び欠失が含まれる。当該保存的配列改変がなされたアミノ酸配列としては、改変前のアミノ酸配列と90%以上、好ましくは95%以上、更に好ましくは99%以上の配列同一性を有するアミノ酸配列が挙げられる。当該改変は、部位特異的変異誘発及びPCR媒介変異誘発のような、当技術分野において公知の標準的な技術によって本発明の抗体に導入することができる。保存的アミノ酸置換としては、アミノ酸残基が類似の側鎖を有するアミノ酸残基(アミノ酸残基のファミリー)に置換されることが挙げられる。斯かるアミノ酸残基のファミリーは、当技術分野において定義されており、塩基性側鎖(例えば、リジン、アルギニン、ヒスチジン)、酸性側鎖(例えば、アスパラギン酸、グルタミン酸)、非荷電極性側鎖(例えば、グリシン、アスパラギン、グルタミン、セリン、トレオニン、チロシン、システイン、トリプトファン)、非極性側鎖(例えば、アラニン、バリン、ロイシン、イソロイシン、プロリン、フェニルアラニン、メチオニン)、β-分岐側鎖(例えば、トレオニン、バリン、イソロイシン)、及び芳香族側鎖(例えば、チロシン、フェニルアラニン、トリプトファン、ヒスチジン)を有するアミノ酸が含まれる。
尚、アミノ酸配列間の同一性とは、2つのアミノ酸配列をアラインメントしたときに両方の配列において同一のアミノ酸残基が存在する位置の数の全長アミノ酸残基数に対する割合(%)をいう。具体的には、例えばリップマン-パーソン法(Lipman-Pearson法;Science, 227, 1435, (1985))によって計算され、遺伝情報処理ソフトウェアGenetyx-Win(Ver.5.1.1;ソフトウェア開発)のホモロジー解析(Search homology)プログラムを用いて、Unit size to compare(ktup)を2として解析を行なうことにより算出できる。
上記(1)で表されるアミノ酸配列は、H鎖可変領域を示す配列番号2で示されるアミノ酸配列の50~62番目の13アミノ酸残基からなる領域をコードするものである。
したがって、上記抗活性型GIP抗体としては、H鎖可変領域として配列番号2で示されるアミノ酸配列又はその保存的配列改変からなる領域を含むものであるのがより好ましい。また更に、H鎖可変領域として配列番号2で示されるアミノ酸配列又はその保存的配列改変からなる領域を含み、且つL鎖可変領域として配列番号4で示されるアミノ酸配列又はその保存的配列改変からなる領域を含むものがより好ましい。
H鎖可変領域として配列番号2で示されるアミノ酸配列からなる領域を含み、且つL鎖可変領域として配列番号4で示されるアミノ酸配列からなる領域を含む抗活性型GIP抗体としては、後記製造例1に示すハイブリドーマ9B9H5-B9株から産生されるモノクローナル抗体が挙げられる。
したがって、上記抗活性型GIP抗体としては、H鎖可変領域として配列番号2で示されるアミノ酸配列又はその保存的配列改変からなる領域を含むものであるのがより好ましい。また更に、H鎖可変領域として配列番号2で示されるアミノ酸配列又はその保存的配列改変からなる領域を含み、且つL鎖可変領域として配列番号4で示されるアミノ酸配列又はその保存的配列改変からなる領域を含むものがより好ましい。
H鎖可変領域として配列番号2で示されるアミノ酸配列からなる領域を含み、且つL鎖可変領域として配列番号4で示されるアミノ酸配列からなる領域を含む抗活性型GIP抗体としては、後記製造例1に示すハイブリドーマ9B9H5-B9株から産生されるモノクローナル抗体が挙げられる。
本発明の抗GIP抗体は、上記反応性を有する限り、当該抗体のフラグメント、例えば、F(ab')2、F(ab')、一本鎖Fv(scFv)、VH及びVL中のシステイン残基に置換されたアミノ酸残基がジスルフィド結合を介して結合しているジスルフィド結合Fv(dsFv)若しくはこれらの重合体、又はscFvが二量体化した二量体化V領域(Diabody)であっても良い。更に、上記反応性を有する限り、抗活性型GIP抗体の一部を含むペプチド、すなわち抗体を構成するアミノ酸配列の一部を備えるペプチドであって上記反応性を有するものも当該抗体のフラグメントに含まれる。
また、本発明の抗GIP抗体のイムノグロブリンクラスは特に限定されるものではなく、IgG、IgM、IgA、IgE、IgD、IgYのいずれのイムノグロブリンクラスであってもよく、好ましくはIgGである。また、本発明の抗体はいずれのアイソタイプの抗体をも包含するものである。
また本発明の抗GIP抗体は、非ヒト動物の抗体、ヒト型キメラ抗体、ヒト化抗体及びヒト抗体の何れであっても良い。非ヒト動物の抗体としては、例えば、マウス、ラット、ハムスター、モルモット等の抗体を挙げることができ、好ましくはマウスの抗体である。
ここで、「ヒト型キメラ抗体」とは、非ヒト動物由来であってGIPと特異的に結合する抗体の定常領域をヒトの抗体と同じ定常領域を有するように遺伝子工学的に改変した抗体のことであり、好ましくは、ヒト・マウス・キメラ抗体である。また、「ヒト化抗体」とは、非ヒト動物由来であってGIPと特異的に結合する抗体のH鎖とL鎖の相補性決定領域(CDR)以外の一次構造をヒトの抗体に対応する一次構造に遺伝子工学的に改変した抗体のことである。また、「ヒト抗体」とは、完全にヒト由来の抗体遺伝子の発現産物であるヒト抗体を意味する。
ここで、「ヒト型キメラ抗体」とは、非ヒト動物由来であってGIPと特異的に結合する抗体の定常領域をヒトの抗体と同じ定常領域を有するように遺伝子工学的に改変した抗体のことであり、好ましくは、ヒト・マウス・キメラ抗体である。また、「ヒト化抗体」とは、非ヒト動物由来であってGIPと特異的に結合する抗体のH鎖とL鎖の相補性決定領域(CDR)以外の一次構造をヒトの抗体に対応する一次構造に遺伝子工学的に改変した抗体のことである。また、「ヒト抗体」とは、完全にヒト由来の抗体遺伝子の発現産物であるヒト抗体を意味する。
抗GIP抗体は、市販のポリクローナル抗体(Bioss社)の他、公知の手段を用いて作製されたモノクローナル抗体を使用することができる。哺乳動物由来のモノクローナル抗体としては、ハイブリドーマから産生されるもの、及び抗体遺伝子又は抗体フラグメント遺伝子を設計し周知の遺伝子工学的手法を用いて生産されるものが包含される。
例えば、上述した抗活性型GIP抗体は、H鎖可変領域をコードするDNA(例えば、配列番号1で示される塩基配列からなるDNA)と、L鎖可変領域をコードするDNA(例えば、配列番号3で示される塩基配列からなるDNA)を、それぞれ適当なベクターのプロモーター下流に挿入した組換え体ベクターを造成し、それを宿主細胞に導入した形質転換体からH鎖及びL鎖を製造し、これらを可能性のあるペプチドで連結させる、或いはH鎖可変領域をコードするDNA(例えば、配列番号1で示される塩基配列からなるDNA)とL鎖可変領域をコードするDNA(例えば、配列番号3で示される塩基配列からなるDNA)を、公知のリンカーをコードするDNAで繋いで適当なベクターのプロモーター下流に挿入した組換え体ベクターを造成し、それを宿主細胞内で発現させる、等により抗原結合能を持った一本鎖の組換え抗体タンパク質(scFv)を生産することが挙げられる。(MacCfferty, J. et al., Nature, 348, 552-554, 1990、Tim Clackson et al, Nature, 352, 642-628, 1991等参照)。また、更に、可変領域をコードするDNAと定常領域をコードするDNAとを結合させて発現させたものを生産することであってもよい。この場合、定常領域は、可変領域の由来する抗体と同一のものであっても、あるいは異なる抗体に由来するものであってもよい。
上記の如く機能的に同等なポリペプチドを調製するためのアミノ酸変異の導入は、例えば、部位特異的変異誘発法などを用いて行うことができる。
例えば、上述した抗活性型GIP抗体は、H鎖可変領域をコードするDNA(例えば、配列番号1で示される塩基配列からなるDNA)と、L鎖可変領域をコードするDNA(例えば、配列番号3で示される塩基配列からなるDNA)を、それぞれ適当なベクターのプロモーター下流に挿入した組換え体ベクターを造成し、それを宿主細胞に導入した形質転換体からH鎖及びL鎖を製造し、これらを可能性のあるペプチドで連結させる、或いはH鎖可変領域をコードするDNA(例えば、配列番号1で示される塩基配列からなるDNA)とL鎖可変領域をコードするDNA(例えば、配列番号3で示される塩基配列からなるDNA)を、公知のリンカーをコードするDNAで繋いで適当なベクターのプロモーター下流に挿入した組換え体ベクターを造成し、それを宿主細胞内で発現させる、等により抗原結合能を持った一本鎖の組換え抗体タンパク質(scFv)を生産することが挙げられる。(MacCfferty, J. et al., Nature, 348, 552-554, 1990、Tim Clackson et al, Nature, 352, 642-628, 1991等参照)。また、更に、可変領域をコードするDNAと定常領域をコードするDNAとを結合させて発現させたものを生産することであってもよい。この場合、定常領域は、可変領域の由来する抗体と同一のものであっても、あるいは異なる抗体に由来するものであってもよい。
上記の如く機能的に同等なポリペプチドを調製するためのアミノ酸変異の導入は、例えば、部位特異的変異誘発法などを用いて行うことができる。
抗活性型GIP抗体産生ハイブリドーマは、基本的には公知技術を使用し、以下のようにして作製できる。
例えば、活性型GIP又はそのN末端のアミノ酸配列を有するペプチド(配列番号5の1~15番目のアミノ酸配列からなるペプチド)を、必要に応じて、適当なキャリアー蛋白質、例えばキーホールリンペットヘモシアニン(KLH)やウシ血清アルブミンなどと結合することによって、より免疫原性を高め、非ヒト哺乳動物に免疫することにより作製することができる。尚、感作抗原(免疫源)として用いられる活性型GIP又は上記ペプチドは、遺伝子工学的手法又は化学合成により作製することができる。
例えば、活性型GIP又はそのN末端のアミノ酸配列を有するペプチド(配列番号5の1~15番目のアミノ酸配列からなるペプチド)を、必要に応じて、適当なキャリアー蛋白質、例えばキーホールリンペットヘモシアニン(KLH)やウシ血清アルブミンなどと結合することによって、より免疫原性を高め、非ヒト哺乳動物に免疫することにより作製することができる。尚、感作抗原(免疫源)として用いられる活性型GIP又は上記ペプチドは、遺伝子工学的手法又は化学合成により作製することができる。
感作抗原で免疫される哺乳動物としては、特に限定されるものではないが、細胞融合に使用する親細胞である哺乳動物のミエローマ細胞との適合性を考慮して選択するのが好ましく、一般的にはげっ歯類の動物、例えば、マウス、ラット、ハムスター等が使用される。
感作抗原を動物に免疫するには、公知の方法に従って行われる。例えば、感作抗原を哺乳動物の腹腔内又は、皮下に注射することにより行われる。具体的には、感作抗原をPBS(Phosphate-Buffered Saline)や生理食塩水等で適当量に希釈、懸濁したものを所望により通常のアジュバント、例えば、フロイント完全アジュバントを適量混合し、乳化後、動物の皮下、皮内、腹腔などに投与して一時刺激後、必要に応じて同様の操作を繰り返し行う。抗原の投与量は投与経路、動物種に応じて適宣決定されるが、通常の投与量は1回当たり10μg~1mg程度が好ましい。このように免疫し、血清中に所望の抗体レベルが上昇するのを確認した後に、抗体レベルが上昇した哺乳動物から免疫細胞を取り出し、細胞融合を行う。細胞融合を行う際の好ましい免疫細胞としては、特に脾細胞が挙げられる。
前記免疫細胞と融合される他方の親細胞としての哺乳動物のミエローマ細胞は、すでに、公知の種々の細胞株、例えばP3X63、NS-1、MPC-11、SP2/0等が適宜使用される。

前記免疫細胞とミエローマ細胞の細胞融合は公知の方法、たとえば、ケラーらの方法(Kohler et al., Nature, vol, 256, p495-497(1975))等に準じて行うことができる。すなわち、ポリエチレングリコール(平均分子量1000~6000のPEG、30~60%濃度)、センダイウィルス(HVJ)等の細胞融合促進剤の存在下、所望によりジメチルスルホキシド等の補助剤を添加し、RPMI1640培養液、MEM培養液等の栄養培養液中で、免疫細胞とミエローマ細胞を混合することによって、融合細胞(ハイブリドーマ)の形成が行われる。
融合により形成されたハイブリドーマをヒポキサンチン、チミジン及びアミノプテリンを含む培地(HAT培地)等の選択培地で1日~7日間培養し、未融合細胞と分離する。得られたハイブリドーマをその産生する抗体(活性型GIPに結合し、且つ非活性型GIPとは実質的に結合しない抗体)により更に選択する。
選択したハイブリドーマを公知の限界希釈法に従って単一クローン化し、単一クローン性抗体産生ハイブリドーマとして樹立する。
ハイブリドーマが産生する抗体の活性を検出する方法は、公知の方法を使用することができる。例えばELISA法、凝集反応法、ラジオイムノアッセイ法が挙げられる。
選択したハイブリドーマを公知の限界希釈法に従って単一クローン化し、単一クローン性抗体産生ハイブリドーマとして樹立する。
ハイブリドーマが産生する抗体の活性を検出する方法は、公知の方法を使用することができる。例えばELISA法、凝集反応法、ラジオイムノアッセイ法が挙げられる。
得られたハイブリドーマからモノクローナル抗体を取得するには、当該ハイブリドーマを通常の方法に従って培養し、その培養上清として得る方法、あるいはハイブリドーマをこれと適合性がある哺乳動物に投与して増殖させ、その腹水として得る方法などが採用される。
抗体の精製は、塩析法、ゲル濾過法、イオン交換クロマト法又はアフィニティークロマト法等の公知の精製手段を用いて行なうことができる。
本発明において、「GIP受容体拮抗剤」としては、例えば、国際公開第2003/097031号に記載のメチリデンヒドラジド化合物、具体的には、4-ヒドロキシ安息香酸(2-ブロモベンジリデン)ヒドラジド、3-シアノ-4-ヒドロキシ安息香酸[1-(2,3,5,6-テトラメチルベンジル)インドール-4-イル]メチリデンヒドラジド、3-クロロ-4-ヒドロキシ安息香酸(4-メトキシナフタレン-1-イル)メチリデンヒドラジド、3-クロロ-4-ヒドロキシ安息香酸[1-(5-クロロチオフェン-2-イルメチル)-1H-インドール-5-イル]メチリデンヒドラジド等が挙げられる。
本発明において、「GIP分泌又は上昇抑制剤」としては、例えば、BMPP(3-ブロモ-5-メチル-2-フェニルピラゾロ[1,5-a]ピリミジン-7-オール)(国際公開第2001/87341号)、アルギン酸(特開2013-166741号公報)、フォスファチジルエタノールアミン(特開2010-222284号公報)、ポリグルタミン酸(特開2012-144486号公報)、キラヤ(特開2012-171914号公報)、リゾフォスファチジルイノシトール(特開2012-171915号公報)、セルロースナノファイバ(特開2009-126837号公報)、β-キチンナノファイバー(特開2010-241713号公報)、ジアシルグリセロール(特開2006-342084号公報)、ヒドロキシプロピル化澱粉(特開2006-342085号公報)、モノアシルグリセロール(特開2007-290989号公報)、炭素数20以上の超長鎖脂肪酸(例えば、アラキジン酸、ベヘン酸、リグノセリン酸、セロチン酸、モンタン酸、メリシン酸、ラクセル酸、ガドレイン酸、ジホモ-γ-リノレン酸、アラキドン酸、エイコサペンタエン酸、エルカ酸、ドコサペンタエン酸、ドコサヘキサエン酸、ネルボン酸、ヘキサコセン酸、オクタコセン酸等;特開2011-225458号公報)、構成脂肪酸の1質量%以上がドコサヘキサエン酸であり、1質量%以上がエイコサペンタエン酸であるトリアシルグリセロール(特開2013-063937号公報)、長鎖不飽脂肪酸エタノールアミド(例えば、オレイルエタノールアミド、リノレイルエタノールアミド、リノレニルエタノールアミド、homo-γ-リノレニルエタノールアミド、アラキドニルエタノールアミド、7,10,13,16-ドコサテトラエニルエタノールアミド;特開2010-180203号公報)、米糠抽出物(特表2012-515139号公報)、構成脂肪酸の10質量%以上がα-リノレン酸であるトリアシルグリセロール(特開2013-075887号公報)、グリセロール骨格の2位にC14~C18飽和脂肪酸が結合したアシルグリセロール(例えば、ラウリン酸(12:0)、ミリスチン酸(14:0)、パルミチン酸(16:0)、リノール酸(18:2)、オレイン酸(18:1)、ステアリン酸(18:0)、又はアラキドン酸(20:4)が2位に結合した2-アシルモノグリセロール;特開2016-047805号公報)、ツクリタケの圧搾物又は抽出物(PCT/JP2017/043101)、小麦ふすま(特願2016-234973)等が挙げられる。
後記実施例に示すように、抗GIP抗体は、GIPの投与により生じる運動調節機能の低下を抑制し、老齢マウスにおける運動調節機能を向上する作用を有する。
したがって、抗GIP抗体のようなGIP機能阻害剤は、運動調節機能向上剤となり得、またこれらを製造するために使用できる。
また、GIP機能阻害剤は、運動調節機能向上のために使用することができる。ここで、当該使用は、ヒト若しくは非ヒト動物、又はそれらに由来する検体における使用であり得、また治療的使用であっても非治療的使用であってもよい。尚、「非治療的」とは、医療行為を含まない概念、すなわち人間を手術、治療又は診断する方法を含まない概念、より具体的には医師又は医師の指示を受けた者が人間に対して手術、治療又は診断を実施する方法を含まない概念である。
したがって、抗GIP抗体のようなGIP機能阻害剤は、運動調節機能向上剤となり得、またこれらを製造するために使用できる。
また、GIP機能阻害剤は、運動調節機能向上のために使用することができる。ここで、当該使用は、ヒト若しくは非ヒト動物、又はそれらに由来する検体における使用であり得、また治療的使用であっても非治療的使用であってもよい。尚、「非治療的」とは、医療行為を含まない概念、すなわち人間を手術、治療又は診断する方法を含まない概念、より具体的には医師又は医師の指示を受けた者が人間に対して手術、治療又は診断を実施する方法を含まない概念である。
本発明において、「運動調節機能」とは、平衡感覚やバランス能力、敏捷性等に関する動作を調節する機能であること意味し、「運動調節機能の向上」とは、平衡感覚やバランス能力、敏捷性等に関する動作を調節する機能を高めることを意味する。
本発明の運動調節機能向上剤は、運動調節機能の向上効果を発揮する、ヒト若しくは動物用の医薬品となり、また医薬品に配合して使用される素材又は製剤となり得る。
本発明の運動調節機能向上剤を医薬品として用いる場合、当該医薬品は任意の投与形態で投与され得る。投与形態としては、例えば錠剤、カプセル剤、顆粒剤、散剤、シロップ剤等による経口投与又は注射剤、坐剤、吸入薬、経皮吸収剤、外用剤等による非経口投与が挙げられるが、好ましい形態は非経口投与である。
このような種々の剤型の医薬製剤は、本発明のGIP機能阻害剤を単独で、又は他の薬学的に許容される賦形剤、結合剤、増量剤、崩壊剤、界面活性剤、滑沢剤、分散剤、緩衝剤、保存剤、嬌味剤、香料、被膜剤、担体、希釈剤等を適宜組み合わせて調製することができる。
このような種々の剤型の医薬製剤は、本発明のGIP機能阻害剤を単独で、又は他の薬学的に許容される賦形剤、結合剤、増量剤、崩壊剤、界面活性剤、滑沢剤、分散剤、緩衝剤、保存剤、嬌味剤、香料、被膜剤、担体、希釈剤等を適宜組み合わせて調製することができる。
本発明の運動調節機能向上剤におけるGIP機能阻害剤の含有量は、好ましくは0.001質量%以上、より好ましくは0.01質量%以上、且つ好ましくは10質量%以下、より好ましくは5質量%以下、より好ましくは1質量%以下、より好ましく0.1質量%以下であり、また好ましくは0.001~10質量%、好ましくは0.001~5質量%、好ましくは0.001~1質量%、より好ましくは0.01~0.1質量%である。
本発明の運動調節機能向上剤の投与量又は摂取量は、対象者の状態、体重、性別、年齢又はその他の要因に従って変動し得るが、経口投与又は摂取の場合成人1人当たり、GIP機能阻害剤として、1日あたり好ましくは1mg以上、より好ましくは5mg以上であり、且つ好ましくは500mg以下、より好ましくは100mg以下、より好ましくは20mg以下である。
また、本発明の運動調節機能向上剤の投与対象者としては、運動器の障害により要介護になるリスクの高い運動器症候群(ロコモティブシンドローム)のヒトが好ましい。
本発明の運動調節機能向上剤の評価又は選択方法は、(I)被験物質のGIP機能阻害活性を測定する工程と、(II)(I)の結果に基づいて被験物質のGIP機能阻害活性を評価する工程、(III)GIP機能阻害活性を増加又は増強させる被験物質を運動調節機能向上剤として評価又は選択する工程、を含むものである。
ここで、GIP機能阻害活性を測定する方法としては、例えば以下の方法が挙げられる。
1)GIP受容体cDNAを細胞に導入したGIP受容体発現細胞を用い、被験物質存在下で、GIPによりcAMPを産生させる。その後、cAMPを抽出し、イムノアッセイによりcAMPを測定する。
2)GIP受容体cDNAを細胞に導入したGIP受容体発現細胞に、cAMP依存性プロモーターにバクテリア由来lac Z geneを結合させた遺伝子を導入する。この細胞を、被験物質存在下でGIPと反応させる。GIP活性により産生されたcAMPに応じて細胞中に蓄積したβ-galactosidase活性を測定する。
3)GIP受容体cDNAを細胞に導入したGIP受容体発現細胞に、被験物質及び放射ラベルしたGIPを細胞に加え、インキュベーションの後、放射活性を測定する。
4)GIPを産生する腸管由来細胞、又は神経由来細胞等を用いて、被験物質による刺激後、細胞中もしくは培養上清中に産生されるGIPを定量する。
5)被験物質存在下で、脂質や糖質等の栄養素によるGIP分泌量をELISA法により測定する。
ここで、GIP機能阻害活性を測定する方法としては、例えば以下の方法が挙げられる。
1)GIP受容体cDNAを細胞に導入したGIP受容体発現細胞を用い、被験物質存在下で、GIPによりcAMPを産生させる。その後、cAMPを抽出し、イムノアッセイによりcAMPを測定する。
2)GIP受容体cDNAを細胞に導入したGIP受容体発現細胞に、cAMP依存性プロモーターにバクテリア由来lac Z geneを結合させた遺伝子を導入する。この細胞を、被験物質存在下でGIPと反応させる。GIP活性により産生されたcAMPに応じて細胞中に蓄積したβ-galactosidase活性を測定する。
3)GIP受容体cDNAを細胞に導入したGIP受容体発現細胞に、被験物質及び放射ラベルしたGIPを細胞に加え、インキュベーションの後、放射活性を測定する。
4)GIPを産生する腸管由来細胞、又は神経由来細胞等を用いて、被験物質による刺激後、細胞中もしくは培養上清中に産生されるGIPを定量する。
5)被験物質存在下で、脂質や糖質等の栄養素によるGIP分泌量をELISA法により測定する。
被験物質のGIP機能阻害活性の評価は、GIP機能阻害活性を増加又は増強する被験物質を同定することにより行われる。
例えば、異なる濃度の被験物質を添加した場合に測定されるGIP機能阻害活性を比較することによって行うことができる。より具体的な例としては、より高濃度の被験物質添加群とより低濃度の被験物質添加群との間;被験物質添加群とプラセボ添加群との間;又は被験物質添加前後で、GIP機能阻害活性を比較する。被験物質の添加により、又はより高濃度の被験物質の添加によりGIP機能阻害活性が増加又は増強される場合、当該被験物質を、当該GIP機能阻害活性を増加又は増強させる物質として同定することができる。
そして、GIP機能阻害活性を増加又は増強させるものとして同定された被験物質が、運動調節機能向上剤として評価又は選択される。
尚、被験物質としては、運動調節機能向上に使用することを所望する物質であれば、特に制限されず、天然に存在する物質であっても、化学的又は生物学的方法等で人工的に合成した物質であってもよく、また化合物であっても、組成物若しくは混合物であってもよい。

尚、被験物質としては、運動調節機能向上に使用することを所望する物質であれば、特に制限されず、天然に存在する物質であっても、化学的又は生物学的方法等で人工的に合成した物質であってもよく、また化合物であっても、組成物若しくは混合物であってもよい。

製造例1 抗活性型GIP抗体の作成
(1)免疫用ペプチドの合成
活性型GIPのN末端15アミノ酸(GIP(1-15))にポリエチレングリコールを付加(PEG(polyethylene glycol)化)後、キーホールリンペットヘモシアニン(KLH;keyhole limpet hemocyanin)を化学的に結合させて、KLH結合PEG化GIP(1-15)を作製し、免疫抗原とした。活性型GIPのN末端15アミノ酸(GIP(1-15))をPEG化したものを測定用抗原(1)、非活性型GIPのN末端13アミノ酸(GIP(3-15))をPEG化したものを測定用抗原(2)とした。
(1)免疫用ペプチドの合成
活性型GIPのN末端15アミノ酸(GIP(1-15))にポリエチレングリコールを付加(PEG(polyethylene glycol)化)後、キーホールリンペットヘモシアニン(KLH;keyhole limpet hemocyanin)を化学的に結合させて、KLH結合PEG化GIP(1-15)を作製し、免疫抗原とした。活性型GIPのN末端15アミノ酸(GIP(1-15))をPEG化したものを測定用抗原(1)、非活性型GIPのN末端13アミノ酸(GIP(3-15))をPEG化したものを測定用抗原(2)とした。
(2)免疫
BALB/cマウス(オリエンタル酵母工業(株))を用い、背部皮下に免疫した。初回の免疫では、上記で作成した抗原と完全フロイントアジュバンドとを混合したエマルジョンを投与した。初回免疫から2週間毎に抗原と不完全フロイントアジュバンドを混合したエマルジョンを用いて追加免疫を実施した。一度に免疫する抗原量は0.1~0.2mgの範囲で行った。初回免疫実施7週後、マウスより採取した血清を用いて抗体価測定を実施し、抗体価の上昇を確認した。
BALB/cマウス(オリエンタル酵母工業(株))を用い、背部皮下に免疫した。初回の免疫では、上記で作成した抗原と完全フロイントアジュバンドとを混合したエマルジョンを投与した。初回免疫から2週間毎に抗原と不完全フロイントアジュバンドを混合したエマルジョンを用いて追加免疫を実施した。一度に免疫する抗原量は0.1~0.2mgの範囲で行った。初回免疫実施7週後、マウスより採取した血清を用いて抗体価測定を実施し、抗体価の上昇を確認した。
(3)細胞融合
抗体価の上昇したマウスから脾臓を摘出し、脾細胞を得た。得られた脾細胞とマウスミエローマ細胞株P3U1をPEG法により融合した。その後、96ウェルプレート20枚へ播種(1×105cells/ウェル)した。

(4)スクリーニング
測定用抗原(1)及び(2)を固相化したELISA法でハイブリドーマ培養上清と抗原(1)及び(2)との反応を評価し、抗原(1)に陽性かつ抗原(2)に陰性を示したハイブリドーマを抗活性型GIPモノクローナル抗体産生ハイブリドーマとして選択した。
測定用抗原(1)及び(2)を固相化したELISA法でハイブリドーマ培養上清と抗原(1)及び(2)との反応を評価し、抗原(1)に陽性かつ抗原(2)に陰性を示したハイブリドーマを抗活性型GIPモノクローナル抗体産生ハイブリドーマとして選択した。
(5)クローニング
上記で得られたハイブリドーマを限界希釈法によって単一のコロニーが得られるように培養することにより、抗体産生ハイブリドーマのクローニングを行い、シングルコロニー形成ウェルを再びELISA測定し、抗原(1)に陽性かつ抗原(2)に陰性を示す抗体を産生する9B9H5-B9株を樹立した(国際公開第2016/104439号)。
得られた抗体産生ハイブリドーマの保存は、当該ハイブリドーマを培養し、対数増殖期に回収した後、FBS(Fetal Bovine Serum)含有細胞凍結液にて細胞濃度が1×106cells/mLとなるように調製した後、1×106cells/チューブとなるように凍結チューブに分注し、バイセル中にて-80℃で保存した。
上記で得られたハイブリドーマを限界希釈法によって単一のコロニーが得られるように培養することにより、抗体産生ハイブリドーマのクローニングを行い、シングルコロニー形成ウェルを再びELISA測定し、抗原(1)に陽性かつ抗原(2)に陰性を示す抗体を産生する9B9H5-B9株を樹立した(国際公開第2016/104439号)。

(6)抗体生産
得られた抗体産生ハイブリドーマの凍結バイアルを起眠し、Hybridoma-SFM(Serum-Free Medium)へ無血清馴化した。拡大培養後、ローラーボトル2本(500mL×2本、1L)で培養し、培養上清を回収した。回収した培養上清を、Protein Aを用いたアフィニティークロマトグラフィーにより、モノクローナル抗体として精製した。
得られた抗体産生ハイブリドーマの凍結バイアルを起眠し、Hybridoma-SFM(Serum-Free Medium)へ無血清馴化した。拡大培養後、ローラーボトル2本(500mL×2本、1L)で培養し、培養上清を回収した。回収した培養上清を、Protein Aを用いたアフィニティークロマトグラフィーにより、モノクローナル抗体として精製した。
試験例1 ELISA法による活性型GIPとの反応性
製造例1で得られたモノクローナル抗体と活性型GIPとの反応性をELISA法により確認した。NH2基ビオチン化キット(Dojindo社製)にて抗活性型GIPモノクローナル抗体のアミノ基のビオチン化を行った。Human(total)GIP ELISAキット(Millipore社製)に付属の検出抗体’GIP detection antibody(ビオチン化抗total GIPモノクローナル抗体)の代替として、作製したビオチン化抗活性型GIPモノクローナル抗体1μg/mLを用いてELISAを行った。GIP(1-42)あるいはGIP(3-42)の2000pg/mL溶液を最高濃度とした4倍希釈系列を6段階(8.2~2000pg/mL)まで作製し、捕捉抗体として抗total GIPモノクローナル抗体(Millipore社製 Human GIP(total)ELISAキットに付属)、検出抗体としてビオチン化抗活性型GIPモノクローナル抗体、検出にペルオキシダーゼ‐ストレプトアビジン結合物を用いたサンドイッチELISA系により、GIP濃度をX軸、吸光度450nm-590nmをY軸とした検量線を作製した(図1)。
図1のように、GIP(3-42)では高濃度域においても吸光度が上昇せず、GIP(1-42)にのみ濃度依存的に吸光度が上昇したことから、製造例1で得られたモノクローナル抗体はGIP(3-42)には交差性がなく、GIP(1-42)を特異的に検出できる抗体であることを確認した。
製造例1で得られたモノクローナル抗体と活性型GIPとの反応性をELISA法により確認した。NH2基ビオチン化キット(Dojindo社製)にて抗活性型GIPモノクローナル抗体のアミノ基のビオチン化を行った。Human(total)GIP ELISAキット(Millipore社製)に付属の検出抗体’GIP detection antibody(ビオチン化抗total GIPモノクローナル抗体)の代替として、作製したビオチン化抗活性型GIPモノクローナル抗体1μg/mLを用いてELISAを行った。GIP(1-42)あるいはGIP(3-42)の2000pg/mL溶液を最高濃度とした4倍希釈系列を6段階(8.2~2000pg/mL)まで作製し、捕捉抗体として抗total GIPモノクローナル抗体(Millipore社製 Human GIP(total)ELISAキットに付属)、検出抗体としてビオチン化抗活性型GIPモノクローナル抗体、検出にペルオキシダーゼ‐ストレプトアビジン結合物を用いたサンドイッチELISA系により、GIP濃度をX軸、吸光度450nm-590nmをY軸とした検量線を作製した(図1)。
図1のように、GIP(3-42)では高濃度域においても吸光度が上昇せず、GIP(1-42)にのみ濃度依存的に吸光度が上昇したことから、製造例1で得られたモノクローナル抗体はGIP(3-42)には交差性がなく、GIP(1-42)を特異的に検出できる抗体であることを確認した。
実施例1 GIPによる運動調節機能低下作用と抗GIP抗体による運動調節機能低下抑制作用
(1)動物および飼育方法
C57BLKS/J系統の雄性、レプチン受容体異常マウス(db/dbマウス,オリエンタル酵母(株))を6週齢で搬入後(室温23℃,湿度55±10%,明期;7:00~19:00)、自由摂餌、自由飲水下で飼育した。餌はCE-2(日本クレア(株))を用い、上記環境下で2週間馴化を行った後、試験に供した。
(1)動物および飼育方法

(2)GIP溶液および抗体抗原反応によるGIP結合型抗GIP抗体溶液の作製
生理食塩水にマウス由来GIP(Ana spec社製)を500nMの濃度で溶解し、GIP溶液とした。生理食塩水にマウス由来GIP(Ana spec社製)および製造例1で作製した抗活性型GIP抗体をそれぞれ500nMおよび0.1mg/mLの濃度で溶解し、1~2時間室温でインキュベートした溶液をGIP結合型抗GIP抗体溶液とした。
生理食塩水にマウス由来GIP(Ana spec社製)を500nMの濃度で溶解し、GIP溶液とした。生理食塩水にマウス由来GIP(Ana spec社製)および製造例1で作製した抗活性型GIP抗体をそれぞれ500nMおよび0.1mg/mLの濃度で溶解し、1~2時間室温でインキュベートした溶液をGIP結合型抗GIP抗体溶液とした。
(3)投与量および投与方法
マウス(8週齢)に生理食塩水(対照群)、GIP溶液(5nmol/kg体重)(GIP投与群)、又はGIP結合型抗GIP抗体溶液(GIP;5nmol/kg体重,抗GIP抗体;1mg/kg体重)(GIP+抗GIP抗体投与群)を毎朝(AM9:00~10:00)腹腔内投与した。投与28日後にロータロッドテストを行い、運動調節機能を計測した。
マウス(8週齢)に生理食塩水(対照群)、GIP溶液(5nmol/kg体重)(GIP投与群)、又はGIP結合型抗GIP抗体溶液(GIP;5nmol/kg体重,抗GIP抗体;1mg/kg体重)(GIP+抗GIP抗体投与群)を毎朝(AM9:00~10:00)腹腔内投与した。投与28日後にロータロッドテストを行い、運動調節機能を計測した。
(4)ロータロッド(運動調節機能)テスト
ロータロッド(MK-600A,室町機械社製)を用い、各種速度で回転するロッドの上での滞在能力を測ることで、運動調節機能(バランス能力、および敏捷性を反映した調節力)を評価した。静止したロッド上にマウスを置き、6rpm(1分)→12rpm(1分)→16rpm(1分)→20rpm(1分)→24rpm(1分)→28rpm(1分)のプログラムで落下回数を測定した。
ロータロッド(MK-600A,室町機械社製)を用い、各種速度で回転するロッドの上での滞在能力を測ることで、運動調節機能(バランス能力、および敏捷性を反映した調節力)を評価した。静止したロッド上にマウスを置き、6rpm(1分)→12rpm(1分)→16rpm(1分)→20rpm(1分)→24rpm(1分)→28rpm(1分)のプログラムで落下回数を測定した。
(5)統計解析
解析結果は平均値(Ave.)±標準誤差(SE)で示した。統計解析には2-way ANOVA followed by Bonferroni’s post hoc検定を用い、P値が0.05以下の場合においては統計学的に有意差ありと判定した。

対照群と比較して、GIP投与群で著しい運動調節機能の低下(回転速度の上昇に伴った落下回数の増加)が認められた。GIP+抗GIP抗体投与群では、GIP投与群で認められた運動調節機能の低下は認められず、対照群と同程度の運動調節機能であった(図2)。

C57BL/6J雄性マウス(日本クレア(株))を4週齢で搬入後(室温23℃,湿度55±10%,明期;7:00~19:00)、自由摂餌、自由飲水下で飼育した。餌はCE-2(日本クレア(株))で1週間馴化を行った後、通常食(D12450K,Research Diets,Inc)、又は高脂肪食(D12451,Research Diets,Inc)で95週間飼育した。
(1)動物および飼育方法
C57BL/6J雄性マウス(日本クレア(株))を4週齢で搬入後(室温23℃,湿度55±10%,明期;7:00~19:00)、自由摂餌、自由飲水下で飼育した。餌はCE-2(日本クレア(株))で1週間馴化を行った後、通常食(D12450K,Research Diets,Inc)、又は高脂肪食(D12451,Research Diets,Inc)で95週間飼育した。
(2)採血
各週齢(5,10,15,20,30,40,50,65,80,100週齢)のマウスに対して、イソフルラン麻酔下にて腹部大静脈より全血を採取した。

(5)結果
加齢に伴って血中GIP濃度の上昇が観察され、特に高脂肪食摂取群では通常食群と比較して、有意な血中GIP量の上昇が認められた(図3)。トータルGIPと活性型GIP量は連動して変化することが知られているため(国際公開2012-121302号)、活性型GIP量も上昇していると考えられる。
加齢に伴って血中GIP濃度の上昇が観察され、特に高脂肪食摂取群では通常食群と比較して、有意な血中GIP量の上昇が認められた(図3)。トータルGIPと活性型GIP量は連動して変化することが知られているため(国際公開2012-121302号)、活性型GIP量も上昇していると考えられる。
実施例3 老齢マウスに対する抗GIP抗体の運動調節機能改善作用
(1)動物および飼育方法
C57BL/6J雄性マウス(日本クレア(株))を4週齢で搬入後(室温23℃,湿度55±10%,明期;7:00~19:00)、自由摂餌(D12450K, Research Diets,Inc)、自由飲水下で111週間飼育した。

生理食塩水に製造例1で作製した抗活性型GIP抗体を0.05mg/mLの濃度で溶解し、抗GIP抗体溶液とした。

ロータロッド(MK-600A,室町機械社製)を用い、各種速度で回転するロッドの上での滞在能力を測ることで、運動調節機能(バランス能力、および敏捷性を反映した調節力)を評価した。試験には50週齢(非投与群)および115週齢(非投与群、対照群、抗GIP抗体投与群)のマウスを用いた。静止したロッド上にマウスを置き、6rpm(1分)→12rpm(1分)→16rpm(1分)→20rpm(1分)→24rpm(1分)→28rpm(1分)のプログラムで落下回数を測定した。

非投与(50週齢)群と比較して、非投与(115週齢)群および対照(115週齢)群で著しい運動調節機能の低下(回転速度の上昇に伴った落下回数の増加)が認められた。非投与(115週齢)群および対照(115週齢)群と比較して、抗GIP抗体投与(115週齢)群では運動調節機能の改善が認められ、非投与(50週齢)群と同程度の運動調節機能であった(図4)。
非投与(50週齢)群と比較して、非投与(115週齢)群および対照(115週齢)群で著しい運動調節機能の低下(回転速度の上昇に伴った落下回数の増加)が認められた。非投与(115週齢)群および対照(115週齢)群と比較して、抗GIP抗体投与(115週齢)群では運動調節機能の改善が認められ、非投与(50週齢)群と同程度の運動調節機能であった(図4)。
実施例4 老齢マウスに対するGIP分泌又は上昇抑制剤の運動調節機能改善作用
(1)動物および飼育方法
C57BL/6J雄性マウス(日本クレア(株))を4週齢で搬入後(室温23℃,湿度55±10%,明期;7:00~19:00)、自由摂餌、自由飲水下で飼育した。餌は高脂肪食(D12451,Research Diets,Inc)で57週間(61週齢まで)飼育を行った後、各群体重が同等になるように群分けした。試験食として、30%の脂質を含む高脂肪食(高脂肪食群)、20%ツクリタケ圧搾物を含む30%高脂肪食(ツクリタケ摂取群)、又は30%小麦ふすまを含む30%高脂肪食(小麦ふすま摂取群)をそれぞれ9週間(70週齢)給餌した。飼育期間中、体重は週1回、摂食量は週3回測定した。なお、飼育期間中は自由摂食、自由摂水とした。70週齢でロータロッドテストにて運動調節機能を計測した。

C57BL/6J雄性マウス(日本クレア(株))を4週齢で搬入後(室温23℃,湿度55±10%,明期;7:00~19:00)、自由摂餌、自由飲水下で飼育した。餌は高脂肪食(D12451,Research Diets,Inc)で57週間(61週齢まで)飼育を行った後、各群体重が同等になるように群分けした。試験食として、30%の脂質を含む高脂肪食(高脂肪食群)、20%ツクリタケ圧搾物を含む30%高脂肪食(ツクリタケ摂取群)、又は30%小麦ふすまを含む30%高脂肪食(小麦ふすま摂取群)をそれぞれ9週間(70週齢)給餌した。飼育期間中、体重は週1回、摂食量は週3回測定した。なお、飼育期間中は自由摂食、自由摂水とした。70週齢でロータロッドテストにて運動調節機能を計測した。
(2)飼料の調製および組成比
ツクリタケ(Agaricus bisporus)の子実体(11.3kg)を、室温条件下でスロージューサー(HUROM(株))にて圧搾した後、圧搾液を凍結乾燥し、ツクリタケ圧搾物の粉末(308.2g)を調製した。
小麦ふすまは、日清ファルマ社より購入したものを、エクストルーダー(EA-20、SUEHIRO EPM CORPORATION)にて蒸煮処理後、小麦ふすま重量の35%相当の水を混合し、エクストルーダーにて120℃、5.0MPaの条件で処理を行ったものを蒸煮処理小麦ふすまとして用いた。
ツクリタケ圧搾物および蒸煮処理小麦ふすまの成分組成は食品成分分析センターにて測定した。ツクリタケ圧搾物および蒸煮処理小麦ふすまの組成比を下記表1、表2に示す。また、試験に供した食餌の組成比を下記表3に示す。なおツクリタケ圧搾物、又は小麦ふすまを配合した餌は、各成分組成を考慮し、対照となる高脂肪食と同等の栄養組成・カロリーとなるように成分を置き換える形で混合した。

Claims (26)
- GIP機能阻害剤を有効成分とする運動調節機能向上剤。
- GIP機能阻害剤が、抗GIP抗体、GIP受容体拮抗剤又はGIP分泌又は上昇抑制剤である請求項1記載の運動調節機能向上剤。
- 抗GIP抗体が、抗活性型GIP抗体である請求項2記載の運動調節機能向上剤。
- GIP受容体拮抗剤が、4-ヒドロキシ安息香酸(2-ブロモベンジリデン)ヒドラジド、又は3-シアノ-4-ヒドロキシ安息香酸〔1-(2,3,5,6-テトラメチルベンジル)インドール-4-イル〕メチリデンヒドラジド、3-クロロ-4-ヒドロキシ安息香酸(4-メトキシナフタレン-1-イル)メチリデンヒドラジド、又は3-クロロ-4-ヒドロキシ安息香酸[1-(5-クロロチオフェン-2-イルメチル)-1H-インドール-5-イル]メチリデンヒドラジドである請求項2記載の運動調節機能向上剤。
- GIP分泌又は上昇抑制剤が、ツクリタケの圧搾物又は抽出物、小麦ふすま、3-ブロモ-5-メチル-2-フェニルピラゾロ[1,5-a]ピリミジン-7-オール、アルギン酸、フォスファチジルエタノールアミン、ポリグルタミン酸、キラヤ、リゾフォスファチジルイノシトール、セルロースナノファイバ、β-キチンナノファイバー、ジアシルグリセロール、ヒドロキシプロピル化澱粉、モノアシルグリセロール、炭素数20以上の超長鎖脂肪酸、構成脂肪酸の1質量%以上がドコサヘキサエン酸であり、1質量%以上がエイコサペンタエン酸であるトリアシルグリセロール、長鎖不飽脂肪酸エタノールアミド、米糠抽出物、構成脂肪酸の10質量%以上がα-リノレン酸であるトリアシルグリセロール、又はグリセロール骨格の2位にC14~C18飽和脂肪酸が結合したアシルグリセロールである請求項2記載の運動調節機能向上剤。
- 運動調節機能向上剤を製造するための、GIP機能阻害剤の使用。
- GIP機能阻害剤が、抗GIP抗体、GIP受容体拮抗剤又はGIP分泌又は上昇抑制剤である請求項6記載の使用。
- 抗GIP抗体が、抗活性型GIP抗体である請求項7記載の使用。
- GIP受容体拮抗剤が、4-ヒドロキシ安息香酸(2-ブロモベンジリデン)ヒドラジド、又は3-シアノ-4-ヒドロキシ安息香酸〔1-(2,3,5,6-テトラメチルベンジル)インドール-4-イル〕メチリデンヒドラジド、3-クロロ-4-ヒドロキシ安息香酸(4-メトキシナフタレン-1-イル)メチリデンヒドラジド、又は3-クロロ-4-ヒドロキシ安息香酸[1-(5-クロロチオフェン-2-イルメチル)-1H-インドール-5-イル]メチリデンヒドラジドである請求項7記載の使用。
