Abstract
본 발명은 양수 내 태아 유래 중간엽 줄기세포의 배양액에 관한 것으로, 보다 구체적으로는 역분화 인자 나노그(Nanog)를 과발현시킨 양수 내 태아 유래 중간엽 줄기세포의 배양액을 유효성분으로 포함하는 발모 촉진 또는 탈모 방지용 조성물에 관한 것이다. 또한, 본 발명은 나노그(Nanog)가 도입된 양수 내 태아 유래 중간엽 줄기세포를 컨디션드 배지에서 배양하는 단계 및 상기 배양액을 수거하는 단계를 포함하는 상기 조성물의 제조방법에 관한 것이다. 본 발명에 따른 컨디션드 배지 조성물은 모발 성장 촉진 효과를 나타내므로, 육모 촉진용 화장료 조성물 및 약학적 조성물로써 활용이 가능하다.

본 발명은 역분화 인자 나노그(Nanog)를 과발현시킨 양수 내 태아 유래 중간엽 줄기세포의 배양액을 유효성분으로 포함하는 육모 촉진용 조성물 및 이의 제조방법에 관한 것이다.
줄기세포(stem cell)는 신체 내 외의 환경 및 자극을 통해 신체를 구성하는 각종 세포로 분화할 수 있는 능력을 갖추고 있으며, 자가증식 능력을 갖추고 있는 세포로서, 초기 배아에서 분리한 배아 줄기세포(embryonic stem cell, ES 세포), 배아기의 원시 생식세포에서 분리한 배아 생식세포(embryonic germ cell. EG 세포), 및 성체의 골수에서 분리한 다능성 성체 줄기세포(multipotent adult progenitor cell, MAPC 세포) 등 3종이 잘 알려져 있다.
성체 줄기세포의 한 종류인 골수에서 유래한 중간엽 줄기세포는 오래전부터 사용되어 왔고 다양한 효과도 입증되었다. 또한, 최근에는 지방조직이나 다른 조직에서 분리한 세포들도 골수 유래 중간엽 줄기세포와 유사한 특성이 있다는 것이 밝혀지고 있다.
본 발명자들은 산모나 태아에게서 쉽게 분리 가능한 양수에 주목하였다. 양수를 통하여 태아의 건강에 대한 여러 가지 정보를 알아보는 검사를 하며, 임신이 시작되는 순간부터 출산 직후까지 모체에 해를 끼치지 않고 양수를 추출할 수 있다. 검사에 사용된 양수 세포는 검사 후 폐기하게 되는데 환자의 동의를 얻을 경우 폐기하지 않고 연구용 목적으로 사용할 수 있으므로, 양수 세포는 기존에 연구되어 오던 다른 성체 줄기세포에 비해 쉽게 다량의 세포를 획득할 수 있다는 장점을 가진다.
한편, 사람의 모발은 각각의 독자적인 성장주기, 즉 성장기(Anagen) -> 퇴행기 -> 휴지기 -> 성장기의 모주기(hair cycle)를 가지고 있기 때문에 털갈이 없이 항상 일정한 모발의 수를 유지하게 된다. 그러나 대머리가 진행되면 모근에 존재하는 모유두가 작아지고, 모유두가 작아지면 머리털의 굵기도 가늘어지고 동시에 모주기도 짧아지며, 새로 자라나온 털은 더욱 가늘어지게 된다. 따라서 대머리가 진행되면 머리털은 솜털로 변하며 모발의 성장주기는 더욱 짧아져 조금 자란 후 빠지게 된다. 대머리의 가장 큰 원인은 유전이고, 대머리 유전자의 발현에는 남성호르몬인 테스토스테론이 관여하는 것으로 알려져 있다. 유전적으로 대머리가 아니더라도 노화, 스트레스 등으로 탈모현상이 일어나는 경우도 많다. 노화로 인한 탈모는 두피 세포의 감소와 두피 지방질의 축적량 증가에 따른 모공 폐쇄로 산소공급이 감소되고 그로 인해 모공 주위의 모세혈관이 압박을 받아 혈액 순환이 원활하게 이루어지지 않는 데에 그 원인이 있는 것으로 알려져 있다. 또한, 스트레스, 불규칙한 생활, 환경오염 등도 탈모 현상을 일으키는 원인으로 생각되고 있다.
대머리나 탈모 현상으로 당사자들이 느끼는 정신적인 고통은 매우 크나, 현재 개발되어 판매되고 있는 발모제들은 대부분 발모 효과가 일시적이거나 제한적이어서 사용자의 욕구를 충분히 만족시키지 못하고 있다. 어느 정도 효과가 검증되어 사용되고 있는 바르는 발모제인 미녹시딜은 혈관 확장작용에 의해, 경구용인 프로페시아는 피나스테라이드를 주제로 한 남성 호르몬의 활성화 억제작용에 의해 탈모방지 효과가 우수한 제제로 많이 이용되고 있다. 그러나 전술한 발모제들은 탈모의 방지에는 어느 정도의 효과를 나타내지만 발모와 관련된 효과는 미미하다. 즉, 상기 발모제들을 사용하다가 중단할 시에는 다시 탈모 현상이 일어나게 되고, 장기 사용시에는 부작용의 우려가 크며, 또한 장기 사용에 따른 비용문제도 있어 대부분의 환자들이 치료를 포기하고 있는 실정이다.
이에, 본 발명자들은 양수 내 태아 유래 중간엽 줄기세포의 생장 및 수명을 연장하고 컨디션드 배지의 발모 효능을 증진시킬 수 있는 역분화 인자를 발굴하여 컨디션드 배지의 어떠한 성분들이 발모 효능에 유효한지 밝혀내었다. 또한, 본 발명자들은 인 비보(in vivo) 실험을 통해 상기 컨디션드 배지 조성물의 모발의 밀도 및 발모 촉진 효과를 확인함으로써 본 발명을 완성하였다.
본 발명의 목적은 나노그(Nanog)가 과발현된 양수 내 태아 유래 중간엽 줄기세포로부터 인간 성장인자를 생산하는 방법을 제공하기 위한 것이다.
본 발명의 다른 목적은 나노그(Nanog)가 과발현된 양수 내 태아 유래 중간엽 줄기세포로부터 생산된 인간 성장인자가 함유된 컨디션드 배지를 유효성분으로 포함하는 육모 촉진용 조성물의 제조방법을 제공하기 위한 것이다.
본 발명의 또 다른 목적은 나노그(Nanog)가 과발현된 양수 내 태아 유래 중간엽 줄기세포로부터 생산된 인간 성장인자가 함유된 컨디션드 배지를 유효성분으로 포함하는 육모 촉진용 조성물, 및 이의 화장료 조성물 또는 약학적 조성물을 제공하기 위한 것이다.

본 발명은 나노그(Nanog)가 과발현된 양수 내 태아 유래 중간엽 줄기세포로부터 인간 성장인자를 생산하는 방법을 제공하며, 보다 구체적으로 나노그(Nanog)가 과발현된 양수 내 태아 유래 중간엽 줄기세포로부터 bFGF, IGF, Wnt7a 및 PDGF-AA로 이루어진 군으로부터 선택된 어느 하나 이상의 인간 성장인자를 생산하는 방법을 제공하며, 더욱 구체적으로
(a) 산모로부터 얻은 양수에서 태아 유래 세포를 분리하는 단계;
(b) 상기 분리된 태아 유래 세포를 FBS(fetal bovine serum), bFGF(basic fibroblast growth factor), 셀레늄(selenium) 및 아스코르브산(ascorbic acid) 함유 배지에서 계대배양하여 양수 내 태아 유래 중간엽 줄기세포를 수득하는 단계; 및

(d) 상기 수득된 나노그(Nanog)가 과발현된 양수 내 태아 유래 중간엽 줄기세포를 무혈청 배지에서 1 내지 5일간 배양하여 bFGF(basic fibroblast growth factor, 기본 섬유아세포성장인자), IGF(Insulin-like Growth Factor), Wnt7a(Wingless-type MMTV integration site family member 7A) 및 PDGF-AA(Platelet-derived Growth Factor)로 이루어진 군으로부터 선택된 어느 하나 이상의 인간 성장인자가 함유된 컨디션드 배지를 제조하는 단계;를 포함하는 나노그(Nanog)가 과발현된 양수 내 태아 유래 중간엽 줄기세포로부터 bFGF, IGF, Wnt7a 및 PDGF-AA로 이루어진 군으로부터 선택된 어느 하나 이상의 인간 성장인자를 생산하는 방법을 제공한다.
상기 단계 (a)에서 태아 유래 세포는 산모로부터 얻은 양수에 포함되어 있을 수 있다.
본 명세서에서 사용된 용어, "중간엽 줄기세포(mesenchymal stem cells; MSCs)"란 골, 연골, 지방, 골수간질, 근육, 신경 등을 만드는데 원조가 되는 세포로서, 성인에서는 일반적으로 골수에 머물러 있지만 제대혈, 말초혈액, 기타 조직 등에도 존재하며, 이들로부터 수득할 수 있는 세포를 의미한다. 산모로부터 얻은 양수에는 태아의 몸으로부터 나온 여러 가지 화학물질들이 포함되어 인체에 있는 대부분의 세포를 생성할 수 있고 이의 채취가 쉬운 편이다. 또한, 양수에는 이종(heterogenous) 모양의 세포들이 존재하는데, 그 중에서 중간엽 줄기세포의 특징인 섬유아세포(fibroblast)와 같은 모양을 가지는 동종(homogenous)의 중간엽 줄기세포가 존재하는 것을 확인하였다.
본 명세서에서 사용된 용어, "배지(culture medium)"란 인 비트로(in vitro) 상에서 양수 내 태아 유래 세포들의 성장 및 생존을 지지할 수 있게 하는 배지를 의미하고, 양수 내 태아 유래 세포의 배양에 적절한 당업계에서 사용되는 통상의 배지를 모두 포함한다. 배지 및 배양 조건은 세포의 종류에 따라 선택할 수 있다. 배양에 사용되는 배지는 바람직하게는 세포 배양 최소 배지(cell culture minimum medium; CCMM)로, 일반적으로 탄소원, 질소원 및 미량원소 성분을 포함한다. 이러한 세포 배양 최소 배지에는 예를 들어, DMEM(Dulbecco's Modified Eagle's Medium), MEM(Minimal essential Medium), BME(Basal Medium Eagle), RPMI1640, F-10, F-12, αMEM(α Minimal essential Medium), GMEM(Glasgow's Minimal essential Medium) 및 IMEM( Iscove's Modified Dulbecco's Medium) 등이 있다. 또한, 상기 세포 배양 최소 배지는 페니실린(penicillin), 스트렙토마이신(streptomycin) 또는 겐타마이신(gentamicin) 등의 항생제를 포함할 수 있다.
상기 단계 (b)에서 양수 내 태아 유래 중간엽 줄기세포는 양수에서 분리한 세포들을 FBS, bFGF, 셀레늄(selenium) 및 아스코르브산(ascorbic acid)을 함유하는 기본 배지에서 배양함으로써 수득할 수 있으며, 바람직하게는 FBS가 포함된 로우 글루코즈 DMEM 배지에 bFGF, 셀레늄(Selenium), 아스코르브산(ascorbic acid)을 첨가하여 배양함으로써 수득할 수 있고, 보다 바람직하게는 10% FBS 로우 글루코즈 DMEM 배지에 bFGF 4ng/ml, 셀레늄(Selenium) 5ng/ml, 아스코르브산(ascorbic acid) 50㎍/ml, 1% L-글루타민 및 1% 페니실린-스트렙토마이신을 포함할 수 있으나, 이에 한정되지 않는다.
상기 단계 (c)에서 나노그(Nanog)는 레트로바이러스 벡터를 이용하여 도입하는 것일 수 있으며, 바람직하게는 pMXs 바이러스 벡터를 이용할 수 있으나, 이에 한정되지 않는다.
상기 단계 (c)에서 나노그(Nanog)는 서열번호 1로 표시되는 염기서열로 이루어지는 유전자일 수 있으며, 인간(Homo sapiens) 유래의 나노그(Nanog) 유전자인 것이 바람직하나, 이에 한정되지 않는다.
본 발명의 일 실시예에서, 중간엽 줄기세포에 나노그(Nanog)를 도입하고 배양하는 단계에 의해서 중간엽 줄기세포의 성장 및 증식이 증가하는 것을 확인하였다.
본 명세서에서 사용된 용어, "나노그(Nanog)"란 역분화 인자로, 역분화 인자는 2006년 야마나카 교수팀에 의해 도입된 개념인 역분화(Reprogramming)에서 비롯되었다. 성체의 모든 조직은 정상 발달 과정을 거치면서 분화되지 않은 상태에서 점차적으로 분화되어 각 기증이 전문화된 세포로 변화한다. 그 중 수정란의 세포들은 전능성(Totipotent)을 가지고 있고 이 후 발달 단계가 진행되면서 배반포가 되면 내부 세포괴(inner cell mass) 세포들과 바깥쪽 세포들로 구분이 가능하며, 내부 세포괴 세포들은 배아 체세포와 생식세포로 발생할 수 있으며 이를 만능성(pluripotent)라 부른다. 이 내부 세포괴 세포는 배아 줄기세포로도 불리며 만능성 특이적 유전자(역분화 인자)를 발현하는데, 그 대표적인 예가 Oct4, Sox2, 나노그(Nanog), Lin28 등이다. 역분화는 체세포에 이러한 만능성 특이적 유전자 발현을 유도하여 배아 줄기세포와 유사한 성질로 되돌리는 기술이다. 이러한 배경에서 본 발명은 다양한 역분화 인자 중 나노그(Nanog)를 단독적으로 사용하였으며, 그 이유로는 1) 여러 외래 유전자 도입 시 세포 특성 변성의 우려, 2) 바이러스를 통한 여러 인자 도입 시 성공적인 세포주 구축의 어려움, 3) 각 만능성 특이적 유전자를 도입한 후 세포주의 유지가 지속되는 유전자를 조사한 결과 나노그(Nanog)가 단독 도입되었을 시 지속적 유지가 가능한 점(도 15 참조)이 있다.
또한, 본 발명은 상기 단계 (d)에 의해 제조된 컨디션드 배지, 보다 구체적으로 나노그(Nanog)가 과발현된 양수 내 태아 유래 중간엽 줄기세포를 배양하여 제조한 컨디션드 배지, 더욱 구체적으로 나노그(Nanog)가 과발현된 양수 내 태아 유래 중간엽 줄기세포를 배양한 bFGF(basic fibroblast growth factor, 기본 섬유아세포성장인자), IGF(Insulin-like Growth Factor), Wnt7a(Wingless-type MMTV integration site family member 7A) 및 PDGF-AA(Platelet-derived Growth Factor)로 이루어진 군으로부터 선택된 어느 하나 이상의 인간 성장인자가 함유된 컨디션드 배지를 제공할 수 있다.
본 명세서에서 사용된 용어, "컨디션드 배지(conditioned medium)" 란 세포를 액체 현탁 배양하여 세포분열 최성기인 대수 성장기에 도달했을 때 분열세포를 원심분리 또는 여과하여 제거하고 배양액만 채취하여 이를 배양기질에 혼합한 배지를 말한다. 이는 분열중인 세포로부터 배지 내에서 추출되어 나오는 미지의 성장요소(growth factor)를 이용하는 것으로서, 저밀도의 세포 플레이팅이나 원형질체 배양에 많이 이용된다.
상기 단계 (d)에 의해 제조된 컨디션드 배지는 태아 유래 중간엽 줄기세포를 배양한 배지에서 태아 유래 중간엽 줄기세포를 제거한 배지로서, 태아 유래 중간엽 줄기세포에서 유래한 성장인자 등의 물질이 풍부하게 함유되어 있는 배지를 말한다.
또한, 본 발명은 나노그(Nanog)가 과발현된 양수 내 태아 유래 중간엽 줄기세포를 배양하여 제조한 컨디션드 배지를 유효성분으로 포함하는 육모 촉진용 조성물의 제조방법을 제공하며, 보다 구체적으로
(a) 산모로부터 얻은 양수에서 태아 유래 세포를 분리하는 단계;
(b) 상기 분리된 태아 유래 세포를 FBS(fetal bovine serum), bFGF(basic fibroblast growth factor), 셀레늄(selenium) 및 아스코르브산(ascorbic acid) 함유 배지에서 계대배양하여 양수 내 태아 유래 중간엽 줄기세포를 수득하는 단계;
(c) 상기 수득된 양수 내 태아 유래 중간엽 줄기세포에 나노그(Nanog)를 도입하여 나노그(Nanog)가 과발현된 양수 내 태아 유래 중간엽 줄기세포를 수득하는 단계;
(d) 상기 수득된 나노그(Nanog)가 과발현된 양수 내 태아 유래 중간엽 줄기세포를 무혈청 배지에서 1 내지 5일간 배양하여 bFGF(basic fibroblast growth factor, 기본 섬유아세포성장인자), IGF(Insulin-like Growth Factor), Wnt7a(Wingless-type MMTV integration site family member 7A) 및 PDGF-AA(Platelet-derived Growth Factor)로 이루어진 군으로부터 선택된 어느 하나 이상의 인간 성장인자가 함유된 컨디션드 배지를 제조하는 단계; 및
(e) 상기 제조한 컨디션드 배지를 유효성분으로 포함하는 조성물을 제조하는 단계;를 포함하는, 나노그(Nanog)가 과발현된 양수 내 태아 유래 중간엽 줄기세포로부터 생산된 bFGF, IGF, Wnt7a 및 PDGF-AA로 이루어진 군으로부터 선택된 어느 하나 이상의 인간 성장인자가 함유된 컨디션드 배지를 유효성분으로 포함하는 육모 촉진용 조성물의 제조방법을 제공한다.
본 명세서에서 사용된 용어, "육모 촉진"이란, 모발 성장 효과 및 모발 성장 시기의 촉진에 따른 모발 성장 촉진을 의미한다.
상기 단계 (d)에 의해 제조된 컨디션드 배지는 bFGF(basic Fibroblast Growth Factor), IGF(Insulin-like Growth Factor), PDGF-AA(Platelet-derived Growth Factor) 또는 Wnt7a(Wingless-type MMTV integration site family member 7A), 및 이들의 2 이상의 조합의 인간 성장인자를 함유하는 것이 바람직하나, 이에 한정되지 않는다. 상기 bFGF(Du Cros et al. Fibroblast growth factor and epidermal growth factor in hair development. J Invest Dermalto. 101:106S-113S, 1993), IGF(Nicole et al. PDGF isoforms induce and maintain anagen phase of murine hair follicles. Journal of dermatological science 43(2):105-15, 2006) 또는 Wnt7a(Kishimoto et al. Wnt signaling maintains the hair-inducing activity of the dermal papilla. Genes&Development 14:1191-1185, 2000)는 모발 성장을 촉진하는 성장인자로 알려져 있다.
또한, 상기 단계 (d)에 의해 제조된 컨디션드 배지는 Apo-1(apoptosis antigen 1)/Fas, 표피세포 성장인자(EGF, Epidermal growth factor), IP-10(Interferon-γ inducible protein-10), 렙틴(Leptin), MIP4(Macrophage inflammatory protein 4), MMP3(matrix metalloproteinase 3), 란테스(Rantes), 인터페론γ(IFNγ, Interferon γ), 인간형질전환 성장인자β(TGFβ, Transforming growth factor β), 종양괴사인자α(TNFα, Tumor necrosis factors α), 종양괴사인자 수용체Ⅰ(TNFRⅠ, Tumor necrosis factors receptor Ⅰ), 종양괴사인자 수용체Ⅱ(TNFRⅡ, Tumor necrosis factors receptor Ⅱ), 세포부착인자 1(ICAM1, Intercellular Adhesion Molecule 1), 혈관세포부착인자 1(VCAM1, vascular cell adhesion molecule 1), 맥관내피세포 성장인자(vascular endothelial growth factor), 인터루킨-1β(IL-1β, Interleukin-1β), 인터루킨-1 수용체α(IL-1Rα, Interleukin-1 receptorα), 인터루킨-2, 인터루킨-3, 인터루킨-4, 인터루킨-5, 인터루킨-6, 인터루킨-6R, 인터루킨-7, 인터루킨-8, 인터루킨-12, 인터루킨-15 등의 성장인자를 추가로 더 포함할 수 있으나, 이러한 성장인자들의 모발 성장 촉진 효과는 증명된 전례가 없는 것으로 알려져 있다.
따라서, 상기 컨디션드 배지를 유효성분으로 포함하는 육모 촉진용 조성물의 제조방법을 이용하면, 모발 성장 효과가 있는 것으로 알려진 bFGF, IGF, Wnt7a, PDFG-AA, 또는 이들의 2이상의 조합의 성장인자의 발현량이 현저히 증가함으로써 모발 성장 증진 효과가 더욱 증대될 수 있다.
본 발명의 일 실시예에서, 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포로부터 수득한 컨디션드 배지를 처리한 그룹에서 모낭 조직이 더 많이 생성되고, 조직적 분석에서 성장기 단계 구별 시 성장기 후기 단계의 형태를 나타냄을 확인하였으며 모발 성장단계에서 많이 발현하는 ALP(amplified length polymorphism), LEF, 베르시칸(Versican) 및 Hey의 mRNA가 더 많이 발현하는 것을 확인하였다.
또한, 본 발명은 나노그(Nanog)가 과발현된 양수 내 태아 유래 중간엽 줄기세포를 배양한 컨디션드 배지를 유효성분으로 포함하는 육모 촉진용 조성물, 보다 구체적으로 나노그(Nanog)가 과발현된 양수 내 태아 유래 중간엽 줄기세포로부터 생산된 bFGF, IGF, Wnt7a 및 PDFG-AA로 이루어진 군으로부터 선택된 어느 하나 이상의 인간 성장인자가 함유된 컨디션드 배지를 유효성분으로 포함하는 육모 촉진용 조성물, 더욱 구체적으로 상기 육모 촉진용 조성물의 제조방법에 의해 제조된 육모 촉진용 조성물을 제공한다.
본 발명의 일 실시예에서, 나노그(Nanog)가 과발현된 양수 유래 중간엽 줄기세포를 배양한 컨디션드 배지는 배지 내 모발 성장 촉진 인자들의 발현량이 현저하게 증가하고, 이를 마우스에 처리하였을 경우 빠른 모발 성장을 나타내어 육모 촉진, 모발 재생 및 탈모 방지 효과를 확인하였다.
또한, 본 발명은 상기 육모 촉진용 조성물을 포함하는 육모 촉진용 화장료 조성물을 제공한다.
상기 화장료 조성물은 육모 촉진, 모발 재생 및 탈모 방지 효과를 갖는 모발 화장품에 다양하게 이용될 수 있다.
상기 화장료 조성물은 전체 화장료 조성물 100 중량부에 대해 0.001 내지 10 중량부를 포함할 수 있다.
상기 화장료 조성물은 지방 물질, 유기용매, 용해제, 농축제, 겔화제, 연화제, 항산화제, 현탁화제, 안정화제, 발포제(foaming agent), 방향제, 계면활성제, 물, 이온형 또는 비이온형 유화제, 충전제, 금속이온봉쇄제, 킬레이트화제, 보존제, 비타민, 차단제, 습윤화제, 필수 오일, 염료, 안료, 친수성 또는 친유성 활성제, 지질소낭 또는 화장품에 통상적으로 사용되는 임의의 다른 성분과 같은 화장품학 또는 피부과학 분야에서 통상적으로 사용되는 보조제를 함유할 수 있다. 상기 보조제는 화장품학 또는 피부과학 분야에서 일반적으로 사용되는 양으로 도입된다.
상기 화장료 조성물의 외형은 화장품학 또는 피부과학적으로 허용 가능한 매질 또는 기제를 함유한다. 이는 국소적용에 적합한 모든 제형으로, 예를 들면, 용액, 겔, 고체, 반죽 무수 생성물, 수상에 유상을 분산시켜 얻은 에멀젼, 현탁액, 마이크로에멀젼, 마이크로캡슐, 미세과립구 또는, 이온형(리포좀) 및 비이온형의 소낭 분산제의 형태로, 또는 크림, 스킨, 로션, 파우더, 연고, 스프레이 또는 콘실 스틱의 형태로 제공될 수 있다. 이들 조성물은 당해 분야의 통상적인 방법에 따라 제조될 수 있다. 본 발명에 따른 조성물은 또한 포말(foam)의 형태로 또는 압축된 추진제를 더 함유한 에어로졸 조성물의 형태로도 사용될 수 있다.
본 발명의 화장료 조성물은 그 제형에 있어서 특별히 한정되는 바가 없으며, 예를 들면, 유연화장수, 수렴화장수, 영양화장수, 영양크림, 마사지크림, 에센스, 아이크림, 아이에센스, 클렌징크림, 클렌징폼, 클렌징워터, 팩, 파우더, 바디로션, 바디크림, 바디오일, 바디에센스, 헤어토닉, 헤어컨디셔너, 헤어에센스, 헤어로션, 헤어영양로션, 헤어샴푸, 헤어린스, 헤어트리트먼트, 헤어크림, 헤어영양크림, 헤어모이스처크림, 헤어맛사지크림, 헤어왁스, 헤어 에어로졸, 헤어팩, 헤어영양팩, 헤어비누, 헤어클렌징폼, 머릿기름, 모발건조제, 모발보존처리제, 모발염색제, 모발용 웨이브제, 모발탈색제, 헤어겔, 헤어글레이즈, 헤어드레싱어, 헤어래커, 헤어모이스처라이저, 헤어무스 또는 헤어스프레이 등의 화장품으로 제형화될 수 있다.
상술한 바와 같은 화장료 조성물은 피부에 바르는 형태로 적용될 수 있고, 마이크로 니들 등을 이용하여 피부 내부로 흡수되는 형태로 적용될 수도 있다.
또한, 본 발명은 상기 육모 촉진용 조성물을 포함하는 육모 촉진용 약학적 조성물을 제공한다.
상기 약학적 조성물은 육모 촉진, 모발 재생 및 탈모 방지 효과를 갖는 모발 화장품에 다양하게 이용될 수 있다.
상기 약학적 조성물은 전체 약학적 조성물 100 중량부에 대해 0.001 내지 10 중량부를 포함할 수 있다.
상기 약학적 조성물은 육모 촉진용 정제, 캡슐제, 산제, 과립제, 주사제, 젤, 에멀젼, 시럽, 에어로졸, 패취, 분무제, 크림, 연고제, 경고제, 로션제, 리니멘트제, 파스타제 또는 카타플라스마제 형태의 약학 조성물로 제조하여 사용할 수 있으나, 이에 한정하는 것은 아니다.
상기 약학적 조성물을 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제된다. 경구투여를 위한 고형 제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형 제제는 하나 이상의 화합물에 적어도 하나 이상의 부형제 예를 들면, 전분, 탄산칼슘, 수크로오스(sucrose) 또는 락토오스(lactose), 젤라틴 등을 섞어 조제된다. 또한 단순한 부형제 이외에 스테아린산 마그네슘, 탈크 등과 같은 윤활제들도 사용된다. 경구투여를 위한 액상 제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데 흔히 사용되는 단순 희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수성 용제, 현탁제, 유제, 동결건조 제제, 좌제가 포함된다. 비수성 용제, 현탁 용제로는 프로필렌글리콜(propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세로젤라틴 등이 사용될 수 있다.
상기 약학적 조성물의 투여 형태는 이들의 약학적 허용 가능한 염, 단독 또는 타 약학적 활성 화합물과 결합뿐만 아니라 적당한 집합의 형태로 사용될 수 있다. 상기 염으로는 약학적으로 허용되는 것이면 특별히 한정되지 않으며, 예를 들어 염산, 황산, 질산, 인산, 불화수소산, 브롬화수소산, 포름산 아세트산, 타르타르산, 젖산, 시트르산, 푸마르산, 말레산, 숙신산, 메탄술폰산, 벤젠술폰산, 톨루엔술폰산, 나프탈렌술폰산 등을 사용할 수 있다.
상기 약학적 조성물은 목적하는 바에 따라 비경구 투여하거나 경구 투여할 수 있으며, 하루에 체중 1 ㎏당 0.1~500 ㎎, 1~100 ㎎의 양으로 투여되도록 1 내지 수회에 나누어 투여할 수 있다. 특정 환자에 대한 투여용량은 환자의 체중, 연령, 성별, 건강 상태, 식이, 투여 시간, 투여 방법, 배설률, 질환의 중증도 등에 따라 변화될 수 있다.
상기 약학적 조성물은 쥐, 생쥐, 가축, 인간 등의 포유동물에 비경구, 경구 등의 다양한 경로로 투여될 수 있으며, 투여의 모든 방식은 예상될 수 있는데, 예를 들면, 경구, 직장 또는 정맥, 근육, 피하, 자궁내 경막 또는 뇌혈관내(intracerebroventricular) 주사에 의해 투여될 수 있다.
중간엽 줄기세포의 컨디션드 배지 생산에 있어 주된 한계점은 줄기세포의 노화와 크게 연관이 있다. 세포의 노화는 세포 성장 저조를 유발하고 분비하는 성장인자 및 사이토카인(cytokine)의 양을 감소시키며 세포의 사멸을 야기한다. 또한, 세포 유래 컨디션드 배지의 양 및 질적 손실을 유발하고 균일한 조성을 유지하기 어렵게 하며, 이는 결국 컨디션드 배지 및 이를 유효성분으로 포함하는 조성물의 대량생산 및 산업화 시 어려움을 초래한다. 이에, 본 발명자들은 중간엽 줄기세포 배양에 역분화 기술을 도입하였다. 선행연구에 의하면, 중간엽 줄기세포에서 발현하는 만능성 인자들(Pluripotent factors)의 발현에 따라 줄기세포능의 양상에 변화가 일어난다고 알려져 있다. 이에 착안하여 양수 유래 중간엽 줄기세포에 다양한 만능성 인자들인 Oct4, 나노그(Nanog), Lin28을 도입하였고, 그 중 나노그(Nanog)를 도입한 양수 유래 중간엽 줄기세포는 세포주를 확립 가능하며 지속적인 배양이 가능함에 반해 타 유전자의 도입은 세포 사멸이 유래되었으며 지속적인 세포 배양이 어려움을 알 수 있었다(도 15 참조). 더욱이 나노그(Nanog)를 도입한 양수 줄기세포에서 세포 성장의 증대 및 줄기세포능의 보존 효과가 기존 양수 줄기세포보다 향상되었음을 확인하였다.
본 발명의 일 실시예에서, 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포 수는 양수 유래 줄기세포에 비해 현저히 많아(도 4), 나노그(Nanog)가 양수 유래 중간엽 줄기세포의 세포 성장 증진에 긍정적인 영향을 미치는 것을 확인하였다. 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포는 양수 유래 중간엽 줄기세포보다 β-갈락토시다아제를 덜 발현하였으며(도 5 좌측), p53 및 p21의 발현량이 감소하였다(도 5 우측). 따라서, 나노그(Nanog)의 도입으로 인하여 양수 유래 중간엽 줄기세포의 수명 연장 효과가 있음을 확인하였다. 또한, 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포에서 만능성 특이적 마커 Oct4 및 Sox2의 발현이 향상되어(도 7), 나노그(Nanog)의 도입에도 양수 유래 중간엽 줄기세포의 줄기세포능이 보존됨을 확인하였다.
본 발명의 일 실시예에서, 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포에서 모발 성장을 촉진하는 성장인자인 bFGF(basic fibroblast growth factor), IGF(Insulin-like Growth Factor), Wnt7a 및 PDGF-AA(Platelet-derived Growth Factor)의 발현이 양수 유래 중간엽 줄기세포보다 증가하였음을 확인하였으므로(도 8 및 도 9), 나노그(Nanog)의 도입으로 인해 양수 유래 중간엽 줄기세포에서 모발 성장인자들의 분비가 촉진됨을 확인하였다.
본 발명의 일 실시예에서, 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포로부터 수득한 컨디션드 배지를 처리한 그룹에서 모낭 조직이 더 많이 생성되는 것을 관찰하였다(도 12). 또한, 조직적 분석에서 성장기 단계를 구별하였을 때, 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포의 컨디션드 배지를 처리한 그룹이 성장기 후기 단계의 형태를 나타냄을 확인하였으며(도 13), 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포이 컨디션드 배지를 처리한 그룹에서 모발 성장단계에서 많이 발현하는 ALP(amplified length polymorphism), LEF, 베르시칸(Versican) 및 Hey의 mRNA가 더 많이 발현하는 것을 확인하였다(도 14). 따라서, 나노그(Nanog)를 도입한 양수 유래 중간엽 줄기세포 컨디션드 배지를 적용 시 체외 및 체내에서 모발 성장 촉진 효과를 나타냄을 확인하였다.
본 발명은 양수 내 태아 유래 중간엽 줄기세포에 역분화 인자인 나노그(Nanog)를 도입하여 과발현함으로써 중간엽 줄기세포의 성장, 줄기세포능, 수명을 증대하고, 이로부터 분비되는 성장인자의 발현을 증대하였다. 또한, 나노그(Nanog)가 도입된 양수 내 태아 유래 중간엽 줄기세포를 배양한 컨디션드 배지는 모발 성장 촉진 효과를 나타내므로, 육모 촉진용 화장료 조성물 및 약학적 조성물로써 활용이 가능하다.

이하, 본 발명의 이해를 돕기 위하여 실시예를 들어 상세하게 설명하기로 한다. 다만 하기의 실시예는 본 발명의 내용을 예시하는 것일 뿐 본 발명의 범위가 하기 실시예에 한정되는 것은 아니다. 본 발명의 실시예는 당업계에서 평균적인 지식을 가진 자에게 본 발명을 보다 완전하게 설명하기 위해 제공되는 것이다.

1> 양수 유래

줄기세포 분리 및 배양
산모로부터 얻은 양수 내에서 분리한 태아 유래 세포를 10% FBS(Fetal bovine serum, 소태아혈청), 1% 페니실린/스트렙토마이신, 1% L-글루타민, bFGF(basic fibroblast growth factor, 기본 섬유아세포성장인자) 4ng/ml, 셀레늄(Selenium) 5ng/ml, 아스코르브산(Ascorbic acid) 50㎍/ml이 포함된 로우 글루코오스(low glucose) DMEM(Dulbecco's Modified Eagle's Medium) 배지에서 계대배양하여 양수 내 태아 유래 중간엽 줄기세포를 수득하였다. 이러한 배지 조건은 bFGF 처리, bFGF와 Selenium 처리, bFGF와 아스코르브산(Ascorbic acid) 처리 조건보다 더 향상된 양수 유래 중간엽 줄기세포 성장 효과를 보인다 (도 1).
양수 유래 중간엽 줄기세포의 성장 효과는 2일 또는 4일 동안 배양한 후 성장률을 조사하였으며, 정량화는 크리스탈 바이올렛 염색(crystal violet staining)을 통해 진행하였다. 크리스탈 바이올렛 염색 기법은 세포를 동일한 수로 준비한 뒤 2일 또는 4일 간격으로 10% 포르말린 고정(formalin fixation)을 진행한 후 크리스탈 바이올렛 염색을 20분간 진행하였다. 이를 10% 아세트산(acetic acid)으로 탈색(destaining)하여 아세트산의 595nm 흡광도(absorbance)를 Ultrospec 2100pro 기기를 이용하여 측정한 후 정량 분석하였다.
수득한 양수 내 태아 유래 중간엽 줄기세포는 위와 동일한 조건의 DMEM 배지에서 배양하였다.

레트로바이러스 벡터 시스템(Retroviral vector system)을 이용해 상기 실시예 1에서 수득한 양수 유래 중간엽 줄기세포에 나노그(Nanog) 유전자를 도입하여 나노그(Nanog)의 과발현을 유도함으로써 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포주들(#1, #2, #3)를 제조하였다(도 2).
구체적으로, pBS-Nanog 벡터(Yamanaka lab's plasmid stock #A4, JAPAN)에서 제한효소(restriction enzyme)를 이용하여 나노그 유전자(NCBI GenBank Accession number NM_024865.3)(서열번호 1)를 분리하였다. 분리한 나노그 유전자를 pMXs 벡터(Cell biolab, JAPAN)에 T4 연결효소(ligase)로 라이게이션(ligation)하여 pMXs-Nanog를 제작하였다. pMXs-Nanog 벡터를 293GPG cell로 형질도입 시약(transfection reagent)을 이용하여 6시간 동안 형질도입(transfection)하였다. 72시간 동안 나노그 유전자를 가진 바이러스를 생산하고, 상층액을 분리한 후, 2000rpm에서 10분간 원심 분리한 뒤 그 상층액을 0.45um 필터에 필터링하였다. 이 바이러스를 80% confluence 상태의 양수 유래 중간엽 줄기세포에 6시간 동안 처리하여 세포 내로 감염(Infection)시켰다.
제조한 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포는 실시예 1의 배지에서 배양하였을 때, 지속적이고 정상적으로 세포가 성장하였다.
< 실시예 3> 나노그(Nanog)가 도입된 양수 유래 중간엽 줄기세포의 나노그(Nanog) 발현 검정
실시예 2에서 제조한 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포주들(#1, #2, #3)에서 나노그(Nanog)의 과발현이 제대로 일어나고 있는지 확인하기 위해, 나노그(Nanog) mRNA에 대한 RT-PCR(Reverse transcription polymerase chain reaction) 및 면역형광 염색(immunofluorescence staining)을 수행하였다.
RT-PCR 수행을 위해, 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포를 트리졸(TRIzol) 시약(Invitrogen, USA)을 이용하여 제조자의 지시에 따라 총 RNA를 분리하였다. 분리된 총 RNA 500ug을 cDNA 제조 믹스(Bioneer)를 이용하여 cDNA로 역전사하였다. 실험 튜브들을 45℃에서 60 분간 정치한 후, 95℃에서 5분간 정치하였고, 이후 사용시까지 -20℃에서 보관되었다. cDNA는 Taq 합성효소(Promega)와 나노그(Nanog) mRNA에 특이적인 프라이머(Exo-Nanog foward: 5'-GCTTGGATACACGCCGC-3' (서열번호 2); 및 Exo-Nanog reverse: 5'-GATTGTTCCAGGATTGGGTG-3' (서열번호 3))를 통해 증폭되었다. RT-PCR은 94℃ 20초, 60℃ 30초, 및 72℃로 2분의 사이클을 35회 반복한 후, 최종 합성을 위해 72℃로 10분간 진행하였다. PCR 결과물을 1% 아가로즈 젤에 전기영동하고 이를 분석하였다.
면역형광 염색을 위해, 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포를 PBS로 세척한 뒤 4% 파라포름알데히드로 1시간 동안 고정한 후 0.5% PBST(0.5% Triton-100 첨가된 PBS(Phosphate-buffered saline))로 5분씩 3번 세척하였다. 3% fatal bovine 혈청이 포함된 PBST로 1시간 동안 블로킹(blocking)한 후 나노그(Nanog) 수용체를 탐지하는 항체(AF276, R&D)를 1:50의 비율로 1시간 동안 처리하였다. 그 후 0.5% PBST로 5분씩 3번 세척하고 형광이 달린 2차 항체(Alexa Fluor 488 goat anti-human IgG, #A11013, invitrogen, USA)를 1:200의 비율로 희석하여 1시간 동안 처리하였다. 5분 간 핵 염색을 위해 DAPI(4',6-diamidino-2-phenylindole) 용액을 1:1000으로 처리한 후 앞에서와 동일하게 PBST로 3번 세척하였다. 이후 샘플을 형광현미경(Olympus DP70)으로 찍어 이미지화하였다.
그 결과, 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포가 mRNA 수준에서의 외인성(exogenous) 나노그(Nanog) 유전자 발현량과 단백질 수준에서의 나노그(Nanog) 발현량이 양수 유래 중간엽 줄기세포보다 현저히 향상되어, 과발현되었음을 확인하였다(도 3).
<실시예 4> 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포의 세포 성장 증진 효과 검정
실시예 2에서 제조한 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포주들(#1, #2, #3)과 나노그(Nanog) 도입 없이 실시예 1의 배지에서 지속적으로 배양한 양수 유래 줄기세포의 성장률을 비교 분석하였다.
실시예 1의 배지를 이용하여 줄기세포를 13번 계대 배양함과 동시에 각각의 세포주들의 세포 수를 3일 간격으로 Counting chamber(MARIENFELD, Germany)를 이용하여 측정, 비교하였다.
그 결과, 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포 모두 양수 유래 줄기세포에 비해 세포 수가 현저히 많은 것을 확인하였다(도 4). 이는 나노그(Nanog)가 양수 유래 중간엽 줄기세포의 세포 성장 증진에 긍정적인 영향을 미치는 것을 시사하였다.
<실시예 5> 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포의 수명 연장 효과 검정
나노그(Nanog) 도입 양수 유래 중간엽 줄기세포의 수명을 실험하기 위해, 지속적인 계대배양으로 인한 세포 사멸 정도를 β-갈락토시다아제 염색(β-Galactosidase staining) 및 세포 사멸과 관련된 단백질인 p53 및 p21의 mRNA 발현량으로 분석하였다.
β-갈락토시다아제 염색을 위해, 실시예 1의 배지에서 35번 계대배양한 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포를 6웰 플레이트에 5 x 104 cells/웰로 분주하고 하룻밤 동안 부착시킨 후 PBS로 세척하고 고정하였다. β-갈락토시다아제 염색 키트(Cell Signaling Technology, Beverly, MA)의 프로토콜에 따라 pH 6.0의 X-gal 발색 기질(chromogenic substrate)을 37℃에서 하룻밤 동안 배양한 후 현미경(Olympus DP70)으로 100배 확대하여 색깔 변화를 확인하였다.
세포 사멸과 관련된 단백질인 p53 및 p21의 mRNA 발현량을 측정하기 위해, 실시예 3과 동일한 방법으로 35번 계대배양한 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포로부터 총 RNA를 추출하였으며 이로부터 cDNA를 증폭하였다. cDNA는 Taq 합성효소(Promega)와 p53, p21 및 GAPDH(glyceraldehyde-3-phosphate dehydrogenase)의 mRNA에 각각 특이적인 프라이머를 통해 증폭되었다. p53 특이적 프라이머는 p53 forward: 5'-CCTCACCATCATCACACTGG-3' (서열번호 4) 및 p53 reverse: 5'-TTATGGCGGGAGGTAGACTG-3' (서열번호 5)를 사용하였고, p21 특이적 프라이머는 p21 forward: 5'-GGAAGACCATGTGGACCTGT-3' (서열번호 6) 및 p21 reverse: 5'-AGGCAGAAGATGTAGAGCGG-3' (서열번호 7)를 사용하였으며, GAPDH 특이적 프라이머는 GAPDH forward: 5'-GTGGTCTCCTCTGACTTCAACA-3' (서열번호 8) 및 GAPDG reverse: 5'-CTCTTCCTCTTGTGCTCTTGCT-3' (서열번호 9)를 사용하였다. PCR 결과물을 1% 아가로즈 젤에 전기영동하여, GAPDH mRNA를 기준으로 하여 상대적인 mRNA 발현량을 Quantity One 소프트웨어를 이용하여 정량화하였다.
그 결과, 나노그(Nanog) 도입 후 실시예 1의 배지에서 지속적으로 계대 배양한 양수 유래 중간엽 줄기세포가 상대적으로 나노그(Nanog)를 도입하지 않고 실시예 1과 동일한 배지에서 계속적으로 계대 배양을 한 양수 유래 중간엽 줄기세포보다 β-갈락토시다아제를 덜 발현하였으며(도 5 좌측), p53 및 p21의 발현량이 감소하는 것을 확인하였다(도 5 우측). 따라서, 나노그(Nanog)의 도입으로 인하여 양수 유래 중간엽 줄기세포의 수명 연장 효과가 있음을 확인하였다.
< 실시예 6> 나노그 ( Nanog ) 도입 양수 유래 중간엽 줄기세포의 고유 특성 변화 유무 확인
나노그(Nanog) 도입으로 인한 줄기세포 고유의 발현 마커(marker)의 발현 변화가 생겼는지에 대한 여부를 확인하기 위하여 피브로넥틴(Fibronectin), MMP1, Snail, Slug 발현 양상을 RT-PCR을 통하여 확인하였다.
RT-PCR은 실시예3 과 같은 방식으로 RNA 추출 및 PCR을 진행하였으며 각각의 프라이머는 다음과 같다. 피브로넥틴(Fibronectin) 특이적 프라이머는 Fibronectin forward: 5'-GACGACTCCCTTTTCTCCTCTT-3' (서열번호 10) 및 Fibronectin reverse: 5'-TGAGTTCTGTGCTGCTACCTTC-3' (서열번호 11)를 사용하였고, MMP1 특이적 프라이머는 MMP1 forward: 5'-TTGAGAAAGCCTTCCAACTCTG-3' (서열번호 12) 및 MMP1 reverse : 5'-CCGCAACACGATGTAAGTTGTA-3' (서열번호 13)를 사용하였으며, Snail 특이적 프라이머는 Snail forward: 5'-CTCCTTCGTCCTTCTCCTCTACTT-3' (서열번호 14) 및 Snail reverse: 5'-TCTTGACATCTGAGTGGGTCTG-3' (서열번호 15)를 사용하였고, Slug 특이적 프라이머는 Slug forward: 5'-GACCCTGGTTGCTTCAAGGACA-3' (서열번호 16) 및 Slug reverse: 5'-TTGTCATTTGGCTTCGGAGTGA-3' (서열번호 17)를 사용하였다.
RT-PCR의 결과에 따르면, 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포는 양수 유래 중간엽 줄기세포와 중간엽 줄기세포 특이적 마커(marker) 발현에 차이가 없음을 확인할 수 있다(도 6). 따라서, 나노그(Nanog)의 도입은 양수 유래 중간엽 줄기세포의 고유 특성에 큰 영향을 미치지 않았다.
<실시예 7> 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포의 만능성 유전자 발현 검정
다능성 줄기세포는 만능성 특이적 마커인 Oct4 및 Sox2이 소량 발현되는 것으로 알려져 있다(Riekstina et al. Embryonic stem cell marker expression pattern in human mesenchymal stem cells derived from bone marrow, adipose tissue, heart and dermis. Stem cell rep 5(4):378-386. (2009)). 나노그(Nanog) 도입으로 인한 줄기세포의 만능성 유전자 발현 여부를 확인하기 위해, 실시예 2에서 제조한 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포의 Oct4 및 Sox2 mRNA 발현량을 qRT-PCR(quantitative reverse transcription polymerase chain reaction)을 이용하여 분석하였다.
qRT-PCR은 위의 RT-PCR시 RNA prep과 동일하게 RNA를 수득하였으며 동일하게 cDNA를 제작하였다. 이렇게 준비된 cDNA는 CFX-96 PCR system을 통해 qRT-PCR을 수행하였으며, cDNA는 Oct4 및 Sox2의 mRNA에 특이적인 프라이머를 이용하여 증폭하였다. Oct4 특이적 프라이머는 Oct4 forward: 5'-GACAGGGGGAGGGGAGGAGCTAGG-3' (서열번호 18) 및 Oct4 reverse: 5'-CTTCCCTCCAACCAGTTGCCCCAAAC-3' (서열번호 19) 를 사용하였고, Sox2 특이적 프라이머는 Sox2 forward: 5'-ACCAATCCCATCCACACTCACGCA-3' (서열번호 20) 및 Sox2 reverse: 5'-GCAAACTTCCTGCAAAGCTCCTACCG-3' (서열번호 21)를 사용하였다. 표적 mRNA의 상대적 정량은 CT(comparative threshold cycle) 방법(Johnson MR., et al., Anal Biochem 2000; 278: 175-184)으로 분석하였다.
그 결과, 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포에서 만능성 특이적 마커 Oct4 및 Sox2의 발현이 더욱 향상됨을 확인하였다(도 7). 따라서, 나노그(Nanog)의 도입으로 인하여 양수 유래 중간엽 줄기세포의 만능성 특이 유전자의 발현이 증가되었다.
< 실시예 8> 나노그(Nanog)의 과발현으로 인한 양수 유래 중간엽 줄기세포의 모발 성장인자 발현 검정
나노그(Nanog) 도입 양수 유래 중간엽 줄기세포에서의 모발 성장인자 발현 여부를 확인하기 위해, 모발과 관련된 성장인자인 bFGF(basic fibroblast growth factor), IGF(Insulin-like Growth Factor), Wnt7a 및 PDGF-AA(Platelet-derived Growth Factor)의 mRNA 발현량 및 단백질 발현량을 qRT-PCR 및 웨스턴 블롯(Western blot)을 이용하여 분석하였다.
qRT-PCR은 실시예 7과 동일한 방법으로 수행하였으며, cDNA는 bFGF, IGF, Wnt7a 및 PDGF-AA의 mRNA에 특이적인 프라이머를 이용하여 증폭하였다. bFGF 특이적 프라이머는 bFGF forward: 5'-CAGATTAGCGGACGCGGTGC-3' (서열번호 22) 및 bFGF reverse: 5'-TCACGGATGGGTGTCTCCGC-3' (서열번호 23)를 사용하였고, IGF 특이적 프라이머는 IGF forward: 5'-CCATGTCCTCCTCGCATCTCTTCT-3' (서열번호 24) 및 IGF reverse: 5'-CCATACCCTGTGGGCTTGTTGAA-3' (서열번호 25)를 사용하였으며, Wnt7a 특이적 프라이머는 Wnt7a forward: 5'-TCTTTCTCAGCCTGGGCATGGT-3' (서열번호 26) 및 Wnt7a reverse: 5'-TCCTATGACGATGATGGCGTCG-3' (서열번호 27)를 사용하였고, PDFG-AA 특이적 프라이머는 PDGF-AA forward: 5'-CTGCCCATTCGGAGGAAGAGAA-3' (서열번호 28) 및 PDGF-AA reverse: 5'-TGGCACTTGACACTGCTCGTGTT-3' (서열번호 29)를 사용하였다. 표적 mRNA의 상대적 정량은 CT 방법으로 분석하였다.
bFGF, IGF, Wnt7a 및 PDGF-AA에 대한 웨스턴 블롯을 수행하기 위해, 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포를 10% FBS을 포함하는 DMEM 배지에서 100% 포화될 때까지 배양한 후 수득하고, 세포를 파쇄한 후 단백질이 함유된 상층액 30 ㎍을 SDS-PAGE(sodium dodecyl sulfate-polyacrylamide gel electrophoresis)에 전개하여 분리하고, 니트로셀룰로오즈 막(nitrocellulose membrane)으로 이동하여 4% 스킴밀크(skim milk)로 블로킹(blocking)하였다. 이후, 막에 1차 항체로 항-bFGF 항체(Santa Cruz Biotechnology, USA), 항-IGF 항체(Santa Cruz Biotechnology, USA), 항-Wnt7a 항체(Santa Cruz Biotechnology, USA) 및 항-PDGF-AA 항체(Milipore, Germany)를 처리하여 4℃에서 밤새 배양하고 TBST(0.1% Tween 20 첨가된 Tris-Buffered Saline(TBS))로 세척하였다. 2차 항체로 마우스 및 염소 유래의 항-마우스 IgG 항체(goat anti-mouse IgG)(Santa Cruz Biotechnology, USA)를 1% BSA(소혈청알부민, bovine serum albumin)을 포함하는 TBST와 함께 막에 처리하고 한 시간 동안 배양하여 웨스턴 블럿을 수행하였다. 발현의 정도를 비교하기 위한 대조군으로, 항-α-튜불린(tubulin) 항체(R&D, USA)을 1차 항체로 사용하여 상기의 방법과 동일한 방법을 수행하여 α-튜불린의 발현을 확인하였다.
bFGF, IGF, Wnt7a 및 PDGF-AA 단백질들의 분비 정도를 알아보기 위해, 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포에서 생산된 컨디션드 배지에 대하여 효소면역측정법(Enzyme-Linked ImmunoSorbent Assay, ELISA assay)을 수행하였다. 컨디션드 배지 내 bFGF, PDGF-AA 단백질의 양은 ELISA 키트(Raybiotech)를 사용하여 각각의 컨디션드 배지 내 단백질 함량을 확인하였다. ELISA는 컨디션드 배지를 준비한 뒤 standard 및 샘플에 biotin 항체를 1시간 동안 처리를 한 뒤 streptavidin을 45분간 처리하였다. 이후 30분간 TMB substrate reagent를 처리한 후 stop solution을 더하여 반응을 중지시켰다. 결과는 Microplate Spectrophotometer를 통해 450nm 흡광도를 측정한 뒤 정량 분석하였다.
그 결과, 나노그(Nanog) 도입 후 실시예 1에 해당되는 배지에서 3번 계대 배양을 한 양수 유래 중간엽 줄기세포에서 bFGF, IGF, Wnt7a 및 PDGF-AA의 발현이 나노그(Nanog) 도입이 되지 않고 같이 3번 계대 배양을 한 양수 유래 중간엽 줄기세포보다 증가하였음을 확인하였으며(도 8), 각각의 단백질 발현량은 도 8의 결과와 동일하였다(도 9 위). 또한, 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포의 컨디션드 배지에서 양수 유래 중간엽 줄기세포의 컨디션드 배지보다 더 많은 양의 단백질을 확인하였다(도 9 아래). 따라서, 나노그(Nanog)의 도입으로 인해 양수 유래 중간엽 줄기세포에서 모발 성장인자들의 분비가 촉진됨을 확인하였다.
< 실시예 9> 나노그 ( Nanog ) 도입 양수 유래 중간엽 줄기세포의 모발 성장 촉진 효과 인 비트로(in vitro) 검정
나노그(Nanog) 도입 양수 유래 중간엽 줄기세포의 컨디션드 배지가 실제로 모발 성장을 촉진하는지를 인 비트로에서 확인하였다.
High glucose DMEM 무혈청 배지에서 동일한 양수 세포의 수에서부터 배양액을 3일간 획득하여 1000rpm에서 10분간 원심분리(centrifugation)를 진행한 후 0.25um 필터링을 통하여 컨디션드 배지를 제조하였다. 양수 유래 중간엽 줄기세포와 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포로부터 각각 컨디션드 배지를 추출한 후 이를 모낭 세포(hair follicle dermal papilla cell)에 처리하고 2일 간격의 세포의 수를 크리스탈 바이올렛 염색(crystal violet staining) 기법으로 상대적 성장률을 측정하였다. 크리스탈 바이올렛 염색 기법은 세포를 동일한 수로 준비한 뒤 2일 또는 4일 간격으로 10% 포르말린 고정(formalin fixation)을 진행한 후 크리스탈 바이올렛 염색을 20분간 진행하였다. 그 후 10% 아세트산(acetic acid)으로 탈색(destaining)하여 아세트산의 595nm 흡광도(absorbance)를 Ultrospec 2100pro 기기를 이용하여 측정한 후 정량 분석하였다.
그 결과, 양수 유래 중간엽 줄기세포에서 유래한 컨디션드 배지보다 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포에서 유래한 컨디션드 배지를 처리한 그룹에서 모낭 세포 수가 더 많음을 확인하였다(도 10). 따라서, 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포의 컨디션드 배지가 모낭 세포의 성장에 영향을 미친다는 것을 인 비트로에서 확인하였다.
< 실시예 10> 나노그 ( Nanog ) 도입 양수 유래 중간엽 줄기세포의 모발 성장 촉진 효과 인 비보(in vivo) 검정
나노그(Nanog) 도입 양수 유래 중간엽 줄기세포의 컨디션드 배지가 실제로 모발 성장을 촉진하는지를 인 비보에서 확인하였다.
쥐의 목 부위에서부터 꼬리 부위까지 제모 도구를 이용하여 털을 모두 제거한 뒤 실시예 1의 배지에서 15번 계대배양한 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포의 컨디션드 배지를 매일 50ul씩 처리하고, 모발 성장의 초기 단계인 성장기(등 부위에 갈색 내지는 검은색의 털이 생성되는 시기)에 모발 성장 정도를 분석하였다. 음성대조군은 양수 유래 중간엽 줄기세포의 컨디션드 배지를 매일 50ul씩 처리하였고, 양성대조군은 미녹시딜(minox) 10uM/ml를 매일 50ul씩 처리하였다.
조직적 분석을 위해, 동일한 컨디션드 배지를 10일 동안 처리한 쥐의 피부 조직을 채취하여 H&E(헤마톡실린(Hematoxylin)과 에오신(Eosin)) 염색하여 분석하였다.
조직 단위의 발모 촉진 효능을 유전자 단계에서 확인하기 위해, 모발 성장단계에서 많이 발현하는 모발 형성 특이적 마커인 ALP(amplified length polymorphism), LEF, 베르시칸(Versican) 및 Hey의 mRNA 발현량을 RT-PCR로 분석하였다. 쥐의 피부 조직을 채취하여 세포를 분리하고 실시예 3과 동일한 방법으로 세포의 총 RNA를 분리하여 cDNA로 역전사하였으며, cDNA는 Taq 합성효소(Promega)와 ALP, LEF, 베르시칸 및 Hey의 mRNA에 특이적인 프라이머를 통해 증폭되었다. ALP 특이적 프라이머는 ALP forward: 5'-TGGCCCTCTCCAAGACGTACAA-3' (서열번호 30) 및 ALP reverse: 5'-TGGTTCACTCTCGTGGTGGTCA-3' (서열번호 31)를 사용하였고, LEF 특이적 프라이머는 LEF forward: 5'-CTTCCTTGGTGAACGAGTCTG-3' (서열번호 32) 및 LEF reverse: 5'-GTGTTCTCTGGCCTTGTCGT-3' (서열번호 33)를 사용하였으며, 베르시칸 특이적 프라이머는 Versican forward: 5'-AACTAGCCGTTGGAGTGGATTC-3' (서열번호 34) 및 Versican reverse: 5'-AAATGCTCTGTGGCTCTGGA-3' (서열번호 35)를 사용하였고, Hey 특이적 프라이머는 Hey forward: 5'-GCCGACGAGACCGGATCAATAA-3' (서열번호 36) 및 Hey reverse: 5'-TCCCGAAATCCCAAACTCCGAPCR-3' (서열번호 37)를 사용하였다. 결과물을 1% 아가로즈 젤에 전기영동하여, GAPDH mRNA를 기준으로 하여 상대적인 mRNA 발현량을 Quantity One 소프트웨어를 이용하여 정량화하였다.
그 결과, 외형적인 측면에서 성장기가 시작하는 시기의 털 색이 유의미한 차이를 나타냄을 확인하였으며(도 11), 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포로부터 수득한 컨디션드 배지를 처리한 그룹에서 모낭 조직이 더 많이 생성되는 것을 관찰하였다(도 12). 또한, 컨디션드 배지 처리 5일째의 조직적 분석에서 성장기 단계를 구별하였을 때, 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포의 컨디션드 배지를 처리한 그룹이 성장기 후기 단계의 형태를 나타냄을 확인하여, 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포의 컨디션드 배지가 양성대조군인 미녹시딜(minoxidil)에 비해 모발 성장 촉진 효과가 우수함을 확인하였다(도 13). 또한, 나노그(Nanog) 도입 양수 유래 중간엽 줄기세포의 컨디션드 배지를 처리한 그룹에서 ALP, LEF, Versican 및 Hey가 더 많이 발현하는 것을 확인하였다(도 14).
< 비교예 1> 나노그(Nanog)가 아닌 다른 만능성 유전자를 도입시켰을 때의 줄기세포 배양 효과 검정
양수 유래 중간엽 줄기세포에 나노그(Nanog)가 아닌 다른 만능성 유전자를 도입시킨 뒤 지속적인 배양이 가능한지 확인하기 위한 실험을 진행하였다.
구체적으로, 실시예 1에서 수득한 양수 유래 중간엽 줄기세포에 다양한 종류의 만능성 유전자를 단독으로 또는 조합하여 도입하였다. 나노그(Nanog) 유전자 도입은 실시예 2에서 제작한 pMXs-Nanog 벡터를 이용하였고, Oct4 유전자(NCBI GenBank Accession number NM_002701.5) 도입은 pMXs-hOct3/4(Addgene, Plasmid #17217) 벡터를 이용하였으며, Lin28 유전자(NCBI GenBank Accession number NM_024674.4) 도입은 pMXs-hLin28A(Addgene, Plasmid #47902) 벡터를 이용하여 실시예 2와 동일한 방법으로 수행하였다. 복수의 유전자 도입은 각각의 바이러스를 혼합하여 세포 내에 주입하는 방법을 사용하였다. 실험군은 ① Oct4 단독 도입군, ② Nanog 단독 도입군, ③ Lin28 단독 도입군, ④ Oct4 및 Lin28 조합 도입군, ⑤ Oct4 및 Nanog 조합 도입군, 그리고 ⑥ Oct4, Nanog 및 Lin28 조합 도입군(O+N+L)으로 총 6가지 군을 설정하였다. 사진은 현미경(Olypus DP70)으로 40배율로 촬영하였다.
그 결과, 양수 유래 중간엽 줄기세포는 나노그(Nanog)를 단독으로 도입하였을 때 이외에는 지속적인 성장이 불가하였다(도 15).
실시예에 따른 결과로부터, 나노그(Nanog)를 도입한 양수 유래 중간엽 줄기세포 컨디션드 배지를 적용 시 체외 및 체내에서 모발 성장 촉진 효과를 나타냄을 확인하였다.

Claims (7)
- (a) 산모로부터 얻은 양수에서 태아 유래 세포를 분리하는 단계;(b) 상기 분리된 태아 유래 세포를 FBS(fetal bovine serum), bFGF(basic fibroblast growth factor), 셀레늄(selenium) 및 아스코르브산(ascorbic acid) 함유 배지에서 계대배양하여 양수 내 태아 유래 중간엽 줄기세포를 수득하는 단계;(c) 상기 수득된 양수 내 태아 유래 중간엽 줄기세포에 나노그(Nanog)를 도입하여 나노그(Nanog)가 과발현된 양수 내 태아 유래 중간엽 줄기세포를 수득하는 단계; 및(d) 상기 수득된 나노그(Nanog)가 과발현된 양수 내 태아 유래 중간엽 줄기세포를 무혈청 배지에서 1 내지 5일간 배양하여 bFGF(basic fibroblast growth factor, 기본 섬유아세포성장인자), IGF(Insulin-like Growth Factor), Wnt7a(Wingless-type MMTV integration site family member 7A) 및 PDGF-AA(Platelet-derived Growth Factor)로 이루어진 군으로부터 선택된 어느 하나 이상의 인간 성장인자가 함유된 컨디션드 배지를 제조하는 단계;를 포함하는 나노그(Nanog)가 과발현된 양수 내 태아 유래 중간엽 줄기세포로부터 bFGF, IGF, Wnt7a 및 PDGF-AA로 이루어진 군으로부터 선택된 어느 하나 이상의 인간 성장인자를 생산하는 방법.
