Abstract

技术领域
本发明涉及一种用于促进毛发生长的组合物及其制备方法,该组合物包括源自羊水胎儿的间充质干细胞的培养液,其中,逆转录因子Nanog作为活性成分过表达。
背景技术
干细胞是由于人体内外的环境和刺激而具有分化成构成人体的各种类型细胞的能力和自我复制能力的细胞,存在三种类型已知的干细胞,例如从早期胚胎中分离的胚胎干细胞(ES细胞),从胚胎期的原始生殖细胞中分离的胚胎生殖细胞(EG细胞)和从成人骨髓中分离的多潜能成人祖细胞(MAPC细胞)。
骨髓来源的间充质干细胞作为一种类型的成人干细胞已经使用了很长时间,并且已经证明了它们的各种效果。另外,近来有报道称,从脂肪组织或其它类型的组织中分离的细胞具有与骨髓来源的间充质干细胞相似的特征。
本发明人专注于容易从孕妇或胎儿中分离的羊水。他们测试了从羊水中获得关于胎儿健康的各种信息,并且在怀孕初期直到出生后立即提取羊水而不伤害孕妇。测试中使用的羊水细胞在测试后被处置,并且当患者同意时,可以将细胞用于研究而不处置,因此与常规研究的其它成人干细胞相比,可以容易地获得大量的羊水细胞。
同时,由于人毛发具有独特的生长周期,即毛发周期包括:产绒期->退化期->休止期->生长期,始终保持恒定数量的毛发而不脱毛。然而,当秃发进行时,存在于毛发根中的毛发乳头变得更小,毛发的厚度减小,毛发周期变短,并且新生长的毛发变得更薄。因此,当秃发进行时,毛发变成蓬松的毛发,具有较短的生长周期,然后稍微生长后脱落。秃发的主要原因是遗传,并且已知雄性激素睾酮参与秃发基因的表达。许多情况下,由于老龄化、压力等原因脱发,不是因为遗传性秃发。众所周知,老龄化相关的脱发是由毛孔周围受毛细血管压迫导致的血液循环不畅引起的,这是由于根据头皮细胞减少和头皮脂肪累积量的增加,毛孔闭合导致的氧供应减少而产生的。另外,压力、不规则的生活方式和环境污染也被认为会导致秃发。
尽管秃发或脱发患者感受到的情绪痛苦非常大,但是大多数已经开发并且现在可商购的生发剂具有暂时或有限的效果,并且不能充分满足用户的需求。由于血管舒张已被证明在一定程度上是有效的适用生发剂米诺地尔和含有非那甾胺作为主要成分并由于对雄性激素的活化具有抑制作用而表现出其作用的口服生发剂非那雄胺已被广泛用作在防止脱发中效果优异的试剂。然而,上述生发剂在一定程度上有效地防止脱发,但对毛发生长并没有表现出显著影响。也就是说,当停止使用生发剂时,脱发会再次发生,并且由于长期使用,对副作用和成本增加有很大担忧。出于这个原因,大多数患者正在放弃治疗。
因此,本发明人发现了能够延长源自羊水胎儿的间充质干细胞的生长和寿命并且增强条件培养基的毛发生长作用的重编程因子,并且鉴定了条件培养基中的哪些成分对毛发生长作用是有效的。另外,本发明人通过体内实验证实了条件培养基对促进毛发密度和毛发生长的作用,从而完成了本发明。

本发明提供了一种由Nanog过表达的源自羊水胎儿的间充质干细胞生产人生长因子的方法,更特别地,涉及一种由Nanog过表达的源自羊水胎儿的间充质干细胞生产选自由碱性成纤维细胞生长因子(bFGF)、胰岛素类生长因子 (IGF)、无翅型MMTV整合位点家族成员7A(Wnt7a)和血小板来源生长因子 (PDGF-AA)组成的组中的任一种或多种人生长因子的方法,并且更具体地,涉及一种用于由Nanog过表达的源自羊水胎儿的间充质干细胞生产选自由 bFGF、IGF、Wnt7a和PDGF-AA组成的组中的任一种或多种人生长因子的方法,该方法包括:
(a)从由孕妇获得的羊水中分离胎儿细胞;

在步骤(a)中,胎儿细胞可以包括在由孕妇获得的羊水中。
本文使用的术语“间充质干细胞(MSC)”是指生成骨、软骨、脂肪、骨髓基质、肌肉或神经来源的细胞,以及通常在成人中保留在骨髓中的细胞,但也存在于脐带血、外周血和其它组织中,并由此获得。由于由孕妇获得的羊水含有胎儿体内生成的各种化学物质,所以可以生成人体内的大部分细胞,并且很容易进行采样。此外,证实了异质细胞存在于羊水中,并且在这些细胞中,存在具有与成纤维细胞相同形状的均质MSC,这是MSC的特征。
本文使用的术语“培养基”是指能够在体外维持羊水中胎儿细胞的生长和存在的培养基,并且包括本领域常规使用的所有适用于培养羊水中胎儿细胞的培养基。培养基和培养条件可以根据细胞的类型来选择。培养基优选为细胞培养最低限度培养基(CCMM),通常包括碳源、氮源和微量元素。这种CCMM包括Dulbecco改良Eagle培养基(DMEM)、最小必需培养基(MEM)、Eagle基础培养基(BME)、RPMI1640、F-10、F-12、α最小必需培养基(αMEM)、Glasgow 最小必需培养基(GMEM)和Iscove改良Dulbecco培养基(IMEM)。另外,细胞培养最低限度培养基(CCMM)可以包括抗生素,例如青霉素、链霉素或庆大霉素。
在步骤(b)中,可以通过在含有FBS、bFGF、硒和抗坏血酸的碱性培养基中培养从羊水中分离的细胞来获得源自羊水胎儿的MSC,优选地,通过在补充有bFGF、硒和抗坏血酸的含FBS的低葡萄糖DMEM中培养细胞来获得,并且更优选地,通过在补充有4ng/ml的bFGF、5ng/ml的硒、50μg/ml的抗坏血酸、 1%的L-谷氨酰胺和1%的青霉素-链霉素的10%FBS低葡萄糖DMEM中培养细胞来获得,但是本发明并不限于此。
在步骤(c)中,可以使用逆转录病毒载体引入Nanog,并且优选使用pMXs 病毒载体,但是本发明并不限于此。
在步骤(c)中,Nanog可以是由SEQ ID NO:1表示的碱基序列组成的基因,并且优选人(智人)来源的Nanog基因,但是本发明并不限于此。
在本发明的一个示例性实施方式中,证实了根据将Nanog引入到MSC中并培养细胞的步骤增加MSC的生长和增殖。
本文使用的术语“Nanog”是重编程因子,其起源于“重编程”,其是由 Yamanaka教授及其团队在2006年引入的概念。所有成人组织通过正常发展过程逐渐变得差异化,并且改变为功能特异性细胞。其中,受精卵的细胞是全能性的,并且正在发育成囊胚,可以区分为内细胞团细胞和外细胞。内细胞团细胞可以生成胚胎体细胞和生殖细胞,这称为多能性。内细胞团细胞也称为胚胎干细胞,并表达多能性特异性基因(重编程因子)。基因的具体实例是Oct4、Sox2、 Nanog和Lin28。重编程是在体细胞中诱导这种多能性特异性基因表达的技术,并且因此使细胞具有与胚胎干细胞相似的性质。根据这样的背景,本发明在各种重编程因子中独立使用Nanog。因此,1)当引入多个外源基因时,存在对细胞特征变化的担忧,2)当使用病毒引入多种因子时,难以成功构建细胞系,以及3)证实了当独立引入Nanog时,可以连续维持细胞系(参见图15)。

本文使用的术语“条件培养基”是指通过使用液体悬浮培养物使细胞生长,当通过离心或过滤在细胞分裂期间细胞达到指数期时除去分裂的细胞,从而仅获得培养溶液并将培养溶液与培养基质混合而制备的培养基。该培养基使用从培养基中的分裂细胞提取的未知生长因子,并广泛用于低密度细胞培养或原生质体培养。

因此,使用包含条件培养基作为活性成分制备用于促进毛发生长的组合物的方法,已知具有毛发生长作用的生长因子例如bFGF、IGF、Wnt7a、PDFG-AA 或其两种或更多种的组合的表达水平显著增加,因此可以进一步提高增强毛发生长的效果。
在本发明的一个示例性实施方式中,证实了当在组织学分析中区分生长期的步骤时,从Nanog引入的源自羊水的MSC获得的条件培养基处理组中生成的大量毛囊组织显示出在生长期随后的步骤中形成的形状,并且还证实了扩增长度多态性(ALP)、LEF、蛋白聚糖(Versican)和Hey的mRNA在毛发生长期高度表达。
此外,本发明提供了一种用于促进毛发生长的组合物,其包含通过培养 Nanog过表达的源自羊水胎儿的MSC而制备的条件培养基作为活性成分,具体地,提供了一种用于促进毛发生长的组合物,其包含条件培养基作为活性成分,所述条件培养基含有选自由bFGF、IGF、Wnt7a和PDFG-AA组成的组中的任一种或多种人生长因子,并且由Nanog过表达的源自羊水胎儿的MSC产生,更具体地,提供了一种通过制备用于促进毛发生长的组合物的方法制备的用于促进毛发生长的组合物。
在本发明的一个示例性实施方式中,证实了通过培养过表达Nanog的羊水来源的MSC制备的条件培养基显示出培养基中毛发生长促进剂表达水平的显著增加和用该培养基处理小鼠时的快速毛发生长,从而促进毛发生长、毛发再生和防止脱发。
另外,本发明提供了一种用于促进毛发生长的化妆品组合物,其包含用于促进毛发生长的组合物。

化妆品组合物的产品含有美容或皮肤病学可接受的介质或基质。它可以以适用于局部应用的所有剂型而制备,例如溶液、凝胶、固体、无水糊剂产品、通过将油相分散在水相中获得的乳化剂、悬浮液、微乳化剂、微胶囊、微粒细胞或离子(脂质体)和非离子型囊泡分散剂、霜剂、皮肤软化剂、洗剂、粉剂、药膏、喷雾剂或遮瑕膏。该组合物可以通过本领域常规使用的方法制备。根据本发明的组合物还可以以还含有压缩推进剂的泡沫或气雾剂组合物的形式使用。
对本发明的化妆品组合物制剂没有特别限制,其可以配制成化妆品产品,例如保湿洗剂、收敛剂、滋养洗剂、滋养霜、按摩霜、香精、眼霜、眼精华、清洁霜、清洁泡沫、清洁水、面膜、粉剂、身体洗剂、身体霜、身体油、身体精华、头发营养液、护发素、头发精华、头发洗剂、头发滋养洗剂、洗发水、染发液、头发护理剂、护发霜、头发滋养霜、头发保湿霜、头发按摩霜、发蜡、头发气雾剂、头发发膜、头发滋养发膜、头发皂、头发清洁泡沫、头发油、头发干燥制剂、头发保护护理剂、染发剂、烫头制剂、头发脱色剂、发胶、头釉、美发剂、头发啫喱、头发保湿剂、头发摩丝或喷发定型剂。
如上所述的化妆品组合物可以施用于皮肤上,或者使用微针施用以被吸收到皮肤中。
另外,本发明提供了一种用于促进毛发生长的药物组合物,其包含用于促进毛发生长的组合物。
该药物组合物可以用于各种有效促进毛发生长、毛发再生和防止脱发的美发产品中。
相对于100重量份的总药物组合物,其可以包含0.001至10重量份的药物组合物。
药物组合物可以制备成用于促进毛发生长的片剂、胶囊剂、粉剂、颗粒剂、注射剂、凝胶剂、乳化剂、糖浆剂、气雾剂、贴剂、喷雾剂、霜剂、膏剂、膏药、洗剂、擦剂、糊剂或泥敷剂,但本发明并不限于此。

根据目的,药物组合物可以胃肠外或口服施用,并且以每千克体重 0.1-500mg(优选1-100mg)的日剂量施用一次或数次。用于特定患者的剂量可以根据患者的体重、年龄、性别、健康状况、饮食、施用时间、施用方法、施用途径、排泄速率和疾病的严重程度而变化。
药物组合物可以通过各种途径施用,例如胃肠外或口服施用于哺乳动物诸如大鼠、小鼠、家畜和人。可预期所有施用途径,并且可通过口服或通过直肠、静脉内、肌内、皮下、子宫内、硬膜外或脑室内注射进行施用。
生产MSC条件培养基的主要限制与干细胞老龄化高度相关。细胞老龄化诱导低细胞生长、降低生长因子和细胞因子的量,并导致细胞死亡。另外,细胞老龄化也诱导细胞来源的条件培养基的定量和定性损失,并且使得难以维持均一组合物,导致难以大规模生产和条件培养基以及包含其作为一种有效成分的组合物的产业化。因此,本发明人引入重编程技术以培养MSC。根据以前的研究,已知根据MSC中多能性因子的表达发生干细胞方面的变化。在这方面,将各种多能性因子Oct4、Nanog和Lin28引入到羊水来源的MSC中,并且其中可以看出Nanog引入的羊水来源的MSC可以建立为细胞系并且连续培养,但其它基因的引入诱导细胞死亡,并难以连续培养细胞(参照图15)。此外,证实了 Nanog引入的羊水干细胞比传统的羊水干细胞在增强细胞生长和保存干细胞方面的作用中进一步改善。
在本发明的一个示例性实施方式中,证实了由于引入Nanog的羊水来源的 MSC的数量远高于羊水来源的干细胞(图4),所以Nanog对羊水来源的MSC 生长的改善具有积极作用。Nanog引入的羊水来源的MSC较少表达β-半乳糖苷酶(图5左侧),并且表现出比羊水来源的MSC更低表达水平的p53和p21(图 5右侧)。因此,证实了由于Nanog的引入,延长了羊水来源的MSC的寿命的效果。另外,在Nanog引入的羊水来源的MSC中的多能性特异性标记物Oct4和Sox2的表达增强(图7),因此即使通过引入Nanog,羊水来源的MSC的干细胞特性是保守的。
在本发明的一个示例性实施方式中,证实了与羊水来源的MSC相比,在 Nanog引入的羊水来源的MSC中促进毛发生长的生长因子bFGF、IGF、Wnt7a 和PDGF-AA的表达增加(图8和9),并且由于Nanog的引入,在羊水来源的 MSC中促进了毛发生长因子的分泌。
在本发明的一个示例性实施方式中,观察到在从Nanog引入的羊水来源的 MSC获得的条件培养基处理组中产生更多的毛囊组织(图12)。另外,在组织学分析中区分生长期步骤时,证实了用Nanog引入的羊水来源的MSC的条件培养基处理的组显示出在生长期的后续步骤中的形状(图13),并且在用Nanog 引入的羊水来源的MSC的条件培养基处理的组中高度表达在生长期步骤中高度表达的扩增长度多态性(ALP)、LEF、蛋白聚糖和Hey的mRNA(图14)。因此,证实了当应用Nanog引入的羊水来源的MSC的条件培养基时,在体外和体内表现出毛发生长促进作用。
技术效果
在本发明中,将作为重编程因子的Nanog引入并在源自羊水胎儿的MSC中过表达,MSC的生长、干细胞特性和寿命得到改善,并且由细胞分泌的生长因子的表达增加。另外,由于通过培养Nanog引入的源自羊水胎儿的MSC而制备的条件培养基表现出毛发生长促进作用,所以该细胞可以用作用于促进毛发生长的化妆品和药物组合物。

在下文中,将参考有助于理解本发明的实施例来详细描述本发明。然而,仅提供以下实施例来说明本发明,本发明的范围并不限于此。提供本发明的实施例是为了更全面地向本领域的普通技术人员描述本发明。
<实施例1>羊水来源的MSC的分离和培养
通过在含有10%FBS、1%青霉素/链霉素、1%L-谷氨酰胺、4ng/ml bFGF、 5ng/ml硒和50μg/ml抗坏血酸的低葡萄糖Dulbecco改良Egale培养基(DMEM) 中将从孕妇获得的羊水中分离出的胎儿细胞传代培养来获得羊水胎儿来源的 MSC。培养基的这种条件与bFGF处理、bFGF和硒处理、bFGF和抗坏血酸处理的条件相比表现出羊水来源的MSC的生长效果进一步提高(图1)。
通过检查细胞培养2至4天后的生长速率来评估羊水来源MSC的生长效果,并且通过结晶紫染色进行定量。制备相同细胞数的样品,然后以2天或4天的间隔进行10%福尔马林固定,然后对细胞进行结晶紫染色20分钟。细胞用10%乙酸脱色,并通过使用Ultrospec2100pro分光光度计测量595nm处的乙酸的吸光度来进行定量分析。
在与上述相同的条件下,将获得的羊水胎儿来源的MSC在DMEM中培养。
<实施例2>Nanog引入的羊水来源的MSC的制备
通过使用逆转录病毒载体系统将Nanog基因引入到实施例1中获得的羊水来源的MSC中以诱导Nanog过表达来制备Nanog引入的羊水来源的间充质干细胞系(#1、#2、#3)(图2)。
具体而言,用限制性内切酶从pBS-Nanog载体(Yamanaka实验室的质粒储存#A4,日本)分离Nanog基因(NCBI GenBank登录号NM_024865.3;SEQ ID NO:1)。使用T4连接酶将分离的Nanog基因连接到pMXs载体(Cell Biolabs,日本)中,从而制造pMXs-Nanog。使用转染试剂将pMXs-Nanog载体转染到 293GPG细胞中6小时。72小时后,产生具有Nanog基因的病毒,并分离上清液,以2000rpm离心10分钟,并使用0.45μm过滤器过滤。将病毒引入到已生长至80%融合6小时的羊水来源的MSC中,导致感染进入到细胞中。
当制备的Nanog引入的羊水来源的MSC在实施例1的培养基中培养时,细胞连续并正常生长。
<实施例3>证实Nanog引入的羊水来源的MSC的Nanog表达的测定
为了证实实施例2中制备的Nanog引入的羊水来源的MSC系(#1、#2、#3) 中的Nanog过表达是否合适地发生,在Nanog mRNA上进行逆转录聚合酶链反应(RT-PCR)和免疫荧光染色。
为了进行RT-PCR,根据制造商的方案,用TRIzol试剂(Invitrogen,美国) 处理Nanog引入的羊水来源的MSC以分离总RNA。使用cDNA制备混合物 (Bioneer)将500μg分离的总RNA逆转录为cRNA。试管在45℃保持60分钟,在95℃保持5分钟,然后保存于-20℃备用。使用Taq聚合酶(Promega)和Nanog mRNA特异性引物(Exo-Nanog正向:5'-GCTTGGATACACGCCGC-3'(SEQ ID NO:2);和Exo-Nanog反向:5'-GATTGTTCCAGGATTGGGTG-3'(SEQ ID NO: 3)扩增cDNA。在94℃20秒、60℃30秒和72℃2分钟的35个循环中重复进行 RT-PCR,并进一步在72℃10分钟以进行最终合成。通过在1%琼脂糖凝胶中电泳分析PCR产物。
对于免疫荧光染色,将Nanog引入的羊水来源的MSC用PBS洗涤,用4%多聚甲醛固定1小时,并用添加0.5%Triton-100的磷酸盐缓冲盐水(PBS)(0.5% PBST)洗涤三次,每次5分钟。用含有3%FBS的PBST封闭细胞1小时,以 1:50的比例用检测Nanog受体的抗体(AF276,R&D)处理1小时。之后,用 0.5%PBST洗涤细胞3次,每次5分钟,然后将以1:200比例稀释的荧光标记的二抗(Alexa Fluor 488山羊抗人IgG,#A11013,Invitrogen,美国)处理1小时。用1:1000的4',6-二脒基-2-苯基吲哚(DAPI)溶液处理5分钟以进行核染色后,以与上述相同的方式用PBST洗涤细胞三次。之后,使用荧光显微镜(Olympus DP70)使样品可视化。
作为结果,与羊水来源的MSC相比,Nanog引入的羊水来源的MSC显示出外源Nanog基因表达水平在mRNA水平上和Nanog表达水平在蛋白水平上的显著增加,证实了Nanog基因的过表达(图3)。
<实施例4>证实Nanog引入的羊水来源的MSC的细胞生长改善效果的测试
比较在实施例1的培养基中连续培养的实施例2中制备的Nanog引入的羊水来源的间充质干细胞系(#1、#2、#3)和不引入Nanog的羊水来源的干细胞的生长速率并进行分析。
使用实施例1的培养基对干细胞进行13次传代培养,每隔3天使用Countingchamber(MARIENFELD,Germany)测定各细胞系的细胞数,随后进行比较。
作为结果,证实了Nanog引入的羊水来源的MSC的细胞数显著高于羊水来源的干细胞(图4)。该结果显示出Nanog对羊水来源的MSC的细胞生长改善具有积极的效果。
<实施例5>证实Nanog引入的羊水来源的MSC的延长寿命效果的测试
为了测试Nanog引入的羊水来源的MSC的寿命,通过β-半乳糖苷酶染色和细胞死亡相关蛋白p53和p21的mRNA表达水平分析由连续传代培养引起的细胞死亡程度。
对于β-半乳糖苷酶染色,将在实施例1的培养基中传代培养35次的Nanog 引入的羊水来源的MSC以5×104个细胞/孔的密度接种在6孔板中,接着过夜,用PBS洗涤,然后固定。根据β-半乳糖苷酶染色试剂盒(Cell Signaling Technology,Beverly,MA)的方案,将pH值6.0X-gal发色底物在37℃培养过夜,然后用放大倍数为100倍的显微镜(OlympusDP70)观察颜色变化。
为了测量细胞死亡相关蛋白p53和p21的mRNA表达水平,通过与实施例 3中所述相同的方法从传代培养35次的Nanog引入的羊水来源的MSC中提取总RNA,并从总RNA中扩增cDNA。用Taq聚合酶(Promega)和对p53、p21 和甘油醛-3-磷酸脱氢酶(GAPDH)的mRNA特异性的引物扩增cDNA。p53特异性引物是p53正向引物:5’-CCTCACCATCATCACACTGG-3’(SEQ IDNO:4) 和p53反向引物:5’-TTATGGCGGGAGGTAGACTG-3’(SEQ ID NO:5),p21 特异性引物是p21正向引物:5’-GGAAGACCATGTGGACCTGT-3’(SEQ ID NO: 6)和p21反向引物:5’-AGGCAGAAGATGTAGAGCGG-3’(SEQ ID NO:7),并且GAPDH特异性引物是GAPDH正向引物: 5’-GTGGTCTCCTCTGACTTCAACA-3’(SEQ ID NO:8)和GAPDG反向引物: 5’-CTCTTCCTCTTGTGCTCTTGCT-3’(SEQ ID NO:9)。通过在1%琼脂糖凝胶中电泳分析PCR产物,并使用基于GAPDH mRNA的Quantity One软件定量相对mRNA表达水平。
作为结果,证实了在实施例1的培养基中连续传代培养羊水来源的MSC(引入Nanog后)相比于在与实施例1相同的培养基中连续传代培养羊水来源的MSC (不引入Nanog),β-半乳糖苷酶表达较少(图5的左侧),并且p53和p21表达水平降低(图5的右侧)。因此,证实了Nanog引入导致延长羊水来源的MSC 的寿命的效果。
<实施例6>证实Nanog引入的羊水来源的MSC的独特性质的变化
为了证实干细胞特异性表达标志物的表达是否因Nanog引入而变化,通过 RT-PCR确定纤连蛋白、MMP1、Snail和Slug表达模式。
对于RT-PCR,通过与实施例3中所述相同的方法进行RNA提取和PCR,并且本文使用的引物如下。纤连蛋白特异性引物是纤连蛋白正向引物: 5’-GACGACTCCCTTTTCTCCTCTT-3’(SEQ ID NO:10)和纤连蛋白反向引物: 5’-TGAGTTCTGTGCTGCTACCTTC-3’(SEQ ID NO:11),MMP1特异性引物是 MMP1正向引物:5’-TTGAGAAAGCCTTCCAACTCTG-3’(SEQ ID NO:12)和MMP1反向引物:5’-CCGCAACACGATGTAAGTTGTA-3’(SEQ ID NO:13), Snail特异性引物是Snail正向引物:5’-CTCCTTCGTCCTTCTCCTCTACTT-3’ (SEQ ID NO:14)和Snail反向引物:5’-TCTTGACATCTGAGTGGGTCTG-3’ (SEQ ID NO:15),并且Slug特异性引物是Slug正向引物:5’-GACCCTGGTTGCTTCAAGGACA-3’(SEQ ID NO:16)和Slug反向引物: 5’-TTGTCATTTGGCTTCGGAGTGA-3’(SEQ ID NO:17)。
根据RT-PCR的结果,证实了Nanog引入的羊水来源的MSC和羊水来源的 MSC之间MSC特异性标记物的表达没有差异(图6)。因此,Nanog引入对羊水来源的MSC的独特性质没有太大效果。
<实施例7>证实Nanog引入的羊水来源的MSC中的多能性基因表达的测试
已知多能性干细胞表达少量多能性特异性标记物Oct4和Sox2(Riekstina 等,Embryonic stem cell marker expression pattern in human MSCs derived from bonemarrow,adipose tissue,heart and dermis.Stem cell rep 5(4):378-386.(2009))。为了证实多能性干细胞是否由于Nanog引入而在干细胞中表达,通过定量逆转录聚合酶链式反应(qRT-PCR)分析实施例2中制备的Nanog引入的羊水来源的MSC Oct4和Sox2的mRNA表达水平。
以与上述RT-PCR中的RNA制备相同的方式进行qRT-PCR,从而获得RNA 并制造cDNA。使用CFX-96PCR系统对由此制备的cDNA进行qRT-PCR,并使用Oct4和Sox2的mRNA特异性引物进行扩增。Oct4特异性引物是Oct4正向引物:5’-GACAGGGGGAGGGGAGGAGCTAGG-3’(SEQ ID NO:18)和Oct4 反向引物:5’-CTTCCCTCCAACCAGTTGCCCCAAAC-3’(SEQ ID NO:19),Sox2 特异性引物是Sox2正向引物:5’-ACCAATCCCATCCACACTCACGCA-3’(SEQ ID NO:20)和Sox2反向引物:5’-GCAAACTTCCTGCAAAGCTCCTACCG-3’ (SEQ ID NO:21)。通过比较阈值(CT)循环分析靶标mRNA的相对量(Johnson MR等,Anal Biochem 2000;278:175-184)。
作为结果,证实了Nanog引入的羊水来源的MSC中的多能性特异性标记物 Oct4和Sox2的表达进一步增强(图7)。因此,由于Nanog引入,羊水来源的 MSC中的多能性特异性基因的表达增加。
<实施例8>证实由于Nanog过表达的羊水来源的MSC中的毛发生长因子表达的测试
为了证实Nanog引入的羊水来源的MSC中毛发生长因子的表达,通过 qRT-PCR和免疫印迹(western blotting)分析毛发相关生长因子如bFGF、IGF、 Wnt7a和PDGF-AA的mRNA表达水平和蛋白表达水平。
通过与实施例7中所述相同的方法进行qRT-PCR,并使用对bFGF、IGF、 Wnt7a和PDGF-AA的mRNA特异性的引物扩增cDNA。bFGF特异性引物是 bFGF正向引物:5’-CAGATTAGCGGACGCGGTGC-3’(SEQ ID NO:22)和bFGF 反向引物:5’-TCACGGATGGGTGTCTCCGC-3’(SEQ ID NO:23),IGF特异性引物是IGF正向引物:5’-CCATGTCCTCCTCGCATCTCTTCT-3’(SEQ ID NO: 24)和IGF反向引物:5’-CCATACCCTGTGGGCTTGTTGAA-3’(SEQ ID NO: 25),Wnt7a特异性引物是Wnt7a正向引物: 5’-TCTTTCTCAGCCTGGGCATGGT-3’(SEQ ID NO:26)和Wnt7a反向引物: 5’-TCCTATGACGATGATGGCGTCG-3’(SEQ ID NO:27),并且PDFG-AA特异性引物是PDGF-AA正向引物:5’-CTGCCCATTCGGAGGAAGAGAA-3’(SEQ ID NO:28)和PDGF-AA反向引物:5’-TGGCACTTGACACTGCTCGTGTT-3’ (SEQ ID NO:29)。通过CT方法分析靶标mRNA的相对量。
为了对bFGF、IGF、Wnt7a和PDGF-AA进行免疫印迹,在含有10%FBS 的DMEM中培养以生长直至100%融合并破碎后获得Nanog引入的羊水来源的 MSC,然后用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)显色30μg含有蛋白的上清液以分离蛋白。将蛋白转移到硝酸纤维素膜上,用4%脱脂奶粉封闭。之后,用一抗如抗-bFGF抗体(Santa CruzBiotechnology,美国)、抗-IGF 抗体(Santa Cruz Biotechnology,美国)、抗-Wnt7a抗体(Santa Cruz Biotechnology,美国)和抗-PDGF-AA抗体(Millipore,德国)处理该膜,在4℃培养过夜,并用TBST(添加0.1%吐温20的Tris缓冲盐水(TBS))洗涤。用小鼠和山羊来源的抗鼠IgG抗体(山羊抗小鼠IgG;Santa Cruz Biotechnology,美国)作为二抗和含有1%牛血清白蛋白(BSA)的TBST处理该膜,培养1小时,然后进行免疫印迹。作为比较表达程度的对照,使用抗α-微管蛋白抗体(R&D,美国) 作为一抗,通过与上述相同的方法证实α-微管蛋白的表达。
为了确定bFGF、IGF、Wnt7a和PDGF-AA蛋白的分泌程度,在由Nanog 引入的羊水来源的MSC产生的条件培养基上进行酶联免疫吸附测定(ELISA测定)。条件培养基中的bFGF和PDGF-AA蛋白的量通过使用ELISA试剂盒 (RayBiotech)测量条件培养基中的蛋白含量来确定。对于ELISA,制备条件培养基,将标准品和样品用生物素抗体处理1小时,然后用链霉亲和素处理45分钟。之后,将标准品和样品用TMB底物试剂处理30分钟,然后用停止溶液处理以停止反应。使用微孔板分光光度计通过测量450nm处的吸光度对结果进行定量分析。
作为结果,证实了在对应于实施例1的培养基中传代培养三次的羊水来源的MSC(Nanog引入后)中的bFGF、IGF、Wnt7a和PDGF-AA的表达高于不引入Nanog的传代培养三次的羊水来源的MSC(图8),并且蛋白的表达水平与图8所示的结果相同(图9的上部)。另外,在Nanog引入的羊水来源的MSC 的条件培养基中比在羊水来源的MSC的条件培养基(图9的底部)中鉴定出更大量的蛋白。因此,证实了由于Nanog引入而导致促进了羊水来源的MSC中的毛发生长因子的分泌。
<实施例9>证实Nanog引入的羊水来源的MSC促进毛发生长效果的体外测试
进行体外测试以证实Nanog引入的羊水来源的MSC的条件培养基是否实际上促进毛发生长。
通过从高葡萄糖无血清DMEM获得相同数量羊水来源的细胞的培养液三天,以1000rpm进行离心10分钟,并使用0.25μm过滤器过滤所得产物来制备条件培养基。从羊水来源的MSC和Nanog引入的羊水来源的MSC中提取条件培养基,并用于处理毛囊细胞(毛囊皮肤乳头细胞),然后每隔一天通过结晶紫染色测量细胞数以确定相对生长率。在样品制备成具有相同数量的细胞后20分钟进行结晶紫染色,并以2或4天的间隔进行10%福尔马林固定。之后,细胞用10%乙酸脱色,并通过使用Ultrospec 2100pro分光光度计测量595nm处的乙酸的吸光度以定量分析。
作为结果,证实了用源自Nanog引入的羊水来源的MSC的条件培养基处理组中的毛囊细胞数量高于源自羊水来源的MSC的条件培养基的处理组中的毛囊细胞数量(图10)。因此,体外证实了Nanog引入的羊水来源的MSC的条件培养基影响毛囊细胞的生长。
<实施例10>证实Nanog引入的羊水来源的MSC促进毛发生长效果的体内测试
进行体内测试以证实Nanog引入的羊水来源的MSC的条件培养基是否实际上促进毛发生长。
使用剃刮工具将小鼠从头到尾彻底剃光,每天用50μl在实施例1的培养基中传代培养15次的Nanog引入的羊水来源的MSC条件培养基处理,分析头发生长早期的头发生长程度,即生长期(在背部产生棕色或黑色毛发的时期)。作为阴性对照,每天用50μl羊水来源的MSC条件培养基处理,并且作为阳性对照,每天用50μl 10μM/ml米诺地尔处理。
对于组织学分析,收集用相同的条件培养基处理10天的小鼠的表皮组织,并通过苏木精和伊红(H&E)染色进行分析。
为了鉴定基因水平的组织单位毛发生长促进效率,通过RT-PCR分析在毛发生长步骤中高表达的毛发形成特异性标记物例如ALP、LEF、蛋白聚糖和Hey 的mRNA表达水平。收集小鼠的表皮组织,分离细胞,用与实施例3中所述相同的方法分离细胞的总RNA,并逆转录为cDNA,并使用Taq聚合酶(Promega) 和对ALP、LEF、蛋白聚糖和Hey mRNA特异性的引物扩增cDNA。ALP特异性引物是ALP正向引物:5’-TGGCCCTCTCCAAGACGTACAA-3’(SEQ ID NO: 30)和ALP反向引物:5’-TGGTTCACTCTCGTGGTGGTCA-3’(SEQ ID NO:31), LEF特异性引物是LEF正向引物:5’-CTTCCTTGGTGAACGAGTCTG-3’(SEQ ID NO:32)和LEF反向引物:5’-GTGTTCTCTGGCCTTGTCGT-3’(SEQ ID NO: 33),蛋白聚糖特异性引物是蛋白聚糖正向引物:5’-AACTAGCCGTTGGAGTGGATTC-3’(SEQ ID NO:34)和蛋白聚糖反向引物: 5’-AAATGCTCTGTGGCTCTGGA-3’(SEQ ID NO:35),Hey特异性引物是Hey 正向引物:5’-GCCGACGAGACCGGATCAATAA-3’(SEQ ID NO:36)和Hey 反向引物:5’-TCCCGAAATCCCAAACTCCGAPCR-3’(SEQ ID NO:37)。通过在1%琼脂糖凝胶中的电泳分析所得产物,使用基于GAPDH mRNA的Quantity One软件定量相对mRNA表达水平。
作为结果,证实了就外观而言,当生长期开始时,毛发颜色存在显著差异 (图11),并且观察到在用由Nanog引入的羊水来源的MSC获得的条件培养基处理组中产生更多的毛囊组织(图12)。另外,在处理条件培养基后的第5天,在组织学分析中区分生长期的步骤时,证实了用Nanog引入的羊水来源的MSC 的条件培养基处理组显示出在生长期的后期步骤中的形状,从而证实了Nanog 引入的羊水来源的MSC的条件培养基比阳性对照米诺地尔具有更好的毛发生长促进效果(图13)。另外,证实了ALP、LEF、蛋白聚糖和Hey在用Nanog引入的羊水来源的MSC的条件培养基处理组中更高度地表达(图14)。
<比较例1>证实在引入不同多能性基因代替Nanog时干细胞培养效果的测试
进行了测试以证实在引入不同多能性基因而不是Nanog后,羊水来源的 MSC是否能够继续培养。
具体而言,将各种类型的多能性基因单独或以其组合引入到实施例1中获得的羊水来源的MSC中。使用实施例2中制造的pMXs-Nanog载体引入Nanog 基因,使用pMXs-hOct3/4(Addgene,质粒#17217)载体引入Oct4基因(NCBI GenBank登录号NM_002701.5),并使用pMXs-hLin28A(Addgene,质粒#47902) 载体引入Lin28基因(NCBI GenBank登录号NM_024674.4),并通过与实施例2 中所述相同的方法进行这种引入。通过将病毒组合注入细胞来进行多种基因的引入。存在6个实验组,例如①仅引入Oct4的组,②仅引入Nanog的组,③仅引入Lin28的组,④引入Oct4和Lin28组合的组,⑤引入Oct4和Nanog组合的组,以及⑥引入Oct4、Nanog和Lin28组合的组(O+N+L)。使用显微镜(Olympus DP70)以40倍放大率拍摄图像。
作为结果,除了仅将Nanog引入到细胞中时,羊水来源的MSC不可能连续生长(图15)。
根据实施例的结果,证实了当施用Nanog引入的羊水来源的MSC的条件培养基时,体外和体内表现出毛发生长促进效果。
<制备例1>头发洗剂的制造
根据制造头发洗剂的传统方法,如表1所示,制造包含本发明的Nanog引入的羊水来源的MSC的条件培养基作为活性成分的头发洗剂。

按照传统的方法制造亲水性软膏,如表2所示,制造包含本发明的Nanog 引入的羊水来源的MSC的条件培养基作为活性成分的亲水性软膏。

Claims (4)
1.一种由Nanog过表达的源自羊水胎儿的间充质干细胞生产包括碱性成纤维细胞生长因子(bFGF)、胰岛素类生长因子(IGF)、无翅型MMTV整合位点家族成员7A(Wnt7a)和血小板来源生长因子PDGF-AA的人生长因子的方法,所述方法包括:
(a)从由孕妇获得的羊水中分离胎儿细胞;
(b)通过在含有胎牛血清(FBS)、bFGF、硒和抗坏血酸的培养基中传代培养分离的所述胎儿细胞来获得源自羊水胎儿的间充质干细胞;
(c)通过将Nanog引入到所获得的源自羊水胎儿的间充质干细胞中来获得Nanog过表达的源自羊水胎儿的间充质干细胞;以及
(d)通过在无血清培养基中培养所获得的Nanog过表达的源自羊水胎儿的间充质干细胞1至5天来制备含有bFGF、IGF、Wnt7a和PDGF-AA的条件培养基。
2.根据权利要求1所述的方法,其中,在步骤(c)中,使用逆转录病毒载体引入Nanog。
3.一种制备用于促进毛发生长的组合物的方法,所述组合物包含条件培养基作为活性成分,所述条件培养基含有碱性成纤维细胞生长因子(bFGF)、胰岛素类生长因子(IGF)、无翅型MMTV整合位点家族成员7A(Wnt7a)和血小板来源生长因子PDGF-AA,并且来自Nanog过表达的源自羊水胎儿的间充质干细胞,所述方法包括:
(a)从由孕妇获得的羊水中分离胎儿细胞;
(b)通过在含有胎牛血清(FBS)、bFGF、硒和抗坏血酸的培养基中传代培养分离的所述胎儿细胞来获得源自羊水胎儿的间充质干细胞;
(c)通过将Nanog引入到所获得的源自羊水胎儿的间充质干细胞中来获得Nanog过表达的源自羊水胎儿的间充质干细胞;
(d)通过在无血清培养基中培养所获得的Nanog过表达的源自羊水胎儿的间充质干细胞1至5天来制备含有bFGF、IGF、Wnt7a和PDGF-AA的条件培养基;以及
(e)制备包含所制备的条件培养基作为活性成分的组合物。
4.根据权利要求3所述的方法,其中,在步骤(c)中,使用逆转录病毒载体引入Nanog。
