Abstract
[課題]本発明は、低い製造コストで、有効成分を第一胃における放出や分解から保護し、目的とする臓器で放出挙動を増大させるという目的を同時に達成するルーメンバイパス製剤の製造方法とこれにより得られた顆粒剤を提供する。 [解決手段]本発明によれば、例えば、ルーメンバイパス製剤を製造する方法であって、ルーメンバイパス製剤用の被覆剤と、ルーメンをバイパスさせる栄養素との溶融物が含まれている、少なくとも1つの噴射口を有するダイヘッドまたは前記溶融物に振動を加え、これにより、溶融物を噴射口から噴射させることを含む、方法が提供される。
Description
本発明は、低い製造コストで、有効成分を第一胃における放出や分解から保護し、目的とする臓器で放出挙動を増大させるという目的を同時に達成するルーメンバイパス製剤の製造方法とこれにより得られたルーメンバイパス製剤に関する。
ルーメンバイパス製剤は、反芻動物において栄養成分等の有効成分を第一胃における放出や分解から保護し、これにより反芻動物による有効成分の吸収を改善するために開発が進められ、製品化されているものも存在する。ルーメンバイパス製剤の形状には、顆粒剤やペレット状の形状を有するものが存在する。だが、ペレット状等の肥大化した剤形においては、第一胃での有効成分の放出や分解が保護されやすいものの、咀嚼による口腔内での崩壊や、有効成分の放出阻害を招きやすく、その利用効率が低下するという問題が存在する。
特許文献1は、振動するノズルから有効成分と賦形剤とを含む混合物の溶融物を放出し、塔内を落下させて塔の底部において持続性もしくは迅速な放出挙動を示す顆粒を得る方法が記載されている。しかし、この方法によって得られた顆粒は一定の孔容積を有し、有効成分が迅速もしくは持続的に放出するため、反芻動物の第一胃における有効成分の放出や分解から保護するという課題を解決するには困難が伴う。
本発明は、低い製造コストで、有効成分を第一胃における放出や分解から保護し、目的とする臓器で放出挙動を増大させるという目的を同時に達成するルーメンバイパス製剤の製造方法とこれにより得られたルーメンバイパス製剤を提供する。
本発明者らは、ルーメンバイパス製剤の製造において、ダイヘッドに充填された前記製剤成分の溶融物に振動を与えつつ溶融物をダイヘッドから噴射させることにより、粒径が均一で有り、特定粒径範囲の顆粒が多く得られることを明らかにした。本発明者らはまた、特に溶融物に直接的に振動子を接触させて振動を与えること、および、与える振動の振動数を1,000Hz~10,000Hzとすることにより、粒径の均一度が向上することを明らかにした。本発明者らはさらに、得られた製剤がルーメンバイパス製剤として優れた溶出特性および安定性を有していることを明らかにした。本発明はこのような知見に基づく発明である。
すなわち、本発明によれば以下の発明が提供される。
(1)ルーメンバイパス製剤を製造する方法であって、
ルーメンバイパス製剤用の被覆剤と、ルーメンをバイパスさせる栄養素との溶融物が含まれている、少なくとも1つの噴射口を有するダイヘッドまたはダイヘッドに含まれる前記溶融物に振動を加え、これにより、溶融物を噴射口から噴射させることを含む、方法。
(2)振動が1000Hz~10000Hzの範囲の振動である、上記(1)に記載の方法。
(3)振動が3000Hz~7000Hzの範囲の振動である、上記(1)または(2)に記載の方法。
(4)振動が、ダイヘッド内腔に露出し溶融物に直接接触する振動子を介して溶融物に与えられる、上記(1)~(3)のいずれかに記載の方法。
(5)被覆剤が硬化油である、上記(1)~(4)のいずれかに記載の方法。
(6)硬化油がパーム硬化油および菜種硬化油からなる群から選択される1以上の硬化油である、上記(5)に記載の方法。

本発明によれば、粒径700μm以上の粒径を有するルーメンバイパス製剤が低コストで効率よく得られ、有利である。本発明によればまた、得られたルーメンバイパス製剤は、第一胃における溶解に対して耐性を有し、また、長期保存に対する安定性が高く、有利である。
本明細書で用いられる「ルーメンバイパス製剤」とは、ルーメン(すなわち、第一胃)をバイパスさせる成分と混合されたルーメンバイパス用の担体を含む顆粒剤であるか、またはルーメンをバイパスさせる成分とその周囲に担体(または被覆層)を有する顆粒剤であり、これにより前記成分が反芻動物の第一胃で溶出したり分解されたりするのを防ぐことができる製剤をいう。本明細書では「ルーメンバイパス用の担体」は、ルーメンにおいて有用成分を保護することに用いられる担体(単に「保護剤」または「ルーメンバイパス用の保護剤」ということもある)をいい、「被覆剤」は、前記担体であって、有用成分を被覆するものをいう。本明細書では、「ルーメンバイパス用の担体」という用語は、「保護剤」または「ルーメンバイパス用の保護剤」と同義であり、相互互換的に用いられ得る。
本明細書で用いられる「粒径」は、日本薬局方の粒度測定法に基づいて測定される粒径である。本明細書では、他に断りの無い限り、「平均粒径」は数平均の粒径を意味する。
本発明では、ルーメンバイパス製剤を製造する方法であって、
ルーメンバイパス用の担体と、ルーメンをバイパスさせる成分との溶融物が含まれている、少なくとも1つの噴射口を有するダイヘッドまたは前記溶融物に振動を加え、これにより、溶融物を噴射口から噴射させることを含む方法が提供される。

ルーメンバイパス用の担体は、溶融状態においてスプレー法で用いることができる担体を用いることができ、当業者であれば適宜選択して用いることができる。本発明で用いることができる担体の例は以下の通りである。ルーメンバイパス用の担体は、融点が40℃以上のものを用いることができる。ルーメンバイパス用の担体はまた、融点がルーメンをバイパスさせる成分が分解される温度よりも低い温度とすることができ、例えば、80℃以下とすることができる。従って、ルーメンバイパス用の担体は、融点が、例えば、40℃~90℃である担体とすることができ、例えば、50℃~70℃、60℃~80℃、40℃~70℃、および40℃~60℃から選択される温度範囲の融点を有する担体とすることができる。
このようなルーメンバイパス用の担体の具体的な例としては、特に限定されないが、ロウ、脂肪酸、脂肪酸塩、グリセロリン脂質および硬化油が挙げられる。硬化油としては、ヒマシ硬化油、パーム硬化油および菜種硬化油が挙げられる。グリセロリン脂質としては、レシチンが挙げられる。
本明細書で、「溶融物」と述べた場合には、ルーメンをバイパスさせる成分が担体に完全に溶解したものだけではなく、ルーメンをバイパスさせる成分が顆粒形態で担体の溶融物中に分散しているものも含んだ意味で用いられる。通常は、ルーメンバイパス用の担体は、脂溶性であるので、脂溶性成分を有効成分として用いる場合には、有効成分は担体に溶解させることができ、水溶性成分を有効成分として用いる場合には、水溶成分を含有する固形粒子を担体に分散させることができる。
このようなルーメンバイパス用の担体の具体的な例としては、特に限定されないが、ロウ、脂肪酸、脂肪酸塩、グリセロリン脂質および硬化油が挙げられる。硬化油としては、ヒマシ硬化油、パーム硬化油および菜種硬化油が挙げられる。グリセロリン脂質としては、レシチンが挙げられる。
本明細書で、「溶融物」と述べた場合には、ルーメンをバイパスさせる成分が担体に完全に溶解したものだけではなく、ルーメンをバイパスさせる成分が顆粒形態で担体の溶融物中に分散しているものも含んだ意味で用いられる。通常は、ルーメンバイパス用の担体は、脂溶性であるので、脂溶性成分を有効成分として用いる場合には、有効成分は担体に溶解させることができ、水溶性成分を有効成分として用いる場合には、水溶成分を含有する固形粒子を担体に分散させることができる。
ルーメンをバイパスさせる成分としては、例えば、栄養素が挙げられる。栄養素をルーメンバイパス製剤により反芻動物に供給することで、例えば、反芻動物の疾病予防や健康維持に役立つ。本発明で用いることができる栄養素としては、例えば、アミノ酸類およびビタミン類並びにこれらの塩が挙げられる。アミノ酸としては、例えば、メチオニンおよびリジンなどのアミノ酸が挙げられる。ビタミンとしては、例えば、ビタミンB1、B2、パントテン酸、葉酸、ニコチン酸、ビタミンC、ビタミンEが挙げられる。ルーメンをバイパスさせる成分は、その他添加剤(例えば、食品添加物)を含んでいてもよい。添加剤の例としては、特に限定されないが、ブドウ糖およびトリメチルグリシン(ベタイン)が挙げられる。
ルーメンをバイパスさせる成分は、粒子状であってよく、溶融物中に分散させることができる。ルーメンをバイパスさせる成分が粒子状の形態で提供される場合、粒子の粒径は、数平均で600μm以下、500μm以下、400μm以下、300μm以下、200μm以下、または100μm以下とすることができる。下記に説明するように、噴出口のホール径よりも小さければよいであろう。また、前記粒子の粒径は、小さいほど得られる顆粒剤の粒径の均質性が高まる。
ダイヘッドは、溶融物の導入口と溶融物の噴射口を備える。本発明の方法では、少なくとも1つの噴射口を有するダイヘッドまたは前記溶融物に振動を加えることにより、ダイヘッドから溶融物を噴射させ、顆粒剤を得ることができる。振動を加える際には、ダイヘッドは全体を振動させることもできるし、ダイヘッドの内腔に振動子を露出させ、露出した振動子と溶融物とを直接的に接触させて振動子を振動させてもよい。
振動は、溶融物が噴射されるために十分な振動を与えることができる。振動は、例えば、周波数において1,000Hz~10,000Hzとすることができ、3,000Hz~7,000Hzとすることができ、5,000~7000Hzとすることができる。振動数は、可変であってもよいが、好ましくは一定であってもよい。
振動は、溶融物が噴射されるために十分な振動を与えることができる。振動は、例えば、周波数において1,000Hz~10,000Hzとすることができ、3,000Hz~7,000Hzとすることができ、5,000~7000Hzとすることができる。振動数は、可変であってもよいが、好ましくは一定であってもよい。
本発明のある態様では、振動を加える際には、ダイヘッドは全体を振動させ、振動数は、1,000Hz~10,000Hzとすることができ、3,000Hz~7,000Hzとすることができ、5,000~7000Hzとすることができる。
本発明のある態様では、振動を加える際には、ダイヘッドの内腔に振動子を露出させ、露出した振動子と溶融物とを直接的に接触させて振動子を振動させ、振動数は、1,000Hz~10,000Hzとすることができ、3,000Hz~7,000Hzとすることができ、5,000~7000Hzとすることができる。
本発明のある態様では、振動を加える際には、ダイヘッドの内腔に振動子を露出させ、露出した振動子と溶融物とを直接的に接触させて振動子を振動させ、振動子を3,000~7,000Hzの範囲の一定の振幅により振動させることができる。
本発明のある態様では、振動は、P-P値が、1mm~10mmとすることができる。ある態様では、振動はP-P値が3mm~7mm、4mm~6mm、または約5mmとすることができる。本発明のある態様では、振動は、正弦波である。
以下、ダイヘッドについて図1を参照して説明する。但し、本発明は、図1の非限定的例示に限定されて解釈されるものではない。
ダイヘッド100は、溶融物の導入口21と内腔30と溶融物の噴射口22を備える。図1では、噴射口を2つ備え、それぞれ22aおよび22bで示されているが、噴射口の数は1つまたは複数とすることができ、例えば8~32個とすることができ、特に限定されない。ダイヘッドは、内腔30を備え、導入口21から導入された溶融物で満たすことができる。また、内腔は、溶融物の噴射口22につながっており、溶融物は内腔30から噴射口22を介して噴射される。
ダイヘッド100は、さらに振動子11を備える振動発生装置10と連結している。振動発生装置10は、振動子11を図1の矢印方向に振動させることができる。振動子11は、ダイヘッドの内腔30に露出しており、振動子11が振動することにより、内腔30に満たされている溶融物に振動を与えることができる。溶融物は、振動を加えることにより噴射口22から噴射され、顆粒40を形成する。
噴射口22は、例えば、ダイホール径を0.5~1.0mm、例えば、0.6~0.8mm、例えば、略0.7mmとすることができる。本発明のある態様では、得たい顆粒剤の粒径の約半分程度の径を有するダイホールが好ましく用いられ得る。
ダイヘッド100は、溶融物の導入口21と内腔30と溶融物の噴射口22を備える。図1では、噴射口を2つ備え、それぞれ22aおよび22bで示されているが、噴射口の数は1つまたは複数とすることができ、例えば8~32個とすることができ、特に限定されない。ダイヘッドは、内腔30を備え、導入口21から導入された溶融物で満たすことができる。また、内腔は、溶融物の噴射口22につながっており、溶融物は内腔30から噴射口22を介して噴射される。
ダイヘッド100は、さらに振動子11を備える振動発生装置10と連結している。振動発生装置10は、振動子11を図1の矢印方向に振動させることができる。振動子11は、ダイヘッドの内腔30に露出しており、振動子11が振動することにより、内腔30に満たされている溶融物に振動を与えることができる。溶融物は、振動を加えることにより噴射口22から噴射され、顆粒40を形成する。
噴射口22は、例えば、ダイホール径を0.5~1.0mm、例えば、0.6~0.8mm、例えば、略0.7mmとすることができる。本発明のある態様では、得たい顆粒剤の粒径の約半分程度の径を有するダイホールが好ましく用いられ得る。
本発明において、溶融物に振動を与えることに、上記ダイヘッド100を用いることができる。
本発明の方法は、噴射された溶融物を冷却することをさらに含んでいてもよい。噴射された溶融物は、担体の融点以下の温度に冷却されると固化して顆粒剤となる。冷却することにより効率よく顆粒剤を得ることができる。固化には一定の時間が必要となるので、顆粒剤は、固化させるために十分な落下時間を確保できる高さから、溶融物を冷却された空気中に噴射させ、固化した状態で着地させて回収できる。
ルーメンをバイパスさせる成分とその周囲に担体(または被覆層)を有する固形製剤について、本発明の方法により得られ得るルーメンバイパス製剤の形状の模式図を図2に示す。顆粒剤40aは、バイパスさせる成分の粒子41が被覆剤42によって被覆された略球形状を理想的に有する。図2では、粒子41は、顆粒剤40aに一つだけ含まれているが、2つ以上含まれていてもよい。バイパスさせる成分の粒子41の粒径は、例えば、500μm以下、400μm以下、300μm以下、200μm以下、または100μm以下とすることができ、前記粒径が小さいほど得られる顆粒剤40の形状は球形状に近づく。また、ルーメンをバイパスさせる成分を溶解した担体を有する固形製剤について、本発明の方法により得られ得るルーメンバイパス製剤の形状の模式図を図3に示す。顆粒剤40bは、バイパスさせる成分を溶解した担体45から構成される。
本発明の方法は、ルーメンバイパス用の担体と、ルーメンをバイパスさせる栄養素との溶融物をダイヘッドに導入することをさらに含んでいてもよい。
従って、本発明では、ルーメンバイパス製剤を製造する方法であって、
ルーメンバイパス用の担体と、ルーメンをバイパスさせる成分との溶融物を、少なくとも1つの噴射口を有するダイヘッドに導入することと、
ダイヘッド内腔に充填された前記溶融物に振動を加え、これにより、溶融物を噴射口から噴射させることと、

本発明のある態様では、本発明の方法により得られたルーメンバイパス製剤は、顆粒剤であり、700μm以上の粒径を有する。本発明のある態様では、本発明のルーメンバイパス製剤は、顆粒剤で有り、1500μm以下の粒径を有する。本発明のある態様では、本発明のルーメンバイパス製剤は、顆粒剤であり、700μm以上1500μm以下の粒径を有する。本発明のある態様では、本発明のルーメンバイパス製剤は、顆粒剤であり、得られた顆粒の40重量/重量%、50重量/重量%以上、60重量/重量%、好ましくは70重量/重量%以上が700μm以上1500μm以下の粒径を有する。本発明のある態様では、本発明のルーメンバイパス製剤は、顆粒剤であり、得られた顆粒の40重量/重量%、50重量/重量%以上、60重量/重量%、好ましくは70重量/重量%以上が1000μm以上1500μm以下の粒径を有する。本発明によれば、本発明のルーメンバイパス製剤は、本発明の方法を用いて製造することができる。
本発明のある態様では、本発明の方法により得られたルーメンバイパス製剤が提供される。本発明のある態様では、本発明の方法により得られたルーメンバイパス製剤は、5μL/g以上、10μL/g以上、20μL/g以上、30μL/g以上、40μL/g以上、50μL/g以上、60μL/g以上、70μL/g以上、80μL/g以上、90μL/g以上、または100μL/g以上の孔容積を有し得る。
本発明のある態様では、本発明の方法により得られたルーメンバイパス製剤が提供される。本発明のある態様では、本発明の方法により得られたルーメンバイパス製剤は、5μL/g以上、10μL/g以上、20μL/g以上、30μL/g以上、40μL/g以上、50μL/g以上、60μL/g以上、70μL/g以上、80μL/g以上、90μL/g以上、または100μL/g以上の孔容積を有し得る。
得られた製剤の安定性は、例えば、得られた製剤を40℃、75%RHの条件下で、例えば、2ヶ月間保管し、保管後の有効成分の含有量を定量することにより評価することができる。
得られた製剤からの含有成分の溶出は、疑似第一胃液中で例えば40℃で10~20時間(例えば、16時間)攪拌し、溶出した成分量を分析することにより評価することができる。分析は、特に限定されないが例えば、高速液体クロマトグラフィーにより行うことができる。疑似第一胃液としては、例えば、リン酸水素二ナトリウム12水和物(Na2HPO4・12H2O)6.3gとリン酸二水素カリウム(KH2PO4)6.7g/Lを含む水溶液900mL(pH6.4)を用いることができる。
以下、具体例により発明を説明するが、発明は、以下の実施例に限定されるものではない。
実施例1:ルーメンバイパスビタミンD

製剤の調製および得られた製剤の分析
実施例1-1:ルーメンバイパスビタミンD 3 製剤の調製およびその粒度分布の測定
本実施例では、ビタミンD3製剤を調製し、その粒度分布を示す。

パーム硬化油100kgを72℃で溶融した。この溶融液に大豆油に溶解させたビタミンD3を6.25g添加し均一に攪拌した。得られた溶液をダイホール径0.7mmを有する振動造粒装置に送液し、1000~10000Hzの振動を与え冷気中に噴射した。凝固したルーメンバイパスビタミンD3については、日本薬局方の粒度測定法(ふるい分け法)に基づき粒度分布を測定した。振動は、p-p値が5mmの正弦波とした。
上記において、本実施例で用いた振動造粒装置100は、図1に示される通りであった。図1に示されるように本実施例で用いた振動造粒装置100は以下の構成を備えた。振動造粒装置100は、ダイヘッド20と振動発生装置10と、振動を溶液に直接伝達する振動伝達部11を備えた。ダイヘッド20には、分散溶液の溶融物30の導入口21と、溶融物30を噴射する噴射口22(図には噴射口22aと22bの2つが描かれているが、噴射口は2つに限られない)を有した。本実施例では、振動造粒装置100を以下の様に作動させた。すなわち、上記分散溶液の溶融物30は、振動造粒装置の溶融物導入口21を介してダイヘッド20に導入された。ダイヘッド20を溶融物30で充填した後に、ダイヘッド20に溶融物30を送液しながら、振動発生装置10を作動させて振動伝達部11を図1の矢印に示されるように上下に振動させ、この振動を利用して溶融物20を噴射口22から噴射させた。噴射させた溶融物31は、冷気中に放出され、冷やされて固化した。
3000Hzの振動を与えて得られた顆粒を観察したところ、図2に示されるように、通常のスプレー法では、粒径が不均一であり、かつ、粒径が非常に小さいのに対して、本願実施例の方法では、粒径が均一であり、かつ、粒径が1000μm以上のものが大半を占めた。
また、表1に示されるように、全ての周波数において振動を与えて噴射させて作製した顆粒は、粒径が1000μm以上であり、特に3000Hz~10000Hzの振動を与えた場合には、1000μm~1519μmの粒径を有する顆粒が極めて効率よく得られた。
実施例1-2:得られた顆粒の構造的分析
得られた顆粒から日本薬局方のふるい分け法に従って1000μm~1519μmの粒径を有する顆粒を得て更なる詳細の分析を行った。本実施例では、特に、細孔サイズの分布について分析した。

細孔サイズは、水銀ポシロメータを用いて水銀圧入法により測定した。測定は、Auto Pore III (Micrometrics社製)を用いて行った。上記で得られた1000μm~1519μmの粒径を有する顆粒を脱気して、水銀を顆粒中に圧入し、顆粒中に侵入した水銀量とそのときに加えた圧力との関係を調べた。細孔サイズはWashburnの式:

図5に示されるように、本実施例で得られた顆粒を分析すると、顆粒中に0.04μm~0.3μmの細孔が頻度高く観察された。なお、積算細孔容積について、図5の「a」の領域で示される細孔直径は、顆粒内部の細孔ではなく、異なる顆粒間の距離を示すものであると考えられた。図5の「b」の領域で示される細孔直径を示す細孔はほとんど観察されなかった。図5の「c」の領域で示される細孔直径を有する細孔は多く観察された。図5の「d」の領域は、水銀圧入法による測定誤差の大きい領域であり、細孔の存在を示唆する根拠とはならない。
図5の「c」の領域に含まれる細孔のデータから「b」の領域をバックグラウンドとして差し引いて、「c」に含まれるサイズを有する細孔について、顆粒中の孔の容積を計算すると、0.0388mL/g(38.8μL/g)程度の孔容積を有することが分かった。
実施例1-3:得られた顆粒の溶出試験
上記実施例1-1で得られた顆粒には、1000μm以上の粒径を有する顆粒が高い割合で含まれていた。粒径の大きい顆粒は、ルーメンでの溶出耐性が高いと考えられるので、本実施例では、疑似第一胃液を用いて、実施例1-1で得られた顆粒の耐溶出性を検討した。具体的には以下の手順で耐溶出性を確認した。
上記実施例1-1で得られた顆粒には、1000μm以上の粒径を有する顆粒が高い割合で含まれていた。粒径の大きい顆粒は、ルーメンでの溶出耐性が高いと考えられるので、本実施例では、疑似第一胃液を用いて、実施例1-1で得られた顆粒の耐溶出性を検討した。具体的には以下の手順で耐溶出性を確認した。
パーム硬化油100kgを72℃で溶融した。この溶融液に大豆油に溶解させたビタミンD3を6.25g添加し均一に攪拌した。得られた溶液をダイホール径0.7mmを有する振動造粒装置に送液し、1000~10000Hzの振動を与え冷気中に噴射した。凝固したルーメンバイパスビタミンD3については、1000μm~1519μmの粒径を有する顆粒を分取し溶出試験を行った。溶出試験では、具体的には、疑似第一胃液としては、リン酸水素二ナトリウム12水和物(Na2HPO4・12H2O)6.3gとリン酸二水素カリウム(KH2PO4)6.7g/Lを含む水溶液(pH6.4)900mLを用い、1000μm~1519μmの粒径を有する顆粒を上記疑似第一胃液に浸漬し、40℃で16時間攪拌した。攪拌後、溶出した有効成分溶出したビタミンD3はElabscience Biotechnology社製VD3 ELISA キットを用いて定量した。なお、本実施例における検出限界は、含有量の2.37%であった。
その結果、1000Hz、3000Hz、5000Hz、7000Hzおよび10000Hzのいずれの振動を与えて得た顆粒についても、疑似第一胃液中での溶出量は、検出限界以下であり、溶出を検出することができなかった。
このように、本実施例によれば、粒径1000μm以上顆粒が高効率で得られるが、これらの顆粒は、疑似第一胃液中で高い耐溶出性を示し、ルーメンバイパス製剤として好適に用いうることが明らかである。
実施例2:ルーメンバイパスリジン製剤の調製および得られた製剤の分析
実施例2-1:ルーメンバイパスリジン製剤の調製およびその粒度分布の測定
本実施例では、ルーメンバイパスリジン製剤を調製し、その粒度分布を示す。

菜種硬化油54.2kg、ステアリン酸5.0kg、レシチン3.0kgを80℃で溶融混合した。この溶融混合液にリジン塩酸塩37.5kg、プロピオン酸カルシウム0.3kgを含む粒子(粒径:0.3mm)を添加し、均一に分散させた。得られた分散溶液を溶融物としてダイホール径0.7mmを有する振動造粒装置に送液し、3000~7000Hzの範囲の一定の振動数を有する振動を与えることにより、冷気中に噴射した。凝固したルーメンバイパスリジン製剤については、日本薬局方の「1.3.顆粒剤」の粒度測定法(ふるい分け法)に従って粒度分布を測定した。具体的には、様々な細孔サイズを有するふるいを用いて造粒された顆粒をふるい分けし、ふるい分けられた各画分の重量(g)を求めた。
表2に示されるように、すべての振動において、粒径710~1519μmの均一な顆粒が得られ、特に粒径が1000~1519μmの均一な顆粒が多く得られ、5000Hz~7000Hzにおいては、得られた顆粒の大半が1000~1519μmの均一な顆粒であった。
実施例2-2:得られた顆粒の溶出試験
上記実施例2-1において、1000Hz、3000Hz、5000Hzまたは10000Hzの振動により得られた顆粒から日本薬局方のふるい分け法に従って、それぞれ1000μm~1519μmの粒径を有する顆粒を得て更なる詳細の分析を行った。本実施例では、特に、得られた顆粒からのリジンの溶出を下記溶出試験により調べた。
上記実施例2-1において、1000Hz、3000Hz、5000Hzまたは10000Hzの振動により得られた顆粒から日本薬局方のふるい分け法に従って、それぞれ1000μm~1519μmの粒径を有する顆粒を得て更なる詳細の分析を行った。本実施例では、特に、得られた顆粒からのリジンの溶出を下記溶出試験により調べた。
溶出試験では、具体的には、疑似第一胃液としては、リン酸水素二ナトリウム12水和物(Na2HPO4・12H2O)6.3gとリン酸二水素カリウム(KH2PO4)6.7g/Lを含む水溶液(pH6.4)900mLを用いた。上記により得られた1000μm~1519μmの粒径を有する顆粒を上記疑似第一胃液に浸漬し、40℃で16時間攪拌した。攪拌後、溶出した有効成分は、常法により、ニンヒドリン溶液(ニンヒドリン5mg、塩化第二銅二水和物8.5mg、クエン酸24mg、2-メトキシエタノール、375μL/mL)を溶出成分を含む溶液と反応させ、475nmの吸光を測定することにより求めた。結果は、表2-1に示される通りであった。
表2-1に示されるように、実施例2により得られた製剤は、第一胃液に対する含有成分の溶出に抵抗性を示した。このことから、得られた製剤は、ルーメンバイパス製剤として有用であることが明らかとなった。
実施例2-3:得られた顆粒中の細孔の分析 実施例1と同様に得られた顆粒から日本薬局方のふるい分け法に従って1000μm~1519μmの粒径を有する顆粒を得て細孔サイズの分析を行った。結果は図6に示される通りであった。
図6に示される通り、本実施例で得られた顆粒を分析すると、顆粒中に0.04μm~0.3μmの細孔が頻度高く観察された。図6の「c」の領域に含まれる細孔のデータから「b」の領域をバックグラウンドとして差し引いて、「c」に含まれるサイズを有する細孔について、顆粒中の孔の容積を計算すると、0.0182mL/g(18.2μL/g)程度の孔容積を有することが分かった。
実施例3:ルーメンバイパスビタミンB-メチオニン製剤の調整および得られた製剤の分析

本実施例では、ビタミンB-メチオニン製剤を調製し、その粒度分布を示す。

ステアリン酸22.5kg、パーム硬化油10.0kg、菜種硬化油、33.7kg、レシチン3.0kgを80℃で溶融混合した。この溶融混合液に、ニコチン酸2kg、D-パントテン酸カルシウム3.3kg、DL-メチオニンを15kg、ベタインを10kg、無水ケイ酸を0.5kgを含む粒子(粒径0.5mm)を添加し、均一に分散した。実施例1に記載される通り、分散した溶液をダイホール径0.7mmを有する振動造粒装置に送液し、5000~10000Hzの振動を与え冷気中に噴射した。凝固したルーメンバイパスビタミンB-メチオニン製剤については、日本薬局方の粒度測定法(ふるい分け法)に基づき粒度分布を測定した。
表3に示されるように、5000~10000Hzの振動を与えて噴射させて作製した顆粒は、大半が1000~1519μmの粒径を有していた。
このように、実施例1~3では、様々な栄養素を内包させたルーメンバイパス製剤を作製したところ、本発明の方法により、粒度が700μm以上、好ましくは1000~1519μmである顆粒を効率よく得ることができた。
実施例3-2:得られた顆粒の溶出試験
上記実施例3-1において、5000Hz、7000Hzまたは10000Hzの振動により得られた顆粒から日本薬局方のふるい分け法に従って、それぞれ1000μm~1519μmの粒径を有する顆粒を得て更なる詳細の分析を行った。本実施例では、特に、得られた顆粒からのビタミン等の溶出を下記溶出試験により調べた。
上記実施例3-1において、5000Hz、7000Hzまたは10000Hzの振動により得られた顆粒から日本薬局方のふるい分け法に従って、それぞれ1000μm~1519μmの粒径を有する顆粒を得て更なる詳細の分析を行った。本実施例では、特に、得られた顆粒からのビタミン等の溶出を下記溶出試験により調べた。
溶出試験では、疑似第一胃液中でのニコチン酸、メチオニン、およびパントテン酸カルシウムの溶出割合を調べた。具体的には、疑似第一胃液としては、リン酸水素二ナトリウム12水和物(Na2HPO4・12H2O)6.3gとリン酸二水素カリウム(KH2PO4)6.7g/Lを含む水溶液900mLを用いた。上記により得られた1000μm~1519μmの粒径を有する顆粒を上記疑似第一胃液に浸漬し、40℃で16時間攪拌した。攪拌後、溶出した有効成分を高速液体クロマトグラフィー(カラム: wakopak Handy ODS (wako pure chemical Ind.)(Φ 4.6×150 mm))により分析した。各溶出成分量は、常法により、UV吸光度検出器(製品名:UV-970、製造者名:日本分光)を用い210nmの吸光を測定することにより測定した。結果は、表3-1~表3-3に示される通りであった。
表3-1~表3-3に示されるように、実施例3により得られた製剤は、第一胃液に対する含有成分の溶出に抵抗性を示した。このことから、得られた製剤は、ルーメンバイパス製剤として有用であることが明らかとなった。
実施例3-3:得られた顆粒中の細孔の分析
実施例1と同様に得られた顆粒から日本薬局方のふるい分け法に従って1000μm~1519μmの粒径を有する顆粒を得て細孔サイズの分析を行った。結果は図7に示される通りであった。

図7に示される通り、本実施例で得られた顆粒を分析すると、顆粒中に0.04μm~0.3μmの細孔が頻度高く観察された。図7の「c」の領域に含まれる細孔のデータから「b」の領域をバックグラウンドとして差し引いて、「c」に含まれるサイズを有する細孔について、顆粒中の孔の容積を計算すると、0.1514mL/g(151.4μL/g)程度の孔容積を有することが分かった。
上記実施例から、ルーメンをバイパスさせる成分の粒径は小さいほど、得られるルーメンバイパス製剤の粒径がより均質になることが分かる。一方で、ダイホール径0.7mmに対して0.5mmの粒径を有する有効成分の顆粒を用いても、得られるルーメンバイパス製剤の粒径の均質性は保たれた。
実施例4:放出制御型ビタミンD 3 製剤の安定性試験
本実施例では、ルーメンバイパスビタミンD3製剤の安定性を試験した。

放出制御型製剤を販売するためには、長期間保存した際の化学的変化の予測及び流通期間中に起こり得る保存条件の短期的な逸脱の影響を評価しなければならない。そこで、放出制御型ビタミンD3製剤を40℃、75%RHの条件下で2ヶ月間保存し、保存後のビタミンD3の含有量をp-anisaldehyde法を用いて定量した。結果は、表4に示される通りであった。
表4に示されるように、710μm未満の放出制御型ビタミンD製剤では保存後30日及び60日で規定値の20%以上の逸脱が認められた。一方、710~1510μmの粒径を有する顆粒におけるビタミンD3の定量値は、保存前と比較し大きな変動は認められなかった。

本実施例では、顆粒の粒径とバイパス率(すなわち、第一胃を通過する割合)との関係を調べた。

実施例2の記載の通りにリジンバイパス製剤を作成した。バイパス製剤を粒径に応じてふるい分けて粒径毎のバイパス率を調査した。バイパス率は、

疑似第一胃液は、リン酸水素二ナトリウム12水和物(Na2HPO4・12H2O)6.3gとリン酸二水素カリウム(KH2PO4)6.7g/Lを含む水溶液(pH6.4)900mLとし、40℃で16時間攪拌した。結果は表5に示される通りであった。
表5に示されるように、粒径が709μm以下の場合には、バイパス率が30%以下となり、バイパス率が低いのに対して、粒径が710μmを超えると、50%を超えるバイパス率が実現され、特に粒径が1000μm以上の場合にバイパス率が60%を超え、さらに粒径が1000μm~1519μmの場合にバイパス率が約80%に達した。
本発明によれば、700μm以上の均一な粒径、特に1000μm~1510μmの均一な粒径を有する顆粒を効果的に得ることができる。従って、実施例4の結果に基づけば、得られた顆粒は長期安定性に優れる。また、実施例5の結果に基づけば、得られる顆粒は、その粒径範囲がバイパス率の高い粒径範囲となる。
なお、粒径が2mmを超えると、反芻動物の咀嚼により顆粒剤が砕かれる可能性が高まるので、1500μm以下の顆粒が効率よく得られる方法は、産業上極めて有用であると考えられる。また、粒径が700μm以上であると、上記のように製剤の安定性と第一胃のバイパス率に優れるので、700μm以上の顆粒が効率よく得られる方法は、産業上極めて有用であると考えられる。従って、700μm以上の均一な粒径、特に1000μm~1510μmの均一な粒径を有する顆粒を効果的に得ることができる本発明は、産業上極めて有用な発明である。

Claims (13)
- ルーメンバイパス製剤を製造する方法であって、
ルーメンバイパス製剤用の被覆剤と、ルーメンをバイパスさせる栄養素との溶融物が含まれている、少なくとも1つの噴射口を有するダイヘッドまたはダイヘッドに含まれる前記溶融物に振動を加え、これにより、溶融物を噴射口から噴射させることを含む、方法。 - 振動が1000Hz~10000Hzの範囲の振動である、請求項1に記載の方法。
- 振動が3000Hz~7000Hzの範囲の振動である、請求項1または2に記載の方法。
- 振動が、ダイヘッド内腔に露出し溶融物に直接接触する振動子を介して溶融物に与えられる、請求項1~3のいずれか一項に記載の方法。
- 被覆剤が硬化油である、請求項1~4のいずれか一項に記載の方法。
- 硬化油がパーム硬化油および菜種硬化油からなる群から選択される1以上の硬化油である、請求項5に記載の方法。
