Abstract
본 발명은 골 질환 및 지질 관련 대사성 질환의 예방, 개선 또는 치료용 기능성 조성물에 관한 것으로, 보다 상세하게는 프로바이오틱스 및 비타민 복합체를 동시에 사용함으로써 항산화 효과, 혈중 지질 농도의 감소, 체내 칼슘 및 철분의 흡수 촉진에 대한 시너지 효과에 의해 골 질환, 비만 및 지질 관련 대사성 질환을 효과적으로 예방, 개선 또는 치료할 수 있는 조성물에 관한 것이다.
Description
본 발명은 골 질환, 비만 및 지질 관련 대사성 질환의 예방, 개선 또는 치료용 기능성 조성물에 관한 것이다.
최근 우리나라는 경제성장과 더불어 식생활이 서구화되면서 섭취하는 열량이 늘어나고, 그에 따라 비만 인구도 증가하는 추세이다. 구체적으로 2013년 보건복지통계연보에 따르면, 만 19세 이상 성인들 중에서 비만인의 비율이 31.4%로 2010년에 조사된 30.9%보다 0.5% 증가하였으며, 소아 비만율도 빠르게 증가하고 있는 추세이다.
이러한 비만은 그 자체로도 체중이 증가하고 몸집이 비대해져 생활에 불편을 주지만 더 큰 문제는 혈중 지질 농도를 높여, 동맥경화와 심장병을 발생시키고, 인슐린 저항성을 높여 당뇨, 생리불순, 암 등의 합병증을 유발하며, 고지혈증, 고혈압, 관상동맥 및 뇌졸중 등의 만성 성인병 질환의 원인이 되기 때문에 비만의 치료와 예방은 필수적이다(Lee JH, J. Kor. Soc. Obes., 1:21-24, 1992; Lew EA, Ann. Intern. Med., 103:1024-1029, 1985; Kim KI et al., Korean J. Food Sci. Technol., 35:720-725, 2003).
비만을 유발하는 원인은 유전적 영향, 서구화되는 식생활에 의한 환경적인 영향, 스트레스에 의한 심리적인 영향 등에 의해 유발되는 것으로 알려져 있으나 그 정확한 원인이나 기작에 대해서는 명확히 밝혀져 있지 않다.
과거 지방세포는 단순히 인체의 잉여에너지를 중성지방의 형태로 저장하고 외부로부터 충격을 완충해주는 작용을 하는 세포로 인식되어 왔다. 하지만 최근에 지방세포는 공복, 대사, 인슐린 민감도(insulin sensitivity)를 조절하는 아디포사이토카인(adipocytokine)을 분비하는 내분비기관으로서 인식되고 있다. 구체적으로, 아디포넥틴(adiponectin), 렙틴(leptin), 레지스틴(resistin), 종양괴사인자-α(tumor necrosis factor alpha; TNF-α), 인터루킨-6(interleukin-6; IL-6) 등의 아디포사이토카인(adipocytokines)이 항상성 유지와 에너지 대사 조절에 중요한 역할을 한다고 알려져 있다(Matsuzawa, Y. et al., Ann. Ny. Acad. Sci., 892:146-154, 1999; Saltiel, A.R., Nat. Med., 7:887-888, 2001).
특히, 비만 유전자에서 생성되는 렙틴은 지방세포의 양과 비례하므로 비만자의 혈청 렙틴의 농도가 정상 체중자에 비하여 높게 나타나고, 내장지방보다는 피하지방에서 더 많이 생성되는데, 이러한 렙틴은 지방세포의 내분비 개념으로 비만에 중요한 역할을 하고, 섭식조절뿐만 아니라 에너지 소비와 생식기능까지 광범위하게 작용하며, 식욕을 억제하고 교감신경계를 통하여 열 생성을 증가시킨다. 또한, 뇌를 순환하면서 시상하부의 수용체에 작용하여 식이를 억제하고, 체내지방이 증가하면 뇌에서 분비되는 물질로서 뇌에 포만감을 관장하는 만복중추(satiety center)를 자극해 식욕을 저해하는 물질 중 하나로 밝혀졌다(Mantzoros, C.S., Ann Intern. Med., 130:671-680, 1999; Roemmich, J.N. and A.D., Am. J. Hum. Biol., 11:209-224, 1999).
이와 같은 비만을 치료하기 위한 목표는 크게 두 가지로 구분할 수 있다. 첫 번째는 과량의 지방을 연소시켜 체중을 감소시키는 것이며, 두 번째는 대사성 불균형(metabolic imbalance)을 개선시키는 것이다. 현재 비만의 치료는 체중감량뿐만 아니라, 조기에 심혈관 질환을 유발하는 요인을 제거하여 대사 이상을 개선하는 데 그 목표가 있다. 또한 식이섭취 조절 및 에너지 소비 조절을 통해 비만을 억제하는 연구도 활발히 이루어지고 있다. 음식을 섭취하는 행동을 조절하는 기관으로 시상하부와 운동신경, 자율신경 및 말초신경계가 모두 관여하는데 비만의 병인론에 있어서 중추신경계 중 특히 시상하부가 중요한 역할을 하며 뉴로펩타이드 Y, POMC/CART, 멜라노코틴 수용체, 노르에피네프린, 세로토닌 등이 시상하부에서 분비되는 대표적인 인자들이다. 현재 비만치료제 개발 전략은 식사량 감소, 열량흡수의 억제, 발열반응 촉진, 에너지 대사 조절, 신경계를 통한 신호전달조절과 같은 것들이다(박미정,Korean J Pediatr 48(2), 2005).
현재까지 알려진 비만치료제는 작용 기작에 따라 크게 포만감 항진제, 지방흡수 억제제, 항정신성 식욕 억제제로 나뉘며, 가장 대표적인 약물들로는 제니칼(XenicalTM, 로슈제약회사, 스위스), 리덕틸(ReductilTM, 애보트사, 미국), 엑소리제(ExoliseTM, 아토파마, 프랑스) 등이 있으나 지방변, 장내가스발생, 복부팽만감, 배변 실금 등을 유발하고, 심장질환, 호흡기 질환, 신경계질환 등의 부작용을 동반하며, 그 효능의 지속성이 낮다는 문제점이 있다.
이에 상기와 같은 인공적으로 합성된 물질의 부작용을 최소화하기 위해 천연물질로부터 체중조절에 효과적인 기능성 물질의 개발이 이루어지고 있다. 이러한 대표적 예로 녹차, 인산, 당귀, 차가버섯, 홍삼, 다래나무, 갈조류 등이 있다. 그러나 이러한 천연물질로부터 추출한 항비만 물질의 경우 효능을 나타내는 물질의 유효농도가 약하고, 농경지 등에서 재배하여야 하므로 많은 비용이 소용되는 등의 문제가 있다.
한편, 골 손실 및 골절 위험이 높은 것을 특징으로 하는 골다공증은 특히 노인에서는 가장 흔한 질환 중 하나이며, 이는 전세계적으로는 대략 1억 여명의 사람이 병에 걸린 것으로 추정된다. 노인에서의 뼈 골절 발생률은 일반적으로 증가하고 있지만, 치료학상의 선택 범위는 한정되어 있다. 현재, 골흡수 억제제(예컨대, 비스포스포네이트, 데노수맙, 호르몬 요법)가 가장 보편적으로 사용되는 골다공증 치료법이다. 상기 작용제는 골 재형성을 저속화시키고, 골 밀도를 증가시키도록 디자인된 것이다. 그러나 이는 턱의 골괴사, 비정형 골절, 심방 세동, 및 뇌졸중 또는 암의 위험 증가를 비롯한, 유의적인 부작용과 연관된다. 동화제는 골다공증 환자에서 새로 골을 생성하는 데 사용될 수 있다. 그러나 골량을 증가시키고, 골아세포매개 골 형성과 골수 지방축적 사이의 균형을 조절하는 동화 인자를 찾는 것이 도전 과제가 되어 왔다. 추가로, 상업적으로 이용 가능한 유일의 동화제(테리파라티드)는 매우 비싸고, 투여하기 어려울 뿐만 아니라, 혈압 하락, 구역, 통증, 쇠약, 및 우울증을 비롯한 부작용과도 관련이 있다. 또한, 래트에서의 테리파라티드의 사용은 악성 종양 성장(골원성 암종)을 유발하는 것으로 나타났다. 일반적으로, 골다공증을 위한 치료학상의 선택 범위는 한정되어 있고, 골 형성을 자극하는 새로운 치료학적 접근법의 개발이 우선시되고 있다.
본 발명의 일 목적은 항산화 효과와 혈중 지질 및 콜레스테롤의 함량을 감소시키는 효과가 뛰어나 비만 및 지질 관련 대사성 질환을 예방, 개선 또는 치료할 수 있는 조성물을 제공하고자 한다.
본 발명의 다른 목적은 칼슘 및 철분의 흡수를 촉진하여 골 질환을 예방, 개선 또는 치료할 수 있는 조성물을 제공하고자 한다.
그러나 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 당업계에서 통상의 지식을 가진 자에게 명확하게 이해될 수 있을 것이다.
본 발명의 발명자들은 유산균과 비타민 복합체를 함께 사용하는 경우 항산화 효과, 혈중 지질 농도의 감소, 체내 칼슘 및 철분의 흡수 촉진에 시너지 효과가 부여됨을 발견하여 본 발명에 이르게 되었다.
본 발명의 일 구현 예에 따르면, 프로바이오틱스(probiotics) 및 비타민 복합체를 유효 성분으로 포함하는 비만 및 지질 관련 대사성 질환의 예방 또는 치료용 약학 조성물에 관한 것이다.
본 발명에서, "프로바이오틱스(probiotics)"란, 장의 미생물 균형을 개선함으로써 숙주에 유리한 영향을 미치는 살아있는 미생물 식품 보충제로, 보다 광범위하게는 특정한 수로 섭취될 경우 본래의 기초적인 영양을 너머 건강상의 효과를 부여하는 살아 있는 미생물로서 정의될 수 있다. 다양한 미생물, 특히 락토바실러스(Lactobacillus) 및 비피도박테리엄의 혼합제 (cocktail)는 전통적으로 건강을 향상시키기 위해 발효된 유제품에 사용되어왔다. 그러나, 효과적이기 위해, 상기 프로바이오틱은 제조 공정, 포장 및 저장 조건에서 생존해야 할 뿐만 아니라, 프로바이오틱 물질이 긍정적인 건강 효과를 갖기 위해 생육 가능하게 남아있도록 위장관을 통해 생존 가능하게 통과되어야 한다는 요건을 필요로 한다.
본 발명에서 상기 프로바이오틱스 유산균 균주들은 유산균을 위한 일반적이 배양 방법으로 성장되고, 원심분리와 같은 분리과정으로 회수되며, 건조, 이에 제한되지 아니하지만 동결건조에 의해 생균제 형태로 제조하여 이용될 수 있다.
본 발명에서 상기 프로바이오틱스의 종류는 특별히 제한하지 않으나, 예를 들면, 스트렙토코커스(Streptococcus) 속, 락토코커스(Lactococcus) 속, 엔테로코커스(Enterococcus) 속, 락토바실러스(Lactobacillus) 속, 페디오코커스(Pediococcus) 속, 류코노스톡(Leuconostoc) 속, 비셀라(Weissella) 속 및 비피도박테리움(Bifidobacterium) 속으로 이루어진 군에서 선택된 하나 이상의 유산균 균주를 포함할 수 있으며, 바람직하게는 엔테로코커스 페슘(Enterococcus
faecium), 락토바실러스 람노수스(Lactobacillus
rhamnosus) 또는 이들의 혼합물을 포함할 수 있다.
본 발명에서 상기 프로바이오틱스는 조성물의 총 중량에 대하여 3 X 109 내지 6 X 109 CFU/g의 함량으로 포함될 수 있으나, 이에 제한되는 것은 아니다. 프로바이오틱스의 함량이 3 X 109 CFU/g 이하에 해당하는 경우, 포함되는 함량이 적어 항산화 효과를 증가시키는 효과를 얻을 수 없고,프로바이오틱스의 함량이 6 X 109 CFU/g을 초과하는 경우에는 조성물의 성상이 나쁠 수 있다.
또한, 본 발명에서 상기 비타민 복합체로는 수용성 비타민, 지용성 비타민 또는 이들의 혼합물 중 2종 이상의 비타민 혼합물일 수 있다. 바람직하게는 상기 비타민 복합체는 카로티노이드(예, 베타-카로틴, 제아잔틴, 루테인, 리코펜), 비오틴, 콜린, 이노시톨, 엽산, 판토텐산, 콜린, 비타민 A, 티아민(비타민 B1), 리보플라빈(비타민B2), 니아신(비타민 B3), 피리독신(비타민 B6), 시아노코발라민(비타민 B12), 아스코르브산(비타민 C), 비타민 D, 비타민E, 비타민 K, 이들의 염 및 이들의 유도체로 이루어진 군에서 선택된 2종 이상 일 수 있으나, 이에 국한되는 것은 아니다.
본 발명에서 상기 비타민 A는 레티놀이 비가역적으로 산화된 형태인 레티노산의 형태로 상피 세포 등에서 호르몬과 같이 중요한 성장 인자로서 기능을 할 수 있다.
본 발명에서 상기 비타민 E는 주로 세포막, 적혈구막 그리고 미토콘드리아막에서 유리기 포착제로서 산화적 손상에 대한 보호작용을 하고, 막 속의 인지질과 황을 함유한 단백질과 결합하여 막 안정화에 기여하며, 항산화 작용을 할 수 있다.
본 발명에서 상기 비타민 D는 태양광선에 통해 광합성 되고 체내에 흡수된 칼슘, 뼈와 치아에 축적되고 흉선에서 면역세포 생산에 작용하는 일종의 호르몬이다. 또한, 비타민 D는 식이를 통해 비타민 D2와 비타민 D3의 형태로 섭취가 된다. 비타민 D는 체내에서 세포증식과 분화의 조절작용, 면역기능조절작용, 항암작용을보이지만, 주요기능은 칼슘염과 인삼염의 흡수를 촉진하여 골격형성에 도움을 줄 수 있다.
본 발명에서 상기 비타민 B1 (Thiamin)은 신경계 질환인 각기병을 예방하고 치료하며, 비타민 B2 (Riboflavin)는 비타민 G라고도 하며 각종 대사에 중요한 역할을 하는 조효소 구성 성분으로 세포가 탄수화물, 지방, 단백질로부터 에너지를 공급받는 물질대사에 참여한다. 또한, 비타민 B6는 피리독신(pyridoxine), 피리독살(pyridoxal), 피리독사민(pyridoxamine)의 세가지 화합물로 구성되어 있으며 아미노산 대사, 헴합성, 탄수화물 대사 신경전달물질 합성, 비타민 형성, 면역대사와 지질대사에 관여할 수 있다.
본 발명에서 상기 비타민 C는 항산화 물질로 자유라디칼의 억제제로서 지질과 산화연쇄반응을 차단시켜 간 조직의 지방대사에 영향을 줄 수 있고, 일부 상황에 따라 활성산소를 제거하기도 하고 반대로 오히려 생성하기도 한다. 또한, 면역 체계도 강화시킬 뿐만 아니라 결합조직과 지지조직의 형성에 가담하여 피부와 잇몸의 건강유지에도 기여한다.
본 발명에서는 상기한 비타민 복합체에 유산균을 함께 사용함으로써 항산화 효과와 혈중 지질의 감소에 큰 시너지 효과를 부여할 수 있고, 보다 상세하게는 혈중 LDL(Low density lipoprotein) 콜레스테롤, LDL(Low density lipoprotein)/VLDL(Very-low-density lipoprotein) 콜레스테롤 및 중성지방(triglyceride)의 농도를 현저히 낮춤으로써 비만 및 지질 관련 대사성 질환을 예방 또는 치료할 수 있다.
본 발명에서 상기 비만은 에너지 불균형으로 의하여 과다한 체지방을 가진 상태(condition) 또는 질환(disease)을 의미할 수 있다.
본 발명에서 상기 지질 관련 대사성 질환은 생체 내 과도한 지질 축적에 의해 발생하는 질환을 의미한다. 구체적인 예로, 당뇨, 고지혈증, 지방간, 간염, 간경화, 동맥경화, 고혈압, 심혈관질환 및 상기 질환들이 동시다발적으로 발생하는 대사증후군 등으로 이루어진 군으로부터 선택된 어느 하나일 수 있다.
본 발명의 다른 구현 예에 따르면, 프로바이오틱스(probiotics) 및 비타민 복합체를 유효 성분으로 포함하는 골 질환의 예방 또는 치료용 약학 조성물에 관한 것이다.
본 발명에서 상기 프로바이오틱스의 종류는 특별히 제한하지 않으나, 예를 들면, 스트렙토코커스(Streptococcus) 속, 락토코커스(Lactococcus) 속, 엔테로코커스(Enterococcus) 속, 락토바실러스(Lactobacillus) 속, 페디오코커스(Pediococcus) 속, 류코노스톡(Leuconostoc) 속, 비셀라(Weissella) 속 및 비피도박테리움(Bifidobacterium) 속으로 이루어진 군에서 선택된 하나 이상의 유산균 균주를 포함할 수 있으며, 바람직하게는 엔테로코커스 페슘(Enterococcus

본 발명에서 상기 프로바이오틱스는 조성물의 총 중량에 대하여 3 X 109 내지 6 X 109 CFU/g의 함량으로 포함될 수 있으나, 이에 제한되는 것은 아니다. 프로바이오틱스의 함량이 3 X 109 CFU/g 이하에 해당하는 경우, 포함되는 함량이 적어 항산화 효과를 증가시키는 효과를 얻을 수 없고, 프로바이오틱스의 함량이 6 X 109 CFU/g을 초과하는 경우에는 조성물의 성상이 나쁠 수 있다.
또한, 본 발명에서 상기 비타민 복합체로는 수용성 비타민, 지용성 비타민 또는 이들의 혼합물 중 2종 이상의 비타민 혼합물일 수 있다. 바람직하게는 상기 비타민 복합체는 카로티노이드(예, 베타-카로틴, 제아잔틴, 루테인, 리코펜), 비오틴, 콜린, 이노시톨, 엽산, 판토텐산, 콜린, 비타민 A, 티아민(비타민 B1), 리보플라빈(비타민B2), 니아신(비타민 B3), 피리독신(비타민 B6), 시아노코발라민(비타민 B12), 아스코르브산(비타민 C), 비타민 D, 비타민E, 비타민 K, 이들의 염, 및 이들의 유도체로 이루어진 군에서 선택된 2종 이상 일 수 있으나, 이에 국한되는 것은 아니다.
본 발명에서는 상기한 비타민 복합체에 프로바이오틱스를 함께 사용함으로써 체내로 칼슘 및 철분의 흡수 촉진에 큰 시너지 효과를 부여할 수 있고, 이로써 다양한 골 질환을 예방 또는 치료할 수 있다.
본 발명에서 상기 골 질환은 뼈 내의 파골세포와 조골세포의 불균형에 의해 유발되는 질환으로, 구체적인 예로는 골다공증(osteoporosis), 골연화증(osteomalacia), 치주염(periodontitis), 류마티스 관절염(rheumatoid arthritis) 또는 대사성 골질환 등이 있으나, 바람직하게는 골다공증일 수 있다.
본 발명에서 상기 "골다공증(osteoporosis)"은 골형성과 골흡수의 균형이 파괴되어 골량의 전반적인 감소로 발생하는 대사성 질환으로 뼈의 무기질과 단백질이 줄어들어 골조직이 엉성해지고 뼈가 물러지는 현상이며, 크게 폐경 후 골다공증과 노인성 골다공증으로 나눌 수 있다. 폐경 후 골다공증의 경우 폐경 후 에스트로겐 결핍으로 인해 골 흡수가 증가되고, 칼슘의 흡수 및 비타민 D의 생성이 감소함에 따라 골밀도 및 골강도의 감소가 급격히 진행되어 발생하며, 노인성 골다공증은 노화에 따른 골량감소 및 칼슘 흡수 감소가 주된 원인으로 노령인구에서 성별에 구별 없이 발생한다.
한편, 본 발명에서, "예방"은 본 발명의 약학 조성물을 이용하여 비만, 지질 관련 대사성 질환 또는 골 질환의 증상을 차단하거나, 그 증상의 억제 또는 지연시키는 모든 행위라면 제한없이 포함할 수 있다.
또한, 본 발명에서, "치료"는 본 발명의 약학 조성물을 이용하여 비만, 지질 관련 대사성 질환 또는 골 질환의 증상이 호전되거나 이롭게 되는 모든 행위라면 제한없이 포함할 수 있다.
본 발명에 있어서, 상기 약학 조성물은 캡슐, 정제, 과립, 주사제, 연고제, 분말 또는 음료 형태임을 특징으로 할 수 있으며, 상기 약학 조성물은 인간을 대상으로 하는 것을 특징으로 할 수 있다.
본 발명의 약학 조성물은 이들로 한정되는 것은 아니지만, 각각 통상의 방법에 따라 산제, 과립제, 캡슐, 정제, 수성 현탁액 등의 경구형 제형, 외용제, 좌제 및 멸균 주사용액의 형태로 제형화하여 사용될 수 있다. 본 발명의 약학 조성물은 약제적으로 허용 가능한 담체를 포함할 수 있다. 약제학적으로 허용되는 담체는 경구 투여 시에는 결합제, 활탁제, 붕해제, 부형제, 가용화제, 분산제, 안정화제, 현탁화제, 색소, 향료 등을 사용할 수 있으며, 주사제의 경우에는 완충제, 보존제, 무통화제, 가용화제, 등장제, 안정화제 등을 혼합하여 사용할 수 있으며, 국소투여용의 경우에는 기제, 부형제, 윤활제, 보존제 등을 사용할 수 있다. 본 발명의 약학 조성물의 제형은 상술한 바와 같은 약제학적으로 허용되는 담체와 혼합하여 다양하게 제조될 수 있다. 예를 들어, 경구 투여시에는 정제, 트로키, 캡슐, 엘릭서(elixir), 서스펜션, 시럽, 웨이퍼 등의 형태로 제조할 수 있으며, 주사제의 경우에는 단위 투약 앰플 또는 다수회 투약 형태로 제조할 수 있다. 기타, 용액, 현탁액, 정제, 캡슐, 서방형 제제 등으로 제형화할 수 있다.
한편, 제제화에 적합한 담체, 부형제 및 희석제의 예로는, 락토즈, 덱스트로즈, 수크로즈, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말디톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로즈, 폴리비닐피롤리돈, 물, 메틸하이드록시벤조에이트, 프로필하이드록시벤조에이트, 탈크, 마그네슘 스테아레이트 또는 광물유 등이 사용될 수 있다. 또한, 충진제, 항응집제, 윤활제, 습윤제, 향료, 유화제, 방부제 등을 추가로 포함할 수 있다.
본 발명에 따른 약학 조성물의 투여 경로는 이들로 한정되는 것은 아니지만 구강, 정맥내, 근육내, 동맥내, 골수내, 경막내, 심장내, 경피, 피하, 복강내, 비강내, 장관, 국소, 설하 또는 직장이 포함된다. 경구 또는 비경구 투하가 바람직하다.
본 발명에서 사용된 용어 "비경구"는 피하, 피내, 정맥내, 근육내, 관절내, 활액낭내, 흉골내, 경막내, 병소내 및 두개골내 주사 또는 주입기술을 포함한다. 본 발명의 약학 조성물은 또한 직장 투여를 위한 좌제의 형태로 투여될 수 있다.
본 발명의 약학 조성물은 사용된 특정 화합물의 활성, 연령, 체중, 일반적인 건강, 성별, 정식, 투여시간, 투여경로, 배출율, 약물 배합 및 예방 또는 치료될 특정 질환의 중증을 포함한 여러 요인에 따라 다양하게 변할 수 있고, 상기 약학 조성물의 투여량은 환자의 상태, 체중, 질병의 정도, 약물 형태, 투여경로 및 기간에 따라 다르지만 당업자에 의해 적절하게 선택될 수 있고, 1일 0.0001 내지 50mg/kg 또는 0.001 내지 50mg/kg으로 투여할 수 있다. 투여는 하루에 한번 투여할 수도 있고, 수회 나누어 투여할 수도 있다. 상기 투여량은 어떠한 면으로든 본 발명의 범위를 한정하는 것은 아니다. 본 발명에 따른 약학 조성물은 환제, 당의정, 캡슐, 액제, 겔, 시럽, 슬러리, 현탁제로 제형화 될 수 있다.
본 발명의 또 다른 구현 예에 따르면, 프로바이오틱스 및 비타민 복합체를 유효 성분으로 포함하는 비만 및 지질 관련 대사성 질환의 예방 또는 개선용 식품 조성물에 관한 것이다.
본 발명의 식품 조성물에서 상기 프로바이오틱스, 비타민 복합체, 비만 및 지질 관련 대사성 질환에 관한 내용은 상기 약학 조성물에서 기재한 바와 중복되어, 이하 구체적인 기재를 생략한다.
본 발명의 또 다른 구현 예에 따르면, 프로바이오틱스 및 비타민 복합체를 유효 성분으로 포함하는 골 질환의 예방 또는 개선용 식품 조성물에 관한 것이다.
본 발명의 식품 조성물에서 상기 프로바이오틱스, 비타민 복합체 및 골 질환에 관한 내용은 상기 약학 조성물에서 기재한 바와 중복되어, 이하 구체적인 기재를 생략한다.
한편, 본 발명에서, "개선"은 본 발명의 약학 조성물을 이용하여 비만, 지질 관련 대사성 질환 및 골 질환의 증상이 호전 또는 이롭게 변경되는 모든 행위라면 제한없이 포함할 수 있다.
본 발명의 조성물을 유효성분으로 포함하는 식품 조성물은 각종 식품류, 예를 들어, 음료, 껌, 차, 비타민 복합제, 분말, 과립, 정제, 캡슐, 과자, 떡, 빵 등의 형태로 제조될 수 있다. 본 발명의 식품 조성물은 독성 및 부작용이 거의 없는 식물추출물로 구성된 것이므로 예방 목적으로 장기간 복용 시에도 안심하고 사용할 수 있다.
본 발명의 조성물이 식품 조성물에 포함될 때 그 양은 전체 중량의 0.1 내지 50%의 비율로 첨가할 수 있다.
여기서, 상기 식품 조성물이 음료 형태로 제조되는 경우 지시된 비율로 상기 식품 조성물을 함유하는 것 외에 특별한 제한점은 없으며 통상의 음료와 같이 여러가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 즉, 천연 탄수화물로서 포도당 등의 모노사카라이드, 과당 등의 디사카라이드, 슈크로스 등의 및 폴리사카라이드, 덱스트린, 시클로덱스트린 등과 같은 통상적인 당 및 자일리톨, 소르비톨, 에리트리톨 등의 당알콜 등을 포함할 수 있다. 상기 향미제로서는 천연 향미제(타우마틴, 스테비아 추출물(예를 들어 레바우디오시드 A, 글리시르히진등) 및 합성 향미제(사카린, 아스파르탐 등) 등을 들 수 있다.
그 외 본 발명의 식품 조성물은 여러 가지 영양제, 비타민, 광물(전해질), 합성 풍미제 및 천연 풍미제 등의 풍미제, 착색제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알콜, 탄산 음료에 사용되는 탄산화제 등을 함유할 수 있다.
이러한 성분은 독립적으로 또는 조합하여 사용할 수 있다. 이러한 첨가제의 비율은 그렇게 중요하진 않지만 본 발명의 조성물 100 중량부 당 0.1 내지 약 50 중량부의 범위에서 선택되는 것이 일반적이다.
또한, 본 발명의 식품 조성물은 밀크 또는 유장계 발효 유제품과 같은 식품 또는 영양 제품으로서 또는 식품 보조제 또는 건강기능성 식품으로 섭취될 수 있다. 구체적으로, 유제품, 음료수, 주스, 수프 또는 어린이용 식품과 같은 식품을 예로들수 있지만, 이에 제한되는 것은 아니다. 상기 유제품은 다양한 지방 함량을 갖는 임의의 액체 또는 반고체 밀크 또는 유장계 제품을 의미한다. 상기 유제품은 예를 들어 우유, 염소의 젖, 양의 젖, 크림, 전지유(full-fat milk), 전유(whole milk), 저지방유(low-fat milk) 또는 탈지유(skim milk), 한외 여과유, 이중 여과유(diafilteredmilk), 미세 여과유(microfiltered milk), 임의의 가공을 통해 분유 또는 유장으로부터 얻은 재조합유, 가공 제품, 예를 들어, 요거트, 응유, 산유, 산전유(sour whole milk), 버터유, 기타 발효유 제품, 예를 들어 빌리(viili), 스낵바의 필링(filling of snack bars)등 일 수 있다. 또 다른 중요한 그룹은 유음료(milk beverage), 예를 들어, 유장 음료, 발효유, 농축유, 유아 및 아기용 밀크, 아이스크림, 단것과 같은 유함유 식품이 포함될 수 있다.
본 발명의 또 다른 구현 예에 따르면, 치료를 필요로 하는 대상에게 프로바이오틱스 및 비타민 복합체를 포함하는 조성물을 투여하는 단계를 포함하는, 비만 및 지질 관련 대사성 질환의 예방 또는 치료 방법에 관한 것이다.
본 발명에서 상기 치료를 필요로 하는 대상은 비만 및 지질 관련 대사성 질환이 있거나, 그 증상이 의심되는 개체로, 여기서, 상기 지질 관련 대사성 질환은 생체 내 과도한 지질 축적에 의해 발생하는 질환을 의미한다. 구체적인 예로, 당뇨, 고지혈증, 지방간, 간염, 간경화, 동맥경화, 고혈압, 심혈관 질환 및 상기 질환들이 동시다발적으로 발생하는 대사증후군 등으로 이루어진 군으로부터 선택된 어느 하나일 수 있으나, 이에 제한되지 아니한다.
본 발명에서는 상기한 프로바이오틱스와 비타민 복합체를 동시 투여함으로써 상기한 개체의 항산화 및 혈중 지질의 감소에 큰 시너지 효과를 부여하여, 비만 및 지질 관련 대사성 질환을 효과적으로 예방 또는 치료할 수 있다.
본 발명에서 상기 프로바이오틱스와 비타민 복합체에 관한 내용은 상기 약학 조성물에 기재된 바와 중복되어, 이하에서는 그 자세한 기재를 생략한다.
본 발명의 또 다른 구현 예에 따르면, 치료를 필요로 하는 대상에게 프로바이오틱스 및 비타민 복합체를 포함하는 조성물을 투여하는 단계를 포함하는, 골 질환의 예방 또는 치료 방법에 관한 것이다.
본 발명에서 상기 치료를 필요로 하는 대상은 골 질환이 있거나, 그 증상이 의심되는 개체로, 여기서, 상기 골 질환은 뼈 내의 파골세포와 조골세포의 불균형에 의해 유발되는 질환이며, 구체적인 예로는 골다공증(osteoporosis), 골연화증(osteomalacia), 치주염(periodontitis), 류마티스 관절염(rheumatoid arthritis) 또는 대사성 골질환 등이 있으나, 바람직하게는 골다공증일 수 있다.

실험 동물의
준비
프로바이오틱스 및 비타민 복합체의 조합에 의한 골 질환, 비만 및 지질 관련 대사성 질환 등의 효과를 검증하기 위하여, 생후 4주된 흰쥐(male, SD Rat, 새론바이오, 한국)를 구입하였다. 상기 동물은 폴리프로필렌(polypropylene) 케이지에 2마리씩 넣고, 고형사료(pellet type)와 음용수를 자유롭게 섭취하도록 유도하는 순화기간을 1주일 거치도록 하였다. 또한, 사육 환경은 온도 24±2℃, 상대습도 40±20% 및 명암주기 12시간의 조건에서 진행하였다. 상기 순화기간 동안 상기 쥐의 건강을 확인하였고, 그 중 활동성이 떨어지지 않는 쥐를 선별하여 이후 실험에 사용하였다.
고지혈증 관련 지질대사 장애 예방 및 치유 시험을 위해 동물의 식이는 93G based 사료에 2.5% calcium, 10 ppm Fe이 추가된 사료를 기본 식이 조성으로 하여 시험 기간 중 제한 급이를 시행하고, 음용수의 섭취는 자유공급 하였다.

1] 흰쥐의 체중 변화 및 식이 섭취량 변화
상기 실시예 1에서 5주간 실험군 프로바이오틱스 및 비타민 복합체를 투여하였을 때 개체의 안정성 및 표현형(phenotype)을 확인하기 위하여, 상기 실시예 1의 투여를 수행하기 전일부터 투여 시작 후 5주간 상기 흰쥐의 체중 변화 양상을 확인하여, 하기 표 2 및 도 1에 나타내었다. 또한, 3일 간격으로 식이 섭취량을 측정한 후 체중 증가량을 상기 식이 섭취량으로 나누어 식이 효율(food efficiency ratio, FER)을 계산한 뒤 그 결과는 하기 표 3 및 도 2에 나타내었다.

상기 표 2 및 3과, 도 1 및 2에서 보는 바와 같이, 비타민 복합체 또는 프로바이오틱스와 비타민 복합체를 병용 투여한 경우 프로바이오틱스만 투여한 경우에 비하여 체중이 약간 감소하였으나, 식이효율 상 큰 차이가 없는 것을 확인할 수 있었다. 따라서, 본 발명에 따른 프로바이오틱스 및 비타민 복합체를 동시에 투여한 경우에도 이들을 각각 투여한 경우와 동등하게 개체에 독성이 없는 것을 알 수 있다.

실험예
2] 흰쥐의 혈청 내 생화학적 성분 변화
상기 실시예 1에서 쥐에게 5주간 실험군 프로바이오틱스 및 비타민 복합체를 투여한 뒤 14시간 동안 절식시킨 후, CO2 마취 하에 복부대동맥에서 혈액을 채취하였다. 채혈된 혈액은 실온에서 30분 동안 방치한 후 3,000 RPM에서 30분 동안 원심분리하여 혈청을 분리하고 -80℃에 보관하였다.
골 질환 치료 효과를 확인하기 위하여 자동 생화학 분석기(Modular analytics, Roche, Germany)를 이용하여 분리한 혈청 내 Fe 및 Ca 성분의 함량 변화를 측정하여 그 결과를 하기 표 4 및 5와, 도 3 및 4에 나타내었다. 또한, 지질 성분의 감소 효과를 확인하기 위하여, 혈청 내 콜레스테롤(total cholesterol, CHOL)의 함량의 변화를 측정하여 그 결과를 하기 표 6 및 도 5에 나타내었다.

상기 표 4 및 5와, 도 3 및 4에서 보는 바와 같이, 본 발명에 따른 프로바이오틱스 및 비타민 복합체를 동시에 투여한 경우 이들을 각각 투여한 경우와 비교하여 혈청 내 칼슘 및 철분 농도가 현저히 증가한 것을 볼 수 있다.
또한, 상기 표 6 및 도 5에서 보는 바와 같이, 본 발명에 따른 프로바이오틱스 및 비타민 복합체를 동시에 투여한 경우 이들을 각각 투여한 경우와 비교하여 혈중 콜레스테롤의 함량이 현저히 감소한 것을 볼 수 있다.
따라서, 본 발명에서 프로바이오틱스 및 비타민 복합체를 함께 사용하는 경우 이들을 단독으로 사용한 경우에 비하여 체내 칼슘 및 철분 흡수 촉진 및 혈중 지질 감소에 예측하기 어려운 시너지 효과를 부여함을 알 수 있다.

실험예
3] 흰쥐의 혈청 내 비타민 복합체 흡수율 확인
혈청 내 비타민 복합체의 흡수율을 확인하기 위하여, 상기 실시예 1에서 5주간 실험군 프로바이오틱스 및 비타민복합체를 투여한 뒤 14시간 동안 절식시키고 이산화탄소 마취 하에 복부대동맥에서 혈액을 채취하였다. 채혈된 혈액은 실온에서 30분간 방치한 후 3,000RPM에서 30분간 원심분리하여 혈청을 분리하고, -80℃에서 보관하였다.
또한, 혈액 내 비타민 복합체의 흡수율을 확인하기 위해, 비타민 A, C, E의 수치를 HPLC(high performance liquid chromatography) 분석법을 이용하여 혈청 내 존재하는 상기 비타민의 양을 각각 측정하여 그 결과를 하기 표 7 내지 9 및 도 6 내지 8에 나타내었다.

상기 표 7 내지 9 및 도 6 내지 8에서 보는 바와 같이, 본 발명에 따른 프로바이오틱스 및 비타민 복합체를 동시에 투여하는 경우에는 혈청 내 비타민의 농도가 이들을 각각 투여한 경우보다 현저히 증가한 것을 확인할 수 있었다.
따라서, 본 발명에서 프로바이오틱스 및 비타민 복합체를 함께 사용하는 경우 이들을 단독으로 사용한 경우에 비하여 체내로 비타민의 흡수 촉진에 예측하기 어려운 시너지 효과를 부여함을 알 수 있다.

실험예
4] 흰쥐의 장 내 유산균 분포 변화
본 발명에 따른 프로바이오틱스 및 비타민 복합체의 투여로 인한 장내 미생물의 변화를 확인하기 위하여, 상기 실시예 1의 G1 내지 G4 실험군의 장내 미생물의 유산균 유전자 발현을 측정하였다.
상기 실시예 1의 G1 내지 G4에서 경구 투여 전 및 5주 후 분리한 분변을MP bio stool 키트를 이용하여, 제조사가 제공한 프로토콜에 의하여 DNA를 추출하였다.상기 추출된 DNA를 이용하여 L. 람노수스(L.
rhamnosus) 및 E. 페슘(E.facecium)에 특이적인 프라이머를 이용하여 실시간 중합효소연쇄반응(Real time PCR)을 실시하여,그 결과를 하기 표 10 및 11과, 도 9 및 10에 나타내었다.

상기 표 10 및 11과, 도 9 및 10에서 보는 바와 같이, 본 발명에 따른 프로바이오틱스 및 비타민 복합체를 동시에 투여한 경우 이들을 각각 투여한 경우와 비교하여 장내 유산균 분포가 현저히 증가한 것을 볼 수 있다.
상기 결과를 이상에서 본 발명에 대하여 상세하게 설명하였지만 본 발명의 권리범위는 이에 한정되는 것은 아니고, 청구범위에 기재된 본 발명의 기술적 사상을 벗어나지 않는 범위 내에서 다양한 수정 및 변형이 가능하다는 것은 당 기술분야의 통상의 지식을 가진 자에게는 자명할 것이다.
본 발명은 골 질환, 비만 및 지질 관련 대사성 질환의 예방, 개선 또는 치료용 기능성 조성물에 관한 것이다.
Claims (16)
- 프로바이오틱스(probiotics) 및 비타민 복합체를 유효 성분으로 포함하는 비만 및 지질 관련 대사성 질환의 예방 또는 치료용 약학 조성물.
- 제1항에 있어서,상기 프로바이오틱스는 스트렙토코커스(Streptococcus) 속, 락토코커스(Lactococcus) 속, 엔테로코커스(Enterococcus) 속, 락토바실러스(Lactobacillus) 속, 페디오코커스(Pediococcus) 속, 류코노스톡(Leuconostoc) 속, 비셀라(Weissella) 속 및 비피도박테리움(Bifidobacterium) 속으로 이루어진 군에서 선택된 하나 이상의 유산균 균주를 포함하는, 약학 조성물.
- 제1항에 있어서,상기 프로바이오틱스는 엔테로코커스 페슘(Enterococcus faecium), 락토바실러스 람노수스(Lactobacillus rhamnosus) 또는 이들의 혼합물을 포함하는, 약학 조성물.
- 제1항에 있어서,상기 프로바이오틱스는 상기 조성물의 총 중량에 대하여 3 X 109 내지 6 X 109 CFU/g의 함량으로 포함되는, 약학 조성물.
- 제1항에 있어서,상기 비타민 복합체는 카로티노이드, 비오틴, 콜린, 이노시톨, 엽산, 판토텐산, 콜린, 비타민 A, 비타민 B1, 비타민 B2, 비타민 B3, 비타민 B6, 비타민 B12, 비타민 C, 비타민 D, 비타민 E, 비타민 K, 이들의 염, 및 이들의 유도체로 이루어진 군에서 선택된 2종 이상인, 약학 조성물.
- 제1항에 있어서,상기 지질 관련 대사성 질환은 당뇨, 고지혈증, 지방간, 간염, 간경화, 동맥경화, 고혈압, 심혈관질환 및 상기 질환들이 동시다발적으로 발생하는 대사증후군으로 이루어진 군에서 선택된 어느 하나인, 약학 조성물.
- 프로바이오틱스(probiotics)및 비타민 복합체를 유효 성분으로 포함하는 골 질환의 예방 또는 치료용 약학 조성물.
- 제7항에 있어서,상기 프로바이오틱스는 스트렙토코커스(Streptococcus) 속, 락토코커스(Lactococcus) 속, 엔테로코커스(Enterococcus) 속, 락토바실러스(Lactobacillus) 속, 페디오코커스(Pediococcus) 속, 류코노스톡(Leuconostoc) 속, 비셀라(Weissella) 속 및 비피도박테리움(Bifidobacterium) 속으로 이루어진 군에서 선택된 하나 이상의 유산균 균주를 포함하는, 약학 조성물.
- 제7항에 있어서,상기 프로바이오틱스는 엔테로코커스 페슘(Enterococcus faecium), 락토바실러스 람노수스(Lactobacillus rhamnosus) 또는 이들의 혼합물을 포함하는, 약학 조성물.

- 제7항에 있어서,상기 비타민 복합체는 카로티노이드, 비오틴, 콜린, 이노시톨, 엽산, 판토텐산, 콜린, 비타민 A, 비타민 B1, 비타민 B2, 비타민 B3, 비타민 B6, 비타민 B12, 비타민 C, 비타민 D, 비타민E, 비타민 K, 이들의 염 및 이들의 유도체로 이루어진 군에서 선택된 2종 이상인, 약학 조성물.
- 제7항에 있어서,상기 골 질환은 골다공증(osteoporosis), 골연화증(osteomalacia), 치주염(periodontitis), 류마티스 관절염(rheumatoid arthritis) 및 대사성 골질환으로 이루어진 군에서 선택된 어느 하나인, 약학 조성물.
- 프로바이오틱스(probiotics) 및 비타민 복합체를 유효 성분으로 포함하는 비만 및 지질 관련 대사성 질환의 예방 또는 개선용 식품 조성물.
- 프로바이오틱스(probiotics) 및 비타민 복합체를 유효 성분으로 포함하는 골 질환의 예방 또는 개선용 식품 조성물.
- 치료를 필요로 하는 대상에게 프로바이오틱스 및 비타민 복합체를 포함하는 조성물을 투여하는 단계를 포함하는, 비만 및 지질 관련 대사성 질환의 예방 또는 치료 방법.
- 치료를 필요로 하는 대상에게 프로바이오틱스 및 비타민 복합체를 포함하는 조성물을 투여하는 단계를 포함하는, 골 질환의 예방 또는 치료 방법.
