Abstract
Description
<1> 下記一般式(1)で表されるヌクレオチド単位及び下記一般式(2)で表されるヌクレオチド単位の少なくとも1つの単位が、3個~50個連続する構造を含み、塩基長が8塩基~50塩基であるRNAハイブリッド形成用核酸オリゴマー。
<2> 前記一般式(1)及び前記一般式(2)中、Bs1は、下記一般式(3)又は一般式(4)で表される基である<1>に記載のRNAハイブリッド形成用核酸オリゴマー。
ここで、相補鎖との二重鎖形成能とは、相補鎖となるRNAと、二重鎖の形成しやすさをいう。二重鎖形成能は、例えば、本発明のRNAハイブリッド形成用核酸オリゴマーと、天然型相補鎖RNAとの二重鎖の融解温度(Tm)で表すことができる。Tmは、例えば、温度の可変が可能なセルを装備した紫外可視分光光度計を用い、260nmにおける吸光度を各温度で測定し、融解曲線を得ることによって求めることができる。
従って、核酸塩基がピリミジンである場合には、該硫黄原子やボラノ基との強い相互作用が生じることにより、分子内の構造がより安定化する結果、さらに相補鎖との結合もより強固となり、結果としてA型のらせん構造がより安定化するものと考えられる。
本明細書において「モノマー」とは、核酸オリゴマーを製造する際に、核酸オリゴマーのヌクレオチド単位を導入するために使用される核酸をいう。
また本明細書において「~」は、その前後に記載される数値をそれぞれ最小値および最大値として含む範囲を示すものとする。
さらに本明細書において組成物中の各成分の量は、組成物中に各成分に該当する物質が複数存在する場合、特に断らない限り、組成物中に存在する当該複数の物質の合計量を意味する。
以下、本発明について説明する。
下記一般式(1)で表されるヌクレオチド単位及び下記一般式(2)で表されるヌクレオチド単位の少なくとも1つの単位が、3個~50個連続する構造を含み、塩基長が8塩基~50塩基であるRNAハイブリッド形成用核酸オリゴマーである。
RNAハイブリッド形成用核酸オリゴマーの塩基配列は、二重鎖形成能の観点から、標的となるRNAの塩基配列に対して相補的な塩基配列となるように適宜選択されるが、場合によって、異なる塩基を1つ以上含む塩基配列であってもよい。
アルケニルオキシ基としては、例えば、ビニルオキシ基、アリルオキシ基、1-プロペニルオキシ基、イソプロペニルオキシ基、2-メチル-1-プロペニルオキシ基、2-メチルアリルオキシ基、2-ブテニルオキシ基等を挙げることができる。
アシルオキシ基としては、例えば、炭素数1から6のアルキル-カルボニルオキシ基(例えば、メチルカルボニルオキシ基、エチルカルボニルオキシ基等)、炭素数6から10のアリール-カルボニルオキシ(例えば、ベンゾイルオキシ基)などが挙げられる。
トリアルキルシリルオキシ基としては、例えば、トリメチルシリルオキシ基、トリエチルシリルオキシ基などを挙げることができる。
ハロゲニル基としては、例えば、フルオロ基、クロロ基、ブロモ基などを挙げることができる。
R1としては、二重鎖形成能の点から、上記の中でも、水素原子、アルコキシ基及びトリアルキルシリルオキシ基が好ましい。
また、アルコキシ基としては、炭素数1~12のアルコキシ基が好ましく、さらに炭素数1~6のアルコキシ基がより好ましい。
また、ハロゲニル基としてはフルオロ基がより好ましい。
前記シクロアルキル基としては、シクロプロピル基及びシクロヘキシル基が挙げられる。
前記アルケニル基としては、炭素数2~10の直鎖、分岐鎖もしくは環状のものを挙げることができる。具体的には、例えばビニル基、プロペニル基、ブテニル基、ペンテニル基、ヘキセニル基、ヘプテニル基、オクテニル基、ノネニル基及びアリル基などがあげられる。
前記アリール基としては、炭素数6~12のものを挙げることができる。具体的には、例えば、フェニル、1-ナフチル、2-ナフチル、ビフェニル等を挙げることができる。
前記ヘテロアリール基としては、炭素数3~10のものを挙げることができる。具体的には、1,2,3-トリアゾール、イミダゾール及びチオフェン等が挙げられる。
前記アルキニル基としては、炭素数2~10の直鎖もしくは分岐のものを挙げることができる。具体的には、例えば、エチニル基、1-プロピニル基、2-プロピニル基、1-ブチニル基、2-ブチニル基、3-ブチニル基、1-メチル-2-プロピニル基、2-メチル-3-ブチニル基、1-ペンチニル基、2-ペンチニル基、3-ペンチニル基、4-ペンチニル基、1-メチル-2-ブチニル基、2-メチル-3-ペンチニル基、1-ヘキシニル基、1,1-ジメチル-2-ブチニル基等などがあげられる。
前記アシル基としては、例えば、直鎖状又は分岐鎖状の炭素数1~10のアルカノイルを挙げることができる。具体的には、例えば、アセチル基、n-プロピオニル基、イソプロピオニル基、n-ブチリル基及びヘキサノイル基などがあげられる。
上記の置換基のうち、二重鎖形成能の観点からは、炭素数2~5のアルキル基、炭素数2~5のアルケニル基、炭素数2~5のアルキニル基及びハロゲニル基が好ましく、さらに、炭素数2~5のアルキニル基及びハロゲニル基がより好ましく、炭素数3~4のアルキニル基が特に好ましい。
また、R2及びR3は、互いに結合して環を形成してもよい。具体的には、例えば、R2R3、及びR2が結合するR3以外の炭素原子が、シクロプロパン環、シクロブタン環、シクロヘキサン環等の環状構造を形成してもよい。
炭素数2~10のアルケニル基としては、ビニル基、プロペニル基、ブテニル基、ペンテニル基、ヘキセニル基などが挙げられる。
炭素数2~10のアルキニル基としては、エチニル基、1-プロピニル基、2-プロピニル基、1-ブチニル基、2-ブチニル基、3-ブチニル基、1-メチル-2-プロピニル基、2-メチル-3-ブチニル基、1-ペンチニル基、2-ペンチニル基、3-ペンチニル基、4-ペンチニル基、1-メチル-2-ブチニル基、などが挙げられる。
また、以下の縮合工程に用いるモノマーの立体化学的純度としては、例えば、97%以上のものを用いることが好ましい。
前記文献に記載のモノマーの縮合反応では、リン原子の絶対立体配置を高度に維持して縮合することが可能であるため、上記モノマー合成工程で得られたモノマーを用いて当該縮合反応させることにより、結果として、立体化学的純度の高い本発明のRNAハイブリッド形成用核酸オリゴマーを得ることができる。また、モノマーの縮合においては、用いるモノマー中のピリミジン塩基の5位の置換基の種類として、例えば、すべて一定の置換基が結合した塩基を有するモノマーを用いてもよく、異なる置換基が結合した塩基を有するモノマーを用いてもよい。
各種分析器は以下に示した機種を用いた。

なお、1H-NMRにはテトラメチルシラン(TMS)を、13C-NMRにはCDCl3(δ77.16ppm)を内部標準として用い、31P-NMRは85%H3PO4を外部標準として用いた。

オキサザホスホリジンモノマーは、DMTr基で保護した各ヌクレオシド(化合物3a~3e)とアミノアルコールから誘導した化合物(化合物D2又は化合物L2)とを反応させて合成した。
以下、詳細について述べる。
アミノアルコールである化合物D1(1.77g、10mmol)をトルエンで繰り返し共沸乾燥し、該トルエン溶液5mLにメチルモルホリン2.20mL(20mmol)を加え混合溶液とした。該混合溶液に三塩化リン870μL(10mmol)を含むトルエン5.0mL溶液を、0℃で撹拌しつつ添加し、さらに室温で2時間撹拌して反応を行った。反応後に生じた塩をAr雰囲気下、-78℃でろ別し、アルゴン雰囲気下、減圧濃縮することで、黄色オイル状の2-クロロ-1,3,2-オキサザホスホリジン(化合物D2)2.09gを得た。得られた化合物D2はこれ以上の精製はおこなわず、次の化合物の合成に用いた。
以下、各オキサザホスホリジンモノマーの合成に用いる、5’-O-ジ(p-メトキシフェニル)フェニルメチル(DMTr)-2’-デオキシウリジン(化合物3a)、5’-O-ジ(p-メトキシフェニル)フェニルメチル(DMTr)-チミジン(化合物3b)、5’-O-ジ(p-メトキシフェニル)フェニルメチル(DMTr)-2’-デオキシブロモウリジン(化合物3c)、5’-O-ジ(p-メトキシフェニル)フェニルメチル(DMTr)-2’-デオキシヨードウリジン(化合物3d)及び5’-O-ジ(p-メトキシフェニル)フェニルメチル(DMTr)-2’-デオキシプロピニルウリジン(化合物3e)は、文献[J.Am.Chem.Soc.,Vol.104,1316-1319]に記載の方法を参考に、2’-デオキシウリジン(TCI社製)、チミジン(和光純薬工業株式会社)、2’-デオキシブロモウリジン(和光純薬工業株式会社)、2’-デオキシヨードウリジン(和光純薬工業株式会社)から合成した。5’-O-ジ(p-メトキシフェニル)フェニルメチル(DMTr)-2’-デオキシプロピニルウリジン(化合物3e)は、Tetrahedron Lett.,Vol.45,2457-2461に記載の方法を参考に5’-O-ジ(p-メトキシフェニル)フェニルメチル(DMTr)-2’-デオキシヨードウリジン(化合物3d)から合成した。
<化合物(Sp)-4aの合成>
化合物3a(531mg、1.0mmol)をピリジン、トルエンで繰り返し共沸し、テトラヒドロフラン5mL溶液とし、トリエチルアミン0.97mL(7mmol)を加えて混合し-78℃に冷却し、化合物D2(725mg)を溶解したテトラヒドロフラン(THF)6mLを滴下した後、室温で2時間攪拌した。その後、クロロホルム300mL、飽和炭酸水素水溶液(100mL)を加えて分液操作を行い、有機相を飽和炭酸水素水溶液100mLで2回洗浄し、さらに、回収した洗浄液にクロロホルムを添加した後分液操作を行い、有機層を回収した。回収したすべての有機層を無水硫酸ナトリウムで乾燥後、ろ過し、溶媒を減圧蒸留去して残渣を得た。得られた残渣をシリカゲルカラムクロマトグラフィー(16gのNH-シリカゲル、富士シリシア社製)[展開溶媒:トルエン/酢酸エチル/トリエチルアミン(70/30/0.1、v/v/v)]で分離精製後、溶媒を減圧留去することで、(Sp)-4a(313mg、0.43mmol)を収率43%で得た。

化合物3b(544mg、1.0mmol)をピリジン、トルエンで繰り返し共沸し、テトラヒドロフラン5mL溶液とし、トリエチルアミン0.97mL(7mmol)を加えて混合し-78℃に冷却し、化合物D2(725mg)を溶解したテトラヒドロフラン(THF)6mLを滴下した後、室温で2時間攪拌した。その後、化合物(Sp)-4aの方法と同様の方法によって残渣を得た。得られた残渣をシリカゲルカラムクロマトグラフィー(15gのNH-シリカゲル、富士シリシア社製)[展開溶媒:トルエン/酢酸エチル/トリエチルアミン(70/30/0.1、v/v/v)]で分離精製後、溶媒を減圧留去することで、(Sp)-4b(330mg、0.44mmol)を収率44%で得た。
化合物3c(611mg、1.0mmol)をピリジン、トルエンで繰り返し共沸し、テトラヒドロフラン5mL溶液とし、トリエチルアミン0.97mL(7mmol)を加えて混合し-78℃に冷却し、化合物D2(725mg)を溶解したテトラヒドロフラン(THF)6mLを滴下した後、室温で2時間攪拌した。その後、化合物(Sp)-4aの方法と同様の方法によって残渣を得た。得られた残渣をシリカゲルカラムクロマトグラフィー(15gのNH-シリカゲル、富士シリシア社製)[展開溶媒:トルエン/酢酸エチル/トリエチルアミン(80/20/0.5(v/v/v)から0/100/0.5(v/v/v))]で分離精製後、溶媒を減圧留去することで、化合物(Sp)-4c(280mg、0.44mmol)を収率30%で得た。

化合物3d(658mg、1mmol)をピリジン、トルエンで繰り返し共沸し、テトラヒドロフラン5mL溶液とし、トリエチルアミン0.97mL(7mmol)を加えて混合し-78℃に冷却し、化合物D2(725mg)を溶解したテトラヒドロフラン(THF)6mLを滴下した後、室温で2時間攪拌した。その後、化合物(Sp)-4aの方法と同様の方法によって残渣を得た。得られた残渣をシリカゲルカラムクロマトグラフィー(17gのNH-シリカゲル、富士シリシア社製)[展開溶媒:トルエン/酢酸エチル/トリエチルアミン(80/20/0.1(v/v/v)から0/100/0.1(v/v/v))]で分離精製後、溶媒を減圧留去することで、化合物(Sp)-4d(420mg、0.49mmol)を収率49%で得た。

化合物3a(530mg、1mmol)をピリジン、トルエンで繰り返し共沸し、テトラヒドロフラン5mL溶液とし、トリエチルアミン0.97mL(7mmol)を加えて混合し-78℃に冷却し、化合物L2(725mg)を溶解したテトラヒドロフラン(THF)6mLを滴下した後、室温で2時間攪拌した。その後、化合物(Sp)-4aの方法と同様の方法によって残渣を得た。得られた残渣をシリカゲルカラムクロマトグラフィー(15gのNH-シリカゲル、富士シリシア社製)[展開溶媒:トルエン/酢酸エチル/トリエチルアミン(70/30/0.1(v/v/v)]で分離精製後、溶媒を減圧留去することで、化合物(Rp)-4a(360mg、0.49mmol)を収率49%で得た。

化合物3b(545mg、1mmol)をピリジン、トルエンで繰り返し共沸し、テトラヒドロフラン5mL溶液とし、トリエチルアミン0.97mL(7mmol)を加えて混合し-78℃に冷却し、化合物L2(725mg)を溶解したテトラヒドロフラン(THF)6mLを滴下した後、室温で2時間攪拌した。その後、化合物(Sp)-4aの方法と同様の方法によって残渣を得た。得られた残渣をシリカゲルカラムクロマトグラフィー(15gのNH-シリカゲル、富士シリシア社製)[展開溶媒:トルエン/酢酸エチル/トリエチルアミン(70/30/0.1(v/v/v)]で分離精製後、溶媒を減圧留去することで、化合物(Rp)-4b(350mg、0.35mmol)を収率47%で得た。
化合物3c(611mg、1mmol)をピリジン、トルエンで繰り返し共沸し、テトラヒドロフラン5mL溶液とし、トリエチルアミン0.97mL(7mmol)を加えて混合し-78℃に冷却し、化合物L2(725mg)を溶解したテトラヒドロフラン(THF)6mLを滴下した後、室温で2時間攪拌した。その後、化合物(Sp)-4aの方法と同様の方法によって残渣を得た。得られた残渣をシリカゲルカラムクロマトグラフィー(15gのNH-シリカゲル、富士シリシア社製)[展開溶媒:トルエン/酢酸エチル/トリエチルアミン(80/20/0.1(v/v/v)から0/100/0.1(v/v/v))]で分離精製後、溶媒を減圧留去することで、化合物(Rp)-4c(330mg、0.41mmol)を収率41%で得た。

化合物3d(657mg、1mmol)をピリジン、トルエンで繰り返し共沸し、テトラヒドロフラン5mL溶液とし、トリエチルアミン0.97mL(7mmol)を加えて混合し-78℃に冷却し、化合物L2(725mg)を溶解したテトラヒドロフラン(THF)6mLを滴下した後、室温で2時間攪拌した。その後、化合物(Sp)-4aの方法と同様の方法によって残渣を得た。得られた残渣をシリカゲルカラムクロマトグラフィー(15gのNH-シリカゲル、富士シリシア社製)[展開溶媒:トルエン/酢酸エチル/トリエチルアミン(80/20/0.1(v/v/v)から0/100/0.5(v/v/v))]で分離精製後、溶媒を減圧留去することで、化合物(Rp)-4d(360mg、0.42mmol)を収率42%で得た。

化合物3e(531mg、1mmol)をピリジン、トルエンで繰り返し共沸し、テトラヒドロフラン5mL溶液とし、トリエチルアミン0.97mL(7mmol)を加えて混合し-78℃に冷却し、化合物L2(725mg)を溶解したテトラヒドロフラン(THF)6mLを滴下した後、室温で2時間攪拌した。その後、化合物(Sp)-4aの方法と同様の方法によって残渣を得た。得られた残渣をシリカゲルカラムクロマトグラフィー(17gのNH-シリカゲル、富士シリシア社製)[展開溶媒:トルエン/酢酸エチル/トリエチルアミン(80/20/0.1(v/v/v)から0/100/0.1(v/v/v))]で分離精製後、溶媒を減圧留去することで、化合物(Rp)-4e(420mg、0.29mmol)を収率40%で得た。

以下、各オキサザホスホリジンモノマーの合成に用いる、5’-O-ジ(p-メトキシフェニル)フェニルメチル(DMTr)-2’-デオキシシチジン(化合物5a)、5’-O-ジ(p-メトキシフェニル)フェニルメチル(DMTr)-メチルシチジン(化合物5b)、5’-O-ジ(p-メトキシフェニル)フェニルメチル(DMTr)-2’-デオキシヨードシチジン(化合物5d)及び5’-O-ジ(p-メトキシフェニル)フェニルメチル(DMTr)-2’-デオキシプロピニルシチジン(化合物5e)は、文献[J.Am.Chem.Soc.,Vol.104,1316-1319]に記載の方法を参考に、2’-デオキシシジチン(TCI社製)、5’-メチルシチジン(米国登録特許4,754,026号に記載の方法によりチミジンから合成した社)、2’-デオキシブロモシチジン(和光純薬工業株式会社)、2’-デオキシヨードシチジン(和光純薬工業株式会社)から合成した。5’-O-ジ(p-メトキシフェニル)フェニルメチル(DMTr)-2’-デオキシプロピニルシチジン(化合物5e)は、Tetrahedron Lett.,Vol.45,2457-2461に記載の方法を参考に5’-O-ジ(p-メトキシフェニル)フェニルメチル(DMTr)-2’-デオキシヨードシチジン(化合物5d)から合成した。
化合物(Sp)-6aは、化合物3aに代えて化合物5a(0.96g、1.5mmol)を用いたこと以外は、上記した化合物(Sp)-4aの合成と同様にして、化合物(Sp)-6aを合成した。また、精製においてシリカゲルカラムクロマトグラフィーの条件を[展開溶媒:トルエン/酢酸エチル/トリエチルアミン(80/20/0.1、v/v/v)]とした以外は、化合物(Sp)-4aの精製と同様にして無色の化合物(Sp)-6a(0.70g、0.84mmol、収率56%)を得た。
化合物(Sp)-6bは、化合物3bに代えて化合物5b(0.97g、1.5mmol)を用いた以外は、上記した化合物(Sp)-4bの合成と同様にして、化合物(Sp)-6bを合成および精製した。無色の(Sp)-6bを得た(0.74g、0.87mmol、収率58%)。

化合物(Sp)-6eは、化合物3eに代えて化合物5e(0.67g、1mmol)を用いたこと、および化合物D2の量を483mgに変更した以外は、上記した化合物(Sp)-4eの合成と同様にして、化合物(Sp)-6eを合成した。また、精製においてシリカゲルカラムクロマトグラフィーの条件を[展開溶媒:トルエン/酢酸エチル/トリエチルアミン(90/10/0.5、v/v/vから80/20/0.5、v/v/v)]とした以外は、化合物(Sp)-4eの精製と同様に精製を行って、無色の化合物(Sp)-6eを得た(0.40g、0.45mmol、収率45%)。

化合物(Rp)-6aは、化合物3aに代えて化合物5a(1.27g、2mmol)を用いたこと、及び化合物L2の量を967mgに変更した以外は、上記した化合物(Rp)-4aの合成と同様にして、化合物(Rp)-6aを合成した。また、精製においてシリカゲルカラムクロマトグラフィーの条件を[展開溶媒:トルエン/酢酸エチル/トリエチルアミン(90/10/0.1、v/v/vから80/20/0.1、v/v/v)]とした以外は、化合物(Rp)-4aの精製と同様に精製を行って、無色の(Rp)-6aを得た(0.54g、0.65mmol、収率33%)。
化合物(Rp)-6bは、化合物3bに代えて化合物5b(0.97g、1.5mmol)を用いた以外は、上記した化合物(Rp)-4bの合成と同様にして、化合物(Rp)-6bを合成した。また、精製においてシリカゲルカラムクロマトグラフィーの条件を[展開溶媒:トルエン/酢酸エチル/トリエチルアミン(90/10/0.1、v/v/vから80/20/0.1、v/v/v)]とした以外は、化合物(Rp)-4bと同様に精製を行って、無色の(Rp)-6bを得た(0.54g、0.63mmol、42%)。

化合物(Rp)-6eは、化合物3eに代えて化合物5e(0.68g、1mmol)を用いたこと、および化合物L2の量を483mgに変更した以外は、上記した化合物(Rp)-4eの合成と同様にして、化合物(Rp)-6eを合成した。また、精製においてシリカゲルカラムクロマトグラフィーの条件を[展開溶媒:トルエン/酢酸エチル/トリエチルアミン(80/20/0.5、v/v/v)]とした以外は、化合物(Rp)-4eの精製と同様に精製を行って、無色の(Rp)-6eを得た(0.53g、0.60mmol、収率60%)。

額賀らの文献[額賀陽平; 岡夏央; 前田雄介; 和田猛, リン原子の立体を制御したPS/POキメラオリゴヌクレオチドの固相合成, アンチセンス・遺伝子・デリバリーシンポジウム2014 講演予稿集, p. 52]及びOkaらの文献[J.Am.Chem.Soc.130,16031-16037]に記載の方法に従って、5’末端及び3’末端に天然型のCGの塩基配列を有するキメラオリゴマーであるホスホロチオエート型核酸オリゴマー(5’-CGTTTTTTTTCG-3’、12mer)、末端に天然型のATA及びTATの塩基配列を有するキメラオリゴマーであるホスホロチオエート型核酸オリゴマー(5’-ATACCCCCCTAT-3’、12mer)又はホスホロチオエート結合に代えてリン酸結合により結合する核酸オリゴマーを、上記で得られた各種オキサザホスホリジンモノマー等を用いて合成した。なお、ハロゲン修飾体であるモノマー((SP)-4C、(RP)-4C、(SP)-4d及び(RP)-4d)を用いる場合には、上記文献に記載の“ウルトラマイルド”の条件に従って合成を行った。
具体的な条件の一部を表2に示す。
また、「ps」は、5-メチルウラシル基を有するヌクレオチド残基(T)同士、ウラシル基を有するヌクレオチド残基(U)同士、シチジル基を有するヌクレオチド残基(C)同士又は5位が修飾されたウラシル基を有するヌクレオチド残基同士もしくは5位が修飾されたシチジル基を有するヌクレオチド残基同士、がホスホロチオエート結合していることを示し、例えば、「(Ups)8」は、連続した8個のウラシル基を有するヌクレオチド残基同士が、ホスホロチオエート結合していることを示す。また、(Tps)8は、8個の連続した5-メチルウラシル基を有するヌクレオチド残基同士が、ホスホロチオエート結合していることを示し、(Cps)6は、6個の連続したシチジル基を有するヌクレオチド残基同士が、ホスホロチオエート結合していることを示し、(Cps)2は、2個の連続したシチジル基を有するヌクレオチド残基同士が、ホスホロチオエート結合していることを示す。(BrUps)8は、5位に臭素原子(Br)が結合した8個のウラシル基を有するヌクレオチド残基同士が、ホスホロチオエート結合していることを示し、(IUps)8は、5位にヨウ素原子(I)が結合した8個のウラシル基を有するヌクレオチド残基同士が、ホスホロチオエート結合していることを示し、(pUps)8は、5位にプロピニル基が結合した8個のウラシル基を有するヌクレオチド残基同士が、ホスホロチオエート結合していることを示し、(MeCps)6は、5位にメチル基(Me)が結合した6個のシチジル基を有するヌクレオチド残基同士が、ホスホロチオエート結合していることを示す。(Cps)2(pCps)2(Cps)2は、2個のシチジル基を有するヌクレオチド残基と、5位にプロピニル基が結合した2個のシチジル基を有するヌクレオチド残基と、2個のシチジル基を有するヌクレオチド残基とが、この順でホスホロチオエート結合していることを示す。
また、表3-1及び3-2中のすべてのオリゴマー中、CGはオリゴマーの5’位及び3’位に、シトシン(C)及びグアニン(G)の2残基が天然型で結合していることを示し、ATAまたはTATは、オリゴマーの5’位及び3’位に、アデニン(A)及びチミン(T)の3残基が天然型で結合していることを示す。
mix-d(CG(Tps)8CG)は、5’-O-ジ(p-メトキシフェニル)フェニルメチル(DMTr)-2’-デオキシチミジンのホスホロアミダイトモノマー(Rp体及びSp体の1:1混合物、GlenRsearch社製)を用い、上記と同様の方法でホスホロチオエート型核酸オリゴマーを合成した。mix-d(CG(Tps)8CGにおいては、(Tps)8のTがランダムにRp体またはSp体である。
mix-d(CG(pUps)8CG)は、5-プロピニル - 5’-O-ジ(p-メトキシフェニル)フェニルメチル(DMTr)-2’-デオキシチミジンのホスホロアミダイトモノマー(Rp体及びSp体の1:1混合物、GlenRsearch社製)を用い、上記と同様の方法でホスホロチオエート型核酸オリゴマーを合成した。mix-d(CG(pUps)8CG)においては、(pUps)8のpUがランダムにRp体またはSp体である。
以下に示すリン酸結合型核酸オリゴマーを調製した。
・d(CG(U)8CG)
・d(CG(T)8CG)
・d(CG(BrU)8CG)
・d(CG(IU)8CG)
・d(CG(pU)8CG)
・d(ATA(C)6TAT)
・d(ATA(MeC)6TAT)
塩基に「ps」が付されていないことは、5-メチルウラシル基を有するヌクレオチド残基(T)同士、ウラシル基を有するヌクレオチド残基(U)同士、シチジル基を有するヌクレオチド残基(C)同士又は5位が修飾されたウラシル基を有するヌクレオチド残基同士もしくは5位が修飾されたシチジル基を有するヌクレオチド残基同士がリン酸結合していることを示し、例えば、「(U)8」は、連続した8個のウラシル基同士が、天然型であるリン酸結合で結合していることを示す。(T)8は、8個の連続した5-メチルウラシル基同士が、リン酸結合していることを示す。(C)6は、6個の連続したシチジル基同士が、リン酸結合していることを示す。(BrU)8は、5位に臭素原子(Br)が結合した8個のウラシル基同士がリン酸結合していることを示し、(IU)8は、5位にヨウ素原子(I)が結合した8個のウラシル基同士がリン酸結合していることを示し、(pU)8は、5位にプロピニル基が結合した8個のウラシル基同士がリン酸結合していることを示し、(MeC)6は、5位にメチル基が結合した6個のシチジル基同士がリン酸結合していることを示す。
標的核酸となる天然型のRNA及びDNAは、配列番号1~2に示す塩基配列のRNAオリゴマー(配列番号1)及びDNAオリゴマー(配列番号2)を、日本バイオサービス社の受託合成(HPLCグレード)により入手した。
<実施例1~5及び比較例1-1~5-3:各核酸オリゴマーとRNAの二重鎖、参考例1~8:各核酸オリゴマーとDNAの二重鎖における融解温度測定>
〔実施例1~5〕
以下の表3の実施例1~5に示す核酸オリゴマーのRp体(核酸オリゴマー2、4、6、9、10)の各々0.4nmolと、標的核酸(天然型相補鎖RNA)(配列番号1)の0.4nmolを10mM NaH2PO4-Na2HPO4、100mM NaCl 緩衝液(pH 7.0)160μLに溶かし、それぞれ、8連セルに140μL分を分注した。紫外可視分光光度計のセル温度を室温から5℃/minの速度で95℃まで上昇させた後、95℃の状態を5分間保持し、-0.5℃/分の速度で0℃になるまで温度を下降させ、アニーリングを行なった。オリゴマー溶液をそのまま0℃で30間分静置した後、核酸オリゴマー溶液をセルに入れ、窒素雰囲気下、0.5℃/minの速度で95℃まで温度を上昇させながら、0.25℃間隔で260nmにおける吸光度を測定し、融解曲線を得た。得られた二重鎖融解曲線から中線法により、Tm値を算出した。
核酸オリゴマーのRp体(核酸オリゴマー2、4、6、9、10)を核酸オリゴマーのSp体(核酸オリゴマー1、3、5、7、8)又はリン酸結合型核酸オリゴマーに変更した以外は、実施例1~5と同様にして測定を行い、表3の比較例1-1~5-3に示す通り、二重鎖融解温度(Tm)を得た。
参考例1~4及び参考例6~8は、実施例1~5において、核酸オリゴマーのRp体(核酸オリゴマー2、4、6)を、核酸オリゴマーのSp体(核酸オリゴマー1、3、5、7)に変更し、さらに標的核酸である天然型相補鎖RNAを、天然型相補鎖DNA(配列番号2)に変更した以外は実施例1~5と同じにして、実施例1~5と同様に測定を行い、表3の参考例1~4及び参考例6~8にあるように、二重鎖融解温度(Tm)を得た。また、参考例5については、上記で合成したmix-d(CG(Tps)8CG)を用いて、上記参考例1~4等と同様に、測定を行い、表3の参考例5にあるように、二重鎖融解温度(Tm)を得た。また、参考例1~4及び参考例6~8のTm値の算出に用いた融解曲線の結果を図2に示す。図2(A)は、核酸オリゴマーのSp体(核酸オリゴマー1、3、5、7、8)と天然型相補鎖DNAとの二重鎖の融解曲線を示し、図2(B)は、核酸オリゴマーのRp体(核酸オリゴマー2、4、6、9)と天然型相補鎖DNAとの二重鎖の融解曲線を示す。
これに対して、標的核酸をDNAとした参考例1~8では、Rp体のTm値とSpのTm値が同程度であり、さらに、参考例1~4及び6~7では、むしろRp体のTm値がSpのTm値よりも若干低下していることが示された。
表3に示した、以下のRp体のホスホロチオエート型核酸オリゴマーのESI MSによる測定結果を示す。

(リン酸型核酸オリゴマーのTm測定)
実施例1~5に用いられた核酸オリゴマー(核酸オリゴマー2、4、6、9、10)に代えてリン酸型核酸オリゴマーであるd(ATA(C)6TAT)又はd(ATA(MeC)6TAT)に変更した以外は実施例1~5と同様にして測定を行い、二重鎖融解温度(Tm)を得た。結果を表5に示す。
実施例6、比較例6-1および比較例6-2の二重鎖融解曲線を図3Aに示す。実施例7、比較例7-1および比較例7-2の二重鎖融解曲線を図3Bに示す。実施例9および比較例9-1の二重鎖融解曲線を図3BC示す。

Claims (4)
- 下記一般式(1)で表されるヌクレオチド単位及び下記一般式(2)で表されるヌクレオチド単位の少なくとも1つの単位が、3個~50個連続する構造を含み、塩基長が8塩基~50塩基であるRNAハイブリッド形成用核酸オリゴマー。
(一般式(1)中、R1は、水素原子、アルコキシ基、アルケニルオキシ基、アシルオキシ基、トリアルキルシリルオキシ基又はハロゲニル基を表す。一般式(1)及び一般式(2)中、各Bs1は、それぞれ独立して、保護基を有していてもよいピリミジン塩基であり、前記ピリミジン塩基の5位の炭素原子に結合する水素原子は、水素原子以外の基で置換されていてもよい。一般式(1)及び一般式(2)中、Xは、それぞれ独立して、S-Z+又はBH3 -Z+を表す。Z+は、カウンターカチオンを示す。R2及びR3は、それぞれ独立して、水素原子又は炭素数1~10のアルキル基を表す。R2及びR3は互いに結合して環を形成してもよい。) - 前記塩基長が、10塩基~30塩基である請求項1又は請求項2に記載のRNAハイブリッド形成用核酸オリゴマー。
- 前記一般式(1)で表されるヌクレオチド単位及び前記一般式(2)で表されるヌクレオチド単位の少なくとも1つの単位が、5個~30個連続する構造を含む請求項1~請求項3のいずれか1項に記載のRNAハイブリッド形成用核酸オリゴマー。
