Abstract

본 명세서에는 유효성분으로 3β,23,28-트리하이드록시-12-올레아넨 23-카페에이트(3β,23,28-trihydroxy-12-oleanene 23-caffeate), 이의 염, 이의 이성질체, 이의 수화물 또는 이의 용매화물; 및 3β,23,28-트리하이드록시-12-올레아넨 3β-카페에이트(3β,23,28-trihydroxy-12-oleanene 3β-caffeate), 이의 이성질체, 이의 수화물 또는 이의 용매화물; 중 어느 하나 이상을 유효성분으로 포함하는 피부 보습 또는 피부 미백용 조성물에 관한 것으로, 상기 조성물은 피부 외용제 조성물 또는 화장료 조성물로 사용될 수 있고, 상기 조성물은 피부 보습 또는 피부 미백의 효과가 있다.

Description

펜타사이클릭 트리터펜 카페익 액씨드 에스터를 포함하는 피부 보습 또는 피부 미백용 조성물
본 명세서는 유효성분으로 펜타사이클릭 트리터펜 카페익 액씨드 에스터를 포함하는 피부 보습 또는 피부 미백용 조성물에 관한 것이다.
무궁화 목근피 추출물이 항노화, 미백, 보습등에 효과가 있음은 알려져 있다. 다만, 무궁화 목근피 추출물의 성분 중 하나인 Pentacyclic triterpene caffeic acid esters에 대하여는 지질과산화 억제 효과 및 암세포에 대한 독성효과에 대하여는 알려진 바가 있으나, 상기 Pentacyclic triterpene caffeic acid esters가 화장료 조성물등에 사용되는 경우에 있어서 피부 보습 또는 피부 미백 효과에 대하여는 전혀 알려진 바가 없다.
이에 본 발명의 발명자들은 상기 Pentacyclic triterpene caffeic acid esters의 피부 보습 및 피부 미백 효과에 대하여 연구하여 본 발명에 이르게 되었다.

상기와 같은 문제점에 착안하여, 본 발명의 발명자들은 Pentacyclic triterpene caffeic acid esters에 관한 연구를 하여 상기 Pentacyclic triterpene caffeic acid esters에 피부 보습 및 피부 미백 효과가 있음을 알게 되어 본 발명에 이르게 되었다.

본 발명의 일 측면에서, 3β,23,28-트리하이드록시-12-올레아넨 3β-카페에이트(3β,23,28-trihydroxy-12-oleanene 3β-caffeate)는 상기 화학식 3 에서 R1이 화학식 4이며, R2가 -OH 일 수 있다.
본 명세서에서 상기 화학식 1의 3β,23,28-트리하이드록시-12-올레아넨 23-카페에이트 또는 화학식 2의 3β,23,28-트리하이드록시-12-올레아넨 3β-카페에이트는 목근피에서 유도될 수 있다. 본 발명에서 사용된 "목근피(木槿皮, Hisbiscior cortex)"는, 무궁화과에 속하는 갈잎떨기나무인 무궁화 (Hibiscus syriacus L.)의 뿌리껍질 또는 줄기껍질을 말린 것이다. 봄에 뿌리껍질 또는 줄기껍질을 벗겨 햇볕 에 말린다. 맛은 달고 쓰며, 성질은 평하다. 간경, 비경, 대장경, 소장경에 작용한다. 열을 내리고 습을 없애며, 해독하고 살충한다. 또 혈을 잘 돌게 하고 출혈을 멈춘다. 장출혈, 이질, 탈항, 대하, 옴, 무좀, 치질 등에 쓴다. 하루 3~9 g을 탕약으로 먹거나 외욕약으로 쓸 때는 달인 물로 씻거나 술에 우려서 바른다.

본 발명의 일측면에서, 상기 조성물은 3β,23,28-트리하이드록시-12-올레아넨 23-카페에이트(3β,23,28-trihydroxy-12-oleanene 23-caffeate), 이의 염, 이의 이성질체, 이의 수화물 또는 이의 용매화물; 및 3β,23,28-트리하이드록시-12-올레아넨 3β-카페에이트(3β,23,28-trihydroxy-12-oleanene 3β-caffeate), 이의 염, 이의 이성질체, 이의 수화물 또는 이의 용매화물;를 포함하는, 조성물일 수 있다.
본 발명의 일측면에서, 상기 3β,23,28-trihydroxy-12-oleanene 23-caffeate 및 3β,23,28-trihydroxy-12-oleanene 3β-caffeate는 1 : 0.5 내지 1.5 의 중량비로 포함될 수 있다. 구체적으로, 상기 3β,23,28-trihydroxy-12-oleanene 23-caffeate 및 3β,23,28-trihydroxy-12-oleanene 3β-caffeate는 1 : 0.6 내지 1.4의 중량비, 1 : 0.8 내지 1.3의 중량비, 1 : 0.9 내지 1.2의 중량비, 1 : 0.9 내지 1.1의 중량비 또는 1 : 1의 중량비일 수 있다. 상기 3β,23,28-trihydroxy-12-oleanene 23-caffeate 및 3β,23,28-trihydroxy-12-oleanene 3β-caffeate의 중량비가 상기 범위내 일 때, 피부 보습 또는 피부 미백 효과가 우수하다.
본 발명의 일측면에서, 상기 피부 보습용 조성물은 세라마이드를 더 포함하는 것을 특징으로 하는, 조성물이다.
본 명세서에서 세라마이드라 함은 천연 세라마이드 및 유사(Pseudo) 세라마이드로 이루어진 군에서 선택되는 하나 이상인 것일 수 있다. 상기 천연 세라마이드는 자연에 존재하는 세라마이드로서, 세라마이드 1, 세라마이드 2, 세라마이드 3, 세라마 이드 4, 세라마이드 5, 세라마이드 6, 세라마이드 7, 세라마이드 8 등이 포함되고, 상기 유사(Pseudo) 세라마이드는 세라마이드 104 및 세라마이드 102등을 포함한다.
본 발명에서 유사 세라마이드는 천연 세라마이드의 구조인 이중사슬 라멜라(lamellar) 구조를 가지는 화합물을 통칭하는 것이다.
본 발며의 일 측면에서, 펜타사이클릭 트리터펜 카페익 액씨드 에스터인 상기 유효성분 및 상기 세라마이드는 1 : 0.1 내지 1.5 의 중량비로 포함될 수 있다. 구체적으로 상기 펜타사이클릭 트리터펜 카페익 액씨드 에스터 및 상기 세라마이드는 1 : 0.15 내지 1의 중량비, 1 : 0.18 내지 0.9의 중량비, 1 : 0.19 내지 0.8의 중량비, 1 : 0.2 내지 0.5의 중량비 또는 1 : 0.2의 중량비일 수 있다. 상기 펜타사이클릭 트리터펜 카페익 액씨드 에스터 및 상기 세라마이드의 중량비가 상기 범위내 일 때, 피부 보습 또는 피부 미백 효과가 우수하다.
본 발명의 일측면에서, 상기 조성물은 피부 외용제 조성물인, 조성물이다. 상기 피부 외용제 조성물은 피부 건조 치료제 또는 피부 미백 치료제의 약학 조성물로 사용될 수 있다. 상기 피부 외용제는 피부 외부에서 도포되는 어떠한 것이라도 포함될 수 있는 총칭이며, 다양한 제형의 화장품, 의약품이 여기에 포함될 수 있다. 상기 피부 외용제는 또한, 특별히 제한되지 않으나, 예를 들어 피부 보습용 또는 피부 미백용 피부 외용제일 수 있다.
본 발명의 일측면에서, 상기 조성물은 화장료 조성물인, 조성물이다.
상기 화장료 조성물은 제형이 특별히 한정되지 않으며, 목적하는 바에 따라 적절히 선택할 수 있다. 예를 들어, 유연화장수(스킨로션 및 밀크로션), 영양화장수, 에센스, 영양크림, 마사지크림, 팩, 젤, 에센스, 아이크 림, 아이에센스, 클렌징크림, 클렌징폼, 클렌징워터, 팩, 파우더, 보디로션, 보디크림, 보디오일 및 보디 에센스로 이루어진 군으로부터 선택된 어느 하나 이상의 제형으로 제조될 수 있으나, 이에 제한되는 것은 아니다. 상기 유효 성분의 함량은 특별히 제한되지 않으나, 조성물 총 중량을 기준으로 0.0001 내지 10 중량%로 포함될 수 있다. 상기 유효 성분이 상기 함량을 만족하는 경우 부작용 없이 우수한 효능을 나타낼 수 있다. 상기 화장료 조성물은 또한 조성물이 피부에 도포될 때 분포되기 용이하도록, 희석제, 분산제 또는 담체로서 작 용하는 화장용으로 허용 가능한 부형제를 포함할 수 있다. 바람직하게는, 조성물은 수중유형(oil-in-water, O/W) 유화액이고, 유화액은 바람직하게는 물을 부형제의 80 중량% 이상으로 함유한다. 그러나 이에 한정되는 것은 아니며, 공지의 화장용으로 허용 가능한 부형제이면 어느 것이라도 사용 가능하다. 또한, 상기 화장료 조성물은 본 발명의 분야에서 통상 사용되는 화장 보조제들, 이를테면 지방물질, 유기용매, 규소, 농후제, 연화제, 보충적인 태양광선 차단제, 발포억제제, 보습제, 향료, 보존제, 표면활성제, 총전재, 봉쇄제, 양이온성, 음이온성, 비이온성 혹은 양쪽성 중합체들 또는 그것들의 혼합물, 추진제, 염기화 작용제 또는 산성화 작용제, 염료, 안료 혹은 나노안료(nanopigment) (특히, 자외선조사를 물리적으로 차단시킴으로써 광보 호 효과를 보충적으로 제공할 수 있도록 만들어진 것), 또는 화장품에서 통상 사용되는, 특히 태양 광선 차단조성물의 제조에 사용되는 기타 여러 가지 성분들을 포함할 수가 있다. 유기용매들 중에서는, 저급알코올 및 폴리올, 이를테면 에탄올, 이소프로판올, 프로필렌글리콜, 글리세롤 및 솔비톨 등이 언급될 수 있다. 지방물질은 오일이나 왁스 또는 그것들의 혼합물, 지방산, 지방산에스테르, 지방알코올, 페트롤라텀, 파라핀, 라놀린, 수소화라놀린 또는 아세틸화라놀린으로 구성될 수 있다. 오일은 식물성, 광물성 또는 합성오일로, 특히 수소화팜유, 수소화피마자유, 액체페트롤라텀, 액체파라핀, 푸르셀린유(purcellin oil), 휘발성 또는 비휘발성 실리콘유 그리고 이소파라핀에서 선택될 수 있다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 예시하기 위한 것으로, 본 발명의 범위가 이들 실시예에 의해 제한되는 것으로 해석되지 않는 것은 당업계에서 통상의 지식을 가진 자에게 있어서 자명할 것이다.
실시예 1 내지 4 및 비교예 1
㈜ 건화약품으로부터 목근피 구입하여 목근피 추출물을 비교예 1로 사용하였다.
본 발명의 3β,23,28-trihydroxy-12-oleanene 23-caffeate는 실시예 1(PTCE 1)로, 3β,23,28-trihydroxy-12-oleanene 3β-caffeate는 실시예 2(PTCE 2)로 사용하였다.
비교예 1 추출방법
A. 건조된 목근피를 70% 에탄올 50℃에서 3시간 추출한다.
B. 1.2um filtering 하여 일반 추출물을 분리하고, 상기 분리된 추출물을 비교예 1로 활용하였다.(목근피 추출물: HE)
상기 실시예 1 내지 2를 하기 표 1에 따라 실시예 1 내지 4로 하여 하기 피부 보습 및 피부 미백 실험에 사용하였다.

실험예 1 : 피부 보습 평가(히알루론산 생합성 효소( HAS2 , hyaluronic acid synthase2)와 caspase 14 Real Time PCR 정량)
HAS2(Hyaluronan synthase 2)는 히알루논(Hyaluronan) 또는 히알루론산(hyaluronic acid, HA) 를 만드는 유전자다. Caspase 14는 케라티노사이트의 말단 분화(keratinocyte terminal differentiation)에 관여하여 피부 장벽(skin barrier)에 주요한 단백질이다. 피부 보습에 있어, 상기 HA는 안에서 채워지며, Caspase 14는 장벽을 튼튼하게 하여 수분 손실을 막아주며, 또한 filaggrin를 분해하여 천연보습인자성분(NMF성분)으로 만드는데도 관여한다. 이러한 보습에 중요한 역할을 하는 HAS2와 Caspase 14의 발현량을 상기 비교예 1 및 실시예 1 내지 3으로 실험하였다.
A. 세포배양
각질 형성 세포인 HcCaT 세포를 DMEM 배지에 10% FBS, Streptomycin을 포함한 배양액을 사용하여 37℃, 5% CO2 인큐베이터 조건 하에 배양하였다. 배양시 최대 90% 정도의 confluency를 유지하였으며 그 이후엔 subcultication 하였다.
B. Real time PCR
상기 A.를 통해 배양된 HaCat 세포를 12 well plate에 well당 1x104개씩 seeding 한 후 24시간 배양 후에 실시예 1 내지 3과 비교예 1을 처리한다. 세포들을 24시간 뒤에 RNA추출을 위하여 harvest한다. Invirogen사의 TriZol로 Prep한 RNA를 Invitrogen사의 superscript 를 이용하여 RT-PCR을 수행한 후 얻어진 cDNA를 taqman probe를 이용하여 real time qPCR을 수행한다. 상대적인 mRNA 레벨은 아이사이클러(ICycler) 소프트웨 어를 이용하여 SYBR 그린 I 형광 변화를 측정함으로써 분석하였다. HAS2 및 caspase 14 프라이머는 Life technologies 社를 이용하였다(HAS2: Hs00193435_m1, caspase14: Hs00201637_m1). 내부 표준물질로 GAPDH(glyceraldehyde 3-phosphate dehydrogenase)를 사용하여 유전자의 정량적 발현 수준을 보정하였다. HAS및 caspase-14 유전자의 발현 정도는 각각 도 1 내지 2에 나타내었다.
피부 보습 실험결과, 도 1 내지 도 2와 같이, 무처리군에 비해 본원발명 실시예 1 내지 3 처리 군이 HAS및 caspase-14 유전자의 발현 정도가 우수하였으므로, 피부 보습 효과가 우수한 것이 확인된다. 특히, 실시예 3이 HAS및 caspase-14 유전자의 발현 정도가 가장 우수하였다. 이에 더해, 비교예 1의 목근피 추출물(HE)에 비해 실시예 1 내지 3 처리 군이 HAS및 caspase-14 유전자의 발현에 있어 현저한 효과를 보였다.
실험예 2 : 피부 보습 평가
피부 보습에 중요한 인자로 알려진 filaggrin 단백질을 면역 형광 염색을 통해 이미지를 촬영하여, 실시예 1 및 실시예 4의 보습 능력을 평가하였다. 면역 형광 이미지는 타겟하는 단백질의 양에 따라 비례한다.
인공피부인 AmoreReskin TM의 표면에 세라마이드 PC 104 2ppm, 실시예 1 및 실시예 4를 4일간 처리후, 면역 형광 염색 진행하였다. 면역 형광 염색은 하기 실험방법에 따라 진행하였다.
면역형광염색 방법
i. 인공피부를 OCT 용액(Optimal Cutting Temperature 용액, Tissue Tek 社)에 담근후 -70℃ 냉장 보관
ii. Cryotome 이용하여 10㎛ 두께로 절편을 제작하여 슬라이드에 고정
iii. PAP pen을 이용하여 인공피부 절편 주위 선을 그려준다
iv. 슬라이드를 상온에서 8시간 이상 건조
v. PBS로 인공피부 절편을 washing하여 OCT 용액에 녹인다
vi. Blocking solution(1% BSA in PBS)을 30분간 처리
vii. 필라그린 단백질에 대한 항체(Anti-filaggrin antibody(ab24584), Abcam 社)를 2시간 처리
viii. Texas red anti-rabbit 항체(T6391, Life technologies 社) 를 1:200으로 blocking solution에 희석하여 1시간 처리
ix. DAPI(4',6-diamidino-2-phenylindole) staining 5분 진행
x. PBS로 3회 washing
xi. 남은 PBS 제거하여 mounting solution을 인공피부 절편위에 넣은후 커버글라스로 덮어 굳힌다
xii. LSM510 현미경을 이용하여 관찰
면역 형광 실험에 따른 실험 결과는 도 3에 도시하였다. 도 3의 원본에서 파란색으로 관찰되는 부분이 필라그린 단백질 부분이었다. 그러나 상기 부분은 흑백도면에서 표시되지 않으므로, 도 3의 파란색 부분을 표시할 수 있기 위해 흑백도면으로 변환하면서, 원본의 파란색 부분을 백색 부분으로 변환하였다. 따라서, 흑백도면인 도 3에서 백색 부분이 필라그린 단백질 부분이다.
실험결과, 도 3에 도시된 것과 같이 백색으로 관찰되는 부분인 필라그린 단백질 부분의 증가가 실시예 1 및 4에서 뚜렷이 관찰되었음을 확인하였다. 따라서, 실시예 1 및 4에서 필라그린 단백질 부분이 증가하여 보습력이 향상되었음을 확인하였다.
실험예 3 : 피부 미백 평가(B16 melanoma cell을 이용한 melanin assay)
B16 melanoma cell을 12well plate에 well당 1.5x105 cell이 되도록 seeding한 다음 24h동안 배양한다. 실시예 1 내지 3, 비교예 1과 양성대조군 처치후 배지는 6일간 처리하며 3일에 한번 새배지로 교환한다. 시험물질 처리 완료 후 cell을 DPBS로 washing한 다음 2N NaOH(10%DMSO)를 넣고 60도에서 1h 동안 용해시킨다. 세포 배양액과 용해된 cell lysate를 96 well plate에 옮겨 475nm에서 흡광도를 측정한다. 측정된 값은 synthetic melanin에 대한 표준 곡선으로부터 melanin content로 산출된다. 산출된 melanin content는 각 실험군의 단백질 총량으로 표준화한 다음 대조군과 비교하여 효능을 평가한다.
실험 결과를 도 4에 도시하였다. 도 4에서 확인된 바와 같이, 미백 효능에 있어, 본원발명 실시예 1 내지 3은 비교예 1 및 양성대조군에 비해 우수한 미백 효과를 보임을 확인하였다.
이하, 본 발명에 따른 조성물의 제형 예를 설명하나, 이는 본 발명을 한정하고자 함이 아닌 단지 구체적으로 설명하고자 함이다.

  1. 3β,23,28-트리하이드록시-12-올레아넨 23-카페에이트(3β,23,28-trihydroxy-12-oleanene 23-caffeate), 이의 염, 이의 이성질체, 이의 수화물 또는 이의 용매화물; 및 3β,23,28-트리하이드록시-12-올레아넨 3β-카페에이트(3β,23,28-trihydroxy-12-oleanene 3β-caffeate), 이의 염, 이의 이성질체, 이의 수화물 또는 이의 용매화물; 중 어느 하나 이상을 유효성분으로 포함하는 피부 보습용 조성물.
  2. 3β,23,28-트리하이드록시-12-올레아넨 23-카페에이트(3β,23,28-trihydroxy-12-oleanene 23-caffeate), 이의 염, 이의 이성질체, 이의 수화물 또는 이의 용매화물; 및 3β,23,28-트리하이드록시-12-올레아넨 3β-카페에이트(3β,23,28-trihydroxy-12-oleanene 3β-caffeate), 이의 염, 이의 이성질체, 이의 수화물 또는 이의 용매화물; 중 어느 하나 이상을 유효성분으로 포함하는 피부 미백용 조성물.
