Abstract
根菜類のような原料を用いた場合でも、低コストであり、蛋白質の等電点以上の酸性pH域において安定な酸性蛋白飲食品を得ることができるような、水溶性多糖類を提供することを目的とする。 湿潤状態の根菜由来の原料からpH2.7~3.7の酸性条件下において熱水抽出される水溶性多糖類が、蛋白質の等電点以上のpH域において酸性蛋白飲食品を良好に安定化できる。
Description
本発明は、根菜類由来の水溶性多糖類、およびその製造方法に関する。
酸性蛋白質飲食品はさわやかな酸味を持つ飲食品として好まれるが、酸性下では、蛋白質が凝集沈殿してしまい商品としての価値が著しく損なわれる。そこで、従来より酸性蛋白飲食品の製造に際しては、蛋白質の等電点以下のpH域にて、蛋白粒子の凝集、沈殿等を防止する目的で高メトキシペクチン、カルボキシメチルセルロース、高分子型水溶性大豆多糖類により安定化する技術が開示されている(特許文献1)。
一方、近年、等電点以上の酸性pH域のおける蛋白の分散安定剤についての要望が高まってきている。例えば、中性からp H 5 . 2 までの微酸性p H域において、有機酸塩を加えることにより蛋白成分を安定化する技術(特許文献2)が開示されている。また、根菜類をpH3.8~5.3条件下で熱水抽出して得られるペクチンを加えることにより蛋白成分を安定化する技術(特許文献3)が開示されている。
しかし、特許文献2の技術では、安定化された蛋白液の乳濁性が消失し、加えた有機酸塩の影響により良好な酸味が得られないという問題があった。また、特許文献3の技術では、特許文献2のような風味の問題はないものの、実際、実施例で具体的に効果を確認しているのは、原料として乾燥したコストの高いものを用いた場合であり、乾燥工程を経ていない原料で、特許文献3の抽出pHで水溶性多糖類を製造したところpH5.0付近の酸性飲料の蛋白安定性効果が十分満足できるものではないことがわかった。従って、風味を良くするため、根菜類のような原料を用いた場合でも、低コストであり、蛋白質の酸性pH域において安定な酸性蛋白飲食品を得ることができるような、水溶性多糖類を提供することを目的とする。
本発明者らは、上記課題の解決を指向して鋭意研究した結果、湿潤状態の根菜由来の原料からpH2.7~3.7の酸性条件下において熱水抽出される水溶性多糖類が、蛋白質の等電点以上の酸性pH域において酸性蛋白飲食品を良好に安定化できるという知見を得た。本発明は、かかる知見に基づいて完成されたものである。
すなわち本発明は、
(1)湿潤状態の根菜類由来の原料からpH2.7~3.7で熱水抽出することを特徴とする、根菜類由来の水溶性多糖類の製造方法、
(2)抽出pHが、pH2.8~3.6である、(1)記載の根菜類由来の水溶性多糖類の製造方法、
(3)根菜類由来の原料が馬鈴薯である、(1)または(2)記載の根菜類由来の水溶性多糖類の製造方法、
(4)HPLCのゲル濾過クロマトグラフィーの分析の方法で測定したときに、下記を特徴とする根菜類由来の水溶性多糖類、
(A)(第二ピーク面積/第一ピーク面積)×100の数値が10%以上、
(B)(第一ピークにおけるピーク強度/第二ピークにおけるピーク強度)×100の数値が70%以上、

本発明により、乾燥工程を経ていない湿潤状態の根菜類由来の原料を用いることで、低コストで等電点以上の酸性pH域で酸性蛋白飲食品を安定化できる水溶性多糖類を得ることができる。
さらに、製造された酸性蛋白食品は、レトルト殺菌などの加熱後も安定な状態を保持できるようになる。

以下に本発明について具体的に説明する。
(水溶性多糖類)
本発明の水溶性多糖類は、湿潤状態の根菜類由来の原料からpH2.7~3.7で熱水抽出することにより得られる。
また、本発明の根菜類由来の水溶性多糖類は、HPLCのゲル濾過クロマトグラフィーの分析の方法で測定したときに以下の(A)、(B)の特徴を有する。
(A)(第二ピーク面積/第一ピーク面積)×100の数値が10%以上。
(B)(第一ピークにおけるピーク強度/第二ピークにおけるピーク強度)×100の数値が70%以上。
なお、第一ピーク面積は第一ピークと接線で囲まれた面積であり、第二ピーク面積は第二ピークと接線で囲まれた面積を示し、ゲルろ過において保持時間が早いピークより、第一ピーク、第二ピークとする。ピーク強度は、各ピークにおける頂点の高さを示す。
より、具体的な説明を以下に行う。
本発明の水溶性多糖類は、湿潤状態の根菜類由来の原料からpH2.7~3.7で熱水抽出することにより得られる。
また、本発明の根菜類由来の水溶性多糖類は、HPLCのゲル濾過クロマトグラフィーの分析の方法で測定したときに以下の(A)、(B)の特徴を有する。
(A)(第二ピーク面積/第一ピーク面積)×100の数値が10%以上。
(B)(第一ピークにおけるピーク強度/第二ピークにおけるピーク強度)×100の数値が70%以上。
なお、第一ピーク面積は第一ピークと接線で囲まれた面積であり、第二ピーク面積は第二ピークと接線で囲まれた面積を示し、ゲルろ過において保持時間が早いピークより、第一ピーク、第二ピークとする。ピーク強度は、各ピークにおける頂点の高さを示す。
より、具体的な説明を以下に行う。
(ピーク強度、ピーク面積の算出方法)
ゲル濾過クロマトグラフィー分析により分離処理した溶出ピークのうち、保持時間の早いピークより、第一ピーク、第二ピーク、第三ピークとする。なお、本発明ではショルダーピークもピークとして扱う。
クロマトグラフ曲線の第一ピークと第二ピーク間にある下に凸のカーブと、第二ピークと第三ピーク間にある下に凸のカーブの曲線の微分(傾き)を求める。このとき、両方の傾きが同じく、かつ、その2つの接線が、もう一方の接点を通る接線を作成する。その接線と曲線で囲まれたピーク面積を「第二ピーク面積」とする。その接線を第一ピーク側へ延長した時に囲まれるピーク面積を「第一ピーク面積」とする(図1)。
また、第一ピークのピーク強度は、第一ピークの、ピークの頂点の高さのことであり、同様に、第二ピークのピーク強度は第二ピークの、ピークの頂点の高さのことである(図2)。
以下の(A)、(B)の式からそれぞれ数値を算出する。
(A)(第二ピーク面積/第一ピーク面積)×100の数値。
(B)(第一ピークにおけるピーク強度/第二ピークにおけるピーク強度)×100の数値。
本発明の水溶性多糖類は、(A)の数値が10%以上、かつ、(B)の数値が70%以上である。(A)の数値は好ましくは、12%以上であり、(B)の数値は好ましくは75%以上である。
ゲル濾過クロマトグラフィー分析により分離処理した溶出ピークのうち、保持時間の早いピークより、第一ピーク、第二ピーク、第三ピークとする。なお、本発明ではショルダーピークもピークとして扱う。
クロマトグラフ曲線の第一ピークと第二ピーク間にある下に凸のカーブと、第二ピークと第三ピーク間にある下に凸のカーブの曲線の微分(傾き)を求める。このとき、両方の傾きが同じく、かつ、その2つの接線が、もう一方の接点を通る接線を作成する。その接線と曲線で囲まれたピーク面積を「第二ピーク面積」とする。その接線を第一ピーク側へ延長した時に囲まれるピーク面積を「第一ピーク面積」とする(図1)。
また、第一ピークのピーク強度は、第一ピークの、ピークの頂点の高さのことであり、同様に、第二ピークのピーク強度は第二ピークの、ピークの頂点の高さのことである(図2)。
以下の(A)、(B)の式からそれぞれ数値を算出する。
(A)(第二ピーク面積/第一ピーク面積)×100の数値。
(B)(第一ピークにおけるピーク強度/第二ピークにおけるピーク強度)×100の数値。
本発明の水溶性多糖類は、(A)の数値が10%以上、かつ、(B)の数値が70%以上である。(A)の数値は好ましくは、12%以上であり、(B)の数値は好ましくは75%以上である。
(ゲル濾過クロマトグラフィー) HPLCのゲル濾過クロマトグラフィーの分析条件は以下の通りである。
水溶性多糖類の1重量%水溶液(50mM酢酸ナトリウム水溶液, pH5.0)を調製し、0.8μmのフィルターにて濾過した濾液を分析に供する。予め、50mM酢酸ナトリウム水溶液, pH5.0, 40℃で平衡化したTSK-gel G-5000PWXLカラム(東ソー;φ7.8mm×300mm)に濾液20μL添加し、流速:1.0mL/minで分離する。分離した多糖類は、示差屈折検出器(RI検出器)にて検出する。

(原料)
本発明の湿潤状態の根菜類由来の原料として、馬鈴薯、甘藷、里芋、山芋、コンニャクなどのイモ類、ゴボウ、ニンジン、大根、ハス、ビートなどが例示できるが、特に馬鈴薯類が好ましい。このような馬鈴薯類は、デンプン産業の加工副産物として生成される湿潤状態のデンプン粕を使用することが好ましい。
なお、本発明において「湿潤状態」とは、乾燥工程を経ていない水分を含む、いわゆる生原料を意味する。例えば、デンプン産業の加工副産物であるデンプン粕の場合、保存性の観点から乾燥されることが多いが、この乾燥工程を経ていない状態のものをいう。湿潤状態の原料の水分は概ね40~99.9重量%である。好ましくは、50~99重量%である。
本発明の湿潤状態の根菜類由来の原料として、馬鈴薯、甘藷、里芋、山芋、コンニャクなどのイモ類、ゴボウ、ニンジン、大根、ハス、ビートなどが例示できるが、特に馬鈴薯類が好ましい。このような馬鈴薯類は、デンプン産業の加工副産物として生成される湿潤状態のデンプン粕を使用することが好ましい。
なお、本発明において「湿潤状態」とは、乾燥工程を経ていない水分を含む、いわゆる生原料を意味する。例えば、デンプン産業の加工副産物であるデンプン粕の場合、保存性の観点から乾燥されることが多いが、この乾燥工程を経ていない状態のものをいう。湿潤状態の原料の水分は概ね40~99.9重量%である。好ましくは、50~99重量%である。
(抽出pH)
湿潤状態の根菜類からのペクチン性多糖類の抽出は、pH2.7~3 . 7の酸性下にて行うことが必須である。このpH域から外れた範囲において抽出されたペクチン性多糖類、および、HPLCのゲル濾過クロマトグラフィーの分析の方法で測定したときに特徴的な第二ピークが現れないペクチン性多糖類は、等電点以上のpH域での蛋白質の十分な分散安定化機能は発現されない。
抽出pHは、好ましくはpH2.8~3.6であり、より好ましくはpH3.0~3.6である。
湿潤状態の根菜類からのペクチン性多糖類の抽出は、pH2.7~3 . 7の酸性下にて行うことが必須である。このpH域から外れた範囲において抽出されたペクチン性多糖類、および、HPLCのゲル濾過クロマトグラフィーの分析の方法で測定したときに特徴的な第二ピークが現れないペクチン性多糖類は、等電点以上のpH域での蛋白質の十分な分散安定化機能は発現されない。
抽出pHは、好ましくはpH2.8~3.6であり、より好ましくはpH3.0~3.6である。
(抽出温度)
また、上記のpH範囲における水溶性多糖類の抽出温度は、100℃を超える温度にて行うのが好ましい。100℃以下の温度で抽出を行った場合には、水溶性多糖類の溶出に時間がかかり経済的に不利である。一方、温度が高温になるに従って抽出は短時間で済むが、余りに高温にし過ぎると風味、色調に悪影響を及ぼすと共に水溶性多糖類の低分子化が進み機能の発現効果が低下する場合があるので、130℃以下で行うのが好ましい。
また、上記のpH範囲における水溶性多糖類の抽出温度は、100℃を超える温度にて行うのが好ましい。100℃以下の温度で抽出を行った場合には、水溶性多糖類の溶出に時間がかかり経済的に不利である。一方、温度が高温になるに従って抽出は短時間で済むが、余りに高温にし過ぎると風味、色調に悪影響を及ぼすと共に水溶性多糖類の低分子化が進み機能の発現効果が低下する場合があるので、130℃以下で行うのが好ましい。
(精製)
抽出した水溶性多糖類は、夾雑物等の精製を行なうことができ、エタノールやイソプロパノール等の親水性極性溶媒による分配、脱塩、活性炭処理など何れの方法も利用できる。

抽出した水溶性多糖類は、水溶液の状態でも用いることはできるが、水溶液を乾燥して粉体で用いることもできる。乾燥方法として、例えば、噴霧乾燥や凍結乾燥、間接加熱による乾燥等が挙げられる。
(分散安定剤)
本発明の水溶性多糖類は酸性蛋白飲食品の分散安定剤として使用することができる。
本発明の水溶性多糖類の使用量としては、標準的に最終製品に対して好ましくは0.05~10重量% 、より好ましくは0. 2~5重量%であるが、蛋白濃度の相違などに応じて変化し得るので、この使用量は本発明の範囲を制限するものではない。
本発明の水溶性多糖類は酸性蛋白飲食品の分散安定剤として使用することができる。
本発明の水溶性多糖類の使用量としては、標準的に最終製品に対して好ましくは0.05~10重量% 、より好ましくは0. 2~5重量%であるが、蛋白濃度の相違などに応じて変化し得るので、この使用量は本発明の範囲を制限するものではない。
また、本発明の酸性蛋白飲食品の製造に際して、従来よりある安定剤、例えばリンゴまたは柑橘類由来のペクチン、水溶性大豆多糖類、カルボキシメチルセルロースナトリウム、アルギン酸プロピレングリコールエステル、カラギーナン、微結晶セルロース、キトサン、有機酸塩、重合リン酸塩、乳化剤、加熱変性蛋白質などと併用しても良く、それにより安定なpH域の拡大等を図ることができる。
(酸性蛋白質飲食品)
本発明の酸性蛋白質飲食品とは、動植物、微生物由来の蛋白質を含有する酸性蛋白質飲食品であり、酸性蛋白飲料や、酸性蛋白食品が挙げられる。
酸性蛋白飲料として、牛乳、豆乳等の動植物性蛋白を使用した飲料に柑橘類果汁又はその他の果汁、或いはクエン酸、乳酸などの有機酸もしくは燐酸などの無機酸を添加してなる飲料や、乳酸菌飲料(生菌、殺菌タイプを含む)、ドリンクヨーグルト類、ケフィア等の発酵乳飲料等が挙げられる。
また、酸性蛋白食品として、乳製品を酸性にした酸性乳飲料やアイスクリームなどの乳成分入りの冷菓に果汁等を加えた酸性アイス、フローズンヨーグルトなどの酸性冷菓、プリン、ババロア等のゲル化食品に果汁などを加えた酸性デザートや、ヨーグルト等が挙げられる。
これらの中でも特に酸性蛋白飲料が好ましい。
本発明で酸性とはpH 6. 5以下のpH域を指し、好ましくは、pH6.0以下である。また、pHの下限は好ましくはpH4.2以上、より好ましくはpH4.5以上である。酸性蛋白質飲食品のpHを上記のような範囲に設定することで、本発明の水溶性多糖類の分散安定化効果がより効果的に発揮される。
本発明の酸性蛋白質飲食品とは、動植物、微生物由来の蛋白質を含有する酸性蛋白質飲食品であり、酸性蛋白飲料や、酸性蛋白食品が挙げられる。

これらの中でも特に酸性蛋白飲料が好ましい。

以下、実施例により本発明の実施態様を説明する。なお、例中、部及び%は何れも重量基準を意味する。
(実施例1) 水溶性多糖類(イ)の調製
湿潤状態の馬鈴薯デンプン粕(水分86%) 200 gを水360 gに懸濁した後に、pHを3. 5に調整後、110℃、90分間加熱して、水溶性多糖類を抽出した。冷却後、遠心分離(10000×g、30分間)を行い、水溶性多糖類抽出液と沈殿部に分離した。分離した沈殿部に等重量の水を加えて再度遠心分離を行い、上澄み液を先のペクチン抽出液と混合した後に、そのまま凍結乾燥して各粗水溶性多糖類を得た。60重量%になるようにエタノールを加えて水溶性多糖類を沈殿させ、90重量%の含水エタノールで洗浄し、得られた沈殿を風乾して、水溶性多糖類(イ)を得た。
湿潤状態の馬鈴薯デンプン粕(水分86%) 200 gを水360 gに懸濁した後に、pHを3. 5に調整後、110℃、90分間加熱して、水溶性多糖類を抽出した。冷却後、遠心分離(10000×g、30分間)を行い、水溶性多糖類抽出液と沈殿部に分離した。分離した沈殿部に等重量の水を加えて再度遠心分離を行い、上澄み液を先のペクチン抽出液と混合した後に、そのまま凍結乾燥して各粗水溶性多糖類を得た。60重量%になるようにエタノールを加えて水溶性多糖類を沈殿させ、90重量%の含水エタノールで洗浄し、得られた沈殿を風乾して、水溶性多糖類(イ)を得た。
(実施例2~4) 水溶性多糖類(ロ、ハ、ニ)の調製
抽出時のpHを3.7、3.0、2.8に代えた他は、実施例1と同様にして、それぞれ、水溶性多糖類(ロ)~(ニ)を得た。

(比較例1~4) 水溶性多糖類(ホ、ヘ、ト、チ)の調製
抽出時のpHを5.0、4.5、4.0、2.5に代えた他は、実施例1と同様にして、それぞれ、水溶性多糖類(ホ)~(チ)を得た。
抽出時のpHを5.0、4.5、4.0、2.5に代えた他は、実施例1と同様にして、それぞれ、水溶性多糖類(ホ)~(チ)を得た。
(比較例5) 水溶性多糖類(リ)の調製
原料を湿潤から乾燥に代えた他は、実施例1と同様にして水溶性多糖類(リ)を得た。

回収した各水溶性多糖類を使用して表1の配合によりpH 5 . 0での蛋白質の分散安定化機能の評価に供した。評価結果を表2に示した。また、各水溶性多糖類のゲルろ過の分析結果を表2に示した。pH5.0での蛋白質の良好な分散安定性を示した。
○安定性の評価
調製した乳飲料について、沈殿率にて安定性を評価した。尚、測定方法及び評価方法を以下に示す。
[沈殿率]
酸性乳飲料20gを遠心管に量り取り、遠心分離機(型式H-103N、株式会社コクサン製)にて2,000rpmで20分間遠心分離する。上清をデカンテーションで除去し、沈殿重量を測定する。尚、沈殿率は以下の計算式により算出する。
沈殿率(%)=(沈殿物重量)/(分取した酸性乳飲料重量)×100
沈殿率は、沈殿率1%未満を◎、沈殿率1%以上3%未満を○、沈殿率3%以上7%未満を△、沈殿率7%以上を×と表記する。
調製した乳飲料について、沈殿率にて安定性を評価した。尚、測定方法及び評価方法を以下に示す。
[沈殿率]
酸性乳飲料20gを遠心管に量り取り、遠心分離機(型式H-103N、株式会社コクサン製)にて2,000rpmで20分間遠心分離する。上清をデカンテーションで除去し、沈殿重量を測定する。尚、沈殿率は以下の計算式により算出する。
沈殿率(%)=(沈殿物重量)/(分取した酸性乳飲料重量)×100
沈殿率は、沈殿率1%未満を◎、沈殿率1%以上3%未満を○、沈殿率3%以上7%未満を△、沈殿率7%以上を×と表記する。
水溶性多糖類(イ)~(ニ)は、pH5.0での蛋白質の良好な分散安定性を示した。一方、本発明の抽出pHの範囲外で抽出された水溶性多糖類(ホ)~(チ)は、ゲルろ過の分析値が本発明からは逸脱し、pH5.0での蛋白質の分散安定性は悪かった。また、原料として乾燥原料を用いた水溶性多糖類(リ)の場合、本発明の抽出pHの条件で抽出してもゲルろ過の分析値が本発明からは逸脱し、pH5.0での蛋白質の分散安定性は悪かった。
(実施例5~6)
実施例1で回収した水溶性多糖類(イ)を使用し、クエン酸液のpH調整を4.5、5.5に代えた他は、表1の配合と同様にしてpH 4.5、5.5での蛋白質の分散安定化機能の評価に供した。評価結果を表3に示した。各pHでの蛋白質の良好な分散安定性を示した。
実施例1で回収した水溶性多糖類(イ)を使用し、クエン酸液のpH調整を4.5、5.5に代えた他は、表1の配合と同様にしてpH 4.5、5.5での蛋白質の分散安定化機能の評価に供した。評価結果を表3に示した。各pHでの蛋白質の良好な分散安定性を示した。
Claims (11)
- 湿潤状態の根菜類由来の原料からpH2.7~3.7で熱水抽出することを特徴とする、根菜類由来の水溶性多糖類の製造方法。
- 抽出pHが、pH2.8~3.6である、請求項1記載の根菜類由来の水溶性多糖類の製造方法。
- 根菜類由来の原料が馬鈴薯である、請求項1または2記載の根菜類由来の水溶性多糖類の製造方法。
- HPLCのゲル濾過クロマトグラフィーの分析の方法で測定したときに、下記を特徴とする根菜類由来の水溶性多糖類。
(A)(第二ピーク面積/第一ピーク面積)×100の数値が10%以上。
(B)(第一ピークにおけるピーク強度/第二ピークにおけるピーク強度)×100の数値が70%以上。
なお、第一ピーク面積は第一ピークと接線で囲まれた面積であり、第二ピーク面積は第二ピークと接線で囲まれた面積を示し、ゲルろ過において保持時間が早いピークより、第一ピーク、第二ピークとする。ピーク強度は、各ピークにおける頂点の高さを示す。 - (A)(第二ピーク面積/第一ピーク面積)×100の数値が12%以上、
(B)(第一ピークにおけるピーク強度/第二ピークにおけるピーク強度)×100の数値が75%以上、
である、請求項4記載の根菜類由来の水溶性多糖類。 - 請求項4または5記載の水溶性多糖類を含有する酸性蛋白飲食品。
- 酸性蛋白飲食品のpHが4.2~6.5である、請求項6記載の酸性蛋白飲食品。
- 酸性蛋白飲食品のpHが4.5~6.5である、請求項6記載の酸性蛋白飲食品。
- 酸性蛋白飲食品のpHが4.2~6.0である、請求項6記載の酸性蛋白飲食品。
- 酸性蛋白飲食品のpHが4.5~6.0である、請求項6記載の酸性蛋白飲食品。
- 酸性蛋白飲食品が酸性蛋白飲料である、請求項6~10何れか1項に記載の酸性蛋白飲食品。
